    • Angiotensin-converting enzyme 2 (ACE2) and accessory proteases (TMPRSS2 and CTSL) are needed for severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) cellular entry, and their expression may shed light on viral tropism and impact across the body. We assessed the cell-type-specific expression of ACE2, TMPRSS2 and CTSL across 107 single-cell RNA-sequencing studies from different tissues. ACE2, TMPRSS2 and CTSL are coexpressed in specific subsets of respiratory epithelial cells in the nasal passages, airways and alveoli, and in cells from other organs associated with coronavirus disease 2019 (COVID-19) transmission or pathology. We performed a meta-analysis of 31 lung single-cell RNA-sequencing studies with 1,320,896 cells from 377 nasal, airway and lung parenchyma samples from 228 individuals. This revealed cell-type-specific associations of age, sex and smoking with expression levels of ACE2, TMPRSS2 and CTSL. Expression of entry factors increased with age and in males, including in airway secretory cells and alveolar type 2 cells. Expression programs shared by ACE2(+)TMPRSS2(+) cells in nasal, lung and gut tissues included genes that may mediate viral entry, key immune functions and epithelial-macrophage cross-talk, such as genes involved in the interleukin-6, interleukin-1, tumor necrosis factor and complement pathways. Cell-type-specific expression patterns may contribute to the pathogenesis of COVID-19, and our work highlights putative molecular pathways for therapeutic intervention. An integrated analysis of over 100 single-cell and single-nucleus transcriptomics studies illustrates severe acute respiratory syndrome coronavirus 2 viral entry gene coexpression patterns across different human tissues, and shows association of age, smoking status and sex with viral entry gene expression in respiratory cell populations.

  • Relationship of plasma biomarkers to digital cognitive tests in Alzheimer's disease.

    • A major limitation in Alzheimer's disease (AD) research is the lack of the ability to measure cognitive performance at scale-robustly, remotely, and frequently. Currently, there are no established online digital platforms validated against plasma biomarkers of AD.We used a novel web-based platform that assessed different cognitive functions in AD patients (N=46) and elderly controls (N=53) who were also evaluated for plasma biomarkers (amyloid beta 42/40 ratio, phosphorylated tau ([p-tau]181, glial fibrillary acidic protein, neurofilament light chain). Their cognitive performance was compared to a second, larger group of elderly controls (N=352).Patients with AD were significantly impaired across all digital cognitive tests, with performance correlating with plasma biomarker levels, particularly p-tau181. The combination of p-tau181 and the single best-performing digital test achieved high accuracy in group classification.These findings show how online testing can now be deployed in patients with AD to measure cognitive function effectively and related to blood biomarkers of the disease.This is the first study comparing online digital testing to plasma biomarkers.Alzheimer's disease patients and two independent cohorts of elderly controls were assessed.Cognitive performance correlated with plasma biomarkers, particularly phosphorylated tau (p-tau)181.Glial fibrillary acidic protein and neurofilament light chain, and less so the amyloid beta 42/40 ratio, were also associated with performance.The best cognitive metric performed at par to p-tau181 in group classification.

  • A Combined Multi-Level Perspective and Agent-Based Modeling in Low-Carbon Transition Analysis

    • Low-carbon transitions are long-term complex processes that are driven by multiple factors. To provide a theoretical and practical framework of this process, we argue that the combination of the multi-level perspective (MLP) and agent-based modeling (ABM) enables us to reach a deeper and detailed analysis of low-carbon transitions. As an extensively applied theoretical form, MLP conceptualizes low-carbon transitions as a nonlinear process and allows a system to be analyzed and organized into multiple dimensions (landscape, regime, and niche). However, MLP cannot explain the many details of complex transitions, whereas ABM can estimate the influence of interacting behaviors in a complex system. Therefore, the main advantages of the combined approach for the analysis of low-carbon transition are verified: the MLP can contribute to the overall design of ABM, and ABM can provide a dynamic, continuous, and quantitative description of the MLP. To construct this combination framework, this paper offers a guiding principle that combines the two perspectives under a low-carbon transitional background to create an integrated strategy using three procedures: defining the common concepts, their interaction, and their combination. Through the proposed framework, the goal of this work was to reach a better understanding of social system evolution from the present high-carbon state to a low-carbon state under the pressure of ambitious climate goals, providing specific policy recommendations.

    • The evolution of a society is inextricably linked to technological transition, which is based on both innovation and dissemination of technologies. To protect the vulnerable new generation of technology, government subsidies are one of the most common and effective tools. However, not all subsidy policies can lead to a healthy development of market shares. Subsidy fraud is one of the most problematic issues that can arise under an imperfect system. This paper identifies an interesting subsidy fraud like phenomenon via a validated agent-based model. After analysing the mechanism of the transition of technology in the model, we drive the condition upon which subsidy fraud could occur.

  • Multi-agent simulation of the effects of Japanese electricity market policies on the low-carbon transition

    • Market policies play a crucial role in facilitating the transition to a low-carbon society by restructuring the electricity market and influencing stakeholder behavior. Policymakers are concerned with how to implement these policies in terms of their intensity, combination, and timing. However, existing research lacks effective simulation tools that can accurately capture the impact of market policies on individual decision-making in the electricity sector, which is essential to represent the complex impacts of the policy mix. To address this gap, we present an agent-based model for analyzing the Low Carbon Transition (LCT) in the electricity sector. Using the Japanese electricity sector as a case study, we design various subsidies, incentives, and liberalization policy scenarios to evaluate the role of market policies in facilitating LCT. We observed that, within the Feed-in Premium (FIP) system, above a subsidy threshold of 2 JPY/kWh or 20% of the electricity cost leads to overcompensation, resulting in a stagnation of LCT promotion. To address this stagnation, it is imperative to not only enhance demand-side incentives, such as carbon taxes but also expedite the advancement of the free trade market to prevent market-induced stagnation. A synergistic implementation of these three policies is crucial for the most efficient progression of LCT.
