    • The morphology and physiology of pheromone receptors on the antennae of male pine sawflies, Diprion pini L., were investigated. Using scanning electron microscopy, five sensillar types were recognized. The type shown to be pheromone sensitive has a long (50-70 μm) cuticular hair, is single-walled, and is innervated by 8 or 9 sensory cells as revealed by transmission electron microscopy. Electroantennography (EAG) showed similar activity of the acetate and propionate of (2S,3R,7R)-3,7-dimethyl-2-tridecanol, precursor of the main constituent of the female-produced sex pheromone. No other isomer induced any significant response. Single-sensillum recordings confirmed the results of the EAG, and also showed that several neurons were excited by the active compound. EAG recordings and combined gas chromatographic-electroantennographic detection indicated that esters of three 3,7-dimethyl-2-pentadecanol (diprionol) isomers were active, but field tests could not demonstrate any behavioural effect. Diprionol esters are used as sex pheromones by all other pine sawflies investigated so far, and D. pini is thus the first diprionid species shown to use a different sex pheromone.

    • The pine sawfly pheromone precursor 3,7-dimethyl-2-pentadecanol (diprionol) was quantified by gas chromatography in different body parts of virgin female Neodiprion sertifer. About one-third of the total amount (approx. 10 ng per female) was found in each of head + thorax, abdominal segments 1-3, and the remaining abdomen. Diprionol was also found in the respective parts of pupae, but in lower amounts. This suggests that at least the final steps of the pheromone biosynthesis take place in many parts of the body, possibly in the cuticle. The diprionol content remained relatively constant over the whole lifetime of virgin females. By using coupled gas chromatography-electroantennogram detection three active compounds in the acetylated female extract were observed. These corresponded to the pheromone, (2S,3S,7S)-diprionyl acetate, and its homologues with the chain shortened by one and two carbons respectively, as determined by gas chromatography-mass spectrometry. In addition, the homologue having a chain elongated by one carbon was found. The homologues occurred in amounts of 2-5% of the diprionyl acetate. When synthetic homologues were tested in the field, alone or in combination with diprionyl acetate, they were not behaviourally active to male sawflies, despite their electroantennogram activity. A threo-isomer of diprionol, most likely (2S,3R,7R), was present in the females in amounts less than 0.5% of the (2S,3S,7S)-content. The acetate of this threo-isomer is known as both synergist and inhibitor to the (2S,3S,7S)-isomer. trans-Perillenal was found in the abdominal segments 1-3 of both males and females and its possible biosynthetic relationship to diprionol is discussed.

  • Peripheral modulation of olfaction by physiological state in the Egyptian leaf worm Spodoptera littoralis (Lepidoptera: Noctuidae)

    • Insects show behavioural plasticity based on their physiological state. Deprivation from a resource will normally make them more responsive to it or to perform behaviour increasing the probability of encountering such a resource. Modulation of the olfactory system has been shown mainly in the central nervous system, but also in the periphery. In this study, antennal sensitivity of females of the Egyptian cotton leaf worm Spodoptera littoralis to different plant and sex pheromone odours was measured using electroantennography (EAG). Different mating status, age, and feeding status were used to investigate peripheral changes in olfactory responses. Virgin females were found to be more sensitive to plant odours and sex pheromone compared to mated females. Age also had an impact on antennal sensitivity, 2 and 4 days old females being more sensitive than just-emerged ones. Such an increase in sensitivity could be explained by maturation of olfactory receptor neurons after emergence or by an increase in motivation as the available expected life time remaining decreases. Finally, feeding status did not modify the antennal sensitivity of females. A peripheral modulation could thus be demonstrated in the olfactory system of female Spodoptera littoralis at certain life stages. (C) 2009 Elsevier Ltd. All rights reserved.

