  • A meta-analysis of methane mitigation potential of feed additives evaluated in vitro

    • A systematic literature review of in vitro studies was performed to identify methane (CH4) mitigation interventions with a potential to reduce CH4 emission in vivo. Data from 277 peer-reviewed studies published between 1979 and 2018 were reviewed. Individual CH4 mitigation interventions were classified into 14 categories of feed additives based on their type, chemical composition, and mode of action. Response variables evaluated were absolute CH4 emission (number of treatment means comparisons = 1,325); total volatile fatty acids (VFA; n = 1,007), acetate (n = 783), propionate (n = 792), and butyrate (n = 776) concentrations; acetate to propionate ratio (A:P; n = 675); digestibility of dry matter (DM; n = 489), organic matter (OM; n = 277), and neutral detergent fiber (NDF; n = 177). Total gas production was used as an explanatory variable in the model for CH4 production. Relative mean difference between treatment and control means reported in the studies were calculated and used for statistical analysis. Robust variance estimation method was used to analyze the effects of CH4 mitigation interventions. In vitro CH4 production was decreased by antibodies (−38.9%), chemical inhibitors (−29.2%), electron sinks (−18.9%), essential oils (−18.2%), plant extracts (−14.5%), plants inclusion (−11.7%), saponins (−14.8%), and tannins (−14.5%). Overall effects of direct fed microbials, enzymes, macroalgae, and organic acids supplementation did not affect CH4 production in the current meta-analysis. When considering the effects of individual mitigation interventions containing a minimum number of 4 degrees of freedom within feed additives categories, Enterococcus spp. (i.e., direct fed microbial), nitrophenol (i.e., electron sink), and Leucaena spp. (i.e., tannins) decreased CH4 production by 20.3, 27.1, and 23.5%, respectively, without extensively, or only slightly, affecting ruminal fermentation and digestibility of nutrients. It should be noted, however, that although the total number of publications (n = 277) and treatment means comparisons (n = 1,325 for CH4 production) in the current analysis were high, data for most mitigation interventions were obtained from less than 5 observations (e.g., maximum number of observations was 4, 7, and 22 for nitrophenol, Enterococcus spp., and Leucaena spp., respectively), because of limited data available in the literature. These should be further evaluated in vitro and in vivo to determine their true potential to decrease enteric CH4 production, yield, and intensity. Some mitigation interventions (e.g., magnesium, Heracleum spp., nitroglycerin, β-cyclodextrin, Leptospermum pattersoni, Fructulus Ligustri, Salix caprea, and Sesbania grandiflora) decreased in vitro CH4 production by over 50% but did not have enough observations in the database. These should be more extensively investigated in vitro, and the dose effect must be considered before adoption of mitigation interventions in vivo.

  • Dairy cow longevity and farm economic performance: Evidence from Swedish dairy farms

    • The longevity of dairy cows is mainly determined by farmers' subjective culling decisions and can be linked to the environmental impact of dairy production and to the social acceptance of the industry. Still, the economic impacts of dairy cow longevity are not well understood. The aim of this study was to examine how herd average dairy cow longevity is related to the farm economic outcome. We used 3 indicators of economic outcome: technical efficiency, profitability, and average milk yield per cow. We used 2 indicators of dairy cow longevity: average herd length of life and average herd length of productive life. The study was based on a unique and detailed dataset from Swedish dairy agriculture, where herd-management data from the national dairy herd recording scheme were combined with farm-level economic variables from the Swedish Farm Accountancy Survey, for a total of 1,959 observations from 2010 to 2018. The regression results highlight that both measures of average herd dairy cow longevity have an overall positive and significant association with farm-level economic performance. These associations had an inverted U-shape, which implies that the association is first positive and then declines. Descriptive statistics indicate that the point where the maximum economic performance is attained varied across the economic indicators. Our results are relevant for individual dairy farmers and their advisors, who are interested in understanding how herd average longevity relates to economic performance on the farms. Our results are also important from a greater sustainability perspective, because linking them to previous knowledge about the environmental and social sustainability benefits of keeping cows longer highlights longevity-associated trade-offs between those benefits and the farm economic outcomes.

  • Rapid eye movement sleep time in dairy cows changes during the lactation cycle

    • The importance of rest and sleep is well established; we know, for example, that lack of sleep impairs immune function in rats and increases pain sensitivity in humans. However, little is known about sleep in dairy cows, but a lack of rest and sleep is discussed as a possible welfare problem in cattle. A first step toward a better understanding of sleep in dairy cows is to quantify the time cows spend awake and asleep in different stages of lactation. Using electrophysiological recordings on 7 occasions in wk -2, 2, 7, 13, 22, 37, and 45 in relation to calving, we investigated changes in rapid eye movement (REM) sleep time as well as non-rapid eye movement (NREM) sleep, drowsing, awake, and rumination in 19 dairy cows of the Swedish Red breed kept in single pens with ad libitum access to feed and water. The recordings on wk 2 and 45 were conducted during the dry period, and all others during lactation. The PROC MIXED procedure in SAS (SAS Institute Inc., Cary, NC) was used to test for significant differences in REM, NREM, drowsing, awake, and rumination between the different stages of lactation cycle. Pairwise comparisons between all recording occasions showed that total REM sleep duration was shorter for cows in wk 2 relative to calving compared with wk -2, and the number of REM sleep bouts were fewer in wk 2 compared with wk -2. The HEM sleep was recorded during both the day (0500-2100 h) and night (2100-0500 h), but predominantly performed at night compared with daytime, and the bout duration was longer during nighttime compared with daytime. A tendency was observed for time spent in NREM sleep to be shorter in wk 2 relative to calving compared with wk -2. The duration spent drowsing was shorter for cows in wk 2 and 13 relative to calving compared with wk -2. We found no effect of stage of lactation cycle on the duration of awake or ruminating. Our study is the first to assess sleep distribution during a lactation cycle, and our results show that stage of lactation is important to consider when moving forward with sleep investigations in dairy cows. The shortest REM sleep duration was found for cows 2 wk after calving and longest 2 wk before calving, and the difference was due a higher number of REM sleep bouts in the recording 2 wk before calving. The REM sleep and rumination predominantly occurred at night but were recorded during both day and night.

    • Dairy cow longevity combines all functional traits and is thought to be especially important in organic production, which is an established, increasing part of Swedish dairy production, representing approximately 6% of the market. The aim of this study was to compare dynamics in culling reasons between organic and conventional production and to analyze genotype by environment interactions for longevity. The data contained information from all organic herds with information available from official recording (n = 402) and from approximately half of the conventional herds (n = 5,335). Records from Swedish Holsteins (n = 155,379) and Swedish Red cows (n = 160,794) that had their first calf between January 1998 and September 2003 were included. The opportunity period for longevity was at least 6 yr. Six longevity traits were defined: length of productive life; survival through first, second, and third lactations; fertility-determined survival; and udder health-determined survival. Twenty codes were used to describe the cause of culling, and these were divided into 8 groups: udder health, low fertility, low production, leg problems, metabolic diseases, other diseases, other specified causes, and unspecified cause. The main reason for culling cows in organic herds was poor udder health, whereas for cows in conventional herds it was low fertility. Furthermore, the shift in main culling reason from fertility, which was most common in first lactation regardless of production system, to udder health occurred at a lower age in organic production. Heritabilities and genetic correlations for the longevity traits expressed in organic and conventional herds were estimated from a bivariate animal model. The genetic correlations were close to unity (>0.88), except for fertility-determined survival in the Swedish Red breed (0.80). Heritabilities were low to moderate, and no clear pattern was identified for production system or breed. In general, the results indicate that farmers' culling criteria differ between organic and conventional production. Different preferences may influence the need for alternative selection indexes for organic production, with different weightings of traits, or a separate breeding program. However, no genotype by environment interaction of importance was found between the production systems.

  • Planned Behavior, Social Networks, and Perceived Risks: Understanding Farmers' Behavior toward Precision Dairy Technologies

    • Precision dairy tools (PDT) can provide timely information on individual cow's physiological and behavioral parameters, which can lead to more efficient management of the dairy farm. Although the economic rationale behind the adoption of PDT has been extensively discussed in the literature, the socio-psychological aspects related to the adoption of these technologies have received far less attention. Therefore, this paper proposes a socio-psychological model that builds upon the theory of planned behavior and develops hypotheses regarding cognitive constructs, their interaction with the farmers' perceived risks and social networks, and their overall influence on adoption. These hypotheses are tested using a generalized structural equation model for (a) the adoption of automatic milking systems (AMS) on the farms and (b) the PDT that are usually adopted with the AMS. Results show that adoption of these technologies is affected directly by intention, and the effects of subjective norms, perceived control, and attitudes on adoption are mediated through intention. A unit increase in perceived control score is associated with an increase in marginal probability of adoption of AMS and PDT by 0.05 and 0.19, respectively. Subjective norms are associated with an increase in marginal probability of adoption of AMS and PDT by 0.009 and 0.05, respectively. These results suggest that perceived control exerts a stronger influence on adoption of AMS and PDT, particularly compared with their subjective norms. Technology-related social networks are associated with an increase in marginal probability of adoption of AMS and PDT by 0.026 and 0.10, respectively. Perceived risks related to AMS and PDT negatively affect probability of adoption by 0.042 and 0.16, respectively, by having negative effects on attitudes, perceived self-confidence, and intentions. These results imply that integrating farmers within knowledge-sharing networks, minimizing perceived risks associated with these technologies, and enhancing farmers' confidence in their ability to use these technologies can significantly enhance uptake.

