  • Robust estimation of bacterial cell count from optical density
  • 2020
  • Ingår i: Communications Biology. - : Springer Science and Business Media LLC. - 2399-3642. ; 3:1
  • Tidskriftsartikel (refereegranskat)abstract
    • Optical density (OD) is widely used to estimate the density of cells in liquid culture, but cannot be compared between instruments without a standardized calibration protocol and is challenging to relate to actual cell count. We address this with an interlaboratory study comparing three simple, low-cost, and highly accessible OD calibration protocols across 244 laboratories, applied to eight strains of constitutive GFP-expressing E. coli. Based on our results, we recommend calibrating OD to estimated cell count using serial dilution of silica microspheres, which produces highly precise calibration (95.5% of residuals <1.2-fold), is easily assessed for quality control, also assesses instrument effective linear range, and can be combined with fluorescence calibration to obtain units of Molecules of Equivalent Fluorescein (MEFL) per cell, allowing direct comparison and data fusion with flow cytometry measurements: in our study, fluorescence per cell measurements showed only a 1.07-fold mean difference between plate reader and flow cytometry data.

  • Individual nanostructure optimization in donor and acceptor phases to achieve efficient quaternary organic solar cells
  • 2019
  • Ingår i: Nano Energy. - : ELSEVIER. - 2211-2855 .- 2211-3282. ; 66
  • Tidskriftsartikel (refereegranskat)abstract
    • Fullerene derivative (PC71BM) and high crystallinity molecule (DR3TBDTT) are employed into PTB7-Th:FOIC based organic solar cells (OSCs) to cooperate an individual nanostructure optimized quaternary blend. PC71BM functions as molecular adjuster and phase modifier promoting FOIC forming "head-to-head" molecular packing and neutralizing the excessive FOIC crystallites. A multi-scale modified morphology is present thanks to the mixture of FOIC and PC71BM while DR3TBDTT disperses into PTB7-Th matrix to reinforce donors crystal-linity and enhance domain purity. Morphology characterization highlights the importance of individually optimizated nanostructures for donor and acceptor, which contributes to efficient hole and electron transport toward improved carrier mobilities and suppressed non-geminated recombination. Therefore, a power conversion efficiency of 13.51% is realized for a quaternary device which is 16% higher than the binary device (PTB7-Th:FOIC). This work demonstrates that utilizing quaternary strategy for simultaneous optimization of donor and acceptor phases is a feasible way to realize high efficient OSCs.

  • Association of Ultra-Processed Food Intake with Cardiovascular And Respiratory Disease Multimorbidity : A Prospective Cohort Study
  • 2023
  • Ingår i: Molecular Nutrition & Food Research. - : Wiley. - 1613-4133 .- 1613-4125. ; 67:11
  • Tidskriftsartikel (refereegranskat)abstract
    • SCOPE: Evidence suggests a positive association between ultra-processed food (UPF) consumption and the incidence of cardiovascular disease (CVD). We aimed to investigate associations between UPF intake and respiratory disease, CVD, and their multimorbidity in a large prospective cohort.METHODS AND RESULTS: Within the UK Biobank, participants who were free from respiratory disease or CVD at baseline and completed at least two times 24-h dietary records were included in this study. After adjusting for socioeconomic status and lifestyle factors, the hazard ratios (95% confidence interval) for each ten percent increase in UPF were 1.06 (1.04, 1.09) for CVD, 1.04 (1.02, 1.06) for respiratory disease, 1.15 (1.08, 1.22) for CVD mortality, and 1.06 (1.01, 1.12) for their multimorbidity, respectively. In addition, replacing 20% of UPF weight in diet with an equivalent proportion of unprocessed or minimally processed foods was estimated to be associated with 11% lower risk of CVD, 7% lower risk of respiratory disease, 25% lower risk of CVD mortality and 11% lower risk of CVD and respiratory disease multimorbidity.CONCLUSION: In this prospective cohort study, higher consumption of UPF was associated with higher risks of CVD and respiratory disease multimorbidity. Further longitudinal studies are needed to confirm our findings. This article is protected by copyright. All rights reserved.

