  • Organic carbon budget for the Gulf of Bothnia

    • We calculated input of organic carbon to the unproductive, brackish water basin of the Gulf of Bothnia from rivers, point sources and the atmosphere. We also calculated the net exchange of organic carbon between the Gulf of Bothnia and the adjacent marine system, the Baltic Proper. We compared the input with sinks for organic carbon; permanent incorporation in sediments and mineralization and subsequent evasion of CO2 to the atmosphere. The major fluxes were riverine input (1500 Gg C year(-1)), exchange with the Baltic Proper (depending on which of several possible DOC concentration differences between the basins that was used in the calculation, the flux varied between an outflow of 466 and an input of 950 Gg C year(-1)), sediment burial (1100 Gg C year) and evasion to the atmosphere (3610 Gg C year(-1)). The largest single net flux was the emission of CO2 to the atmosphere, mainly caused by bacterial mineralization of organic carbon. Input and output did not match in our budget which we ascribe uncertainties in the calculation of the exchange of organic carbon between the Gulf of Bothnia and the Baltic Proper, and the fact that CO2 emission, which in our calculation represented 1 year (2002) may have been overestimated in comparison with long-term means. We conclude that net heterotrophy of the Gulf of Bothnia was due to input of organic carbon from both the catchment and from the Baltic Proper and that the future degree of net heterotrophy will be sensible to both catchment export of organic carbon and to the ongoing eutrophication of the Baltic Proper.

    • A processing scheme for the fabrication of embedded W-GaAs contacts has been established and the resulting contact characteristics have been evaluated. The main advantage of these contacts is that they are stable during high-temperature epitaxial overgrowth. The fabrication scheme is based on a liftoff process with electron beam evaporation of tungsten and subsequent epitaxial overgrowth using metalorganic vapor phase epitaxy. Various methods were used to characterize the buried contacts. First, the structural properties of GaAs surrounding embedded W features, with widths down to 50 nm, were characterized by high-resolution transmission electron microscopy. Measurements of the conductivity in individual, buried wires were performed in order to study the influence of the overgrowth process on the properties of the tungsten. We also evaluated the current-voltage characteristics for macroscopic contacts, which revealed a clear dependence on processing parameters. Optimized processing conditions could thus be established under which limited contact degradation occurred during overgrowth. Finally, we used the overgrowth technique to perform a detailed investigation of the electrical and optical properties of floating-potential embedded nano-Schottky contacts by space-charge spectroscopy.

  • Tissue variations of mosaic genome-wide paternal uniparental disomy and phenotype of multi-syndromal congenital hyperinsulinism

    • Mosaic genome-wide paternal uniparental disomy (GW-pUPD) is a rarely recognised disorder. The phenotypic manifestations of multilocus imprinting defects (MLIDs) remain unclear. We report of an apparently non-syndromic infant with severe congenital hyperinsulinism (CHI) and diffuse pancreatic labelling by 18F*-DOPA-PET/CT leading to near-total pancreatectomy. The histology was atypical with pronounced proliferation of endocrine cells comprising >70% of the pancreatic tissue and a small pancreatoblastoma. Routine genetic analysis for CHI was normal in the blood and resected pancreatic tissue. At two years’ age, Beckwith-Wiedemann Syndrome (BWS) stigmata emerged, and at five years a liver tumour with focal nodular hyperplasia and an adrenal tumour were resected. pUPD was detected in 11p15 and next in the entire chromosome 11 with microsatellite markers. Quantitative fluorescent PCR with amplification of chromosome-specific DNA sequences for chromosomes 13, 18, 21 and X indicated GW-pUPD. A next generation sequencing panel with 303 SNPs on 21 chromosomes showed pUPD in both blood and pancreatic tissue. The mosaic distribution of GW-pUPD ranged from 31 to 35% in blood and buccal swap to 74% in the resected pancreas, 80% in a non-tumour liver biopsy, and 100% in the liver focal nodular hyperplasia and adrenal tumour. MLID features included transient conjugated hyperbilirubinaemia and lack of macrosomia from BWS (pUPD6); and behavioural and psychomotor manifestations of Angelman Syndrome (pUPD15) on follow-up. In conclusion, atypical pancreatic histology in apparently non-syndromic severe CHI patients may be the first clue to BWS and multi-syndromal CHI from GW-pUPD. Variations in the degree of mosaicism between tissues explained the phenotype.

