  • Mutual Favours : The social and scientific practice of eighteenth-century Swedish chemistry

    • The dissertation is a study of the creation of chemistry as a science in eighteenth-century Sweden. It is argued that the chemists in the study participated in a network for exchange of scientific facts and all kinds of favours, in which science was both conducted, negotiated and created. A number of relationships between chemists are analyzed with regards to two central eighteenth-century institutions: the patron-client relationship and the egalitarian ideal of reciprocity articulated in the eighteenth-century Republic of Letters.In the first half the background to the success of Swedish chemistry is sketched out. It is discussed which groups supported chemistry and for what reasons. There is a discussion of the theoretical and methodological changes that were initiated by Torbern Bergman when he took over the chair of chemistry in Uppsala. Bergman's attempts to marginalize his two major opponents, Johan Gottschalk Wallerius, the previous holder of the Uppsala chair and Gustav von Engeström, the head of the Board of Mines laboratory in Stockholm, are also analyzed.In the second half the focus shifts to the interaction of university chemistry with industry. It is shown how industrial processes gradually came to be redefined as a kind of “coarse chemistry”, a process which benefited both engineers employed at industrial installations and university chemists. The many themes explored in the study are brought together in an analysis of Carl Wilhelm Scheele’s adoption into the network of Swedish chemists. The dissertation concludes with a survey of the more general conclusions.

  • Biopolitics and Reflexivity : A Study of GMO Policymaking in the European Union

    • The political discourse that has emerged as a consequence of establishing a European regulatory framework for GMOs has not been without problems. This dissertation addresses the political and regulatory challenges created by the development and use of genetically modified organisms (GMOs) in the EU. The underlying hypothesis in the dissertation is that the emergence of a European policy in the field of GMOs has appeared through new reflexive forms of governance. The aim of this research is to understand how these reflexive forms of regulation have emerged and operate within the EU, with a particular focus on the two GMO directives 90/220/EEC and 2001/18/EC. However, the study scrutinises the regulatory regimes from the 1970s onwards by investigating how the regulatory framework regarding GMOs has been developed and implemented in the EU. This is done through an analysis of the notion of ‘risk’ and the ‘precautionary principle’ since these concepts have been at the forefront of the GMO regulation debate. The empirical approach focuses on how the European Commission, the European Parliament and the European Council as well as other actors and institutions articulate ambivalence, interpretations and uncertainties in the decision-making processes regarding regulative measures for GMOs, with an accent on risk and the precautionary principle. The main empirical material has been documents concerning the inter-institutional process involved in the regulatory process of making the two directives. The analysis indicates that during the process of implementing GMO regulations, new steering strategies have appeared within the EU’s decision-making institutions when the objective of the regulation has taken centre stage in political and scientific controversies.

    • beta-1,4-Gatactosyltransferase 7 (beta 4GalT7) is an essential enzyme in the biosynthesis of glycosaminoglycan (GAG) chains of proteoglycans (PGs). Mammalian cells produce PGs, which are involved in biological processes such as cell growth and differentiation. The PGs consist of a core protein, with one or several GAG chains attached. Both the structure of the PGs and the GAG chains, and the expression of the enzymes involved in their biosynthesis and degradation, vary between normal cells and tumor cells. The biosynthesis of GAG chains is initiated by xylosylation of a serine residue of the core protein, followed by galactosylation by beta 4GalT7. The biosynthesis can also be initiated by exogenously added beta-D-xylopyranosides with hydrophobic aglycons, which thus can act as acceptor substrates for beta 4GalT7. To determine the structural requirements for beta 4GalT7 activity, we have cloned and expressed the enzyme and designed a focused library of 2-naphthyl beta-D-xylopyranosides with modifications of the xylose moiety. Based on enzymatic studies, that is galactosylation and its inhibition, conformational analysis and molecular modeling using the crystal structure, we propose that the binding pocket of beta 4GalT7 is very narrow, with a precise set of important hydrogen bonds. Xylose appears to be the optimal acceptor substrate for galactosylation by beta 4GalT7. However, we show that modifications of the xylose moiety of the beta-D-xylopyranosides can render inhibitors of galactosylation. Such compounds will be valuable tools for the exploration of GAG and PG biosynthesis and a starting point for development of anti-tumor agents.