  • Association of Ultraprocessed Food Consumption With Risk of Dementia : A Prospective Cohort
  • 2022
  • Ingår i: Neurology. - 1526-632X. ; 99:10, s. 1056-1066
  • Tidskriftsartikel (refereegranskat)abstract
    • BACKGROUND: There has been a growing body of evidence associating consumption of ultra-processed foods (UPF) with adverse health outcomes including depression, cardiovascular disease, all-cause mortality. However, whether UPF are associated with dementia is unknown. The authors investigated the associations between UPF and dementia incidence in UK biobank.METHODS: We included 72,083 participants (55 years or older) who were free from dementia at baseline and provided at least two times 24-h dietary assessments from the UK Biobank study. Follow-up occurred through March 2021. UPF were defined according to the NOVA classification. Incident all-cause dementia comprising Alzheimer's disease and vascular dementia was ascertained through electronic linkages to hospital and mortality records. Cox proportional hazards were used to estimate the association between the proportion (%) of UPF in the diet and the subsequent risk of dementia. In addition, substitution analysis was used to estimate the risk of dementia when substituting UPF with an equivalent proportion of unprocessed or minimally processed foods.RESULTS: During a total of 717,333 person-years of follow-up (median 10.0 years), 518 participants developed dementia, of which 287 developed Alzheimer's disease and 119 developed vascular dementia. In the fully adjusted model, consumption of UPF was associated with higher risk of dementia (hazard ratio (HR) for 10% increase in UPF: 1.25; 95% confidence interval (CI): 1.14, 1.37), Alzheimer's disease (HR: 1.14; 95% CI: 1.00, 1.30) and vascular dementia (HR: 1.28; 95% CI: 1.06, 1.55), respectively. In addition, replacing 10% of UPF weight in diet with an equivalent proportion of unprocessed or minimally processed foods was estimated to be associated with a 19% lower risk of dementia (HR: 0.81; 95% CI: 0.74, 0.89).CONCLUSIONS: In this prospective cohort study, higher consumption of UPF was associated with higher risk of dementia, while substituting unpr2ocessed or minimally processed foods for UPF was associated lower risk of dementia.

  • Data-Driven Controlled Synthesis of Oriented Quasi-Spherical CsPbBr3 Perovskite Materials
  • 2024
  • Ingår i: Angewandte Chemie International Edition. - : WILEY-V C H VERLAG GMBH. - 1433-7851 .- 1521-3773. ; 63:14
  • Tidskriftsartikel (refereegranskat)abstract
    • Controlled synthesis of lead-halide perovskite crystals is challenging yet attractive because of the pivotal role played by the crystal structure and growth conditions in regulating their properties. This study introduces data-driven strategies for the controlled synthesis of oriented quasi-spherical CsPbBr3, alongside an investigation into the synthesis mechanism. High-throughput rapid characterization of absorption spectra and color under ultraviolet illumination was conducted using 23 possible ligands for the synthesis of CsPbBr3 crystals. The links between the absorption spectra slope (difference in the absorbance at 400 nm and 450 nm divided by a wavelength interval of 50 nm) and crystal size were determined through statistical analysis of more than 100 related publications. Big data analysis and machine learning were employed to investigate a total of 688 absorption spectra and 652 color values, revealing correlations between synthesis parameters and properties. Ex situ characterization confirmed successful synthesis of oriented quasi-spherical CsPbBr3 perovskites using polyvinylpyrrolidone and Acacia. Density functional theory calculations highlighted strong adsorption of Acacia on the (110) facet of CsPbBr3. Optical properties of the oriented quasi-spherical perovskites prepared with these data-driven strategies were significantly improved. This study demonstrates that data-driven controlled synthesis facilitates morphology-controlled perovskites with excellent optical properties.

