    • Optical density (OD) is widely used to estimate the density of cells in liquid culture, but cannot be compared between instruments without a standardized calibration protocol and is challenging to relate to actual cell count. We address this with an interlaboratory study comparing three simple, low-cost, and highly accessible OD calibration protocols across 244 laboratories, applied to eight strains of constitutive GFP-expressing E. coli. Based on our results, we recommend calibrating OD to estimated cell count using serial dilution of silica microspheres, which produces highly precise calibration (95.5% of residuals <1.2-fold), is easily assessed for quality control, also assesses instrument effective linear range, and can be combined with fluorescence calibration to obtain units of Molecules of Equivalent Fluorescein (MEFL) per cell, allowing direct comparison and data fusion with flow cytometry measurements: in our study, fluorescence per cell measurements showed only a 1.07-fold mean difference between plate reader and flow cytometry data.

    • Humans rely increasingly on sensors to address grand challenges and to improve quality of life in the era of digitalization and big data. For ubiquitous sensing, flexible sensors are developed to overcome the limitations of conventional rigid counterparts. Despite rapid advancement in bench-side research over the last decade, the market adoption of flexible sensors remains limited. To ease and to expedite their deployment, here, we identify bottlenecks hindering the maturation of flexible sensors and propose promising solutions. We first analyze challenges in achieving satisfactory sensing performance for real-world applications and then summarize issues in compatible sensor-biology interfaces, followed by brief discussions on powering and connecting sensor networks. Issues en route to commercialization and for sustainable growth of the sector are also analyzed, highlighting environmental concerns and emphasizing nontechnical issues such as business, regulatory, and ethical considerations. Additionally, we look at future intelligent flexible sensors. In proposing a comprehensive roadmap, we hope to steer research efforts towards common goals and to guide coordinated development strategies from disparate communities. Through such collaborative efforts, scientific breakthroughs can be made sooner and capitalized for the betterment of humanity.

    • Understanding the fundamental principle of catalytic performance and the mechanism of multimetal-based electrocatalysts is essential for the rational design of advanced renewable energy systems. Here, highly crystalline MMMoB4 (M = Fe, Co) compounds with controllable compositions of multiple active metal atoms and polyacene-type boron networks were synthesized delicately by a one-step high-pressure technique to explore electrocatalytic selectivity and activity. CoFeMoB4 and Co2MoB4 are revealed to be highly active and durable oxygen evolution reaction (OER) electrocatalysts under alkaline conditions. The mutually promotive activation of metals with amorphous clusters and ultra-small grains on the surface are responsible for the enhanced activity of CoFeMoB4. More specifically, Co and Fe coupling in CoFeMoB4 facilitates surface reconstruction into active Co hydroxide and Fe oxyhydroxide, in contrast to Co oxyhydroxide in Co2MoB4 and Fe oxides in Fe2MoB4. Dissolving Mo may provide potential space for adsorbing hydroxyl, and the optimized electronic structure with boron is mainly responsible for the long-term durability. In contrast, Mo atoms are responsible for hydrogen evolution reaction (HER) properties, and the optimized d-band center and density of states at the Fermi level make Co2MoB4 a superior HER catalyst. Our findings provide insight into distinguishing the catalytic pathway of multi-metal borides with improved OER activity and different roles of Mo and Co/Fe in the HER and OER.

  • Realizing Efficient Charge/Energy Transfer and Charge Extraction in Fullerene-Free Organic Photovoltaics via a Versatile Third Component

    • Solution-processed organic photovoltaics (OPVs) based on bulk-heterojunctions have gained significant attention to alleviate the increasing demend of fossil fuel in the past two decades. OPVs combined of a wide bandgap polymer donor and a narrow bandgap nonfullerene acceptor show potential to achieve high performance. However, there are still two reasons to limit the OPVs performance. One, although this combination can expand from the ultraviolet to the near-infrared region, the overall external quantum efficiency of the device suffers low values. The other one is the low open-circuit voltage (V-OC) of devices resulting from the relatively downshifted lowest unoccupied molecular orbital (LUMO) of the narrow bandgap. Herein, the approach to select and incorporate a versatile third component into the active layer is reported. A third component with a bandgap larger than that of the acceptor, and absorption spectra and LUMO levels lying within that of the donor and acceptor, is demonstrated to be effective to conquer these issues. As a result, the power conversion efficiencies (PCEs) are enhanced by the elevated short-circuit current and V-OC; the champion PCEs are 11.1% and 13.1% for PTB7-Th:IEICO-4F based and PBDB-T:Y1 based solar cells, respectively.

