    • In 2008 we published the first set of guidelines for standardizing research in autophagy. Since then, research on this topic has continued to accelerate, and many new scientists have entered the field. Our knowledge base and relevant new technologies have also been expanding. Accordingly, it is important to update these guidelines for monitoring autophagy in different organisms. Various reviews have described the range of assays that have been used for this purpose. Nevertheless, there continues to be confusion regarding acceptable methods to measure autophagy, especially in multicellular eukaryotes. A key point that needs to be emphasized is that there is a difference between measurements that monitor the numbers or volume of autophagic elements (e.g., autophagosomes or autolysosomes) at any stage of the autophagic process vs. those that measure flux through the autophagy pathway (i.e., the complete process); thus, a block in macroautophagy that results in autophagosome accumulation needs to be differentiated from stimuli that result in increased autophagic activity, defined as increased autophagy induction coupled with increased delivery to, and degradation within, lysosomes (in most higher eukaryotes and some protists such as Dictyostelium) or the vacuole (in plants and fungi). In other words, it is especially important that investigators new to the field understand that the appearance of more autophagosomes does not necessarily equate with more autophagy. In fact, in many cases, autophagosomes accumulate because of a block in trafficking to lysosomes without a concomitant change in autophagosome biogenesis, whereas an increase in autolysosomes may reflect a reduction in degradative activity. Here, we present a set of guidelines for the selection and interpretation of methods for use by investigators who aim to examine macroautophagy and related processes, as well as for reviewers who need to provide realistic and reasonable critiques of papers that are focused on these processes. These guidelines are not meant to be a formulaic set of rules, because the appropriate assays depend in part on the question being asked and the system being used. In addition, we emphasize that no individual assay is guaranteed to be the most appropriate one in every situation, and we strongly recommend the use of multiple assays to monitor autophagy. In these guidelines, we consider these various methods of assessing autophagy and what information can, or cannot, be obtained from them. Finally, by discussing the merits and limits of particular autophagy assays, we hope to encourage technical innovation in the field.

    • The Visual Object Tracking challenge VOT2018 is the sixth annual tracker benchmarking activity organized by the VOT initiative. Results of over eighty trackers are presented; many are state-of-the-art trackers published at major computer vision conferences or in journals in the recent years. The evaluation included the standard VOT and other popular methodologies for short-term tracking analysis and a “real-time” experiment simulating a situation where a tracker processes images as if provided by a continuously running sensor. A long-term tracking subchallenge has been introduced to the set of standard VOT sub-challenges. The new subchallenge focuses on long-term tracking properties, namely coping with target disappearance and reappearance. A new dataset has been compiled and a performance evaluation methodology that focuses on long-term tracking capabilities has been adopted. The VOT toolkit has been updated to support both standard short-term and the new long-term tracking subchallenges. Performance of the tested trackers typically by far exceeds standard baselines. The source code for most of the trackers is publicly available from the VOT page. The dataset, the evaluation kit and the results are publicly available at the challenge website (http://votchallenge.net).

