    • Changes in the latitudinal position and strength of the Southern Hemisphere westerlies (SHW) are thought to be tightly coupled to important climate processes, such as cross-equatorial heat fluxes, Atlantic Meridional Overturning Circulation (AMOC), the bipolar seesaw, Southern Ocean ventilation and atmospheric CO2 levels. However, many uncertainties regarding magnitude, direction, and causes and effects of past SHW shifts still exist due to lack of suitable sites and scarcity of information on SHW dynamics, especially from the last glacial. Here we present a detailed hydroclimate multiproxy record from a 36.4-18.6 kyr old lake sediment sequence on Nightingale Island (NI). It is strategically located at 37ĝF S in the central South Atlantic (SA) within the SHW belt and situated just north of the marine Subtropical Front (SF). This has enabled us to assess hydroclimate changes and their link to the regional climate development as well as to large-scale climate events in polar ice cores. The NI record exhibits a continuous impact of the SHW, recording shifts in both position and strength, and between 36 and 31 ka the westerlies show high latitudinal and strength-wise variability possibly linked to the bipolar seesaw. This was followed by 4 kyr of slightly falling temperatures, decreasing humidity and fairly southerly westerlies. After 27 ka temperatures decreased 3-4 ĝC, marking the largest hydroclimate change with drier conditions and a variable SHW position. We note that periods with more intense and southerly-positioned SHW seem to be related to periods of increased CO2 outgassing from the ocean, while changes in the cross-equatorial gradient during large northern temperature changes appear as the driving mechanism for the SHW shifts. Together with coeval shifts of the South Pacific westerlies, our results show that most of the Southern Hemisphere experienced simultaneous atmospheric circulation changes during the latter part of the last glacial. Finally we can conclude that multiproxy lake records from oceanic islands have the potential to record atmospheric variability coupled to large-scale climate shifts over vast oceanic areas..

  • Archaeal community changes in Lateglacial lake sediments: Evidence from ancient DNA

    • The Lateglacial/early Holocene sediments from the ancient lake at Hässeldala Port, southern Sweden provide an important archive for the environmental and climatic shifts at the end of the last ice age and the transition into the present Interglacial. The existing multi-proxy data set highlights the complex interplay of physical and ecological changes in response to climatic shifts and lake status changes. Yet, it remains unclear how microorganisms, such as Archaea, which do not leave microscopic features in the sedimentary record, were affected by these climatic shifts. Here we present the metagenomic data set of Hässeldala Port with a special focus on the abundance and biodiversity of Archaea. This allows reconstructing for the first time the temporal succession of major Archaea groups between 13.9 and 10.8 ka BP by using ancient environmental DNA metagenomics and fossil archaeal cell membrane lipids. We then evaluate to which extent these findings reflect physical changes of the lake system, due to changes in lake-water summer temperature and seasonal lake-ice cover. We show that variations in archaeal composition and diversity were related to a variety of factors (e.g., changes in lake water temperature, duration of lake ice cover, rapid sediment infilling), which influenced bottom water conditions and the sediment-water interface. Methanogenic Archaea dominated during the Allerød and Younger Dryas pollen zones, when the ancient lake was likely stratified and anoxic for large parts of the year. The increase in archaeal diversity at the Younger Dryas/Holocene transition is explained by sediment infilling and formation of a mire/peatbog.

  • The fate of fixed nitrogen in marine sediments with low organic loading : an in situ study

