    • An evaluation of modelled estimates for C release following early land clearance at the global level based on new model assumptions suggests that earlier studies may have underestimated its magnitude, chiefly because of underestimation of the mid-Holocene global population. Alternative information sources for population and land utilisation support both a greater total CO(2) release and a greater Neolithic contribution. Indeed, we show that the quantity of terrestrial C release due to early farming, even using the most conservative assumptions, greatly exceeds the net terrestrial C release estimated by inverse modelling of ice core data by Elsig et al. (Elsig J, Schmitt J, Leuenberger D, Schneider R, Eyer M, Leuenberger M et al. ( 2009) Stable isotope constraints on Holocene carbon cycle changes from an Antarctic ice core. Nature 461: 507-510), though uncertainty about past global population estimates precludes calculation of a precise value.

  • Safe and just operating spaces for regional social-ecological systems

    • Humanity faces a major global challenge in achieving wellbeing for all, while simultaneously ensuring that the biophysical processes and ecosystem services that underpin wellbeing are exploited within scientifically informed boundaries of sustainability. We propose a framework for defining the safe and just operating space for humanity that integrates social wellbeing into the original planetary boundaries concept (Rockstrom et al., 2009a,b) for application at regional scales. We argue that such a framework can: (1) increase the policy impact of the boundaries concept as most governance takes place at the regional rather than planetary scale; (2) contribute to the understanding and dissemination of complexity thinking throughout governance and policy-making; (3) act as a powerful metaphor and communication tool for regional equity and sustainability. We demonstrate the approach in two rural Chinese localities where we define the safe and just operating space that lies between an environmental ceiling and a social foundation from analysis of time series drawn from monitored and palaeoecological data, and from social survey statistics respectively. Agricultural intensification has led to poverty reduction, though not eradicated it, but at the expense of environmental degradation. Currently, the environmental ceiling is exceeded for degraded water quality at both localities even though the least well-met social standards are for available piped water and sanitation. The conjunction of these social needs and environmental constraints around the issue of water access and quality illustrates the broader value of the safe and just operating space approach for sustainable development.

    • Understanding social-ecological system dynamics is a major research priority for sustainable management of landscapes, ecosystems and resources. But the lack of multi-decadal records represents an important gap in information that hinders the development of the research agenda. Without improved information on the long-term and complex interactions between causal factors and responses, it will be difficult to answer key questions about trends, rates of change, tipping points, safe operating spaces and pre-impact conditions. Where available long-term monitored records are too short or lacking, palaeoenvironmental sciences may provide continuous multi-decadal records for an array of ecosystem states, processes and services. Combining these records with conventional sources of historical information from instrumental monitoring records, official statistics and enumerations, remote sensing, archival documents, cartography and archaeology produces an evolutionary framework for reconstructing integrated regional histories. We demonstrate the integrated approach with published case studies from Australia, China, Europe and North America.

    • Land-use history, soil ersosion, lake trophy and lake-level fluctuations during the last 3000 years werereconstructed through a multidisciplinary palaeolimnological study (pollen, plant macrofossils, diatoms,physical and chemical analysis, magnetic measurements and radiometric methods) of a small eutrophiclake in southern Sweden (Bj~resjOsjOn, Scania). There are striking responses in diatom, chemical,sediment yield and magnetic records to land-use changes documented by pollen analysis or historicalsources, and to lake-level changes identified from sedimentary changes. Our multidisciplinary approachassists interpretation of the processes controlling long-term changes and separation of the effects ofdifferent factors (land-use changes, lake-level fluctuations) on individual biostratigraphical records.Climate has controlled processes in the lake indirectly, through lake-level fluctuations, from the LateBronze Age to the Viking Age (700 BC-AD 800). Since the Viking Age, land-use controlled most of thechanges observed in the lake's development and soil erosion processes. Major changes in lake developmentoccurred during the last 200 years, due to a drastic increase in soil erosion and water eutrophicationduring a period of agricultural modernization.