  • Acid suppressants use and risk of atherosclerotic cardiovascular disease in middle-aged and older adults
  • 2022
  • Ingår i: Atherosclerosis. - : Elsevier BV. - 0021-9150. ; 358, s. 47-54
  • Tidskriftsartikel (refereegranskat)abstract
    • Background and aims: Concerns regarding adverse events associated with the use of acid suppressants have increased. However, the impact of proton pump inhibitors (PPIs) and histamine‐2 receptor antagonists (H2RAs) on the risk of atherosclerotic cardiovascular disease (ASCVD) remains unknown. This study aimed to estimate the risk of ASCVD in association with the use of PPIs and H2RAs. Methods: This prospective cohort study included participants without cardiovascular diseases or anti-hypertensive treatment at baseline (2006–2010) in the UK Biobank. The outcomes were ASCVD and each subtype (coronary artery disease, myocardial infarction, peripheral artery disease, and ischemic stroke). The association was estimated by Cox proportional-hazards models. Results: Among 316,730 individuals (aged 50–88 years), during a median of 12.5 years of follow-up, we documented 13,503 (4.3%) incident ASCVD. Regular PPIs use was associated with a higher risk of ASCVD (HR: 1.16, 95% CI: 1.09–1.23) and every subtype of ASCVD. Among each type of PPIs, omeprazole (HR: 1.19, 95% CI: 1.11–1.28), lansoprazole (HR: 1.11, 95% CI: 1.02–1.22), and pantoprazole (HR: 1.40, 95% CI: 1.00–1.97) were associated with a higher risk of ASCVD. Stratification analysis showed that PPIs use was associated with a higher risk of ASCVD among individuals without indications of medications for PPIs. In addition, use of H2RAs was not related to the risk of ASCVD (HR: 0.97, 95% CI: 0.85–1.11). Conclusions: PPIs were associated with increased risk of ASCVD, particularly amongst participants without indications for medication. Our findings are of important practical significance and suggest that clinicians should be cautious in prophylactic use of PPIs.

  • Effect of psychotropics on the risk of COVID-19 in middle-aged and older adults
  • 2023
  • Ingår i: European Neuropsychopharmacology. - : Elsevier BV. - 0924-977X. ; 66, s. 67-77
  • Tidskriftsartikel (refereegranskat)abstract
    • Older adults have been markedly impacted by the coronavirus disease 19 (COVID-19) pandemic, and many reports have cited concerns regarding potential psychiatric sequelae of coronavirus disease (COVID-19), but the actual effects of psychotropics on the COVID-19 are unclear. In this study, multivariate logistic regression was used to evaluate associations between the prescription of psychotropics and the risk of SARS-CoV-2 infection, and COVID-19-related death among the participants who were tested for severe acute respiratory syndrome coronavirus-2 (SARS-CoV-2) before October 18, 2021, in UK Biobank. The psychotropics included 18 types of medications. Among 168,173 participants who underwent testing for SARS-CoV-2 RNA, 30,577 (18.2%) were positive, and 14,284 (8.5%) participants used psychotropics. Among 30,577 participants who were infected with SARS-CoV-2, 1,181 (3.9%) were COVID-19-related deaths, and 2,542 (8.3%) participants used psychotropics. In multivariate logistic regression, psychotropics use was significantly associated with the risk of SARS-CoV-2 infection (odds ratio [OR], 0.95; 95% confidence interval [CI], 0.88–0.98), and COVID-19-related death (OR, 0.78; 95% CI, 0.64–0.98). Interestingly, the use of diazepam was significantly associated with a 31% lower risk of SARS-CoV-2 infection (OR, 0.69; 95% CI, 0.53–0.88). The use of sertraline was significantly associated with a 89% lower risk of COVID-19-related death (OR, 0.11; 95% CI, 0.02–0.39). In conclusion, our findings suggested that the use of psychotropics was associated with a lower risk of SARS-CoV-2 infection and COVID-19-related deaths.

