  • Evaluation of starvation status in the early developmental stages of black rockfish (Sebastes schlegelii) based on morphological and histological characteristics

    • Assessing the nutritional status and identifying major causes of mortality in larvae experiencing varying degrees of starvation are crucial for establishing appropriate feeding protocols and enhancing the welfare of hatchery-reared fish. The black rockfish Sebastes schlegelii is an important species in aquaculture and stock enhancement efforts in China, Japan, and Korea. This study aimed to identify optimal diagnostic morphometric indicators of starvation in newly hatched (0–6 days post-hatch, DPH) and postlarval stages (27–37 DPH) of this valuable fish species through histological analyses. Our findings revealed that certain morphometric parameters, including body length, the ratios of eye diameter to head height, body height to body length, and abdomen height to body height, exhibit sensitivity to starvation during both larval and postlarval stages. Particularly, the ratios of body height to body length and abdomen height to body height emerged as the most sensitive morphometric indicators of starvation. Histological examinations of the digestive system revealed rapid alterations in the morphology of hepatic parenchymal cells, accompanied by a significant decrease in the number of lipid cells in the liver during episodes of food deprivation. Starvation induced cellular degeneration in the digestive organs, manifested by reduced heights of epithelial cells and mucosal layers in the intestine, oesophagus, and stomach, along with degeneration and separation of muscle fibers. Among these variables, the height of the intestinal submucosa and muscle layer emerged as the most sensitive indicators reflecting nutritional conditions in newly hatched larvae. In contrast, the height of intestinal striated borders and mucosal folds proved to be the most sensitive indicators in the postlarval stage. Furthermore, the height of intestinal epithelial cells and the number of lipid vacuoles in enterocytes exhibited high sensitivity to food deprivation in both newly hatched larvae and postlarvae. These findings underscore the varying resilience of fish to starvation during different developmental phases and highlight the utility of morphological sensitivity characteristics as reliable diagnostic indices for assessing nutritional status in relation to starvation or suboptimal feeding during the early developmental stages of black rockfish in hatchery-reared processes.

  • Endothelial cell heterogeneity and microglia regulons revealed by a pig cell landscape at single-cell level

    • Pigs are valuable large animal models for biomedical and genetic research, but insights into the tissue- and cell-type-specific transcriptome and heterogeneity remain limited. By leveraging single-cell RNA sequencing, we generate a multiple-organ single-cell transcriptomic map containing over 200,000 pig cells from 20 tissues/organs. We comprehensively characterize the heterogeneity of cells in tissues and identify 234 cell clusters, representing 58 major cell types. In-depth integrative analysis of endothelial cells reveals a high degree of heterogeneity. We identify several functionally distinct endothelial cell phenotypes, including an endothelial to mesenchymal transition subtype in adipose tissues. Intercellular communication analysis predicts tissue- and cell type-specific crosstalk between endothelial cells and other cell types through the VEGF, PDGF, TGF-beta, and BMP pathways. Regulon analysis of single-cell transcriptome of microglia in pig and 12 other species further identifies MEF2C as an evolutionally conserved regulon in the microglia. Our work describes the landscape of single-cell transcriptomes within diverse pig organs and identifies the heterogeneity of endothelial cells and evolutionally conserved regulon in microglia.

  • Common variants in breast cancer risk loci predispose to distinct tumor subtypes

