    • The transient receptor potential melastin 8 ion channel (TRPM8) is implicated in bladder sensing but limited information on TRPM8 antagonists in bladder overactivity is available. This study characterizes a new TRPM8-selective antagonist (DFL23448 [5-(2-ethyl-2H-tetrazol-5-yl)-2-(3-fluorophenyl)-1,3-thiazol-4-ol]) and evaluates it in cold-induced behavioral tests and tests on bladder function and experimental bladder overactivity in vivo in rats. DFL23448 displayed IC50 values of 10 and 21 nM in hTRPM8 human embryonic kidney 293 cells activated by Cooling Agent 10 or cold, but it had limited activity (IC50 > 10 mu M) at transient receptor potential vanilloids TRPV1, TRPA1, or TRPV4 or at various G protein-coupled receptors. In rats, DFL23448 administered intravenously or orally had a half-life of 37 minutes or 4.9 hours, respectively. DLF23448 (10 mg/kg i.v.) reduced icilin-induced "wet dog-like" shakes in rats. Intravesical DFL23448 (10 mg/l), but not vehicle, increased micturition intervals, micturition volume, and bladder capacity. During bladder overactivity by intravesical prostaglandin E-2 (PGE(2)), vehicle controls exhibited reductions in micturition intervals, micturition volumes, and bladder capacity by 37%-39%, whereas the same parameters only decreased by 12%-15% (P < 0.05-0.01 versus vehicle) in DFL23448-treated rats. In vehicle-treated rats, but not in DFL23448-treated rats, intravesical PGE(2) increased bladder pressures. Intravenous DFL23448 at 10 mg/kg, but not 1 mg/kg DFL23448 or vehicle, increased micturition intervals, micturition volumes, and bladder capacity. During bladder overactivity by intravesical PGE(2), micturition intervals, micturition volumes, and bladder capacity decreased in vehicle- and 1 mg/kg DFL23448-treated rats, but not in 10 mg/kg DFL23448-treated rats. Bladder pressures increased less in rats treated with DFL23448 10 mg/kg than in vehicle-or 1 mg/kg DFL23448-treated rats. DFL23448 (10 mg/kg i.v.), but not vehicle, prevented cold stress-induced bladder overactivity. Our results support a role for bladder TRPM8-mediated signals in experimental bladder overactivity.

  • Dietary Diversity and Its Association with Diet Quality and Health Status of European Children, Adolescents, and Adults : Results from the I.Family Study

    • Dietary diversity (DD) plays a crucial role in fostering high-quality diets, but its association with health outcomes, particularly body adiposity and non-communicable diseases (NCDs), is inconsistent. This may be due to a lack of a standardized method for estimating DD. Our study investigates the association between two DD indices, namely the dietary diversity score (DDS) and food variety score (FVS), and anthropometric measures, biochemical parameters, and diet quality in a large population sample from the I.Family study across research centers in eight European countries. In our cross-sectional analysis of 3035 participants, DDSs varied among countries, with a higher prevalence in the third DDS tertile among those with higher education. DDS showed a positive association with diet quality across all age groups. Higher DDS tertile individuals showed increased fiber, fruit, and vegetable intake, greater meal frequency, and lower ultra-processed food consumption. No relevant biochemical differences were observed across DDS tertiles, and a higher DDS was associated with lower overweight/obesity prevalence only in adults. No significant associations were found with FVS. Our findings emphasize the need to consider food groups for a more accurate estimation of diet quality. This aligns with studies suggesting DDS alone is not an independent risk factor for obesity in children and adolescents. Public health programs should prioritize food diversity to promote improved nutrition and overall well-being in communities.

    • Purpose: We assessed whether spinal inhibition of the cannabinoid degrading enzyme FAAH would have urodynamic effects in normal rats and rats with bladder overactivity induced by partial urethral obstruction or prostaglandin E2. We also determined the expression of FAAH, and the cannabinoid receptors CB1 and CB2 in the sacral spinal cord. Materials and Methods: We used 44 rats for functional (cystometry) and Western blot experiments. The FAAH inhibitor oleoyl ethyl amide (3 to 300 nmol) was administered intrathecally (subarachnoidally) or intravenously. The expression of FAAH and CB1/CB2 receptors was determined by Western blot. Results: Oleoyl ethyl amide given intrathecally affected micturition in normal rats and rats with bladder overactivity but effects were more pronounced in the latter. In normal rats oleoyl ethyl amide only decreased micturition frequency, while it decreased frequency and bladder pressures in rats with bladder overactivity. Intravenous oleoyl ethyl amide (3 to 300 nmol) had no urodynamic effect. FAAH and CB1/CB2 receptors were expressed in the rat sacral spinal cord. The expression of CB1/CB2 receptors but not FAAH was higher in obstructed than in normal rats. Conclusions: FAAH inhibition in the sacral spinal cord by oleoyl ethyl amide resulted in urodynamic effects in normal rats and rats with bladder overactivity. The spinal endocannabinoid system may be involved in normal micturition control and it appears altered when there is bladder overactivity.

