  • Size-segregated particle number and mass concentrations from different emission sources in urban Beijing

    • Although secondary particulate matter is reported to be the main contributor of PM2.5 during haze in Chinese megacities, primary particle emissions also affect particle concentrations. In order to improve estimates of the contribution of primary sources to the particle number and mass concentrations, we performed source apportionment analyses using both chemical fingerprints and particle size distributions measured at the same site in urban Beijing from April to July 2018. Both methods resolved factors related to primary emissions, including vehicular emissions and cooking emissions, which together make up 76% and 24% of total particle number and organic aerosol (OA) mass, respectively. Similar source types, including particles related to vehicular emissions (1.6 +/- 1.1 mu gm(-3); 2.4 +/- 1.8 x 10(3) cm(-3) and 5.5 +/- 2.8 x 10(3) cm(-3) for two traffic-related components), cooking emissions (2.6 +/- 1.9 mu gm(-3) and 5.5 +/- 3.3 x 10(3) cm(-3)) and secondary aerosols (51 +/- 41 mu gm(-3) and 4.2 +/- 3.0 x 10(3) cm(-3)), were resolved by both methods. Converted mass concentrations from particle size distributions components were comparable with those from chemical fingerprints. Size distribution source apportionment separated vehicular emissions into a component with a mode diameter of 20 nm (traffic-ultrafine) and a component with a mode diameter of 100 nm (traffic-fine). Consistent with similar day- and nighttime diesel vehicle PM2.5 emissions estimated for the Beijing area, traffic-fine particles, hydrocarbon-like OA (HOA, traffic-related factor resulting from source apportionment using chemical fingerprints) and black carbon (BC) showed similar diurnal patterns, with higher concentrations during the night and morning than during the afternoon when the boundary layer is higher. Traffic-ultrafine particles showed the highest concentrations during the rush-hour period, suggesting a prominent role of local gasoline vehicle emissions. In the absence of new particle formation, our re-sults show that vehicular-related emissions (14% and 30% for ultrafine and fine particles, respectively) and cooking-activity-related emissions (32 %) dominate the particle number concentration, while secondary particulate matter (over 80 %) governs PM2.5 mass during the non-heating season in Beijing.

  • Building bridges to operationalise one health : A Sino-Swedish collaboration to tackle antibiotic resistance

    • Antibiotic resistance is a complex global health challenge. The recent Global Action Plan on antimicrobial resistance highlights the importance of adopting One Health approaches that can cross traditional disciplinary boundaries. We report on the early experiences of a multisectoral Sino-Swedish research project that aims to address gaps in our current knowledge and seeks to improve the situation through system-wide interventions. Our research project is investigating antibiotic use and resistance in a rural area of China through a combination of epidemiological, health systems and laboratory investigations. We reflect here on the challenges inherent in conducting long distance cross-disciplinary collaborations, having now completed data and sample collection for a baseline situation analysis. In particular, we recognise the importance of investing in aspects such as effective communication, shared conceptual frameworks and leadership. We suggest that our experiences will be instructive to others planning to develop similar international One Health collaborations.

  • Progress and Challenges in Studying Regional Permafrost in the Tibetan Plateau Using Satellite Remote Sensing and Models

    • © Copyright © 2020 Jiang, Zheng, Yi, Chen, Zhang, Yang and Miller. Recent climate change has induced widespread soil thawing and permafrost degradation in the Tibetan Plateau. Significant advances have been made in better characterizing Tibetan Plateau soil freeze/thaw dynamics, and their interaction with local-scale ecohydrological processes. However, factors such as sparse networks of in-situ sites and short observational period still limit our understanding of the Tibetan Plateau permafrost. Satellite-based optical and infrared remote sensing can provide information on land surface conditions at high spatial resolution, allowing for better representation of spatial heterogeneity in the Tibetan Plateau and further infer the related permafrost states. Being able to operate at “all-weather” conditions, microwave remote sensing has been widely used to retrieve surface soil moisture, freeze/thaw state, and surface deformation, that are critical to understand the Tibetan Plateau permafrost state and changes. However, coarse resolution (>10km) of current passive microwave sensors can add large uncertainties to the above retrievals in the Tibetan Plateau area with high topographic relief. In addition, current microwave remote sensing methods are limited to detections in the upper soil layer within a few centimetres. On the other hand, algorithms that can link surface properties and soil freeze/thaw indices to permafrost properties at regional scale still need improvements. For example, most methods using InSAR (interferometric synthetic aperture radar) derived surface deformation to estimate active layer thickness either ignore the effects of vertical variability of soil water content and soil properties, or use site-specific soil moisture profiles. This can introduce non-negligible errors when upscaled to the broader Tibetan Plateau area. Integrating satellite remote sensing retrievals with process models will allow for more accurate representation of Tibetan Plateau permafrost conditions. However, such applications are still limiting due to a number of factors, including large uncertainties in current satellite products in the Tibetan Plateau area, and mismatch between model input data needs and information provided by current satellite sensors. Novel approaches to combine diverse datasets with models through model initialization, parameterization and data assimilation are needed to address the above challenges. Finally, we call for expansion of local-scale observational network, to obtain more information on deep soil temperature and moisture, soil organic carbon content, and ground ice content.