    • BackgroundGenome-wide association studies (GWAS) have identified multiple common breast cancer susceptibility variants. Many of these variants have differential associations by estrogen receptor (ER) status, but how these variants relate with other tumor features and intrinsic molecular subtypes is unclear.MethodsAmong 106,571 invasive breast cancer cases and 95,762 controls of European ancestry with data on 173 breast cancer variants identified in previous GWAS, we used novel two-stage polytomous logistic regression models to evaluate variants in relation to multiple tumor features (ER, progesterone receptor (PR), human epidermal growth factor receptor 2 (HER2) and grade) adjusting for each other, and to intrinsic-like subtypes.ResultsEighty-five of 173 variants were associated with at least one tumor feature (false discovery rate < 5%), most commonly ER and grade, followed by PR and HER2. Models for intrinsic-like subtypes found nearly all of these variants (83 of 85) associated at p < 0.05 with risk for at least one luminal-like subtype, and approximately half (41 of 85) of the variants were associated with risk of at least one non-luminal subtype, including 32 variants associated with triple-negative (TN) disease. Ten variants were associated with risk of all subtypes in different magnitude. Five variants were associated with risk of luminal A-like and TN subtypes in opposite directions.ConclusionThis report demonstrates a high level of complexity in the etiology heterogeneity of breast cancer susceptibility variants and can inform investigations of subtype-specific risk prediction.

  • Co-option of PPARα in the regulation of lipogenesis and fatty acid oxidation in CLA-induced hepatic steatosis

    • Nonalcoholic-fatty-liver-disease (NAFLD) is the result of imbalances in hepatic lipid partitioning and is linked to dietary factors. We demonstrate that conjugated linoleic acid (CLA) when given to mice as a dietary supplement, induced an enlarged liver, hepatic steatosis, and increased plasma levels of fatty acid (FA), alanine transaminase, and triglycerides. The progression of NAFLD and insulin resistance was reversed by GW6471 a small-molecule antagonist of peroxisome proliferator-activated receptor α (PPARα). Transcriptional profiling of livers revealed that the genes involved in FA oxidation and lipogenesis as two core gene programs controlled by PPARα in response to CLA and GW6471 including Acaca and Acads. Bioinformatic analysis of PPARα ChIP-seq data set and ChIP-qPCR showed that GW6471 blocks PPARα binding to Acaca and Acads and abolishes the PPARα-mediated local histone modifications of H3K27ac and H3K4me1 in CLA-treated hepatocytes. Thus, our findings reveal a dual role of PPARα in the regulation of lipid homeostasis and highlight its druggable nature in NAFLD.

  • Maternal high-protein diet modulates hepatic growth axis in weaning piglets by reprogramming the IGFBP-3 gene

    • PurposeThe aim of this study was to investigate the effects of maternal high dietary protein intake on the hepatic growth axis in offspring.MethodsFourteen primiparous purebred Meishan sows were fed either a standard-protein (SP, n = 7) diet or a high-protein (HP, 150% of SP, n = 7) diet during pregnancy. Offspring (one male and one female per group, n = 14) on day 70 of the embryonic stage and on days 1, 35 and 180 after birth were selected, weighed and killed. Serum samples were analyzed for Tch, insulin and insulin-like growth factor-binding protein 3 (IGFBP-3) levels. Liver samples were analyzed for IGFBP-3 and IGF-I mRNA expression by qRT-PCR and for IGFBP-3, IGF1R and growth hormone receptor (GHR) protein expression by Western blotting. The underlying mechanism of IGFBP-3 regulation was determined by methylated DNA immunoprecipitation (MeDIP) and chromatin immunoprecipitation (ChIP).ResultsHigh-protein exposure resulted in significantly higher body and liver weights of piglets, and it increased their serum T3 and T4 levels at birth and/or at weaning. Furthermore, the IGFBP-3 protein content in the liver and serum was significantly reduced in the HP-exposed weaning piglets, whereas at the transcriptional level IGFBP-3 mRNA expression was downregulated in the livers of HP group piglets. Finally, DNA hypermethylation and higher enrichment of the histone repressive marks H3K27me3 and H3K9me3 were observed.ConclusionsTaken together, these results suggest that a maternal high-protein diet during gestation epigenetically reprograms IGFBP-3 gene expression to modulate the hepatic growth axis in weaning piglets.