    • Over the last decades, the impact of human activities on the global nitrogen (N) cycle has drastically increased. Consequently, benthic N cycling has mainly been studied in anthropogenically impacted estuaries and coasts, while in oligotrophic systems its understanding is still scarce. Here we report on benthic solute fluxes and on rates of denitrification, anammox, and dissimilatory nitrate reduction to ammonium (DNRA) studied by in situ incubations with benthic chamber landers during two cruises to the Gulf of Bothnia (GOB), a cold, oligotrophic basin located in the northern part of the Baltic Sea. Rates of N burial were also inferred to investigate the fate of fixed N in these sediments. Most of the total dissolved fixed nitrogen (TDN) diffusing to the water column was composed of organic N. Average rates of dinitrogen (N-2) production by denitrification and anammox (range: 53-360 mu mol Nm(-2) day(-1)) were comparable to those from Arctic and subarctic sediments worldwide (range: 34-344 mu mol Nm(-2) day(-1)). Anammox accounted for 18-26% of the total N2 production. Absence of free hydrogen sulfide and low concentrations of dissolved iron in sediment pore water suggested that denitrification and DNRA were driven by organic matter oxidation rather than chemolithotrophy. DNRA was as important as denitrification at a shallow, coastal station situated in the northern Bothnian Bay. At this pristine and fully oxygenated site, ammonium regeneration through DNRA contributed more than one-third to the TDN efflux and accounted, on average, for 45% of total nitrate reduction. At the offshore stations, the proportion of DNRA in relation to denitrification was lower (0-16% of total nitrate reduction). Median value and range of benthic DNRA rates from the GOB were comparable to those from the southern and central eutrophic Baltic Sea and other temperate estuaries and coasts in Europe. Therefore, our results contrast with the view that DNRA is negligible in cold and well-oxygenated sediments with low organic carbon loading. However, the mechanisms behind the variability in DNRA rates between our sites were not resolved. The GOB sediments were a major source (237 kt yr(-1), which corresponds to 184% of the external N load) of fixed N to the water column through recycling mechanisms. To our knowledge, our study is the first to document the simultaneous contribution of denitrification, DNRA, anammox, and TDN recycling combined with in situ measurements.

  • Modes of carbon fixation in an arsenic and CO2-rich shallow hydrothermal ecosystem

    • The seafloor sediments of Spathi Bay, Milos Island, Greece, are part of the largest arsenic-CO2-rich shallow submarine hydrothermal ecosystem on Earth. Here, white and brown deposits cap chemically distinct sediments with varying hydrothermal influence. All sediments contain abundant genes for autotrophic carbon fixation used in the Calvin-Benson-Bassham (CBB) and reverse tricaboxylic acid (rTCA) cycles. Both forms of RuBisCO, together with ATP citrate lyase genes in the rTCA cycle, increase with distance from the active hydrothermal centres and decrease with sediment depth. Clustering of RuBisCO Form II with a highly prevalent Zetaproteobacteria 16S rRNA gene density infers that iron-oxidizing bacteria contribute significantly to the sediment CBB cycle gene content. Three clusters form from different microbial guilds, each one encompassing one gene involved in CO2 fixation, aside from sulfate reduction. Our study suggests that the microbially mediated CBB cycle drives carbon fixation in the Spathi Bay sediments that are characterized by diffuse hydrothermal activity, high CO2, As emissions and chemically reduced fluids. This study highlights the breadth of conditions influencing the biogeochemistry in shallow CO2-rich hydrothermal systems and the importance of coupling highly specific process indicators to elucidate the complexity of carbon cycling in these ecosystems.

  • Pyrococcus kukulkanii sp nov., a hyperthermophilic, piezophilic archaeon isolated from a deep-sea hydrothermal vent

    • A novel hyperthermophilic, piezophilic, anaerobic archaeon, designated NCB100(T), was isolated from a hydrothermal vent flange fragment collected in the Guaymas basin at the hydrothermal vent site named 'Rebecca's Roost' at a depth of 1997 m. Enrichment and isolation were performed at 100 degrees C under atmospheric pressure. Cells of strain NCB100(T) were highly motile, irregular cocci with a diameter of -1 mu m. Growth was recorded at temperatures between 70 and 112 degrees C (optimum 105 degrees C) and hydrostatic pressures of 0.1-80 MPa (optimum 40-50 MPa). Growth was observed at pH 3.5-8.5 (optimum pH 7) and with 1.5-7% NaCl (optimum at 2.5-3 %). Strain NCB100(T) was a strictly anaerobic chemo-organoheterotroph and grew on complex proteinaceous substrates such as yeast extract, peptone and tryptone, as well as on glycogen and starch. Elemental sulfur was required for growth and was reduced to hydrogen sulfide. The fermentation products from complex proteinaceous substrates were CO2 and H-2. The G+C content of the genomic DNA was 41.3 %. Phylogenetic analysis of the 16S rRNA gene sequence revealed that strain NCB100(T) belongs to the genus Pyrococcus, showing 99% similarity with the other described species of the genus Pyrococcus. On the basis of physiological characteristics, DNA G+C content, similarity level between ribosomal proteins and an average nucleotide identity value of 79 %, strain NCB100(T) represents a novel species for which the name Pyrococcus kukulkanii sp. nov. is proposed. The type strain is NCB100(T) (=DSM 101590(T) =Souchotheque de Bretagne BG1337(T)).

