  • ARF suppression by MYC but not MYCN confers increased malignancy of aggressive pediatric brain tumors

    • Medulloblastoma, the most common malignant pediatric brain tumor, often harbors MYC amplifications. Compared to high-grade gliomas, MYC-amplified medulloblastomas often show increased photoreceptor activity and arise in the presence of a functional ARF/p53 suppressor pathway. Here, we generate an immunocompetent transgenic mouse model with regulatable MYC that develop clonal tumors that molecularly resemble photoreceptor-positive Group 3 medulloblastoma. Compared to MYCN-expressing brain tumors driven from the same promoter, pronounced ARF silencing is present in our MYC-expressing model and in human medulloblastoma. While partial Arf suppression causes increased malignancy in MYCN-expressing tumors, complete Arf depletion promotes photoreceptor-negative high-grade glioma formation. Computational models and clinical data further identify drugs targeting MYC-driven tumors with a suppressed but functional ARF pathway. We show that the HSP90 inhibitor, Onalespib, significantly targets MYC-driven but not MYCN-driven tumors in an ARF-dependent manner. The treatment increases cell death in synergy with cisplatin and demonstrates potential for targeting MYC-driven medulloblastoma.

  • MRTFA overexpression promotes conversion of human coronary artery smooth muscle cells into lipid-laden foam cells

    • Objective: Smooth muscle cells contribute significantly to lipid-laden foam cells in atherosclerotic plaques. However, the underlying mechanisms transforming smooth muscle cells into foam cells are poorly understood. The purpose of this study was to gain insight into the molecular mechanisms regulating smooth muscle foam cell formation. Approach and results: Using human coronary artery smooth muscle cells we found that the transcriptional co-activator MRTFA promotes lipid accumulation via several mechanisms, including direct transcriptional control of LDL receptor, enhanced fluid-phase pinocytosis and reduced lipid efflux. Inhibition of MRTF activity with CCG1423 and CCG203971 significantly reduced lipid accumulation. Furthermore, we demonstrate enhanced MRTFA expression in vascular remodeling of human vessels. Conclusions: This study demonstrates a novel role for MRTFA as an important regulator of lipid homeostasis in vascular smooth muscle cells. Thus, MRTFA could potentially be a new therapeutic target for inhibition of vascular lipid accumulation.

  • Speed characteristics of urban streets based on driver behaviour studies and simulation

    • The objective of the study was to gain in-depth knowledge of speed relationships for urban streets. The speed characteristics were examined using a number of methods for data collection. Throughout the research, a special focus was placed on capturing the influence on driver speed of interactions with pedestrians, cyclists and other road users, called sidefriction events in this study. First, driver behaviour and travel time data was collected from field and driving simulator studies for a range of street types and traffic conditions. The collected data was used to calibrate a microscopic traffic simulation model. Production runs with this model were performed for various traffic conditions. Second, aggregated speed data was collected at the link level, i.e. the macro level, for three street types. In combination with street site variables, speed and flow data was analysed using multiple regression techniques with space mean speed as dependent variable. This analysis was also performed for average travel speed data produced by microscopic traffic simulation. Two central results were attained and utilized for the model development: - In-depth knowledge of which factors influence speed choice on urban street links with minor intersections, on a micro and macro level. - A comprehensive research methodology for study of speed characteristics on urban streets in which the knowledge gained at the micro and macro level was applied. Results from the micro study showed that Average number of crossing pedestrians and Traffic flow had significant impact on average travel speed (R2=0.91). Results from the macro study performed for three street types showed that Street function and Number of lanes also had a high degree of explanation (R2 close to 0.70). The variables Separated bicycle lane, Roadside parking permitted and Number of minor intersections per 1 km were significant for some of the street types modelled in the macro study. The variables Ratio of through vehicles and Gender of the driver were also investigated and were found not to influence space-mean speed. The macro study demonstrated that speed choice and driver behaviour were consistent for each street type investigated regardless of city type and population size. The speed-flow relationships of the micro model for an urban street type showed good agreement with the macro model for traffic flows in the upper range. In conclusion, the research effort showed that the included side-friction variables added explanatory value to the estimation of speed, and thus can enhance the knowledge of traffic impacts of different urban street designs.