    • Background: Gonadotropin-releasing hormone (GnRH) antagonists have been reported to have beneficial effects on lower urinary tract symptoms in patients with benign prostatic hyperplasia. Objective: Our aim was to investigate the effects of ganirelix, a GnRH receptor antagonist, on bladder function and detrusor overactivity (DO) in female rats. Design, setting, and participants: Female Sprague-Dawley rats received 2 wk of daily systemic (0.1 mg/kg) or acute intravesical administration (IVES; 0.14 mg/l or 1.4 mg/l) ganirelix or vehicle (controls). Measurements: Assessments were obtained using cystometry in awake rats, organ bath studies, enzyme-linked immunosorbent assay, and western blot (WB). Results and limitations: Luteinising hormone levels were lower in rats treated systemically with ganirelix than in controls. No differences were observed in body or bladder weights. Micturition interval (MI), micturition volume (MV), residual volume, and bladder capacity (BC) were similar in both groups at baseline. No differences in urodynamic pressure parameters were observed between groups at baseline. Intravesical prostaglandin E-2 reduced MI, MV, and BC, and it increased basal pressure (BP), threshold pressure (TP), flow pressure (FP), and maximum pressure (MP) in all rats. MI, MV, and BC were reduced by 43% +/- 4%, 50% +/- 4%, and 43% +/- 4% (controls) versus 22% +/- 3%, 23% +/- 3%, and 21% +/- 3% (ganirelix-treated rats; p andlt; 0.001). TP and FP increased by 38% +/- 8% and 30% +/- 4% (controls) versus 16% +/- 7% and 16% +/- 5% (ganirelix; p andlt; 0.05). The maximal force of contractions for carbachol was larger in detrusor from ganirelix-treated rats (231% vs 177% of 60 mM K+-induced contractions). At 0.14 mg/l, but not 0.14 mg/l, IVES ganirelix increased MI, MV, and BC and decreased BP, TP, FP, and MP. In vitro, ganirelix had no effect on detrusor function. The gonadotropin-releasing hormone receptor was expressed (by WB) in the bladder mucosa. Conclusions: Systemic treatment with ganirelix counteracted experimental DO in female rats. Because bladder preparations from these rats exhibited larger contractions to carbachol and because intravesical ganirelix affected both micturition intervals and urodynamic pressure profiles, a peripheral site of action of ganirelix in the urinary bladder cannot be excluded.

    • Despite the encouraging results of the innovative therapeutic treatments, complete remission is uncommon in patients affected by chronic lymphocytic leukaemia, which remains an essentially incurable disease. Recently, clinical trials based on BH3-mimetic drugs showed positive outcomes in subjects with poor prognostic features. However, resistance to treatments occurs in a significant number of patients. We previously reported that the multi-kinase inhibitor quercetin, a natural flavonol, restores sensitivity to ABT-737, a BH3-mimetic compound, in both leukemic cell lines and B-cells isolated from patients. To identify the molecular target of quercetin, we employed a new cell line, HG3, obtained by immortalization of B-cells from a chronic lymphocytic leukaemia patient at the later stage of disease. We confirmed that quercetin in association with ABT-737 synergistically enhances apoptosis in HG3 (combination index < 1 for all fractions affected). We also reported that the cellular uptake of quercetin is extremely rapid, with an intracellular concentration of about 38.5 ng/106 cells, after treatment with 25 μM for 5 min. We demonstrated that the activity of protein kinase CK2, which positively triggers PI3K/Akt pathway by inactivating PTEN phosphatase, is inhibited by quercetin immediately after its addition to HG3 cells (0-2 min). PI3K activity was also inhibited by quercetin within 60 min from the treatment. The combined inhibition of CK2 and PI3K kinase activities by quercetin restored ABT-737 sensitivity and increased lethality in human leukemia cells.