    • OBJECTIVE: : The aim of this study is to determine Swedish parents' willingness to pay (WTP) for coeliac disease (CD) screening of their child. SUBJECTS AND METHODS: : CD screening was undertaken involving 10,041 12-year-old children, with 7567 (75%) agreeing to participate. Blood samples from the children were analysed for CD serological markers. Parents received a questionnaire including a scenario describing the health-related risks of having CD and screening and diagnostic procedures. Parents were also asked whether they were willing to pay for CD screening, should this not be offered free of charge, and, if so, what their maximum WTP would be. Their WTP was compared with the average cost per child for the screening and case ascertainment procedures. RESULTS: : The questionnaire was answered by 6524 parents, and of 6057 valid responses 63% stated that they were willing to pay something. The mean WTP was 79 EUR and the median 10 EUR. The average cost per child for the screening and case ascertainment procedures was 47 EUR, which 23% of the parents stated they were willing to pay. Parents' WTP increased with higher education and income, and with child symptoms that may indicate CD. CONCLUSIONS: : Swedish parents' WTP for school-based CD screening of their child was higher than the average cost per child; however, only a minority of the parents were willing to pay that amount.

  • A clinical single-pass perfusion investigation of the dynamic in vivo secretory response to a dietary meal in human proximal small intestine

    • Purpose  To investigate the gastrointestinal secretory and enzymatic responses to a liquid meal during in vivo perfusion of the proximal human jejunum. Methods  Human intestinal fluid was collected from the proximal jejunum by single-pass in vivo perfusion (Loc-I-Gut). The fluid was quantitatively collected at 10-min intervals during 90 min while perfusing a nutritional drink at 2 mL/min. Quantification of lipids in the fluid leaving the segment was performed by using novel chromatographic methods. Results  The overall bile acid concentration varied between 0.5 and 8.6 mM with a peak level 40 min after the start of the liquid meal perfusion. The total concentration of phospholipids was between 0.1 and 3.9 mM and there was a rapid degradation of phosphatidylcholine to lysophosphatidylcholine. The tri-, di-, monoglycerides and free fatty acid levels increased sharply in the beginning and reached steady-state levels between 7 and 9.5 mM. Conclusions  There is a rapid secretion of bile in response to food. Most of the dietary lipids are found in the form of their degradation products in vivo in human jejunum. This novel in vivo characterization, based on direct and high-recovery sampling of intestinal fluids, forms a basis for further development of improved in vitro drug dissolution test media.

  • Measuring the value of older people's production : a diary study

    • Background: The productive capacity of retired people is usually not valued. However, some retirees produce much more than we might expect. This diary-based study identifies the activities of older people, and suggests some value mechanisms. One question raised is whether it is possible to scale up this diary study into a larger representative study.Methods: Diaries kept for one week were collected among 23 older people in the north of Sweden. The texts were analysed with a grounded theory approach; an interplay between ideas and empirical data.Results: Some productive activities of older people must be valued as the opportunity cost of time or according to the market value, and others must be valued with the replacement cost. In order to make the choice between these methods, it is important to consider the societal entitlement. When there is no societal entitlement, the first or second method must be used; and when it exists, the third must be used.Conclusions: An explicit investigation of the content of the entitlement is needed to justify the choice of valuation method for each activity. In a questionnaire addressing older people's production, each question must be adjusted to the type of production. In order to fully understand this production, it is important to consider the degree of free choice to conduct an activity, as well as health-related quality of life.