  • Climate change drives rapid decadal acidification in the Arctic Ocean from 1994 to 2020
  • 2022
  • Ingår i: Science. - : American Association for the Advancement of Science (AAAS). - 0036-8075 .- 1095-9203. ; 377:6614, s. 1544-1550
  • Tidskriftsartikel (refereegranskat)abstract
    • The Arctic Ocean has experienced rapid warming and sea ice loss in recent decades, becoming the first open-ocean basin to experience widespread aragonite undersaturation [saturation state of aragonite (Warag) < 1]. However, its trend toward long-term ocean acidification and the underlying mechanisms remain undocumented. Here, we report rapid acidification there, with rates three to four times higher than in other ocean basins, and attribute it to changing sea ice coverage on a decadal time scale. Sea ice melt exposes seawater to the atmosphere and promotes rapid uptake of atmospheric carbon dioxide, lowering its alkalinity and buffer capacity and thus leading to sharp declines in pH and Warag. We predict a further decrease in pH, particularly at higher latitudes where sea ice retreat is active, whereas Arctic warming may counteract decreases in Warag in the future.

  • A hybrid Markov-based model for human mobility prediction
  • 2018
  • Ingår i: Neurocomputing. - : Elsevier BV. - 0925-2312 .- 1872-8286. ; 278:SI, s. 99-109
  • Tidskriftsartikel (refereegranskat)abstract
    • Human mobility behavior is far from random, and its indicators follow non-Gaussian distributions. Predicting human mobility has the potential to enhance location-based services, intelligent transportation systems, urban computing, and so forth. In this paper, we focus on improving the prediction accuracy of non-Gaussian mobility data by constructing a hybrid Markov-based model, which takes the non-Gaussian and spatio-temporal characteristics of real human mobility data into account. More specifically, we (1) estimate the order of the Markov chain predictor by adapting it to the length of frequent individual mobility patterns, instead of using a fixed order, (2) consider the time distribution of mobility patterns occurrences when calculating the transition probability for the next location, and (3) employ the prediction results of users with similar trajectories if the recent context has not been previously seen. We have conducted extensive experiments on real human trajectories collected during 21 days from 3474 individuals in an urban Long Term Evolution (LTE) network, and the results demonstrate that the proposed model for non-Gaussian mobility data can help predicting people’s future movements with more than 56% accuracy.