    • An early Quaternary shallow submarine hydrothermal iron formation (IF) in the Cape Vani sedimentary basin (CVSB) on Milos Island, Greece, displays banded rhythmicity similar to Precambrian banded iron formation (BIF). Field-wide stratigraphic and biogeochemical reconstructions show two temporal and spatially isolated iron deposits in the CVSB with distinct sedimentological character. Petrographic screening suggests the presence of a photoferrotrophic-like microfossil-rich IF (MFIF), accumulated on a basement consisting of andesites in a ∼ 150m wide basin in the SW margin of the basin. A banded nonfossiliferous IF (NFIF) sits on top of the Mn-rich sandstones at the transition to the renowned Mn-rich formation, capping the NFIF unit. Geochemical data relate the origin of the NFIF to periodic submarine volcanism and water column oxidation of released Fe(II) in conditions predominated by anoxia, similar to the MFIF. Raman spectroscopy pairs hematite-rich grains in the NFIF with relics of a carbonaceous material carrying an average δ13Corg signature of ∼ −25‰. A similar δ13Corg signature in the MFIF could not be directly coupled to hematite by mineralogy. The NFIF, which postdates large-scale Mn deposition in the CVSB, is composed primarily of amorphous Si (opal-SiO2 ⋅ nH2O) while crystalline quartz (SiO2) predominates the MFIF. An intricate interaction between tectonic processes, changing redox, biological activity, and abiotic Si precipitation are proposed to have collectively formed the unmetamorphosed BIF-type deposits in a shallow submarine volcanic center. Despite the differences in Precambrian ocean–atmosphere chemistry and the present geologic time, these formation mechanisms coincide with those believed to have formed Algoma-type BIFs proximal to active seafloor volcanic centers.

    • The Bering Strait connects the Arctic and Pacific oceans and separates the North American and Asian landmasses. The presently shallow (similar to 53 m) strait was exposed during the sea level lowstand of the last glacial period, which permitted human migration across a land bridge today referred to as the Bering Land Bridge. Proxy studies (stable isotope composition of foraminifera, whale migration into the Arctic Ocean, mollusc and insect fossils and paleobotanical data) have suggested a range of ages for the Bering Strait reopening, mainly falling within the Younger Dryas stadial (12.9-11.7 cal ka BP). Here we provide new information on the deglacial and post-glacial evolution of the Arctic-Pacific connection through the Bering Strait based on analyses of geological and geophysical data from Herald Canyon, located north of the Bering Strait on the Chukchi Sea shelf region in the western Arctic Ocean. Our results suggest an initial opening at about 11 cal ka BP in the earliest Holocene, which is later than in several previous studies. Our key evidence is based on a well-dated core from Herald Canyon, in which a shift from a near-shore environment to a Pacific-influenced open marine setting at around 11 cal ka BP is observed. The shift corresponds to meltwater pulse 1b (MWP1b) and is interpreted to signify relatively rapid breaching of the Bering Strait and the submergence of the large Bering Land Bridge. Although the precise rates of sea level rise cannot be quantified, our new results suggest that the late deglacial sea level rise was rapid and occurred after the end of the Younger Dryas stadial.