    • Seed-eating Apion weevils (Coleoptera: Apionidae) cause large economic losses in white and red clover seed production across Europe. Monitoring and control of clover weevils would be facilitated by semiochemical-based methods. Until now, however, nothing was known about physiological or behavioral responses to semiochemicals in this insect group. Here we analyzed the antenna of the white clover (Trifolium repens L.) specialist Apion fulvipes Geoffroy with scanning electron microscopy, and used single sensillum recordings with a set of 28 host compounds to characterize 18 classes of olfactory sensory neurons (OSNs). Nine of the OSN classes responded strongly to synthetic compounds with high abundance in clover leaves, flowers, or buds. Eight classes responded only weakly to the synthetic stimuli, whereas one collective class responded exclusively to volatiles released from a crushed clover leaf. The OSNs showed a remarkable degree of specificity, responding to only one or a few chemically related compounds. In addition, we recorded a marked difference in the temporal dynamics of responses between different neurons, compounds, and doses. The identified physiologically active compounds will be screened for behavioral activity, with the ultimate goal to develop an odor-based control strategy for this pest.

  • Specificity and redundancy in the olfactory system of the bark beetle Ips typographus: Single-cell responses to ecologically relevant odors

    • We screened 150 olfactory sensilla in single-sensillum recordings to unravel the mechanisms underlying host selection in the spruce bark beetle, Ips typographus (Coleoptera: Curculionidae: Scolytinae). Odor stimuli comprised of pheromone (various bark beetle spp.). host, and non-host compounds elicited strong and selective responses from 106 olfactory receptor neurons (ORNs), 45 of which were tuned to pheromone compounds, 37 to host compounds, and 24 to non-host volatiles (NHV). In addition, 26 ORNs responded only weakly to any odor stimulus. Strongly responding ORNs were classified into 17 classes. Seven classes responded primarily to the Ips pheromone compounds: cis-verbenol, ipsenol, ipsdienol (two classes), 2-methyl-3-buten-2-ol, amitinol, or verbenone, respectively. Six classes responded to the host compounds: alpha-pinene, myrcene, p-cymene, myrcene and p-cymene, 1,8-cineole, or Delta 3-carene, respectively. Four classes responded to NHV: 3-octanol, 1-octen-3-ol, trans-conophthorin, or indiscriminately to the repellent green leaf volatiles (GLVs) 1-hexanol, Z3-hexen-1-ol and E2-hexen-1-ol, respectively. Indiscriminate responses from GLV neurons might explain a behavioral redundancy among these GLVs. This is the first description of individual bark beetle ORNs dedicated to NHV perception. These comprise almost 25% of the strongly responding neurons, demonstrating that a large proportion of the olfactory system is devoted to signals from plants that the insect avoids.   • Collective choice in ants : The role of protein and carbohydrates ratios

    • In a foraging context, social insects make collective decisions from individuals responding to local information. When faced with foods varying in quality, ants are known to be able to select the best food source using pheromone trails. Until now, studies investigating collective decisions have focused on single nutrients, mostly carbohydrates. In the environment, the foods available are a complex mixture and are composed of various nutrients, available in different forms. In this paper, we explore the effect of protein to carbohydrate ratio on ants' ability to detect and choose between foods with different protein characteristics (free amino acids or whole proteins). In a two-choice set up, Argentine ants Linepithema humile were presented with two artificial foods containing either whole protein or amino acids in two different dietary conditions: high protein food or high carbohydrate food. At the collective level, when ants were faced with high carbohydrate foods, they did not show a preference between free amino acids or whole proteins, while a preference for free amino acids emerged when choosing between high protein foods. At the individual level, the probability of feeding was higher for high carbohydrates food and for foods containing free amino acids. Two mathematical models were developed to evaluate the importance of feeding probability in collective food selection. A first model in which a forager deposits pheromone only after feeding, and a second model in which a forager always deposits pheromone, but with greater intensity after feeding. Both models were able to predict free amino acid selection, however the second one was better able to reproduce the experimental results suggesting that modulating trail strength according to feeding probability is likely the mechanism explaining amino acid preference at a collective level in Argentine ants.

