  • Associations of Insomnia With Insulin Resistance Traits : A Cross-sectional and Mendelian Randomization Study

    • Context: Insomnia is associated with insulin resistance (IR) in observational studies; however, whether insomnia is causally associated with IR remains unestablished.Objective: This study aims to estimate the causal associations of insomnia with IR and its related traits.Methods: In primary analyses, multivariable regression (MVR) and 1-sample Mendelian randomization (1SMR) analyses were performed to estimate the associations of insomnia with IR (triglyceride-glucose index and triglyceride to high-density lipoprotein cholesterol [TG/HDL-C] ratio) and its related traits (glucose level, TG, and HDL-C) in the UK Biobank. Thereafter, 2-sample MR (2SMR) analyses were used to validate the findings from primary analyses. Finally, the potential mediating effects of IR on the pathway of insomnia giving rise to type 2 diabetes (T2D) were examined using a 2-step MR design.Results: Across the MVR, 1SMR, and their sensitivity analyses, we found consistent evidence suggesting that more frequent insomnia symptoms were significantly associated with higher values of triglyceride-glucose index (MVR, beta = 0.024, P < 2.00E-16; 1SMR, beta = 0.343, P < 2.00E-16), TG/HDL-C ratio (MVR, beta = 0.016, P = 1.75E-13; 1SMR, beta = 0.445, P < 2.00E-16), and TG level (MVR, beta = 0.019 log mg/dL, P < 2.00E-16, 1SMR: beta = 0.289 log mg/dL, P < 2.00E-16) after Bonferroni adjustment. Similar evidence was obtained by using 2SMR, and mediation analysis suggested that about one-quarter (25.21%) of the association between insomnia symptoms and T2D was mediated by IR.Conclusions: This study provides robust evidence supporting that more frequent insomnia symptoms are associated with IR and its related traits across different angles. These findings indicate that insomnia symptoms can be served as a promising target to improve IR and prevent subsequent T2D.

  • Causal associations of short and long sleep durations with 12 cardiovascular diseases : linear and nonlinear Mendelian randomization analyses in UK Biobank

    • Aims Observational studies have suggested strong associations between sleep duration and many cardiovascular diseases (CVDs), but causal inferences have not been confirmed. We aimed to determine the causal associations between genetically predicted sleep duration and 12 CVDs using both linear and nonlinear Mendelian randomization (MR) designs. Methods and results Genetic variants associated with continuous, short (<= 6 h) and long (>= 9 h) sleep durations were used to examine the causal associations with 12 CVDs among 404 044 UK Biobank participants of White British ancestry. Linear MR analyses showed that genetically predicted sleep duration was negatively associated with arterial hypertension, atrial fibrillation, pulmonary embolism, and chronic ischaemic heart disease after correcting for multiple tests (P <0.001). Nonlinear MR analyses demonstrated nonlinearity (L-shaped associations) between genetically predicted sleep duration and four CVDs, including arterial hypertension, chronic ischaemic heart disease, coronary artery disease, and myocardial infarction. Complementary analyses provided confirmative evidence of the adverse effects of genetically predicted short sleep duration on the risks of 5 out of the 12 CVDs, including arterial hypertension, pulmonary embolism, coronary artery disease, myocardial infarction, and chronic ischaemic heart disease (P< 0.001), and suggestive evidence for atrial fibrillation (P < 0.05). However, genetically predicted long sleep duration was not associated with any CVD. Conclusion This study suggests that genetically predicted short sleep duration is a potential causal risk factor of several CVDs, while genetically predicted long steep duration is unlikely to be a causal risk factor for most CVDs.     • Cellulose and chitin are two of the most abundant biopolymers in nature, but they cannot be effectively utilized in industry due to their recalcitrance. This limitation was overcome by the advent of lytic polysaccharide monooxygenases (LPMOs), which promote the disruption of biopolymers through oxidative mechanism and provide a breakthrough in the action of hydrolytic enzymes. In the application of LPMOs to biomass degradation, the key to consistent and effective functioning lies in their stability. The efficient transformation of biomass resources using LPMOs depends on factors that interfere with their stability. This review discussed three aspects that affect LPMO stability: general external factors, structural factors, and factors in the enzyme-substrate reaction. It explains how these factors impact LPMO stability, discusses the resulting effects, and finally presents relevant measures and considerations, including potential resolutions. The review also provides suggestions for the application of LPMOs in polysaccharide degradation.

