  • Robust estimation of bacterial cell count from optical density

    • Optical density (OD) is widely used to estimate the density of cells in liquid culture, but cannot be compared between instruments without a standardized calibration protocol and is challenging to relate to actual cell count. We address this with an interlaboratory study comparing three simple, low-cost, and highly accessible OD calibration protocols across 244 laboratories, applied to eight strains of constitutive GFP-expressing E. coli. Based on our results, we recommend calibrating OD to estimated cell count using serial dilution of silica microspheres, which produces highly precise calibration (95.5% of residuals <1.2-fold), is easily assessed for quality control, also assesses instrument effective linear range, and can be combined with fluorescence calibration to obtain units of Molecules of Equivalent Fluorescein (MEFL) per cell, allowing direct comparison and data fusion with flow cytometry measurements: in our study, fluorescence per cell measurements showed only a 1.07-fold mean difference between plate reader and flow cytometry data.

    • This paper attempts to address the failings of a predominant paradigm in IS research and practice that emphasises technological determinism. This paradigm makes use of a false belief in the power of rationality in organizational decision-making, and a mythology in which organizational actors can be viewed as passive ‘users’ of technology. We wish to create a discussion of the nature and role of professionalism as an expression of more than technical competence. Both system analysts and organizational stakeholders (e.g. ‘users’) are to be viewed as professionals. We discuss desire, exercise of will and their role in professional judgment in relation to transcendent values espoused within communities of practice. We go on to relate this to the environments of Information Systems research and practice. It is pointed out that many researchers, over a number of years, have dealt with these issues in relation to effective management of technological development and organizational change. The paper attempts to encourage renewed attention to interpretivist perspectives on IS development and organizational change, including recognition of the importance of contextual dependencies.

  • A Comparative Nitrogen Balance and Productivity Analysis of Legume and Non-legume Supported Cropping Systems: The Potential Role of Biological Nitrogen Fixation

    • The potential of biological nitrogen fixation (BNF) to provide sufficient N for production has encouraged re-appraisal of cropping systems that deploy legumes. It has been argued that legume-derived N can maintain productivity as an alternative to the application of mineral fertilizer, although few studies have systematically evaluated the effect of optimizing the balance between legumes and non N-fixing crops to optimize production. In addition, the shortage, or even absence in some regions, of measurements of BNF in crops and forages severely limits the ability to design and evaluate new legume-based agroecosystems. To provide an indication of the magnitude of BNF in European agriculture, a soil-surface N-balance approach was applied to historical data from 8 experimental cropping systems that compared legume and non-legume crop types (e.g., grains, forages and intercrops) across pedoclimatic regions of Europe. Mean BNF for different legume types ranged from 32 to 115 kg ha(-1) annually. Output in terms of total biomass (grain, forage, etc.) was 30% greater in non-legumes, which used N to produce dry matter more efficiently than legumes, whereas output of N was greater from legumes. When examined over the crop sequence, the contribution of BNF to the N-balance increased to reach a maximum when the legume fraction was around 0.5 (legume crops were present in half the years). BNF was lower when the legume fraction increased to 0.6-0.8, not because of any feature of the legume, but because the cropping systems in this range were dominated by mixtures of legume and non-legume forages to which inorganic N as fertilizer was normally applied. Forage (e.g., grass and clover), as opposed to grain crops in this range maintained high outputs of biomass and N. In conclusion, BNF through grain and forage legumes has the potential to generate major benefit in terms of reducing or dispensing with the need for mineral N without loss of total output.