  • Soil freeze/thaw dynamics strongly influences runoff regime in a Tibetan permafrost watershed: Insights from a process-based model

    • The Tibetan Plateau (TP) is a region rich in extensive frozen ground and the source of many major Asian rivers. However, how soil freeze/thaw (F/T) dynamics influence runoff production at the catchment scale in the TP is poorly understood. This study employs a process-based permafrost hydrology model with a new soil parameterization to investigate soil F/T dynamics and their impact on runoff in a TP permafrost watershed, i.e., the source region of Yangtze River (SRYR). The revised model separates soil evaporation and plant transpiration, and accounts for the influence of soil gravel and organic carbon content, as well as variation in saturated hydraulic conductivity along the soil profile. Validation results demonstrate that the revised model accurately simulates daily soil temperature (mean RMSE of 1.3 degrees C), soil moisture (mean ubRMSE of 0.05 cm3 cm-3), and runoff discharge (NSE = 0.82). The results reveal different altitudinal patterns of warming trend between permafrost and seasonally frozen ground (SFG). Warming rates in SFG area increase monotonously with elevation, while a turning point is observed in permafrost region around 4800 m. With active layer deepening, deep-soil water content increases but primarily replenishes soil water storage rather than directly contributing to runoff recharge, while rootzone and the middle part of the active layer become drier. Soil F/T cycles in the permafrost region exert stronger influences on runoff process compared to SFG. Delayed soil thaw onset generally results in higher spring runoff coefficient, while delayed soil freeze onset is related to slower runoff recession. The freezing zero-curtain period is likely to impact the continuity of runoff recession processes by affecting the connectivity of groundwater flow channels. These findings uncover the regulatory mechanisms of soil F/T dynamics on runoff production and river discharge characteristics, providing a fundamental basis for predicting permafrost hydrology responses to future climate change in the TP.

    • INTRODUCTION: To effectively minimise the emergence and dissemination of antibiotic resistant bacteria, a holistic One Health approach is called for. The Sino-Swedish Integrated Multisectoral Partnership for Antibiotic Resistance Containment is a cross-sectoral and integrated project on antibiotic resistance, conducted in Shandong Province in China. This paper outlines the overall study protocol for the project. To our knowledge, this is the first research programme aiming to take a true holistic approach across multiple sectors simultaneously in China, and the first to incorporate both antibiotic use and infection prevention and control in addition to antibiotic resistance patterns. The project aims to address gaps in current knowledge and seeks to improve the situation through a system-wide intervention. By using a One Health approach we can address important research questions that individual discipline investigations are unable to. The results obtained should thus more closely reflect the world in which human health, animal health and the environment are inextricably and intimately interlinked.METHODS AND ANALYSIS: Both quantitative and qualitative studies are included for households from 12 villages, their surrounding environment and a tertiary care hospital in a nearby town. The studies include analyses of antibiotic consumption for humans and pigs; qualitative and quantitative data on perceptions, knowledge and attitudes; faecal carriage of extended spectrum β-lactamase and carbapenemase-producing Enterobacteriaceae from pigs and humans, and occurrence in household drinking water, surface water, waste water and clinical bacterial isolates from the hospital. Carriage of methicillin-resistant Staphylococcus aureus in humans, household pigs and clinical bacterial isolates is also investigated. Furthermore, potential inter-relationships between these sources are analysed. A multifaceted One Health intervention is designed and implemented in 6 of the 12 villages. Repeated and continuous data collections take place over 2 years, where the repeated data collection is performed after 1 year of intervention. Comparisons are made between intervention and control villages, before and after the intervention.ETHICS: Ethics approval was obtained from the first Affiliated Hospital, College of Medicine, Zhejiang University, China, reference number 2015#185 and 2015#283.

    • To evaluate the early stages of the host response to chancroid bacterium Haemophilus ducreyi, we investigated the in vitro responses of monocyte-derived dendritic cells (DCs) and macrophages (MQs) to this pathogen and Haemophilus influenzae. The phagocytic activities and pro-inflammatory cytokine secretion profiles of the antigen-presenting cells (APCs) were analyzed after exposure to gentamycin-killed bacteria, H. ducreyi lipooligosaccharide (LOS), and purified cytolethal distending toxin (HdCDT). T-cell proliferation and cytokine release were examined after co-culturing isolated autologous CD4+ T cells with antigen-pulsed APCs. Both the DCs and MQs phagocytosed H. ducreyi and H. influenzae, as estimated by flow cytometry. All of the strains induced APC secretion of TNF-alpha, IL-6, IL-8, and IL-12, as measured by ELISA. Other human cells, particularly endothelial cells and fibroblasts, also produced cytokines when stimulated with these bacteria. Purified LOS at concentration 1 microg/ml induced two to threefold lower levels of cytokines than the whole bacteria, which indicates that other components are involved in immune activation. HdCDT inhibited partially the production of the aforementioned cytokines. High levels of IFN-gamma, but not of IL-4 and IL-13, were secreted by T cells after activation by either DCs or MQs that were pre-exposed to bacteria, indicating the Th1 nature of the immune response. The levels of T-cell proliferation induced by H. ducreyi were lower than those induced by H. influenzae. HdCDT-treated APCs did not display cytokine responses or T-cell proliferation. These results indicate that HdCDT intoxication, which results in progressive apoptosis of APCs, may hamper early stage immune responses.