    • IMPORTANCE The prevalence of cardiometabolic multimorbidity is increasing.OBJECTIVE To estimate reductions in life expectancy associated with cardiometabolic multimorbidity.DESIGN, SETTING, AND PARTICIPANTS Age-and sex-adjusted mortality rates and hazard ratios (HRs) were calculated using individual participant data from the Emerging Risk Factors Collaboration (689 300 participants; 91 cohorts; years of baseline surveys: 1960-2007; latest mortality follow-up: April 2013; 128 843 deaths). The HRs from the Emerging Risk Factors Collaboration were compared with those from the UK Biobank (499 808 participants; years of baseline surveys: 2006-2010; latest mortality follow-up: November 2013; 7995 deaths). Cumulative survival was estimated by applying calculated age-specific HRs for mortality to contemporary US age-specific death rates. EXPOSURES A history of 2 or more of the following: diabetes mellitus, stroke, myocardial infarction (MI).MAIN OUTCOMES AND MEASURES All-cause mortality and estimated reductions in life expectancy.RESULTS In participants in the Emerging Risk Factors Collaboration without a history of diabetes, stroke, or MI at baseline (reference group), the all-cause mortality rate adjusted to the age of 60 years was 6.8 per 1000 person-years. Mortality rates per 1000 person-years were 15.6 in participants with a history of diabetes, 16.1 in those with stroke, 16.8 in those with MI, 32.0 in those with both diabetes and MI, 32.5 in those with both diabetes and stroke, 32.8 in those with both stroke and MI, and 59.5 in those with diabetes, stroke, and MI. Compared with the reference group, the HRs for all-cause mortality were 1.9 (95% CI, 1.8-2.0) in participants with a history of diabetes, 2.1 (95% CI, 2.0-2.2) in those with stroke, 2.0 (95% CI, 1.9-2.2) in those with MI, 3.7 (95% CI, 3.3-4.1) in those with both diabetes and MI, 3.8 (95% CI, 3.5-4.2) in those with both diabetes and stroke, 3.5 (95% CI, 3.1-4.0) in those with both stroke and MI, and 6.9 (95% CI, 5.7-8.3) in those with diabetes, stroke, and MI. The HRs from the Emerging Risk Factors Collaboration were similar to those from the more recently recruited UK Biobank. The HRs were little changed after further adjustment for markers of established intermediate pathways (eg, levels of lipids and blood pressure) and lifestyle factors (eg, smoking, diet). At the age of 60 years, a history of any 2 of these conditions was associated with 12 years of reduced life expectancy and a history of all 3 of these conditions was associated with 15 years of reduced life expectancy.CONCLUSIONS AND RELEVANCE Mortality associated with a history of diabetes, stroke, or MI was similar for each condition. Because any combination of these conditions was associated with multiplicative mortality risk, life expectancy was substantially lower in people with multimorbidity.

  • A GA-based NZEB-cluster planning and design optimization method for mitigating grid overvoltage risk

    • Net-zero energy buildings (NZEBs) are considered as a promising method to mitigating the energy problems. Due to the intermittent characteristics of renewable energy (e.g., solar energy), NZEBs need to frequently exchange energy with the grid, which imposes severe negative impacts on the grid especially the overvoltage risk. Both planning and design are essential for reducing NZEB connected grid overvoltage, but most existing studies isolated the efforts from planning to design, thereby failing to achieve the best cumulative result. More importantly, existing studies oversimplified overvoltage quantification by using aggregated power interactions to represent overvoltage risk, which cannot consider the complex voltage influences among grid nodes. Due to the isolated efforts and the quantification oversimplification, existing studies can hardly achieve overvoltage risk minimization. Therefore, this study proposes a novel GA (genetic algorithm)-based method in which the key planning and design parameters are optimized sequentially for mitigating the overvoltage risk. Meanwhile, distribution network model has been adopted to precisely quantify the grid overvoltage. The study results show that the proposed method is highly effective in reducing NZEB cluster connected grid overvoltage risk. The proposed method can be used in practice for improving NZEB cluster planning and system design as grid interaction is considered. © 2021 Elsevier Ltd

