  • Robust estimation of bacterial cell count from optical density

    • Optical density (OD) is widely used to estimate the density of cells in liquid culture, but cannot be compared between instruments without a standardized calibration protocol and is challenging to relate to actual cell count. We address this with an interlaboratory study comparing three simple, low-cost, and highly accessible OD calibration protocols across 244 laboratories, applied to eight strains of constitutive GFP-expressing E. coli. Based on our results, we recommend calibrating OD to estimated cell count using serial dilution of silica microspheres, which produces highly precise calibration (95.5% of residuals <1.2-fold), is easily assessed for quality control, also assesses instrument effective linear range, and can be combined with fluorescence calibration to obtain units of Molecules of Equivalent Fluorescein (MEFL) per cell, allowing direct comparison and data fusion with flow cytometry measurements: in our study, fluorescence per cell measurements showed only a 1.07-fold mean difference between plate reader and flow cytometry data.

  • Logistics-involved service composition in a dynamic cloud manufacturing environment : A DDPG-based approach

    • Service composition as an important technique for combining multiple services to construct a value-added service is a major research issue in cloud manufacturing. Highly dynamic environments present great challenges to cloud manufacturing service composition (CMfg-SC). Most of previous studies employ heuristic algorithms to solve service composition issues in cloud manufacturing, which, however, are designed for specific problems and lack adaptability necessary to dynamic environment. Hence, CMfg-SC calls for new adaptive approaches. Recent advances in deep reinforcement learning (DRL) provide a new means for solving this issue. Based on DRL, we propose a Deep Deterministic Policy Gradient (DDPG)-based service composition approach to cloud manufacturing, with which optimal service composition solutions can be learned through repeated training. Performance of DDPG in solving CMfg-SC in both static and dynamic environments is examined. Results obtained with another DRL algorithm -Deep Q-Networks (DQN) and the traditional Ant Colony Optimization (ACO) are also presented. Comparison indicates that DDPG has better adaptability, robustness, and extensibility to dynamic environments than ACO, although ACO converges faster and its steady QoS value of the service composition solution is higher than that of DDPG by 0.997%. DDPG outperforms DQN in convergence speed and stability, and the QoS value of the service composition solution of DDPG is higher than that of DQN by 3.249%.

    • Background The Global Burden of Diseases, Injuries, and Risk Factors Study (GBD) 2017 comparative risk assessment (CRA) is a comprehensive approach to risk factor quantification that offers a useful tool for synthesising evidence on risks and risk-outcome associations. With each annual GBD study, we update the GBD CRA to incorporate improved methods, new risks and risk-outcome pairs, and new data on risk exposure levels and risk- outcome associations. Methods We used the CRA framework developed for previous iterations of GBD to estimate levels and trends in exposure, attributable deaths, and attributable disability-adjusted life-years (DALYs), by age group, sex, year, and location for 84 behavioural, environmental and occupational, and metabolic risks or groups of risks from 1990 to 2017. This study included 476 risk-outcome pairs that met the GBD study criteria for convincing or probable evidence of causation. We extracted relative risk and exposure estimates from 46 749 randomised controlled trials, cohort studies, household surveys, census data, satellite data, and other sources. We used statistical models to pool data, adjust for bias, and incorporate covariates. Using the counterfactual scenario of theoretical minimum risk exposure level (TMREL), we estimated the portion of deaths and DALYs that could be attributed to a given risk. We explored the relationship between development and risk exposure by modelling the relationship between the Socio-demographic Index (SDI) and risk-weighted exposure prevalence and estimated expected levels of exposure and risk-attributable burden by SDI. Finally, we explored temporal changes in risk-attributable DALYs by decomposing those changes into six main component drivers of change as follows: (1) population growth; (2) changes in population age structures; (3) changes in exposure to environmental and occupational risks; (4) changes in exposure to behavioural risks; (5) changes in exposure to metabolic risks; and (6) changes due to all other factors, approximated as the risk-deleted death and DALY rates, where the risk-deleted rate is the rate that would be observed had we reduced the exposure levels to the TMREL for all risk factors included in GBD 2017.

  • CBD2 : A functional biomarker database for colorectal cancer

    • The rapidly evolving landscape of biomarkers for colorectal cancer (CRC) necessitates an integrative, updated repository. In response, we constructed the Colorectal Cancer Biomarker Database (CBD), which collected and displayed the curated biomedicine information for 870 CRC biomarkers in the previous study. Building on CBD, we have now developed CBD2, which includes information on 1569 newly reported biomarkers derived from different biological sources (DNA, RNA, protein, and others) and clinical applications (diagnosis, treatment, and prognosis). CBD2 also incorporates information on nonbiomarkers that have been identified as unsuitable for use as biomarkers in CRC. A key new feature of CBD2 is its network analysis function, by which users can investigate the visible and topological network between biomarkers and identify their relevant pathways. CBD2 also allows users to query a series of chemicals, drug combinations, or multiple targets, to enable multidrug, multitarget, multipathway analyses, toward facilitating the design of polypharmacological treatments for CRC.   • Evaluation of starvation status in the early developmental stages of black rockfish (Sebastes schlegelii) based on morphological and histological characteristics

