    • This initiative examined systematically the extent to which a large set of archival research findings generalizes across contexts. We repeated the key analyses for 29 original strategic management effects in the same context (direct reproduction) as well as in 52 novel time periods and geographies; 45% of the reproductions returned results matching the original reports together with 55% of tests in different spans of years and 40% of tests in novel geographies. Some original findings were associated with multiple new tests. Reproducibility was the best predictor of generalizability-for the findings that proved directly reproducible, 84% emerged in other available time periods and 57% emerged in other geographies. Overall, only limited empirical evidence emerged for context sensitivity. In a forecasting survey, independent scientists were able to anticipate which effects would find support in tests in new samples.

    • When conditions change, unicellular organisms rewire their metabolism to sustain cell maintenance and cellular growth. Such rewiring may be understood as resource re-allocation under cellular constraints. Eukaryal cells contain metabolically active organelles such as mitochondria, competing for cytosolic space and resources, and the nature of the relevant cellular constraints remain to be determined for such cells. Here, we present a comprehensive metabolic model of the yeast cell, based on its full metabolic reaction network extended with protein synthesis and degradation reactions. The model predicts metabolic fluxes and corresponding protein expression by constraining compartment-specific protein pools and maximising growth rate. Comparing model predictions with quantitative experimental data suggests that under glucose limitation, a mitochondrial constraint limits growth at the onset of ethanol formation-known as the Crabtree effect. Under sugar excess, however, a constraint on total cytosolic volume dictates overflow metabolism. Our comprehensive model thus identifies condition-dependent and compartment-specific constraints that can explain metabolic strategies and protein expression profiles from growth rate optimisation, providing a framework to understand metabolic adaptation in eukaryal cells.

    • The new Scientific Decade 2013-2022 of IAHS, entitled Panta RheiEverything Flows, is dedicated to research activities on change in hydrology and society. The purpose of Panta Rhei is to reach an improved interpretation of the processes governing the water cycle by focusing on their changing dynamics in connection with rapidly changing human systems. The practical aim is to improve our capability to make predictions of water resources dynamics to support sustainable societal development in a changing environment. The concept implies a focus on hydrological systems as a changing interface between environment and society, whose dynamics are essential to determine water security, human safety and development, and to set priorities for environmental management. The Scientific Decade 2013-2022 will devise innovative theoretical blueprints for the representation of processes including change and will focus on advanced monitoring and data analysis techniques. Interdisciplinarity will be sought by increased efforts to connect with the socio-economic sciences and geosciences in general. This paper presents a summary of the Science Plan of Panta Rhei, its targets, research questions and expected outcomes.

  • Timing Of Primary Surgery for cleft palate (TOPS): protocol for a randomised trial of palate surgery at 6 months versus 12 months of age

    • Introduction Cleft palate is among the most common birth abnormalities. The success of primary surgery in the early months of life is crucial for successful feeding, speech, hearing, dental development and facial growth. Over recent decades, age at palatal surgery in infancy has reduced. This has led to palatal closure in one-stage procedures being carried out around the age of 12 months, but in some cases as early as 6 months. The primary objective of the Timing Of Primary Surgery for Cleft Palate (TOPS)trial is to determine whether surgery for cleft palate performed at 6 or 12 months of age is most beneficial for speech outcomes. Methods and analysis Infants with a diagnosis of non-syndromic isolated cleft palate will be randomised to receive standardised primary surgery (Sommerlad technique) for closure of the cleft at either 6 months or 12 months, corrected for gestational age. The primary outcome will be perceived insufficient velopharyngeal function at 5 years of age. Secondary outcomes measured across 12 months, 3 years and 5 years will include growth, safety of the procedure, dentofacial development, speech, hearing level and middle ear function. Video and audio recordings of speech will be collected in a standardised age-appropriate manner and analysed independently by multiple speech and language therapists. The trial aims to recruit and follow-up 300 participants per arm. Data will be analysed according to the intention-to-treat principle using a 5% significance level. All analyses will be prespecified within a full and detailed statistical analysis plan. Ethics and dissemination Ethical approval has been sought in each participating country according to country-specific procedures. Trial results will be presented at conferences, published in peer-reviewed journals and disseminated through relevant patient support groups.

  • Tunable critical correlations in kagome ice

    • We present a comprehensive experimental and theoretical study of the kagome ice Coulomb phase that explores the fine-tuning of critical correlations by applied field, temperature, and crystal orientation. The continuous modification of algebraic correlations is observed by polarized neutron scattering experiments and is found to be well described by numerical simulations of an idealized model. We further clarify the thermodynamics of field-tuned Kasteleyn transitions and demonstrate some dramatic finite-size-scaling properties that depend on how topological string defects wind around the system boundaries. We conclude that kagome ice is a remarkable example of a critical and topological state in a real system that may be subject to fine experimental control.

  • Particle tracer transport in a sloping soil lysimeter under periodic, steady state conditions

    • Colloid transport through complex and dynamic (i.e. non-steady-state) hydrologic systems is rarely studied, owing to the difficulty of constraining initial and boundary conditions and quantifying colloid-porous media and colloid-colloid interactions in transient flow systems. Here we present a particle tracer experiment conducted on a sloped lysimeter receiving periodic rainfall events for 10 days. Four unique, DNA-labelled particle tracers were injected both in sequence and in parallel, together with a conservative tracer (deuterium), over the course of the first day and allowed to move through the system. Discharge-particle tracer concentration curves and the spatial distribution of particle tracer mass retained in the soil at the end of the experiment were found to be highly dependent on the timing of the tracer injection and the precipitation input and subsequent dynamic response of the water table. Overall, neglecting the total DLT recovery rate, the DLT particle tracer breakthrough trend (DNA-labelled particle tracer 4) was similar to deuterium and decreased over time with the exception of a few peaks later in the experiment. The individual particle tracer breakthrough curves suggest a complex system with different fast transport mechanisms (e.g. capillary barrier and size exclusion effect) and slow retention-release mechanisms (e.g. straining, physical-chemical adsorption), which resulted in particle tracers transferring faster than deuterium in the first 10 h of the experiment but being exceeded by deuterium soon after deuterium started to break through. The experiment not only highlights the interaction of repeated colloidal pollution events in hydrologic systems with different pre-event saturation conditions, but also the benefits of using multiple synchronous or sequential tracer applications to dissect explicit formulations of water flow and colloid transport processes in complex and dynamic hydrological systems. Such explicit process formulations could help improve understanding hydrologically-controlled transport through catchments and the quantitative prediction of these processes with water quality models.
