    • Genetic variants within nearly 1000 loci are known to contribute to modulation of blood lipid levels. However, the biological pathways underlying these associations are frequently unknown, limiting understanding of these findings and hindering downstream translational efforts such as drug target discovery. To expand our understanding of the underlying biological pathways and mechanisms controlling blood lipid levels, we leverage a large multi-ancestry meta-analysis (N=1,654,960) of blood lipids to prioritize putative causal genes for 2286 lipid associations using six gene prediction approaches. Using phenome-wide association (PheWAS) scans, we identify relationships of genetically predicted lipid levels to other diseases and conditions. We confirm known pleiotropic associations with cardiovascular phenotypes and determine novel associations, notably with cholelithiasis risk. We perform sex-stratified GWAS meta-analysis of lipid levels and show that 3-5% of autosomal lipid-associated loci demonstrate sex-biased effects. Finally, we report 21 novel lipid loci identified on the X chromosome. Many of the sex-biased autosomal and X chromosome lipid loci show pleiotropic associations with sex hormones, emphasizing the role of hormone regulation in lipid metabolism.Taken together, our findings provide insights into the biological mechanisms through which associated variants lead to altered lipid levels and potentially cardiovascular disease risk.

  • Gene-educational attainment interactions in a multi-ancestry genome-wide meta-analysis identify novel blood pressure loci
  • 2021
  • Ingår i: Molecular Psychiatry. - : Springer Nature. - 1359-4184 .- 1476-5578. ; 26:6, s. 2111-2125

    • Educational attainment is widely used as a surrogate for socioeconomic status (SES). Low SES is a risk factor for hypertension and high blood pressure (BP). To identify novel BP loci, we performed multi-ancestry meta-analyses accounting for gene-educational attainment interactions using two variables, “Some College” (yes/no) and “Graduated College” (yes/no). Interactions were evaluated using both a 1 degree of freedom (DF) interaction term and a 2DF joint test of genetic and interaction effects. Analyses were performed for systolic BP, diastolic BP, mean arterial pressure, and pulse pressure. We pursued genome-wide interrogation in Stage 1 studies (N = 117 438) and follow-up on promising variants in Stage 2 studies (N = 293 787) in five ancestry groups. Through combined meta-analyses of Stages 1 and 2, we identified 84 known and 18 novel BP loci at genome-wide significance level (P < 5 × 10-8). Two novel loci were identified based on the 1DF test of interaction with educational attainment, while the remaining 16 loci were identified through the 2DF joint test of genetic and interaction effects. Ten novel loci were identified in individuals of African ancestry. Several novel loci show strong biological plausibility since they involve physiologic systems implicated in BP regulation. They include genes involved in the central nervous system-adrenal signaling axis (ZDHHC17, CADPS, PIK3C2G), vascular structure and function (GNB3, CDON), and renal function (HAS2 and HAS2-AS1, SLIT3). Collectively, these findings suggest a role of educational attainment or SES in further dissection of the genetic architecture of BP.

  • Multiancestry Genome-Wide Association Study of Lipid Levels Incorporating Gene-Alcohol Interactions
  • 2019
  • Ingår i: American Journal of Epidemiology. - : Oxford University Press. - 0002-9262 .- 1476-6256. ; 188:6, s. 1033-1054
  • Tidskriftsartikel (refereegranskat)abstract
    • A person's lipid profile is influenced by genetic variants and alcohol consumption, but the contribution of interactions between these exposures has not been studied. We therefore incorporated gene-alcohol interactions into a multiancestry genome-wide association study of levels of high-density lipoprotein cholesterol, low-density lipoprotein cholesterol, and triglycerides. We included 45 studies in stage 1 (genome-wide discovery) and 66 studies in stage 2 (focused follow-up), for a total of 394,584 individuals from 5 ancestry groups. Analyses covered the period July 2014-November 2017. Genetic main effects and interaction effects were jointly assessed by means of a 2-degrees-of-freedom (df) test, and a 1-df test was used to assess the interaction effects alone. Variants at 495 loci were at least suggestively associated (P < 1 x 10(-6)) with lipid levels in stage 1 and were evaluated in stage 2, followed by combined analyses of stage 1 and stage 2. In the combined analysis of stages 1 and 2, a total of 147 independent loci were associated with lipid levels at P < 5 x 10(-8) using 2-df tests, of which 18 were novel. No genome-wide-significant associations were found testing the interaction effect alone. The novel loci included several genes (proprotein convertase subtilisin/kexin type 5 (PCSK5), vascular endothelial growth factor B (VEGFB), and apolipoprotein B mRNA editing enzyme, catalytic polypeptide 1 (APOBEC1) complementation factor (A1CF)) that have a putative role in lipid metabolism on the basis of existing evidence from cellular and experimental models.

