    • Genetic modifications during development of paediatric groups 3 and 4 medulloblastoma are responsible for their highly metastatic properties and poor patient survival rates. PRUNE1 is highly expressed in metastatic medulloblastoma group 3, which is characterized by TGF-β signalling activation, c-MYC amplification, and OTX2 expression. We describe the process of activation of the PRUNE1 signalling pathway that includes its binding to NME1, TGF-β activation, OTX2 upregulation, SNAIL (SNAI1) upregulation, and PTEN inhibition. The newly identified small molecule pyrimido-pyrimidine derivative AA7.1 enhances PRUNE1 degradation, inhibits this activation network, and augments PTEN expression. Both AA7.1 and a competitive permeable peptide that impairs PRUNE1/NME1 complex formation, impair tumour growth and metastatic dissemination in orthotopic xenograft models with a metastatic medulloblastoma group 3 cell line (D425-Med cells). Using whole exome sequencing technology in metastatic medulloblastoma primary tumour cells, we also define 23 common ‘non-synonymous homozygous’ deleterious gene variants as part of the protein molecular network of relevance for metastatic processes. This PRUNE1/TGF-β/OTX2/PTEN axis, together with the medulloblastoma-driver mutations, is of relevance for future rational and targeted therapies for metastatic medulloblastoma group 3.

    • (101) anatase TiO2 surface in water ambient is an important system for the interaction of biocompatible nanodevices with biological environment. Following the experimental evidence showing that water molecules are mobile at temperature as low as 190 K and tend to form clusters along the [11 (1) over bar]/[1 (11) over bar] surface directions, a complete theoretical characterization of the dynamical properties of the first water layer on the (101) anatase TiO2 surface is presented. A variety of computational techniques have been employed in the context of the transition-state theory in the harmonic regime, ranging from first-principles total energy ground-state calculations, to density functional perturbation theory, minimum energy path search, and kinetic Monte Carlo simulations, to explain the experimental results on water kinetics on the (101) anatase TiO2 surface. We have calculated the migration energy barrier of water molecules, the vibrational prefactor through the phonon density of states, and the hopping rate along two principal directions. Lastly, in a kinetic Monte Carlo context, we have simulated and clarified the dynamical processes that are on the basis of the observed experimental behavior.

    • Dissociative and molecular water adsorption on the anatase (001) surface is studied in the context of state-of-the-art density functional theory in large supercells suited for adsorption studies at various water coverage ratios. At low coverage values below 1/4 ML, water adsorption remains dissociative and a network of hydrogen bonds between the so formed hydroxyl groups favors the formation of a ridge surface structure. The hydroxyl patterned (4 X 4) surface thus undergoes a (2 X 4) reconstruction that causes the relief of the large tensile stress measured in the unreconstructed surface along the direction orthogonal to the ridge. This phenomenology is accompanied by the loss of reactivity of the reconstructed surface with respect to the dissociative water adsorption that becomes molecular above 1/4 ML. We also show that the molecular adsorption on the terrace is weaker than the one on the ridge. The present water reconstruction model is discussed and compared to the well-known ADM model of the reconstructed anatase (001) surface in dry environment.

    • Water on metal oxides interfaces generate a variety of ordered motifs that depend on the structural properties of the exposed solid surfaces. Here we emphasize the importance of considering the thermodynamic state of the surrounding liquid to find the interface structures in real systems. In particular, using ab initio molecular dynamics, we have studied the thermodynamic behavior of the water induced reconstructed (WIR) anatase (0 0 1) surface under full hydration. The long standing issue of the reconstruction symmetry in this facet of the anatase, that is the TiO2 stable phase at the nanoscale, is addressed showing that the stable state for a WIR surface in vacuum and in bulk water are different, the latter depending on the thermodynamic state of the system. Thermally activated surface phase transitions between (2x4) and (2x3) symmetries are lead by the surface relaxation caused by the molecular adsorption and release phenomena at the interface. Our approach enables the validation to aqueous environment of surface-confined water structures derived in vacuum, emphasizing the role of the thermodynamics conditions for characterizing solid-liquid interfaces especially for nano sized systems.