    • Ripening is the most crucial process step in cheese manufacturing and constitutes multiple biochemical alterations that describe the final cheese quality and its perceived sensory attributes. The assessment of the cheese-ripening process is challenging and requires the effective analysis of a multitude of biochemical changes occurring during the process. This study monitored the biochemical and sensory attribute changes of paraffin wax-covered long-ripening hard cheeses (n = 79) during ripening by collecting samples at different stages of ripening. Near-infrared hyperspectral (NIR-HS) imaging, together with free amino acid, chemical composition, and sensory attributes, was studied to monitor the biochemical changes during the ripening process. Orthogonal projection-based multivariate calibration methods were used to characterize ripening-related and orthogonal components as well as the distribution map of chemical components. The results approve the NIR-HS imaging as a rapid tool for monitoring cheese maturity during ripening. Moreover, the pixelwise evaluation of images shows the homogeneity of cheese maturation at different stages of ripening. Among the chemical compositions, fat content and moisture are the most important variables correlating to NIR-HS images during the ripening process.

  • Herd-level risk factors associated with cow mortality in Swedish dairy herds

    • An increase in on-farm mortality (euthanasia and death) in dairy herds has been reported in several countries in the last decade. This does not only imply possible problems with animal welfare, but it also causes economic losses to the farmer. The objective of this study was to evaluate time trends in on-farm dairy cow mortality in Sweden and identify potential herd-level risk factors. Data were retrieved on all Swedish dairy herds enrolled in the milk recording scheme between 2002 and 2010. Herds with a herd size of <20 cows or a mortality rate (MR) of >40 dead or euthanized cows per 100 cow-years were excluded. Two different models were used: 1 multiple-year analysis, which included 6,898 herds during the period 2002 to 2010 and 1 single-year analysis including 4,252 herds for the year 2010, where other variables that were not present during the entire multiple year study were analyzed. The outcome variable was the number of euthanized and dead cows per year and season. A negative binomial regression model, adjusted for clustering within herd, was applied to both models. Fixed effects in the multiple-year analysis were breed, calving interval, herd size, milk yield, region, season, pasture period, and year. The fixed effects in the single-year analysis were breed, calving interval, conventional versus organic farming, herd size, housing system, milk yield, region, and season. The results demonstrated that MR gradually increased from 5.1 to 6.6 events per 100 cow-years during the study period. Swedish MR are consequently on par with, or even greater than, MR among dairy herds in other comparable countries. Higher mortality was associated with larger herd size, longer calving intervals, and herds that had Swedish Holstein as the predominant breed. Lower mortality was observed in herds with a higher herd average milk yield, during the fall and winter, and in organically managed herds. There were regional differences in mortality. An interaction between herd size and season was found in both models. Also, an interaction between housing system and milk yield was found in the single-year analysis. This first assessment of on-farm mortality in Swedish dairy herds confirmed that the MR has increased over the last few years. The study also identified some herd-level risk factors.

  • Effects of dry period length on metabolic status, fertility, udder health, and colostrum production in 2 cow breeds

    • Shortening the dry period improves postpartum energy balance, which has potential positive effects on metabolic health. This concept has been mainly studied in Holstein cows. The aim of this study was to assess the effect of a short dry period of 4 wk, compared with a standard dry period of 8 wk, on the metabolic status, progesterone profiles, health, and colostrum quality of dairy cows of 2 breeds, Swedish Red (SR) and Swedish Holstein (SH), not treated with antibiotics at dry off. The IgG uptake in calves was also studied to reflect the colostrum quality when shortening the dry period. Cows of both SH and SR were blocked by breed and parity and then randomly allocated to a short dry period of 4 wk (4W, n = 43) or a conventional dry period of 8 wk (8W, n = 34). Blood samples were collected wk 8, 4, 2, 1 and 1, 2, 3, 4, 6, 9, and 12 relative to calving. Prepartum, cows with a 4-wk dry period had higher concentrations of nonesterified fatty acids and lower concentrations of insulin-like growth factor-1. and insulin than 8W cows. Postpartum, plasma concentration of nonesterified fatty acids was lower, whereas plasma insulin and insulin-like growth factor-1 tended to be higher for 4W cows than for 8W cows. Plasma concentration of beta-hydroxybutyrate did not differ between dry period lengths. Swedish Holstein cows with a 4W dry period responded with a lower concentration of insulin prepartum than SR and SH on an 8W dry period. The dry period length had no effect on the proportion of disturbed progesterone profiles; disturbed progesterone profiles occurred in 30% of the 4W cows and 47% of the 8W cows. In this trial, only 48.8% of the SR cows had a normal progesterone profile, which differed from the SH where 76.5% had a normal profile. Fertility-related diseases (endometritis, pyometra, anestrus, ovarian cyst) did not differ between the 2 dry period groups: 21% in the 8W group versus 12% in the 4W group, whereas mastitis tended to be more common: 26% of the 4W cows versus 9% of the 8W cows. A short dry period resulted in less colostrum but with a higher content of protein and somatic cell count. Calves were fed colostrum from their dam, and the IgG and total protein in plasma did not differ between calves to mothers with different a dry period length. Shortening the dry period could improve metabolic status in cows of both SH and SR breed postpartum, without compromising the colostrum quality. Health and progesterone profiles were not affected by the dry period length for SH or SR in this study.

  • The occurrence of noncoagulating milk and the association of bovine milk coagulation properties with genetic variants of the caseins in 3 Scandinavian dairy breeds

    • Substantial variation in milk coagulation properties has been observed among dairy cows. Consequently, raw milk from individual cows and breeds exhibits distinct coagulation capacities that potentially affect the technological properties and milk processing into cheese. This variation is largely influenced by protein composition, which is in turn affected by underlying genetic polymorphisms in the major milk proteins. In this study, we conducted a large screening on 3 major Scandinavian breeds to resolve the variation in milk coagulation traits and the frequency of milk with impaired coagulation properties (noncoagulation). In total, individual coagulation properties were measured on morning milk collected from 1,299 Danish Holstein (DH), Danish Jersey (DJ), and Swedish Red (SR) cows. The 3 breeds demonstrated notable interbreed differences in coagulation properties, with DJ cows exhibiting superior coagulation compared with the other 2 breeds. In addition, milk samples from 2% of DH and 16% of SR cows were classified as noncoagulating. Furthermore, the cows were genotyped for major genetic variants in the alpha(S1)- (CSN1S1), beta- (CSN2), and kappa-casein (CSN3) genes, revealing distinct differences in variant frequencies among breeds. Allele I of CSN2, which had not formerly been screened in such a high number of cows in these Scandinavian breeds, showed a frequency around 7% in DH and DJ, but was not detected in SR. Genetic polymorphisms were significantly associated with curd firming rate and rennet coagulation time. Thus, CSN1S1 C, CSN2 B, and CSN3 B positively affected milk coagulation, whereas CSN2 A(2), in particular, had a negative effect. In addition to the influence of individual casein genes, the effects of CSN1S1-CSN2-CSN3 composite genotypes were also examined, and revealed strong associations in all breeds, which more or less reflected the single gene results. Overall, milk coagulation is under the influence of additive genetic variation. Optimal milk for future cheese production can be ensured by monitoring the frequency of unfavorable variants and thus preventing an increase in the number of cows producing milk with impaired coagulation. Selective breeding for variants associated with superior milk coagulation can potentially increase raw milk quality and cheese yield in all 3 Scandinavian breeds.

  • Detecting and predicting changes in milk homogeneity using data from automatic milking systems.

    • To ensure milk quality and detect cows with signs of mastitis, visual inspection of milk by prestripping quarters before milking is recommended in many countries. An objective method to find milk changed in homogeneity (i.e., with clots) is to use commercially available inline filters to inspect the milk. Due to the required manual labor, this method is not applicable in automatic milking systems (AMS). We investigated the possibility of detecting and predicting changes in milk homogeneity using data generated by AMS. In total, 21,335 quarter-level milk inspections were performed on 5,424 milkings of 624 unique cows on 4 farms by applying visual inspection of inline filters that assembled clots from the separate quarters during milking. Images of the filters with clots were scored for density, resulting in 892 observations with signs of clots for analysis (77% traces or mild cases, 15% moderate cases, and 8% heavy cases). The quarter density scores were combined into 1 score indicating the presence of clots during a single cow milking and into 2 scores summarizing the density scores in cow milkings during a 30-h sampling period. Data generated from the AMS, such as milk yield, milk flow, conductivity, and online somatic cell counts, were used as input to 4 multilayer perceptron models to detect or predict single milkings with clots and to detect milking periods with clots. All models resulted in high specificity (98-100%), showing that the models correctly classified cow milkings or cow milking periods with no clots observed. The ability to successfully classify cow milkings or cow periods with observed clots had a low sensitivity. The highest sensitivity (26%) was obtained by the model that detected clots in a single milking. The prevalence of clots in the data was low (2.4%), which was reflected in the results. The positive predictive value depends on the prevalence and was relatively high, with the highest positive predictive value (72%) reached in the model that detected clots during the 30-h sampling periods. The misclassification rate for cow milkings that included higher-density scores was lower, indicating that the models that detected or predicted clots in a single milking could better distinguish the heavier cases of clots. Using data from AMS to detect and predict changes in milk homogeneity seems to be possible, although the prediction performance for the definitions of clots used in this study was poor.

  • Longitudinal study of Staphylococcus aureus genotypes isolated from bovine clinical mastitis

    • Bovine clinical mastitis is an important problem for the dairy industry, and Staphylococcus aureus is a common mastitis-causing pathogen in many countries. Detailed knowledge on genetic variation of Staph. aureus strains within the bovine population, including changes over time, can be useful for mastitis control programs, because severity of disease and effects on milk production are at least partly strain-associated. Therefore, the major aim of this study was to compare sequence types of Staph. aureus isolated from cases of bovine clinical mastitis from 2002 to 2003 with sequence types of a more recent set of isolates collected from 2013 to 2018, using core genome multi-locus sequence typing (cgMLST). We also wanted to compare antibiotic resistance genes of isolates from the 2 sets, to identify changes that may have occurred over time in the Staph. aureus population. A total of 157 isolates of Staph. aureus, almost equally distributed between the 2 time periods, were subjected to high-throughput sequencing and cgMLST. The results showed that the most prevalent sequence types found among the 2002 to 2003 isolates belonged to the clonal complexes CC97, CC133, and CC151, and that those complexes still dominated among the isolates from 2013 to 2018. However, a population shift from CC133 to CC97 and CC151 over time was observed. Likewise, no important differences in prevalence of antibiotic resistance genes were found between the 2 sets of isolates. As expected, genes belonging to the major facilitator superfamily of transporter proteins, and multidrug and toxic compound extrusion transporters, were very common. Moreover, several genes and mutations conferring resistance to fosfomycin were present, but not in CC97 isolates. The beta-lactamase gene blaZ was found in only 3 out of 81 isolates from 2002 to 2003 and 1 out of 76 isolates in 2013 to 2018. In conclusion, the results indicate that mastitis-associated Staph. aureus strains circulating among dairy cows in Sweden exhibit a remarkable genotypic persistence over a time frame of close to 15 yr.

