  • Robust estimation of bacterial cell count from optical density

    • Optical density (OD) is widely used to estimate the density of cells in liquid culture, but cannot be compared between instruments without a standardized calibration protocol and is challenging to relate to actual cell count. We address this with an interlaboratory study comparing three simple, low-cost, and highly accessible OD calibration protocols across 244 laboratories, applied to eight strains of constitutive GFP-expressing E. coli. Based on our results, we recommend calibrating OD to estimated cell count using serial dilution of silica microspheres, which produces highly precise calibration (95.5% of residuals <1.2-fold), is easily assessed for quality control, also assesses instrument effective linear range, and can be combined with fluorescence calibration to obtain units of Molecules of Equivalent Fluorescein (MEFL) per cell, allowing direct comparison and data fusion with flow cytometry measurements: in our study, fluorescence per cell measurements showed only a 1.07-fold mean difference between plate reader and flow cytometry data.

    • The Visual Object Tracking challenge VOT2019 is the seventh annual tracker benchmarking activity organized by the VOT initiative. Results of 81 trackers are presented; many are state-of-the-art trackers published at major computer vision conferences or in journals in the recent years. The evaluation included the standard VOT and other popular methodologies for short-term tracking analysis as well as the standard VOT methodology for long-term tracking analysis. The VOT2019 challenge was composed of five challenges focusing on different tracking domains: (i) VOT-ST2019 challenge focused on short-term tracking in RGB, (ii) VOT-RT2019 challenge focused on "real-time" short-term tracking in RGB, (iii) VOT-LT2019 focused on long-term tracking namely coping with target disappearance and reappearance. Two new challenges have been introduced: (iv) VOT-RGBT2019 challenge focused on short-term tracking in RGB and thermal imagery and (v) VOT-RGBD2019 challenge focused on long-term tracking in RGB and depth imagery. The VOT-ST2019, VOT-RT2019 and VOT-LT2019 datasets were refreshed while new datasets were introduced for VOT-RGBT2019 and VOT-RGBD2019. The VOT toolkit has been updated to support both standard short-term, long-term tracking and tracking with multi-channel imagery. Performance of the tested trackers typically by far exceeds standard baselines. The source code for most of the trackers is publicly available from the VOT page. The dataset, the evaluation kit and the results are publicly available at the challenge website(1).

    • To investigate the nature of the psi(3770) resonance and to measure the cross section for e(+)e(-) -> D (D) over bar, a cross-section scan data sample, distributed among 41 center-of-mass energy points from 3.73 to 3.89 GeV, was taken with the BESIII detector operated at the BEPCII collider in the year 2010. By analyzing the large angle Bhabha scattering events, we measure the integrated luminosity of the data sample at each center-of-mass energy point. The total integrated luminosity of the data sample is 76.16 +/- 0.04 +/- 0.61 pb(-1), where the first uncertainty is statistical and the second systematic.

  • Pharmacological modulation of T cell immunity results in long-term remission of autoimmune arthritis

    • Chronic inflammatory diseases like rheumatoid arthritis are characterized by a deficit in fully functional regulatory T cells. DNA-methylation inhibitors have previously been shown to promote regulatory T cell responses and, in the present study, we evaluated their potential to ameliorate chronic and acute animal models of rheumatoid arthritis. Of the drugs tested, decitabine was the most effective, producing a sustained therapeutic effect that was dependent on indoleamine 2,3-dioxygenase (IDO) and was associated with expansion of induced regulatory T cells, particularly at the site of disease activity. Treatment with decitabine also caused apoptosis of Th1 and Th17 cells in active arthritis in a highly selective manner. The molecular basis for this selectivity was shown to be ENT1, a nucleoside transporter, which facilitates intracellular entry of the drug and is up-regulated on effector T cells during active arthritis. It was further shown that short-term treatment with decitabine resulted in the generation of a population of regulatory T cells that were able to suppress arthritis upon adoptive transfer. In summary, a therapeutic approach using an approved drug is described that treats active inflammatory disease effectively and generates robust regulatory T cells with the IDO-dependent capacity to maintain remission.

