    • The aim of the study was to provide a standardisation of self-reported competencies and emotional/behavioural problems among Swedish adolescents, using the Youth Self-Report (YSR). The YSR was completed by 2522 adolescents aged 13-18 years, recruited from secondary and upper secondary schools in different regions in Sweden. The results showed that effects of gender and age were small but significant with girls scoring higher than boys on most problem scales, and 15- to 16-year-olds scoring higher than younger and older adolescents on the problem scales. Small effects were also found for residence as well as for parental SES. The correlations between internalising and externalising problems were 0.51 for boys and 0.49 for girls, whereas the correlation between competence and problem scores was low. We conclude that the individual variation in YSR-scores is much greater than can be attributed to factors such as gender, age, SES, or residential area. Consequently, the YSR has the potential to serve as an instrument for assessing individual adolescents' self-reported competencies and problems in Sweden. Given the almost orthogonal relation between self-reported competencies and problems, the competence scale is surprisingly little used in psychopathology research.

  • Dimers of dipyrrometheneboron difluoride (BODIPY) with light spectroscopic applications in chemistry and biology.

    • A ground-state dimer (denoted D(I)) exhibiting a strong absorption maximum at 477 nm (epsilon = 97 000 M(-1)cm(-1)) can form between adjacent BODIPY groups attached to mutant forms of the protein, plasminogen activator inhibitor type 1 (PAI-1). No fluorescence from excited D(I) was detected. A locally high concentration of BODIPY groups was also achieved by doping lipid phases (micelles, vesicles) with BODIPY-labeled lipids. In addition to an absorption band located at about 480 nm, a new weak absorption band is also observed at ca. 570 nm. Both bands are ascribed to the formation of BODIPY dimers of different conformation (D(I) and D(II)). Contrary to D(I) in PAI-1, the D(II) aggregates absorbing at 570 nm are emitting light observed as a broad band centered at about 630 nm. The integrated absorption band of D(I) is about twice that of the monomer, which is compatible with exciton coupling within a dimer. The Förster radius of electronic energy transfer between a BODIPY excited monomer and the ground-state dimer (D(I)()) is 57 +/- 2 A. A simple model of exciton coupling suggests that in D(I) two BODIPY groups are stacked on top of each other in a sandwich-like configuration with parallel electronic transition dipoles. For D(II) the model suggests that the S(0) --> S(1) transition dipoles are colinear. An explanation for the previously reported (J. Am. Chem. Soc. 1994, 116, 7801) exceptional light spectroscopic properties of BODIPY is also presented. These are ascribed to the extraordinary electric properties of the BODIPY chromophore. First, changes of the permanent electric dipole moment (Delta(mu) approximately -0.05 D) and polarizability (-26 x 10(-40) C m(2) V(-1)) between the ground and the first excited states are small. Second, the S(0) <--> S(1) electronic transition dipole moments are perpendicular to Delta(mu).

    • The use of molecular genetics for introducing fluorescent molecules enables the use of donor-donor energy migration to determine intramolecular distances in a variety of proteins. This approach can be applied to examine the overall molecular dimensions of proteins and to investigate structural changes upon interactions with specific target molecules. In this report, the donor-donor energy migration method is demonstrated by experiments with the latent form of plasminogen activator inhibitor type 1. Based on the known x-ray structure of plasminogen activator inhibitor type 1, three positions forming the corners of a triangle were chosen. Double Cys substitution mutants (V106C-H185C, H185C-M266C, and M266C-V106C) and corresponding single substitution mutants (V106C, H185C, and M266C) were created and labeled with a sulfhydryl specific derivative of BODIPY (=the D molecule). The side lengths of this triangle were obtained from analyses of the experimental data. The analyses account for the local anisotropic order and rotational motions of the D molecules, as well as for the influence of a partial DD-labeling. The distances, as determined from x-ray diffraction, between the C(alpha)-atoms of the positions V106C-H185C, H185C-M266C, and M266C-V106C were 60.9, 30.8, and 55.1 A, respectively. These are in good agreement with the distances of 54 +/- 4, 38 +/- 3, and 55 +/- 3 A, as determined between the BODIPY groups attached via linkers to the same residues. Although the positions of the D-molecules and the C(alpha)-atoms physically cannot coincide, there is a reasonable agreement between the methods.