    • This study examined the feasibility of using pregnancy-associated glycoproteins (PAG) in milk within breeding for pregnancy maintenance and assessed the genetic variation in pregnancy loss traits. A total of 374,206 PAG samples from 41,889 Swedish Red (SR) and 82,187 Swedish Holstein (SH) cows were collected at monthly test-day milkings in 1,119 Swedish herds. Pregnancy status was defined based on PAG levels and confirmed by data on artificial insemination (AI), calving, and culling from d 1 postinsemination to calving. Pregnancy loss traits were defined as embryonic loss (diagnosed 28 d to 41 d after AI), fetal loss (42 d after AI until calving), and total pregnancy loss. Least squares means (± standard error, %) and genetic parameters were estimated using mixed linear models. Heritability was estimated to be 0.02, 0.02, and 0.03 for embryonic loss, fetal loss, and total pregnancy loss, respectively. Cows with pregnancy loss had lower PAG concentrations than cows which successfully maintained pregnancy and calved. PAG recording was limited to monthly test-day milking, resulting in low estimated embryonic loss (17.5 ± 0.4 and 18.7 ± 0.4 in SR and SH, respectively) and higher fetal loss (32.8 ± 0.5 and 35.1 ± 0.5 in SR and SH, respectively). Pregnancy loss might have occurred earlier but remained undetected until the next test-day milking, when it was recorded as fetal loss rather than embryonic loss. Estimated genetic correlation between embryonic and fetal pregnancy loss traits and classical fertility traits were in general high. Identification of novel genetic traits from PAG data can be highly specific, as PAG are only secreted by the placenta. Thus, PAG could be useful indicators in selection to genetically improve pregnancy maintenance and reduce reproductive losses in milk production. Further studies are needed to clarify how these results could be applied in breeding programs concurrent with selection for classical fertility traits.

    • This study compared the abilities of virgin heifer genomically enhanced breeding values (GEBV) and parent average breeding values (PA) to predict future cow performance. To increase confidence in genomic technology among farmers, a clear demonstration of the relationship between genomic predictions and future phenotypes is needed. We analyzed 12 different traits in first parity, including production, conformation, fer-tility, and other functional traits. Phenotype data were obtained from national milk recording schemes and breeding values from the Nordic Cattle Genetic Evalu-ation. Direct genomic breeding values were calculated using genomic BLUP and combined with traditional breeding values, using bivariate blending. The data covered 14,862 Red Dairy Cattle, 17,145 Holstein, and 7,330 Jersey genotyped virgin heifers born between 2013 and 2015 in Denmark, Finland, and Sweden. Phe-notypes adjusted for systematic environmental effects were used as measures of cow performance. Two meth-ods were used to compared virgin heifer GEBV and PA regarding their ability to predict future cow per-formance: (1) correlations between breeding values and adjusted phenotypes, (2) ranking cows into 4 quartiles for their virgin heifer GEBV or PA, and calculating actual cow performance for each quartile. We showed that virgin heifer GEBV predicted cow performance significantly better than PA for the vast majority of analyzed traits. The correlations with adjusted pheno-types were 38 to 136% higher for GEBV than for PA in Red Dairy Cattle, 42 to 194% higher for GEBV in Holstein, and 11 to 78% higher for GEBV in Jersey. The relative change between GEBV bottom and top quartiles compared with that between PA bottom and top quartiles ranged from 9 to 261% for RDC, 42 to 138% for Holstein, and 4 to 90% for Jersey. Hence, farmers in Denmark, Finland, and Sweden can have confidence in using genomic technology on their herds.

    • In this study, we explored mating allocation in Hol-stein using genomic information for 24,333 Holstein fe-males born in Denmark, Finland, and Sweden. We used 2 data sets of bulls: the top 50 genotyped bulls and the top 25 polled genotyped bulls on the Nordic total merit scale. We used linear programming to optimize economic scores within each herd, considering genetic level, genetic relationship, semen cost, the economic impact of genetic defects, polledness, and 0-casein. We found that it was possible to reduce genetic relation-ships and eliminate expression of genetic defects with minimal effect on the genetic level in total merit index. Compared with maximizing only Nordic total merit in-dex, the relative frequency of polled offspring increased from 13.5 to 22.5%, and that of offspring homozygous for 0-casein (A2A2) from 66.7 to 75.0% in one genera-tion, without any substantial negative impact on other comparison criteria. Using only semen from polled bulls, which might become necessary if dehorning is banned, considerably reduced the genetic level. We also found that animals carrying the polled allele were less likely to be homozygous for 0-casein (A2A2) and more likely to be carriers of the genetic defect HH1. Hence, adding economic value to a monogenic trait in the economic score used for mating allocation sometimes negatively affected another monogenetic trait. We rec-ommend that the comparison criteria used in this study be monitored in a modern genomic mating program.

We used linear programming to optimize different economic scores within each herd, considering genetic level, semen cost, the economic impact of recessive genetic defects, and genetic relationships. We selected 9,841 genotyped females born in Denmark, Finland, or Sweden in 2019 for mating allocations. We used 2 different pedigree relationship coefficients, the first tracing the pedigree 3 generations back from the parents of the potential mating and the second based on all available pedigree information. We used 3 different genomic relationship coefficients, 1 SNP-by-SNP genomic relationship and 2 based on shared genomic segments. We found high correlations (≥0.83) between the pedigree and genomic relationship measures. The mating results showed that it was possible to reduce the different genetic relationships between parents with minimal effect on genetic level. Including the cost of known recessive genetic defects eliminated expression of genetic defects. It was possible to reduce genomic relationships between parents with pedigree measures, but it was best done with genomic measures. Linear programming maximized the economic score for all herds studied within seconds, which means that it is suitable for implementation in mating software to be used by advisors and farmers.

    • The aim of this study was to define endocrine fertility traits from in-line milk progesterone (P4) records and to estimate genetic parameters for these traits. Correlations of classical fertility (calving interval and calving to first service) and milk production traits with endocrine fertility traits were also estimated. In-line milk P4 records (n = 160,952) collected from June 2009 through November 2013 for 2,273 lactations of 1,561 Holstein-Friesian cows in 12 commercial herds in the Netherlands were analyzed for (the log of) the number of days from calving till commencement of luteal activity (1nC-LA), proportion of samples between 25 and 60 d in milk with luteal activity (PLA), presence or absence of luteal activity for a cow between 25 and 60 d in milk, interval from commencement of luteal activity to first service (CLAFS), first luteal phase length, length of first interluteal interval, and length of first interovulatory interval. Milk P4 records were sampled, on average, every 2 d. Genetic parameters were estimated using a mixed linear animal model. Heritability estimates ( SE) of endocrine fertility traits were 0.12 +/- 0.05 for 1nC-LA, 0.12 0.05 for PLA, and 0.11 +/- 0.06 for CLAFS, and their repeatability estimates were 0.29 +/- 0.04, 0.21 +/- 0.04, and 0.15 +/- 0.06, respectively. The genetic correlation of 1nC-LA with PLA was 0.91 +/- 0.06 and with CLAFS was 0.56 +/- 0.25. The genetic correlations of 1nC-LA were 0.26 +/- 0.33 with calving interval and 0.37 +/- 0.21 with calving to first service. Genetic correlations of the milk production traits with 1nC-LA ranged from 0.04 to 0.18 and 0.07 to 0.65 with classical fertility traits. The phenotypic correlations of all endocrine fertility traits with milk production traits were close to zero (0.01 to 0.07). This study shows that in-line P4 records can be used to define and explore several heritable endocrine fertility traits in dairy cows and might help in selection for improved fertility.

    • Our hypothesis was that grazing time, the number of days (duration) and number of hours per day, affected claw health. From Swedish freestall herds that fulfilled our criteria of claw-trimming routines, 201 herds were randomly selected for a telephone interview regarding grazing management. Herd data were retrieved from the Swedish Official Milk Recording Scheme. Claw disorders to be analyzed were recorded at maintenance claw trimming before and after the grazing period and included mild and severe dermatitis, severe heel-horn erosion, and sole ulcer (including severe sole hemorrhage). Any remark included one or more of these recorded disorders. The odds for having a recorded claw disorder at the autumn trimming in relation to grazing management, as well as to herd- and cow-related parameters, was tested using multilevel logistic regression models. The final statistical analysis included 17,600 cows in 174 herds, which were distributed from the south to the north of Sweden with decreasing length of mandatory grazing period because of climate. Grazing duration was statistically associated with the risk of sole ulcer, but it was not linear. However, grazing duration was not statistically associated with the odds for any remark, dermatitis, or heel-horn erosion. The odds for dermatitis were lower with access to pasture for 24 h compared with either day or night access. Otherwise, the number of hours that the animals had access to grazing per day was not significantly associated with any of the other analyzed claw disorders. Higher pasture stocking density (number of cow hours per day per hectare) was associated with a higher odds for dermatitis and sole ulcer. For all recorded claw disorders, the highest odds for having a disorder after the grazing period were consistently when the cow had the same claw disorder before the release to pasture. The positive effects of grazing on claw health were less than expected, and the previous known effects of breed, days in milk, parity, production system, housing environment, and management were verified for most claw disorders. If cows in today's loose housing systems have a more restrictive grazing than cows in tie-stall herds previously experienced, one cannot expect as strong an effect even if grazing is mandatory in all Swedish dairy cattle. Despite some positive effects of grazing, good stall environment and management during the housing period seem to be more important to obtain good claw health.