    • For the first time in serveral years, the El Nino-Southern Oscillation did not dominate regional climate conditions around the globe. A weak La Ni a dissipated to ENSOneutral conditions by spring, and while El Nino appeared to be emerging during summer, this phase never fully developed as sea surface temperatures in the eastern conditions. Nevertheless, other large-scale climate patterns and extreme weather events impacted various regions during the year. A negative phase of the Arctic Oscillation from mid-January to early February contributed to frigid conditions in parts of northern Africa, eastern Europe, and western Asia. A lack of rain during the 2012 wet season led to the worst drought in at least the past three decades for northeastern Brazil. Central North America also experienced one of its most severe droughts on record. The Caribbean observed a very wet dry season and it was the Sahel's wettest rainy season in 50 years. Overall, the 2012 average temperature across global land and ocean surfaces ranked among the 10 warmest years on record. The global land surface temperature alone was also among the 10 warmest on record. In the upper atmosphere, the average stratospheric temperature was record or near-record cold, depending on the dataset. After a 30-year warming trend from 1970 to 1999 for global sea surface temperatures, the period 2000-12 had little further trend. This may be linked to the prevalence of La Ni a-like conditions during the 21st century. Heat content in the upper 700 m of the ocean remained near record high levels in 2012. Net increases from 2011 to 2012 were observed at 700-m to 2000-m depth and even in the abyssal ocean below. Following sharp decreases in to the effects of La Ni a, sea levels rebounded to reach records highs in 2012. The increased hydrological cycle seen in recent years continued, with more evaporation in drier locations and more precipitation in rainy areas. In a pattern that has held since 2004, salty areas of the ocean surfaces and subsurfaces were anomalously salty on average, while fresher areas were anomalously fresh. Global tropical cyclone activity during 2012 was near average, with a total of 84 storms compared with the 1981-2010 average of 89. Similar to 2010 and 2011, the North Atlantic was the only hurricane basin that experienced above-normal activity. In this basin, Sandy brought devastation to Cuba and parts of the eastern North American seaboard. All other basins experienced either near-or below-normal tropical cyclone activity. Only three tropical cyclones reached Category 5 intensity-all in Bopha became the only storm in the historical record to produce winds greater than 130 kt south of 7 N. It was also the costliest storm to affect the Philippines and killed more than 1000 residents. Minimum Arctic sea ice extent in September and Northern Hemisphere snow cover extent in June both reached new record lows. June snow cover extent is now declining at a faster rate (-17.6% per decade) than September sea ice extent (-13.0% per decade). Permafrost temperatures reached record high values in northernmost Alaska. A new melt extent record occurred on 11-12 July on the Greenland ice sheet; 97% of the ice sheet showed some form of melt, four times greater than the average melt for this time of year. The climate in Antarctica was relatively stable overall. The largest maximum sea ice extent since records begain in 1978 was observed in September 2012. In the stratosphere, warm air led to the second smallest ozone hole in the past two decades. Even so, the springtime ozone layer above Antarctica likely will not return to its early 1980s state until about 2060. Following a slight decline associated with the global 2 emissions from fossil fuel combustion and cement production reached a record 9.5 +/- 0.5 Pg C in 2011 and a new record of 9.7 +/- 0.5 Pg C is estimated for 2012. Atmospheric CO2 concentrations increased by 2.1 ppm in 2012, to 392.6 ppm. In spring 2012, 2 concentration exceeded 400 ppm at 7 of the 13 Arctic observation sites. Globally, other greenhouse gases including methane and nitrous oxide also continued to rise in concentration and the combined effect now represents a 32% increase in radiative forcing over a 1990 baseline. Concentrations of most ozone depleting substances continued to fall.

  • Effect of tunnel cross section on gas temperatures and heat fluxes in case of large heat release rate

    • Tests with liquid and solid fuels in model tunnels (1:20) were performed and analysed in order to study the effect of tunnel cross section (width and height) together with ventilation velocity on ceiling gas temperatures and heat fluxes. The model tunnel was 10m long with varying width (0.3m, 0.45m and 0.6m) and height (0.25m and 0.4m). Test results show that the maximum temperature under the ceiling is a weak function of heat release rate (HRR) and ventilation velocity for cases with HRR more than 100MW at full scale. It clearly varies with the tunnel height and is a weak function of the tunnel width. With a lower tunnel height, the ceiling is closer to the base of continuous flame zone and the temperatures become higher. Overall, the gas temperature beneath the ceiling decreases with the increasing tunnel dimensions, and increases with the increasing longitudinal ventilation velocity. The HRR is also an important factor that influences the decay rate of excess gas temperature, and a dimensionless HRR integrating HRR and other two key parameters, tunnel cross-sectional area and distance between fuel centre and tunnel ceiling, was introduced to account for the effect. An equation for the decay rate of excess gas temperature, considering both the tunnel dimensions and HRR, was developed. Moreover, a larger tunnel cross-sectional area will lead to a smaller heat flux.