    • Angular correlations between charged trigger and associated particles are measured by the ALICE detector in p-Pb collisions at a nucleon-nucleon centre-of-mass energy of 5.02 TeV for transverse momentum ranges within 0.5 < P-T,P-assoc < P-T,P-trig < 4 GeV/c. The correlations are measured over two units of pseudorapidity and full azimuthal angle in different intervals of event multiplicity, and expressed as associated yield per trigger particle. Two long-range ridge-like structures, one on the near side and one on the away side, are observed when the per-trigger yield obtained in low-multiplicity events is subtracted from the one in high-multiplicity events. The excess on the near-side is qualitatively similar to that recently reported by the CMS Collaboration, while the excess on the away-side is reported for the first time. The two-ridge structure projected onto azimuthal angle is quantified with the second and third Fourier coefficients as well as by near-side and away-side yields and widths. The yields on the near side and on the away side are equal within the uncertainties for all studied event multiplicity and p(T) bins, and the widths show no significant evolution with event multiplicity or p(T).     • Accurate and precise rainfall records are crucial for hydrological applications and water resources management. The accuracy and continuity of ground-based time series rely on the density and distribution of rain gauges over territories. In the context of a decline of rain gauge distribution, how to optimize and design optimal networks is still an unsolved issue. In this work, we present a method to optimize a ground-based rainfall network using satellite-based observations, maximizing the information content of the network. We combine Climate Prediction Center MORPhing technique (CMORPH) observations at ungauged locations with an existing rain gauge network in the Rio das Velhas catchment, in Brazil. We use a greedy ranking algorithm to rank the potential locations to place new sensors, based on their contribution to the joint entropy of the network. Results show that the most informative locations in the catchment correspond to those areas with the highest rainfall variability and that satellite observations can be successfully employed to optimize rainfall monitoring networks.

  • On the Use of Satellite Rainfall Data to Design a Dam in an Ungauged Site

    • The estimation of the design peak discharge is crucial for the hydrological design of hydraulic structures. A commonly used approach is to estimate the design storm through the intensity-duration-area-frequency (IDAF) curves and then use it to generate the design discharge through a hydrological model. In ungauged areas, IDAF curves and design discharges are derived throughout regionalization studies, if any exist for the area of interest, or from using the hydrological information of the closest and most similar gauged place. However, many regions around the globe remain ungauged or are very poorly gauged. In this regard, a unique opportunity is provided by satellite precipitation products developed and improved in the last decades. In this paper, we show weaknesses and potentials of satellite data and, for the first time, we evaluate their applicability for design purposes. We employ CMORPH-Climate Prediction Center MORPHing technique satellite precipitation estimates to build IDAF curves and derive the design peak discharges for the Pietrarossa dam catchment in southern Italy. Results are compared with the corresponding one provided by a regionalization study, i.e., VAPI-VAlutazione delle Piene in Italia project, usually used in Italy in ungauged areas. Results show that CMORPH performed well for the estimation of low duration and small return periods storm events, while for high return period storms, further research is still needed.

    • From state-based developmentalism to community-based initiatives to market-based conservation, the Brazilian Amazon has been a laboratory of development interventions for over 50 years. The region is now confronting a devastating COVID-19 pandemic amid renewed environmental pressures and increasing social inequities. While these forces are shaping the present and future of the region, the Amazon has also become an incubator of local innovations and efforts confronting these pressures. Often overlooked, place-based initiatives involving individual and collective-action have growing roles in promoting regional sustainability. We review the history of development interventions influencing the emergence of place-based initiatives and their potential to promoting changes in productive systems, value-aggregation and market-access, and governance arrangements improving living-standards and environmental sustainability. We provide examples of initiatives documented by the AGENTS project, contextualizing them within the literature. We reflect on challenges and opportunities affecting their trajectories at this critical juncture for the future of the region.

    • Background: A medical treatment for urethral stricture (US) is not yet available. Objective: To evaluate if local injection of human adipose tissue-derived stem cells (hADSC) prevents urethral fibrosis in a rat model of US. Design, setting, and participants: Male rats were divided into three groups: sham, US, and hADSC (n = 12 each). Sham rats received a vehicle injection in the urethral wall. US and hADSCs were incised and injected with the fibrosis-inducer transforming growth factor-β1 in the urethral wall. Intervention: One day later, hADSCs were injected in the urethral wall of hADSC rats whereas sham and US rats were injected with the vehicle. After 4 wk, the rats underwent cystometries and tissues were then harvested for functional and molecular analyses. Outcome measurements and statistical analysis: Cystometry, microultrasound, histochemistry, organ bath studies, reverse transcription polymerase chain reaction, and western blot. Results and limitations: US rats exhibited 49-51% shorter micturition intervals, 35-51% smaller micturition volumes and bladder capacity, 33-62% higher threshold pressures and flow pressures, and 35-37% lower bladder filling compliance compared with hADSC-treated rats and sham rats (p