  • Novel genetic associations for blood pressure identified via gene-alcohol interaction in up to 570K individuals across multiple ancestries
  • 2018
  • Ingår i: PLOS ONE. - : Public library science. - 1932-6203. ; 13:6

    • Heavy alcohol consumption is an established risk factor for hypertension; the mechanism by which alcohol consumption impact blood pressure (BP) regulation remains unknown. We hypothesized that a genome-wide association study accounting for gene-alcohol consumption interaction for BP might identify additional BP loci and contribute to the understanding of alcohol-related BP regulation. We conducted a large two-stage investigation incorporating joint testing of main genetic effects and single nucleotide variant (SNV)-alcohol consumption interactions. In Stage 1, genome-wide discovery meta-analyses in approximate to 131 K individuals across several ancestry groups yielded 3,514 SNVs (245 loci) with suggestive evidence of association (P <1.0 x 10(-5)). In Stage 2, these SNVs were tested for independent external replication in individuals across multiple ancestries. We identified and replicated (at Bonferroni correction threshold) five novel BP loci (380 SNVs in 21 genes) and 49 previously reported BP loci (2,159 SNVs in 109 genes) in European ancestry, and in multi-ancestry meta-analyses (P < 5.0 x 10(-8)). For African ancestry samples, we detected 18 potentially novel BP loci (P< 5.0 x 10(-8)) in Stage 1 that warrant further replication. Additionally, correlated meta-analysis identified eight novel BP loci (11 genes). Several genes in these loci (e.g., PINX1, GATA4, BLK, FTO and GABBR2 have been previously reported to be associated with alcohol consumption. These findings provide insights into the role of alcohol consumption in the genetic architecture of hypertension.

  • Loci influencing blood pressure identified using a cardiovascular gene-centric array

  • Ingår i: Human Molecular Genetics. - : Oxford University Press (OUP). - 0964-6906 .- 1460-2083. ; 22:8, s. 1663-1678

    • Blood pressure (BP) is a heritable determinant of risk for cardiovascular disease (CVD). To investigate genetic associations with systolic BP (SBP), diastolic BP (DBP), mean arterial pressure (MAP) and pulse pressure (PP), we genotyped 50 000 single-nucleotide polymorphisms (SNPs) that capture variation in 2100 candidate genes for cardiovascular phenotypes in 61 619 individuals of European ancestry from cohort studies in the USA and Europe. We identified novel associations between rs347591 and SBP (chromosome 3p25.3, in an intron of HRH1) and between rs2169137 and DBP (chromosome1q32.1 in an intron of MDM4) and between rs2014408 and SBP (chromosome 11p15 in an intron of SOX6), previously reported to be associated with MAP. We also confirmed 10 previously known loci associated with SBP, DBP, MAP or PP (ADRB1, ATP2B1, SH2B3/ATXN2, CSK, CYP17A1, FURIN, HFE, LSP1, MTHFR, SOX6) at array-wide significance (P 2.4 10(6)). We then replicated these associations in an independent set of 65 886 individuals of European ancestry. The findings from expression QTL (eQTL) analysis showed associations of SNPs in the MDM4 region with MDM4 expression. We did not find any evidence of association of the two novel SNPs in MDM4 and HRH1 with sequelae of high BP including coronary artery disease (CAD), left ventricular hypertrophy (LVH) or stroke. In summary, we identified two novel loci associated with BP and confirmed multiple previously reported associations. Our findings extend our understanding of genes involved in BP regulation, some of which may eventually provide new targets for therapeutic intervention.