    • Inom projektet Biokombi Rya har ett flertal olika forskargrupper samarbetat för att studera system¬effekterna av förgasning av biobränsle ur olika aspekter. Syftet med projektet är att öka kunskapen om biobränsleförgasning i Sverige samt att utreda förutsättningar för att sådana anläggningar ska vara ekonomiskt och miljömässigt intressanta. En referensgrupp har varit kopplad till projektet där förutsättningar, resultat och slutsatser har behandlats.I denna underlagsrapport har slutrapporterna från projektets delprojekt samlats. De beskriver förutsättningar, metodansatser, använda data och resultat utförligt och utgör på så sätt ett viktigt komplement till den mer övergripande beskrivningen i projektets syntesrapport. De delrapporter som ingår har valts för att täcka in samtliga delar av projektet som är av allmänt intresse. Projektresultat som publicerats på annat sätt berörs dock mer kortfattat.Projektet Biokombi Rya har pågått under två år (2005-2006) och drivits av Chalmers EnergiCentrum. Förutom de omfattande analysinsatser som författarna till denna rapport står för, har Avdelningen för kemisk teknologi vid KTH, Siemens Industrial Turbines AB och Göteborg Energi AB bidragit med expertstöd.     • BACKGROUND: Cigarette smoke induces inflammation and remodels immune response. Genetic and epigenetic alterations might be involved in the pathogenesis of smoking related diseases. In this study, we investigated the effect of smoking on systemic inflammation biomarkers and epigenetic changes at microRNA (miRNA) expression level. We also examined if the levels of inflammatory biomarkers were associated with selected single nucleotide polymorphisms (SNPs).METHOD: From 39 smokers and 101 non-smokers, levels of total white blood cells (WBCs) and its subpopulations, plasma cytokines/chemokines/proteins and miRNAs were analysed. For three biomarkers, C-reactive protein (CRP), MCP-1 and IFN-γ that were affected by smoking, the influence of SNPs was analyzed.RESULT: Elevated levels of total WBCs, neutrophils, monocytes, lymphocytes, CRP, MCP-1, IFN-γ and lower levels of miR-21 were detected in smokers. The elevated levels of IFN-γ in smokers was only statistically significantly associated with rs2069705 AG/GG SNP-genotype.CONCLUSIONS: A lower level of oncomir miRNA-21 and a higher level of immune modelling cytokine IFN-γ detected in smokers could be a protective immune response to cigarette smoke. The higher level of IFN-γ in smokers with a specific SNP genotype also suggests that a genetic interaction with smoking might predict the pathobiology of smoking related disease.

    • PURPOSE: Tumor TNM staging is the main basis for prognosis and treatment decision for head and neck squamous cell carcinoma (HNSCC) despite significant heterogeneity in terms of outcome among patients with the same clinical stage. In this study, a possible role of plasma interleukin-2 (IL-2), interleukin-6 (IL-6), granulocyte-macrophage colony-stimulating factor (GM-CSF), tumor necrosis factor-α (TNF-α) and C-reactive protein (CRP) as biomarkers for survival of HNSCC patients was investigated.METHODS: In this prospective study, plasma levels of IL-2, IL-6, GM-CSF, TNF-α and CRP in patients (n = 100) and controls (n = 48) were analyzed.RESULTS: Significantly elevated levels of CRP and TNF-α (p < 0.001) were found in the patients. Combination of upregulated CRP and TNF-α in the patient plasma was significantly related to shorter patient survival, independent of clinical stage.CONCLUSIONS: Our findings indicate that CRP and TNF-α might be suitable as biomarkers in combination with tumor TNM staging for predicting survival and individualized treatment of HNSCC patients. Plasma CRP and TNF-α analysis are simple, rapid, cost effective and suitable for clinical practice.

  • Technology Forecast 2012: Military utility of ten technologies : a report from seminars at the SNDC Department of Military Technology