    • Introduction. Cyclic adenosine 35 monophosphate (cAMP) is produced by adenylate cyclase after activation by, e.g., vasoactive intestinal polypeptide or prostaglandin E1 (PGE1). The cAMP-degrading phosphodiesterase 4 (PDE4) is expressed in the vagina and clitoris, but no information is available on the functional role for PDE4-related signals in the female neurovascular genital response. Aim. The aim of this study is to study the effect of inhibition of PDE4 with rolipram on nerve- and PGE1-induced vaginal and clitoral blood flow responses of rat. Methods. Measure of clitoral and vaginal blood flow and blood pressure in anesthetized rats during activation of the dorsal clitoral nerve (DCN) before and after intraperitoneal administration of rolipram or sildenafil (phosphodiesterase type 5 inhibitors [PDE5]) and nitro-L-arginine (L-NNA) (nitric oxide synthase inhibitor). Effect by topical administration of PGE1 on genital blood flow was also evaluated. Main Outcome Measure. Blood flow was recorded as tissue perfusion units (TPU) by a Laser Doppler Flowmeter. Mean arterial blood pressure (MAP) was recorded (cmH2O) in the carotid artery. Blood flow responses are expressed as TPU/MAP. Unpaired t-test and an analysis of variance were used. Results. Compared with control stimulations, rolipram (0.3mg/kg) caused a twofold increase in peak blood flow (Pandlt;0.05) and fourfold increase of the rate of clitoral blood flow during activation of the DCN (Pandlt;0.05). Simultaneously, a twofold increase in peak blood flow and threefold increase in rate of blood flow were noted in the vagina (Pandlt;0.05). Similar effects were noted for sildenafil (0.2mg/kg) (Pandlt;0.05). Inhibitory effects by L-NNA (60mg/kg) on blood flow responses to DCN activation were significantly lower for rats treated with rolipram than with sildenafil (Pandlt;0.05). PGE1-induced (10g) blood flow responses were significantly higher (Pandlt;0.05) in rats treated with rolipram than with sildenafil. Conclusions. These findings suggest that the cAMP/PDE4 system may be of similar functional importance as the nitric oxide/cyclic guanosine monophosphate/PDE5 pathway for neurovascular genital responses of the female rat.     • In 2008 we published the first set of guidelines for standardizing research in autophagy. Since then, research on this topic has continued to accelerate, and many new scientists have entered the field. Our knowledge base and relevant new technologies have also been expanding. Accordingly, it is important to update these guidelines for monitoring autophagy in different organisms. Various reviews have described the range of assays that have been used for this purpose. Nevertheless, there continues to be confusion regarding acceptable methods to measure autophagy, especially in multicellular eukaryotes. A key point that needs to be emphasized is that there is a difference between measurements that monitor the numbers or volume of autophagic elements (e.g., autophagosomes or autolysosomes) at any stage of the autophagic process vs. those that measure flux through the autophagy pathway (i.e., the complete process); thus, a block in macroautophagy that results in autophagosome accumulation needs to be differentiated from stimuli that result in increased autophagic activity, defined as increased autophagy induction coupled with increased delivery to, and degradation within, lysosomes (in most higher eukaryotes and some protists such as Dictyostelium) or the vacuole (in plants and fungi). In other words, it is especially important that investigators new to the field understand that the appearance of more autophagosomes does not necessarily equate with more autophagy. In fact, in many cases, autophagosomes accumulate because of a block in trafficking to lysosomes without a concomitant change in autophagosome biogenesis, whereas an increase in autolysosomes may reflect a reduction in degradative activity. Here, we present a set of guidelines for the selection and interpretation of methods for use by investigators who aim to examine macroautophagy and related processes, as well as for reviewers who need to provide realistic and reasonable critiques of papers that are focused on these processes. These guidelines are not meant to be a formulaic set of rules, because the appropriate assays depend in part on the question being asked and the system being used. In addition, we emphasize that no individual assay is guaranteed to be the most appropriate one in every situation, and we strongly recommend the use of multiple assays to monitor autophagy. In these guidelines, we consider these various methods of assessing autophagy and what information can, or cannot, be obtained from them. Finally, by discussing the merits and limits of particular autophagy assays, we hope to encourage technical innovation in the field.