  • The structure of a serpin–protease complex revealed by intramolecular distance measurements using donor–donor energy migration and mapping of interaction sites

    • Background: The inhibitors that belong to the serpin family are widely distributed regulatory molecules that include most protease inhibitors found in blood. It is generally thought that serpin inhibition involves reactive-centre cleavage, loop insertion and protease translocation, but different models of the serpin–protease complex have been proposed. In the absence of a spatial structure of a serpin–protease complex, a detailed understanding of serpin inhibition and the character of the virtually irreversible complex have remained controversial.Results: We used a recently developed method for making precise distance measurements, based on donor–donor energy migration (DDEM), to accurately triangulate the position of the protease urokinase-type plasminogen activator (uPA) in complex with the serpin plasminogen activator inhibitor type 1 (PAI-1). The distances from residue 344 (P3) in the reactive-centre loop of PAI-1 to residues 185, 266, 313 and 347 (P1′) were determined. Modelling of the complex using this distance information unequivocally placed residue 344 in a position at the distal end from the initial docking site with the reactive-centre loop fully inserted into β sheet A. To validate the model, seven single cysteine substitution mutants of PAI-1 were used to map sites of protease–inhibitor interaction by fluorescence depolarisation measurements of fluorophores attached to these residues and cross-linking using a sulphydryl-specific cross-linker.Conclusions: The data clearly demonstrate that serpin inhibition involves reactive-centre cleavage followed by full-loop insertion whereby the covalently linked protease is translocated from one pole of the inhibitor to the opposite one.

    • Plasminogen activator inhibitor 1 (PAI-1) is the main inhibitor of plasminogen activators and plays an important role in many pathophysiological processes. Like other members of the serpin family, PAI-1 has a reactive center consisting of a mobile loop (RCL) with P1 and P1′ residues acting as a “bait” for cognate protease. In contrast to the other serpins, PAI-1 loses activity by spontaneous conversion to an inactive latent form. This involves full insertion of the RCL into β-sheet A. To search for molecular determinants that could be responsible for conversion of PAI-1 to the latent form, we studied the conformation of the RCL in active PAI-1 in solution. Intramolecular distance measurements by donor–donor energy migration and probe quenching methods reveal that the RCL is located much closer to the core of PAI-1 than has been suggested by the recently resolved X-ray structures of stable PAI-1 mutants. Disulfide bonds can be formed in double-cysteine mutants with substitutions at positions P11 or P13 of the RCL and neighboring residues in β-sheet A. This suggests that the RCL may be preinserted up to residue P13 in active PAI-1, and possibly even to residue P11. We propose that the close proximity of the RCL to the protein core, and the ability of the loop to preinsert into β-sheet A is a possible reason for PAI-1 being able to convert spontaneously to its latent form.

    • The extended Förster theory (EFT) is for the first time applied to the quantitative determination of the intramolecular distances in proteins. It is shown how the EFT (J. Chem. Phys., 1996, 105, 10896) can be adapted to the analyses of fluorescence depolarisation experiments based on the time-correlated single photon counting technique (TCSPC). The protein system studied was the latent form of plasminogen activator inhibitor type I (PAI-1), which was mutated and labelled by the thiol reactive BODIPY® derivative {N-(4,4-difluoro-5,7-dimethyl-4-bora-3a,4a-diaza-s-indacene-3-yl)methyl iodoacetamide}. The energy migration occurs within pairs of photophysically identical donor groups that undergo reorientational motions on the timescales of energy migration and fluorescence relaxation. Unlike all models currently used for analysing fluorescence TCSPC data, the EFT explicitly accounts for the time-dependent reorientations that influence the rate of electronic energy transfer/migration in a complex manner. The complexity is related to the “κ2 problem”, which has been discussed for years. The EFT brings the analyses of DDEM data to the same level of molecular description as in ESR and NMR spectroscopy, i.e. it yields microscopic information about the reorientation correlation times, the order parameters, as well as inter-chromophoric distances.

    • The relations between age, metabolic control, disease adjustment, and psychological factors in boys and girls with recently diagnosed insulin-dependent diabetes mellitus (IDDM) were studied. Older girls had significant higher postremission glycosylated haemoglobin A (Hb AIc) levels (p = 0.008). Girls with more hospitalizations had a lower developmental level (p = 0.05), and had significantly more problems in the behavioural rating (p = 0.05). Boys with more hospitalizations had a more external locus of control (p = 0.01), more difficulties with disease adjustment, more emotional problems, and were also clinically assessed as having more behavioural problems. Boys showing more difficulties in psychological adjustment to the disease also had higher postremission Hb AIc levels (p = 0.02). Although Swedish children with IDDM of short disease duration do not differ from healthy children in important psychological aspects, older girls and a small group of problematic younger boys are at risk of developing metabolic imbalance after a short disease duration.

  • Conduct problems in Russian adolescents : the role of personality and parental rearing.