  • Oral traditional Chinese medication for adhesive small bowel obstruction
  • 2012
  • Ingår i: Cochrane Database of Systematic Reviews. - 1469-493X. ; :5
  • Forskningsöversikt (refereegranskat)abstract
    • Background Small bowel obstruction (SBO) is one of the most common emergent complications of general surgery. Intra-abdominal adhesions are the leading cause of SBO. Because surgery can induce new adhesions, non-operative management is preferred in the absence of signs of peritonitis or strangulation. Oral traditional Chinese herbal medicine has long been used as a non-operative therapy to treat adhesive SBO in China. Many controlled trials have been conducted to investigate its therapeutic value in resolving adhesive SBO. Objectives The aim of this review was to assess the efficacy and safety of oral traditional Chinese medicine (TCM) for adhesive small bowel obstruction. Search methods We searched the following databases, without regard to language or publishing restrictions: the Cochrane Central Register of Controlled Trials (CENTRAL), MEDLINE, EMBASE, Chinese Biomedical Database (CBM), China National Knowledge Infrastructure/Chinese Academic Journals full-text Database (CNKI), and VIP (a full-text database of Chinese journals). The searches were conducted in November 2011. Selection criteria Randomised controlled trials and quasi-randomised controlled trials comparing Chinese medicines administered orally, via the gastric canal, or both with a placebo or conventional therapy in participants diagnosed with adhesive SBO were considered. We also considered trials of TCM (oral administration, gastric tube perfusion, or both) plus conventional therapy compared with conventional therapy alone for patients with adhesive SBO. Studies addressing the safety and efficacy of oral traditional Chinese medicinal agents in the treatment of adhesive SBO were also considered. Data collection and analysis Two authors collected the data independently. We assessed the risk of bias according to the following methodological criteria: random sequence generation, allocation concealment, blinding, incomplete outcome data, selective outcome reporting and other sources of bias. Dichotomous data are presented as risk ratios (OR) and 95% confidence intervals (CI); continuous outcomes are presented as mean differences (MD) and 95% CIs. The data analyses were carried out using Review Manager 5.1. For cases in which necessary information was not reported in the paper, we contacted the primary authors for additional information. Main results Five randomised trials involving 664 participants were analysed. Five different herbal medicines were tested in these trials, including Huo-Xue-Tong-Fu decoction, Xiao-Cheng-Qi-Tang decoction, a combination of Xiao-Cheng-Qi-Tang and Si-Jun-Zi-Tang decoctions, Chang-Nian-Lian-Song-Jie-Tang decoction, and Fufang-Da-Cheng-Qi-Tang decoction. There were variations in the tested herbal compositions and methods of medicine administration. The main outcomes reported in the trials were effects on abdominal pain, abdominal distension, constipation defection, time of first defecation after treatment, and reoperation rate during the course of the disease. Secondary outcomes selected for this review were not available, including complications such as small bowel perfusion (bowel resection, system complications, and other possible complications), length of hospital stay, cost of hospitalisation, and time from admission to surgical intervention. The results of five trials showed that patients receiving TCM combined with conventional therapy seemed to have improved outcomes compared with patients receiving conventional treatment alone (OR 4.24, 95% CI 2.83 to 6.36). However, we cannot conclusively determine the efficacy of TCM in this review due to inadequate reporting, low methodological quality, and the prevalence of various biases in the reviewed studies. Furthermore, because none of the reviewed trials discussed adverse events, we could not evaluate the safety of TCM for adhesive SBO patients. All trials were conducted and published in China. Authors' conclusions Although many studies have assessed the use of TCM products for adhesive SBO, most were excluded from this review due to their methodological limitations. This systematic review did not find sufficient evidence to support the objective efficacy and safety of TCM for patients with adhesive SBO. The positive evidence should be interpreted with caution given the insufficient number of studies with large sample sizes, the absence of well-designed, high-quality trials, and the lack of safety information. Therefore, further studies with larger sample sizes and high-quality, randomised, and controlled trials are necessary to produce more accurate and meaningful data on the efficacy of Chinese herbal medicines for adhesive SBO.

  • 2015
  • Ingår i: Scientific Reports. - : Springer Science and Business Media LLC. - 2045-2322. ; 5
  • Tidskriftsartikel (refereegranskat)abstract
    • We report on the prediction of the zinc-blende structure BP into a novel C2/m phase from 113 to 208 GPa which possesses zigzag phosphorus chain structure, followed by another P4(2)/mnm structure above 208 GPa above using the particle-swarm search method. Strong electron-phonon coupling lambda in compressed BP is found, in particular for C2/m phase with the zigzag phosphorus chain, which has the highest lambda (0.56-0.61) value among them, leading to its high superconducting critical temperature T-c (9.4 K-11.5 K), which is comparable with the 4.5 Kto 13 Kvalue of black phosphorus phase I (orthorhombic, Cmca). This is the first system in the boron phosphides which shows superconductivity from the present theoretical calculations. Our results show that pressure-induced zigzag phosphorus chain in BP exhibit higher superconducting temperature T-C, opening a new route to search and design new superconductor materials with zigzag phosphorus chains.

