    • To accommodate the demand of exponentially increased global wireless data traffic, the prospective data rates for wireless communication in the market place will soon reach 100 Gbit/s and beyond. In the lab environment, wireless transmission throughput has been elevated to the level of over 100 Gbit/s attributed to the development of photonic-assisted millimeter wave (MMW) and THz technologies. However, most of recent demonstrations with over 100 Gbit/s data rates are based on spatial or frequency division multiplexing techniques, resulting in increased system's complexity and energy consumption. Here, we experimentally demonstrate a single channel 0.4 THz photonic-wireless link achieving a net data rate of beyond 100 Gbit/s by using a single pair of THz emitter and receiver, without employing any spatial/frequency division multiplexing techniques. The high throughput up to 106 Gbit/s within a single THz channel is enabled by combining spectrally efficient modulation format, ultra-broadband THz transceiver and advanced digital signal processing (DSP) routine. Besides that, our demonstration from system-wide implementation viewpoint also features high transmission stability, and hence shows its great potential to not only decrease the system's complexity, but also meet the requirements of prospective data rates for bandwidth-hungry short-range wireless applications.

  • A draft Diabrotica virgifera virgifera genome : insights into control and host plant adaption by a major maize pest insect

    • Background: Adaptations by arthropod pests to host plant defenses of crops determine their impacts on agricultural production. The larval host range of western corn rootworm, Diabrotica virgifera virgifera (Coleoptera: Chrysomelidae), is restricted to maize and a few grasses. Resistance of D. v. virgifera to crop rotation practices and multiple insecticides contributes to its status as the most damaging pest of cultivated maize in North America and Europe. The extent to which adaptations by this pest contributes to host plant specialization remains unknown. Results: A 2.42 Gb draft D. v. virgifera genome, Dvir_v2.0, was assembled from short shotgun reads and scaffolded using long-insert mate-pair, transcriptome and linked read data. K-mer analysis predicted a repeat content of ≥ 61.5%. Ortholog assignments for Dvir_2.0 RefSeq models predict a greater number of species-specific gene duplications, including expansions in ATP binding cassette transporter and chemosensory gene families, than in other Coleoptera. A majority of annotated D. v. virgifera cytochrome P450s belong to CYP4, 6, and 9 clades. A total of 5,404 transcripts were differentially-expressed between D. v. virgifera larvae fed maize roots compared to alternative host (Miscanthus), a marginal host (Panicum virgatum), a poor host (Sorghum bicolor) and starvation treatments; Among differentially-expressed transcripts, 1,908 were shared across treatments and the least number were between Miscanthus compared to maize. Differentially-expressed transcripts were enriched for putative spliceosome, proteosome, and intracellular transport functions. General stress pathway functions were unique and enriched among up-regulated transcripts in marginal host, poor host, and starvation responses compared to responses on primary (maize) and alternate hosts. Conclusions: Manual annotation of D. v. virgifera Dvir_2.0 RefSeq models predicted expansion of paralogs with gene families putatively involved in insecticide resistance and chemosensory perception. Our study also suggests that adaptations of D. v. virgifera larvae to feeding on an alternate host plant invoke fewer transcriptional changes compared to marginal or poor hosts. The shared up-regulation of stress response pathways between marginal host and poor host, and starvation treatments may reflect nutrient deprivation. This study provides insight into transcriptomic responses of larval feeding on different host plants and resources for genomic research on this economically significant pest of maize.

  • Biocatalytic Access to 1,4-Diazepanes via Imine Reductase-Catalyzed Intramolecular Asymmetric Reductive Amination

    • An enzymatic intramolecular asymmetric reductive amination has been developed for the synthesis of chiral 1,4-diazepanes. Several enantiocomplementary IREDs were identified for the synthesis of (R)- and (S)-5-chloro-2-(5-methyl-1,4-diazepan-1-yl)benzo[d]oxazole with high enan- tioselectivity. The catalytic efficiency of (R)-selective IRED from Leishmania major (IR1) and (S)-selective IRED from Micromonospora echinaurantiaca IR25) was 0.027 and 0.962 s(-1) mM(-1), respectively. To further improve the catalytic efficiency of IR1, its double mutant Y194F/D232H was identified by saturation mutagenesis and iterative combinatorial mutagenesis, which exhibited 61-fold in the catalytic efficiency relative to that of wild-type enzyme. The density functional calculations and molecular dynamics simulations provided some insights into the molecular basis for the improved activity of mutant Y194F/D232H. Furthermore, Y194F/D232H and IR25 were applied to access a range of different substituted 1,4-diazepanes with high enantiomeric excess (from 93 to >99%). This study offers an effective method for construction of chiral 1,4-diazepanes of pharmaceutical importance via imine reductase-catalyzed intramolecular reductive amination of the corresponding aminoketones.

  • Quantifying the conductivity of a single polyene chain by lifting with an STM tip

    • Conjugated polymers are promising candidates for molecular wires in nanoelectronics, with flexibility in mechanics, stability in chemistry and variety in electrical conductivity. Polyene, as a segment of polyacetylene, is a typical conjugated polymer with straightforward structure and wide-range adjustable conductance. To obtain atomic scale understanding of charge transfer in polyene, we have measured the conductance of a single polyene-based molecular chain via lifting it up with scanning tunneling microscopy tip. Different from semiconducting characters in pristine polyene (polyacetylene), high conductance and low decay constant are obtained, along with an electronic state around Fermi level and characteristic vibrational mode. These observed phenomena result from pinned molecular orbital owing to molecule-electrode coupling at the interface, and weakened bond length alternation due to electron-phonon coupling inside single molecular chain. Our findings emphasize the interfacial characteristics in molecular junctions and promising properties of polyene, with single molecular conductance as a vital tool for bringing insights into the design and construction of nanodevices. Polyene is a segment of polyacetylene, a conductive polymer. Here, the authors measured the conductance of single molecular chain of trans-polyene and found a high conductivity and low decay constant, attributed to the alignment of the energy levels.
