    • In this document, technical details of the upgrade plan of the J-PARC neutrino beamline for the extension of the T2K experiment are described. T2K has proposed to accumulate data corresponding to 2×1022 protons-on-target in the next decade, aiming at an initial observation of CP violation with 3σ or higher significance in the case of maximal CP violation. Methods to increase the neutrino beam intensity, which are necessary to achieve the proposed data increase, are described.

  • Guideline adherence and survival of patients with candidaemia in Europe: results from the ECMM Candida III multinational European observational cohort study

    • Background The European Confederation of Medical Mycology (ECMM) collected data on epidemiology, risk factors, treatment, and outcomes of patients with culture-proven candidaemia across Europe to assess how adherence to guideline recommendations is associated with outcomes.Methods In this observational cohort study, 64 participating hospitals located in 20 European countries, with the number of eligible hospitals per country determined by population size, included the first ten consecutive adults with culture-proven candidaemia after July 1, 2018, and entered data into the ECMM Candida Registry (FungiScope CandiReg). We assessed ECMM Quality of Clinical Candidaemia Management (EQUAL Candida) scores reflecting adherence to recommendations of the European Society of Clinical Microbiology and Infectious Diseases and the Infectious Diseases Society of America guidelines.Findings 632 patients with candidaemia were included from 64 institutions. Overall 90-day mortality was 43% (265/617), and increasing age, intensive care unit admission, point increases in the Charlson comorbidity index score, and Candida tropicalis as causative pathogen were independent baseline predictors of mortality in Cox regression analysis. EQUAL Candida score remained an independent predictor of mortality in the multivariable Cox regression analyses after adjusting for the baseline predictors, even after restricting the analysis to patients who survived for more than 7 days after diagnosis (adjusted hazard ratio 1 & BULL;08 [95% CI 1 & BULL;04-1 & BULL;11; p<0 & BULL;0001] in patients with a central venous catheter and 1 & BULL;09 [1 & BULL;05-1 & BULL;13; p<0 & BULL;0001] in those without one, per one score point decrease). Median duration of hospital stay was 15 days (IQR 4-30) after diagnosis of candidaemia and was extended specifically for completion of parenteral therapy in 100 (16%) of 621 patients. Initial echinocandin treatment was associated with lower overall mortality and longer duration of hospital stay among survivors than treatment with other antifungals.Interpretation Although overall mortality in patients with candidaemia was high, our study indicates that adherence to clinical guideline recommendations, reflected by higher EQUAL Candida scores, might increase survival. New antifungals, with similar activity as current echinocandins but with longer half-lives or oral bioavailability, are needed to reduce duration of hospital stay.

  • Contrasting changes in the abundance and diversity of North American bird assemblages from 1971 to 2010

    • Although it is generally recognized that global biodiversity is declining, few studies have examined long-term changes in multiple biodiversity dimensions simultaneously. In this study, we quantified and compared temporal changes in the abundance, taxonomic diversity, functional diversity, and phylogenetic diversity of bird assemblages, using roadside monitoring data of the North American Breeding Bird Survey from 1971 to 2010. We calculated 12 abundance and diversity metrics based on 5-year average abundances of 519 species for each of 768 monitoring routes. We did this for all bird species together as well as for four subgroups based on breeding habitat affinity (grassland, woodland, wetland, and shrubland breeders). The majority of the biodiversity metrics increased or remained constant over the study period, whereas the overall abundance of birds showed a pronounced decrease, primarily driven by declines of the most abundant species. These results highlight how stable or even increasing metrics of taxonomic, functional, or phylogenetic diversity may occur in parallel with substantial losses of individuals. We further found that patterns of change differed among the species subgroups, with both abundance and diversity increasing for woodland birds and decreasing for grassland breeders. The contrasting changes between abundance and diversity and among the breeding habitat groups underscore the relevance of a multifaceted approach to measuring biodiversity change. Our findings further stress the importance of monitoring the overall abundance of individuals in addition to metrics of taxonomic, functional, or phylogenetic diversity, thus confirming the importance of population abundance as an essential biodiversity variable.

