    • BackgroundMyelodysplastic Syndromes (MDSS) are pre-leukemic disorders with increasing incident rates worldwide, but very limited treatment options. Little is known about small regulatory RNAs and how they contribute to pathogenesis, progression and transcriptome changes in MDS.MethodsPatients' primary marrow cells were screened for short RNAs (RNA-seq) using next generation sequencing. Exon arrays from the same cells were used to profile gene expression and additional measures on 98 patients obtained. Integrative bioinformatics algorithms were proposed, and pathway and ontology analysis performed.ResultsIn low-grade MDS, observations implied extensive post-transcriptional regulation via microRNAs (miRNA) and the recently discovered Piwi interacting RNAs (piRNA). Large expression differences were found for MDS-associated and novel miRNAs, including 48 sequences matching to miRNA star (miRNA*) motifs. The detected species were predicted to regulate disease stage specific molecular functions and pathways, including apoptosis and response to DNA damage. In high-grade MDS, results suggested extensive post-translation editing via transfer RNAs (tRNAs), providing a potential link for reduced apoptosis, a hallmark for this disease stage. Bioinformatics analysis confirmed important regulatory roles for MDS linked miRNAs and TFs, and strengthened the biological significance of miRNA*. The "RNA polymerase II promoters" were identified as the tightest controlled biological function. We suggest their control by a miRNA dominated feedback loop, which might be linked to the dramatically different miRNA amounts seen between low and high-grade MDS.DiscussionThe presented results provide novel findings that build a basis of further investigations of diagnostic biomarkers, targeted therapies and studies on MDS pathogenesis.

  • Shocked chromites in fossil L chondrites : A Raman spectroscopy and transmission electron microscopy study

    • Chromites from Middle Ordovician fossil L chondrites and from matrix and shock-melt veins in Catherwood, Tenham, and Coorara L chondrites were studied using Raman spectroscopy and TEM. Raman spectra of chromites from fossil L chondrites showed similarities with chromites from matrix and shock-melt veins in the studied L chondrite falls and finds. Chromites from shock-melt veins of L chondrites show polycrystallinity, while the chromite grains in fossil L chondrites are single crystals. In addition, chromites from shock-melt veins in the studied L chondrites have high densities of planar fractures within the subgrains and many subgrains show intergrowths of chromite and xieite. Matrix chromite of Tenham has similar dislocation densities and planar fractures as a chromite from the fossil meteorite Golvsten 001 and higher dislocation densities than in chromite from the fossil meteorite Sextummen 003. Using this observation and knowing that the matrix of Tenham experienced 20-22 GPa and <1000° C, an upper limit for the P,T conditions of chromite from Golvsten 001 and Sextummen 003 can be estimated to be 20-22 GPa and 1000° C (shock stage S3-S6) and 20 GPa and 1000° C (S3-S5), respectively, and we conclude that the studied fossil meteorite chromites are from matrix.

  • Catalytically Active Oil-based Lubricant Additives Enabled by Calcining Ni-Al Layered Double Hydroxides

    • Layered double hydroxides (LDHs) have lately been hailed as robust lubricant additives for improving tribological properties and as ideal catalysts for synthesizing carbon-based nanomaterials. In this paper, in situ analytical tools are used to track the evolution of the crystal structure and chemical composition of LDHs during calcination. Nickel oxide and elemental nickel can be produced by calcining NiAl-LDH in air (LDH-C-Air) and argon (LDH-C-Ar), respectively. For the base oil with 1 wt % LDH-C-Air, negligible wear can be detected even after a 2 h friction test under a severe contact pressure (∼637 MPa). A relatively thick tribofilm (∼60 nm) with a better mechanical property is formed, which protects the solid surface from severe wear. In addition, the possible formed carbon debris may also prevent the direct collision of asperities and effectively improve the wear resistance. This work provides a unique vision for the application of calcined LDHs with the combination of catalysis and tribology.

  • Genome and proteomic analysis of risk factors for fatal outcome in children with severe community-acquired pneumonia caused by human adenovirus 7

    • Introduction: Human adenovirus 7 (HAdV-7) is an important viral pathogen of severe pneumonia in children and a serious threat to health. Methods: A cohort of 45 pediatric patients diagnosed with HAdV-7-associated severe pneumonia and admitted to the Pediatric Intensive Care Unit at the Children's Hospital of Chongqing Medical University from May 2018 to January 2020 were included. Risk factors of death were analyzed by the Cox proportional risk mode with Clinical data, serum, and nasopharyngeal aspirate adenovirus load, Genome analysis, Olink proteomics, and cytokine profile between dead and surviving patients were also analyzed. Results: A total of 45 children with a median age of 12.0 months (interquartile range [IQR]: 6.5, 22.0) were included (female 14), including 14 (31.1%) who died. High serum viral load was an independent risk factor for mortality (hazard ratio [HR] = 2.16, 95% confidence interval [CI], 1.04-4.49, p = 0.039). BTB and CNC homology 1 (BACH1), interleukin-5 (IL-5), and IL-9 levels were significantly correlated with serum viral load (p = 0.0400, 0.0499, and 0.0290; r = 0.4663, 0.3339, and -0.3700, respectively), with significant differences between the dead and survival groups (p = 0.021, 0.001, and 0.021). Conclusions: Severe cytokine storm-associated high serum viral load after HAdV-7 infection may be the main mechanism responsible for poor prognosis in children.