  • Application of Rainbow Trout CYP1 Gene Expression Patterns in Gill and Liver for Haihe River Bio-monitoring

    • CYP1 subfamily genes in gills and liver of rainbow trout as biomarkers were studied to establish methods for quantitative mRNA expression analysis of these genes and to determine their expression pattern. Fish caged in various waters in the Haihe River (Tianjin) were analyzed. The mRNA expression patterns observed in Machangjian River and estuary site of Haihe River were markedly similar but at different levels, reflecting that those sites shared the similar pollution components but with different local pollution load. CYP1C1 and 1C3 were only induced at Gegu site and estuary site of Haihe River, indicating different types of CYP1 agonists in Machangjian River. Response patterns of multiple CYP1 genes in gills and liver could be applied in the monitoring strategy. The response patterns of CYP1 genes could be used for better understanding the relationship between complex mixtures of pollutants and biological response of organisms in aquatic environments.

    • It is of importance for restaurants to identify their competitors to gain competitiveness. Meanwhile, opinion-rich resources like online reviews sites can be used to understand others opinion toward restaurant services. We thus propose a novel model to extract comparative relations from online reviews, and then construct three types of comparison relation networks, enabling competitiveness analysis for three tasks. The first network help restaurants analyze market structure for their positioning. The second network enables to identify top competitors using competitive index and dissimilarity index. The third network help restaurants identify strengths and weaknesses through aspects-comparison relation mining. Finally, the market environment is illustrated in a visual way according to the three types of networks. Experimental results reveal the effectiveness of the proposed competitiveness analysis using text analytics, which can identify top competitors and evaluate the market environment, as well as help the focal restaurant effectively develop a service improvement strategy.

    • All-inorganic perovskite quantum dots (PeQDs) have aroused great research interest in white light-emitting diodes (WLED) due to their excellent optoelectronic properties, but the poor stability, caused by dynamically binding long-chain capping ligands, hinders their future practical applications. To address this issue, here, we exploit short-chain butyric acid (BA) to replace long-chain oleic acid (OA) as capping ligand of CsPbX3 PeQDs by a hot-injection method. The addition of BA not only makes the morphology of CsPbBr3 PeQDs uniform and improves the crystallinity but also effectively suppresses nonradiative recombination, achieving a near unit photoluminescence quantum yield of 96%. The BA capped CsPbBr3 PeQDs exhibit high stability up to 180 d stored in ambient environment and also significantly improved resistance against polar solvent, ultra-violet lamp irradiation, and heat, which is rationalized by the strong binding capacity and shortened distance of BA to the PeQDs through ab initio calculations. Furthermore, by combining green-emission CsPbBr3 and red-emission CsPbBr0.8I2.2 PeQDs with blue GaN chip, we achieved WLEDs with excellent luminous properties, showing their great potential in practical application of wide-color-gamut display and lighting.

  • Experimental study of the effect of air split ratio on thermoacoustic instability in a centrally-staged swirl burner

    • This study focuses on one of the critical design parameters of lean premixed (LPM) combustors-the air split ratio (ASR) between the pilot and main stages. The effect of the air split ratio on thermoacoustic instability is experimentally studied based on a centrally staged model combustor. It is found that the thermoacoustic instability is suppressed with the ASR higher than 20%. The acoustic modes of ASR lower than 20% are dominated by the main stage mode, while the others are controlled by the pilot stage mode. The time-averaged flame shapes and the flame dynamics are processed and discussed. With the increase in ASR, the pilot flame becomes longer, whereas the main flame becomes shorter. Meanwhile, the pilot and main flames become less separated. Moreover, the flame in the outer shear layer (OSL) is extinguished, which is the key to suppressing the thermoacoustic instability. The flame with lower ASR exhibits large-scale periodic axial motion of flame dynamics. The flow fields of ASR = 15% (unstable) and ASR = 30% (stable) are analyzed and discussed to help reveal the differences in thermoacoustic instability between the two cases. It is found that the primary recirculation zone is smaller with ASR = 30%. Higher vorticity and strain rate distributions found in the OSL are believed to quench the flame there. The present study illustrates that a higher air split ratio improves the thermoacoustic stability, which is helpful in designing LPM combustors.

