    • The Miocene epoch, spanning 23.03-5.33 Ma, was a dynamic climate of sustained, polar amplified warmth. Miocene atmospheric CO2 concentrations are typically reconstructed between 300 and 600 ppm and were potentially higher during the Miocene Climatic Optimum (16.75-14.5 Ma). With surface temperature reconstructions pointing to substantial midlatitude and polar warmth, it is unclear what processes maintained the much weaker-than-modern equator-to-pole temperature difference. Here, we synthesize several Miocene climate modeling efforts together with available terrestrial and ocean surface temperature reconstructions. We evaluate the range of model-data agreement, highlight robust mechanisms operating across Miocene modeling efforts and regions where differences across experiments result in a large spread in warming responses. Prescribed CO2 is the primary factor controlling global warming across the ensemble. On average, elements other than CO2, such as Miocene paleogeography and ice sheets, raise global mean temperature by similar to 2 degrees C, with the spread in warming under a given CO2 concentration (due to a combination of the spread in imposed boundary conditions and climate feedback strengths) equivalent to similar to 1.2 times a CO2 doubling. This study uses an ensemble of opportunity: models, boundary conditions, and reference data sets represent the state-of-art for the Miocene, but are inhomogeneous and not ideal for a formal intermodel comparison effort. Acknowledging this caveat, this study is nevertheless the first Miocene multi-model, multi-proxy comparison attempted so far. This study serves to take stock of the current progress toward simulating Miocene warmth while isolating remaining challenges that may be well served by community-led efforts to coordinate modeling and data activities within a common analytical framework.

    • The Miocene (23.03-5.33 Ma) is recognized as a period with close to modern-day paleogeography, yet a much warmer climate. With large uncertainties in future hydroclimate projections, Miocene conditions illustrate a potential future analog for the Earth system. A recent opportunistic Miocene Model Intercomparison Project 1 (MioMIP1) focused on synthesizing published Miocene climate simulations and comparing them with available temperature reconstructions. Here, we build on this effort by analyzing the hydrological cycle response to Miocene forcings across early-to-middle (E2MMIO; 20.03-11.6 Ma) and middle-to-late Miocene (M2LMIO; 11.5-5.33 Ma) simulations with CO2 concentrations ranging from 200 to 850 ppm and providing a model-data comparison against available precipitation reconstructions. We find global precipitation increases by similar to 2.1 and 2.3% per degree of warming for E2MMIO and M2LMIO simulations, respectively. Models generally agree on a wetter than modern-day tropics; mid and high-latitude, however, do not agree on the sign of subtropical precipitation changes with warming. Global monsoon analysis suggests most monsoon regions, except the North American Monsoon, experience higher precipitation rates under warmer conditions. Model-data comparison shows that mean annual precipitation is underestimated by the models regardless of CO2 concentration, particularly in the mid- to high-latitudes. This suggests that the models may not be (a) resolving key processes driving the hydrological cycle response to Miocene boundary conditions and/or (b) other boundary conditions or processes not considered here are critical to reproducing Miocene hydroclimate. This study highlights the challenges in modeling and reconstructing the Miocene hydrological cycle and serves as a baseline for future coordinated MioMIP efforts. This study looks at Earth's hydrological cycle during the Miocene (23-5 million years ago). During this period, the Earth's climate was 3-7 degrees C warmer than today, with carbon dioxide (CO2) estimates ranging between 400 and 850 ppm. Understanding how the hydrological cycle responded during warmer climate conditions can give us insight into what might happen as the Earth gets warmer. We analyzed a suite of Miocene paleoclimate simulations with different CO2 concentrations in the atmosphere and compared them against fossil plant data, which gives an estimate of the average annual rainfall during the period. We found that during the Miocene global rainfall increased by about 2.1%-2.3% for each degree of warming. The models agree that the tropics, mid- and high-latitude, became wetter than they are today but have lower agreement on whether subtropical areas got wetter or drier as they warmed. Compared to proxies, models consistently underestimated how much rain fell in a year, especially in the mid- to high-latitude. This illustrates the challenges in reconstructing the Miocene's hydrological cycle and suggests that the models might not fully capture the range of uncertainties associated with changes in the hydrological cycle due to warming or other factors that differentiated the Miocene.     • Schizophrenia is a highly heritable disorder with a heterogeneous symptomatology. Research increasingly indicates the importance of the crucial and often overlooked glial perturbations within schizophrenia. Within this study, we examined an isoform of quaking (a gene encoding an RNA-binding protein that is exclusively expressed in glial cells), known as QKI6B, and a prototypical astrocyte marker, glial fibrillary acidic protein (GFAP), postulated to be under the regulation of QKI. The expression levels of these genes were quantified across post-mortem brain samples from 55 schizophrenic individuals, and 55 healthy controls, using real-time PCR. We report, through an analysis of covariance (ANCOVA) model, an upregulation of both QKI6B, and GFAP in the prefrontal cortex of brain samples of schizophrenic individuals, as compared to control samples. Previous research has suggested that the QKI protein directly regulates the expression of several genes through interaction with a motif in the target's sequence, termed the Quaking Response Element (QRE). We therefore examined if QICI6B expression can predict the outcome of GFAP, and several oligodendrocyte-related genes, using a multiple linear regression approach. We found that QKI6B significantly predicts the expression of GFAP, but does not predict oligodendrocyte-related gene outcome, as previously seen with other QKI isoforms.