    • A series of small scale tests was conducted to investigate the influence of sidewall on flame characteristics of heptane pool fires in a channel, considering pool shape (aspect ratio: 1, 2, 4 and 8) and pool orientation relative to sidewall. Distance between fire and sidewall was changed systematically. Both transverse flame development (along the direction of channel width) and longitudinal flame development (along the direction of channel length) were recorded by digital video. Results show that for a fixed fire location, the heat release rate increases with the increasing pool aspect ratio (namely a larger pool perimeter), which indicates more air entrainment and more intense combustion. In wall fire cases, when the long pool rim is perpendicular with channel sidewall, the flame can obtain more air entrainment with a weaker boundary restriction from the sidewall, compared to the case with the long pool rim being parallel with sidewall. Comparison of some previously established correlations based on various experimental conditions with our test results is made. Due to the fact without considering sidewall effect and fuel shape on the air entrainment of fire plume, the classic correlations need to be further improved. Therefore, an integral flame length model considering both sidewall effect and fuel shape is developed, which correlates well with all the data from cases with various pool positions, orientations and aspect ratios. © 2016 The Combustion Institute.   • Assessment of Central Asian heat extremes by statistical downscaling: Validation and future projection for 2015‒2100

    • Increasing heatwaves and extreme temperatures have recently been observed across Central Asia (CA). Accurately assessing and projecting the changing climate extremes at the local (station) scale required for climate risk management are therefore highly important. However, global and regional climate models often fail to represent the statistical distributions of observed daily extreme variables and hence extremes in complex terrain. In this work, we developed a statistical downscaling (SD) model to project summer daily maximum temperature (Tmax) and heatwave indices for 65 meteorological stations in CA toward 2100. The SD model involves first-order autoregression and multiple linear regression using large-scale Tmax and circulation indices (CIs) as predictors, and the model is cross-validated against historical observations. The local Tmax and heatwave indices are then projected for 2015–2100 driven by the output of a global climate model (CNRM-CM6-1) under four Shared Socioeconomic Pathways (SSP126, SSP245, SSP370, and SSP585). The application of the SD model significantly improves forecasting of the probability distribution (10th/90th percentiles) of Tmax at stations, particularly across mountainous regions. The model also captures interannual variability and the long-term trend in Tmax, consistent with synoptic-scale inputs. SD projections demonstrate strong warming trends of summer Tmax in CA toward 2100 with rates between 0.35–0.64 °C per decade based on the SSP245 and SSP370 scenarios. Consequently, heatwave occurrence is projected to rise by 1.0–5.0 and 2.0–7.0 d per decade under the SSP245 and SSP370 scenarios, respectively, by 2100. Duration, intensity, and amplitude of heatwaves rise at greater rates under higher-emission scenarios, particularly in southeastern CA. The proposed SD model serves as a useful tool for assessing local climate extremes, which are needed for regional risk management and policymaking for adaption to climate change.

  • Molecular Functionalization of NiO Nanocatalyst for Enhanced Water Oxidation by Electronic Structure Engineering

    • Tuning the local environment of nanomaterial-based catalysts has emerged as an effective approach to optimize their oxygen evolution reaction (OER) performance, yet the controlled electronic modulation around surface active sites remains a great challenge. Herein, directed electronic modulation of NiO nanoparticles was achieved by simple surface molecular modification with small organic molecules. By adjusting the electronic properties of modifying molecules, the local electronic structure was rationally tailored and a close electronic structure-activity relationship was discovered: the increasing electron-withdrawing modification readily decreased the electron density around surface Ni sites, accelerating the reaction kinetics and improving OER activity, and vice versa. Detailed investigation by operando Raman spectroelectrochemistry revealed that the electron-withdrawing modification facilitates the charge-transfer kinetics, stimulates the catalyst reconstruction, and promotes abundant high-valent gamma-NiOOH reactive species generation. The NiO-C(6)F(5)catalyst, with the optimized electronic environment, exhibited superior performance towards water oxidation. This work provides a well-designed and effective approach for heterogeneous catalyst fabrication under the molecular level.