  • One thousand plant transcriptomes and the phylogenomics of green plants
  • 2019
  • Ingår i: Nature. - : Nature Publishing Group. - 0028-0836 .- 1476-4687. ; 574:7780, s. 679-

    • Green plants (Viridiplantae) include around 450,000-500,000 species(1,2) of great diversity and have important roles in terrestrial and aquatic ecosystems. Here, as part of the One Thousand Plant Transcriptomes Initiative, we sequenced the vegetative transcriptomes of 1,124 species that span the diversity of plants in a broad sense (Archaeplastida), including green plants (Viridiplantae), glaucophytes (Glaucophyta) and red algae (Rhodophyta). Our analysis provides a robust phylogenomic framework for examining the evolution of green plants. Most inferred species relationships are well supported across multiple species tree and supermatrix analyses, but discordance among plastid and nuclear gene trees at a few important nodes highlights the complexity of plant genome evolution, including polyploidy, periods of rapid speciation, and extinction. Incomplete sorting of ancestral variation, polyploidization and massive expansions of gene families punctuate the evolutionary history of green plants. Notably, we find that large expansions of gene families preceded the origins of green plants, land plants and vascular plants, whereas whole-genome duplications are inferred to have occurred repeatedly throughout the evolution of flowering plants and ferns. The increasing availability of high-quality plant genome sequences and advances in functional genomics are enabling research on genome evolution across the green tree of life.

  • One-Step Synthesis of Precursor Oligomers for Organic Photovoltaics: A Comparative Study between Polymers and Small Molecules
  • 2015
  • Ingår i: ACS Applied Materials and Interfaces. - : American Chemical Society (ACS). - 1944-8244 .- 1944-8252. ; 7:49, s. 27106-27114
  • Tidskriftsartikel (refereegranskat)abstract
    • Two series of oligomers TQ and rhodanine end-capped TQ-DR were synthesized using a facile one-step method. Their optical, electrical, and thermal properties and photovoltaic performances were systematically investigated and compared. The TQ series of oligomers were found to be amorphous, whereas the TQ-DR series are semicrystalline. For the TQ oligomers, the results obtained in solar cells show that as the chain length of the oligomers increases, an increase in power conversion efficiency (PCE) is obtained. However, when introducing 3-ethylrhodanine into the TQ oligomers as end groups, the PCE of the TQ-DR series of oligomers decreases as the chain length increases. Moreover, the TQ-DR series of oligomers give much higher performances compared to the original amorphous TQ series of oligomers owing to the improved extinction coefficient (epsilon) and crystallinity afforded by the rhodanine. In particular, the highly crystalline oligomer TQ5-DR, which has the shortest conjugation length shows a high hole mobility of 0.034 cm(2) V-1 s(-1) and a high PCE of 3.14%, which is the highest efficiency out of all of the six oligomers. The structure-property correlations for all of the oligomers and the TQ1 polymer demonstrate that structural control of enhanced intermolecular interactions and crystallinity is a key for small molecules/oligomers to achieve high mobilities, which is an essential requirement for use in OPVs.

  • 9.0% power conversion efficiency from ternary all-polymer solar cells
  • 2017
  • Ingår i: Energy and Environmental Science. - : Royal Society of Chemistry (RSC). - 1754-5692 .- 1754-5706. ; 10:10, s. 2212-2221
  • Tidskriftsartikel (refereegranskat)abstract
    • Integration of a third component into a single-junction polymer solar cell (PSC) is regarded as an attractive strategy to enhance the performance of PSCs. Although binary all-polymer solar cells (all-PSCs) have recently emerged with compelling power conversion efficiencies (PCEs), the PCEs of ternary all-PSCs still lag behind those of the state-of-the-art binary all-PSCs, and the advantages of ternary systems are not fully exploited. In this work, we realize high-performance ternary all-PSCs with record-breaking PCEs of 9% and high fill factors (FF) of over 0.7 for both conventional and inverted devices. The improved photovoltaic performance benefits from the synergistic effects of extended absorption, more efficient charge generation, optimal polymer orientations and suppressed recombination losses compared to the binary all-PSCs, as evidenced by a set of experimental techniques. The results provide new insights for developing high-performance ternary all-PSCs by choosing appropriate donor and acceptor polymers to overcome limitations in absorption, by affording good miscibility, and by benefiting from charge and energy transfer mechanisms for efficient charge generation.