    • The Visual Object Tracking Segmentation VOTS2023 challenge is the eleventh annual tracker benchmarking activity of the VOT initiative. This challenge is the first to merge short-term and long-term as well as single-target and multiple-target tracking with segmentation masks as the only target location specification. A new dataset was created; the ground truth has been withheld to prevent overfitting. New performance measures and evaluation protocols have been created along with a new toolkit and an evaluation server. Results of the presented 47 trackers indicate that modern tracking frameworks are well-suited to deal with convergence of short-term and long-term tracking and that multiple and single target tracking can be considered a single problem. A leaderboard, with participating trackers details, the source code, the datasets, and the evaluation kit are publicly available at the challenge website1

  • Emerging contaminants: A One Health perspective

    • Environmental pollution is escalating due to rapid global development that often prioritizes human needs over planetary health. Despite global efforts to mitigate legacy pollutants, the continuous introduction of new substances remains a major threat to both people and the planet. In response, global initiatives are focusing on risk assessment and regulation of emerging contaminants, as demonstrated by the ongoing efforts to establish the UN's Intergovernmental Science-Policy Panel on Chemicals, Waste, and Pollution Prevention. This review identifies the sources and impacts of emerging contaminants on planetary health, emphasizing the importance of adopting a One Health approach. Strategies for monitoring and addressing these pollutants are discussed, underscoring the need for robust and socially equitable environmental policies at both regional and international levels. Urgent actions are needed to transition toward sustainable pollution management practices to safeguard our planet for future generations.

    • Using 14M psi(2S) events taken with the BESII detector, chi(cJ) -> 2(K+K-) decays are studied. For the four-kaon final state, the branching fractions are B(chi(c0,1,2) ->.2(K+K-)) = (3.48 +/- 0.23 +/- 0.47) x 10(-3), (0.70 +/- 0.13 +/- 0.10) x 10(-3), and (2.17 +/- 0.20 +/- 0.31) x 10(-3). For the phi K+K- final state, the branching fractions, which are measured for the first time, are B(chi(c0,1,2) -> phi K+K-) = (1.03 +/- 0.22 +/- 0.15) x 10(-3), (0.46 +/- 0.16 +/- 0.06) x 10(-3), and (1.67 +/- 0.26 +/- 0.24) x 10(-4). For the phi phi final state, B(chi(c0,2) -> phi phi) = (0.94 +/- 0.21 +/- 0.13) x 10(-3) and (1.70 +/- 0.30 +/- 0.25) x 10(-3).

  • Non-Interpenetrated Single-Crystal Covalent Organic Frameworks

    • Growth of covalent organic frameworks (COFs) as single crystals is extremely challenging. Inaccessibility of open-structured single-crystal COFs prevents the exploration of structure-oriented applications. Herein we report for the first time a non-interpenetrated single-crystal COF, LZU-306, which possesses the open structure constructed exclusively via covalent assembly. With a high void volume of 80 %, LZU-306 was applied to investigate the intrinsic dynamics of reticulated tetraphenylethylene (TPE) as the individual aggregation-induced-emission moiety. Solid-state(2)H NMR investigation has determined that the rotation of benzene rings in TPE, being the freest among the reported cases, is as fast as 1.0x10(4) Hz at 203 K to 1.5x10(7) Hz at 293 K. This research not only explores a new paradigm for single-crystal growth of open frameworks, but also provides a unique matrix-isolation platform to reticulate functional moieties into a well-defined and isolated state.

    • The three-dimensional and temperature-dependent electronic structures of the heavy-fermion superconductor CePt2In7 are investigated. Angle-resolved photoemission spectroscopy using variable photon energy establishes the existence of quasi-two- and three-dimensional Fermi surface topologies. Temperature-dependent 4d-4f on-resonance photoemission spectroscopies data reveal that heavy quasiparticle bands begin to form at a temperature well above the characteristic (coherence) temperature T+. The emergence of low-lying crystal electric field excitation may be responsible for the "relocalization" or the precursor to the establishment of heavy electrons coherence in heavy-fermion compounds. These findings provide critical insight into understanding the hybridization in heavy-fermion systems.

