  • Water and heat coupling processes and its simulation in frozen soils : Current status and future research directions

    • To date, most studies on coupled-water-and-heat processes in frozen soils haves focused on the mechanism of changes in frozen soil and the contribution of climate change, hydrological processes, and ecosystems in cold regions. Several studies have demonstrated considerable improvements in the accuracy of simulating water and heat transfer processes in cold regions. However, substantial differences remain among the different models and parameterizations because of the lack of observations and in-depth understanding of the water and heat transfer processes. Hence, it is necessary to summarize recent advances in the simulation of water-and-heat-coupling processes and challenges for further research. Therefore, we present a theory-focused summary of progress in this field considering the aspects of water flow and coupled-water-and-heat transfer. The simulation progress is discussed in terms of physical process models; one type of model only considers the heat conduction transfer processes without water flow, and the other considers coupled-water-and-heat transfer processes. Aspects of model deficiencies related to non-conductive heat transfer and soil water transfer processes in the frozen soil are also summarized. Moreover, the major parameterizations are reviewed, including phase changes, freeze–thaw fronts, thermal conductivity, hydraulic conductivity, snow processes, surface parameterization schemes, ground ice, and lower boundary conditions. While models and parameterizations can suitably capture local-scale water and heat transfer processes in frozen soil, their applications are spatiotemporally constrained, requiring further improvement. We provide a theoretical basis for further studying water and heat transfer processes in frozen soil and suggest that future research should enhance the accuracy of frozen soil parameterization and improve the understanding of the coupling of water and heat transfer processes based on improved observation techniques and high-resolution data.

  • Convergence and divergence emerging in climatic controls of polynomial trends for northern ecosystem productivity over 2000–2018

    • Southwest China has been the largest terrestrial carbon sink in China over the past 30 years, but has recently experienced a succession of droughts caused by high precipitation variability, potentially threatening vegetation productivity in the region. Yet, the impact of precipitation anomalies on the vegetation primary productivity is poorly known. We used an asymmetry index (AI) to explore possible asymmetric productivity responses to precipitation anomalies in Southwest China from 2003 to 2018, using a precipitation dataset, combined with gross primary productivity (GPP), net primary productivity (NPP), and vegetation optical depth (VOD) products. Our results indicate that the vegetation primary productivity of Southwest China shows a negative asymmetry, suggesting that the increase of vegetation primary productivity during wet years exceeds the decrease during dry years. However, this negative asymmetry of vegetation primary productivity was shifted towards a positive asymmetry during the period of analysis, suggesting that the resistance of vegetation to drought, has increased with the rise in the occurrence of drought events. Among the different biomes, grassland vegetation primary productivity had the highest sensitivity to precipitation anomalies, indicating that grasslands are more flexible than other biomes and able to adjust primary productivity in response to precipitation anomalies. Furthermore, our results showed that the asymmetry of vegetation primary productivity was influenced by the effects of temperature, precipitation, solar radiation, and anthropogenic and topographic factors. These findings improve our understanding of the response of vegetation primary productivity to climate change over Southwest China.

  • Storage, patterns, and environmental controls of soil organic carbon stocks in the permafrost regions of the Northern Hemisphere

    • Large stocks of soil organic carbon (SOC) accumulated in the Northern Hemisphere permafrost regions may be vulnerable to climatic warming, but global estimates of SOC distribution and magnitude in permafrost regions still have large uncertainties. Based on multiple high-resolution environmental variables and a compiled soil sample dataset (>3000 soil profiles), we used machine-learning methods to estimate the size and spatial distribution of SOC for the top 3 m soils in the Northern Hemisphere permafrost regions. We also identified key environmental predictors of SOC. The results showed that the SOC storage for the top 3 m soil was 1079 ± 174 Pg C across the Northern Hemisphere permafrost regions (20.8 × 106 km2), including 1057 ± 167 Pg C in the northern permafrost regions and 22 ± 7 Pg C in the Third Pole permafrost regions. The mean annual air temperature and NDVI are the main controlling factors for the spatial distribution of SOC stocks in the northern and the Third Pole permafrost regions. Our estimations were more accurate than the existing global SOC stock maps. The results improve our understanding of the regional and global permafrost carbon cycle and their feedback to the climate system.

