  • Timing and quality of sleep in a rural Brazilian family-based cohort, the Baependi Heart Study

    • Sleep is modulated by several factors, including sex, age, and chronotype. It has been hypothesised that contemporary urban populations are under pressure towards shorter sleep duration and poorer sleep quality. Baependi is a small town in Brazil that provides a window of opportunity to study the influence of sleep patterns in a highly admixed rural population with a conservative lifestyle. We evaluated sleep characteristics, excessive daytime sleepiness, and chronotype using the Pittsburgh Sleep Quality Index, Epworth Sleepiness Scale and Morningness-Eveningness Questionnaire questionnaires, respectively. The sample consisted of 1,334 subjects from the Baependi Heart study (41.5% male; age: 46.5 ± 16.2 y, range: 18–89 years). Average self-reported sleep duration was 07:07 ± 01:31 (bedtime 22:32 ± 01:27, wake up time: 06:17 ± 01:25 hh:min), sleep quality score was 4.9 + 3.2, chronotype was 63.6 ± 10.8 and daytime sleepiness was 7.4 ± 4.8. Despite a shift towards morningness in the population, chronotype remained associated with reported actual sleep timing. Age and sex modulated the ontogeny of sleep and chronotype, increasing age was associated with earlier sleep time and shorter sleep duration. Women slept longer and later, and reported poorer sleep quality than men (p < 0.0001). This study provides indirect evidence in support of the hypothesis that sleep timing was earlier prior to full urbanisation.

  • Selection for sexual male characters and their effects on other fitness related traits in white leghorn chickens

    • The size and shape of the cockerel comb, ran be regarded as secondary sexual characters or sexual ornaments. Sexual characters are assumed to be costly to express and the expression of a secondary sexual character is suggested to be favourably correlated with the bearer's condition or fitness. The aim of this study was to clarify the effects of selection for male sexual characters, the correlated responses in other male traits and whether selection for sexual characters affects viability. Two selection lines (lines S and H) and a control line was used for 10 generations. Line S was selected for male comb size at 29 weeks of age and from generation six onwards, comb shape (the way the cockerel bear his comb) was added. Line H was selected for high concentration of hyaluronic acid (HA) in the comb at 28 weeks of age. The average number of animals of each sex and;election line was 560 and the randomly mated control line comprised an average of 150 animals of each sex per generation. Traits recorded an analysed were comb size (CS) and body weight (BW) at 29 weeks of age, comb shape (SH) at 32 weeks of age, comb weight (CW) after slaughter and mortality (M). In line S, the genetic and phenotypic trends increased for CS, CW and BW. Both CS and SH are traits involved in the impression of comb size as visualized by females during male choice and by males during male-male competition. With artificial upward selection for the male character CS (line S), CS, CW, BW and M also increased but SH was impaired. When adding SH to the selection criteria in line S, the negative generic trend for SH was changed to positive. As CS approaches its environmental limit, the heritability and genetic progress can be expected to decline. It seems that 10 generations of selection for increased CS is not enough to reach the environmental limit at which CS is expected to stabilize at an optimum size determined by natural selection.

  • Distribution and heritability of diurnal preference (chronotype) in a rural Brazilian family-based cohort, the Baependi study

    • Diurnal preference (chronotype) is a useful instrument for studying circadian biology in humans. It harbours trait-like dimensions relating to circadian period and sleep homeostasis, but also has ontogenetic components (morningness increases with age). We used the Morningness-Eveningness questionnaire (MEQ) in the Baependi study, a family-based cohort study based in a small town in Minas Gerais, Brazil. The population is highly admixed and has a cohesive and conservative lifestyle. 825 individuals (497 female) aged 18–89 years (average ± SD = 46.4 ± 16.3) and belonging to 112 different families participated in this study. The average MEQ score was 63.5 ± 11.2 with a significant (P < 0.0001) linear increase with age. Morningness was significantly (P < 0.0001) higher in the rural (70.2 ± 9.8) than in the municipal zone (62.6 ± 11.1) and was also significantly (P = 0.025) higher in male (64.6 ± 10.9) than in female (62.8 ± 11.2) participants. Thus, in spite of universal access to electricity, the Baependi population was strongly shifted towards morningness, particularly in the rural zone. Heritability of MEQ score was 0.48 when adjusted for sex and age, or 0.38 when adjusted for sex, age and residential zone. The reported MEQ score heritability is more akin to those of previous twin studies than previous family studies.

  • Absence of phosphoglucose isomerase-1 in retinal photoreceptor, pigment epithelium and Muller cells

    • Macroarray analysis was used to compare equal amounts of cDNA from wild-type and rd/rd (retinal degeneration) mice, collected at P90 when photoreceptor degeneration is virtually complete. A stronger signal for the glycolytic enzyme phosphoglucose isomerase (Gpi1) was observed in the rd/rd sample. Extracellularly, Gpi1 may act as a cytokine, independently described as neuroleukin and autocrine motility factor. Retinal Gpi1 expression was investigated by Northern and Western blot analysis and immunohistochemistry. Double-labelling was performed with antibodies against Gpi1 and calbindin-D, glutamine synthetase, RPE65, calretinin and ultraviolet opsin in order to provide positive cell type identification. Northern and Western blots showed double expression levels per microgram of RNA and protein, respectively, in the rd/rd retina compared with wild-type. However, the total amount of Gpi1 protein per retina was indistinguishable. Gpi1 immunoreactivity was found in ganglion, amacrine, horizontal and bipolar cells, but not in rods, cones, pigment epithelium and Muller cells. This distribution explains why the absolute amounts of Gpi1 protein were not appreciably different between wild-type and the rd/rd phenotype, where rods and cones are absent, whilst the relative contribution of Gpi1 to the total protein and RNA pools differed. Some extracellular immunoreactivity was observed in the photoreceptor matrix around cones in freshly fixed tissue only, which could possibly reflect a role as a cytokine. We propose that glycolysis in Gpi1-negative cells proceeds entirely through the pentose phosphate pathway, creating NADPH at the cost of organic carbon. We hypothesize that the unique metabolic needs of photoreceptors justify this trade-off.