  • Place-based solutions for global social-ecological dilemmas : An analysis of locally grounded, diversified, and cross-scalar initiatives in the Amazon

    • The Amazon has a diverse array of social and environmental initiatives that adopt forest-based land-use practices to promote rural development and support local livelihoods. However, they are often insufficiently recognized as transformative pathways to sustainability and the factors that explain their success remain understudied. To address this gap, this paper proposes that local initiatives that pursue three particular pathways are more likely to generate improvements in social-ecological outcomes: (1) maintaining close connections with local grassroots, (2) pursuing diversity in productive activities performed and partnership choices, and (3) developing cross-scale collaborations. To test these ideas we collected and analyzed observations of 157 initiatives in Brazil and Peru, applying a combination of quantitative and qualitative analyses. Our results show that initiatives maintaining groundedness in representing the interests and concerns of local actors while partnering with other organizations at multiple scales are more likely to develop joint solutions to social-ecological problems. Partnerships and support from external organizations may strengthen and enhance local capabilities, providing a platform for negotiating interests and finding common ground. Such diversified pathways demonstrate the power of local actors to transcend their own territories and have broader impacts in sustainability objectives. Our findings highlight the need to make governmental and non-governmental support (e.g., financial, technical, political) available according to local needs to enable local initiatives' own ways of addressing global environmental change.

  • Assessment of Rainfall Kinetic-Energy-Intensity Relationships

    • Raindrop-impact-induced erosion starts when detachment of soil particles from the surface results from an expenditure of raindrop energy. Hence, rain kinetic energy is a widely used indicator of the potential ability of rain to detach soil. Although it is widely recognized that knowledge of rain kinetic energy plays a fundamental role in soil erosion studies, its direct evaluation is not straightforward. Commonly, this issue is overcome through indirect estimation using another widely measured hydrological variable, namely, rainfall intensity. However, it has been challenging to establish the best expression to relate kinetic energy to rainfall intensity. In this study, first, kinetic energy values were determined from measurements of an optical disdrometer. Measured kinetic energy values were then used to assess the applicability of the rainfall intensity relationship proposed for central Italy and those used in the major equations employed to estimate the mean annual soil loss, that is, the Universal Soil Loss Equation (USLE) and its two revised versions (RUSLE and RUSLE2). Then, a new theoretical relationship was developed and its performance was compared with equations found in the literature.

    • For the study and modeling of hydrological phenomena, both in urban and rural areas, a proper estimation of the areal reduction factor (ARF) is crucial. In this paper, we estimated the ARF from observed rainfall data as the ratio between the average rainfall occurring in a specific area and the point rainfall. Then, we compared the obtained ARF values with some of the most widespread empirical approaches in literature which are used when rainfall observations are not available. Results highlight that the literature formulations can lead to a substantial over- or underestimation of the ARF estimated from observed data. These findings can have severe consequences, especially in the design of hydraulic structures where empirical formulations are extensively applied. The aim of this paper is to present a new analytical relationship with an explicit dependence on the rainfall duration and area that can better represent the ARF-area trend over the area case of study. The analytical curve presented here can find an important application to estimate the ARF values for design purposes. The test study area is the Lazio Region (central Italy).

    • Study region: We investigate samples from two Italian regions, i.e. Lazio and Sicily, located in central and south Italy, respectively, and characterized by two diverse climates. Study focus: In engineering practice, the study of maxima daily rainfall values is commonly dealt with light-tailed probability distribution functions, such as the Gumbel. The choice of a distribution rather than another may cause estimation errors of rainfall values associated to specific return periods. Recently, several studies demonstrate that heavy-tailed distributions are preferable for extreme events modelling. Here, we opt for six theoretical probability distribution functions and evaluate their performance in fitting extreme precipitation samples. We select the samples with two common methods, i.e. the Peak-Over-Threshold and the Annual Maxima. We assess the best fitting distribution to the empirical samples of extreme values through the Ratio Mean Square Error Method and the Kolmogorov-Smirnov test. New hydrological insights for the region: The assessment of the best fitting distribution to daily rainfall of the two different areas investigated here leads to interesting remarks. Despite the diversity of their climate, results suggest that heavy-tailed distributions describe more accurately empirical data rather than light-tailed ones. Therefore, extreme events may have been largely underestimated in the past in both areas. The proposed investigation can prompt the choice of the best fitting probability distribution to evaluate the design hydrological quantities supporting common engineering practice.