  • Dormant SOX9-positive cells behind MYC-driven medulloblastoma recurrence

    • Tumor recurrence is a slow biological process involving therapy resistance, immune escape, and metastasis and is the leading cause of death in medulloblastoma, the most frequent malignant pediatric brain tumor. By studying paired primary-recurrent patient samples and patient-derived xenografts we identified a significant accumulation of SOX9-positive cells in relapses and metastases. They exist as rare, quiescent cells in Group 3 and Group 4 patients that constitute two-thirds of medulloblastoma. To follow relapse at the single-cell level we developed an inducible dual Tet model of MYC-driven MB, where MYC can be directed from treatment-sensitive bulk cells to resistant, dormant SOX9-positive cells by doxycycline. SOX9 promoted immune es-cape, DNA repair suppression and was essential for recurrence. Tumor cell dormancy was non-hierarchical, migratory, and depended on MYC suppression by SOX9 to promote relapse. By using computational modeling and treatment we further showed how doxorubicin and MGMT inhibitors are specifically targeting relapsing cells.

  • CDK2 as a therapeutic target in MYC-driven medulloblastoma

    • Group 3 medulloblastoma (MB) is a malignant pediatric brain tumor that shows aberrant cell cycle activation, therapy resistance, and poor prognosis. Here, we identified that MYC expression and poor prognosis in Group 3 MB correlates with elevated levels of core cell cycle members CDK2 and cyclin A2, suggesting they would be promising targets for direct inhibition. Tumor cells in a novel transgenic MYC-driven MB mouse model further displayed increased p27 levels, decreased viability, and cell growth in vitro upon conditional CDK2 depletion using tamoxifen-induced recombination. Human Group 3 MB cells transduced with dominant-negative CDK2 mutants similarly exhibited decreased viability and increased p27 activation. As compared to controls, CDK2-depleted cells responded less to CDK2-specific inhibitors but were not more sensitive to BET inhibition or CDK4/6 inhibition as previously proposed. We finally used global transcriptional profiling and found that mTOR and B-Myb/ZMYM2 signaling pathways are compensating for CDK2 loss in Group 3MB cells. Our analysis suggests that specific inhibitors of these pathways could in combination with approved cell cycle inhibitors provide more efficient treatments for this severe childhood brain cancer.

    • Relapse is the leading cause of death in patients with medulloblas-toma, the most common malignant pediatric brain tumor. A better understanding of the mechanisms underlying recurrence could lead to more effective therapies for targeting tumor relapses. Here, we observed that SOX9, a transcription factor and stem cell/glial fate marker, is limited to rare, quiescent cells in high-risk medulloblastoma with MYC amplification. In paired primary-recurrent patient samples, SOX9-positive cells accumulated in medulloblastoma relapses. SOX9 expression anti-correlated with MYC expression in murine and human medulloblastoma cells. However, SOX9-positive cells were plastic and could give rise to a MYC high state. To follow relapse at the single-cell level, an inducible dual Tet model of medulloblastoma was developed, in which MYC expression was redirected in vivo from treatment-sensitive bulk cells to dormant SOX9-positive cells using doxycycline treatment. SOX9 was essential for relapse initiation and depended on suppression of MYC activity to promote therapy resistance, epithelial-mesenchymal transition, and immune escape. p53 and DNA repair pathways were downregulated in recurrent tumors, whereas MGMT was upregulated. Recurrent tumor cells were found to be sensitive to treatment with an MGMT inhibitor and doxorubicin. These findings suggest that recurrence-specific targeting coupled with DNA repair inhibition comprises a potential therapeutic strategy in patients affected by medulloblastoma relapse.Significance: SOX9 facilitates therapy escape and recurrence in medulloblastoma via temporal inhibition of MYC/MYCN genes, revealing a strategy to specifically target SOX9-positive cells to prevent tumor relapse.