    • Grass silage-based diets often result in poor nitrogen utilization when fed to dairy cows. Perennial ryegrass cultivars with high concentrations of water-soluble carbohydrates (WSC) have proven potential for correcting this imbalance when fed fresh, and have also been shown to increase feed intake, milk production, and N utilization. The possibility of achieving corresponding effects with silage-based diets was investigated in change-over experiments in an incomplete block design with 16 (yr 1) or 12 (yr 2) Swedish Red dairy cows in mid lactation. Measurements on N excretion and rumen parameters were performed on subgroups of 8 and 4 cows, respectively. In yr 1, 2 ryegrass cultivars (standard = Fennema; high-WSC = Aberdart) and 2 cuts (first and second) were compared. In all treatments, ryegrass silage was mixed 75/25 on a dry matter (DM) basis, with red clover silage before feeding out. In yr 2, 1 basic mixture from the different cuts of these 2 cultivars was used and experimental factors were red clover silage inclusion (25 or 50%) and sucrose addition (0 or 10%) on a silage DM basis. Differences in WSC concentration in the silage mixtures in yr 1 were minor, whereas the differences between cuts were more substantial: 100 compared with 111 g/kg of DM for first-cut silage and 39 compared with 47 g/kg of DM for second-cut silage. The silages fed in yr 2 had a WSC concentration of 115 or 102 g/kg of DM (25 or 50% red clover, respectively), but when sucrose was added WSC concentration reached 198 and 189 g/kg of DM, respectively. Milk production (kg/d) did not differ between treatments in either year. Red clover inclusion to 50% of silage DM increased milk protein. Nitrogen efficiency (milk N/feed N) increased from 0.231 to 0.254 with sucrose inclusion in yr 2 (average for the 2 red clover levels). Overall rumen pH was 5.99 and increased sucrose level did not affect pH level or daily pH pattern. Sucrose addition reduced neutral detergent fiber digestibility, particularly at higher inclusion rates of clover. Rumen pool of total purines did not differ between treatments, nor did protein production assessed from urinary allantoin. The NorFor feed evaluation model overestimated digestibility of neutral detergent fiber and N, but underestimated N excretion in feces. We concluded that addition of WSC to dairy cow diets at levels up to 3 kg of WSC per day (> 14% of DM) does not dramatically affect cow performance.

    • Providing more space per animal, soft bedding, and free roaming in animal housing systems is widely presumed to be beneficial for the welfare of the animals. This observational study aimed to investigate the basis of this assumption in free-walk housing systems (FWS) for dairy cows in Europe. The dairy cattle Welfare Quality assessment protocol was adapted for application to FWS, and the focus was on animal-based measures, from individual cow scoring to comfort around resting. The study was conducted on 41 farms [21 FWS and 20 cubicle housing (CH)] from 6 European countries (Germany, the Netherlands, Italy, Austria, Slovenia, and Sweden) displaying a variety of management systems. A total of 4,036 animals were scored. We found differences in animal welfare under different management conditions. The hindquarters and lower hind legs of cows from FWS were dirtier than those of cows in CH, but we found no difference in the dirtiness of udders or teats. Cows from FWS showed fewer hairless patches in all body areas except the neck; fewer lesions in the lower hind legs and hindquarters; and less swelling in the lower hind legs, flanks, and carpus than cows from CH. The prevalence of sound cows appeared to be higher in FWS, and moderate lameness prevalence was lower compared with CH. We found no difference in the prevalence of severe lameness between systems. We conducted a total of 684 observation sessions of comfort around resting, consisting of 830 lying down and 849 rising up movements. Cows in FWS took less time to lie down, had less difficulty rising up, and had fewer collisions with the environment during both behaviors than cows in CH. Cows lay partly or completely outside the supposed lying area less frequently in FWS than in CH. Cows in FWS adopted comfortable lying positions more often compared with CH, showing a higher occurrence of long and wide positions than cows in CH. Short positions were more common in FWS, and narrow positions were slightly more common in CH. We found large variations in animal-based measures between study herds and within housing systems. However, the observed patterns associated with each system demonstrated differences in cow scoring and comfort around resting. This study shows that a wide range of good and bad management practices exist in FWS, especially related to cow hygiene.

    • Reduction of milk yield is one of the principal components in the cost of mastitis. However, past research into the association between milk yield and mastitis indicators is limited. Past research has not been based on online or in-line daily measurements and has not fully explored nonlinearity and the thresholds at which milk yield starts to decrease. In dairy herds with automated milking systems equipped with sensors, mastitis indicators of individual cows are measured on an intraday frequency, which provides unprecedented avenues to explore such effects in detail. The aim of this observational study was primarily to investigate the nonlinear associations of lactate dehydrogenase (LDH), electrical conductivity (EC), and somatic cell count (SCC) with milk yield at various stages of lactation, parity, and mastitis chronicity status (i.e., whether the cow had SCC >= 200,000 SCC/mL for the last 28 d). We also investigated thresholds at which mastitis indicators (LDH, EC, and SCC) started to be negatively associated with milk yield. We used data from 21 automated milking system herds measuring EC and online SCC. Of these herds, 7 of the 21 additionally measured online LDH. We operationalized milk yield as milk synthesis rate in kilograms per hour. Applying a generalized additive model, we estimated the milk synthesis rate as a function of the 3 mastitis indicators for 3 different subgroups based on parity, stage of lactation, and mastitis chronicity. Partial dependence plots of the mastitis indicators were used to evaluate the milk synthesis rate to study if the milk synthesis rate was associated with mastitis indicators at a specific level. Results showed that milk synthesis rate decreased with increasing SCC, LDH, and EC, but in a nonlinear fashion. The thresholds at which milk synthesis rate started to decrease were 2.5 LnSCC (12,000 SCC/mL) to 3.75 LnSCC (43,000 SCC/mL), 0 to 1 LnLDH (1-2.7 U/L), and 5.0 to 6.0 mS/cm for EC. Additionally, another substantial decrease of milk synthesis rate was observed at thresholds of 5.625 LnSCC (277,000 SCC/mL) and 3 LnLDH (20 LDH U/L) but not for EC. Having chronic mastitis decreased milk synthesis rate in all models. The identified nonlinearities between mastitis indicators and milk synthesis rate should be incorporated in statistical models for more accurate estimations of milk loss due to mastitis.

    • In automatic milking systems (AMS), sensors can measure cow behavior and milk composition at every milking. The aim of this observational study of previously collected data was to gain insight into the differences in dynamics of udder inflammation indicators between cows that recover and those that do not recover after detection of an initial inflammation. Milk diversion (milk separated from the bulk tank and thus indicating farmer intervention), conductivity, and somatic cell count (SCC) data from 4 wk before the initial inflammation to 12 wk after the initial inflammation were used to analyze 2,584 cases of udder inflammation. An udder inflammation case was defined as an initial observation of SCC >= 200,000 cells/mL as well as 1 additional SCC measurement >200,000 cells/mL within 10 d after the initial case, among other requirements. The data originated from 15 AMS herds in 6 countries. Four subsets of cows were created based on whether milk was diverted after the initial inflammation and whether the udder inflammation cases recovered, using a 10-d rolling average SCC threshold of 200,000 cells/mL and checking whether this rolling mean was below the threshold within 90 d after the initial inflammation as the indication of recovery. This formed the following subsets of cow lactations: milk diverted-recovered, milk diverted-not recovered, no milk diverted-not recovered, no milk diverted-recovered. Thresholds of 100,000 SCC/mL and 300,000 SCC/mL for the definition of case and recovery were also applied in a sensitivity analysis but with no substantial difference in results. Linear mixed models were used for each subset to study the variation in SCC (natural logarithm of SCC divided by 1,000) and sigma-conductivity (natural logarithm of standard deviation of quarter conductivities). When observing the fraction of cows with SCC <200,000 cells/mL in the recovery subsets, most cows recovered within 20 d after the initial inflammation. In the recovery subsets, both sigma-conductivity and SCC stabilized, mostly within 3 to 4 wk after the initial inflammation. sigma-Conductivity stabilized above the pre-onset level in all subsets and did not show a clear increase in the no-milk-diverted subgroups, whereas SCC stabilized closer to the pre-onset level. Overall, this study indicated a cutoff point between nonchronic and chronic changes in indicators 3 to 4 wk after the initial inflammation for SCC and sigma-conductivity.

    • A meta-analysis based on an individual-cow data set was conducted to investigate the effects of between-cow variation and related animal variables on predicted CH4 emissions from dairy cows. Data were taken from 40 change-over studies consisting of a total of 637 cow/period observations. Animal production and rumen fermentation characteristics were measured for 154 diets in 40 studies; diet digestibility was measured for 135 diets in 34 studies, and ruminal digestion kinetics was measured for 56 diets in 15 studies. The experimental diets were based on grass silage, with cereal grains or by-products as energy supplements, and soybean or canola meal as protein supplements. Average forage:concentrate ratio across all diets on a dry matter basis was 59:41. Methane production was predicted from apparently fermented substrate using stoichiometric principles. Data were analyzed by mixed-model regression using diet and period within experiment as random effects, thereby allowing the effect of experiment, diet, and period to be excluded. Dry matter intake and milk yield were more repeatable experimental measures than rumen fermentation, nutrient outflow, diet digestibility, or estimated CH4 yield. Between-cow coefficient of variation (CV) was 0.010 for stoichiometric CH4 per mol of volatile fatty acids and 0.067 for predicted CH4 yield (CH4/dry matter intake). Organic matter digestibility (OMD) also displayed little between-cow variation (CV = 0.013), indicating that between-cow variation in diet digestibility and rumen fermentation pattern do not markedly contribute to between cow-variation in CH4 yield. Digesta passage rate was much more variable (CV = 0.08) between cows than OMD or rumen fermentation pattern. Increased digesta passage rate is associated with improved energetic efficiency of microbial N synthesis, which partitions fermented substrate from volatile fatty acids and gases to microbial cells that are more reduced than fermented carbohydrates. Positive relationships were observed between CH4 per mol of volatile fatty acids versus OMD and rumen ammonia N concentration versus OMD; and negative relationships between the efficiency of microbial N synthesis versus OMD and digesta passage rate versus OMD, suggesting that the effects of these variables on CH4 yield were additive. It can be concluded that variations in OMD and efficiency in microbial N synthesis resulting from variations in digesta passage contribute more to between-animal variation in CH4 emissions than rumen fermentation pattern.