    • Assessing the nutritional status and identifying major causes of mortality in larvae experiencing varying degrees of starvation are crucial for establishing appropriate feeding protocols and enhancing the welfare of hatchery-reared fish. The black rockfish Sebastes schlegelii is an important species in aquaculture and stock enhancement efforts in China, Japan, and Korea. This study aimed to identify optimal diagnostic morphometric indicators of starvation in newly hatched (0–6 days post-hatch, DPH) and postlarval stages (27–37 DPH) of this valuable fish species through histological analyses. Our findings revealed that certain morphometric parameters, including body length, the ratios of eye diameter to head height, body height to body length, and abdomen height to body height, exhibit sensitivity to starvation during both larval and postlarval stages. Particularly, the ratios of body height to body length and abdomen height to body height emerged as the most sensitive morphometric indicators of starvation. Histological examinations of the digestive system revealed rapid alterations in the morphology of hepatic parenchymal cells, accompanied by a significant decrease in the number of lipid cells in the liver during episodes of food deprivation. Starvation induced cellular degeneration in the digestive organs, manifested by reduced heights of epithelial cells and mucosal layers in the intestine, oesophagus, and stomach, along with degeneration and separation of muscle fibers. Among these variables, the height of the intestinal submucosa and muscle layer emerged as the most sensitive indicators reflecting nutritional conditions in newly hatched larvae. In contrast, the height of intestinal striated borders and mucosal folds proved to be the most sensitive indicators in the postlarval stage. Furthermore, the height of intestinal epithelial cells and the number of lipid vacuoles in enterocytes exhibited high sensitivity to food deprivation in both newly hatched larvae and postlarvae. These findings underscore the varying resilience of fish to starvation during different developmental phases and highlight the utility of morphological sensitivity characteristics as reliable diagnostic indices for assessing nutritional status in relation to starvation or suboptimal feeding during the early developmental stages of black rockfish in hatchery-reared processes.

  • Synthesis of chiral TiO2 nanofibre with electron transition-based optical activity

    • The optical chirality induced at the absorption bands due to electronic exciton coupling of the transition dipole moments between chromophores in close proximity is ubiquitous in helical organic materials. However, inorganic materials with optical activity resulting from electronic transitions have not been explored. Here we report the synthesis of chiral TiO2 fibres via transcription of the helical structure of amino acid-derived amphiphile fibres through coordination bonding interactions between the organics and the TiO2 source. Upon calcination, the as-prepared amorphous TiO2 double-helical fibres with a pitch length of ~100 nm were converted to double-helical crystalline fibres with stacks of anatase nanocrystals in an epitaxial helical relationship. Both the amorphous and anatase crystalline helical TiO2 fibres exhibited optical response to circularly polarized light at the absorption edge around ~350 nm. This was attributed to the semiconductor TiO2-based electronic transitions from the valence band to the conduction band under an asymmetric electric field.

  • Glycolytic enzyme Enolase-1 regulates insulin gene expression in pancreatic β-cell

    • Enolase-1 (Eno1) plays a critical role in regulating glucose metabolism; however, its specific impact on pancreatic islet β-cells remains elusive. This study aimed to provide a preliminary exploration of Eno1 function in pancreatic islet β-cells. The findings revealed that the expression of ENO1 mRNA in type 2 diabetes donors was significantly increased and positively correlated with HbA1C and negatively correlated with insulin gene expression. A high level of Eno1 in human insulin-secreting rat INS-1832/13 cells with co-localization with intracellular insulin proteins was accordingly observed. Silencing of Eno1 using siRNA or inhibiting Eno1 protein activity with an Eno1 antagonist significantly reduced insulin secretion and insulin content in β-cells, while the proinsulin/insulin content ratio remained unchanged. This reduction in β-cells function was accompanied by a notable decrease in intracellular ATP and mitochondrial cytochrome C levels. Overall, our findings confirm that Eno1 regulates the insulin secretion process, particularly glucose metabolism and ATP production in the β-cells. The mechanism primarily involves its influence on insulin production, suggesting that Eno1 represents a potential target for β-cell protection and diabetes treatment.