    • This study describes the morphology and function of the antennal sensilla in two gall midge species, Contarinia nasturtii and Mayetiola destructor, where multi-component sex pheromones have been identified. Both species possess sensilla trichodea, s. coeloconica, s. chaetica and s. circumfila. Sensilla circumfila, which consist of several sensilla that bifurcate and fuse into one structure, are unique for the gall midges. In C. nasturtii s. circumfila are sexually dimorphic. In males, they form elongated loops suspended on cuticular spines, whereas in females they run like worm-like structures directly on the antennal surface. Single sensillum recordings demonstrated that olfactory sensory neurons housed in male s. circumfila in C. nasturtii responded to the female sex pheromone. In M. destructor, S. circumfila were attached to the antennal surface in both sexes, and displayed no response to sex pheromone components.A sexual dimorphism was also found in the number of s. trichodea per antennal segment in both C. nasturtii (male 1 vs. female 7) and M. destructor (male 13 vs. female 10). OSNs located in male M. destructor s. trichodea responded to the sex pheromone. This is the first gall midge single sensillum study, and the first demonstration of the functional significance of s. circumfila. (C) 2010 Elsevier Ltd. All rights reserved.

    • Specific mate communication and recognition underlies reproduction and hence speciation. Our study provides new insights in Drosophila melanogaster premating olfactory communication. Mate communication evolves during adaptation to ecological niches and makes use of social signals and habitat cues. Female-produced, species-specific volatile pheromone (Z)-4-undecenal (Z4-11Al) and male pheromone (Z)-11-octadecenyl acetate (cVA) interact with food odour in a sex-specific manner. Furthermore, Z4-11Al, which mediates upwind flight attraction in both sexes, also elicits courtship in experienced males. Two isoforms of the olfactory receptor Or69a are co-expressed in the same olfactory sensory neurons. Z4-11Al is perceived via Or69aB, while the food odorant (R)-linalool is a main ligand for the other variant, Or69aA. However, only Z4-11Al mediates courtship in experienced males, not (R)-linalool. Behavioural discrimination is reflected by calcium imaging of the antennal lobe, showing distinct glomerular activation patterns by these two compounds. Male sex pheromone cVA is known to affect male and female courtship at close range, but does not elicit upwind flight attraction as a single compound, in contrast to Z4-11Al. A blend of the food odour vinegar and cVA attracted females, while a blend of vinegar and female pheromone Z4-11Al attracted males, instead. Sex-specific upwind flight attraction to blends of food volatiles and male and female pheromone, respectively, adds a new element to Drosophila olfactory premating communication and is an unambiguous paradigm for identifying the behaviourally active components, towards a more complete concept of food-pheromone odour objects.

    • Protapion trifolii and P. fulvipes (Coleoptera: Brentidae) are major pests in European clover seed production. Previous studies have reported a high host plant fidelity of these weevils for red and white clover species, respectively, driven by host plant olfactory cues. Given the specific host preferences observed in these weevils, we aimed to elucidate to which extent such selectivity is reflected in their peripheral olfactory systems. Using an electrophysiological approach, we performed the first functional characterisation of olfactory sensory neurons (OSNs) in P. trifolii to a panel of volatile compounds emitted by red clover plants, and compared the results with the reported OSN types of P. fulvipes. Nineteen OSN classes were characterized in P. trifolii, with the majority of these neurons responding strongly to common volatiles released by the host plant. Based on response profiles, eight of these OSN classes have clear matches to OSN classes in P. fulvipes. The OSN colocalisation patterns and antennal frequency of these classes are similar in the two species. Additionally, the responses of these OSNs are generally highly conserved in the two species, with clear response shifts only revealed for two of the OSN classes. These response shifts in combination with additional response dissimilarities for compounds that vary in abundance between red and white clover plants may underlie the species-specific host preferences. Further behavioural and field experiments should focus on these differentially detected compounds to elucidate their potential role in host selection and use in semiochemical-based control of these pests.