    • This study evaluated the effects of gradual replacement of a mixture of late-cut grass silage (LS) and barley with early-cut grass silage (ES) on milk production, CH4 emissions, and N utilization in Swedish Red cows. Two grass silages were prepared from the same primary growth of timothy grass sward but harvested 2 wk apart [11.0 and 9.7 MJ of metabolizable energy/kg of dry matter (DM)]. Four diets, fed as a total mixed ration, were formulated to meet the metabolizable energy and protein requirements of 35 kg of energy-corrected milk (ECM) by gradually replacing a mixture of LS and barley with ES (0, 33, 67, and 100% of the forage component of the diet), whereas the proportion of barley decreased from 47.2 to 26.6% of diet DM. Expeller canola meal was used as a protein supplement. Sixteen Swedish Red cows were used in 4 replicated 4 x 4 Latin squares. Cows were offered diets ad libitum and milked twice daily. Each period of 28 d comprised 14 d of diet adaptation followed by 14 d of data collection. Intake and milk yield were recorded daily, and milk samples were collected on d 19 to 21 and d 26 to 28 of each period. Diet digestibility was determined by grab sampling using indigestible neutral detergent fiber as an internal marker. Gas emissions were measured using the Green Feed system (C-Lock Inc., Rapid City, SD). Dry matter intake (DMI) linearly decreased from 22.6 to 19.3 kg/d as the proportion of ES increased in the diet. The ECM yield did not differ among treatments, but milk protein yield decreased with increasing proportion of ES in the diet. Because of reduced DMI with increasing ES, feed efficiency (ECM/DMI) improved with an increased proportion of ES in the diet. Nitrogen efficiency (milk N/N intake) did not change despite a linear increase in milk urea N concentration from 9.7 (LS alone) to 11.9 mg/dL (ES alone) with graded replacement of LS and barley by ES in the diet. Lower DMI responses in ES diets were partly compensated for by increased organic matter digestibility (656 g/kg of DM for LS alone; 715 g/kg of DM for ES alone) related to improved forage digestibility at early harvesting. Total CH4 emissions and CH4 intensity (CH4/ECM) were not influenced by diet, but CH4 yield (CH4/DMI) increased linearly from 19.5 to 23.0 g/kg of DMI with greater inclusion of ES in the diet. In conclusion, replacing LS and barley with ES improved the conversion of feed to milk without increasing CH4 emissions or compromising N efficiency.

    • The objective of this experiment was to quantify the effects of graded replacement of late-harvested grass silage and barley with early-harvested silage on nutrient digestion and rumen fermentation. Four experimental diets were fed to 4 multiparous rumen-cannulated Nordic Red cows in 4 x 4 Latin square design with 21-d periods. Dietary treatments consisted of late-cut grass silage (LS) and rolled barley, which was gradually replaced with early-cut grass silage [ES; 0, 33, 67, and 100% of the forage component (ES + LS) of the diet]. With increased proportion of ES in the diet, the proportion of barley decreased from 47.2 to 26.6% on a dry matter basis. Early-and late-cut silages were harvested at 2-wk intervals (predicted concentrations of metabolizable energy 11.0 and 9.7 MJ/kg of dry matter). The 4 diets were formulated to support the same milk production. Nutrient flows were quantified using omasal sampling technique applying the triple-marker method (Cr, Yb, and indigestible neutral detergent fiber) and N-15 as a microbial marker. Feed intake decreased with graded replacement of LS and barley with ES, but milk production was not influenced by diet. Digestibility of nutrients improved with graded addition of ES in the diet with the greatest difference observed in digestibility of neutral detergent fiber (NDF) and potentially digestible NDF (pdNDF). The results suggested that improved cell wall digestibility with graded level of ES in the diet was partly related to higher intrinsic digestibility of ES than LS, and partly due to negative associative effects with an increased proportion of LS and barley in the diet. Efficiency of microbial N synthesis was not influenced by the diet, but ruminal protein degradability increased with ES in the diet. Rumen fermentation pattern was not affected by the diet despite large difference in the profile of dietary carbohydrates. Rumen pool size of NDF and pdNDF, and ruminal turnover time of NDF decreased with graded addition of ES in the diet, whereas digestion rate of pdNDF improved. The results of this study indicate that increased CH4 yield in a parallel production study with graded addition of ES in the diet were more related to greater ruminal and total digestibility of organic matter than to the changes in rumen fermentation pattern.

    • Staphylococcus aureus is a common udder pathogen of dairy cows that often causes herd problems. Various mastitis control programs have been used to combat the problem but have not always been efficient in preventing new Staph. aureus infections, indicating the presence of possible sources of infection other than those traditionally considered. Therefore, the aim of the study was to identify potential sources of infection relevant for Staph. aureus mastitis within 5 dairy herds with udder health problems caused by Staph. aureus. Samples were collected from milk of lactating cows, from body sites, and from the environment of lactating cows, dry cows, late pregnant heifers, young heifers 4 to 12 mo old, and heifer calves 0 to 3 mo old. Isolates of Staph. aureus were identified and compared using pulsed-field gel electrophoresis. Four to 7 unique Staph. aureus pulsotypes were found within each herd, with one strain predominating in milk in each herd. The milk pulsotypes were also frequently isolated in body samples, especially on hock skin, and in the immediate environment of lactating cows, and were sometimes found in other animal groups, especially in dry cows and heifer calves 0 to 3 mo old. The prevalence of Staph. aureus in milk and other types of samples varied markedly between herds. Staphylococcus aureus isolates with genotypes indistinguishable from those found in milk also dominated in extra-mammary sites within the dairy herds studied, and hock skin was identified as an important reservoir of Staph. aureus. The results contribute new knowledge necessary to improve strategies for udder health control in herds.

    • The objective of this study was to investigate how useful data from automatic milking systems used in commercial herds are for genetic analysis of milkability traits. Data were available from 4,968 Swedish Holstein and Swedish Red cows over a span of 5 yr (2004-2009) from 19 herds. The analyzed milkability traits were average flow rate, box time, milking interval, and number of milkings per day. Variance components were estimated for genetic, permanent environmental, and residual effects in first and later (second and third) lactations, and were used for estimation of heritabilities and repeatablilites. The experiences of the data quality and editing procedures showed that almost half of the data and about a quarter of the cows had to be excluded from the analyses due to incomplete or inconsistent information. However, much more data are available than is needed for accurate genetic parameter estimations. For the genetic analysis, a repeatability animal model was used that included the fixed effects of herd, year and season, lactation month, and milk yield. The repeatability coefficients were at a high level: highest for average flow rate, with estimates between 0.8 and 0.9. The estimated heritability coefficients were in the range of 0.37 to 0.48, 0.21 to 0.44, 0.09 to 0.26, and 0.02 to 0.07 for average flow rate, box time, milking interval, and number of milkings, respectively. The results from the present study unraveled large genetic variation in milkability traits. The genetic parameter estimates were well in agreement with previous studies of milkability, which proves the feasibility of using data from automatic milking systems for genetic analysis.

    • The overall objective of this study was to assess the use of in-line recorded milkability information from dairy herds with conventional milking parlors (CMP) and from herds with automatic milking systems (AMS) for genetic evaluation. Some genetic parameters were previously studied on AMS data for 2,053 Swedish Holstein (SH) and 1,749 Swedish Red (SR) cows in 19 herds. These data were combined in the present paper with milkability information from 74 herds with CMP, including 11,123 SH cows and 7,554 SR cows. Genetic parameters were estimated for the CMP data and genetic correlations were estimated between milkability traits measured in the 2 systems. Average flow rate and milking time were derived and used as similar milkability traits for both systems, whereas box time was used only for AMS herds. Estimated heritabilities were in the range from 0.24 to 0.49. Even though the traits were differently defined in the 2 milking systems, the corresponding traits recorded in AMS and CMP were genetically closely related (0.93-1.00). Similarly, close genetic relationships were shown between milkability traits in different lactations in both breeds (0.93-0.99). Thus, it should be possible to treat milkability traits in different lactations and from different milking systems as the same traits in genetic evaluations. The various milkability traits were also highly genetically correlated, indicating that the inclusion of just one trait in the genetic selection program would efficiently select for milkability without the need to consider all measures. Comparisons of repeatability and random regression models, combining all information from the 2 systems for genetic evaluation, were done to find the most suitable model for genetic evaluation purposes. Even though the random regression models were favored in the formal model tests to evaluate suitability, correlation coefficients between test-days within lactation were high (0.7-0.8) and small differences in breeding values resulted among different models. That would indicate that a few test-days per cow would produce accurate breeding values for milkability.