  • Climatic controls on stable carbon and nitrogen isotope compositions of temperate grasslands in northern China

    • Aims The natural abundances of stable carbon (C) and nitrogen (N) isotopes (delta C-13 and delta N-15) are extensively used to indicate the C and N biogeochemical cycles at large spatial scales. However, the spatial patterns of delta C-13 and delta N-15 in plant-soil systems of grasslands in northern China and their main driving factors across regional climatic gradient are still not well understood. Methods We measured plant and soil delta C-13 and delta N-15 compositions as well as their associated environmental factors across 2000 km climatic gradient (-0.2 to 9 degrees C; 152 to 502 mm) in grasslands of northern China. Results The soil delta C-13 and delta N-15 values in surface were lower than those in bottom for temperate typical steppe but had no significant differences for temperate meadow steppe and temperate desert steppe. Soil delta C-13 values declined with increasing soil organic carbon (SOC) but increased as mean annual temperature (MAT). These changes were attributed to the microbial decomposition rate. The delta N-15 values in soil and plant were negatively correlated with MAT and mean annual precipitation (MAP), which were mainly related to the low soil organic matter mineralization rate and the shift of dominant species from C-4 to C-3. Conclusions Our results indicate the spatial patterns and different influencing factors on delta C-13 and delta N-15 values along the climatic gradient in grasslands of northern China. The findings will provide scientific references for future research on the C and N biogeochemical cycles of temperate grasslands.

  • Silicon promotes biomass accumulation in Phragmites australis under waterlogged conditions in coastal wetland

    • Aims Previous studies have shown that silicon (Si) can affect plant growth and yield by regulating the availability of other nutrients. However, the mechanisms by which Si affects plant biomass accumulation in coastal wetlands are not well explored. Methods We conducted a sampling campaign across the whole growing season of Phragmites australis under waterlogging and drought conditions in coastal wetland, and quantified the effects of Si availability on biomass accumulation. Results Compared with drought condition, the waterlogged condition improved the utilization efficiency of nitrogen (N) and phosphorus (P) of P. australis regulated by higher Si contents. Meanwhile, the increased Si contents promoted the utilization of N and P in leaf, suggesting that the increase in Si contents optimizes the photosynthetic process. Lignin contents in P. australis decreased with the increasing Si contents, which confirmed that Si can replace structural carbon components. In addition, principal component analysis (PCA) showed aboveground biomass accumulation of P. australis was synchronized with Si accumulation, indicating that Si was a beneficial element to promote biomass accumulation. Conclusions Our study implies that increasing Si availability is conducive to biomass accumulation of P. australis in waterlogged wetlands, which will provide important scientific references for the management of coastal wetland ecosystem and the increase of global 'blue carbon' sequestration.

    • We report the generation and analysis of functional data from multiple, diverse experiments performed on a targeted 1% of the human genome as part of the pilot phase of the ENCODE Project. These data have been further integrated and augmented by a number of evolutionary and computational analyses. Together, our results advance the collective knowledge about human genome function in several major areas. First, our studies provide convincing evidence that the genome is pervasively transcribed, such that the majority of its bases can be found in primary transcripts, including non-protein-coding transcripts, and those that extensively overlap one another. Second, systematic examination of transcriptional regulation has yielded new understanding about transcription start sites, including their relationship to specific regulatory sequences and features of chromatin accessibility and histone modification. Third, a more sophisticated view of chromatin structure has emerged, including its inter-relationship with DNA replication and transcriptional regulation. Finally, integration of these new sources of information, in particular with respect to mammalian evolution based on inter- and intra-species sequence comparisons, has yielded new mechanistic and evolutionary insights concerning the functional landscape of the human genome. Together, these studies are defining a path for pursuit of a more comprehensive characterization of human genome function.