  • Comparative Greenhouse Gas Footprinting of Online versus Traditional Shopping for Fast-Moving Consumer Goods: A Stochastic Approach

    • Variability in consumer practices and choices is typically not addressed in comparisons of environmental impacts of traditional shopping and e-commerce. Here, we developed a stochastic model to quantify the variability in the greenhouse gas (GHG) footprints of product distribution and purchase of fast-moving consumer goods (FMCGs) via three prevalent retail channels in the United Kingdom (U.K.). We found that shopping via bricks and clicks (click and fulfillment via physical store delivery) most likely decreases the GHG footprints when substituting traditional shopping, while FMCGs purchased through pure players with parcel delivery often have higher GHG footprints compared to those purchased via traditional retail. The number of items purchased and the last-mile travel distance are the dominant contributors to the variability in the GHG footprints of all three retail channels. We further showed that substituting delivery vans with electric cargo bikes can lead to a GHG emission reduction of 26% via parcel delivery. Finally, we showed the differences in the "last mile" GHG footprint of traditional shopping in the U.K. compared to three other countries (China, Netherlands, and the United States), which are primarily caused by the different shares of modes of transport ( walking and by car, bus, and bike).

    • Hepatitis C virus (HCV) has no animal reservoir, infecting only humans. To investigate species barrier determinants limiting infection of rodents, murine liver complementary DNA library screening was performed, identifying transmembrane proteins Cd302 and Cr1l as potent restrictors of HCV propagation. Combined ectopic expression in human hepatoma cells impeded HCV uptake and cooperatively mediated transcriptional dysregulation of a noncanonical program of immunity genes. Murine hepatocyte expression of both factors was constitutive and not interferon inducible, while differences in liver expression and the ability to restrict HCV were observed between the murine orthologs and their human counterparts. Genetic ablation of endogenous Cd302 expression in human HCV entry factor transgenic mice increased hepatocyte permissiveness for an adapted HCV strain and dysregulated expression of metabolic process and host defense genes. These findings highlight human-mouse differences in liver-intrinsic antiviral immunity and facilitate the development of next-generation murine models for preclinical testing of HCV vaccine candidates.

    • We demonstrated that ultra narrowband filtering at 30 GHz dramatically enhances the signal-to-noise ratio to search for dark matter candidates which couple to millimeter waves. A coherent signal relatively locked between a synthesizer and a signal analyzer can be clearly distinguished from incoherent blackbody radiation after real time FFT over a long period. We propose a novel experiment for studying dark matter candidates.

    • Computed optical properties of membrane probes are typically evaluated in the gas phase, i.e. neglecting the influence of the membrane. In this study, we examine how and to what extent a membrane influences the one- and two-photon absorption (1PA and 2PA, respectively) properties for a number of cholesterol analogs and thereby also evaluate the validity of the common gas phase approach. The membrane is modeled using the polarizable embedding scheme both with and without the effective external field extension of the polarizable embedding model. The shifts in excitation energies and 1PA oscillator strengths compared to the gas phase are relatively small, while the 2PA cross section is more affected. The electric field inside the membrane induces a larger change in the permanent electric dipole moment upon excitation of the analogs compared to the gas phase, which leads to an almost 2-fold increase in the 2PA cross section for one cholesterol analog. The relative trends observed in the membrane are the same as in the gas phase, and the use of gas phase calculations for qualitative comparison and design of cholesterol membrane probes is thus a useful and computationally efficient strategy.