  • 2015
  • Ingår i: American Journal of Botany. - : Wiley. - 0002-9122 .- 1537-2197. ; 102:7, s. 1089-1107

    • PREMISE OF THE STUDY: Understanding fern (monilophyte) phylogeny and its evolutionary timescale is critical for broad investigations of the evolution of land plants, and for providing the point of comparison necessary for studying the evolution of the fern sister group, seed plants. Molecular phylogenetic investigations have revolutionized our understanding of fern phylogeny, however, to date, these studies have relied almost exclusively on plastid data. METHODS: Here we take a curated phylogenomics approach to infer the first broad fern phylogeny from multiple nuclear loci, by combining broad taxon sampling (73 ferns and 12 outgroup species) with focused character sampling (25 loci comprising 35 877 bp), along with rigorous alignment, orthology inference and model selection. KEY RESULTS: Our phylogeny corroborates some earlier inferences and provides novel insights; in particular, we find strong support for Equisetales as sister to the rest of ferns, Marattiales as sister to leptosporangiate ferns, and Dennstaedtiaceae as sister to the eupolypods. Our divergence-time analyses reveal that divergences among the extant fern orders all occurred prior to similar to 200 MYA. Finally, our species-tree inferences are congruent with analyses of concatenated data, but generally with lower support. Those cases where species-tree support values are higher than expected involve relationships that have been supported by smaller plastid datasets, suggesting that deep coalescence may be reducing support from the concatenated nuclear data. CONCLUSIONS: Our study demonstrates the utility of a curated phylogenomics approach to inferring fern phylogeny, and highlights the need to consider underlying data characteristics, along with data quantity, in phylogenetic studies.

  • A multi-ancestry genome-wide study incorporating gene-smoking interactions identifies multiple new loci for pulse pressure and mean arterial pressure
  • 2019
  • Ingår i: Human Molecular Genetics. - : Oxford University Press. - 0964-6906 .- 1460-2083. ; 28:15, s. 2615-2633

    • Elevated blood pressure (BP), a leading cause of global morbidity and mortality, is influenced by both genetic and lifestyle factors. Cigarette smoking is one such lifestyle factor. Across five ancestries, we performed a genome-wide gene–smoking interaction study of mean arterial pressure (MAP) and pulse pressure (PP) in 129 913 individuals in stage 1 and follow-up analysis in 480 178 additional individuals in stage 2. We report here 136 loci significantly associated with MAP and/or PP. Of these, 61 were previously published through main-effect analysis of BP traits, 37 were recently reported by us for systolic BP and/or diastolic BP through gene–smoking interaction analysis and 38 were newly identified (P < 5 × 10−8, false discovery rate < 0.05). We also identified nine new signals near known loci. Of the 136 loci, 8 showed significant interaction with smoking status. They include CSMD1 previously reported for insulin resistance and BP in the spontaneously hypertensive rats. Many of the 38 new loci show biologic plausibility for a role in BP regulation. SLC26A7 encodes a chloride/bicarbonate exchanger expressed in the renal outer medullary collecting duct. AVPR1A is widely expressed, including in vascular smooth muscle cells, kidney, myocardium and brain. FHAD1 is a long non-coding RNA overexpressed in heart failure. TMEM51 was associated with contractile function in cardiomyocytes. CASP9 plays a central role in cardiomyocyte apoptosis. Identified only in African ancestry were 30 novel loci. Our findings highlight the value of multi-ancestry investigations, particularly in studies of interaction with lifestyle factors, where genomic and lifestyle differences may contribute to novel findings.

  • Low Band Gap Polymer Solar Cells With Minimal Voltage Losses
  • 2016
  • Ingår i: Advanced Energy Materials. - : Wiley. - 1614-6832 .- 1614-6840. ; 6:18
  • Tidskriftsartikel (refereegranskat)abstract
    • One of the factors limiting the performance of organic solar cells (OSCs) is their large energy losses (E loss) in the conversion from photons to electrons, typically believed to be around 0.6 eV and often higher than those of inorganic solar cells. In this work, a novel low band gap polymer PIDTT-TID with a optical gap of 1.49 eV is synthesized and used as the donor combined with PC71BM in solar cells. These solar cells attain a good power conversion efficiency of 6.7% with a high open-circuit voltage of 1.0 V, leading to the E loss as low as 0.49 eV. A systematic study indicates that the driving force in this donor and acceptor system is sufficient for charge generation with the low E loss. This work pushes the minimal E loss of OSCs down to 0.49 eV, approaching the values of some inorganic and hybrid solar cells. It indicates the potential for further enhancement of the performance of OSCs by improving their V oc since the E loss can be minimized.