    • Composite and nanocomposite samarium doped ceria-carbonates powders were prepared by solidstatereaction, citric acid-nitrate combustion and modified nanocomposite approaches and used aselectrolytes for low temperature solid oxide fuel cells. X-ray Diffraction, Scanning Electron Microscope,low-temperature Nitrogen Adsorption/desorption Experiments, Electrochemical ImpedanceSpectroscopy and fuel cell performance test were employed in characterization of these materials.All powders are nano-size particles with slight aggregation and carbonates are amorphous incomposites. Nanocomposite electrolyte exhibits much lower impedance resistance and higher ionicconductivity than those of the other electrolytes at lower temperature. Fuel cell using the electrolyteprepared by modified nanocomposite approach exhibits the best performance in the whole operationtemperature range and achieves a maximum power density of 839 mW cm−2 at 600 C withH2 as fuel. The excellent physical and electrochemical performances of nanocomposite electrolytemake it a promising candidate for low-temperature solid oxide fuel cells.

    • Ammonia production consumes ~2% of the annual worldwide energy supply, therefore strategic alternatives for the energy-intensive ammonia synthesis through the Haber-Bosch process are of great importance to reduce our carbon footprint. Inspired by MoFe-nitrogenase and the energy-efficient and industrially feasible electrocatalytic synthesis of ammonia, we herein establish a catalytic electrode for artificial nitrogen fixation, featuring a carbon fiber cloth fully grafted by boron-doped molybdenum disulfide (B-MoS2/CFC) nanosheets. An excellent ammonia production rate of 44.09 μg h–1 cm–2 is obtained at −0.2 V versus the reversible hydrogen electrode (RHE), whilst maintaining one of the best reported Faradaic efficiency (FE) of 21.72% in acidic aqueous electrolyte (0.1 M HCl). Further applying a more negative potential of −0.25 V renders the best ammonia production rate of 50.51 μg h–1 cm–2. A strong-weak electron polarization (SWEP) pair from the different electron accepting and back-donating capacities of boron and molybdenum (2p shell for boron and 5d shell for molybdenum) is proposed to facilitate greatly the adsorption of non-polar dinitrogen gas via N≡N bond polarization and the first protonation with large driving force. In addition, for the first time a visible light driven photo-electrochemical (PEC) cell for overall production of ammonia, hydrogen and oxygen from water + nitrogen, is demonstrated by coupling a bismuth vanadate BiVO4 photo-anode with the B-MoS2/CFC catalytic cathode.

    • To study the influence of sidewall effect on flame characteristics and burning rate, a series of experiments with heptane pools was conducted. The results showed that as the fires were placed close to the sidewall, the flames inclined to the sidewall due to the restriction on air entrainment, and the burning rate increased on the whole, which could be mainly due to the enhanced radiation from the heated sidewall and ceiling flame. However, regardless of fuel pool shape, the burning rate obtained the peak value when the fire was near the sidewall, rather than attached to the sidewall, resulting from less flame radiation from the vertical flame part to the fuel in the latter case. The ratio of longitudinal ceiling flame length to transverse length tended to decrease with the fire moving close to the sidewall. For cases with the largest length and wall fires, the ratio was nearly 0.5, which could be explained according to the theory of mirror effect. Also, due to the non-monotonous sidewall effect, a higher burning rate did not necessarily lead to a larger ceiling flame length.

    • Model scale fire tests were performed in tunnels with varying tunnel widths and heights in order to study the effect of tunnel cross-section and ventilation velocity on the heat release rate (HRR) for both liquid pool fires and solid fuel fires. The results showed that for well ventilated heptane pool fires, the tunnel width nearly has no influence on the HRR whilst a lower tunnel height clearly increases the HRR. For well ventilated solid fuel fires, the HRR increases by approximately 25% relative to a free burn test but the HRR is not sensitive to either tunnel width, tunnel height or ventilation velocity. For solid fuel fires that were not well ventilated, the HRRs could be less than those in free burn laboratory tests. In the case of ventilation controlled fires the HRRs approximately lie at the same level as for cases with natural ventilation.