  • SKA Science Data Challenge 2: analysis and results

    • The Square Kilometre Array Observatory (SKAO) will explore the radio sky to new depths in order to conduct transformational science. SKAO data products made available to astronomers will be correspondingly large and complex, requiring the application of advanced analysis techniques to extract key science findings. To this end, SKAO is conducting a series of Science Data Challenges, each designed to familiarize the scientific community with SKAO data and to drive the development of new analysis techniques. We present the results from Science Data Challenge 2 (SDC2), which invited participants to find and characterize 233 245 neutral hydrogen (H i) sources in a simulated data product representing a 2000 h SKA-Mid spectral line observation from redshifts 0.25-0.5. Through the generous support of eight international supercomputing facilities, participants were able to undertake the Challenge using dedicated computational resources. Alongside the main challenge, 'reproducibility awards' were made in recognition of those pipelines which demonstrated Open Science best practice. The Challenge saw over 100 participants develop a range of new and existing techniques, with results that highlight the strengths of multidisciplinary and collaborative effort. The winning strategy - which combined predictions from two independent machine learning techniques to yield a 20 per cent improvement in overall performance - underscores one of the main Challenge outcomes: that of method complementarity. It is likely that the combination of methods in a so-called ensemble approach will be key to exploiting very large astronomical data sets.

    • From 1985 to 2016, the prevalence of underweight decreased, and that of obesity and severe obesity increased, in most regions, with significant variation in the magnitude of these changes across regions. We investigated how much change in mean body mass index (BMI) explains changes in the prevalence of underweight, obesity, and severe obesity in different regions using data from 2896 population-based studies with 187 million participants. Changes in the prevalence of underweight and total obesity, and to a lesser extent severe obesity, are largely driven by shifts in the distribution of BMI, with smaller contributions from changes in the shape of the distribution. In East and Southeast Asia and sub-Saharan Africa, the underweight tail of the BMI distribution was left behind as the distribution shifted. There is a need for policies that address all forms of malnutrition by making healthy foods accessible and affordable, while restricting unhealthy foods through fiscal and regulatory restrictions.

    • Water-based conductive inks are vital for the sustainable manufacturing and widespread adoption of organic electronic devices. Traditional methods to produce waterborne conductive polymers involve modifying their backbone with hydrophilic side chains or using surfactants to form and stabilize aqueous nanoparticle dispersions. However, these chemical approaches are not always feasible and can lead to poor material/device performance. Here, we demonstrate that ground-state electron transfer (GSET) between donor and acceptor polymers allows the processing of water-insoluble polymers from water. This approach enables macromolecular charge-transfer salts with 10,000x higher electrical conductivities than pristine polymers, low work function, and excellent thermal/solvent stability. These waterborne conductive films have technological implications for realizing high-performance organic solar cells, with efficiency and stability superior to conventional metal oxide electron transport layers, and organic electrochemical neurons with biorealistic firing frequency. Our findings demonstrate that GSET offers a promising avenue to develop water-based conductive inks for various applications in organic electronics. Chemical approaches to improve aqueous dispersions of conjugated polymers are limited by the feasibility of modifying the backbone or lead to poor performance. Here, Liu et al. show that ground-state electron transfer in donor:acceptor blends aids aqueous dispersion, for high conductivity and solubility.