    • Understanding microbial mediation in sediment-hosted Mn deposition has gained importance in low-temperature ore genesis research. Here we report Mn oxide ores dominated by todorokite, vernadite, hollandite, and manjiroite, which cement Quaternary microbially induced sedimentary structures (MISS) developed along bedding planes of shallow-marine to tidal-flat volcaniclastic sandstones/sandy tuffs, Cape Vani paleo-hydrothermal vent field, Milos, Greece. This work aims to decipher the link between biological Mn oxide formation, low-T hydrothermalism, and, growth and preservation of Mn-bearing MISS (MnMISS). Geobiological processes, identified by microtexture petrography, scanning and transmission electron microscopy, lipid biomarkers, bulk- and lipid-specific δ13Corganic composition, and field data, and, low-temperature hydrothermal venting of aqueous Mn2+ in sunlit shallow waters, cooperatively enabled microbially-mediated Mn (II) oxidation and biomineralization. The MnMISS biomarker content and δ13Corg signatures strongly resemble those of modern Mn-rich hydrothermal sediments, Milos coast. Biogenic and syngenetic Mn oxide precipitation established by electron paramagnetic resonance (EPR) spectroscopy and petrography, combined with hydrothermal fluid flow-induced pre-burial curing/diagenesis, may account for today’s crystalline Mn oxide resource. Our data suggests that MISS are not unique to cyanobacteria mats. Furthermore, microbial mats inhabited by aerobic methanotrophs may have contributed significantly to the formation of the MnMISS, thus widening the spectrum of environments responsible for marine Mn biometallogenesis.

  • Complexity of the serpentinization lipidome

    • Energy from serpentinization processes potentially fuelled the origin of life on Earth. Thus, if serpentinite-impacted sites facilitate microbial habitability, it is important to understand the source and retention of biological signatures in them. Investigating biological signatures in terrestrial serpentinite-impacted environments is also essential for interpreting molecular signature preservation on extraterrestrial bodies. To expand knowledge on the types of biological signatures derived from microorganisms in the serpentinization microbiome, mass spectrometry analysis was performed on Chimaera serpentinite rocks from Antalya Province, Turkey. The presence of diverse types of core and intact glycerol alkyl diether and tetraether lipids in all samples indicates that methanogenic archaea were present, but not in equal abundance in all rock types (carbonate or brucite) or locations (inside and outside the rock). Bacterial derived brGDGTs (branched glycerol dialkyl glycerol tetraethers) and phospholipid fatty acids were also identified in all samples. Comparing targeted lipid data from this study with other studies and locations reveals consistency across the serpentinization lipidome. Additionally, an untargeted lipidomics method was tested on the intact polar lipid (IPL) extracts, This resulted in detection of a wide range of intact lipids, many requiring further elucidation (e.g., diacyl glycerol IPLs, triacylglycerols, glycosphingolipids, sterol esters and cardiolipins), suggesting that there may be diverse bacterial, archaeal and fungal communities in the different rock samples, highlighting the additional complexity of the serpentinization lipidome dark matter.

  • Separation of Branched and Isoprenoid Glycerol Dialkyl Glycero Tetraether (GDGT) Isomers in Peat Soils and Marine Sediments Using Reverse Phase Chromatography

    • Over the last decade, glycerol dialkyl glycerol tetraethers (GDGTs) have become one of the most investigated lipid classes in marine and terrestrial organic geochemical research. GDGTs are microbial membrane core lipids biosynthesized as multiple homolog series of isoprenoid or methyl-branched isomers [isoprenoid glycerol dialkyl glycerol tetraethers (isoGDGTs) and Branched GDGTs (brGDGTs), respectively], whose relative abundance depend on a range of environmental parameters, including temperature. This has led to the development of GDGT-based temperature proxies. A key aspect in the analysis of GDGTs and the further development of their use as environmental proxies is good chromatographic separation of the full range of structural and stereo-isomers, with potential for discovery of novel GDGT variants. Several HPLC methods have been developed to this extent, but partial co-elution of GDGTs remains an issue despite long run times. In this study, we investigate the effects of different types of reverse phase (RP) chromatography on the separation of GDGT isomers. We found that the use of a Kinetex C18-XB column gives good separation of isoGDGT isomers in comparison to the recently developed double column HILIC analysis operated in normal phase (NP) and has a shorter run time. In marine samples, the regularly reported isoprenoid GDGTs separated in a similar way as in NP, however an earlier eluting group was observed to elute with the crenarchaeol isomer used in the TEX86 proxy. In a Swedish peat bog sample, a large range of isoGDGT isomers were observed. We observed a range of brGDGT isomers in several samples often with near baseline separation. Exact identification of all these isomers remained elusive, due to the different mechanism of separation in RP, and the complexity of the brGDGT family. The C18-XB method is rapid and versatile and can be set up on either low-pressure HPLC systems (max 400 bar) with a sample run time of 25 min for brGDGTs and 45 min to include isoGDGTs. On UHPLC-MS systems (>600 bar) the sample run time is reduced to 15 min. Most importantly, the C18-XB method presented here gives unusual separation of both isoprenoid and brGDGTs and could be a useful tool for the further elucidation of the biological sources and environmental factors that play a role in the production of different GDGT isomers.

