  • The E3 ligase synoviolin controls body weight and mitochondrial biogenesis through negative regulation of PGC-1 beta
  • 2015
  • Ingår i: EMBO Journal. - : EMBO. - 1460-2075 .- 0261-4189. ; 34:8, s. 1042-1055
  • Tidskriftsartikel (refereegranskat)abstract
    • Obesity is a major global public health problem, and understanding its pathogenesis is critical for identifying a cure. In this study, a gene knockout strategy was used in post-neonatal mice to delete synoviolin (Syvn) 1/Hrd1/Der3, an ER-resident E3 ubiquitin ligase with known roles in homeostasis maintenance. Syvn1 deficiency resulted in weight loss and lower accumulation of white adipose tissue in otherwise wild-type animals as well as in genetically obese (ob/ob and db/db) and adipose tissue-specific knockout mice as compared to control animals. SYVN1 interacted with and ubiquitinated the thermogenic coactivator peroxisome proliferator-activated receptor coactivator (PGC)-1 beta, and Syvn1 mutants showed upregulation of PGC-1 beta target genes and increase in mitochondrion number, respiration, and basal energy expenditure in adipose tissue relative to control animals. Moreover, the selective SYVN1 inhibitor LS-102 abolished the negative regulation of PGC-1 beta by SYVN1 and prevented weight gain in mice. Thus, SYVN1 is a novel post-translational regulator of PGC-1 beta and a potential therapeutic target in obesity treatment.

  • Low-temperature annealing of mesoscopic TiO2 films by interfacial microwave heating applied to efficiency improvement of dye-sensitized solar cells
  • 2016
  • Ingår i: Solar Energy Materials and Solar Cells. - : Elsevier. - 0927-0248 .- 1879-3398. ; 147, s. 198-202
  • Tidskriftsartikel (refereegranskat)abstract
    • Novel interfacial microwave heating of a layered structure has been applied to the process of low temperature annealing of titania mesoporous films for dye-sensitized solar cells. Interfacial microwave annealing dramatically enhances the cell performances as compared with conventional heating in an electric oven. The improvement of cell performance is attributed to the electron transport properties in the titania films annealed by MW resulting in longer diffusion length as compared with conventional heating.

  • Variants in KCNQ1 are associated with susceptibility to type 2 diabetes mellitus
  • 2008
  • Ingår i: Nature Genetics. - : Springer Science and Business Media LLC. - 1546-1718 .- 1061-4036. ; 40:9, s. 1092-1097
  • Tidskriftsartikel (refereegranskat)abstract
    • We carried out a multistage genome-wide association study of type 2 diabetes mellitus in Japanese individuals, with a total of 1,612 cases and 1,424 controls and 100,000 SNPs. The most significant association was obtained with SNPs in KCNQ1, and dense mapping within the gene revealed that rs2237892 in intron 15 showed the lowest P value (6.7 x 10(-13), odds ratio (OR) = 1.49). The association of KCNQ1 with type 2 diabetes was replicated in populations of Korean, Chinese and European ancestry as well as in two independent Japanese populations, and meta-analysis with a total of 19,930 individuals (9,569 cases and 10,361 controls) yielded a P value of 1.7 x 10(-42) (OR = 1.40; 95% CI = 1.34-1.47) for rs2237892. Among control subjects, the risk allele of this polymorphism was associated with impairment of insulin secretion according to the homeostasis model assessment of beta-cell function or the corrected insulin response. Our data thus implicate KCNQ1 as a diabetes susceptibility gene in groups of different ancestries.