    • A virgin ant queen has only one opportunity in her lifetime to realize her reproductive fitness when she leaves her nest for a mating flight. After successful mating she sheds her wings, excavates a nest and starts laying eggs to initiate her own colony. Here we report the results of our study on two related species of Camponotus ants - day active Camponotus paria and night active Camponotus compressus - aimed at investigating (i) if there exist inter-species differences in the activity and phototactic behaviors of males and queens, (ii) whether these behaviors in the queen change after mating, and (iii) whether the activity rhythm of queens changes with age. We find that while activity profiles differ between C. paria and C. compressus virgin males and queens, such differences in queens disappear after mating. Once mated, the activity rhythm of queens shows little change with age; the rhythm in virgin queens, on the other hand, changes considerably. As virgins, C. paria queens are positively phototactic, while C. compressus queens are negatively phototactic. After mating, C. paria queens become less phototactic, particularly during the subjective night, while C. compressus queens remain negatively phototactic. These results indicate that there are considerable differences in the activity and phototactic behaviors of virgin queens of the two related species of Camponotus ants. Most of these differences disappear after mating, which suggests that these behaviors may have evolved primarily for the proper execution of pre-mating events.

    • Cyclotides are defense peptides produced by several plant families. Viola spp. (Violaceae) produce an array of cyclotides with varying biological activities. The peach potato aphid Myzus persicae (Sulz.) (Hemiptera: Aphididae) is a generalist that feeds on the secondary hosts of over 40 plant families, including Violaceae. The present work aimed to evaluate the activities of cycloviolacins from Viola odorata L. and V. ulignosa Besser (cyO2, cyO3, cyO13, cyO19) against M. persicae. To investigate the peptides' influence on aphid feeding behavior, we used 20% sucrose diets supplemented with cyclotides and measured the effects with electrical penetration graph (EPG) technique. We also applied anti-cyclotide antibodies and immunohistochemistry to track the peptides in the digestive systems of the aphids. Our study shows that cyclotides affect aphid probing and feeding behavior and limit their diet sap uptake. The cycloviolacin cyclotides: cyO13 (100 mu M) and cyO19 (50 mu M) most strongly impeded aphid ingestion activities when applied in sucrose diet. Sustained ingestion of the diet was blocked by 100 mu M cyO13, and no aphid showed ingestion of the diet for longer than 10 min. Cyclotides were detected in the pharynx, in contact with the epipharyngeal gustatory organ, in the stomach (midgut) and upper intestine. The present study shows the deterrent activity of cycloviolacins on M. persicae. This activity may be related to the peptides' effects on epithelial cells and gustatory organs along the aphid digestive system. We demonstrate that cyclotides may play an important role in plant-aphid interactions.

  • Spectral sensitivity of a colour changing spider

    • Vision plays a paramount role in some spider families such as the Salticidae, Lycosidae and Thomisidae, as it is involved in prey hunting, orientation or choice of substrate. In the thomisid Misumena vatia, for which the substrate colour affects the body colour, vision seems to mediate morphological colour changes. However, nothing is known about which component of visual signals from the substrate might be perceived, nor whether M. vatia possesses the physiological basis for colour vision. The aim of this study is thus to investigate the vision of this spider species by measuring the spectral sensitivities of the different pairs of eyes using electrophysiological methods. Extra- and intracellular electrophysiological recordings combined with selective adaptation revealed the presence of two classes of photoreceptor cells, one sensitive in the UV region of the spectrum (around 340 nm) and one sensitive in the green (around 520 nm) regions in the four pairs of eyes. We conclude that M. vatia possesses the physiological potential to perceive both chromatic and achromatic components of the environment. (C) 2011 Elsevier Ltd. All rights reserved.