    • The orbital effect on the anomalous magnetism and evolution of single crystals with low La doping, LaxY1-xVO3 (x = 0, 0.1, and 0.2), has been studied using single-crystal X-ray diffraction, specific heat, mag-netization, and Raman-scattering techniques. It is found that substituting Y3+ by La3+ increases the de-generacy of the yz/zx orbitals and decreases the Jahn-Teller distortion. These weakens the G-type (antiphase ordering along the c axis) orbital ordering phase. Meanwhile, the substituting decreases the magnetism entropy, indicating the shrinking of the t2g and eg orbital hybridization, eventually destabilizing the C-type (in-phase ordering along the c axis) antiferromagnetic ordering phase. In addition, the mechanism for the shrinking of the diamagnetism with increasing x is analyzed. It may attribute to the competition between the antisymmetric Dzyaloshinsky-Moriya interaction and the single-ion anisotropy.

  • Degradation of Zr-based bulk metallic glasses used in load-bearing implants : A tribocorrosion appraisal

    • Owing to the amorphous structure, Bulk Metallic Glasses (BMGs) have been demonstrating attractive properties for potential biomedical applications. In the present work, the degradation mechanisms of Zr-based BMGs with nominal compositions Zr55Cu30Ni5Al10 and Zr65Cu18Ni7Al10 as potential load-bearing implant material were investigated in a tribocorrosion environment. The composition-dependent micro-mechanical and tribological properties of the two BMGs were evaluated prior to the tribocorrosion tests. The sample Zr65-BMG with a higher Zr content exhibited increased plasticity but relatively reduced wear resistance during the ball-on-disc tests. Both BMGs experienced abrasive wear after the dry wear test under the load of 2N. The cross-sectional subsurface structure of the wear track was examined by Focused Ion Beam (FIB). The electrochemical properties of the BMGs in simulated body fluid were evaluated by means of potentiodynamic polarization and X-ray Photoelectron Spectroscopy (XPS). The spontaneous passivation of Zr-based BMGs in Phosphate Buffer Saline solution was mainly attributed to the highly concentrated zirconium cation (Zr4+) in the passive film. The tribocorrosion performance of the BMGs was investigated using a reciprocating tribometer equipped with an electrochemical cell. The more passive nature of the Zr65-BMG had consequently a negative influence on its tribocorrosion resistance, which induced the wear-accelerated corrosion and eventually speeded-up the degradation process. It has been revealed the galvanic coupling was established between the depassivated wear track and the surrounding passive area, which is the main degradation mechanism for the passive Zr65-BMG subjected to the tribocorrosion environment.

    • A method based on hollow fiber supported liquid membrane extraction coupled with a gas chromatograph equipped with flame ionization detector (GC-FID) was developed for the determination of six short-chain fatty acids including acetic acid, propionic acid, i-butyric acid, n-butyric acid, i-valeric acid and n-valeric acid in serum. Hollow fiber supported liquid membrane extraction was employed for preconcentration and clean-up of the samples. The fatty acids were extracted from the acidic donor (diluted serum) into a liquid membrane formed in the wall of the hollow fiber with 10% tri-n-octylphoshphine oxide (TOPO) in di-n-hexyl ether, and then extracted back into a basic acceptor solution filled in the lumen of the hollow fiber. After being acidified with HCl, the acceptor was directly analyzed by GC-FID. The acceptor concentration, donor pH, membrane liquid and extracting time were optimized giving an enrichment factor up to 155 times. The good linearity (r2 > 0.980), reasonable recovery (87.2-121%), and satisfactory intra-assay (8.2-11.5%) and inter-assay (6.1-11.6%) precision illustrated the good performance of the present method. Limits of detection (LOD) ranged from 0.04 to 0.24 μM and limits of quantification (LOQ) varied from 0.13 to 0.80 μM.