    • Ten technology forecast reports from the Fraunhofer Institute have been reviewed by staff at the Department of Military-Technology at the Swedish National Defence College (Note that there probably are other technology areas, equally interesting, but not included in this study). The task given by FMV was to assess the military utility of the chosen technologies in a time frame from 2025 to 2030, from a SwAF viewpoint.The method used was first to make a summary of each forecast report. The technology was then put into one or more scenarios that are assessed to be the best in order to show possible utility as well as possibilities and drawbacks of the technology. Based on a SWOT-analysis, the contribution to SwAF capabilities and the cost in terms of acquisition, C2 footprint, logistic footprint, doctrine/TTP, training, facilities and R&D were assessed. Conclusions regarding the military utility of the technology were drawn.We introduce our definition of military utility as being activities that efficiently and with the lowest cost in terms of lives and materiel lead to fulfilment of the mission objectives.The technologies were grouped in three classes; technologies with a significant potential, with uncertain potential and with negligible potential.The following technologies were assessed to have a significant potential for military utility;Augmented RealityNano air vehiclesSolid State Laser weaponsIn the scenarios studied, Augmented Reality (AR) is assessed to have a positive impact on several SwAF capabilities, especially for C2 and intelligence. AR is a relatively mature technology, applicable in many different branches. There are examples where AR is already applied with great success, e.g. Head-Up-Displays, HUD. The technology has proven its value. However, there are well known drawbacks to the technology such as weaknesses regarding models, increased weight for dismounted soldiers, power consumption etc. There is also a risk that personnel will have problems solving their tasks when AR systems fail, not being used to fighting without supporting systems.Nano air vehicles (NAV’s) have been assessed to contribute to a large range of capabilities, primarily intelligence. Their lifecycle cost has been assessed to be low, since development in this area is commercially driven, bringing down acquisition costs. Also, FAA has decided to allow NAV’s in controlled air space from 2015, which is expected to lead to an increase in civilian use of NAV’s. The technology is relatively mature even though there are obstacles concerning suitable materials, energy efficient propulsion systems as well as miniaturized microprocessors and software to control them.In the scenario studied, High Energy Solid State Lasers are assessed to have a positive impact on SwAF capabilities to engage targets on surface and in the air. The technology can be used to protect vessels on the surface and thereby increase survivability. The development of SSL in the given timeframe is expected to lower cost per shot and avoid the environmental problems with use of chemical lasers. Neighbouring military powers are expected to use laser weapons in the future, therefore SwAF should monitor the development of the laser weapons technology and develop and purchase adequate countermeasures.The following technologies were assessed to have uncertain potential for military utility;Metamaterial cloakingElectromagnetic gunSmall satellitesUltra-violet communicationMetamaterial cloaking, if realisable in the future, is assessed to be firstly implemented in the acoustic spectrum, since manufacturing of small structured cloaks for the shorter wavelengths in the optic and radar spectra is believed to be more difficult. Cloaking of submarines is primarily assessed to increase the survivability against torpedoes having active sonar. The use of cloaked mines could pose a deterring threat, even to advanced amphibious operations against Sweden. The technological development in this area should be closely monitored and compared to existing, maturing techniques for countermeasures and for the development of broad spectrum active torpedoes. The greatest concern is that cloaking will have negative impact on submarine manoeuvrability.The electro-thermal chemical (ETC) gun seems to be a first step towards a fully electrical gun such as the rail-gun or the coil-gun. The fully electrical guns have been a work in progress for some decades and there are still remaining challenges both concerning electrical power supply and design materials. When or if, they will be operational is difficult to say.The military utility of small satellites is disputed, despite an assessed contribution to several of the SwAF capabilities. The main reason for this is that there seems to be other alternatives which provide the desired capabilities, at a lower cost. Furthermore, the realisability and performance of small productionline manufactured nanosatellites is uncertain. However the scenario has shown that there are benefits to the military utility not met by other resources, e.g. the capability to perform surveillance and reconnaissance in operational areas globally without risking violation of the territorial integrity of other states or the lives of military personnel. Since there is a great interest in the technology area and several programmes are ongoing internationally the knowledgebase is assessed to be significantly better in a five year period. Also, the Swedish in depth study of space exploitation is soon to report.Ultra-violet communication has uncertain potential for military utility within the period, but the technology is assessed to have a positive impact on SwAF capability to maintain communications. The theoretical understanding of the area is low It is therefore uncertain if systems can be realized in the time frame. However, if commercial applications are developed, the prospect of military applications might change. In that case UV-communication could be a complement to RF- communication but is not foreseen to replace it. The following technologies were assessed to have negligible potential for military utility;Biomimetic unmanned underwater vehicles (UUV)Automated behaviour AnalysisEvolutionary RoboticsBiomimetic UUV’s could be used for covert surveillance and inconspicuous naval reconnaissance missions at sea or in amphibious missions. Even though the report focuses on fishlike propulsion, the military utility of UUV’s is assessed to be mostly dependent on the development of advanced automation and learning systems. As of now, we assess other existing technologies as being preferable due to lower cost and less complexity. The performance of UUV’s needed for SwAF capabilities are assessed to be far off into the future. Simpler UUV systems could however be used by potential adversaries for monitoring our own base areas and hence the development should be monitored from a protection point of view.Automated behaviour analysis may be of some relevance for increased security screening and surveillance. The primary military utility of the technology will however probably be for international activities and to a lesser extent for increased base security in Sweden. Generally the main applications for this kind of technology are assessed to be for civilian use in public spaces and close to high value areas like airports, important official buildings and other similar objects.Evolutionary Robotics, here restricted to the sub domain Advanced Robotics, has uncertain potential for military utility within the period. In the scenarios studied the technology is assessed to have a positive impact on a broad range of SwAF capabilities. The area is large and inconsistent comprising sub areas that are assessed to have significant potential, but also those that are believed to have negligible potential or where technological obstacles might retard the development.Our evaluation of the used method shows that there is a risk that the assessment is biased by the participating experts’ presumptions and experiences from their own field of research. The scenarios that were chosen do not cover all possible aspects of the technology and their possible contribution to operational capabilities. It should be stressed that we have assessed the ten technologies’ military utility in the presented scenarios, not the technology itself. The chosen definition of military utility clearly affects the result of the study. The definition is believed to be good enough for this report, but could be further elaborated in the future.The greatest value of the method used is its simplicity, cost effectiveness and the trade off that it promotes learning within the working group. The composition of the working group and the methodology used is believed to provide for a broad and balanced coverage of the technologies under study.