  • 2017
  • Ingår i: Nano Energy. - : Elsevier BV. - 2211-2855 .- 2211-3282. ; 37, s. 24-31
  • Tidskriftsartikel (refereegranskat)abstract
    • By introducing a non-fullerene small molecule acceptor as a third component to typical polymer donor: fullerene acceptor binary solar cells, we demonstrate that the short circuit current density (Jsc), open circuit voltage (Voc), power conversion efficiency (PCE) and thermal stability can be enhanced simultaneously. The different surface energy of each component causes most of the non-fullerene acceptor molecules to self-organize at the polymer/fullerene interface, while the appropriately selected oxidation/reduction potential of the non-fullerene acceptor enables the resulting ternary junction to work through a cascade mechanism. The cascade ternary junction enhances charge generation through complementary absorption between the non-fullerene and fullerene acceptors and aids the efficient charge extraction from fullerene domains. The bimolecular recombination in the ternary blend layer is reduced as the ternary cascade junction increases the separation of holes and electrons during charge transportation and the trap assistant recombination induced by integer charge transfer (ICT) state potentially reduced due to the smaller pinning energy of inserted non-fullerene acceptor, leading to an unprecedented increase in the open circuit voltage beyond the binary reference values.

    • A new strategy for designing ternary solar cells is reported in this paper. A low-bandgap polymer named PTB7-Th and a high-bandgap polymer named PBDTTS-FTAZ sharing the same bulk ionization potential and interface positive integer charge transfer energy while featuring complementary absorption spectra are selected. They are used to fabricate efficient ternary solar cells, where the hole can be transported freely between the two donor polymers and collected by the electrode as in one broadband low bandgap polymer. Furthermore, the fullerene acceptor is chosen so that the energy of the positive integer charge transfer state of the two donor polymers is equal to the energy of negative integer charge transfer state of the fullerene, enabling enhanced dissociation of all polymer donor and fullerene acceptor excitons and suppressed bimolecular and trap assistant recombination. The two donor polymers feature good miscibility and energy transfer from high-bandgap polymer of PBDTTS-FTAZ to low-bandgap polymer of PTB7-Th, which contribute to enhanced performance of the ternary solar cell.

  • 8.0% Efficient All-Polymer Solar Cells with High Photovoltage of 1.1 V and Internal Quantum Efficiency near Unity
  • 2018
  • Ingår i: Advanced Energy Materials. - : Wiley. - 1614-6832 .- 1614-6840. ; 8:1
  • Tidskriftsartikel (refereegranskat)abstract
    • In very recent years, growing efforts have been devoted to the development of all-polymer solar cells (all-PSCs). One of the advantages of all-PSCs over the fullerene-based PSCs is the versatile design of both donor and acceptor polymers which allows the optimization of energy levels to maximize the open-circuit voltage (Voc). However, there is no successful example of all-PSCs with both high Voc over 1 V and high power conversion efficiency (PCE) up to 8% reported so far. In this work, a combination of a donor polymer poly[4,8-bis(5-(2-octylthio)thiophen-2-yl)benzo[1,2-b:4,5-b′]dithiophene-2,6-diyl-alt-(5-(2-ethylhexyl)-4H-thieno[3,4-c]pyrrole-4,6(5H)-dione)-1,3-diyl] (PBDTS-TPD) with a low-lying highest occupied molecular orbital level and an acceptor polymer poly[[N,N′-bis(2-octyldodecyl)-naphthalene-1,4,5,8-bis(dicarboximide)-2,6-diyl]-alt-thiophene-2,5-diyl] (PNDI-T) with a high-lying lowest unoccupied molecular orbital level is used, realizing high-performance all-PSCs with simultaneously high Voc of 1.1 V and high PCE of 8.0%, and surpassing the performance of the corresponding PC71BM-based PSCs. The PBDTS-TPD:PNDI-T all-PSCs achieve a maximum internal quantum efficiency of 95% at 450 nm, which reveals that almost all the absorbed photons can be converted into free charges and collected by electrodes. This work demonstrates the advantages of all-PSCs by incorporating proper donor and acceptor polymers to boost both Voc and PCEs.