  • Global hypo-methylation in a proportion of glioblastoma enriched for an astrocytic signature is associated with increased invasion and altered immune landscape
  • 2022
  • Ingår i: eLIFE. - : eLife Sciences Publications Ltd. - 2050-084X. ; 11
  • Tidskriftsartikel (refereegranskat)abstract
    • We describe a subset of glioblastoma, the most prevalent malignant adult brain tumour, harbouring a bias towards hypomethylation at defined differentially methylated regions. This epigenetic signature correlates with an enrichment for an astrocytic gene signature, which together with the identification of enriched predicted binding sites of transcription factors known to cause demethylation and to be involved in astrocytic/glial lineage specification, point to a shared ontogeny between these glioblastomas and astroglial progenitors. At functional level, increased invasiveness, at least in part mediated by SRPX2, and macrophage infiltration characterise this subset of glioblastoma.

  • Tidskriftsartikel (refereegranskat)abstract
    • Halide double perovskites managed by metal doping approach can exhibit dual emission colors, which have been considered as promising multicolor luminescent materials. However, an independent and stable emission at yellow region is missing owing to limited doping candidates, hindering the further commercialization of multicolor luminescence applications in double perovskites. In this work, we successfully obtain stable multicolor emission with PLQE (photoluminescence quantum yield) as high as 78% through developing the CuI doping strategy in Sb-Cs2NaInCl6. By introducing a high CuI feed ratio in airtight autoclave to compete the oxidization effect, the oxidization of CuI into CuII (detrimental factor for high PLQE due to serious quenching effect) is largely suppressed. With changing the CuI feed ratio, at least four distinct emission colors ranging from blue, purple, pink to yellow can be realized via changing the excitation wavelength. Depending on tunable multicolor emission, we further demonstrate the promise of our co-doped double perovskites in anti-counterfeiting technology and multicolor lighting devices. Our results open the way for enriching the optical applications of double perovskites based on multicolor emission.

  • 2024
  • Ingår i: Cell Reports. - : Elsevier. - 2211-1247. ; 43:6
  • Tidskriftsartikel (refereegranskat)abstract
    • Glioblastomas are the most common malignant brain tumors in adults; they are highly aggressive and heterogeneous and show a high degree of plasticity. Here, we show that methyltransferase-like 7B (METTL7B) is an essential regulator of lineage specification in glioblastoma, with an impact on both tumor size and invasiveness. Single-cell transcriptomic analysis of these tumors and of cerebral organoids derived from expanded potential stem cells overexpressing METTL7B reveal a regulatory role for the gene in the neural stem cell-to-astrocyte differentiation trajectory. Mechanistically, METTL7B downregulates the expression of key neuronal differentiation players, including SALL2, via post-translational modifications of histone marks.

  • Ingår i: IEICE Transactions on Information and Systems. - 0916-8532. ; E100D:6, s. 1303-1315
  • Tidskriftsartikel (refereegranskat)abstract
    • Image quality assessment (IQA) plays an important role in quality monitoring, evaluation and optimization for image processing systems. However, current quality-Aware feature extraction methods for IQA can hardly balance accuracy and complexity. This paper introduces multi-order local description into image quality assessment for feature extraction. The first-order structure derivative and high-order discriminative information are integrated into local pattern representation to serve as the quality-Aware features. Then joint distributions of the local pattern representation are modeled by spatially enhanced histogram. Finally, the image quality degradation is estimated by quantifying the divergence between such distributions of the reference image and those of the distorted image. Experimental results demonstrate that the proposed method outperforms other state-of-The-Art approaches in consideration of not only accuracy that is consistent with human subjective evaluation, but also robustness and stability across different distortion types and various public databases. It provides a promising choice for image quality assessment development.