Denna nionde volym av Lärobok i Militärteknik, benämnd Teori och Metod, behandlar centrala begrepp, teorier och postulat samt metoder för värdering av teknik och består av ett antal texter författade av 16 forskare och lärare vid den militärtekniska avdelningen. Volymen riktar sig främst till de som inlett sin officersutbildning och utgörs till stora delar av ett kompilat av publicerade och opublicerade militärtekniska texter och kan sägas utgöra militärteknikens ”state of the art”.

  • Improved chip design for integrated solid-phase microextraction in on-line proteomic sample preparation

    • A recently introduced silicon microextraction chip (SMEC), used for on-line proteomic sample preparation, has proved to facilitate the process of protein identification by sample clean up and enrichment of peptides. It is demonstrated that a novel grid-SMEC design improves the operating characteristics for solid-phase microextraction, by reducing dispersion effects and thereby improving the sample preparation conditions. The structures investigated in this paper are treated both numerically and experimentally. The numerical approach is based on finite element analysis of the micro-fluidic flow in the microchip. The analysis is accomplished by use of the computational fluid dynamics-module FLOTRAN in the ANSYS(R) software package. The modeling and analysis of the previously reported weir-SMEC design indicates some severe drawbacks, that can be reduced by changing the microextraction chip geometry to the grid-SMEC design. The overall analytical performance was thereby improved and also verified by experimental work. Matrix-assisted laser desorption/ionization mass spectra of model peptides extracted from both the weir-SMEC and the new grid-SMEC support the numerical analysis results. Further use of numerical modeling and analysis of the SMEC structures is also discussed and suggested in this work.

    • Obemannade farkoster används allt oftare, och i allt fler roller, i dagens kon- flikter. Denna rapport ger en bred överblick över området militära obemannade farkoster, samt rekommendationer för inriktningen av framtida FoU-satsningar inom området.Överblicken över området har fokus på både system, förmågor och verksam- heter som är relevanta för Försvarsmakten. Genom att låta de insatsförmågor som definieras i FMUP (Försvarsmaktens utvecklingsplan) gå som en röd tråd genom rapporten, både när specifika system diskuteras och när möjliga scena- rier där obemannade farkoster kan vara till nytta beskrivs, har vi försökt hålla både bredd och relevans i dokumentet.Rekommendationerna vilar på en genomgång av de inriktningsdokument som producerats i Försvarsmakten, t.ex. Perspektivplanneringen och FMUP, besök vid de enheter som dagligen använder obemannade farkoster, UAV-enheten i Karlsborg och Swedec i Eksjö, samt den områdesöverblick som nämns ovan. Slutsatserna är att den effektivaste kompetensuppbyggnaden och kunskapsöver- föringen fås om man skapar breda tvärvetenskapliga projekt inom respektive systemkategori (UAV, UGV, etc) med nära kontakter till materielförsörjnings- processen och perspektivplaneringen. Dessa kan samla kompetensen inom FHS och FOI, övervaka forskningsfronten genom att bevaka tävlingar, konferenser samt delta i internationella samarbeten, samt överföra det samlade resultaten till Försvarsmakten genom demonstrationer av verkliga eller simulerade delsy- stem och interaktiva simuleringar av hela system. Just systemsimuleringar kan göras särskilt realistiska, eftersom interaktionen med de riktiga obemannade systemen till stor del sker igenom kontrollstationernas datorer. På så sätt ska- pas en känsla för både hot och möjligheter med de nya systemen, vilket gagnar både taktikutveckling och materielprocesser.

