  • SEARCH FOR GAMMA-RAY EMISSION FROM DES DWARF SPHEROIDAL GALAXY CANDIDATES WITH FERMI-LAT DATA

    • Due to their proximity, high dark-matter (DM) content, and apparent absence of non-thermal processes, Milky Way dwarf spheroidal satellite galaxies (dSphs) are excellent targets for the indirect detection of DM. Recently, eight new dSph candidates were discovered using the first year of data from the Dark Energy Survey (DES). We searched for gamma-ray emission coincident with the positions of these new objects in six years of Fermi Large Area Telescope data. We found no significant excesses of gamma-ray emission. Under the assumption that the DES candidates are dSphs with DM halo properties similar to the known dSphs, we computed individual and combined limits on the velocity-averaged DM annihilation cross section for these new targets. If the estimated DM content of these dSph candidates is confirmed, they will constrain the annihilation cross section to lie below the thermal relic cross section for DM particles with masses less than or similar to 20 GeV annihilating via the b (b) over bar or pi(+)pi(-) channels.

    • The ESSENCE survey discovered 213 Type Ia supernovae at redshifts 0.1 < z < 0.81 between 2002 and 2008. We present their R- and I-band photometry, measured from images obtained using the MOSAIC II camera at the CTIO Blanco, along with rapid-response spectroscopy for each object. We use our spectroscopic follow-up observations to determine an accurate, quantitative classification, and precise redshift. Through an extensive calibration program we have improved the precision of the CTIO Blanco natural photometric system. We use several empirical metrics to measure our internal photometric consistency and our absolute calibration of the survey. We assess the effect of various potential sources of systematic bias on our measured fluxes, and estimate the dominant term in the systematic error budget from the photometric calibration on our absolute fluxes is similar to 1%.

    • We describe the implementation and optimization of the ESSENCE supernova survey, which we have undertaken to measure the dark energy equation-of-state parameter, w = P/(rho c(2)). We present a method for optimizing the survey exposure times and cadence to maximize our sensitivity to w for a given fixed amount of telescope time. For our survey on the CTIO 4 m telescope, measuring the luminosity distances and redshifts for supernovae at modest redshifts (z approximate to 0: 5 +/- 0: 2) is optimal for determining w. We describe the data analysis pipeline based on using reliable and robust image subtraction to find supernovae automatically and in nearly real time. Since making cosmological inferences with supernovae relies crucially on accurate measurement of their apparent brightnesses, we describe our efforts to establish a thorough calibration of the CTIO 4 m telescope's natural photometric system. In its first four years, ESSENCE has discovered and spectroscopically confirmed 102 Type Ia supernovae, at redshifts from 0.10 to 0.78, identified through an impartial, effective methodology for spectroscopic classification and redshift determination. We present the resulting light curves for all of the Type Ia supernovae found by ESSENCE and used in our measurement of w, presented in a companion paper by Wood-Vasey and coworkers.

  • Observational constraints on the nature of dark energy : First cosmological results from the ESSENCE supernova survey

    • We present constraints on the dark energy equation-of-state parameter, w = P/(rho c(2)), using 60 SNe Ia fromthe ESSENCE supernova survey. We derive a set of constraints on the nature of the dark energy assuming a flat universe. By including constraints on (Omega(M), w) from baryon acoustic oscillations, we obtain a value for a static equation-of-state parameter w = -1:05(-0.12)(+0: 13) (stat 1 sigma) +/- 0: 13 (sys) and Omega(M) = 0:274(-0.020)(+0:033) (stat 1 sigma) with a bestfit chi(2)/dof of 0.96. These results are consistent with those reported by the Supernova Legacy Survey from the first year of a similar program measuring supernova distances and redshifts. We evaluate sources of systematic error that afflict supernova observations and present Monte Carlo simulations that explore these effects. Currently, the largest systematic with the potential to affect our measurements is the treatment of extinction due to dust in the supernova host galaxies. Combining our set of ESSENCE SNe Ia with the first-results Supernova Legacy Survey SNe Ia, we obtain a joint constraint of w = -1:07(-0: 09)(+0:09) (stat 1 sigma) +/- 0: 13 ( sys), Omega(M) 0:267(-0:028)(+0:028) (stat 1 sigma) with a best-fit chi(2)/dof of 0.91. The current global SN Ia data alone rule out empty (Omega(M) = 0), matter-only Omega(M) = 0: 3, and Omega(M) = 1 universes at > 4.5 sigma. The current SN Ia data are fully consistent with a cosmological constant.

    • This article documents the addition of 142 microsatellite marker loci to the Molecular Ecology Resources database. Loci were developed for the following species: Agriophyllum squarrosum, Amazilia cyanocephala, Batillaria attramentaria, Fungal strain CTeY1 (Ascomycota), Gadopsis marmoratus, Juniperus phoenicea subsp. turbinata, Liriomyza sativae, Lupinus polyphyllus, Metschnikowia reukaufii, Puccinia striiformis and Xylocopa grisescens. These loci were cross-tested on the following species: Amazilia beryllina, Amazilia candida, Amazilia rutila, Amazilia tzacatl, Amazilia violiceps, Amazilia yucatanensis, Campylopterus curvipennis, Cynanthus sordidus, Hylocharis leucotis, Juniperus brevifolia, Juniperus cedrus, Juniperus osteosperma, Juniperus oxycedrus, Juniperus thurifera, Liriomyza bryoniae, Liriomyza chinensis, Liriomyza huidobrensis and Liriomyza trifolii.