  • Preparation of magnetic MnFe2O4-Cellulose aerogel composite and its kinetics and thermodynamics of Cu(II) adsorption

    • In this paper, a MnFe2O4-Cellulose magnetic composite aerogel (MnCA) with high adsorption capacity was fabricated by in situ incorporating MnFe2O4 to regenerated cellulose hydrogel matrix, followed by CO2 supercritical drying. A green synthetic strategy was performed by using renewable cellulose materials, environmentally benign cellulose solvents and facile synthetic conditions. The results showed that the obtained magnetic cellulose aerogel had a continuous and tiered three dimensional network with interconnected fibrils of about 30 nm in width, which was similar to those of cellulose aerogel prepared from NaOH/urea solution via CO2 supercritical drying. Meanwhile, they had high specific surface areas of 236-288 m(2)/g and total pore volume of 0.55-0.88 cm(3)/g. In addition, the hybrid aerogel showed superparamagnetism with maximum saturation magnetization reaching up to 18.53 emu/g. The magnetic nanocomposite aerogel could be used for biological and environmental applications. The adsorption test showed that MnCA had rapid adsorption rate and excellent adsorption ability of removing heavy metal ions in aqueous solution which could attain to 63.3 mg/g within 100 min. Moreover, all the composite aerogels exhibited good reusability and could be easily reused from the water after adsorption.

    • The sulphur resistance and regeneration strategy of soot oxidation catalysts are critical to the application of catalytic diesel particulate filters (CDPFs). In the present work, Pt/MnOx-CeO2/Al2O3, Pt/Al2O3 and MnOx-CeO2/Al2O3 catalysts were sulphated and regenerated with ammonia. The soot oxidation activity of poisoned Pt/MnOx-CeO2/Al2O3 was recovered completely after 30 min reduction in 500 ppm NH3 at 550 degrees C. The amount, distribution and structure of sulphates were studied comprehensively for the sulphated and regenerated samples by thermogravimetric analysis (TGA), infrared (IR) spectroscopy, CO chemisorption and transmission electron microscopy (TEM). The NO oxidation of Pt catalysts was found to depend strongly on the particle size distribution, surface exposure and oxidation state of the precious metal. This, as well as the effect of the remaining surface sulphates, helped the regenerated catalysts achieve even higher catalytic activity for soot oxidation.

  • GLP-1 Receptor Activation Abrogates β-Cell Dysfunction by PKA Cα-Mediated Degradation of Thioredoxin Interacting Protein

    • Glucagon-like peptide 1 receptor (GLP-1R) agonist (Exendin-4) is a well-known agent used to improve β-cell dysfunctions via protein kinase A (PKA), but the detailed downstream molecular mechanisms are still elusive. We have now found that PKA Cα mediated- TXNIP phosphorylation and degradation played a vital role in the β-cell protective role of exendin-4. After PKA activator (Exendin-4 or FSK) treatment, PKA Cα could directly interact with TXNIP by bimolecular fluorescence complementation and Co-IP assays in INS-1 cells. And PKA Cα overexpression decreased TXNIP level, whereas TXNIP level was largely increased in PKA Cα-KO β-cells by CRISPR-Cas9. Interestingly, TXNIP overexpression or PKA Cα-KO has impaired β-cell functions, including loss of insulin secretion and activation of inflammation. PKA Cα directly phosphorylated TXNIP at Ser307 and Ser308 positions, leading to its degradation via activation of cellular proteasome pathway. Consistent with this observation, TXNIP (S307/308A) mutant resisted the degradation effects of PKA Cα. However, exendin-4 neither affected TXNIP level in TXNIP (S307/308A) mutant overexpressed β-cells nor in PKA Cα-KO β-cells. Moreover, exendin-4 treatment reduced the inflammation gene expression in TXNIP overexpressed β-cells, but exendin-4 treatment has no effect on the inflammation gene expression in TXNIP (S307/308A) overexpressed β-cells. In conclusion, our study reveals the integral role of PKA Cα/TXNIP signaling in pancreatic β-cells and suggests that PKA Cα-mediated TXNIP degradation is vital in β-cell protective effects of exendin-4.
