    • Optical density (OD) is widely used to estimate the density of cells in liquid culture, but cannot be compared between instruments without a standardized calibration protocol and is challenging to relate to actual cell count. We address this with an interlaboratory study comparing three simple, low-cost, and highly accessible OD calibration protocols across 244 laboratories, applied to eight strains of constitutive GFP-expressing E. coli. Based on our results, we recommend calibrating OD to estimated cell count using serial dilution of silica microspheres, which produces highly precise calibration (95.5% of residuals <1.2-fold), is easily assessed for quality control, also assesses instrument effective linear range, and can be combined with fluorescence calibration to obtain units of Molecules of Equivalent Fluorescein (MEFL) per cell, allowing direct comparison and data fusion with flow cytometry measurements: in our study, fluorescence per cell measurements showed only a 1.07-fold mean difference between plate reader and flow cytometry data.

  • NDFIP1 limits cellular TAZ accumulation via exosomal sorting to inhibit NSCLC proliferation

    • NDFIP1 has been previously reported as a tumor suppressor in multiple solid tumors, but the function of NDFIP1 in NSCLC and the underlying mechanism are still unknown. Besides, the WW domain containing proteins can be recognized by NDFIP1, resulted in the loading of the target proteins into exosomes. However, whether WW domain-containing transcription regulator 1 (WWTR1, also known as TAZ) can be packaged into exosomes by NDFIP1 and if so, whether the release of this oncogenic protein via exosomes has an effect on tumor development has not been investigated to any extent. Here, we first found that NDFIP1 was low expressed in NSCLC samples and cell lines, which is associated with shorter OS. Then, we confirmed the interaction between TAZ and NDFIP1, and the existence of TAZ in exosomes, which requires NDFIP1. Critically, knockout of NDFIP1 led to TAZ accumulation with no change in its mRNA level and degradation rate. And the cellular TAZ level could be altered by exosome secretion. Furthermore, NDFIP1 inhibited proliferation in vitro and in vivo, and silencing TAZ eliminated the increase of proliferation caused by NDFIP1 knockout. Moreover, TAZ was negatively correlated with NDFIP1 in subcutaneous xenograft model and clinical samples, and the serum exosomal TAZ level was lower in NSCLC patients. In summary, our data uncover a new tumor suppressor, NDFIP1 in NSCLC, and a new exosome-related regulatory mechanism of TAZ.

  • A reference method for measuring emissions of SVOCs in small chambers

    • Semi-volatile organic compounds (SVOCs) are indoor air pollutants that may have significant adverse effects on human health. Although emissions of volatile chemicals from building materials and consumer products are usually characterized in small chambers, few chamber studies have been conducted for SVOCs due to the challenges associated with analysis and the lack of validation procedures. There is an urgent need for a reliable and accurate chamber test method to verify these measurements. A reference method employing a specially-designed chamber has been developed and is undergoing extensive evaluation. A pilot inter-laboratory study (ILS) has been conducted with six laboratories performing chamber tests under identical conditions for di-2-ethylhexyl phthalate (DEHP). Results from this study showed inter-laboratory variations of 24% for DEHP emission rates, with closer agreement observed among intra-laboratory measurements for most of the participating laboratories. A mechanistic emission model fits well to the measured concentration profiles, demonstrating the feasibility of the proposed reference method to independently assess laboratory performance and validate SVOC emission tests.

  • Edge-AI-driven framework with efficient mobile network design for facial expression recognition

    • Facial Expression Recognition (FER) in the wild poses significant challenges due to realistic occlusions, illumination, scale, and head pose variations of the facial images. In this article, we propose an Edge-AI-driven framework for FER. On the algorithms aspect, we propose two attention modules, Arbitrary-oriented Spatial Pooling (ASP) and Scalable Frequency Pooling (SFP), for effective feature extraction to improve classification accuracy. On the systems aspect, we propose an edge-cloud joint inference architecture for FER to achieve low-latency inference, consisting of a lightweight backbone network running on the edge device, and two optional attention modules partially offloaded to the cloud. Performance evaluation demonstrates that our approach achieves a good balance between classification accuracy and inference latency.

  • Tunable metal hydroxide-organic frameworks for catalysing oxygen evolution

    • The oxygen evolution reaction is central to making chemicals and energy carriers using electrons. Combining the great tunability of enzymatic systems with known oxide-based catalysts can create breakthrough opportunities to achieve both high activity and stability. Here we report a series of metal hydroxide–organic frameworks (MHOFs) synthesized by transforming layered hydroxides into two-dimensional sheets crosslinked using aromatic carboxylate linkers. MHOFs act as a tunable catalytic platform for the oxygen evolution reaction, where the π–π interactions between adjacent stacked linkers dictate stability, while the nature of transition metals in the hydroxides modulates catalytic activity. Substituting Ni-based MHOFs with acidic cations or electron-withdrawing linkers enhances oxygen evolution reaction activity by over three orders of magnitude per metal site, with Fe substitution achieving a mass activity of 80 A g−1catalyst at 0.3 V overpotential for 20 h. Density functional theory calculations correlate the enhanced oxygen evolution reaction activity with the MHOF-based modulation of Ni redox and the optimized binding of oxygenated intermediates.
