    • The Global Consortium for the Classification of Fungi and fungus-like taxa is an international initiative of more than 550 mycologists to develop an electronic structure for the classification of these organisms. The members of the Consortium originate from 55 countries/regions worldwide, from a wide range of disciplines, and include senior, mid-career and early-career mycologists and plant pathologists. The Consortium will publish a biannual update of the Outline of Fungi and fungus-like taxa, to act as an international scheme for other scientists. Notes on all newly published taxa at or above the level of species will be prepared and published online on the Outline of Fungi website (https://www.outlineoffungi.org/), and these will be finally published in the biannual edition of the Outline of Fungi and fungus-like taxa. Comments on recent important taxonomic opinions on controversial topics will be included in the biannual outline. For example, 'to promote a more stable taxonomy in Fusarium given the divergences over its generic delimitation', or 'are there too many genera in the Boletales?' and even more importantly, 'what should be done with the tremendously diverse 'dark fungal taxa?' There are undeniable differences in mycologists' perceptions and opinions regarding species classification as well as the establishment of new species. Given the pluralistic nature of fungal taxonomy and its implications for species concepts and the nature of species, this consortium aims to provide a platform to better refine and stabilise fungal classification, taking into consideration views from different parties. In the future, a confidential voting system will be set up to gauge the opinions of all mycologists in the Consortium on important topics. The results of such surveys will be presented to the International Commission on the Taxonomy of Fungi (ICTF) and the Nomenclature Committee for Fungi (NCF) with opinions and percentages of votes for and against. Criticisms based on scientific evidence with regards to nomenclature, classifications, and taxonomic concepts will be welcomed, and any recommendations on specific taxonomic issues will also be encouraged; however, we will encourage professionally and ethically responsible criticisms of others' work. This biannual ongoing project will provide an outlet for advances in various topics of fungal classification, nomenclature, and taxonomic concepts and lead to a community-agreed classification scheme for the fungi and fungus-like taxa. Interested parties should contact the lead author if they would like to be involved in future outlines.

  • Long noncoding RNA MEG3 regulates LATS2 by promoting the ubiquitination of EZH2 and inhibits proliferation and invasion in gallbladder cancer

    • Gallbladder cancer (GBC) is the most common type of biliary tract cancer worldwide. Long noncoding RNAs (lncRNAs) play essential roles in physiological and pathological development. LncRNA MEG3, a tumor suppressor, has been reported to play important roles in some cancers, but the role of MEG3 in GBC remains largely unknown. The purpose of the present study was to explore the role of MEG3 in proliferation and invasion and the potential molecular mechanism in GBC. We found that MEG3 was downregulated in GBC tissues and cells, and low expression of MEG3 was correlated with poor prognostic outcomes in patients. Overexpression of MEG3 inhibited GBC cell proliferation and invasion, induced cell apoptosis and decreased tumorigenicity in nude mice. Moreover, we found that MEG3 was associated with EZH2 and attenuated EZH2 by promoting its ubiquitination. Furthermore, MEG3 executed its functions via EZH2 to regulate the downstream target gene LATS2. Taken together, these findings suggest that MEG3 is an effective target for GBC therapy and may facilitate the development of lncRNA-directed diagnostics and therapeutics against GBC.

    • One question in the use of plants as biomonitors for atmospheric mercury (Hg) is to confirm the linear relationships of Hg concentrations between air and leaves. To explore the origin of Hg in the vegetable and grass leaves, open top chambers (OTCs) experiment was conducted to study the relationships of Hg concentrations between air and leaves of lettuce (Lactuca sativa L.), radish (Raphanus sativus L.), alfalfa (Medicago sativa L.) and ryegrass (Lolium perenne L.). The influence of Hg in soil on Hg accumulation in leaves was studied simultaneously by soil Hg-enriched experiment. Hg concentrations in grass and vegetable leaves and roots were measured in both experiments. Results from OTCs experiment showed that Hg concentrations in leaves of the four species were significantly positively correlated with those in air during the growth time (p < 0.05), while results from soil Hg-enriched experiment indicated that soil-borne Hg had significant influence on Hg accumulation in the roots of each plant (p < 0.05), and some influence on vegetable leaves (p < 0.05), but no significant influence on Hg accumulation in grass leaves (p > 0.05). Thus, Hg in grass leaves is mainly originated from the atmosphere, and grass leaves are more suitable as potential biomonitors for atmospheric Hg pollution. The effect detection limits (EDLs) for the leaves of alfalfa and ryegrass were 15.1 and 22.2 ng g(-1), respectively, and the biological detection limit (BDL) for alfalfa and ryegrass was 3.4 ng m(-3).

  • A novel multi-label classification algorithm based on K-nearest neighbor and random walk

    • The multi-label classification problem occurs in many real-world tasks where an object is naturally associated with multiple labels, that is, concepts. The integration of the random walk approach in the multi-label classification methods attracts many researchers’ sight. One challenge of using the random walk-based multi-label classification algorithms is to construct a random walk graph for the multi-label classification algorithms, which may lead to poor classification quality and high algorithm complexity. In this article, we propose a novel multi-label classification algorithm based on the random walk graph and the K-nearest neighbor algorithm (named MLRWKNN). This method constructs the vertices set of a random walk graph for the K-nearest neighbor training samples of certain test data and the edge set of correlations among labels of the training samples, thus considerably reducing the overhead of time and space. The proposed method improves the similarity measurement by differentiating and integrating the discrete and continuous features, which reflect the relationships between instances more accurately. A label predicted method is devised to reduce the subjectivity of the traditional threshold method. The experimental results with four metrics demonstrate that the proposed method outperforms the seven state-of-the-art multi-label classification algorithms in contrast and makes a significant improvement for multi-label classification.

