    • Optical density (OD) is widely used to estimate the density of cells in liquid culture, but cannot be compared between instruments without a standardized calibration protocol and is challenging to relate to actual cell count. We address this with an interlaboratory study comparing three simple, low-cost, and highly accessible OD calibration protocols across 244 laboratories, applied to eight strains of constitutive GFP-expressing E. coli. Based on our results, we recommend calibrating OD to estimated cell count using serial dilution of silica microspheres, which produces highly precise calibration (95.5% of residuals <1.2-fold), is easily assessed for quality control, also assesses instrument effective linear range, and can be combined with fluorescence calibration to obtain units of Molecules of Equivalent Fluorescein (MEFL) per cell, allowing direct comparison and data fusion with flow cytometry measurements: in our study, fluorescence per cell measurements showed only a 1.07-fold mean difference between plate reader and flow cytometry data.

    • The Visual Object Tracking challenge VOT2019 is the seventh annual tracker benchmarking activity organized by the VOT initiative. Results of 81 trackers are presented; many are state-of-the-art trackers published at major computer vision conferences or in journals in the recent years. The evaluation included the standard VOT and other popular methodologies for short-term tracking analysis as well as the standard VOT methodology for long-term tracking analysis. The VOT2019 challenge was composed of five challenges focusing on different tracking domains: (i) VOT-ST2019 challenge focused on short-term tracking in RGB, (ii) VOT-RT2019 challenge focused on "real-time" short-term tracking in RGB, (iii) VOT-LT2019 focused on long-term tracking namely coping with target disappearance and reappearance. Two new challenges have been introduced: (iv) VOT-RGBT2019 challenge focused on short-term tracking in RGB and thermal imagery and (v) VOT-RGBD2019 challenge focused on long-term tracking in RGB and depth imagery. The VOT-ST2019, VOT-RT2019 and VOT-LT2019 datasets were refreshed while new datasets were introduced for VOT-RGBT2019 and VOT-RGBD2019. The VOT toolkit has been updated to support both standard short-term, long-term tracking and tracking with multi-channel imagery. Performance of the tested trackers typically by far exceeds standard baselines. The source code for most of the trackers is publicly available from the VOT page. The dataset, the evaluation kit and the results are publicly available at the challenge website(1).

  • Dipyridamole enhances the anti-cancer ability of aspirin against colorectal cancer by inducing apoptosis in an unfolded protein response-dependent manner

    • PURPOSE: Available evidence indicates that dipyridamole enhances the anti-thrombotic effects of aspirin for the prevention of secondary strokes. Aspirin is a well-known non-steroid anti-inflammatory drug. This anti-inflammatory property has turned aspirin into a potential drug for inflammation-related cancers such as colorectal cancer (CRC). Here, we aimed to explore whether the anti-cancer effect of aspirin against CRC could be improved by combined administration with dipyridamole.METHODS: Population-based clinical data analysis was conducted to assess a possible therapeutic effect of combined dipyridamole and aspirin treatment in inhibiting CRC compared with either monotherapy. This therapeutic effect was further verified in different CRC mouse models, i.e. an orthotopic xenograft mouse model, an AOM/DSS mouse model, an Apc min/+ mouse model and a patient derived xenograft (PDX) mouse model. The in vitro effects of the drugs on CRC cells were tested using CCK8 and flow cytometry assays. RNA-Seq, Western blotting, qRT-PCR and flow cytometry were used to identify the underlying molecular mechanisms. RESULTS: We found that dipyridamole combined with aspirin had a better inhibitory effect on CRC than either monotherapy alone. The enhanced anti-cancer effect of the combined use of dipyridamole with aspirin was found to rely on the induction of an overwhelmed endoplasmic reticulum (ER) stress and subsequent pro-apoptotic unfolded protein response (UPR), which was different from the anti-platelet effect.CONCLUSIONS: Our data indicate that the anti-cancer effect of aspirin against CRC may be enhanced by combined administration with dipyridamole. In case further clinical studies confirm our findings, these may be repurposed as adjuvant agents.

  • Recent Progress on the Synthesis and Biomedical Properties of Natural Biopolymer Composites

    • Background: Natural biopolymers have drawn extensive attention because of their great biocompatibility, biodegradability, renewability, and the availability of various reactive functional groups for modifying and introducing novel components. In the last few years, numerous natural biopolymer composites have been exploited to improve their physical and chemical properties and add new functionalities.Methods: Herein, we summarize the current progress in three common classes of natural biopolymer-based composites, including alginate, chitosan, and gelatin.Results: The morphology characteristics, preparation methods, and unique functionalities of these biopolymer composites are also analyzed and discussed.Conclusion: Finally, the article offers an overview of recent progress in the main biomedical applications such as tissue engineering, wound-healing, and drug delivery, which inspires further progress in biopolymer composites with tailored mechanical property and stable characteristics for pharmaceutical and biomedical applications.

  • Application of an innovative grid-based surveillance strategy to ensure elimination and prevent reintroduction of malaria in high-risk border communities in China

    • Grid management is a grassroots governance strategy widely implemented in China since 2004 to improve the government's efficiency to actively find and solve problems among populated regions. A grid-based strategy surveillancing high-risk groups, including mobile and migrant populations (MMPs), in the China-Myanmar border region has played an indispensable role in promoting and consolidating the malaria elimination efforts by tracking and timely identification of potential importation or re-establishment of malaria among MMPs. A sequential mixed methods was implementated to explore the operational mechanism and best practices of the grid-based strategy including through the focus group discussions (FGDs), comparison of before and after the implementation of a grid-based strategy in the field sites, and data collection from the local health system.This paper distills the implementation mechanism and highlights the role of the grid-based strategy in the elimination and prevention of re-establishment of malaria transmission.

