  • Evidence for FHL1 as a novel disease gene for isolated hypertrophic cardiomyopathy

    • Hypertrophic cardiomyopathy (HCM) is characterized by asymmetric left ventricular hypertrophy, diastolic dysfunction and myocardial disarray. HCM is caused by mutations in sarcomeric genes, but in 40 of patients, the mutation is not yet identified. We hypothesized that FHL1, encoding four-and-a-half-LIM domains 1, could be another disease gene since it has been shown to cause distinct myopathies, sometimes associated with cardiomyopathy. We evaluated 121 HCM patients, devoid of a mutation in known disease genes. We identified three novel variants in FHL1 (c.134delA/K45Sfs, c.459CA/C153X and c.827GC/C276S). Whereas the c.459CA variant was associated with muscle weakness in some patients, the c.134delA and c.827GC variants were associated with isolated HCM. Gene transfer of the latter variants in C2C12 myoblasts and cardiac myocytes revealed reduced levels of FHL1 mutant proteins, which could be rescued by proteasome inhibition. Contractility measurements after adeno-associated virus transduction in rat-engineered heart tissue (EHT) showed: (i) higher and lower forces of contraction with K45Sfs and C276S, respectively, and (ii) prolonged contraction and relaxation with both mutants. All mutants except one activated the fetal hypertrophic gene program in EHT. In conclusion, this study provides evidence for FHL1 to be a novel gene for isolated HCM. These data, together with previous findings of proteasome impairment in HCM, suggest that FHL1 mutant proteins may act as poison peptides, leading to hypertrophy, diastolic dysfunction and/or altered contractility, all features of HCM.

    • Abundance and biomass of cod, haddock, and whiting in the waters off of the west coast of Scotland (wcoS) have undergone large changes in recent years, most notably a recent decline. These three species contribute a considerable part of Scottish demersal landings from this area and as such it is important to understand why these stocks are behaving the way they are. A number of explanations for the decline have been proposed, including: seal predation, pressure from Nephrops trawls, and fishing pressure more generally. We used an ecosystem model of the wcoS continental shelf (<200 m depth) to investigate whether these proposed explanations for declining gadoid stocks are feasible. Results suggest that the rise in the grey seal population over recent years has not led to the decline in gadoid stocks; there is insufficient bycatch by the Nephrops fleet to have a large impact on gadoid stocks; however, fishing, as a key driver of the west of Scotland shelf ecosystem, has impacted stocks and by decreasing fishing levels to maximum sustainable yield cod biomass may increase slightly though not returning to previous levels. Although this means we are little further forward in understanding the cause of recent gadoid declines in the area, the development of this model has enabled us to further our knowledge and understanding of aspects of trophic structure and the impacts of fishing on the wcoS.

  • An Open Dataset Storage Standard for 6G Testbeds

    • The emergence of sixth-generation (6G) networks has spurred the development of novel testbeds, including sub-THz networks, cell-free systems, and 6G simulators. To maximize the benefits of these systems, it is crucial to make the generated data publicly available and easily reusable by others. Although data sharing has become a common practice, a lack of standardization hinders data accessibility and interoperability. In this study, we propose the Dataset Storage Standard (DSS) to address these challenges by facilitating data exchange and enabling convenient processing script creation in a testbed-agnostic manner. DSS supports both experimental and simulated data, allowing researchers to employ the same processing scripts and tools across different datasets. Unlike existing standardization efforts such as SigMF and NI RF Data Recording API, DSS provides a broader scope by accommodating a common definition file for testbeds and is not limited to RF data storage. The dataset format utilizes a hierarchical structure, with a tensor representation for specific experiment scenarios. In summary, DSS offers a comprehensive and flexible framework for enhancing the FAIR principles (Findability, Accessibility, Interoperability, and Reusability) in 6G testbeds, promoting open and efficient data sharing in the research community.

    • Radio frequency (RF) wireless power transfer (WPT) is a promising technology for 6G use cases. It enables a massive yet sustainable deployment of batteryless energy neutral (EN) devices at an unprecedented scale. Recent research on 6G is exploring high operating frequencies up to the THz spectrum, where antenna arrays with large apertures are capable of forming narrow, "laser-like" beams. At sub-10 GHz frequencies, physically large antenna arrays are considered that are operating in the array near field. Transmitting spherical wavefronts, power can be focused to a focal point rather than a beam, which allows for efficient and radiation-safe WPT. We formulate a multipath channel model comprising specular components and diffuse scattering to find the WPT power budget in a realistic indoor scenario. Specular components can be predicted by means of a geometric model. This is used to transmit power via multiple beams simultaneously, increasing the available power budget and expanding the initial access distance. We show that exploiting this "beam diversity" reduces the required fading margin for the initial access to EN devices.

    • Radio stripes (RSs) is an emerging technology in beyond 5G and 6G wireless networks to support the deployment of cell-free architectures. In this paper, we investigate the potential use of RSs to enable joint positioning and synchronization in the uplink channel at sub-6 GHz bands. The considered scenario consists of a single-antenna user equipment (UE) that communicates with a network of multiple-antenna RSs distributed over a wide area. The UE is assumed to be unsynchronized to the RSs network, while individual RSs are time- and phase-synchronized. We formulate the problem of joint estimation of position, clock offset and phase offset of the UE and derive the corresponding maximum-likelihood (ML) estimator, both with and without exploiting carrier phase information. To gain fundamental insights into the achievable performance, we also conduct a Fisher information analysis and inspect the theoretical lower bounds numerically. Simulation results demonstrate that promising positioning and synchronization performance can be obtained in cell-free architectures supported by RSs, revealing at the same time the benefits of carrier phase exploitation through phase-synchronized RSs.

    • The use of radio signal for position tracking (positioning) and navigation has a long tradition. Most notably, global navigation satellite systems (GNSS) are now widely applied for civilian applications, ranging all the way from tracking of cargo containers to gaming. However, GNSS suffers from limited coverage in dense urban areas and indoors and also from a limited position accuracy, which rules out most location-based applications that are concerned with the (natural) interaction of humans with their immediate surroundings, the physical environment in which we live, work, and spend our free time.This chapter highlights the expected benefits of the new wireless technologies proposed for the fifth generation (5G) of mobile communication systems and for the Internet of Things (IoT) for the purpose of improved radiopositioning. It starts with a summary of the most promising future application scenarios for high-accuracy positioning, followed by a discussion on the technical challenges arising from those applications, and on the expected features and limitations of 5G and IoT wireless systems with respect to positioning. These initial considerations are followed by four technical sections, focusing on measurement acquisition and modeling, position estimation, multipath-assisted positioning, and system-level studies. Finally, a range of experimental facilities are described, which have been used to validate the theoretical contributions.
