    • Respiratory droplets are bioaerosols that originate from the respiratory tract. Knowing their deposition characteristics during exhalation would facilitate the understanding of the source of large respiratory droplets and their importance in the spread of respiratory infectious diseases. In this study, computational fluid dynamics is used to simulate the motion and deposition of droplets released from either trachea or bronchi in a realistic reconstruction of the human respiratory tract. Influences of airflow structures and locations of droplet generation on droplet deposition are studied, and droplets with diameters between 1 and 50 mu m are examined. The deposition of droplets is found to be influenced mainly by the droplet diameter and the flow rate of exhalation. The number of droplets released from the trachea or bronchi that can escape into the environment decreases as the flow rate increases. When the flow rate is low (10 L/min), the critical diameter of droplets generated in the lower respiratory system that can escape into the air is approximately 12 mu m, but this diameter is approximately 5 mu m when the flow rate is medium (30 to 60 L/min) or large (90 L/min). The larynx is the dominant site of deposition for droplets smaller than the critical diameter, while trachea and bronchus are more important locations that account for the deposition of larger droplets. This study indicates that the lower respiratory tract is an important source of fine droplets (<5 mu m) in indoor environments, and larger droplets probably originate from the upper respiratory tract, which needs further investigation.     • Lymphocyte-based immunotherapy has emerged as a break-through in cancer therapy for both hematologic and solid malignancies. In a subpopulation of cancer patients, this powerful therapeutic modality converts malignancy to clinically manageable disease. However, the T cell- and chimeric antigen receptor T (CAR-T) cell-mediated antimetastatic activity, especially their impacts on microscopic metastatic lesions, has not yet been investigated. Here we report a living zebrafish model that allows us to visualize the metastatic cancer cell killing effect by tumor- infiltrating lymphocytes (TILs) and CAR-T cells in vivo at the single-cell level. In a freshly isolated primary human melanoma, specific TILs effectively eliminated metastatic cancer cells in the living body. This potent metastasis-eradicating effect was validated using a human lymphoma model with CAR-T cells. Furthermore, cancer-associated fibroblasts protected metastatic cancer cells from T cell-mediated killing. Our data provide an in vivo platform to validate antimetastatic effects by human T cell-mediated immunotherapy. This unique technology may serve as a precision medicine platform for assessing anticancer effects of cellular immunotherapy in vivo before administration to human cancer patients.

  • Ecosystem CO2 Exchange and Its Economic Implications in Northern Permafrost Regions in the 21st Century

    • Climate warming increases carbon assimilation by plant growth and also accelerates permafrost CO2 emissions; however, the overall ecosystem CO2 balance in permafrost regions and its economic impacts remain largely unknown. Here we synthesize in situ measurements of net ecosystem CO2 exchange to assess current and future carbon budgets across the northern permafrost regions using the random forest model and calculate their economic implications under the Shared Socio-economic Pathways (SSPs) based on the PAGE-ICE model. We estimate a contemporary CO2 emission of 1,539 Tg C during the nongrowing season and CO2 uptake of 2,330 Tg C during the growing season, respectively. Air temperature and precipitation exert the most control over the net ecosystem exchange in the nongrowing season, while leaf area index plays a more important role in the growing season. This region will probably shift to a carbon source after 2,057 under SSP5-8.5, with a net emission of 17 Pg C during 2057–2100. The net economic benefits of CO2 budget will be $4.5, $5.0, and $2.9 trillion under SSP1-2.6, SSP2-4.5, and SSP5-8.5, respectively. Our results imply that a high-emission pathway will greatly reduce the economic benefit of carbon assimilation in northern permafrost regions.

    • Tetrabromobisphenol A (TBBPA) is one of the most widely used brominated flame retardants (BFRs) and has been frequently detected in the environment and biota. Recent studies have found that derivatives of TBBPA, such as TBBPA bis(allyl) ether (TBBPA BAE) and TBBPA bis(2,3-dibromopropyl) ether (TBBPA BDBPE) are present in various environmental compartments. In this work, using liquid chromatography coupled with tandem mass spectrometry (LC-MS/MS) and liquid chromatography coupled with quadrupole time-of-flight mass spectrometry (LC-Q-TOF-MS), TBBPA allyl ether (TBBPA AE) and TBBPA 2,3-dibromopropyl ether (TBBPA DBPE) were identified in environmental samples and further confirmed by synthesized standards. Soil, sediment, rice hull, and earthworm samples collected near a BFR manufacturing plant were found to contain these two compounds. In sediments, the concentrations of TBBPA AE and TBBPA DBPE ranged from 1.0 to 346.6 ng/g of dry weight (dw) and from 0.7 to 292.7 ng/g of dw, respectively. TBBPA AE and TBBPA DBPE in earthworm and rice hull samples were similar to soil samples, which ranged from below the method limit of detection (LOD, <0.002 ng/g of dw) to 0.064 ng/g of dw and from below the LOD (<0.008 ng/g of dw) to 0.58 ng/g of dw, respectively. Furthermore, mollusks collected from the Chinese Bohai Sea were used as a bioindicator to investigate the occurrence and distribution of these compounds in the coastal environment. The detection frequencies of TBBPA AE and TBBPA DBPE were 41 and 32%, respectively, and the concentrations ranged from below LOD (<0.003 ng/g of dw) to 0.54 ng/g of dw, with an average of 0.09 ng/g of dw, for TBBPA AE, and from below LOD (<0.008 ng/g of dw) to 1.41 ng/g of dw, with an average of 0.15 ng/g of dw, for TBBPA DBPE.