    • The main objective of this work is the application of variational concepts to microscopic multiple particle systems (MPS) which are assigned to corresponding points of a macroscopic continuum. Due to this underlying micro-structure it is not sufficient to simulate the macroscopic behavior with pre-assumed (overall) material parameters, or rather constitutive-law-based standard methods. Therefore, the challenge is to determine macroscopic material behaviors, by means of e.g. stresses and numerical tangent-stiffnesses, based on the analysis of the underlying multiple particle system. With the assistance of the applied variational principle and the so-called continuization, which corresponds to the limit of an infinite number of particles in the system, the analogy of homogenization of discrete and continuous micro-systems is elaborated. Within this work we focus on the so-called computational homogenization scheme, which provides the stage for a coupling between a macroscopic system simulated by the Finite Element Method and different microscopic simulation techniques.

    • This manuscript investigates possible bifurcations into stationary wave-type solutions in situations where an isotropic elastic bulk is bonded by a non-coherent orthotropic interface to a rigid substrate. The interface is characterised by a tractionseparation-law capturing elastic and inelastic material behaviour. Based on an ansatz for stationary waves at the surface of the bulk being connected to the interface, a bifurcation condition is elaborated. Thereby, the orthotropy of the interface is taken into account by considering the fully three-dimensional elastic half space and allowing for arbitrary orientations of the stationary wave-type solutions with respect to the interface co-ordinate system. The bifurcation condition is studied under the assumption of prevailing ellipticity in the bulk for a number of different examples. The results are graphically explored.

  • Investigation of microcracks in ferroelectric materials by application of a grain--boundary--motivated cohesive law

    • Ferroelectric materials exhibit a huge potential for engineering applications - ranging from electrical actuators (inverse piezoelectric effect) to sensor technology (direct piezoelectric effect). To give an example, lead zirconate titanate (PZT) is a typical perovskite ion crystal possessing ferroelectric properties. In this contribution, we are particularly interested in the modelling of microcracking effects in ferroelectric materials. In view of Finite-Element-based simulations, the geometry of a natural grain structure, as observed on the so-called micro-level, is represented by an appropriate mesh. While the response on the grains themselves is approximated by coupled continuum elements, grain boundaries are numerically incorporated via so-called cohesive-type elements. For the sake of simplicity, switching effects in the bulk material will be neglected. The behaviour of the grain boundaries is modelled by means of cohesive-type laws. Identifying grain boundaries as potential failure zones leading to microcracking, cohesive-type elements consequently offer a great potential for numerical simulations. As an advantage, in the case of failure they do not a priori result in ill-conditioned systems of equations as compared with the application of standard continuum elements to localised deformations. Finally, representative constitutive relations for both the bulk material and the grain boundaries, enable two-dimensional studies of low-cycle-fatigue motivated benchmark boundary value problems.

    • Ferroelectric materials offer a variety of new applications in the field of smart structures and intelligent systems. Accordingly, the modelling of these materials constitutes an active field of research. A critical limitation of the performance of such materials is given when electrical, mechanical, or mixed loading fatigue occurs, combined with, for instance, microcracking phenomena. In this contribution, fatigue effects in ferroelectric materials are numerically investigated by utilisation of a cohesive-type approach. In view of finite element-based simulations, the geometry of a natural grain structure, as observed on the so-called meso-level, is represented by an appropriate mesh. While the response of the grains themselves is approximated by coupled continuum elements, grain boundaries are numerically incorporated via so-called cohesive-type or interface elements. These offer a great potential for numerical simulations: as an advantage, they do not result in bad-conditioned systems of equations as compared with the application of standard continuum elements inhering a very high ratio of length and height. The grain boundary behaviour is modelled by cohesive-type constitutive laws, designed to capture fatigue phenomena. Being a first attempt, switching effects are planned to be added to the grain model in the future. Two differently motivated fatigue evolution techniques are applied, the first being appropriate for low-cycle-fatigue, and a second one adequate to simulate high-cycle-fatigue. Subsequent to a demonstration of the theoretical and numerical framework, studies of benchmark boundary value problems with fatigue-motivated boundary conditions are presented.