    • In nature, ovipositing females may be subjected to multiple extrinsic and intrinsic environmental factors simultaneously. To adequately assess a species response to environmental conditions during oviposition it may therefore be necessary to consider the interaction between multiple intrinsic and extrinsic factors simultaneously. Using the butterfly, Pararge aegeria, this study examined the combined effects of extrinsic (temperature and flight) and intrinsic (body mass and age) factors on ovarian dynamics, egg provisioning and reproductive output, and explored how these effects subsequently influenced offspring fitness when egg-stage development occurred in a low humidity environment. Both temperature- and flight- mediated plasticity in female reproductive output was observed, and there were strong temperature by flight interaction effects for the traits oocyte size and egg mass. As females aged, mean daily fecundity differed across temperature treatments, but not across flight treatments. Overall, temperature had more pronounced effects on ovarian dynamics than flight. Flight mainly influenced egg mass via changes in relative water content. A mismatch between the physiological response of females to high temperature and the requirements of their offspring had a negative impact on offspring fitness via effects on egg hatching success.

  • Visual cues of oviposition sites and spectral sensitivity of Cydia strobilella L.

    • We investigated whether the spruce seed moth (Cydia strobilella L., Tortricidae: Grapholitini), an important pest in seed orchards of Norway spruce (Picea abies (L.) Karst.), can make use of the spectral properties of its host when searching for flowers to oviposit on. Spectral measurements showed that the flowers, and the cones they develop into, differ from a background of P. abies needles by a higher reflectance of long wavelengths. These differences increase as the flowers develop into mature cones. Electroretinograms (ERGs) in combination with spectral adaptation suggest that C. strobilella has at least three spectral types of photoreceptor; an abundant green-sensitive receptor with maximal sensitivity at wavelength λmax = 526 nm, a blue-sensitive receptor with λmax = 436 nm, and an ultraviolet-sensitive receptor with λmax = 352 nm. Based on our spectral measurements and the receptor properties inferred from the ERGs, we calculated that open flowers, which are suitable oviposition sites, provide detectable achromatic, but almost no chromatic contrasts to the background of needles. In field trials using traps of different spectral properties with or without a female sex pheromone lure, only pheromone-baited traps caught moths. Catches in baited traps were not correlated with the visual contrast of the traps against the background. Thus, visual contrast is probably not the primary cue for finding open host flowers, but it could potentially complement olfaction as a secondary cue, since traps with certain spectral properties caught significantly more moths than others.

  • Predator species related adaptive changes in larval growth and digestive physiology

    • Prey species are often non-randomly distributed along predator gradients but according to how they trade off growth against predation risk. The foraging-mediated growth/predation risk trade-off is well established, with increased foraging accelerating growth but also increasing predator induced mortality. While adaptations in digestive physiology may partly modify the relationship between foraging and growth in response to predation risk, studies exploring the impact of digestive physiology on growth in prey subjected to predation risk are still scarce. Larvae of the dragonfly genus Leucorrhinia segregate at the species level between lakes either being dominated by predatory fish (fish-lakes) or predatory invertebrates (dragonfly-lakes). Predators of these two lake types differ dramatically in their hunting style like searching and pursuing mode causing different selection pressure on prey traits including foraging. In a laboratory experiment we estimated growth rate, digestive physiology (ingested food, growth efficiency, assimilation efficiency, conversion efficiency) and metabolic rate (oxygen consumption) in the presence and absence of predator cues. Whereas fish-lake and dragonfly-lake Leucorrhinia species did not differ in growth rate, they evolved different pathways of digestive physiology to achieve similar growth rate. Because fish-lake species expressed a higher metabolic rate than dragonfly-lake species, we assume energy to be differently allocated and used for metabolic demands between species of both predator environments. Further, growth rate, but not digestive physiology was plastic in response to the presence of predator cues. Our results highlight the impact of digestive physiology in shaping the foraging-mediated growth/predation risk trade-off, with digestive physiology contributing to species distribution patterns along predator gradients.