    • The key factor restricting the power conversion efficiency (PCE) of organic solar cells (OSCs) is the energy loss (Eloss), which is the difference between the optical bandgap (Eg) of the active layer and open-circuit voltage (VOC) of the device. To achieve lower Eloss, it is necessary to obtain an appropriate donor-acceptor phase separation size to accelerate exciton dissociation and inhibit the recombination process. However, in most high-efficiency non-fullerene systems, acceptors often exhibit excessive aggregation phenomena. The decrease in the interface area leads to a decrease in exciton dissociation efficiency, which increases the energy loss. Herein, we report a ternary strategy to decrease the crystallization time of the acceptor and inhibit the excessive aggregation condition of a non-fullerene acceptor. We chose a donor poly{[4,8-bis[5-(2-ethylhexyl)-4-fluoro-2-thienyl]benzo[1,2-b:4,5-b ']-dithiophene-2,6-diyl]-alt-[2,5-thiophenediyl[5,7-bis(2-ethylhexyl)-4,8-dioxo-4H,8H-benzo[1,2-c:4,5-c ']dithiophene-1,3-diyl]]} (PM6) and a non-fullerene acceptor (2,2 '-((2Z,2 ' Z)-((12,13-bis(2-ethylhexyl)-3,9-diundecyl-12,13-dihydro-[1,2,5]thiadiazolo[3,4-e]thieno[2 '',3 '':4 ',5 ']thieno[2 ',3 ':4,5]pyrrolo[3,2-g]thieno[2 ',3 ':4,5]thieno[3,2-b]indole-2,10-diyl)bis(methanylylidene))bis(5,6-difluoro-3-oxo-2,3-dihydro-1H-indene-2,1-diylidene))dimalononitrile) (Y6) as the model system. Y6 is prone to forming a tightly packed structure due to its planar curved skeleton. To suppress the excessive aggregation, we chose poly[2,2 '-((2Z,2 ' Z)-((12,13-bis(2-octyldodecyl)-3,9-diundecyl-12,13-dihydro-[1,2,5]thiadiazolo[3,4-e]thieno[2 '',3 '':4 ',5 ']thieno[2 ',3 ':4,5]pyrrolo[3,2-g]thieno[2 ',3 ':4,5]thieno[3,2-b]indole-2,10-diyl)bis(methanylylidene))bis(5,6-difluoro-3-oxo-2,3-dihydro-1H-indene-2,1-diylidene))dimalononitrile-co-2,5-thiophene] (PY-IT) as a second acceptor, which has good compatibility with Y6. By using in situ UV-visible absorption spectroscopy to monitor the film formation kinetics of Y6, it was found that after adding 15 wt% PYIT, the total crystallization time of Y6 decreased and the excessive aggregation of Y6 was inhibited. In the PM6:Y6 system, Y6 only had one crystallization and film-forming process. While in the PM6:Y6+15 wt% PYIT system, the process of film formation became more complex, with two stages of aggregation. PYIT crystallized before Y6, when Y6 began to crystallize, PYIT has occupied a portion of the crystallization growth space. What is more, PYIT delayed the crystallization process of Y6, and the change in the acceptor peak position showed a stable region. After that, Y6 began to aggregate and the crystallization time of Y6 was shorter than that of the binary system. As a result, PYIT alleviated the excessive aggregation of Y6, resulting in better mixing between the non-fullerene acceptor and the donor, increasing the interface area and enabling faster dissociation of excitons. In addition, the vertical phase separation of the active layer has also been optimized, allowing more donors enriched near the anode, enhancing the efficiency of charge extraction. The improved morphology of the active layer results in a better interface area, which can not only ensure exciton dissociation and charge generation, but also reduce the transfer time, which is conducive to reducing energy loss. As a result, Eloss reduced from 0.559 eV to 0. 539 eV, and the optimized ternary OSC exhibited a PCE of 17.05%. PYIT was added to the PM6:Y6 system to delay and shorten the crystallization time of Y6. The ternary strategy has been successfully proven to increase the D/A interface area for faster exciton dissociation. The Eloss decreased (0.559 eV to 0.539 eV), and the PCE increased (15.40% to 17.05%).

  • A critical role of the mechanosensor PIEZO1 in glucose-induced insulin secretion in pancreatic beta-cells

    • Glucose-induced insulin secretion depends on beta-cell electrical activity. Inhibition of ATP-regulated potassium (K-ATP) channels is a key event in this process. However, K-ATP channel closure alone is not sufficient to induce beta-cell electrical activity; activation of a depolarizing membrane current is also required. Here we examine the role of the mechanosensor ion channel PIEZO1 in this process. Yoda1, a specific PIEZO1 agonist, activates a small membrane current and thereby triggers beta-cell electrical activity with resultant stimulation of Ca2+-influx and insulin secretion. Conversely, the PIEZO1 antagonist GsMTx4 reduces glucose-induced Ca2+-signaling, electrical activity and insulin secretion. Yet, PIEZO1 expression is elevated in islets from human donors with type-2 diabetes (T2D) and a rodent T2D model (db/db mouse), in which insulin secretion is reduced. This paradox is resolved by our finding that PIEZO1 translocates from the plasmalemma into the nucleus (where it cannot influence the membrane potential of the beta-cell) under experimental conditions emulating T2D (high glucose culture). beta-cell-specific Piezo1-knockout mice show impaired glucose tolerance in vivo and reduced glucose-induced insulin secretion, beta-cell electrical activity and Ca2+ elevation in vitro. These results implicate mechanotransduction and activation of PIEZO1, via intracellular accumulation of glucose metabolites, as an important physiological regulator of insulin secretion. Insulin secretion depends on action potential firing in pancreatic islet beta-cells, but the underlying mechanism is unclear. Here, the authors show that activation of the mechanosensor ion channel PIEZO1 plays a central role in beta-cell electrical activity and insulin release.