  • Energy management and performance analysis of an off-grid integrated hydrogen energy utilization system
  • 2024
  • Ingår i: Energy Conversion and Management. - 0196-8904. ; 299
  • Tidskriftsartikel (refereegranskat)abstract
    • In integrated hydrogen energy utilization systems, due to the low efficiency of hydrogen/electricity conversion, coordination of energy management and efficient waste heat recovery is required to optimize performance. To address this challenge, this paper presents a comprehensive and sophisticated modeling and energy management strategy to enhance the off-grid energy utilization rate while prolonging the main components' lifetime. The developed model incorporates multiphase flow and heat transport balance for electricity and heat production, enabling a highly accurate representation of real-world behaviors of the system. The proposed off-grid operation strategy is complemented by a designed heat recovery scheme, ensuring the use of energy resources and waste heat. In addition, the proposed energy management strategy monitors the real-time status of each subsystem, actively reducing the number of harmful start-stop cycles of the hydrogen production system, thereby mitigating short-term power impacts and delaying its aging. Specifically, the voltage degradation of the reduction cell is reduced from 4.67 mV to 4.48 mV, the energy utilization rate is increased from 47.6 % to 53.9 %, and the energy efficiency of fuel cells significantly increases from 53.6 % to 78.1 %.

  • Understanding the Role of Triplet-Triplet Annihilation in Non-Fullerene Acceptor Organic Solar Cells

    • Non-fullerene acceptors (NFAs) have enabled power conversion efficiencies exceeding 19% in organic solar cells (OSCs). However, the open-circuit voltage of OSCs remains low relative to their optical gap due to excessive non-radiative recombination, and this now limits performance. Here, an important aspect of OSC design is considered, namely management of the triplet exciton population formed after non-geminate charge recombination. By comparing the blends PM6:Y11 and PM6:Y6, it is shown that the greater crystallinity of the NFA domains in PM6:Y11 leads to a higher rate of triplet-triplet annihilation (TTA). This is attributed to the four times larger ground state dipole moment of Y11 versus Y6, which improves the long range NFA out-of-plane ordering. Since TTA converts a fraction of the non-emissive triplet states into bright singlet states, it has the potential to reduce non-radiative voltage losses. Through a kinetic analysis of the recombination processes under 1-Sun illumination, a framework is provided for determining the conditions under which TTA may improve OSC performance. If these could be satisfied, TTA has the potential to reduce non-radiative voltage losses by up to several tens of millivolts.

  • Epigenetic–smoking interaction reveals histologically heterogeneous effects of TRIM27 DNA methylation on overall survival among early-stage NSCLC patients

  • Ingår i: Molecular Oncology. - : Wiley. - 1574-7891 .- 1878-0261. ; 14:11, s. 2759-2774

    • Tripartite motif containing 27 (TRIM27) is highly expressed in lung cancer, including non-small-cell lung cancer (NSCLC). Here, we profiled DNA methylation of lung adenocarcinoma (LUAD) and lung squamous cell carcinoma (LUSC) tumours from 613 early-stage NSCLC patients and evaluated associations between CpG methylation of TRIM27 and overall survival. Significant CpG probes were confirmed in 617 samples from The Cancer Genome Atlas. The methylation of the CpG probe cg05293407TRIM27 was significantly associated with overall survival in patients with LUSC (HR = 1.65, 95% CI: 1.30–2.09, P = 4.52 × 10−5), but not in patients with LUAD (HR = 1.08, 95% CI: 0.87–1.33, P = 0.493). As incidence of LUSC is associated with higher smoking intensity compared to LUAD, we investigated whether smoking intensity impacted on the prognostic effect of cg05293407TRIM27 methylation in NSCLC. LUSC patients had a higher average pack-year of smoking (37.49LUAD vs 54.79LUSC, P = 1.03 × 10−19) and included a higher proportion of current smokers than LUAD patients (28.24%LUAD vs 34.09%LUSC, P = 0.037). cg05293407TRIM27 was significantly associated with overall survival only in NSCLC patients with medium–high pack-year of smoking (HR = 1.58, 95% CI: 1.26–1.96, P = 5.25 × 10−5). We conclude that cg05293407TRIM27 methylation is a potential predictor of LUSC prognosis, and smoking intensity may impact on its prognostic value across the various types of NSCLC.