  • Rare Earth element enriched birnessite in water-bearing fractures, the Ytterby mine, Sweden

    • Characterization of a black substance exuding from fractured bedrock in a subterranean tunnel revealed a secondary manganese oxide mineralisation exceptionally enriched in rare earth elements (REE). Concentrations are among the highest observed in secondary ferromanganese precipitates in nature. The tunnel is located in the unsaturated zone at shallow depth in the former Ytterby mine, known for the discovery of yttrium, scandium, tantalum and five rare earth elements.Elemental analysis and X-ray diffraction of the black substance establish that the main component is a manganese oxide of the birnessite type. Minor fractions of calcite, other manganese oxides, feldspars, quartz and about 1% organic matter were also found, but no iron oxides were identified. The Ytterby birnessite contains REE, as well as calcium, magnesium and traces of other metals. The REE, which constitute 1% of the dry mass and 2% of the metal content, are firmly included in the mineral structure and are not released by leaching at pH 1.5 or higher. A strong preference for the trivalent REE over divalent and monovalent metals is indicated by concentration ratios of the substance to fracture water. The REE-enriched birnessite has the general formula Mx(Mn3+,Mn4+)2O4·(H2O)n with M = (0.37–0.41) Ca + 0.02 (REE + Y), 0.04 Mg and (0.02–0.03) other metals, and with [Mn3+]/[Mn4+] = 0.86–1.00.The influence of microorganisms on the accumulation of this REE enriched substance is demonstrated by electron paramagnetic resonance spectroscopy. Results show that it is composed of two or more manganese phases, one of which has a biogenic signature. In addition, the occurrence of C31 to C35 extended side chain hopanoids among the identified lipid biomarkers combined with the absence of ergosterol, a fungal lipid biomarker, indicate that the in-situ microbial community is bacterial rather than fungal.

  • REE-Enriched Mn-Oxide Precipitates in Water-Bearing Fractures in the Ytterby Mine, Sweden

    • The Ytterby mine, Sweden, is known for the discovery of eight elements, including yttrium and five of the rare earth elements (REE). The mine was in operation from 1750 to 1933 and was after closure used as a storage depot for fuel from the 1950s to 1995. A tunnel was opened in the 1950s through the bedrock into the mine to allow access to the storage depot. Recent water monitoring campaigns (20122015) in the mine revealed a black substance (denoted YBS) in some fractures opening into the tunnel. Analysis of the YBS (elemental analysis, phase analysis by XRD, SEM with energy dispersive X-ray spectrometry, IR-and EPR-spectroscopy, preferential leaching at pH 4) showed that the main mineral component of the YBS is the manganese oxide birnessite. Also minor quantities of other less well defined manganese oxides were found, as well as silicates (quartz grains, possibly feldspar grains) and calcite. Birnessite has typically the composition Mx(Mn3+, Mn(2)(4+))O(4)xAq, with M= Na, Ca and x= 0.5. The birnessite component in YBS had a Mn3+/Mn4+ ratio of 1.04/0.96 with M = 0.42 Ca + 0.03 (REE+Y), 0.03 Mg and 0.03 other metals. All of these metals were firmly associated with the structure, since no release was observed at pH 4, except for significant fractions of the total Na, Mg, Ca-contents. Thus, REE+Y correspond to 1% of the total YBS mass and up to 3% of the metal content in the birnessite phase. This corresponds to an REE enrichment factor of the order 106 (YBS-birnessite/ fracture water). Birnessite with a substantial fraction of REE in the lattice has not previously been reported. The formation of birnessite is a microbial process. Identification of the microorganisms present in the Ytterby system is in progress.