    • The lake Bjaresjosjon, Southern Scania, Southern Sweden, was studied in the context of the project 'The cultural landscape of the past 6000 years in Southern Sweden'. Pollen, plant macrofossils, diatoms, physical and chemical analysis, magnetic measurements and radiometric methods (Pb-210, C-14) have been used to study palaeoecological changes, i.e. climate, land use, lake trophy and soil erosion during the past 3000 years. This multidisciplinary study shows striking responses of diatom communities, physical and chemical characteristics, sediment yields and magnetic parameters to land-use changes and lake-level fluctuations. Moreover, the latter are closely related to the settlement history at the site, inferred from archaeological records and historical sources. Before 650 AD, the limnological development was affected mainly by lake-level fluctuations, but partly also by human impact (extensive forest clearings and dominant pastoral farming). With the expansion of arable farming (around 650 AD), human impact on the landscape was the major factor influencing soil erosion processes in the catchment and limnological changes in the lake.

    • Cell adhesion molecules play a significant role in the cellular connection of normal cells. The cadherins are believed to act as tumour suppressors, and their altered expression and function have been associated with tumour development.The authors examined the expression of a Ca++ dependent intercellular adhesion molecule, P-cadherin using an immunohistochemical method in 69 surgically resected breast carcinomas.P-cadherin was detected in 30 cases (43.5%, cytoplasmic and/or membrane staining). The expression of P-cadherin was independent of tumour size and lymph node status, but correlated with a high tumour grade (grade III). In contrast, expression of E-cadherin correlated with lower tumour grade (grade I-II). P-cadherin expression was not detected in invasive lobular carcinomas.In general, P-cadherin was expressed at a lower frequency compared to E-cadherin, alpha-, and beta-catenin. These results suggest that an inverse relationship may exist between E- and P-cadherin in relation to grade, and that the expression of P-cadherin may be a marker of aggressiveness.

    • Understanding the transient dynamics of interlinked social-ecological systems (SES) is imperative for assessing sustainability in the Anthropocene. However, how to identify critical transitions in real-world SES remains a formidable challenge. In this study, we present an evolutionary framework to characterize these dynamics over an extended historical timeline. Our approach leverages multidecadal rates of change in socioeconomic data, paleoenvironmental, and cutting-edge sedimentary ancient DNA records from China's Yangtze River Delta, one of the most densely populated and intensively modified landscapes on Earth. Our analysis reveals two significant social-ecological transitions characterized by contrasting interactions and feedback spanning several centuries. Initially, the regional SES exhibited a loosely connected and ecologically sustainable regime. Nevertheless, starting in the 1950s, an increasingly interconnected regime emerged, ultimately resulting in the crossing of tipping points and an unprecedented acceleration in soil erosion, water eutrophication, and ecosystem degradation. Remarkably, the second transition occurring around the 2000s, featured a notable decoupling of socioeconomic development from ecoenvironmental degradation. This decoupling phenomenon signifies a more desirable reconfiguration of the regional SES, furnishing essential insights not only for the Yangtze River Basin but also for regions worldwide grappling with similar sustainability challenges. Our extensive multidecadal empirical investigation underscores the value of coevolutionary approaches in understanding and addressing social-ecological system dynamics.

    • Cities are rapidly increasing in importance as a major factor shaping the Earth system, and therefore, must take corresponding responsibility. With currently over half the world's population, cities are supported by resources originating from primarily rural regions often located around the world far distant from the urban loci of use. The sustainability of a city can no longer be considered in isolation from the sustainability of human and natural resources it uses from proximal or distant regions, or the combined resource use and impacts of cities globally. The world's multiple and complex environmental and social challenges require interconnected solutions and coordinated governance approaches to planetary stewardship. We suggest that a key component of planetary stewardship is a global system of cities that develop sustainable processes and policies in concert with its non-urban areas. The potential for cities to cooperate as a system and with rural connectivity could increase their capacity to effect change and foster stewardship at the planetary scale and also increase their resource security.