  • Field Investigations of Underwater Mounds Formed by Hopper Dredge Discharges in a Coastal Environment
  • 2020
  • Ingår i: Journal of Marine Science and Engineering. - : MDPI. - 2077-1312. ; 8:6
  • Tidskriftsartikel (refereegranskat)abstract
    • In a coastal environment, this paper investigated the formation process and the cumulative shape of subaqueous mounds formed by hopper dredged discharges. Hydrological observations and field tests were performed to examine the flow features and ultimately generated morphology characteristics. A high-precision digital elevation model (DEM) was established by multi-beam depth sweeping (MBDS) in the experiment. Particular attention was paid to the formation of the mounds, the three-dimensional shape and the influence factors. The field measurements showed that the mounds were roughly symmetrical in space, and the tidal current, though of weak strength, played a certain role in shaping the profiles. Cone and volcanic cone mound tops were observed, featuring the main top shapes. The height and covered area of the mounds were proportional to the amount of dumped sediment, and they were also affected a lot by the water depth. The results of superimposed tests showed that the second placement over the existing mound resulted in a similar overall shape, but there was pronounced movement around the mound; additional discharged volumes at the same location mainly increased the mound height. The field tests provided a reference for understanding the sediment dumping in other similar coastal areas.

  • Understanding the Settling Processes of Dredged Sediment Disposed in Open Waters through Experimental Tests and Numerical Simulations
  • 2022
  • Ingår i: Journal of Marine Science and Engineering. - : MDPI. - 2077-1312. ; 10:2

    • During dredging for subsea tunnels and pipelines, the dredged soil is typically dumped in a designated area. Understanding the settling behaviors of the dumped particles is essential for an accurate prediction of the resulting morphology. This study dealt with the settling processes in the open-water column by means of experimental tests and numerical simulations. Both quiescent and ambient current conditions were taken into account. Particular attention was paid to the induced flow patterns, descent sediment movement features and the resulting topography. Regarding the diffusion width and settling velocity, three key effect factors, i.e., dumped volume, particle size and ambient flow strength, were considered. The results show that the dumped sediment in the water forms a particle cloud, and two vortices with opposite rotations occur on both sides of the cloud. During settlement, three stages corresponding to convective descent, dynamic collapse and passive diffusion are observed. Most of the descending sediment is incorporated in a spherical vortex, resembling an upside-down mushroom cloud, and some sediment is also contained in an irregular trailing stem. The dumped particles exhibit initial acceleration, and then they slow down to converge to a fixed value. Subjected to the ambient currents, the initial acceleration phase is prolonged, and the vortex is somewhat distorted; the particle cloud is also advected downstream with a velocity roughly equal to the ambient current.

    • Web services as a new distributed system technology have been widely adopted by industries in the areas, such as enterprise application integration (EAI), business process management (BPM), and virtual organization (VO). However, lack of semantics in the current Web service standards has become a major barrier in service discovery and composition. To tackle the semantic issues of Web services, this paper proposes a comprehensive semantic service description framework – CbSSDF and a two-step service discovery mechanism based on CbSSDF—to help service users to easily locate their required services. The authors give a detailed explanation of CbSSDF, and then evaluate the framework by comparing it with OWL-S to examine how the proposed framework can improve the efficiency and effectiveness of service discovery and composition. The evaluation is carried out by analysing the different proposed solutions based on these two frameworks for achieving a series of tasks in a scenario.

    • Business development or renovation is to introduce newer, more efficient routines and processes through redesign or re-engineering of businesses, which form a set of business patterns. Business patterns encapsulate the best solutions for business practices and tasks confirming business strategies of the enterprise. Nowadays, services with SOA (Service oriented-Architecture) become more and more important in implementing and supporting business routines and processes. An enterprise that can encapsulate their SOA solutions into patterns will make the business more agile and effective. However, with the SOA solutions to automation of locating relevant instance services for its business patterns with minimum human intervention one has to look into the semantic and operational difference between the description of a business pattern and that of an instance service-a gap between the two levels of semantic descriptions. In this paper, we propose a conceptual modeling method to address how to bridge the gap, by a semantic service description for usage contextual approach formalized with the conceptual graphs formalism.