  • Dynamics and waves near multiple magnetic null points in reconnection diffusion region

    • Identifying the magnetic structure in the region where the magnetic field lines break and how reconnection happens is crucial to improving our understanding of three-dimensional reconnection. Here we show the in situ observation of magnetic null structures in the diffusion region, the dynamics, and the associated waves. Possible spiral null pair has been identified near the diffusion region. There is a close relation among the null points, the bipolar signature of the Z component of the magnetic field, and enhancement of the flux of energetic electrons up to 100 keV. Near the null structures, whistler-mode waves were identified by both the polarity and the power law of the spectrum of electric and magnetic fields. It is found that the angle between the fans of the nulls is quite close to the theoretically estimated maximum value of the group-velocity cone angle for the whistler wave regime of reconnection.

    • This article focused on the research of anaerobic digestion conditions using low-temperature pyrolysis oil, including the fresh inoculum acclimatization, the operation conditions of pyrolysis oil (PO) digestion and the influence of biomass pyrolysis parameters. The coupling process was studied by controlling different pyrolysis parameters and anaerobic digestion parameters. It can be concluded that the fresh sludge inoculum acclimatization can significantly improve the tolerance to the inhibitors contained in the PO, thus, the methane production from the PO digestion to a great degree. The mesophilic condition was favorable to the biooil biomethanation for the low POs under 4% as used in the experiment, whereas the thermophilic condition was favorable for the high PO concentration of 10%. Besides, more methane production can be given by 0.85mm biomass particle size, 300℃ pyrolysis temperature in the downstream step of PO digestion.

  • Agyrotropic Electron Distributions in the Terrestrial Foreshock Transients

    • Agyrotropic electron distributions are frequently taken as an indicator of electron diffusion regions of magnetic reconnection. However, they have also been found at electron-scale boundaries of the non-reconnecting magnetopause and are generated by the electron finite gyroradius effect. Here, we present magnetospheric multiscale observations of agyrotropic electron distributions in the foreshock region. These distributions are generated by the electron finite gyroradius effect after magnetic curvature scattering at a thin electron-scale boundary. Meanwhile, the signatures of magnetic reconnection are absent at this boundary. The test-particle simulation is adopted to verify the generation of the agyrotropic electron distributions by assuming one-dimensional magnetic geometry. These observations suggest that agyrotropic electron distributions can be more widely formed at electron-scale boundaries in space plasma environment.Plain Language SummaryThe agyrotropic electron distributions, which could be unstable to generate high frequency electrostatic waves, reveal valuable information of electron dynamics at electron scales. However, due to electron's small mass, the related observational study becomes only possible with the high-resolution magnetospheric multiscale data. In this study, we show that the agyrotropic electron distributions can be also formed in the foreshock transients such as inside an hot flow anomaly, suggesting that agyrotropic electron distributions are ubiquitous in space plasma.

    • Taking into account various available experimental data, an improved internally consistent data set for the SiO2 system is obtained by thermodynamic assessment. The volumetric properties for SiO2 polymorphs are reassessed using the CALPHAD method. A two-coefficient expression is used to describe the thermal expansion and bulk modulus. For liquid phase, one equation is achieved in the temperature range of 298-4000 K either for thermal expansion or for bulk modulus. Phase diagrams have been calculated up to 40 GPa. It has been concluded that, in the pressure range lower than 20 GPa, there is no significant difference between the phase diagrams calculated by the Birch-Murnaghan equation of state (EOS) and by Murnaghan EOS.

    • Idiopathic pulmonary fibrosis is a progressive lung disorder of unknown etiology. Previous studies have shown that aberrant activation of the Wnt/beta-catenin signaling cascade occurs in lungs of patients with idiopathic pulmonary fibrosis. Given the important roles of the Wnt/beta-catenin signaling pathway in the development of pulmonary fibrosis, we targeted this pathway for the intervention of pulmonary fibrosis with XAV939, a small molecule that specifically inhibits Tankyrase 1/2, eventually leading to the degradation of beta-catenin and suppression of the Wnt/beta-catenin signaling pathway. Our results demonstrated that XAV939 significantly inhibited the activation of Wnt/beta-catenin signaling and attenuated bleomycin-induced lung fibrosis in mice, and thus improved the survival of mice with lung injury. Interestingly, previous investigations have confirmed that endogenous and exogenous mesenchymal stem cells could be recruited to the injured lung, although the exact effects of these cells are debatable. To determine the effect of Wnt/beta-catenin signaling in the epithelial differentiation of bone marrow-derived mesenchymal stem cells (BM-MSCs), we established a coculture system that contains BM-MSCs and alveolar type II epithelial cells. The in vitro experiments demonstrated that XAV939 could promote the differentiation of BM-MSCs into an epithelium-like phenotype in the coculture system. We also found that XAV939 could inhibit the proliferation and myofibroblast differentiation of NIH/3T3 fibroblasts. This work supports that inhibition of the Wnt/beta-catenin signaling pathway may be exploited for the treatment of idiopathic pulmonary fibrosis for which effective treatment strategies are still lacking.