  • Effects of size distribution on social interactions and growth of juvenile black rockfish (Sebastes schlegelii)

    • Growth heterogeneity in rearing groups may complicate feeding operations and inducing cannibalism in cultured fish. To reduce these problems size grading has become a common rearing practice. To better understand the relationship between size grading and growth variation in cultured juvenile black rockfish (Sebastes schlegelii) we studied how size heterogeneity affected their growth performance (body weight, SGR, CV,), social interactions (feeding position, aggressive behavior) and endocrine responses (cortisol, growth hormone, 5-hydroxytryptamine). A batch of sibling fish (0.29-1.73 g) were selected and divided into three groups according to their similarity in size. Three types of size-structured groups were established each type containing two treatments with different fish numbers (n = 30 or n = 6): Small fish alone, small and medium fish mixed, small and large fish mixed. The experiment lasted for 30 days. In all treatments, regardless of the number of fish in the group, the growth rates of small-sized fish in size-homogeneous groups were lower than in size-heterogeneous groups. Consequently, size variation decreased significantly with time in size-heterogeneous groups whereas the opposite pattern was found in size-homogenous groups. Overall, growth heterogeneity tended to stabilize with time in all experimental groups. In the size-heterogeneous groups larger fish always occupied the central feeding area. Outside feeding period small fish avoided the central area, likely to avoid cannibalism, but when food was present small fish practiced a sneaky feeding activity. In general, aggressive interactions between small-sized fish were fiercer when larger fish were absent, and within size-heterogeneous groups aggressive interactions were more common between similar-sized larger individuals than between individuals of different size. The physiological analyses supported the growth and behavioural data. Cortisol levels were lower in the most size-heterogeneous groups than in the more homogenous groups whereas the growth hormone levels in the size-homogeneous group were significantly lower than in the more heterogeneous groups. In conclusion, the growth variation in juvenile black rockfish seems to be a consequence of intensive competition between similar-sized individuals rather than growth suppression of smaller in a size-related dominance hierarchy. Thus a size-grading practice where rearing groups retain an acceptable level of size heterogeneity that limits competition and cannibalism would produce the overall best result in terms of growth and survival.   • Effects of rolling motion on helical coil once-through steam generator thermal-hydraulic characteristics

    • Steam generators are essential for the safe, economical, and reliable operation of nuclear reactors. Helical Coil Once-through Steam Generators (HCOTSG) offer unique advantages over other types of steam generators for ship reactors or offshore platforms. To investigate the effect of rolling motion due to ocean conditions on the safe and economic operation of HCOTSG, a new model and a new code are developed in this paper. The program is developed and allows numerical simulation of HCOTSG under ocean conditions. The validity of the code was verified by comparing the simulation results with the design parameters of the Marine Reactor X (MRX) steam generator and the simulation results of other dedicated programs. The code was further used to perform the International Reactor Innovative and Secure (IRIS) transient operating conditions under typical rolling motions. Then, the influences of different swing directions, angles, periods, and positions of swing axes from the origin of the system operating parameters are analyzed. In the cases discussed in this paper, the following conclusions are obtained: (a) The direction of the swing significantly affects the system. The most dangerous situation is around the x-axis, and the case around the z-axis is the safest, in which the rolling situation has little effect on the system because the centripetal force is perpendicular to the tube wall, and the gravitational pressure drop is constant. (b) when the swing angle increases, except the fluctuation speed is faster, the fluctuation range of the parameters also increases. (c) when the swing period changes, the parameters’ fluctuation range also change (d) the orientation of the swing axis from the origin affects the magnitudes of the parameters’ changes (e) the distance of the swing axis from the origin affects the magnitude of the parameter variation and the farther the swing axis is from the origin, the greater the parameter fluctuation.

