    • Background Thoracolumbar myelopathies associated with spinal cord and vertebral column lesions, with a similar clinical phenotype, but different underlying etiologies, occur in pugs. Objectives To further characterize the clinical and neuropathological characteristics of pugs with longstanding thoracolumbar myelopathy. Animals Thirty client-owned pure-bred pugs with a history of more than a month of ataxia and paresis of the pelvic limbs, suggesting a myelopathy localized to the thoracolumbar spinal cord, were included in the study. Methods Prospective clinicopathological study. Included pugs underwent a complete neurological examination and gross and histopathologic postmortem studies with focus on the spinal cord. Computed tomography (n = 18), magnetic resonance imaging (n = 17), and cerebrospinal fluid analysis (n = 27) were performed before or immediately after death. Results Twenty male and 10 female pugs had a median age at clinical onset of 84 months (interquartile range, 66-96). Affected pugs presented with a progressive clinical course and 80% were incontinent. There was circumferential meningeal fibrosis with concomitant focal, malacic, destruction of the neuroparenchyma in the thoracolumbar spinal cord in 24/30 pugs. Vertebral lesions accompanied the focal spinal cord lesion, and there was lympho-histiocytic inflammation associated or not to the parenchymal lesion in 43% of the pugs. Conclusions and Clinical Importance Meningeal fibrosis with associated focal spinal cord destruction and neighboring vertebral column lesions were common findings in pugs with long-standing thoracolumbar myelopathy.

  • Identification of a novel host-specific IgG protease in Streptococcus phocae subsp phocae

    • Streptococcus (S.) phocae subsp. phocae causes bronchopneumonia and septicemia in a variety of marine mammals. Especially in harbor seals infected with phocine distemper virus it plays an important role as an opportunistic pathogen. This study was initiated by the detection of IgG cleavage products in Western blot analysis after incubation of bacterial supernatant with harbor seal serum. Hence, the objectives of this study were the identification and characterization of a secreted IgG cleaving protease in S. phocae subsp. phocae isolated from marine mammals. To further identify the responsible factor of IgG cleavage a protease inhibitor profile was generated. Inhibition of the IgG cleaving activity by iodoacetamide and Z-LVG-CHN2 indicated that a cysteine protease is involved. Moreover, an anti-IdeS antibody directed against the IgG endopeptidase IdeS of S. pyogenes showed cross reactivity with the putative IgG protease of S. phocae subsp. phocae. The IgG cleaving factor of S. phocae subsp. phocae was identified through an inverse PCR approach and designated IdeP (Immunoglobulin G degrading enzyme of S. phocae subsp. phocae) in analogy to the cysteine protease IdeS. Notably, recombinant (r) IdeP is a host and substrate specific protease as it cleaves IgG from grey and harbor seals but not IgG from harbor porpoises or non-marine mammals. The identification of IdeP represents the first description of a protein in S. phocae subsp. phocae involved in immune evasion. Furthermore, the fact that IdeP cleaves solely IgG of certain marine mammals reflects functional adaption of S. phocae subsp. phocae to grey and harbor seals as its main hosts.

  • Liver histopathology of Baltic grey seals (Halichoerus grypus) over three decades

    • The liver plays an important role in the metabolism and elimination of endogenic and exogenic lipid-solublecompounds. Multiple studies have shown that polychlorinated biphenyls (PCB) and dichlorodiphenyl trichloroethane(DDT) lead to morphological changes in liver cells. The aim of the present study was therefore toanalyse liver changes over time in Baltic grey seals (Halichoerus grypus) and to correlate these with historical PCBand DDT contaminations. A total of 191 liver samples were collected between 1981 and 2015 in the Gulf ofBothnia and northern Baltic Proper. Six histological features were evaluated, including portal mononuclear cellinfiltration, random mononuclear cell infiltration, lipid granulomas, hepatocellular fat vacuoles, hepatic stellatecells and mild multifocal bile duct hyperplasia accompanied by portal fibrosis. Three of the six lesions showed asignificant correlation with age. Furthermore, a positive correlation between portal mononuclear cell infiltrationand mild multifocal bile duct hyperplasia was found. Additionally, lipid granulomas were significantly correlatedwith hepatic stellate cells. More importantly, hepatic stellate cells and mild multifocal bile duct hyperplasia werecorrelated with adipose tissue (blubber) concentrations of ƩPCB, measured in a subsample (n = 34) of all individuals.No correlation with lesions and ƩDDT concentrations were found. These results show that age is animportant factor for the development of these liver lesions, but PCBs burden may be an influencing factor. This isin agreement with previous studies of marine mammals in the Baltic Sea as well as in the Arctic. We thereforeconclude that not only age of the animals, but also exposure to PCBs should be taken into account when understandingand evaluating the current health status of Baltic grey seals.