    • Streptococcus pneumoniae (the pneumococcus) is a major cause of mortality and morbidity globally, and the leading cause of death in children under 5 years old. The pneumococcal cytolysin pneumolysin (PLY) is a major virulence determinant known to induce pore-dependent pro-inflammatory responses. These inflammatory responses are driven by PLY-host cell membrane cholesterol interactions, but binding to a host cell receptor has not been previously demonstrated. Here, we discovered a receptor for PLY, whereby pro-inflammatory cytokine responses and Toll-like receptor signalling are inhibited following PLY binding to the mannose receptor C type 1 (MRC-1) in human dendritic cells and mouse alveolar macrophages. The cytokine suppressor SOCS1 is also upregulated. Moreover, PLY-MRC-1 interactions mediate pneumococcal internalization into non-lysosomal compartments and polarize naive T cells into an interferon-gamma(low), interleukin-4(high) and FoxP3(+) immunoregulatory phenotype. In mice, PLY-expressing pneumococci colocalize with MRC-1 in alveolar macrophages, induce lower pro-inflammatory cytokine responses and reduce neutrophil infiltration compared with a PLY mutant. In vivo, reduced bacterial loads occur in the airways of MRC-1-deficient mice and in mice in which MRC-1 is inhibited using blocking antibodies. In conclusion, we show that pneumococci use PLY-MRC-1 interactions to downregulate inflammation and enhance bacterial survival in the airways. These findings have important implications for future vaccine design.

    • Many contemporary societal challenges manifest themselves in the domain of human–environment interactions.There is a growing recognition that responses to these challenges formulated within current disciplinary boundaries, in isolationfrom their wider contexts, cannot adequately address them. Here, we outline the need for an integrated, transdisciplinary synthesisthat allows for a holistic approach, and, above all, a much longer time perspective. We outline both the need for and thefundamental characteristics of what we call “integrated history.” This approach promises to yield new understandings of therelationship between the past, present, and possible futures of our integrated human–environment system. We recommend aunique new focus of our historical efforts on the future, rather than the past, concentrated on learning about future possibilitiesfrom history. A growing worldwide community of transdisciplinary scholars is forming around building this Integrated Historyand future of People on Earth (IHOPE). Building integrated models of past human societies and their interactions with theirenvironments yields new insights into those interactions and can help to create a more sustainable and desirable future. Theactivity has become a major focus within the global change community.

    • The 'Anthropocene' concept provides a conceptual framework that encapsulates the current global situation in which society has an ever-greater dominating influence on Earth System functioning. Simulation models used to understand earth system dynamics provide early warning, scenario analysis and evaluation of environmental management and policies. This paper aims to assess the extent to which current models represent the Anthropocene and suggest ways forward. Current models do not fully reflect the typical characteristics of the Anthropocene, such as societal influences and interactions with natural processes, feedbacks and system dynamics, tele-connections, tipping points, thresholds and regime shifts. Based on an analysis of current model representations of Anthropocene dynamics, we identify ways to enhance the role of modeling tools to better help us understand Anthropocene dynamics and address sustainability issues arising from them. To explore sustainable futures ('safe and operating spaces'), social processes and anthropogenic drivers of biophysical processes must be incorporated, to allow for a spectrum of potential impacts and responses at different societal levels. In this context, model development can play a major role in reconciling the different epistemologies of the disciplines that need to collaborate to capture changes in the functioning of socio-ecological systems. Feedbacks between system functioning and underlying endogenous drivers should be represented, rather than assuming the drivers to be exogenous to the modelled system or stationary in time and space. While global scale assessments are important, the global scale dynamics need to be connected to local realities and vice versa. The diversity of stakeholders and potential questions requires a diversification of models, avoiding the convergence towards single models that are able to answer a wide range of questions, but without sufficient specificity. The novel concept of the Anthropocene can help to develop innovative model representations and model architectures that are better suited to assist in designing sustainable solutions targeted at the users of the models and model results.