    • Cow-calf contact (CCC) systems, although beneficial in many respects, introduce additional challenges to collect reliable data on milk production, which is important to assess individual cow efficiency and dairy farm profitability. Apart from weighing calves before and after each feeding, the amount of saleable milk lost due to calf suckling is practically impossible to measure. Here, we assess 2 indirect methods for estimating loss of saleable milk when housing cows and calves together in a robotic milking unit. In our study, treatment (CCC) cows and calves were kept together full time until the calves were 127 ± 6.6 d old (mean ± SD). Control cows were separated from their calves within 12 h of birth and then kept in the same unit as the treatment cows but with no access to either their own or treatment calves. Milk yield recording of both groups was performed from calving until pasture release at 233 ± 20 d in milk. The first estimation method relied on observed postseparation milk yield data, which were fed into a modified Wilmink regression model to determine the best-fitting lactation curve for the preseparation period. The second method was based on the cows' daily energy intake postseparation, calculated by measuring the daily feed intake and analyzing the energy content of the ration. The calculated energy intake was used to determine the average ratio between energy intake and the observed milk yield the following day for each individual cow, assuming constant rates of mobilization and deposition of body fat. The obtained ratio was then used to calculate the expected daily milk yield based on daily energy intake data during the preseparation period. In this paper, we analyzed data from 17 CCC cows kept together with their calves and 16 control cows; both groups calved from September to October 2020 and were followed up until release to pasture in May 2021. Saleable milk yield was lower in CCC cows than in control cows, both before and after separation. The 2 methods were used on data for control cows and showed milk yield loss using the lactation curve method (average of −3.4 ± 2.8 kg/d) and almost no loss using energy intake data (average of −1.4 ± 2.7 kg/d). Milk yield loss for CCC cows was estimated at average 11.3 ± 4.8 and 7.3 ± 6.6 kg milk/d, respectively. The proposed lactation curve estimation method tends to overestimate milk yield loss, whereas the method based on energy intake is more accurate. However, collecting detailed energy intake data per individual cow requires additional effort and equipment, which is not always feasible on commercial farms. Further research is needed to improve milk loss estimation and to better understand trade-offs in CCC systems.

    • Crossbreeding in dairy cattle has recently become of increased interest. However, farmers in Scandinavian countries are reluctant to implement crossbreeding in their herds, and one reason is the common opinion that only herds at a poor level of management can benefit from crossbreeding. The Danish Cattle Database (SEGES, Aarhus, Denmark) provided data on 14 traits regarding milk yield, udder health, fertility traits, stillbirth, and survival. The data were collected from 103,307 pure Holstein cows and 14,832 F-1 crosses (Holstein darn and Nordic Red sire). The cows were born between 2008 and 2014 and originated from 424 herds that contributed data from at least 5 purebreds and 5 crossbreds across the years. We split the animals into 3 production levels: high, average, and low according to the herd's average production (kg) of 305-d fat plus protein in the given birth year of the cow. We estimated least squares means of breed group (purebred and crossbred) performance within each production level. Crossbred performance in 305-d fat yield in first-parity cows was greater than that of Holstein across all herd production levels; the gain was greater in high- (9 kg more than Holstein) and average-producing herds (7 kg more than Holstein) than in low-producing herds (3 kg more than Holstein). Regardless of production level or parity, crossbreds did not outperform Holstein in terms of 305-d protein yield (0 to 8 kg less). Crossbreds had relatively better udder health than Holstein in both first and second parity (up to 15% less mastitis) within any of the production levels. In terms of fertility, stillbirth, and survival, crossbreds performed better than purebreds, and improved performance was independent of herd production level. We conclude that differences in performance between F-1 crossbreds and Holstein are independent of production level.

    • The development of breeding tools, such as genomic selection and sexed semen, has progressed rapidly in dairy cattle breeding during the past decades. In combination with beef semen, these tools are adopted increasingly at herd level. Dairy crossbreeding is emerging, but the economic and genetic consequences of combining it with the other breeding tools are relatively unknown. We investigated 5 different sexed semen schemes where 0, 50, and 90% of the heifers; 50% of the heifers + 25% of the first-parity cows; and 90% of the heifers + 45% of the first-parity cows were bred to sexed semen. The 5 schemes were combined in scenarios managing pure-breeding or terminal crossbreeding, including genomic testing of all newborn heifers or no testing, and keeping Swedish Red or Swedish Holstein as an initial breed. Thus, 40 scenarios were simulated, combining 2 stochastic simulation models: SimHerd Crossbred (operational returns) and ADAM (genetic returns). The sum of operational and genetic returns equaled the total economic return. Beef semen was used in all scenarios to limit the surplus of replacement heifers. Terminal crossbreeding implied having a nucleus of purebred females, where some were inseminated with semen of the opposite breed. The F-1 crossbred females were inseminated with beef semen. The reproductive performance played a role in improving the benefit of any of the tools. The most considerable total economic returns were achieved when all 4 breeding tools were combined. For Swedish Holstein, the highest total economic return compared with a pure-breeding scenario, without sexed semen and genomic test, was achieved when 90% sexed semen was used in heifers and 45% sexed semen was used for first-parity cows combined with genomic test and crossbreeding (+(sic)58, 33% crossbreds in the herd). The highest total economic return for Swedish Red compared with a pure-breeding scenario, without sexed semen and genomic test, was achieved when 90% sexed semen was used in heifers combined with genomic test and crossbreeding (+(sic)94, 46% crossbreds in the herd). Terminal crossbreeding resulted in lower genetic returns across the herd compared with the corresponding pure-breeding scenarios but was compensated by a higher operational return.

    • Prevention of biofilm formation in milking equipment is important to ensure good hygiene quality of raw milk. Key factors to achieving good results are a successful cleaning procedure and a method to check the cleanliness of milking equipment surfaces. Adenosine triphosphate bioluminescence is a fast and easy method for investigating bacterial contamination of surfaces. However, previous studies on the potential of ATP bioluminescence to assess the hygiene status of milking equipment have been hampered by lack of a validated test procedure. The aim of this work was therefore to establish a test procedure for assessing the cleanliness of milking equipment using ATP bioluminescence, and apply the method on-farm to study the hygiene status of aging rubber material in milking equipment. In developing the test procedure, the effects of sampling location in tubes and liners, sampling of dry versus wet barrels, milking point in the parlor, and acid or alkali detergent on ATP values were investigated. The results showed that, to obtain reproducible results, replicate sampling from the same milking points in the parlor is important. For milk tubes, samples should preferably be taken from the milk meter side, for liners on the inside of the barrel. For best results, sampling should be performed after use of alkali detergent. No beneficial effect was observed of sampling dry liner barrels, so sampling in the standardized test procedure is performed directly after cleaning. The standardized test procedure was used on 3 different commercial farms and sampling was initiated after replacement of old rubber parts. On one of the farms, additional sampling was performed to evaluate total bacteria count and determine the association with ATP level. The results suggest that, provided an efficient cleaning procedure is used, the hygiene quality of milking equipment can be maintained during the recommended lifetime of the rubberware. However, due to occasional variation in cleaning efficiency between milking points and liner barrels, random sampling on single occasions can lead to incorrect conclusions. Replicate sampling over time is therefore important for correct interpretation of ATP bioluminescence data. If ATP levels are very high, complementary sampling for total bacteria count should be used to verify that the level is due to bacterial contamination, and not other organic ATP-contributing material (e.g., milk residues).

    • Natural antibodies (NAb) are produced without any antigenic stimulation as a part of the innate immune system and provide a first line of defense against pathogens. Hence, they may be a useful trait when estimating an animal's potential immune competence and in selection for disease resistance. The aim of this study was to identify genomic regions associated with different NAb traits in milk and potentially describe candidate genes. Milk samples from 1,695 first-lactation Holstein Friesian cows with titer measurements for keyhole limpet hemocyanin, lipopolysaccharide, lipoteichoic acid, and peptidoglycan-binding total NAb and isotypes IgG(1), IgM, and IgA were used. Genome-wide association study analyses were performed using imputed 777K SNP genotypes, accounting for relationships using pedigree information. Functional enrichment analysis was performed on the significantly associated genomic regions to look for candidate genes. For IgM NAb, significant associations (false discovery rate <0.05) were found on Bos taurus autosonie (BTA) 17, 18, and 21 with candidate genies related to immunoglobulin structure and early B cell development. For IgG(1), associations were found on BTA3, and we confirmed a quantitative trait loci on BTA21 previously reported for IgG NAb in serum. Our results provide new insights into the regulation of milk NAb that will help unravel the complex relationship between milk immunoglobulins and disease resistance in dairy cattle.

    • The aim of this study was to evaluate the effect of sampling technique and milk fraction on bovine milk microbiota data and to compare the microbiota in milk to microbiota on the teat end and in the teat canal. Representative milk samples are highly important for assessment of bacteriological findings and microbiota in milk. Samples were obtained from 5 healthy lactating dairy cows at udder quarter level during 1 milking. Swab samples from the teat. end and teat canal, and milk samples collected using different techniques and in different milk fractions were included. Milk was collected by hand stripping and through a teat canal cannula before and after machine milking, through a trans-teat wall needle aspirate after milking, and from udder quarter composite milk. The microbiota of the samples was analyzed with sequencing of the V1-V3 region of the 16S rRNA gene. In addition, somatic cell counts and bacterial cultivability were analyzed in the milk samples. Microbiota data were analyzed using multivariate methods, and differences between samples were tested using analysis of similarity (ANOSIM). Differences between samples were further explored via individual studies of the 10 most abundant genera. The microbiota on the teat end, in the teat canal, and in udder quarter composite milk, collected using a milking machine, differed in composition from the microbiota in milk collected directly from the udder quarter. No differences in milk microbiota composition were detected between hand-stripped milk samples, milk samples taken through a teat canal cannula, or milk samples taken as a trans-teat wall needle aspirate before or after milking. We conclude that for aseptic milk samples collected directly from the lactating udder quarter, sampling technique or milk fraction has minor effect on the microbiota composition.