  • The 4C Approach as a way to understand species interactions determining intercropping productivity

    • Modern agriculture needs to develop transition pathways toward agroecological, resilient and sustainable farming systems. One key pathway for such agroecological intensification is the diversification of cropping systems using intercropping and notably cereal-grain legume mixtures. Such mixtures or intercrops have the potential to increase and stabilize yields and improve cereal grain protein concentration in comparison to sole crops. Species mixtures are complex and the 4C approach is both a pedagogical and scientific way to represent the combination of four joint effects of Competition, Complementarity, Cooperation, and Compensation as processes or effects occurring simultaneously and dynamically between species over the whole cropping cycle. Competition is when plants have fairly similar requirements for abiotic resources in space and time, the result of all processes that occur when one species has a greater ability to use limiting resources (e.g., nutrients, water, space, light) than others. Complementarity is when plants grown together have different requirements for abiotic resources in space, time or form. Cooperation is when the modification of the environment by one species is beneficial to the other(s). Compensation is when the failure of one species is compensated by the other(s) because they differ in their sensitivity to abiotic stress. The 4C approach allows to assess the performance of arable intercropping versus classical sole cropping through understanding the use of abiotic resources.

  • Factors influencing crop rotation strategies on organic farms with different time periods since conversion to organic production

    • Productive crop rotations are central to the success of organic production systems. The selection and sequence of crops are determined by a combination of agronomic and economic factors as well as the principles and standards of organic farming. Semi-structured interviews were conducted with sixteen organic farmers in Central-east Sweden to explore the factors that influence the design of crop rotations and the trade-offs between these factors, taking into account the length of time since conversion to organic production. We discerned three crop rotation strategies: strict, flexible and liberal, based on how crop(s) are repeated over time. A major trade-off for arable farmers was between perennial leys to provide nitrogen and control weeds, and the use of more inputs such as purchased nutrients and mechanical weed control to allow continuous cereal production. Critical considerations for livestock farmers were the length of ley for feed production and weed control, cost of re-seeding leys and decisions about whether to grow crops to feed animals or cereals to sell. Farmers practicing organic for a long time (more than 10years) often had flexible rotations to adapt to changing conditions, but they generally included leys and a selection of annual crops in line with the principles of crop rotation and organic farming. Recently converted organic farmers usually concentrated on controlling weeds and producing sufficient livestock feed by following strict crop rotations. We conclude that farm type and experience strongly influenced rotation strategies and that weed management and market prices were the most important influences.

  • The reflection of principles and values in worldwide organic agricultural research viewed through a crop diversification lens. A bibliometric review

    • Organic agriculture and organic food have expanded in recent decades but have undergone conventionalisation. Some claim that this contradicts some or all of the principles of ‘health’, ‘ecology’, ‘fairness’ and ‘care’ established by the International Federation of Organic Agricultural Movement (IFOAM). It is currently unclear how research on organic food/agriculture is structured, whether it embraces these principles, or how key crop diversification, driving sustainability, are addressed. To fill these knowledge gaps, we conducted a bibliometric analysis of 10,030 peer-reviewed articles published from 1945 to 2021 with topic and textual analysis. Our main findings were the following: (1) research is compartmentalised into scales and disciplines, with at field-scale ‘weed’, ‘soil’, ‘pest and disease’ management and ‘livestock farming’ seldom addressed together, or with environmental assessment separated from socioeconomic studies at farm scale. (2) The proportion of publications on ‘consumers’ preferences’ and ‘product quality’ research almost tripled in 20 years, from 10 to 27%, emphasizing the consumer orientation of research on organic agriculture and organic food. (3) Only 4% of articles covered all four IFOAM principles, while associated values such as ‘resilience’, ‘integrity’, ‘equity’, ‘transparency’ and ‘justice’ were even less frequently addressed. (4) Fewer diversification practices have been tested in organic than in conventional agriculture research, with fewer articles on ‘crop mixtures’ or ‘bee-friendly crops’ and a smaller range of legumes considered. (5) Research on genetic improvement and processing of organic legumes is lacking, which could constrain adoption of legumes in organic farming even more than in conventional agriculture. These results indicate a need for increasing interdisciplinary efforts at field level, with systematic measurement of multiple processes (weed-nutrient-pest dynamics). Future studies on organic agriculture should combine several diversification practices and legumes, with relevant indicators addressing the IFOAM values explicitly, and consider the whole value chain by linking producers with consumers.

