  • Expression profile, molecular functions, and prognostic significance of miRNAs in primary colorectal cancer stem cells

    • MicroRNAs (miRNAs) are known to drive the pathogenesis of colorectal cancer (CRC) via the regulation of cancer stem cells (CSCs). We studied the miRNA expression profile of primary CSCs isolated from patients with CRC (pCRCSCs). Compared to pCRCSC-derived differentiated cells, 98 differentially expressed miRNAs were identified in pCRCSCs. Target genes encoding pCRCSC-related miRNAs were identified using a combination of miRNA target databases and miRNA-mRNA regulatory networks from the same patient. The pCRCSC-related miRNA target genes were associated with pathways contributing to malignant phenotypes, including I-kappa B kinase/NF-kappa B signaling, signal transduction by p53 class mediator, Ras signaling, and cGMP-PKG signaling. The pCRCSC-related miRNA expression signature was independently associated with poor overall survival in both the training and validation cohorts. We have thus identified several pCRCSC-related miRNAs with oncogenic potential that could serve as prognostic biomarkers for CRC.

    • Empirical relationships for estimating Uniaxial Compressive Strength (UCS) of rock from other rock properties are numerous in literature. This is because the laboratory procedure for determination of UCS from compression tests is cumbersome, time consuming, and often considered expensive, especially for small to medium-sized mining engineering projects. However, these empirical models are scattered in literature, making it difficult to access a considerable number of them when there is need to select empirical model for estimation of UCS. This often leads to bias in estimated UCS data as there may be underestimation or overestimation of UCS, because of the site-specific nature of rock properties. Therefore, this study develops large database of empirical relationships between UCS and other rock properties that are reported in literatures. Statistical analysis was performed on the regression equations in the database developed. The typical ranges and mean of data used in developing the regressions, and the range and mean of their R-2 values were evaluated and summarised. Most of the regression equations were found to be developed from reasonable quantity of data with moderate to high R-2 values. The database can be easily assessed to select appropriate regression equation when there is need to estimate UCS for a specific site.

  • An empirical approach for predicting burden velocities in rock blasting

    • An analytical relation between burden velocity and ratio of burden to blasthole diameter is developed in this paper. This relation is found to be consistent with the measured burden velocities of all 37 full-scale blasts found from published articles. These blasts include single-hole blasts, multi-hole blasts, and simultaneously-initiated blasts with various borehole diameters such as 64 mm, 76 mm, 92 mm, 115 mm, 142 mm and 310 mm. All boreholes were fully charged. The agreement between measured and calculated burden velocities demonstrates that this relation can be used to predict the burden velocity of a wide range of full-scale blast with fully-coupled explosive charge and help to determine a correct delay time between adjacent holes or rows in various full-scale blasts involved in tunnelling (or drifting), surface and underground mining production blasts and underground opening slot blasts. In addition, this theoretical relation is found to agree with the measured burden velocities of 9 laboratory small-scale blasts to a certain extent. To predict the burden velocity of a small-scale blast, a further study or modification to the relation is necessary by using more small-scale blasts in the future.

    • Studies of the effects of loading rates on rock fracture were performed for decades. However, the previous work on static or dynamic rock fracture was mainly limited to a macro-experimental study. The present investigation measures the fractal dimensions of the fracture surfaces of the gabbro specimens fractured at various loading rates covering static and dynamic loading, and explores the relationship between the fractal dimensions and the fracture toughness of the rock.