    • The natural-step framework is used by over 100 organizations, including many global corporations in Europe and the United States, to provide strategic direction for their sustainability initiatives. The framework is built on the concept of simplicity without reduction. Out of respect for complexity we designed it to provide a compass, a guide for strategic direction. The framework consists of a backcasting planning process for sustainable development based on four principles (system conditions) for sustainability. The framework does not prescribe detailed actions. Once an organization understands the framework it identifies and specifies the detailed means by which to achieve the strategy, because it knows its business best. The steps in the planning process are understanding and discussing the system conditions for sustainability, describing and discussing how the company relates to the system conditions in today's situation, creating a vision of how the company will fulfill its customers' needs in the futur e while complying with the system conditions, and specifying a program of actions that will take the company from today's situation to the future vision.

    • A qualitative Method for Sustainable Product Development (MSPD) is presented. It consists of three tools: an Integrated Product Development Model (IPDM), a strategic planning process based on backcasting (ABCD-analysis), and a matrix containing hierarchically ordered and guiding questions. This Sustainability Product Analysis (SPA) matrix has conditions for sustainability along one axis and the life cycle of the product along the other axis. The SPA-matrix is used in the ABCD-analysis, which in turn is used in the IPDM. Integrating the tools in this way and using overall conditions for sustainability is unique compared to many other tools and methods for design for the environment. The MSPD is being developed in discussions with ten small- and medium sized enterprises.

    • Medulloblastoma (MB), the most frequent malignant childhood brain tumor, can arise from cellular malfunctions during hindbrain development. Here we generate humanized models for Sonic Hedgehog (SHH)-subgroup MB via MYCN overexpression in primary human hindbrain-derived neuroepithelial stem (hbNES) cells or iPSC-derived NES cells, which display a range of aggressive phenotypes upon xenografting. iPSC-derived NES tumors develop quickly with leptomeningeal dissemination, whereas hbNES-derived cells exhibit delayed tumor formation with less dissemination. Methylation and expression profiling show that tumors from both origins recapitulate hallmarks of infant SHH MB and reveal that mTOR activation, as a result of increased Oct4, promotes aggressiveness of human SHH tumors. Targeting mTOR decreases cell viability and prolongs survival, showing the utility of these varied models for dissecting mechanisms mediating tumor aggression and demonstrating the value of humanized models for a better understanding of pediatric cancers.

    • Since the mid-twentieth century, plastic has become a ubiquitous material. However, its produc- tion, consumption and disposal on a massive scale have led to a range of devastating conse- quences that together form the ‘plastic crisis’. This paper presents a novel mapping of the ways contemporary artistic visual representations narrate and politicize the plastic crisis through their different messages and aesthetics. Drawing on a multifaceted understanding of the plastic crisis and on how art navigates political and aesthetic spheres, an analysis of 35 artworks is con- ducted. Ocean plastic pollution emerges as a dominant theme, together with disposability; and these are connected to consumption patterns and consumer responsibility. However, less atten- tion is given to plastic’s dependence on fossil fuels and possible toxicity. The result is art of strik- ing beauty and emotional resonance, but that downplays the systemic nature of the plastic crisis and the urgent need to hold manufacturers and regulators to account.

    • We present an extensive photophysical study of a series of fluorescent indolylbenzothiadiazole derivatives and their ability to specifically image lipid droplets in astrocytes and glioblastoma cells. All compounds in the series displayed positive solvatochromism together with large Stokes shifts, and π-extended derivatives exhibited elevated brightness. It was shown that the fluorescence properties were highly tunable by varying the electronic character or size of the N-substituent on the indole motif. Three compounds proved capable as probes for detecting small quantities of lipid deposits in healthy and cancerous brain cells. In addition, all twelve compounds in the series were predicted to cross the blood–brain barrier, which raises the prospect for future in vivo studies for exploring the role of lipid droplets in the central nervous system.