    • In the past few years, Geo-spatial data quality has received increasing attention and concerns. As more and more business decisions are made based on data analytic result from geo-spatial related data, low quality data means wrong or inappropriate decisions, which could have substantial effects on a business's future. In this paper, we propose a framework that can systematically ensure and improve geo-spatial data quality throughout the whole life cycle of data.

    • Cell therapy has been shown to be a key clinical therapeutic option for central nervous system diseases or damage. Standardization of clinical cell therapy procedures is an important task for professional associations devoted to cell therapy. The Chinese Branch of the International Association of Neurorestoratology (IANR) completed the first set of guidelines governing the clinical application of neurorestoration in 2011. The IANR and the Chinese Association of Neurorestoratology (CANR) collaborated to propose the current version "Clinical Cell Therapy Guidelines for Neurorestoration (IANR/CANR 2017)". The IANR council board members and CANR committee members approved this proposal on September 1, 2016, and recommend it to clinical practitioners of cellular therapy. These guidelines include items of cell type nomenclature, cell quality control, minimal suggested cell doses, patient-informed consent, indications for undergoing cell therapy, contraindications for undergoing cell therapy, documentation of procedure and therapy, safety evaluation, efficacy evaluation, policy of repeated treatments, do not charge patients for unproven therapies, basic principles of cell therapy, and publishing responsibility.

    • Homozygosity has long been associated with rare, often devastating, Mendelian disorders, and Darwin was one of the first to recognize that inbreeding reduces evolutionary fitness. However, the effect of the more distant parental relatedness that is common in modern human populations is less well understood. Genomic data now allow us to investigate the effects of homozygosity on traits of public health importance by observing contiguous homozygous segments (runs of homozygosity), which are inferred to be homozygous along their complete length. Given the low levels of genome-wide homozygosity prevalent in most human populations, information is required on very large numbers of people to provide sufficient power. Here we use runs of homozygosity to study 16 health-related quantitative traits in 354,224 individuals from 102 cohorts, and find statistically significant associations between summed runs of homozygosity and four complex traits: height, forced expiratory lung volume in one second, general cognitive ability and educational attainment (P < 1 × 10(-300), 2.1 × 10(-6), 2.5 × 10(-10) and 1.8 × 10(-10), respectively). In each case, increased homozygosity was associated with decreased trait value, equivalent to the offspring of first cousins being 1.2 cm shorter and having 10 months' less education. Similar effect sizes were found across four continental groups and populations with different degrees of genome-wide homozygosity, providing evidence that homozygosity, rather than confounding, directly contributes to phenotypic variance. Contrary to earlier reports in substantially smaller samples, no evidence was seen of an influence of genome-wide homozygosity on blood pressure and low density lipoprotein cholesterol, or ten other cardio-metabolic traits. Since directional dominance is predicted for traits under directional evolutionary selection, this study provides evidence that increased stature and cognitive function have been positively selected in human evolution, whereas many important risk factors for late-onset complex diseases may not have been.

  • Standards recommendations for the Earth BioGenome Project
  • 2022
  • Ingår i: Proceedings of the National Academy of Sciences of the United States of America. - : Proceedings of the National Academy of Sciences (PNAS). - 0027-8424 .- 1091-6490. ; 119:4
  • Tidskriftsartikel (refereegranskat)abstract
    • A global international initiative, such as the Earth BioGenome Project (EBP), requires both agreement and coordination on standards to ensure that the collective effort generates rapid progress toward its goals. To this end, the EBP initiated five technical standards committees comprising volunteer members from the global genomics scientific community: Sample Collection and Processing, Sequencing and Assembly, Annotation, Analysis, and IT and Informatics. The current versions of the resulting standards documents are available on the EBP website, with the recognition that opportunities, technologies, and challenges may improve or change in the future, requiring flexibility for the EBP to meet its goals. Here, we describe some highlights from the proposed standards, and areas where additional challenges will need to be met.