  • Cryo-EM structure of the human Kv3.1 channel reveals gating control by the cytoplasmic T1 domain

    • Kv3 channels have distinctive gating kinetics tailored for rapid repolarization in fast-spiking neurons. Malfunction of this process due to genetic variants in the KCNC1 gene causes severe epileptic disorders, yet the structural determinants for the unusual gating properties remain elusive. Here, we present cryo-electron microscopy structures of the human Kv3.1a channel, revealing a unique arrangement of the cytoplasmic tetramerization domain T1 which facilitates interactions with C-terminal axonal targeting motif and key components of the gating machinery. Additional interactions between S1/S2 linker and turret domain strengthen the interface between voltage sensor and pore domain. Supported by molecular dynamics simulations, electrophysiological and mutational analyses, we identify several residues in the S4/S5 linker which influence the gating kinetics and an electrostatic interaction between acidic residues in alpha 6 of T1 and R449 in the pore-flanking S6T helices. These findings provide insights into gating control and disease mechanisms and may guide strategies for the design of pharmaceutical drugs targeting Kv3 channels. Here, Chi et al. report cryo-EM structures of the human Kv3.1a channel, revealing a unique arrangement of the cytoplasmic T1 domain, which allows the interactions with the C-terminal axonal targeting motif and key components of the gating machinery. These findings provide insights into the functional relevance of previously unknown interdomain interactions in Kv3 channels and may guide the design of new pharmaceutical drugs.

    • Background and objectives: Internet gaming disorder (IGD) is associated with health, social, and academic problems but whether these are consequences of the disorder rather than precursors or correlates is unclear. We aimed to evaluate whether IGD in the 1st year of university predicts health, academic and social problems 1 year later, controlling for baseline health, academic and social problems, demographics, and mental health symptoms.Methods: In a prospective cohort study, 1741 university students completed both a baseline online survey in their 1st year and a follow-up survey 1 year later. Log-binomial models examined the strength of prospective associations between baseline predictor variables (IGD, baseline health, academic and social problems, sex, age, and mental health symptoms) and occurrence of health, academic and social problems at follow-up.Results: When extensively adjusted by the corresponding outcome at baseline, any mental disorder symptoms, sex, and age, baseline IGD was associated only with severe school impairment and poor social life (risk ratio [RR] = 1.77; 95% confidence interval [CI] = 1.14-2.75, p = .011; RR = 1.22; 95% CI = 1.07-1.38, p = .002, respectively).Conclusions and scientific significance: University authorities and counselors should consider that incoming 1st-year students that meet criteria for IGD are likely to have increased academic and social impairments during their 1st year for which they may want to intervene. This study adds to the existing literature by longitudinally examining a greater array of negative outcomes of IGD than previously documented.

    • De novo heterozygous variants in KCNC2 encoding the voltage-gated potassium (K+) channel subunit Kv3.2 are a recently described cause of developmental and epileptic encephalopathy (DEE). A de novo variant in KCNC2 c.374G > A (p.Cys125Tyr) was identified via exome sequencing in a patient with DEE. Relative to wild-type Kv3.2, Kv3.2-p.Cys125Tyr induces K+ currents exhibiting a large hyperpolarizing shift in the voltage dependence of activation, accelerated activation, and delayed deactivation consistent with a relative stabilization of the open conformation, along with increased current density. Leveraging the cryogenic electron microscopy (cryo-EM) structure of Kv3.1, molecular dynamic simulations suggest that a strong π-π stacking interaction between the variant Tyr125 and Tyr156 in the α-6 helix of the T1 domain promotes a relative stabilization of the open conformation of the channel, which underlies the observed gain of function. A multicompartment computational model of a Kv3-expressing parvalbumin-positive cerebral cortex fast-spiking γ-aminobutyric acidergic (GABAergic) interneuron (PV-IN) demonstrates how the Kv3.2-Cys125Tyr variant impairs neuronal excitability and dysregulates inhibition in cerebral cortex circuits to explain the resulting epilepsy.

  • The He-Rich Core-Collapse Supernova 2007Y : Observations from X-Ray to Radio Wavelengths

    • A detailed study spanning approximately a year has been conducted on the Type Ib supernova (SN) 2007Y. Imaging was obtained from X-ray to radio wavelengths, and a comprehensive set of multi-band (w2m2w1u'g'r'i'UBVYJHKs ) light curves and optical spectroscopy is presented. A virtually complete bolometric light curve is derived, from which we infer a 56Ni mass of 0.06 M sun. The early spectrum strongly resembles SN 2005bf and exhibits high-velocity features of Ca II and Hα during late epochs the spectrum shows evidence of an ejecta-wind interaction. Nebular emission lines have similar widths and exhibit profiles that indicate a lack of major asymmetry in the ejecta. Late phase spectra are modeled with a non-LTE code, from which we find 56Ni, O, and total-ejecta masses (excluding He) to be 0.06, 0.2, and 0.42 M sun, respectively, below 4500 km s-1. The 56Ni mass confirms results obtained from the bolometric light curve. The oxygen abundance suggests that the progenitor was most likely a ≈3.3 M sun He core star that evolved from a zero-age-main-sequence mass of 10-13 M sun. The explosion energy is determined to be ≈1050 erg, and the mass-loss rate of the progenitor is constrained from X-ray and radio observations to be lsim10-6 M sun yr-1. SN 2007Y is among the least energetic normal Type Ib SNe ever studied.