  • Crop rotational diversity can mitigate climate-induced grain yield losses

    • Diversified crop rotations have been suggested to reduce grain yield losses from the adverse climatic conditions increasingly common under climate change. Nevertheless, the potential for climate change adaptation of different crop rotational diversity (CRD) remains undetermined. We quantified how climatic conditions affect small grain and maize yields under different CRDs in 32 long-term (10-63 years) field experiments across Europe and North America. Species-diverse and functionally rich rotations more than compensated yield losses from anomalous warm conditions, long and warm dry spells, as well as from anomalous wet (for small grains) or dry (for maize) conditions. Adding a single functional group or crop species to monocultures counteracted yield losses from substantial changes in climatic conditions. The benefits of a further increase in CRD are comparable with those of improved climatic conditions. For instance, the maize yield benefits of adding three crop species to monocultures under detrimental climatic conditions exceeded the average yield of monocultures by up to 553 kg/ha under non-detrimental climatic conditions. Increased crop functional richness improved yields under high temperature, irrespective of precipitation. Conversely, yield benefits peaked at between two and four crop species in the rotation, depending on climatic conditions and crop, and declined at higher species diversity. Thus, crop species diversity could be adjusted to maximize yield benefits. Diversifying rotations with functionally distinct crops is an adaptation of cropping systems to global warming and changes in precipitation.Industrial agriculture often relies on one or few crop species grown in monocultures or short crop rotations, making them vulnerable to changes in climatic conditions. Using data from several agricultural experiments in Europe and North America, we show that including more crop species or crop types in rotation can mitigate cereal yield losses caused by increasingly common shifts in climatic conditions, such as increasing temperatures and decreasing precipitation. Hence, increasing crop rotational diversity can support the climate adaptation of the way we produce our food.image

    • Thrombo-inflammation describes the complex interplay between blood coagulation and inflammation that plays a critical role in cardiovascular diseases. The third Maastricht Consensus Conference on Thrombosis assembled basic, translational, and clinical scientists to discuss the origin and potential consequences of thrombo-inflammation in the etiology, diagnostics, and management of patients with cardiovascular disease, including myocardial infarction, stroke, and peripheral artery disease. This article presents a state-of-the-art reflection of expert opinions and consensus recommendations regarding the following topics: (1) challenges of the endothelial cell barrier; (2) circulating cells and thrombo-inflammation, focused on platelets, neutrophils, and neutrophil extracellular traps; (3) procoagulant mechanisms; (4) arterial vascular changes in atherogenesis; attenuating atherosclerosis and ischemia/reperfusion injury; (5) management of patients with arterial vascular disease; and (6) pathogenesis of venous thrombosis and late consequences of venous thromboembolism.

  • Resistance to thyroid hormone induced tachycardia in RTHα syndrome

    • Mutations in thyroid hormone receptor α1 (TRα1) cause Resistance to Thyroid Hormone α (RTHα), a disorder characterized by hypothyroidism in TRα1-expressing tissues including the heart. Surprisingly, we report that treatment of RTHα patients with thyroxine to overcome tissue hormone resistance does not elevate their heart rate. Cardiac telemetry in male, TRα1 mutant, mice indicates that such persistent bradycardia is caused by an intrinsic cardiac defect and not due to altered autonomic control. Transcriptomic analyses show preserved, thyroid hormone (T3)-dependent upregulation of pacemaker channels (Hcn2, Hcn4), but irreversibly reduced expression of several ion channel genes controlling heart rate. Exposure of TRα1 mutant male mice to higher maternal T3 concentrations in utero, restores altered expression and DNA methylation of ion channels, including Ryr2. Our findings indicate that target genes other than Hcn2 and Hcn4 mediate T3-induced tachycardia and suggest that treatment of RTHα patients with thyroxine in high dosage without concomitant tachycardia, is possible.