    • The Three-River Headwaters (TRH) region in the Tibetan Plateau is vulnerable to climate change; changes in summer (June–August) precipitation have a significant impact on water security and sustainability in both local and downstream areas. However, the changes in summer precipitation of different intensities over the TRH region, along with their influencing factors, remain unclear. In this study, we used observational and ERA5 reanalysis data and employed a precipitation categorization and water vapor budget analysis to quantify the categorized precipitation variations and investigate their possible linkages with the water vapor budget. Our results showed an increasing trend in summer precipitation at a rate of 0.9 mm per year (p < 0.1) during 1979–2020, with a significant dry-to-wet transition in 2002. The category ‘very heavy precipitation’ (≥10 mm d−1) contributed 65.1% of the increased summer precipitation, which occurred frequently in the northern TRH region. The dry-to-wet transition was caused by the effects of varied atmospheric circulations in each subregion. Southwesterly water vapor transport through the southern boundary was responsible for the increased net water vapor flux in the western TRH region (158.2%), while southeasterly water vapor transport through the eastern boundary was responsible for the increased net water vapor flux in the central TRH (155.2%) and eastern TRH (229.2%) regions. Therefore, we inferred that the dry-to-wet transition of summer precipitation and the increased ‘very heavy precipitation’ over the TRH was caused by increased easterly and southerly water vapor transport.

    • Background: Virtually all patients with heart failure experience multiple symptoms simultaneously, yet clinicians and researchers usually consider symptoms in isolation. Recognizing and responding early to escalating symptoms is essential to preventing hospitalizations in heart failure, yet patients have considerable difficulty recognizing symptoms. Identification of symptom clusters could improve symptom recognition, but cultural differences may be present that must be considered. Objectives: To identify and compare symptom clusters in heart failure patients from the United States, Europe and Asia. Design: Cross-sectional, observational study. Settings: In- and out-patient settings in three regions of the world: Asia (i.e., China and Taiwan); Europe (i.e., the Netherlands and Sweden); and the United States. Participants: A total of 720 patients with confirmed heart failure. Propensity scoring using New York Heart Association Classification was used to match participants from each of the three regions. Methods: Symptoms were identified using the Minnesota Living with Heart Failure Questionnaire. To identify symptom clusters we used cluster analysis with the hierarchical cluster agglomerative approach. We used the Euclidean distance to measure the similarity of variables. Proximity between groups of variables was measured using Wards method. The resulting clusters were displayed with dendrograms, which show the proximity of variables to each other on the basis of semi-partial R-squared scores. Results: There was a core group of symptoms that formed two comparable clusters across the countries. Dyspnea, difficulty in walking or climbing, fatigue/increased need to rest, and fatigue/low energy were grouped into a cluster, which was labeled as a physical capacity symptom cluster. Worrying, feeling depressed, and cognitive problems were grouped into a cluster, which was labeled as an emotional/cognitive symptom cluster. The symptoms of edema and trouble sleeping were variable among the countries and fell into different clusters. Conclusion: Despite the diversity in cultures studied, we found that symptoms clustered similarly among the cultural groups. Identification of similar symptoms clusters among patients with heart failure may improve symptom recognition in both patients and healthcare providers.

  • Vectorized dataset of roadside noise barriers in China using street view imagery