    • Materials innovation calls for an integrated framework combining physics-based modelling and data-driven informatics. A dislocation-based constitutive model accounting for both transformation-induced plasticity (TRIP) and twinning-induced plasticity (TWIP) was built to interpret the mechanical characteristics of metastable titanium alloys. Particular attention was placed on quantitatively understanding the composition-sensitive phase stability and its influence on the underlying deformation mechanism. For this purpose, a pseudoelastic force balance incorporating thermodynamics and micromechanics was applied to calculate the energy landscapes of beta -> alpha '' martensitic transformation, {332}< 113 & rang; twinning and dislocation slip. Extensive material data were probed, computed and fed to the model. Our results revealed that TRIP and TWIP may operate simultaneously because of the presence of a noticeably overlapped energy domain, and confirmed {332}< 113 & rang; twinning is an energetically favourable deformation mechanism. The model validation further unveiled that the activation of beta -> alpha '' transition remarkably enhances the strain-hardening and plasticity, even though the dynamically formed alpha '' volume fraction is much less than that of deformation twinning. Our work suggests that the synchronised physical metallurgy and data-driven strategy allows to identify the compositional scenarios for developing high-performance engineering alloys.

    • Short-chain fatty acids (SCFAs) have attracted much attention recently because of their positive physiological effects. In this work, a rapid and reliable gas chromatographic method for determination of eight SCFAs, in colonic and faecal samples from rats and humans has been developed and validated. The methodology involves extraction of the SCFAs in water before a direct injection procedure on a FFAP capillary column. A stock standard solution containing acetic acid, propionic acid, n-butyric acid, i-butyric acid, n-valeric acid, i-valeric acid, n-caproic acid and n-heptanoic acid was prepared and used. A high linearity (r(2) > 0.9990), low quantification limit (2.38-30.14 mu M) and high recovery for most acids were obtained. Acidification of faecal samples was found to be crucial for quantitative determination of the SCFAs, and adjustment of pH to 2-3 was regarded as necessary. Glass wool inserted in the glass liner of the injection port proved effective in preventing the contamination of the column by non-volatiles, and 12% formic acid reduced the ghost peak that appeared gradually after several injections. After validation, the methodology was applied on two faecal samples from rats fed diets containing different amount of dietary fibre and one faecal sample from human fed a normal diet to test the accuracy of the developed method.     • Tribocorrosion performance of medical grade A1S1 316L stainless steel with and without surface modification has been studied during exposure to an electrolyte. Some of the samples investigated were surface modified by plasma nitriding (at both 400 and 550 °C), and others by a special arc physical vapor deposition to secure a thin layer of a diamond-like carbon (DLC) coating on the surface of the samples. XRD and SEM were used to characterize the homogeneity and microstructure of all samples. Potentiodynamic polarization measurement and tribocorrosion tests were performed multiple times on all groups of samples. It was established that surface treated SS 316L exhibited totally different passive behaviors, which consequently lead different tribocorrosion performance in the electrolyte. It was also established that the DLC coating performed as a solid lubricant in the ball-on-plate tests, and the resulting wear track after the tribocorrosion tests was almost negligible. On the other hand, the untreated and Nitriding 400 °C samples experienced wear accelerated corrosion due to their passive behavior. The results suggest that the DLC coated SS 316L have the best performance at the combined wear and corrosion environment, while the untreated and Nitriding 400 °C samples should be avoided to apply in biological tribocorrosion conditions.