    • The objective of the study was to evaluate the effect of cashew nut shell extract (CNSE) and glycerol (purity >99%) on enteric methane (CH4) production and microbial communities in an automated gas in vitro system. Microbial communities from the in vitro system were compared with samples from the donor cows, in vivo. Inoculated rumen fluid was mixed with a diet with a 60:40 forage:concentrate ratio and, in total, 5 different treatments were set up: 5 mg of CNSE (CNSE-L), 10 mg of CNSE (CNSE-H), 15 mmol of glycerol/L (glycerol-L), and 30 mmol of glycerol/L (glycerol-H), and a control without feed additive. Gas samples were taken at 2, 4, 8, 24, 32, and 48 h of incubation, and the CH4 concentration was measured. Samples of rumen fluid were taken for volatile fatty acid analysis and for microbial sequence analyses after 8, 24, and 48 h of incubation. In vivo rumen samples from the cows were taken 2 h after the morning feeding at 3 consecutive days to compare the in vitro system with in vivo conditions. The gas data and data from microbial sequence analysis (454 sequencing) were analyzed using a mixed model and principal components analysis. These analyses illustrated that CH4 production was reduced with the CNSE treatment, by 8 and 18%, respectively, for the L and H concentration. Glycerol instead increased CH4 production by 8 and 12%, respectively, for the L and H concentration. The inhibition with CNSE could be due to the observed shift in bacterial population, possibly resulting in decreased production of hydrogen or formate, the methanogenic substrates. Alternatively the response could be explained by a shift in the methanogenic community. In the glycerol treatments, no main differences in bacterial or archaeal population were detected compared with the in vivo control. Thus, the increase in CH4 production may be explained by the increase in substrate in the in vitro system. The reduced CH4 production in vitro with CNSE suggests that CNSE can be a promising inhibitor of CH4 formation in the rumen of dairy cows.

    • Methane production from ruminant livestock varies with the diet as a result of factors such as dry matter intake, diet composition, and digestibility. To estimate the effect of dietary composition and feed additives, CH4 production can be measured in vitro as a first step because large numbers of samples can be incubated and analyzed at the same time. This study evaluated a recently developed in vitro method for prediction of in vivo CH4 production by examining the relationship between predicted and observed CH4 production values. A total of 49 different diets (observations), used in previous 13 in vivo studies, were selected to include diets varying in nutrient composition. Methane production was measured in all in vivo studies by respiration chambers or the GreenFeed system (C-Lock Inc., Rapid City, SD). Overall, the in vitro system predicted CH4 production well (R-2 = 0.96), but the values obtained were slightly underestimated compared with observed in vivo values (mean 399 L/d compared with 418 L/d: root mean square prediction error = 51.6 L/d or 12.3% of observed mean). Further analysis of the effect on residuals showed no significant relationship between CH4 production and most factors known to affect CH4 production such as dry matter intake, digestibility, and dietary concentrations of fat and starch. However, some factors included in the model were not well predicted by the system, with residuals negatively related to neutral detergent fiber concentration and positively related to concentrate proportion. The in vitro system can thus be useful for screening diets and evaluation of feed additives as a first step that can be best interpreted when feeding cows at maintenance level.

    • Genomic selection has been widely implemented in national and international genetic evaluation in the dairy cattle industry, because of its potential advantages over traditional selection methods and the availability of commercial high-density (HD) single nucleotide polymorphism (SNP) panels. However, this method may not be cost-effective for cow selection and for other livestock species, because the cost of HD SNP panels is still relatively high. One possible solution that can enable other species to benefit from this promising method is genomic selection with low-density (LD) SNP panels. In this simulation study, LD SNP panels designed with different strategies and different SNP densities were compared. The effects of number of quantitative trait loci, heritability, and effective population size were evaluated in the framework of genomic selection with LD SNP panels. Methodologies of Bayesian variable selection; BLUP with a trait-specific, marker-derived relationship matrix; and BLUP with a realized relationship matrix were employed to predict genomic estimated breeding values with both HD and LD SNP panels. Up to 95% of accuracy obtained by using an HD panel can be obtained by using only a small proportion of markers. The LD panel with markers selected on the basis of their effects always performs better than the LD panel with evenly spaced markers. Both the genetic architecture of the trait and the effective population size have a significant effect on the performance of the LD panels. We concluded that, to implement genomic selection with LD panels, a training population of sufficient size and genotyped with an HD panel is necessary. The trade-off between the LD panels with evenly spaced markers and selected markers must be considered, which depends on the number of target traits in a breeding program and the genetic architecture of these traits. Genomic selection with LD panels could be feasible and cost-effective, though before implementation, a further detailed genetic and economic analysis is recommended.

    • The aim of this study was to evaluate the effect of shortening or omitting the dry period of dairy cows on milk casein composition. For this study, we analyzed milk samples of 90 cows with a dry period of 0, 30, or 60 d and either a glucogenic or a lipogenic ration in early lactation. Milk was sampled at 6 and 2 wk prepartum and at 2, 6, and 12 wk postpartum. Milk was analyzed for casein (CN) composition by capillary zone electrophoresis, and isoforms of kappa-CN were measured by reversed phase-HPLC. Shortening the dry period from 60 to 30 d reduced the (alpha S1)-CN fraction by 3.8% and increased the alpha(S2)-CN fraction by 5.5%. In milk from cows with a 0-d dry period, the glycosylated is,-CN fraction in late lactation increased from 8 to 12% between 6 and 2 wk prepartum. After calving, the glycosylated kappa-CN fraction in milk was higher for cows with a 0-d dry period (6.7%) compared with cows with a 60-d dry period (5.2%). The glycosylated kappa-CN fraction at 2 wk postpartum was negatively correlated with milk yield, suggesting that glycosylation was related to reduced productivity of mammary epithelial cells. In early lactation, the beta-CN fraction was reduced in milk of cows with a 0-d dry period. A lowered beta-CN fraction was associated with high somatic cell count and greater parity, indicating that it was the result of proteolytic activity. In conclusion, casein composition changes that result from shortening the dry period from 60 to 30 d are not expected to affect processing characteristics of milk. Applying a 0-d dry period may affect processability of milk because of a higher glycosylated kappa-CN fraction, and possibly because of higher proteolytic activity compared with a 60-d dry period.

    • The aim of this study was to evaluate the influence of shortening the dry period of Swedish dairy cows on plasmin activity and casein composition in milk. Swedish Holstein and Swedish Red cows, 45 in total, were assigned to a dry period of either 4 or 8 wk. Milk samples were taken 10 and 5 wk prepartum, and 6 and 12 wk postpartum. Plasmin activity and plasminogen activity were measured with a spectrophotometric assay. Casein composition was measured by capillary zone electrophoresis. Prepartum plasminogen activity increased by 22% between 10 and 5 wk prepartum, whereas no change in plasmin activity was observed during the same period. Cows with a 4-wk dry period had 61% higher plasmin activity in postpartum milk than cows with an 8-wk dry period. Cows of third or greater parity tended to have a stronger increase in plasmin activity as a result of applying a short dry period than cows of second parity. Although the alpha(S1)- and beta-casein fractions declined with increasing plasmin activity, no dry period effects were found. Based on postpartum differences in plasmin activity, it was concluded that particularly multiparous cows require more than 4 wk between lactations for recovery of the mammary epithelium. Changes in casein composition as an effect of plasmin activity are not expected to have a great effect on processing quality of milk, although future work is needed to verify this.

    • A genomic region associated with milk fatty acid (FA) composition has been detected on Bos taurus autosome (BTA)17 based on 50,000 (50K) single nucleotide polymorphism (SNP) genotypes. The aim of our study was to fine-map BTA17 with imputed 777,000 (777K) SNP genotypes to identify candidate genes associated with milk FA composition. Phenotypes consisted of gas chromatography measurements of 14 FA based on winter and summer milk samples. Phenotypes and genotypes were available on 1,640 animals in winter milk, and on 1,581 animals in summer milk samples. Single-SNP analyses showed that several SNP in a region located between 29.0 and 34.0 Mbp were in strong association with C6:0, C8:0, and C10:0. This region was further characterized based on haplotypes. In summer milk samples, for example, these haplotypes explained almost 10% of the genetic variance in C6:0, 9% in C8:0, 3.5% in C10:0, 1.8% in C12:0, and 0.9% in C14:0. Two groups of haplotypes with distinct predicted effects could be defined, suggesting the presence of one causal variant. Predicted haplotype effects tended to increase from C6:0 to C14:0; however, the proportion of genetic variance explained by the haplotypes tended to decrease from C6:0 to C14:0. This is an indication that the quantitative trait locus (QTL) region is involved either in the elongation process or in early termination of de novo synthesized FA. Although many genes are present in this QTL region, most of these genes on BTA17 have not been characterized yet. The strongest association was found close to the progesterone receptor membrane component 2 (PGRMC2) gene, which has not yet been associated with milk FA composition. Therefore, no clear candidate gene associated with milk FA composition could be identified for this QTL.

    • The aim of this study was to fine-map a genomic region associated with milk fatty acids (FA) on Bos taurus autosome (BTA) 17. This genomic region has been discovered with 50,000 (50k) single nucleotide polymorphisms (SNP) imputed to 777,000 (777k) SNP. In this study, high-density genotypes were imputed to whole-genome sequences level to identify candidate gene(s) associated with milk FA composition on BTA17. Phenotypes and genotypes were available for 1,640 cows sampled in winter, and for 1,581 cows sampled in summer. Phenotypes consisted of gas chromatography measurements in winter and in summer milk samples of 6 individual FA and the indicator of de novo synthesis, C6:0-C14:0. Genotypes consisted of imputed 777k SNP, and 89 sequenced ancestors of the population of genotyped cows. In addition, 450 whole-genome sequences from the 1,000 Bull Genome Consortium were available. Using 495 Holstein-Friesian sequences as a reference population, the 777k SNP genotypes of the cows were imputed to sequence level. We then applied single-variant analyses with an animal model, and identified thousands of significant associations with C6:0, C8:0, C10:0, C12:0, C14:0, and C6:0 C14:0. For C8:0 in summer milk samples, the genomic region located between 29 and 34 Mbp on BTA17 revealed a total of 646 significant associations. The most significant associations [ -log(10)(P-value) = 7.82] were 8 SNP in perfect linkage disequilibrium. After fitting one of these 8 SNP as a fixed effect in the model, and re-running the single variant analyses, no further significant associations were found for any of the 6 FA or C6:0 C14:0. These findings suggest that one polymorphism underlying this QTL on BTA17 influences multiple de novo synthesized milk FA. Thirteen genes in the QTL region were identified and analyzed carefully. Six out of the 8 SNP that showed the strongest associations were located in the La ribonucleoprotein domain family, member 1B (LARP1B) gene, and we suggest LARP1B as a primary candidate gene. Another gene of interest for this QTL region might be PKE4. None of these suggested candidate genes have previously been associated with milk fat synthesis or milk FA composition.