    • Since 2008 there has been a rapid buildup of surplus emissions allowances in the EU ETS. The European Commission has presented six structural options to address the oversupply of allowances on a long-term basis. One option would introduce price management mechanisms, which could allow for the use of a price floor. A price floor has been mischaracterized as a tax, an instrument that has historically faced political opposition, and the commission states that an explicit carbon price objective would alter the nature of the EU ETS being a quantity-based market instrument. However, a price floor is structurally different from a tax in multiple ways, and its merits are well documented in the academic literature. This rule-based approach could reinforce the market-based philosophy and investment climate of the ETS.

    • Following the 2009 Copenhagen Summit, momentum for mitigating global climate change has been spurred by the idea that success at the international level must be built around existing national policies. The Mistra Indigo research program —a collaboration among IVL Swedish Environmental Research Institute, the University of Gothenburg’s Department of Economics, and Resources for the Future—was created to analyze this bottom-up approach to climate policy. From 2012-2015 the program brought together leading experts in economics and environmental policy, with the goal of understanding how to design and select environmental policy instruments to promote longterm, cost-effective global climate change mitigation change while taking into account the uncertain ties of international policy.

    • This is a pilot study to investigate icing on wings of wind power turbines. In this report we present and discuss various ways and means to either heat water droplets or melt ice when formed on the wings of wind turbines. The situation is different from icing on wings of airplanes in that (1) the wings of wind turbines spend all of their time in the atmosphere where the risk of icing is highest and (2) the speed of wing to air varies over the wing where it is constant for an airplane. The form of the wind turbine wings also varies from tip to centre, to compensate for the varying relative air speed.We have concentrated on icing conditions at temperatures -10°C – 0°C and droplet sizes of 1-10 μm. Icing occurs also at much lower temperatures, but this will probably be because of direct freezing of water vapour to ice. This is presently outside the scope of our pilot project report.We conclude thatThe form of the wing, especially on the contact area may be crucial to the icing problem.Also the nano-metric structure of the wing surface can probably be designed so that the water droplets have a minimized contact area to the wing.Our pilot investigation also suggests the following:Microwaves are much too inefficient to heat water or melt ice. Direct microwave devices should therefore not be developed. Indirect heating with microwaves is possible.Millimeter waves are sufficiently efficient, but the generation is most probably too inefficient to be of any practical use.Infrared waves are very efficient to heat water and melt ice and should be investigated.Heat conduction is also efficient and should be pursued. Using microwaves to heat the wing surface which then conduct heat to the water/ice is a very efficient and robust method.Our pre-study suggests that the solution to avoid icing or de-ice wings of wind turbines most probably is not one single technology. The form and surface structure of the wings play important role for icing conditions. Both variables have to be modified depending on the latitude and atmospheric climate. The surface structure also has to be designed to vary over the wing, both along and across to be optimized for the mean conditions at the site. In addition, heating of the impact area, or at least the possibility to heat this, may be important to avoid loss of energy output due to ice.Further research is required. We strongly suggest investigating the water droplet flow over the wing as function of the cross section form, and the contact with the wing surface as function of the surface structure (e.g. Lotus effect).The present report is the result of a pre-study project. We will now continue with a deeper project which will concentrate on the form and surface structure suggestions which results from our analysis and flow simulations.