    • The United Nations (UN) Intergovernmental Negotiating Committee (INC) has completed its second round of negotiations on a new international legally binding instrument to ‘end plastic pollution’. This paper examines the results of the second session of negotiations, which took place May 29–June 2, 2023, and analyses the trends, barriers, and themes shaping the emerging legally binding instrument. The paper considers the discussion surrounding the Options Paper as presented by the INC Secretariat and United Nations Environmental Program based upon written submissions received by UN member states prior to the second session of negotiations. These include potential elements the future treaty text may include—namely objectives, core obligations, and implementation and monitoring measures. At the second session of negotiations, progress towards consensus on these items was severely lacking. We draw on close observations via event ethnography of participants during the negotiations and document analysis. We conclude by looking towards the third round of negotiations by discussing the ongoing ‘dance’ of sorts as submissions are being asked of member states and like-minded groups to contribute to the zero draft treaty text.

    • Background & AimsYAP (Yap1) and TAZ (Wwtr1) are transcriptional co-activators and downstream effectors of the Hippo pathway, which play crucial roles in organ size control and cancer pathogenesis. Genetic deletion of YAP/TAZ has shown their critical importance for embryonic development of the heart, vasculature, and gastrointestinal mesenchyme. The aim of this study was to determine the functional role of YAP/TAZ in adult smooth muscle cells in vivo.MethodsBecause YAP and TAZ are mutually redundant, we used YAP/TAZ double-floxed mice crossed with mice that express tamoxifen-inducible CreERT2 recombinase driven by the smooth muscle–specific myosin heavy chain promoter.ResultsDouble-knockout of YAP/TAZ in adult smooth muscle causes lethality within 2 weeks, mainly owing to colonic pseudo-obstruction, characterized by severe distension and fecal impaction. RNA sequencing in colon and urinary bladder showed that smooth muscle markers and muscarinic receptors were down-regulated in the YAP/TAZ knockout. The same transcripts also correlated with YAP/TAZ in the human colon. Myograph experiments showed reduced contractility to depolarization by potassium chloride and a nearly abolished muscarinic contraction and spontaneous activity in colon rings of YAP/TAZ knockout.ConclusionsYAP and TAZ in smooth muscle are guardians of colonic contractility and control expression of contractile proteins and muscarinic receptors. The knockout model has features of human chronic intestinal pseudo-obstruction and may be useful for studying this disease.

  • Role of smooth muscle YAP and TAZ in protection against phenotypic modulation, inflammation, and aneurysm development

    • A ruptured arterial aneurysm, especially in the aorta, represents one of the most acute and mortal conditions encountered in clinical medicine. Population-based screening in elderly men, treatment of risk factors, such as hypertension, and endovascular or open repair of rupture-prone lesions, represent cornerstones in management. Surgical repair has a sizeable effect on life-expectancy, but medical therapy that retards aneurysm growth still represents a considerable and unmet clinical need. In the current review we survey recent findings implicating the mechano-responsive transcriptional co-activators YAP and TAZ in protection from aneurysm development. Arteries from mouse mutants that lack YAP and TAZ in vascular smooth muscle respond inadequately to mechanical stimulation, and they develop aneurysms characterized by elastin fragmentation, proteoglycan infiltration, and severe inflammation at breathtaking speed. This seems to be due, at least in part, to unscheduled activation of STING (stimulator of interferon genes), an arm of innate immunity that responds to double-stranded DNA in the cytoplasm. YAP and TAZ protect from STING activation by securing nuclear integrity. These novel insights suggest unanticipated medical therapies for sporadic and genetic aneurysms alike, involving inhibition of kinases in the Hippo pathway using small molecules, or inhibition of STING signaling itself. Translation of these novel findings into clinical therapies now represents an important priority.