    • We report a photoassisted method to magnetize microcrystal fullerene C-60 at room temperature by exciting it to triplet states via a proper laser radiation and then trapping the spin-polarized states under a strong magnetic field. Novel changes on Raman scattering of the C-60 microcrystals were observed in the presence and absence of the magnetic field. In particular, the Raman spectra were found to exhibit a "hysteresis" phenomenon when the external magnetic field was removed. In light of this, we propose magnetic-field-trapped Raman spectroscopy (MFTRS) and employ first-principle calculations to reproduce the Raman activities of C-60 at different states. Further, MFTRS of the fullerene is demonstrated to originate from its photoassisted magnetization (PAM). The PAM strategy enables the magnetization of materials which consist of only light elements; meanwhile, the MFTRS investigation may open a new research field in Raman spectroscopy.

    • The applicability of surface-enhanced Raman spectroscopy is demonstrated to probe the adsorption behavior of individual molecules on a Ag electrode. High-quality SERS spectra of (R)-di-2-naphthylprolinol (DNP) were obtained from ultradilute solutions (10(-12) M) on the Ag-nanoparticle-modified Ag electrode, which is attributed to the high electromagnetic (EM) effect of the SERS-active system as well as to the strong adsorption and interaction of DNP molecules with Ag. The stable SERS spectra present remarkable potential dependence, which gives evidence for the behavior of individual DNP molecules on the Ag surface. Based on statistical analysis for the probability of DNP molecules located in 'hot spots', we propose an SERS mechanism for individual molecules in the electrode system, in combination with the hot-spot model and orientation of the probemolecules.

    • Nickel oxide (NiO) is considered one of the most promising positive anode materials for electrochromic supercapacitors. Nevertheless, a detailed mechanism of the electrochromic and energy storage process has yet to be unraveled. In this research, the charge storage mechanism of a NiO electrochromic electrode was investigated by combining the in-depth experimental and theoretical analyses. Experimentally, a kinetic analysis of the Li-ion behavior based on the cyclic voltammetry curves reveals the major contribution of surface capacitance versus total capacity, providing fast reaction kinetics and a highly reversible electrochromic performance. Theoretically, our model uncovers that Li ions prefer to adsorb at fcc sites on the NiO(1 1 1) surface, then diffuse horizontally over the plane, and finally migrate in the bulk. More significantly, the calculated theoretical surface capacity (106 mA h g(-1)) accounts for about 77.4% of the total experimental capacity (137 mA h g(-1)), indicating that the surface storage process dominates the whole charge storage, which is in accordance with the experimental results. This work provides a fundamental understanding of transition-metal oxides for application in electrochromic supercapacitors and can also promote the exploration of novel electrode materials for high-performance electrochromic supercapacitors.

  • 2D and 3D Porphyrinic Covalent Organic Frameworks : The Influence of Dimensionality on Functionality

    • The construction of 2D and 3D covalent organic frameworks (COFs) from functional moieties for desired properties has gained much attention. However, the influence of COFs dimensionality on their functionalities, which can further assist in COF design, has never been explored. Now, by selecting designed precursors and topology diagrams, 2D and 3D porphyrinic COFs (2D-PdPor-COF and 3D-PdPor-COF) are synthesized. By model building and Rietveld refinement of powder X-ray diffraction, 2D-PdPor-COF crystallizes as 2D sheets while 3D-PdPor-COF adopts a five-fold interpenetrated pts topology. Interestingly, compared with 2D-PdPor-COF, 3D-PdPor-COF showed interesting properties, including 1) higher CO2 adsorption capacity; 2) better photocatalytic performance; and 3) size-selective photocatalysis. Based on this study, we believe that with the incorporation of functional moieties, the dimensionality of COFs can definitely influence their functionalities.

    • Exploring high-efficient oxygen evolution reaction (OER) catalysts is critical for producing clean H-2 fuel through water splitting. Co and P involved Co-Pi and cobalt phosphide OER catalysts attracted tremendous attention, while limited report on cobalt phosphite as an OER catalyst. Herein, a novel cobalt phosphite nanowires with microporous 1D channel have been succsessfully discovered with remarkable catalytic performance of a low overpotential of (similar to)340 mV and Tafel slope of 60 mV dec(-1) at 10 mA cm(-2) at pH 14. Through comprehensive structural characterization, the effective species has been clearly illustrated to be the in-situ formed heterostructural Co-11(HPO3)(8)(OH)(6)/Co-11(PO3)(8)O-6 core-shell structure nanowires. This work fills the gap for exploring high efficient cobalt phosphites OER catalysts under akaline condition and paves the way for exploring novel high efficiency waster splitting catalysts.