    • The purpose of the present study was 1) to assess the predictive value of conduct problems prior to the age of 12 for the severity of antisocial behaviour during adolescence, and 2) to investigate the relationships between personality traits/parental rearing and childhood conduct problems/teenage antisocial behaviour. A group of 193 delinquents was assessed by means of the Antisocial Behavior Checklist (ABC), the Retrospective Childhood Problems (RETROPROB), the Temperament and Character Inventory (TCI) and the EMBU questionnaire on parental rearing. The extreme groups of delinquents as defined by childhood conduct problems, differed significantly on the experience of a rejecting father and a self-directed character. Furthermore, some specific predictive patterns for current antisocial behaviour by childhood conduct disorder and both personality dimensions and parental rearing factors emerged. The findings are discussed in the light of the interactive nature of relations between personality and parental rearing in the development of antisocial behaviour among adolescents.

  • Coping styles and psychosomatic problems : are they related?

    • A number of recent studies have shown avoidant coping to be related with diverse types of somatic and mental pathology. In order to investigate the relevance of coping for both psychological and somatic problems in delinquent subjects, 185 boys from juvenile correction and 96 boys from secondary schools in Arkhangelsk, Northern Russia, were assessed by means of the Coping Scale for Children and Youth, the Youth Self-Report and the Giessen Subjective Complaints List - youth form. The results from the delinquent subjects partly confirmed previous findings on the dominant role of behavioural avoidance in the relationships between coping styles and both behavior/emotional problems and somatic complaints. Further implications of the results are discussed.

    • A comparison between 192 male delinquent adolescents and 121 controls from Northern Russia using the Temperament and Character Inventory (TCI) and Own Memories of Parental Rearing (EMBU) questionnaire on perceived parental rearing showed significant differences. The delinquent group had a higher level of Novelty Seeking, Harm Avoidance, and Self-transcendence, and also scored lower on Self-directedness. Delinquents who committed nonviolent crimes (thefts) appeared to have a higher level of Harm Avoidance compared with those who committed violent crimes (hooliganism, robbery, rape, and murder). As concerns perceived parental rearing practices, delinquents experienced more parental rejection and overprotection. Most of the personality dimensions were found to be highly correlated with the level of parental emotional warmth. Furthermore, both temperament traits and maternal rearing practices predicted the development of character dimensions. Findings are discussed in light of the interactive nature of parent-child relationships and of character development.

    • OBJECTIVE: The purpose of this study was to assess the level of post-traumatic stress in juvenile male rape victims and to test for its relationships with perceived parental rearing and personality dimensions.METHOD: Fifteen subjects (mean age of 16 years) were recruited from a correctional camp for juvenile criminals in Arkhangelsk, Russia. They were investigated by means of the "Revised Impact of Event Scale," the "Child Self-Report Post-Traumatic Stress Reaction Index," the "Hospital Anxiety and Depression Scale," the "EMBU" (perceived parental rearing) and the "Temperament and Character Inventory" (TCI).RESULTS: The level of post-traumatic stress, in most cases moderate to high, correlated with the temperament dimensions of harm avoidance and reward dependence. As concerns parental rearing, the total level of post-traumatic stress reaction was negatively correlated with paternal emotional warmth and positively with paternal rejection.CONCLUSIONS: The experience of post-traumatic stress is influenced by temperamental characteristics as well as by perceived parental rearing practices. Assumingly, these factors also play a role in becoming a rape victim, which deserves further investigation.

    • The purpose of this study was to investigate possible relationships between parental rearing practices and problem behaviours in a sample of male delinquent adolescents versus controls. A total of 133 subjects from a juvenile correction centre and 108 matched school-children in the Arkhangelsk region, Russia, were assessed by means of the EMBU questionnaire and Youth Self-Report. Delinquents were more severely treated by parents and had more pronounced problem scores. Furthermore, problem scores were found to be highly correlated with parental rejection and lack of emotional warmth in both delinquents and controls. Parental rearing practices may influence the development of problem behaviours. The implications of these findings with regard to preventive measures are discussed.

  • Psychometric properties of Eating Disorder Inventory for children (EDI-C) in Swedish girls with and without a known eating disorder

    • Objective: To evaluate the Swedish version of the Eating Disorder Inventory (EDI-C) for the international consistency, factor structure and ability to discriminate between adolescent girls with and without a known eating disorder (ED).Methods: Completely answered EDI-C questionnaires were available from 201 girls diagnosed with an ED and from 2073 schoolgirls. The girls were adolescents attending grade 7-12.Reslults: International consistency was 0.70-0.91 for the ED group. Eleven factors with eigenvalue >2.2 explained 56% of the variance and showed satisfying correspondence to the proposed constructs of EDI-C. A discriminant analysis classified 85.6% of the cases correctly. The three most important discriminating subscales were Drive for Thinness, Asceticism and Body Dissatisfaction. All subbscales except Maturity Fears showed significantly higher mean values for the ED group.Conclusions:The results indicate that the psychometric properties of EDI-C care comparable to the properties of EDI-2. EDI-C should be preferred for adolescents since its statements suit this age group.