    • This paper develops a one-dimensional thermal–hydraulic analysis program to simulate the main thermal–hydraulic parameter changes of helical coil once-through steam generator (HCOTSG) under inlet thermal–hydraulic parameters perturbation in inclining and vertical conditions. The inclining process is divided into the process of rotating from the vertical position to the inclining position and the stable inclining process. The rotating process is regarded as swaying motion. The swaying and inclining motions are achieved by modifying the momentum equation. What's more, the code was verified through experiments on two-phase heat transfer and friction pressure drop, as well as comparisons with the design parameters of the HCOTSG of the International Reactor Innovative and Secure (IRIS) reactor. The results of the simulation indicate that the direction of incline has an obvious impact on the safety of the HCOTSG, with inclination towards the y-axis having the greatest impact. In the stable inclining process, an increase in the angle of inclining results in a rise in primary-side outlet temperature and a reduction in heat transfer, while the secondary-side pressure drop increases. Furthermore, the HCOTSG underwent testing with eight types of transient perturbations, including four sudden perturbations and four linear perturbations. Results indicated that during sudden perturbations, apart from the secondary-side pressure drop, there were no substantial differences in simulated results between the inclined and vertical states at the same time during the transition process. However, during linear perturbations, due to the slow changes in parameters, the differences between the inclined and vertical states at the same time were more distinct. Regardless of the perturbation type, the inclined state led to a deterioration in the dynamic condition of the system compared to the vertical state.

  • Holocene forcing of aeolian dust activity over the Tibetan Plateau and its surroundings

    • Aeolian deposits on the Tibetan Plateau (TP) and its surroundings provide crucial source materials for the Asian dust cycle, which significantly affects Asian and global ecosystems and climate. However, it is unclear how the dust dynamics of the TP and its surroundings are linked to Earth's climate system. To address this issue, we examined the grain size and accumulation rate of six Holocene aeolian sections on the southern TP (a new, well-dated high-resolution section, two relatively low-resolution sections, and three published sections) and combined them with equivalent aeolian sedimentary records from eastern arid central Asia. The results suggest that dust activity in both regions decreased during the early to middle Holocene and then increased in the late Holocene. We hypothesize that the primary drivers of Holocene dust activity in both regions are similar. Cold-season insolation, as the primary driving factor, combined with ice volume and atmospheric CO2 concentration, collectively controlled the regional temperature, which determined the near-surface wind intensity via its influence on the TP High and Siberian High, respectively, thus ultimately controlling the regional dust activity. In this context, we project that dust activity on the TP and its neighboring areas will decrease under warm scenarios in the 21st century. Overall, our findings provide an extensive overview of the past, present, and future scenarios of Asian dust activity, especially of the TP dust.

  • Heaving motion effect on the transient thermodynamic characteristics of helical coil once-through steam generators in mixed ocean conditions

    • Based on the finite difference method, the fixed boundary method, and a staggered grid method, this paper develops a one-dimensional thermal–hydraulic analysis code to simulate the effects of heave motion and more complex oceanic motions, namely heaving coupled with inclination and heaving coupled with swaying, on the thermal–hydraulic parameters of the International Reactor Innovative and Secure (IRIS) reactor's helical coil once-through steam generator (HCOTSG). The oceanic conditions were achieved by modifying the momentum equation. The validity and accuracy of the code were obtained by comparing the results with the design parameters of HCOTSG for two types of marine nuclear reactors, the SMART reactor and the IRIS reactor, and with the calculations of some similar international prediction programs. The simulation results indicate that (1) when only heave motion is considered, increasing the amplitude and reducing the period both result in a greater magnitude and a faster rate of change in the HCOTSG parameters. (2) When the combined motion of inclining and heaving is present, the dynamic performance of the composite motion inclining towards the positive x-axis deteriorates the most severely, while its thermodynamic conditions improve slightly. In this direction, the inclining angle has little influence on the thermal–hydraulic characteristics of HCOTSG. (3) When the combined motion of heaving and swaying is present, the dynamic performance is more severely affected under swaying motion towards the negative y-axis, despite a slight improvement in thermodynamic conditions. In this direction, as the swaying angle increases, the trend of parameter variation under swaying is similar but with a greater magnitude of change. The deterioration in dynamic performance becomes more severe, although there are small improvements in thermodynamic conditions. With the swaying period increasing, the maximum and minimum values of the secondary side pressure drop show little change. Additionally, the average thermodynamic conditions exhibit little variation.
