  • 9-oxo-ODAs suppresses the proliferation of human cervical cancer cells through the inhibition of CDKs and HPV oncoproteins

    • Mucosal human papillomavirus (HPV) subtypes 16 and 18 are causative agents of cervical cancer, a leading cause of cancer-related deaths among women worldwide. In Japan, eggplant calyx is a folk remedy used to treat common warts. 9-oxo-(10E,12E)-octadecadienoic acid, isolated from eggplant calyx, may have antitumor effects. This study investigated the antitumor effects of 9-oxo-(10E, 12Z)-octadecadienoic acid and 9-oxo-(10E,12E)-octadecadienoic acid (9-oxo-ODAs) on human cervical cancer cells. 9-oxo-ODAs suppressed the proliferation of human cervical cancer cell lines (HeLa, and SiHa) in a concentration-dependent manner (IC50 = 25–50 µM). FCM analysis revealed that 9-oxo-ODAs induced apoptosis. Transcriptome, proteomics, and enrichment analyses revealed that treatment with 9-oxo-ODAs significantly altered the cell cycle and p53 pathways and decreased cyclin-dependent kinase 1 (CDK1) protein expression. Real-time PCR analysis demonstrated that 9-oxo-ODAs reduced CDK1 mRNA expression in a concentration-dependent manner. In vitro, 9-oxo-ODAs reduced the HPV oncoprotein expression. In ex vivo human cervical cancer tissues, 9-oxo-ODAs decreased CDK1 expression and increased cleaved caspase 3, an apoptosis marker. Further, 9-oxo-ODAs showed the potential to suppressed metastatic formation and growth of cervical cancer in vivo. These findings suggest that 9-oxo-ODAs induce cell cycle arrest and apoptosis in HPV-positive human cervical cancer cells, and this process involves CDK1. Consequently, 9-oxo-ODAs may be potential therapeutic agents for cervical cancer.

    • Two novel vanadium complexes, [V(IV)O(bp-O)(HSO4)] (1) and [V(IV)O(bp-OH)Cl2] x CH3OH (2 x CH3OH), where bp-OH is 2-{[bis(pyrid-2-yl)methyl]amine}methylphenol, were prepared and structurally characterised. EPR spectra of methanol solutions of 2 suggest exchange of Cl- for CH3OH and partial conversion to [VO(bp-OH)(CH3OH)3]2+. Speciation studies on the VO2+-bpOH system in a water/dmso mixture (4:1 v/v) revealed [VO(bp-O)(H2O)n]+ as the dominating species in the pH range 2-7. The insulin-mimetic properties of 1 and 2, [V(IV)O(SO4)tpa] (3), [V(IV)O(pic-trpMe)2] (5) and the new mixed-ligand complexes [V(V)O(pic-trpH)tpa]Cl2 (4Cl2) and [V(V)O(pic-OEt)tpa]Cl2 (6Cl2), tpa = tris(pyrid-2-yl)methylamine, picH-trpH = 2-carboxypyridine-5-(L-tryptophan)carboxamide (picH-trpMe is the respective tryptophanmethyl ester), pic-OEt = 5-carboethoxypyridine-2-carboxylic acid, were evaluated with rat adipocytes, employing two lipolysis assays (release of glycerol and free fatty acids (FFA)), respectively and a lipogenesis assay (incorporation of glucose into lipids). The IC50 values for the inhibition of lipolysis in the FFA assay vary between 0.41 (+/-0.03) (5) and 21.2 (+/-0.6) mM (2), as compared to 0.81 (+/-0.2) mM for VOSO4.

    • A hybrid power generation system using biomass gasification and concentrated solar thermal processes (CSTP) was proposed. We analysed the system performance of a combined cycle with a bubbling fluidized bed gasifier (BFG) using CO2-H2O-O2 mixture and CSTP using molten salt. BFG was simulated by a semi-kinetic model, applying a continuously stirred tank reactor (CSTR) model for the bed behaviour and a plug flow reactor (PFR) model for the freeboard reactions. Operating conditions of the plant and heat exchanger network were optimized separately. The current paper shows the effects of gasifying agents and heat input from CSTP on system efficiency among examined parameters. Pure steam at the molar ratio of steam to carbon in biomass around 0.5 was optimum gasifying agent in this system. By increasing heat input from CSTP, marginal efficiency of biomass-to-electricity was enhanced by 6.2-6.5% (from 39.9 ∼41.2% to 46.1∼47.5%).

    • This paper describes the investigation of a hybrid power production system from biomass and solar energy. This paper suggests integration through heat exchanger network as a useful approach to obtain the synergy between biomass and solar. Biomass is first gasified in a bubbling fluidized bed (BFB) gasifier, and then syngas is used in a gas turbine. Excess heat exists in this sub-system and concentrated solar thermal process (CSTP) while there is a demand of steam for generating gasifying agent. Steam Rankine cycle exploits the heat created by these thermal streams to generate power while satisfying the steam demands. Thermodynamic performance was analyzed by process modelling with a semi-kinetic model of BFB gasifier and pinch analyses. The composition and temperature of gasifying agent showed some effect on the overall efficiency of the system. Higher overall efficiency of the system was achieved at higher temperature and higher O2 fraction in the O2-steam mixture as gasifying agent. The increase in thermal input from CSTP had positive effect on overall efficiency of the hybrid system until thermal input from CSTP becomes dominant against thermal stream related to the gasifier and the gas turbine.
