    • Loess-paleosol sequences are the most intensively studied terrestrial archives used for the reconstruction of Late Pleistocene environmental and climatic changes in the Azov Sea region, southwest Russia. Here we present a refined chronostratigraphy and a multiproxy record of Late Pleistocene environmental dynamics of the most complete and representative loess–paleosol sequences (Beglitsa and Chumbur-Kosa sections) from the Azov Sea region. We propose a new chronostratigraphy following the Chinese and Danubean loess stratigraphic models that refines the subdivision of the Last Interglacial paleosol (S1) complex in two Azov Sea sites, resolve the uncertainty of the stratigraphic position of the weakly developed paleosol (L1SSm) in Beglitsa section, and allow for direct correlation of the Azov Sea sections with those in the Danube Basin and the Chinese Loess Plateau. More importantly, it may serve as a basis for better constraining local and regional chronostratigraphic correlations, and facilitate the interpretation of climatic connections and possible forcing mechanisms responsible for the climatic trends in the region. In addition, a general succession of environmental dynamics is reconstructed from these two vital sections, which is broadly consistent with other loess records in the Dnieper Lowland and Lower Danube Basin, demonstrating similar climatic trends at Glacial–Interglacial time scales. Furthermore, our results have important implications for the chronostratigraphic representativeness of Beglitsa as a key regional loess section and for the reconstruction of the temporal and spatial evolution of Late Pleistocene climate in the Azov Sea region.

  • Prevalence of Abdominal Obesity in Chinese Middle-Aged and Older Adults with a Normal Body Mass Index and Its Association with Type 2 Diabetes Mellitus : A Nationally Representative Cohort Study from 2011 to 2018

    • Background: Few studies have focused on the prevalence of abdominal obesity in Chinese middle-aged and older adults with a normal body mass index (BMI). Furthermore, it is still unclear whether abdominal obesity is an independent risk factor for type 2 diabetes mellitus (T2DM). Participants with a normal BMI are usually neglected during assessments of abdominal obesity-associated T2DM risk since the current recommendations for medical interventions are mainly focused on overall body mass index rather than fat deposition patterns. Methods: In this study, 7942 normal-BMI participants aged over 45 years from the China Health and Retirement Longitudinal Study were included to assess the prevalence of abdominal obesity defined by waist circumference (WC) or waist-to-height ratio (WHtR). In addition, 4348 normal-BMI individuals with no diabetes at baseline were included to evaluate the association between abdominal obesity and the risk of T2DM with the Cox proportional hazards model. Results: The prevalence (95% confidence interval, CI) of increased WC and substantially increased WC among adults with a normal BMI was 22.0% (21.1%-22.9%) and 18.1% (17.3%-19.0%), respectively. The adjusted hazard ratios and 95% CIs for T2DM incidence were 1.39 (1.05–1.85) and 1.89 (1.42–2.53) for those with increased WC and substantially increased WC, respectively, compared to the individuals with a normal WC. Similar HRs were obtained for the association between WHtR and the risk of T2DM. In prediabetic patients, the HRs (95% CIs) for new-onset T2DM for those with increased WC and substantially increased WC were 1.85 (1.27–2.69) and 2.46 (1.67–3.61), respectively, when compared with individuals with normal WC. This positive association was observed in women but not in men or adults with normal glucose tolerance (NGT). Conclusion: Abdominal obesity is highly prevalent among middle-aged and older Chinese adults with a normal BMI, and maintaining a normal waist circumference may be beneficial in the prevention of T2DM.

    • Excessive tissue deformation near cartilage lesions and acute inflammation within the knee joint after anterior cruciate ligament (ACL) rupture and reconstruction surgery accelerate the loss of fixed charge density (FCD) and subsequent cartilage tissue degeneration. Here, we show how biomechanical and biochemical degradation pathways can predict FCD loss using a patient-specific finite element model of an ACL reconstructed knee joint exhibiting a chondral lesion. Biomechanical degradation was based on the excessive maximum shear strains that may result in cell apoptosis, while biochemical degradation was driven by the diffusion of pro-inflammatory cytokines. We found that the biomechanical model was able to predict substantial localized FCD loss near the lesion and on the medial areas of the lateral tibial cartilage. In turn, the biochemical model predicted FCD loss all around the lesion and at intact areas; the highest FCD loss was at the cartilage–synovial fluid-interface and decreased toward the deeper zones. Interestingly, simulating a downturn of an acute inflammatory response by reducing the cytokine concentration exponentially over time in synovial fluid led to a partial recovery of FCD content in the cartilage. Our novel numerical approach suggests that in vivo FCD loss can be estimated in injured cartilage following ACL injury and reconstruction. Our novel modeling platform can benefit the prediction of PTOA progression and the development of treatment interventions such as disease-modifying drug testing and rehabilitation strategies.