  • Influence of B2O3 on Viscosity of High Ti-bearing Blast Furnace Slag

    • The influence of B2O3 on the viscosity of high Ti-bearing BF slag is studied under Ar atmosphere from 1 773 K (1 500 degrees C) to about 1 593 K (1 320 degrees C). The results show that the addition of B2O3 can decrease the viscosity of high Ti-bearing BF Slag containing UPC, consequently improve its fluidity. With the existence of 1.5% UPC, when the content of B2O3 increased from 0 to 4.1%, the volume fraction of perovskite decreased from 7.7% to 1.6% at 1 610 K. Meanwhile, the liquidus temperatures decreased from 1 619 K to 1 613 K. At a lower temperature, B2O3 was easy to form a eutectic and decreased the viscosity of molten slag. At a higher temperature, some tetrahedron [BO4](5-) changed into triangle [BO3](3-) which would disintegrate the chains/molecules of molten matrix and decreased its viscosity.

  • Intra- and Inter-Self-Assembly of Identical Supramolecules on Silver Surfaces

    • Self-assembly of identical organometallic supra -molecules into ordered superstructures is of great interest in both chemical science and nanotechnology due to its potential to generate neoteric properties through collective effects. In this work, we demonstrate that large-scale self-organization of atomically precise organometallic supramolecules can be achieved through cascaded on-surface chemical reactions, by the combination of intra-and inter-supramolecular interactions. Supramolecules with defined size and shape are first built through intramolecular reaction and intermolecular metal coordination, followed by the formation of well-ordered two-dimensional arrays with the assistance of Br atoms by-C-HmiddotmiddotmiddotBr interactions. The mechanism of this process has been investigated from the perspectives of thermodynamics and kinetics.

  • De Novo Sequence and Copy Number Variants Are Strongly Associated with Tourette Disorder and Implicate Cell Polarity in Pathogenesis

    • We previously established the contribution of de novo damaging sequence variants to Tourette disorder (TD) through whole-exome sequencing of 511 trios. Here, we sequence an additional 291 TD trios and analyze the combined set of 802 trios. We observe an overrepresentation of de novo damaging variants in simplex, but not multiplex, families; we identify a high-confidence TD risk gene, CELSR3 (cadherin EGF LAG seven-pass G-type receptor 3); we find that the genes mutated in TD patients are enriched for those related to cell polarity, suggesting a common pathway underlying pathobiology; and we confirm a statistically significant excess of de novo copy number variants in TD. Finally, we identify significant overlap of de novo sequence variants between TD and obsessive-compulsive disorder and de novo copy number variants between TD and autism spectrum disorder, consistent with shared genetic risk.

  • Circulation of Circumpolar Deep Water and marine environment traced by I-127 and I-129 speciation in the Amundsen Sea Polynya, Antarctica

    • The long-lived anthropogenic I-129 released from human nuclear activities has been widely employed as an effective oceanographic tracer to investigate circulation of water masses in marine environment. Depth profiles of seawater collected from the Amundsen Sea Polynya, Antarctica were analyzed for total I-129 and I-127, as well as their species of iodide and iodate. The measured I-129 concentrations ((1.15-3.43) x 10(6) atoms/L) and I-129/I-127 atomic ratios ((0.53-1.19) x 10(-11)) indicate that anthropogenic I-129 has not only reached the Antarctic surface marine environment but also the deep water due to a strong vertical mixing of water masses. The Circumpolar Deep Water (CDW) flowed southward along continental shelf towards the ice shelf zone (74.25 degrees S) at a depth of 1025 m and then migrated upward and northward to the polynya and finally to the sea ice zone (71.95 degrees S). The maximum upwelling depth of the CDW was around 200 m in the polynya. The source of I-129(-) in the polynya is predominantly the intrusion of source waters rather than the in-situ reduction of iodate by phytoplankton, implying a considerably slow reduction process of iodate to iodide in this region.

    • Anthropogenic I-129 has been released to the environment in different ways and chemical species by human nuclear activities since the 1940s. These sources provide ideal tools to trace the dispersion of volatile pollutants in the atmosphere. Snow and seawater samples collected in Bellingshausen, Amundsen, and Ross Seas in Antarctica in 2011 were analyzed for I-129 and I-127, including organic forms; it was observed that I-129/I-127 atomic ratios in the Antarctic surface seawater ((6.1-13) x 10(-12)) are about 2 orders of magnitude lower than those in the Antarctic snow ((6.8-9.5) x 10(-1)0), but 4-6 times higher than the prenuclear level (1.5 x 10(-12)), indicating a predominantly anthropogenic source of I-129 in the Antarctic environment. The I-129 level in snow in Antarctica is 24 orders of magnitude lower than that in the Northern Hemisphere, but is not significantly higher than that observed in other sites in the Southern Hemisphere. This feature indicates that I-129 in Antarctic snow mainly originates from atmospheric nuclear weapons testing from 1945 to 1980; resuspension and re-emission of the fallout I-129 in the Southern Hemisphere maintains the I-129 level in the Antarctic atmosphere. I-129 directly released to the atmosphere and re-emitted marine discharged I-129 from reprocessing plants in Europe might not significantly disperse to Antarctica.