  • Epigenome-wide three-way interaction study identifies a complex pattern between TRIM27, KIAA0226, and smoking associated with overall survival of early-stage NSCLC

    • The interaction between DNA methylation of tripartite motif containing 27 (cg05293407TRIM27) and smoking has previously been identified to reveal histologically heterogeneous effects of TRIM27 DNA methylation on early-stage non-small-cell lung cancer (NSCLC) survival. However, to understand the complex mechanisms underlying NSCLC progression, we searched three-way interactions. A two-phase study was adopted to identify three-way interactions in the form of pack-year of smoking (number of cigarettes smoked per day × number of years smoked) × cg05293407TRIM27 × epigenome-wide DNA methylation CpG probe. Two CpG probes were identified with FDR-q ≤ 0.05 in the discovery phase and P ≤ 0.05 in the validation phase: cg00060500KIAA0226 and cg17479956EXT2. Compared to a prediction model with only clinical information, the model added 42 significant three-way interactions using a looser criterion (discovery: FDR-q ≤ 0.10, validation: P ≤ 0.05) had substantially improved the area under the receiver operating characteristic curve (AUC) of the prognostic prediction model for both 3-year and 5-year survival. Our research identified the complex interaction effects among multiple environment and epigenetic factors, and provided therapeutic target for NSCLC patients.

  • Knee-point-conscious battery aging trajectory prediction of lithium-ion based on physics-guided machine learning
  • 2024

    • Early prediction of aging trajectories of lithium-ion (Li-ion) batteries is critical for cycle life testing, quality control, and battery health management. Although data-driven machine learning (ML) approaches are well suited for this task, unfortunately, relying solely on data is exceedingly time-consuming and resource-intensive, even in accelerated aging with complex aging mechanisms. This challenge is rooted in the highly complex and time-varying degradation mechanisms of Li-ion battery cells. We propose a novel method based on physics-guided machine learning (PGML) to overcome this issue. First, electrode-level physical information is incorporated into the model training process to predict the aging trajectory’s knee point (KP). The relationship between the identified KP and the accelerated aging behavior is then explored, and an aging trajectory prediction algorithm is developed. The prior knowledge of aging mechanisms enables a transfer of valuable physical insights to yield accurate KP predictions with small data and weak correlation feature relationship. Based on a Li[NiCoMn]O 2 cell dataset, we demonstrate that only 14 cells are needed to train a PGML model for achieving a lifetime prediction error of 2.02% using the data of the first 50 cycles. In contrast, at least 100 cells are needed to reach this level of accuracy without the physical insights.

  • Film Formation Kinetics of Polymer Donor and Nonfullerene Acceptor Active Layers During Printing Out of 1,2,4-Trimethylbenzene in Ambient Conditions
  • 2023
  • Ingår i: Solar RRL. - : Wiley. - 2367-198X. ; 7:6
  • Tidskriftsartikel (refereegranskat)abstract
    • Slot-die coating is a promising upscaling fabrication method to promote commercialization in the field of organic solar cells. Herein, the nonfullerene active layer blend of a conjugated polymer PffBT4T-2OD and a small molecule acceptor EH-IDTBR, which is printed out of the nonhalogenated solvent 1,2,4-trimethylbenzene, is studied. The film formation kinetics of the active layer PffBT4T-2OD:EH-IDTBR is probed in terms of the temporal evolutions in morphology as well as molecular conformation and aggregation as revealed by in situ grazing-incidence small angle X-ray scattering and UV–vis spectroscopy during the film printing process. A five-regime mesoscale domain growth process is observed in the active layer from the liquid state to the final dry state. The solvent evaporation-induced domain growth is accompanied with molecular stacking in a distinct J-type aggregation of the acceptor and a slight H-type aggregation of the donor molecules. The printed active layers exhibit an edge-on dominated PffBT4T-2OD and a face-on dominated EH-IDTBR crystallite structure. Compared to the neat PffBT4T-2OD and EH-IDTBR films, in the active layer, the crystallite structure deviates slightly in lattice spacing.