  • Novel developments in photonic sintering of inkjet printed functional inks

    • Inkjet printing of electrical tracks in roll-to-roll applications was hampered for a long time since nano-particle inks required thermal sintering at temperatures greater than 120 °C for several minutes. Among a large number of potential R2R compatible techniques, photonic sintering of inkjet-printed metal-based inks was shown to enable very fast sintering times and providing high quality of structural integrity and low electrical resistance [1]. While the above investigations were carried out with a low dutylow frequency irradiation source, novel developments allow for pulse shaping on the timescale of several microseconds and, therefore, the combination of drying and sintering pulses into a single piece of equipment. In this contribution the photonic sintering process was investigated numerically and experimentally for the case of inkjetprinted aqueous copper oxide ink and a Pulse Forge®3200 X2 tool, both implemented onto a NovaCentrix roll-to-roll machine. Our finding support the assumption, that pulse shaping and, therefore, energy tailoring as a function of time, is essential for efficient conversion of wet copper oxide deposits into conductive copper with no impact on the underlying substrate. The paper presents and discusses the resulting electrical resistances of features processed with a conventional hybrid solution using IRradiation for pre-drying as well as a single step drying and sintering using a single radiation source.

  • Subcutaneous daratumumab in patients with relapsed or refractory multiple myeloma: Part 2 of the open-label, multicenter, dose-escalation phase 1b study (PAVO)

    • Intravenous daratumumab is approved for the treatment of multiple myeloma. In Part 1 of the PAVO study, a mix-and-deliver subcutaneous formulation of daratumumab with recombinant human hyaluronidase PH20 (rHuPH20) was well tolerated, with low rates of infusion-related reactions and similar efficacy to intravenous daratumumab. Part 2 of PAVO evaluated a concentrated, pre-mixed co-formulation of daratumumab and rHuPH20 (DARA SC). Patients with ≥2 prior lines of therapy, including a proteasome inhibitor and immunomodulatory drug, received daratumumab (1800 mg) and rHuPH20 (30,000 U) in 15 mL subcutaneously over 3-5 minutes per the approved intravenous monotherapy dosing schedule. Primary endpoints were daratumumab trough concentration at the end of weekly dosing (just prior to the Cycle 3 Day 1 dose) and safety. Twenty-five patients were enrolled in PAVO Part 2. DARA SC achieved daratumumab trough concentrations similar to or greater than intravenous daratumumab 16 mg/kg. The adverse event profile of DARA SC was consistent with intravenous daratumumab, with no new safety concerns and a lower infusion-related reaction rate. At a median follow-up of 14.2 months, the overall response rate was 52%, median duration of response was 15.7 months, and median progression-free survival was 12.0 months. DARA SC 1800 mg was well tolerated in relapsed/refractory multiple myeloma, with a low infusion-related reaction rate and reduced administration time. Daratumumab serum concentrations following DARA SC were consistent with intravenous dosing, and deep and durable responses were observed. Based on these results, ongoing studies are investigating DARA SC in multiple myeloma and other conditions. (ClinicalTrials.gov identifier: 02519452).

  • In vitro contractile responses of detrusor to carbachol and neurokinin A, in children with recurrent urinary tract infection or day wetting

    • OBJECTIVE: To investigate whether a history of recurrent urinary tract infection (UTI) and/or the presence of day-wetting/urge symptoms might influence the contractile responses to the cholinergic agonist carbachol or to the sensory neuropeptide neurokinin A (NKA) in the urinary bladder in children. PATIENTS AND METHODS: Small detrusor strips were taken from the margin of the cystotomy incision of the bladder dome during surgery to correct vesico-ureteric reflux (VUR) in 62 children (aged 4 months to 12 years) or for unrelated bladder conditions in five controls (aged 3 months to 13 years). Concentration-response curves to carbachol and NKA were constructed using organ-bath techniques, and results compared for age, sex, weight of the detrusor strip, UTI history or day-wetting syndrome. RESULTS: The contractile responses to NKA were no different for any of the features investigated. The contractile response to carbachol and NKA in detrusor from control and VUR patients was not significantly different. The children with a history of UTI were significantly older than those without. The contractility in response to carbachol was greater in older girls (aged 4-12 years) than younger girls (< 4 years) and than in all boys (< 4 years and 4-12 years; ANOVA P = 0.013). The mean (SEM) maximum contractile response to carbachol in the group of 20 young children (4-30 months) with previous UTI was 3.0 (0.3) g, whereas the maximum response in the age-matched group of 11 without UTI was 1.8 (0.3) g (P = 0.046). There were no significant differences in maximum responses between those with day-wetting and those without (aged > 4 years), although there was a significant difference in pD2 value, at 6.19 (0.13) and 5.58 (0.14), respectively (P = 0.018). CONCLUSION: Carbachol produced a larger contractile response in detrusor from children with a history of UTI than from those without, indicating possible alterations in muscarinic receptor characteristics. An increased sensitivity to muscarinic stimulation in day-wetting children was also suggested, whereas NKA is unlikely to be involved in any of these pathophysiological conditions.