    • Recently, molecular electronics has become increasingly important. By applying the hybrid density functional theory coupled with the Green's function method, the current-voltage characteristics of the molecular junctions composed of gold-porphyrin-gold and gold-copper porphyrin-gold were investigated. The role of the metal coordination effect in organic molecular electron transport was highlighted. Although the thresholds of the bias voltage for both molecules were almost the same, approximately 0.9 V, the metal compound showed a larger increase in current because of the metal-coordination-enhanced molecule-electrode coupling in the frontier molecular orbitals.

    • Precious metal Pt-based electrocatalysts have been widely used in the methanol catalytic oxidation anodes in direct methanol fuel cells. However, decreasing their cost and improving their efficiency and durability have still been challenging. Herein, ternary PtPdCu nanocatalysts are synthesized through a facile one-step hydrothermal synthesis method. When KI is present with a suitable amount in the synthesis, PtPdCu nanospheres with surface-embedded CuI clusters (CuI/PtPdCu) are fabricated. However, without KI, the prepared PtPdCu catalysts show a distinct hollow structure (h-PtPtCu). CuI/PtPdCu displays the highest specific activity with enhancement 4 times higher than commercial Pt/C for the methanol oxidation reaction in an alkaline medium. The superior activity can be attributed to two aspects: 1) the electronic effect originating from the highly alloyed PtPdCu; 2) the synergetic effect resulting from surface inlaid CuI clusters, which can promote the CO intermediate removal. Furthermore, because of the stable Pt-Pd-rich surface and its special linked hollow structure, the h-PtPtCu catalyst exhibits good durability with only a 3.6% decay in the specific activity.

  • Endothelial cell heterogeneity and microglia regulons revealed by a pig cell landscape at single-cell level

    • Pigs are valuable large animal models for biomedical and genetic research, but insights into the tissue- and cell-type-specific transcriptome and heterogeneity remain limited. By leveraging single-cell RNA sequencing, we generate a multiple-organ single-cell transcriptomic map containing over 200,000 pig cells from 20 tissues/organs. We comprehensively characterize the heterogeneity of cells in tissues and identify 234 cell clusters, representing 58 major cell types. In-depth integrative analysis of endothelial cells reveals a high degree of heterogeneity. We identify several functionally distinct endothelial cell phenotypes, including an endothelial to mesenchymal transition subtype in adipose tissues. Intercellular communication analysis predicts tissue- and cell type-specific crosstalk between endothelial cells and other cell types through the VEGF, PDGF, TGF-beta, and BMP pathways. Regulon analysis of single-cell transcriptome of microglia in pig and 12 other species further identifies MEF2C as an evolutionally conserved regulon in the microglia. Our work describes the landscape of single-cell transcriptomes within diverse pig organs and identifies the heterogeneity of endothelial cells and evolutionally conserved regulon in microglia.

  • Controllable direct-syntheses of delaminated MWW-type zeolites

    • A method for the direct syntheses of partially and fully delaminated MWW zeolites is reported herein. Two organic amines were introduced into the hydrothermal synthetic system: hexamethyleneimine (HMI), which acted as the structure-directing agent for the MWW layered structure; and dicyclohexylamine (DCHA), in the role of an in-situ delaminating agent. By varying the amount of DCHA, partially and fully delaminated MWW zeolites having two MWW structure layers and one single layer, respectively, were obtained. These were denoted as SCM-1 (Sinopec Composite Material) and SCM-6, respectively. The delaminated materials possess ultra-large external surface areas, and the transmission electron microscopy images illustrated their layered nature. In the reaction of liquid phase benzene alkylation with ethylene, SCM-1, the double-layered MWW zeolite, exhibited far superior catalytic performance compared to zeolite MCM-22.

    • The marriage of dynamic covalent chemistry (DCC) and coordination chemistry is a powerful tool for assembling complex architectures from simple building units. Recently, the synthesis of woven covalent organic frameworks (COFs) with topologically fascinating structures has been achieved using this approach. However, the scope is highly limited and there is a need to discover new pathways that can assemble covalently linked organic threads into crystalline frameworks. Herein, we have identified branching pathways leading to the assembly of three-dimensional (3D) woven COFs or one-dimensional (1D) metallo-COFs (mCOFs), where the mechanism is underpinned by the absence or presence of ligand exchange.
