  • Genomic basis of the distinct biosynthesis of β-glucogallin, a biochemical marker for hydrolyzable tannin production, in three oak species

    • Hydrolyzable tannins (HTs), predominant polyphenols in oaks, are widely used in grape wine aging, feed additives, and human healthcare. However, the limited availability of a high-quality reference genome of oaks greatly hampered the recognition of the mechanism of HT biosynthesis. Here, high-quality reference genomes of three Asian oak species (Quercus variabilis, Quercus aliena, and Quercus dentata) that have different HT contents were generated. Multi-omics studies were carried out to identify key genes regulating HT biosynthesis. In vitro enzyme activity assay was also conducted. Dual-luciferase and yeast one-hybrid assays were used to reveal the transcriptional regulation. Our results revealed that β-glucogallin was a biochemical marker for HT production in the cupules of the three Asian oaks. UGT84A13 was confirmed as the key enzyme for β-glucogallin biosynthesis. The differential expression of UGT84A13, rather than enzyme activity, was the main reason for different β-glucogallin and HT accumulation. Notably, sequence variations in UGT84A13 promoters led to different trans-activating activities of WRKY32/59, explaining the different expression patterns of UGT84A13 among the three species. Our findings provide three high-quality new reference genomes for oak trees and give new insights into different transcriptional regulation for understanding β-glucogallin and HT biosynthesis in closely related oak species.

  • Emerging contaminants: A One Health perspective

    • Environmental pollution is escalating due to rapid global development that often prioritizes human needs over planetary health. Despite global efforts to mitigate legacy pollutants, the continuous introduction of new substances remains a major threat to both people and the planet. In response, global initiatives are focusing on risk assessment and regulation of emerging contaminants, as demonstrated by the ongoing efforts to establish the UN's Intergovernmental Science-Policy Panel on Chemicals, Waste, and Pollution Prevention. This review identifies the sources and impacts of emerging contaminants on planetary health, emphasizing the importance of adopting a One Health approach. Strategies for monitoring and addressing these pollutants are discussed, underscoring the need for robust and socially equitable environmental policies at both regional and international levels. Urgent actions are needed to transition toward sustainable pollution management practices to safeguard our planet for future generations.

    • Sturgeon muscle byproduct collected after caviar production is usually not fully utilized, and sometimes may be discarded, thus causing a lot of waste. Yet dietary protein hydrolysates, which may be derived from sturgeon muscle, have been reported to have versatile beneficial biological activities. Studying the biological activities of sturgeon muscle-derived hydrolysates holds much promise for adding value to sturgeon. The current study aimed to study the therapeutic anti-inflammatory effects of sturgeon muscle-derived hydrolysates and the underlying mechanisms. The administration of sturgeon hydrolysates (SH) significantly decreased the severity of DSS-induced damage, evidenced by increased body weight, colon length, and decreased disease activity index (DAI) and histological scores. SH also inhibited myeloperoxidase (MPO) activity and reduced the serum levels of IL-6, IL-1 beta, and TNF-alpha. Western blotting results revealed that SH suppressed DSS-induced activation of the NF-kappa B and MAPK pathways in the colon. Furthermore, SH partially restored the alteration of the gut microbiota in colitic mice. SH increased the Bacteroidetes/Firmicutes ratio and the relative abundance of Ruminococcaceae, Porphyromonadaceae, and Bacteroidetes S24-7, while decreased the abundance of potentially harmful bacteria Erysipelotrichaceae and Enterococcaceae. These results suggest that SH inhibited DSS-induced colitis by regulating the NF-kappa B and MAPK pathways and modulating microbiota composition.

    • MOOC, which stands for massive open online course, has attracted millions of users around the world and it has a promise to be a very important element of future education. However, there is a little research on users' adoption of MOOCs. This paper aims to improve the understanding of users' behaviour intention to use MOOCs. The proposed research model is an extension of technology acceptance model with three factors from the institutional theory. And an empirical study with 279 subjects was conducted to test this model in China. The results indicate that both perceived usefulness and perceived ease of use directly affect users' behaviour intention to use MOOCs significantly. Another interesting finding is that mimetic pressures also have a significant positive influence on users' behaviour intention to use MOOCs.

    • This research aims to investigate the differences in the adoption of smartphones between middle aged adults and older adults in China. Based on a literature review from previous research, a research model with eight research hypotheses was developed by extending UTAUT with a consideration of observability and compatibility from IDT, and perceived enjoyment and price value. This research model was empirically examined using survey data from 196 middle aged adults and 146 older adults respectively from China. The findings indicated that the effects of perceived enjoyment, compatibility, and observability on users’ intention to use smartphones were significant, but no age differences between middle aged adults and older age adults were found to exist. Furthermore, the findings also identified age-related differences in the use and adoption of smartphones. The effects of performance expectancy and social influence on users’ intention to use smartphones were moderated by age, such that it was significant for older adults but insignificant for middle aged adults.

Five research hypotheses were positively significant supported, while three research hypotheses were rejected in this study. The results suggest that social influence, observability, compatibility, performance expectancy and perceived enjoyment, are important determinants for the use and adoption of smartphones with older adults in China.

    • This research aims to study the role of trust towards the adoption of mobile services in China. This study examined users’ adoption of general mobile services by extending TAM with additional trust-related constructs. Based on the literature review from previous research, a research model with 10 research hypotheses is proposed in the study. This research model is empirically evaluated using survey data collected from a sample of 373 subjects. Seven research hypotheses are positively significant supported, while three research hypotheses are rejected in this study. The results indicate that both perceived reputation and perceived structural assurance directly affects the consumers’ trust in mobile services. However, perceived environment risk does not have positive effect on consumers’ trust in mobile services. Another interesting finding is that consumers’ trust in mobile services does not have direct positive effect on users’ intention to use mobile services.

    • Triplet acceptors have been developed to construct high-performance organic solar cells (OSCs) as the long lifetime and diffusion range of triplet excitons may dissociate into free charges instead of net recombination when the energy levels of the lowest triplet state (T-1) are close to those of charge-transfer states ((CT)-C-3). The current triplet acceptors were designed by introducing heavy atoms to enhance the intersystem crossing, limiting their applications. Herein, two twisted acceptors without heavy atoms, analogues of Y6, constructed with large pi-conjugated core and D-A structure, were confirmed to be triplet materials, leading to high-performance OSCs. The mechanism of triplet excitons were investigated to show that the twisted and D-A structures result in large spin-orbit coupling (SOC) and small energy gap between the singlet and triplet states, and thus efficient intersystem crossing. Moreover, the energy level of T-1 is close to (CT)-C-3, facilitating the split of triplet exciton to free charges.

    • To support nonsequential writes, persistent cache (PC) is constructed in drive managed SMR (DM-SMR) drive. However, PC cleaning introduces drastic performance degradation and enlarges tail latencies. In this article, we propose to utilize reinforcement learning (RL) to mitigate the longtail latency of PC cleaning. Our scheme uses the lightweight Q-learning method to monitor and learn the idle time of I/O workloads, based on which PC cleaning is intelligently guided, thus maximally exploit idle time between requests and hiding tail latency from normal requests. In addition, a multiagent RL scheme with clustering algorithm is adopted to further mitigate the tail latencies and adapt to variable workloads. We emulate a DM-SMR drive inside a Linux device driver to implement our proposed scheme. According to the experimental results, our scheme can effectively reduce the tail latency by 59.45% at the 99.9th percentile and the average latency by 48.75% compared with a typical shingled magnetic recording (SMR) design.