    • Background: Hypertension remains a major risk factor for cardiovascular diseases, but the underlying mechanisms are not well understood. We hypothesize that appropriate mechanotransduction and contractile function in vascular smooth muscle cells are crucial to maintain vascular wall integrity. The Hippo pathway effectors YAP (yes-associated protein 1) and TAZ (WW domain containing transcription regulator 1) have been identified as mechanosensitive transcriptional coactivators. However, their role in vascular smooth muscle cell mechanotransduction has not been investigated in vivo. Methods: We performed physiological and molecular analyses utilizing an inducible smooth muscle-specific YAP/TAZ knockout mouse model. Results: Arteries lacking YAP/TAZ have reduced agonist-mediated contraction, decreased myogenic response, and attenuated stretch-induced transcriptional regulation of smooth muscle markers. Moreover, in established hypertension, YAP/TAZ knockout results in severe vascular lesions in small mesenteric arteries characterized by neointimal hyperplasia, elastin degradation, and adventitial thickening. Conclusions: This study demonstrates a protective role of YAP/TAZ against hypertensive vasculopathy.

    • This work presents the pH sensing ability of a fluorescent indolin-3-imine derivative. Protonation of the weakly basic imine (pKa = 8.3 of its conjugate acid) results in a significant redshift of the absorption band. The fluorophore acts as a photobase, with a basicity increase of approximately 6 units upon photoexcitation. This behavior promotes excited state proton transfer from weak acids such as protic solvents. The characteristics of the fluorophore enable sensing of water fractions in organic solvents and differentiation between methanol, ethanol, and longer chain alcohols. Initial cell studies indicated the future potential of indolin-3-imines as fluorophores for bioimaging applications.

    • The study of a fluorescent indolin-3-one derivative is reported that, as opposed to its previously described congeners, selectively undergoes photoactivated ring-opening in apolar solvents. The excited state involved in this photoisomerization was partially deactivated by the formation of singlet oxygen. Cell studies revealed lipid droplet accumulation and efficient light-induced cytotoxicity.

    • In this work, a series of fluorescent 2,1,3-benzothiadiazole derivatives with various N-substituents in the 4- position was synthesized and photophysically characterized in various solvents. Three compounds emerged as excellent fluorescent probes for imaging lipid droplets in cancer cells. A correlation between their high lipophilicity and lipid droplet specificity could be found, with log P ≥ 4 being characteristic for lipid droplet accumulation.

  • Towards a Global Plastics Treaty: Tracing the UN Negotiations

    • In 2022, the United Nations Environmental Assembly (UNEA) adopted resolution 5/14, giving mandate to UN Member States to start negotiations toward an international legally binding instrument on plastic pollution. This report delves into the pre-session submissions by states and coalitions for the second (INC-2) and third (INC-3) rounds of negotiations, held in 2023. Once the contents of the documents had been reviewed, a coding analysis was conducted with the aim of understanding the current policy mix being proposed for the Plastics Treaty. The nine categories to emerge from the coding process cover proposed objectives for the treaty, types and range of measures proposed, and where along the value chain these measures fall. Our results show: (1) that the pre-session submissions give considerable attention to improving waste management and extending recycling infrastructure, while scant attention is given to upstream measures; (2) that relatively few economic measures have been proposed thus far, with pre-session submissions focusing on regulatory and soft measures instead; (3) that this uneven distribution of proposed measure types could weaken the overall effectiveness of the instrument by impeding its ability to address the issue of plastic pollution in all its complexity; (4) that, if current trends continue, we can expect a treaty focused on waste management and recycling, instead of one addressing the full life cycle of plastics; (5) and finally, that the sheer quantity of plastics being produced each year undermines any efforts either to ‘end’ plastic pollution or to reach any net-zero carbon emission targets.