    • Sulfur quantum dots (SQDs), as a novel metal-free fluorescent material, are getting increasingly tremendous attention in metal ion detection, especially for Fe3+, due to the merits of antimicrobial potential, low toxicity, and exciting optoelectronic properties. However, sensing mechanism of SQD based fluorescent probe for Fe3+ is not clear yet, and high sensitivity and large detection range remain a challenge. Here, we report the synthesis of hydrophilic SQDs as a fluorescent nanoprobe for Fe3+ via a fluorescent turn-off mode. We systematically studied the quenching mechanism by ultraviolet–visible absorption spectra, steady-state and time-resolved photoluminescent spectra, and temperature-dependent quenching constants. Results unclearly evidenced the quenching behavior to both inner filter effect and static quenching. Furthermore, the nanoprobe presents a large detection range from 2.5 to 700 μM and a limit of detection low to 53.6 nM, both of which are the record performance to our knowledge. At last, it shows high selectivity toward Fe3+ and presents no ionic strength effect in the range of investigation, which enables surprising results for Fe3+ detection in deionized water with interference ion and real water samples.

  • Optimization of the genotyping-by-sequencing strategy for population genomic analysis in conifers

    • Flexibility and low cost make genotyping-by-sequencing (GBS) an ideal tool for population genomic studies of nonmodel species. However, to utilize the potential of the method fully, many parameters affecting library quality and single nucleotide polymorphism (SNP) discovery require optimization, especially for conifer genomes with a high repetitive DNA content. In this study, we explored strategies for effective GBS analysis in pine species. We constructed GBS libraries using HpaII, PstI and EcoRI-MseI digestions with different multiplexing levels and examined the effect of restriction enzymes on library complexity and the impact of sequencing depth and size selection of restriction fragments on sequence coverage bias. We tested and compared UNEAK, Stacks and GATK pipelines for the GBS data, and then developed a reference-free SNP calling strategy for haploid pine genomes. Our GBS procedure proved to be effective in SNP discovery, producing 7000-11000 and 14751 SNPs within and among three pine species, respectively, from a PstI library. This investigation provides guidance for the design and analysis of GBS experiments, particularly for organisms for which genomic information is lacking.

    • The properties of proton resonances in the p and sd shells are studied with a single-particle potential generated by self-consistent-mean field calculations. Besides, we also propose to calculate the proton-nucleus interaction by a double-folding procedure with the density of the daughter nucleus represented by the result of the same mean-field model. With out raising any adjustable parameter, these calculations provide reasonable estimates for the widths of observed proton resonances in light nuclei. The mean-field-based calculation is particularly useful for the predications of the properties of resonances in light exotic nuclei and the direct proton capture reactions of astrophysics.

    • Background: Persistent inflammation has been proposed to contribute to various stages in the pathogenesis of cardiovascular disease. Interleukin-6 receptor (IL6R) signalling propagates downstream inflammation cascades. To assess whether this pathway is causally relevant to coronary heart disease, we studied a functional genetic variant known to affect IL6R signalling. Methods: In a collaborative meta-analysis, we studied Asp358Ala (rs2228145) in IL6R in relation to a panel of conventional risk factors and inflammation biomarkers in 125 222 participants. We also compared the frequency of Asp358Ala in 51 441 patients with coronary heart disease and in 136 226 controls. To gain insight into possible mechanisms, we assessed Asp358Ala in relation to localised gene expression and to postlipopolysaccharide stimulation of interleukin 6. Findings: The minor allele frequency of Asp358Ala was 39%. Asp358Ala was not associated with lipid concentrations, blood pressure, adiposity, dysglycaemia, or smoking (p value for association per minor allele >= 0.04 for each). By contrast, for every copy of 358Ala inherited, mean concentration of IL6R increased by 34.3% (95% CI 30.4-38.2) and of interleukin 6 by 14.6% (10.7-18.4), and mean concentration of C-reactive protein was reduced by 7.5% (5.9-9.1) and of fibrinogen by 1.0% (0.7-1.3). For every copy of 358Ala inherited, risk of coronary heart disease was reduced by 3.4% (1.8-5.0). Asp358Ala was not related to IL6R mRNA levels or interleukin-6 production in monocytes. Interpretation: Large-scale human genetic and biomarker data are consistent with a causal association between IL6R-related pathways and coronary heart disease.