    • Roadside noise barriers (RNBs) are important urban infrastructures to ensure that cities remain liveable. However, the absence of accurate and large-scale geospatial data on RNBs has impeded the increasing progress of rational urban planning, sustainable cities, and healthy environments. To address this problem, this study creates a vectorized RNB dataset in China using street view imagery and a geospatial artificial intelligence framework. First, intensive sampling is performed on the road network of each city based on OpenStreetMap, which is used as the georeference for downloading 6 x 10(6) Baidu Street View (BSV) images. Furthermore, considering the prior geographic knowledge contained in street view images, convolutional neural networks incorporating image context information (IC-CNNs) based on an ensemble learning strategy are developed to detect RNBs from the BSV images. The RNB dataset presented by polylines is generated based on the identified RNB locations, with a total length of 2667.02 km in 222 cities. Last, the quality of the RNB dataset is evaluated from two perspectives, i.e., the detection accuracy and the completeness and positional accuracy. Specifically, based on a set of randomly selected samples containing 10 000 BSV images, four quantitative metrics are calculated, with an overall accuracy of 98.61 %, recall of 87.14 %, precision of 76.44 %, and F-1 score of 81.44 %. A total length of 254.45 km of roads in different cities are manually surveyed using BSV images to evaluate the mileage deviation and overlap level between the generated and surveyed RNBs. The root mean squared error for the mileage deviation is 0.08 km, and the intersection over union for overlay level is 88.08% +/- 2.95 %. The evaluation results suggest that the generated RNB dataset is of high quality and can be applied as an accurate and reliable dataset for a variety of large-scale urban studies, such as estimating the regional solar photovoltaic potential, developing 3D urban models, and designing rational urban layouts. Besides that, the benchmark dataset of the labeled BSV images can also support more work on RNB detection, such as developing more advanced deep learning algorithms, fine-tuning the existing computer vision models, and analyzing geospatial scenes in BSV.     • Assembly of the functional complexes of the mitochondrial respiratory chain requires sophisticated and efficient regulatory mechanisms. In plants, the subunit composition and assembly factors involved in the biogenesis of cytochrome c oxidase (complex IV) are substantially less defined than in mammals and yeast. In this study, we cloned maize (Zea mays) Small kernel 11 (Smk11) via map-based cloning. Smk11 encodes a mitochondria-localized tetratricopeptide repeat protein. Disruption of Smk11 severely affected the assembly and activity of mitochondrial complex IV, leading to delayed plant growth and seed development. Protein interactions studies revealed that SMK11 might interact with four putative complex IV assembly factors, Inner membrane peptidase 1A (ZmIMP1A), MYB domain protein 3R3 (ZmMYB3R-3), cytochrome c oxidase 23 (ZmCOX23), and mitochondrial ferredoxin 1 (ZmMFDX1), among which ZmMFDX1 might interact with subunits ZmCOX6a and ZmCOX-X1; ZmMYB3R-3 might also interact with ZmCOX6a. The mutation of SMK11 perturbed the normal assembly of these subunits, leading to the inactivation of complex IV. The results of this study revealed that SMK11 serves as an accessory assembly factor required for the normal assembly of subunits into complex IV, which will accelerate the elucidation of the assembly of complex IV in plant mitochondria.

  • Quantifying the photovoltaic potential of highways in China

    • Installing photovoltaic (PV) modules on highways is considered a promising way to support carbon neutrality in China. However, collecting the area of the highway, and precisely assessing the shadow area of the highway under complex terrain remain challenges. That severely hinders the assessment of highway PV potential. To address these challenges, a spatiotemporal model is developed in this study to estimate the annual solar PV potential on highways over the whole Chinese territory. First, the areas of different highway segments are calculated based on highway network and highway toll stations. Second, hourly shadow area on highways created by nearby terrain is estimated based on a digital elevation model (DEM). When calculating the highway PV potential, the solar irradiation received in these shadow areas is regarded as zero. Finally, the PV potential of all lanes and emergency lanes was estimated at the prefecture-level city scale using surface radiation data and radiation assessment models. Based on the highway data with a total mileage of 143,684 km at the end of 2020, the results show that the annual PV potential is 3,932 TW and that the corresponding installed capacity is 700.85 GW, which can generate clean electricity at a rate of up to 629.06 TWh. The annual PV potential of highways in the southeast is greater than that in the northwest owing to the higher highway density in the southeast. This study provides a reference basis for highway PV construction planning and suitably assessment in each region of China for PV highway development.

  • JL-GFDN : A Novel Gabor Filter-Based Deep Network Using Joint Spectral-Spatial Local Binary Pattern for Hyperspectral Image Classification

    • The traditional local binary pattern (LBP, hereinafter we also call it a two-dimensional local binary pattern 2D-LBP) is unable to depict the spectral characteristics of a hyperspectral image (HSI). To cure this deficiency, this paper develops a joint spectral-spatial 2D-LBP feature (J2D-LBP) by averaging three different 2D-LBP features in a three-dimensional hyperspectral data cube. Subsequently, J2D-LBP is added into the Gabor filter-based deep network (GFDN), and then a novel classification method JL-GFDN is proposed. Different from the original GFDN framework, JL-GFDN further fuses the spectral and spatial features together for HSI classification. Three real data sets are adopted to evaluate the effectiveness of JL-GFDN, and the experimental results verify that (i) JL-GFDN has a better classification accuracy than the original GFDN; (ii) J2D-LBP is more effective in HSI classification in comparison with the traditional 2D-LBP.