We postulated that these derived mouthpart structures, positioned at the base of the proboscis, may be particularly important in mediating feeding behaviors. As feeding is spatio-temporally segregated from oviposition in most Tephritidae, this taxonomic group appears quite suitable to parse out sensory breadth and potential functional divergence of palps and antennae. Scanning electron microscopy and anterograde staining underlined the limited palpal olfactory circuit in Tephritidae: only three morphological subtypes of basiconic sensilla were found, each with two neurons, and project to a total of six antennal lobe glomeruli in Bactrocera dorsalis. Accordingly, the palps detected only few volatiles from the headspace of food (fermentation and protein lures) and fruit (guava and mango) compared to the antennae (17 over 77, using gas-chromatography coupled electrophysiology). Interestingly, functionally the antennae were more tuned to fruit volatiles, detecting eight times more fruit than food volatiles (63 over 8), whereas the number of fruit and food volatile detection was more comparable in the palps (14 over 8). As tephritids diverge in oviposition preferences, but converge on food substrates, we postulated that the receptive ranges of palpal circuits would be more conserved compared to the antennae. However, palpal responses of three tephritid species that differed in phylogenetic relatedness and ecologically niche, diverged across ecological rather than phylogenetic rifts. Two species with strongly overlapping ecology, B. dorsalis and Ceratitis capitata, showed inseparable response profiles, whereas the cucurbit specialist Zeugodacus cucurbitae strongly diverged. As Z. cucurbitae is phylogenetically placed between B. dorsalis and C. capitata, the results indicate that ecology overrides phylogeny in the evolution of palpal tuning, in spite of being predisposed to detecting food volatiles.

    • How respiratory structures vary with, or are constrained by, an animal’s environment is of central importance to diverse evolutionary and comparative physiology hypotheses. To date, quantifying insect respiratory structures and their variation has remained challenging due to their microscopic size, hence only a handful of species have been examined. Several methods for imaging insect respiratory systems are available, in many cases however, the analytical process is lethal, destructive, time consuming and labour intensive. Here, we explore and test a different approach to measuring tracheal volume using X-ray micro-tomography (μCT) scanning (at 15 μm resolution) on living, sedated larvae of the cerambycid beetle Cacosceles newmannii across a range of body sizes at two points in development. We provide novel data on resistance of the larvae to the radiation dose absorbed during μCT scanning, repeatability of imaging analyses both within and between time-points and, structural tracheal trait differences provided by different image segmentation methods. By comparing how tracheal dimension (reflecting metabolic supply) and basal metabolic rate (reflecting metabolic demand) increase with mass, we show that tracheal oxygen supply capacity increases during development at a comparable, or even higher rate than metabolic demand. Given that abundant gas delivery capacity in the insect respiratory system may be costly (due to e.g. oxygen toxicity or space restrictions), there are probably balancing factors requiring such a capacity that are not linked to direct tissue oxygen demand and that have not been thoroughly elucidated to date, including CO2 efflux. Our study provides methodological insights and novel biological data on key issues in rapidly quantifying insect respiratory anatomy on live insects.

    • Social insects such as termites live in colonies in which cooperation is assumed by all individuals developing into castes to which specific tasks are allocated. Little has been reported about molecular aspects underlying termite caste-specific gene expression. Genetic regulation has recently been hypothesized to govern caste-specific traits and physiology in social insects. Cytochrome c oxidase (COX) has been shown to be an interesting candidate for expression study in insects. We used the cytochrome c oxidase subunit III gene (COXIII) that was cloned from mRNA in a lower termite, Reticulitermes santonensis De Feytaud (Isoptera; Rhinotermitidae). The full-length cDNA encodes a protein of 262 amino acids that shows high degree of homology with other insects COXIIIs. Reverse transcriptase-PCR and real-time PCR were performed to compare gene expression between larvae, workers, nymphs and soldiers. Analyses performed on head cDNAs revealed that COXIII is differentially expressed between castes. The level of COXIII is caste-regulated with an increase in workers (similar to 1.9-fold) and nymphs (similar to 2.8-fold) and a decrease in soldiers (0.8-fold) compared to the expression level in larvae (1.0-fold). These results may emphasize the physiological importance of COX in the termite brain at different developmental stages.