    • The rennet-induced coagulation ability of milk is important in cheese production. For Swedish Red Dairy Cattle (RDC), this ability is reduced because of a high prevalence of noncoagulating (NC) milk. In this study, we simultaneously combined genetic parameters for NC milk, milk coagulation properties, milk composition, physical traits, and milk protein composition. Our aim was to estimate heritability and genetic and phenotypic correlations for NC milk and 24 traits (milk coagulation properties, milk composition, physical traits, and milk protein composition). Phenotypes and ~7,000 SNP genotypes were available for all 600 Swedish RDC. The genotypes were imputed from ~7,000 SNP to 50,000 SNP. Variance components and genetic parameters were estimated with an animal model. In Swedish RDC, a moderate heritability estimate of 0.28 was found for NC milk. For the other 24 traits, heritability estimates ranged from 0.12 to 0.77 (standard errors from 0.08 to 0.18). A total of 300 phenotypic and genetic correlations were estimated. For phenotypic and genetic correlations, 172 and 95 were significant, respectively. In general, most traits showing significant genetic correlations also showed significant phenotypic correlations. In this study, phenotypic and genetic correlations with NC milk suggest that many correlations between traits exist, making it difficult to predict the real consequences on the composition of milk, if selective breeding is applied on NC milk. We speculate that some of these consequences may lead to changes in the composition of milk, most likely affecting its physical and organoleptic properties. However, our results suggest that κ-casein could be used as an indicator trait to predict the occurrence of NC milk at the herd level.

    • Phosphorylation of caseins (CN) is a crucial post-translational modification that allows caseins to form colloid particles known as casein micelles. Both alpha(S1)-and alpha(S2)-CN show varying degrees of phosphorylation (isoforms) in cow milk and were suggested to be more relevant for stabilizing internal micellar structure than beta- and kappa-CN. However, little is known about the genetic background of individual alpha(S2)-CN phosphorylation isoforms and the phosphorylation degrees of alpha(S1)- and alpha(S2)-CN (alpha(S1)-CN PD and alpha(S2)-CN PD), defined as the proportion of isoforms with higher degrees of phosphorylation in total alpha(S1)- and alpha(S2)-CN, respectively. We aimed to identify genomic regions associated with these traits using 50K single nucleotide polymorphisms for 1,857 Dutch Holstein Friesian cows. A total of 10 quantitative trait loci (QTL) regions were identified for all studied traits on 10 Bos taurus autosomes (BTA1, 2, 6, 9, 11, 14, 15, 18, 24, and 28). Regions associated with multiple traits were found on BTA1, 6, 11, and 14. We showed 2 QTL regions on BTA1, one affecting alpha(S2)-CN production and the other harboring the SLC37A I gene, which encodes a phosphorus antiporter and affects alpha(S1)-and alpha(S2)-CN PD. The QTL on BTA6 harbors the casein gene cluster and affects individual alpha(S1)-CN phosphorylation isoforms. The QTL on BTA11 harbors the PAEP gene that encodes for beta-lactoglobulin and affects relative concentrations of alpha(S2)-CN-10P and alpha(S2)-CN-11P as well as alpha(S1)-CN PD and alpha(S1)-CN PD. The QTL on BTA14 harbors the DGAT1 gene and affects relative concentrations of alpha(S1)-CN-10P and alpha(S2)-CN-11P as well as alpha(S1)-CN PD and alpha(S2)-CN PD. Our results suggest that effects of identified genomic regions on phosphorylation of alpha(S1)-CN and alpha(S2)-CN are related to changes in milk synthesis and phosphorus secretion in milk. The actual roles of SLC37A1, PAEP, and DGAT1 in alpha(S1)- and alpha(S2)- phosphorylation in Dutch Holstein Friesian require further investigation.

  • Risk factors for antimicrobial resistance in fecal Escherichia coli from preweaned dairy calves

    • The primary objective of this study was to investigate calf and farm factors associated with antimicrobial-resistant Escherichia coli in the feces of preweaned dairy calves in Sweden. In particular, we investigated the effects of feeding calves colostrum and milk from cows treated with antimicrobials. The secondary objective was to describe the prevalence of resistant E. coli in feces of preweaned dairy calves in Sweden. Fecal samples from 3 calves, aged 7 to 28 d, from 243 farms were analyzed for the within-sample prevalence of E. coli resistant to nalidixic acid, streptomycin, and cefotaxime using selective agars supplemented with antimicrobials. In addition, resistance to 12 antimicrobials was tested in one randomly selected E. coli isolate per calf. Information was collected from the farmers via questionnaires regarding the use of colostrum and milk from cows treated with antimicrobials as calf feed and other uses of antimicrobials in the herd. Multivariable zero-inflated negative binomial and logistic regression models were used to assess the effect of various risk factors for shedding of resistant E. coli. Escherichia coli resistant to streptomycin, nalidixic acid, or cefotaxime were isolated from 90, 49, and 11% of the calves, respectively. Resistance to at least one antimicrobial was found in a random isolate of E. coli from 48% of the calves. Feeding colostrum from cows treated with antimicrobials at drying off did not affect the prevalence of resistant E. coli. In contrast, feeding milk from cows treated with antimicrobials during lactation resulted in significantly more nalidixic acid- and streptomycin-resistant E. coli than when such milk was discarded; no significant effect was seen for other resistance traits. Furthermore, an interaction was found between feeding milk from cows treated with antimicrobials and use of fluoroquinolones in cows. In general, the prevalence of resistance was lower for older calves and calves on small farms. Other factors that were associated with the shedding of resistant E. coli were administration of oral dihydrostreptomycin to calves, administration of systemic tetracycline and ceftiofur to cows and calves, housing of the calves, predominant breed of the herd, and geographic location of the farm. The presence of resistant E. coli in calves was clearly due to multiple factors, but minimizing the feeding of milk from cows treated with antimicrobials during lactation should lower the prevalence of resistant E. coli in the gastrointestinal tract of the calves.

    • Quinolone resistance may emerge in gut bacteria (e.g., in Escherichia coli) of animals. Such bacteria could cause infections in the animal itself or be transmitted to humans via the food chain. Quinolone resistance is also observed in fecal E. coli of healthy dairy cattle, but the prevalence varies between farms, not solely as a result of varying degree of fluoroquinolone exposure. The objective of this study was to identify risk factors for the fecal shedding of quinolone-resistant E. coli (QREC) from dairy calves and postpartum cows. Rectal swabs from 15 preweaned calves and 5 postpartum cows per farm were collected on 23 Swedish dairy farms to determine the prevalence of QREC. Risk factors for the shedding of QREC were investigated using multivariable statistical models. Quinolone-resistant E. coli were found on all but one farm. Factors associated with QREC shedding by calves were being younger than 18 d, being fed milk from cows treated with antimicrobials, recent use of fluoroquinolones in the herd, carriage of QREC by postpartum cows, and using the calving area never or rarely as a sick pen compared with often. Factors associated with QREC shedding by cows were calving in group pens or freestalls compared with single pens or tiestalls, purchasing cattle, sharing animal transports with other farmers, and poor farm hygiene. Proper biosecurity and improved hygiene, as well as minimizing fluoroquinolone exposure and waste milk feeding, may be important factors to reduce the burden of QREC on dairy farms.

    • When the voluntary waiting period (VWP), defined as the days between calving and when the cow is eligible to receive the first insemination, is extended, high-yielding dairy cows may have better opportunities to regain energy balance before first insemination. This study investigated the effect of an extended (145–215 days in milk [DIM], n = 280) or conventional (25–95 DIM, n = 251) VWP treatment on fertility, disease incidence, and culling rate in cows during their first lactation. The cows were also followed through a second lactation without intervention regarding VWP, during which the farmers could decide when they wished to start the inseminations. This was done in a randomized-controlled study on 16 high-yielding commercial herds in southern Sweden, containing a total of 531 primiparous cows of the Holstein and Red Dairy Cattle breeds. Data from the Swedish national dairy herd recording scheme collected between August 2018 and September 2021 were used in the analysis, including records on breed, calvings, estrus intensity, inseminations, disease, somatic cell count, culling date, and culling reason. During first lactation, more cows receiving the extended VWP treatment showed strong estrus intensity (score 4–5, 55% vs. 48%) and fewer showed moderate estrus intensity (score 3, 35% vs. 43%) at first insemination, compared with cows receiving the conventional VWP treatment. First service conception rate (FSCR) was higher (67% vs. 51%) and number of inseminations per conception (NINS) was lower (1.6 vs. 2.0) during the first lactation for cows receiving the extended compared with the conventional VWP treatment. For disease incidence rate or culling rate expressed as number of events per cow-time in the study, we found no differences between the cows receiving the 2 VWP treatments in any lactation. Calving to first service interval during second lactation was longer (86 vs. 74 d) for cows with extended compared with conventional VWP. In conclusion, primiparous cows with extended VWP showed improved reproductive functions, in the form of higher estrus intensity, greater FSCR, and lower NINS, during the first lactation. However, we observed no apparent effect on these fertility measures during the following lactation (without VWP intervention) and no differences in disease prevalence or culling between cows receiving the 2 different VWP treatments in either lactation. Compliance with the planned VWP treatment was lower for cows with planned extended compared with planned conventional VWP treatment. We studied the “intention-to-treat” effect (i.e., the results for all cows randomized to each treatment regardless of whether the planned VWP was achieved or not) to identify any bias arising due to degree of compliance. However, we found no difference in culling rate between cows randomized to an extended VWP compared with those randomized to a conventional VWP. These findings can be used to support management decisions on VWP length in high-yielding dairy herds.
