    • Research on sustainability transitions has expanded rapidly in the last ten years, diversified in terms of topics and geographical applications, and deepened with respect to theories and methods. This article provides an extensive review and an updated research agenda for the field, classified into nine main themes: understanding transitions; power, agency and politics; governing transitions; civil society, culture and social movements; businesses and industries; transitions in practice and everyday life; geography of transitions; ethical aspects; and methodologies. The review shows that the scope of sustainability transitions research has broadened and connections to established disciplines have grown stronger. At the same time, we see that the grand challenges related to sustainability remain unsolved, calling for continued efforts and an acceleration of ongoing transitions. Transition studies can play a key role in this regard by creating new perspectives, approaches and understanding and helping to move society in the direction of sustainability.

  • Mechanistic insights from high resolution DNA damage analysis to understand mixed radiation exposure

    • Cells exposed to densely ionising high and scattered low linear energy transfer (LET) radiation (50 % dose of each) react more strongly than to the same dose of each separately. The relationship between DNA double strand break location inside the nucleus and chromatin structure was evaluated, using high-resolution transmission electron microscopy (TEM) in breast cancer MDA-MB-231 cells at 30 min post 5 Gy. Additionally, response to high and/or low LET radiation was assessed using single (1 ×1.5 Gy) versus fractionated dose delivery (5 ×0.3 Gy). By TEM analysis, the highest total number of γH2AX nanobeads were found in cells irradiated with alpha radiation just prior to gamma radiation (called mixed beam), followed by alpha, then gamma radiation. γH2AX foci induced by mixed beam radiation tended to be surrounded by open chromatin (lighter TEM regions), yet foci containing the highest number of beads, i.e. larger foci representing complex damage, remained in the heterochromatic areas. The γH2AX large focus area was also greater in mixed beam-treated cells when analysed by immunofluorescence. Fractionated mixed beams given daily induced the strongest reduction in cell viability and colony formation in MDA-MB-231 and osteosarcoma U2OS cells compared to the other radiation qualities, as well as versus acute exposure. This may partially be explained by recurring low LET oxidative DNA damage by every fraction together with a delay in recompaction of chromatin after high LET, demonstrated by low levels of heterochromatin marker H3K9me3 at 2 h after the last mixed beam fraction in MDA-MB-231. In conclusion, early differences in response to complex DNA damage may lead to a stronger cell kill induced by fractionated exposure, which suggest a therapeutic potential of combined high and low LET irradiation.

    • The potential of using supported liquid membrane (SLM) technique, combined with reversed-phase high-performance liquid chromatography (RP-HPLC) has been investigated for the determination of peptides in human blood plasma. The peptides studied were (DL)Leu(DL)Phe, MetLeuPhe, GlyLeuTyr and ValGluProlleProTyr. The carrier (Aliquat 336) was incorporated in membrane phase in order to facilitate the transport of investigated peptides. After extraction, the analyte-enriched acceptor phase was directly injected into an HPLC system for analysis. With SLM, high selectivity and efficiency were achieved for extraction of peptides in aqueous solutions. Lower extraction efficiency was obtained in plasma sample. The limiting factors were the presence of salts and proteins in blood plasma. The necessary of ultrafiltration application before the SLM was required in order to improve its stability. The minimum quantifiable concentration was 90-130 ng/ml of the investigated peptides based on 0.5 ml plasma. The standard curve was linear over the concentration range of 0.05-10 mu g/ml.     • PPFIBP1 encodes for the liprin-β1 protein, which has been shown to play a role in neuronal outgrowth and synapse formation in Drosophila melanogaster. By exome and genome sequencing, we detected nine ultra-rare homozygous loss-of-function variants in 16 individuals from 12 unrelated families. The individuals presented with moderate to profound developmental delay, often refractory early-onset epilepsy, and progressive microcephaly. Further common clinical findings included muscular hyper- and hypotonia, spasticity, failure to thrive and short stature, feeding difficulties, impaired vision, and congenital heart defects. Neuroimaging revealed abnormalities of brain morphology with leukoencephalopathy, ventriculomegaly, cortical abnormalities, and intracranial periventricular calcifications as major features. In a fetus with intracranial calcifications, we identified a rare homozygous missense variant that by structural analysis was predicted to disturb the topology of the SAM domain region that is essential for protein-protein interaction. For further insight into the effects of PPFIBP1 loss of function, we performed automated behavioral phenotyping of a Caenorhabditis elegans PPFIBP1/hlb-1 knockout model, which revealed defects in spontaneous and light-induced behavior and confirmed resistance to the acetylcholinesterase inhibitor aldicarb, suggesting a defect in the neuronal presynaptic zone. In conclusion, we establish bi-allelic loss-of-function variants in PPFIBP1 as a cause of an autosomal recessive severe neurodevelopmental disorder with early-onset epilepsy, microcephaly, and periventricular calcifications.

  • Systematic Studies on Liquid Sodium 4,5-dicyano-2-(trifluoromethyl)imidazolate (NaTDI)-Based Electrolytes and Its Impact on the Cycling Behaviour Against Wet Impregnated WI-NaNMC and Prussian White Cathodes

    • The article describes a research on new salts and cathode materials dedicated to sodium cells. Sodium technology is considered to be one of the most promising (for new battery generations) and has been very actively (re)-developed for more than a decade. The first part of this work is mainly focused on electrochemical studies of the low coordinating Huckel type salt (4,5-dicyano-2-(trifluoromethyl)imidazolate, NaTDI)-based electrolytes with and without solid electrolyte interface modified additives: fluoroethylene carbonate (FEC). In the second part electrolytes are tested in a half-cells with layered oxides. Sodium nickel-manganese-cobalt oxide (NaNMC) and WI-NaNMC-333 and WI-NaNMC-622 layered cathode materials are prepared by wet impregnation synthesis (WI) routine. Prussian White is obtained from University of Uppsala group to compare and initial electrochemical properties against electrolyte with NaTDI. Basic physicochemical properties of the obtained powders are also reported in the manuscript. The best properties are recorded for 0.75 m NaTDI in EC:DMC (1:1 v/v) with 3% FEC additive by its conductivity (11.52 mS cm(-1) for 20 degrees C). The same solution also shows promising results during electrochemical tests in half-cell geometry against WI-NaNMC-622 cathode (114.22 mAh g(-1) after 1 cycle C/20). NaPF6 salt is used as an electrolyte as a commercially available reference.

  • Citizen-led decentralised energy futures: Emerging rationales of energy system organisation

    • The current energy systems are undergoing fundamental changes driven by the climate crisis, unfolding digitalisation and increasing calls for a more active citizens' engagement. The impact of these ongoing trends on the future energy system, however, is far from straightforward. Although there is an increasing shift towards a decentralisation, it is not clear yet how exactly this new decentralised configuration will unfold and materialise. In this article we explore the rationales behind current trends towards a more decentralised electricity system. Given the developments in the electricity system, our study centres on emerging initiatives led by citizens and their values. Theoretically, we first mobilise the notion of socio-technical system as constituted and reproduced by actors, institutions, and technology, operating based on certain shared principles. We use this lens to describe the past and current energy system organisation. Secondly, based on two dimensions of value orientation and steering direction we define four emerging ideal-types socio-technical decentralisation configurations. We examine possible pathways of change, and the institutional, actor and technological preconditions they require, to become predominant forms of decentralisation. We conclude that although all forms of decentralisation may potentially contribute to low carbon objectives, they are likely to co-evolve catering to diverse system needs and the citizen requirements simultaneously.