    • Hemolymph coagulation is vital for larval hemostasis and important in immunity, yet the molecular basis of coagulation is not well understood in insects. Of the larval clotting factors identified in Drosophila, Fondue is not conserved in other insects, but is notable for its effects on the clot's physical properties, a possible function in the cuticle, and for being a substrate of transglutaminase. Transglutaminase is the only mammalian clotting factor found in Drosophila, and as it acts in coagulation in other invertebrates, it is also likely to be important in clotting in Drosophila. Here we describe a Fondue-GFP fusion construct that labels the cuticle and clot, and show that chemical inhibition and RNAi knockdown of the Drosophila transglutaminase gene affect clot properties and composition in ways similar to knockdown of the fon gene. Thus, Fondue appears to be incorporated into the cuticle and is a key transglutaminase substrate in the clot. This is also the first direct functional confirmation that transglutaminase acts in coagulation in Drosophila.

    • Antennal olfactory receptor neurons (ORNs) for pheromone and plant volatile compounds were identified and characterized in male and female clover root weevil, Sitona lepidus (Gyllenhal), using the single sensillum recording technique with five pheromone-related compounds, and 40 host and non-host plant volatile compounds. Overall, seven different types of olfactory sensilla containing specialized ORNs were identified in each sex of S. lepidus. Among them, three different types of sensilla in the males and two types in the females housed ORNs specialized for pheromone-related compounds. The ORNs in males were specialized for 4-methyl-3,5-heptanedione or one or more of four stereoisomers of 5-hydroxy-4-methyl-3-heptanone. In contrast, female sensilla did not contain ORNs sensitive to 4-methyl-3,5-heptanedione while they contained ORNs sensitive to and specialized for the stereoisomers of (4S,5S)-5-hydroxy-4-methyl-3-heptanone. In addition to the pheromone-related ORNs, four types of olfactory sensilla contained ORNs responsive to plant volatile compounds in male S. lepidus, and five types in females. Most of the ORNs identified in S. lepidus showed a high degree of specificity to specific volatile compounds although some of the active compounds showed overlapping response spectra in the ORNs across different types of sensilla. The most active plant volatile compounds were the four green leaf volatile compounds, (E)-2-hexenol, (Z)-2-hexenol, (Z)-3-hexenol and (E)-2-hexenal, and isomers of two monoterpenols, (+/-)-linalool and (+/-)-alpha-terpineol, all eliciting strong responses from relatively large numbers of ORNs in male and female S. lepidus. Our study indicates that S. lepidus has a set of highly sensitive and selective ORNs for pheromone and plant volatile compounds.     • In female Aedes aegypti L mosquitoes, a blood meal induces physiological and behavioral changes. Previous studies have shown that olfactory receptor neurons (ORNs) housed in grooved peg sensilla on the antennae of Ae. aegypti down-regulate their sensitivity to lactic acid, a key component driving host-seeking behavior, which correlates with observed changes in the host-seeking behavior of this species. In the present study, we performed electrophysiological recordings from the most abundant antennal sensillum type, sensilla trichodea. Our results indicate that the response spectra of ORNs contained within most trichoid sensilla do not change in response to blood feeding. However, we observe an increase in sensitivity to primarily indole and phenolic compounds in neurons housed within four of the five functional types of short blunt tipped II trichoid sensilla, both at 24 and 72 h post-blood feeding, which was more pronounced at 24 h than 72 h. Furthermore, sensitivity to undecanone, acetic acid and propionic acid was observed to increase 72 h post-blood meal. Considering the timing of these changes, we believe that these neurons may be involved in driving the orientation behavior of female mosquitoes to oviposition sites, which are known to release these compounds.     • The quality of food eaten by larval insects will affect traits such as gamete production, fat reserves, muscle bulk and body size in the adult. Moreover, larvae also depend on high moisture content in the diet for survival. The almond moth (Ephestia cautella) (W.) (Lepidoptera; Pyralidae) does not feed as an adult although it continues to drink water. We tested the idea that an almond moth could compensate for a low-water diet as a larva by increasing its water intake as an adult. We reared larvae on two different food sources with different moisture regimes; standard laboratory diet with glycerol (relatively wet) and standard diet without glycerol (relatively dry). Half the adult moths from each treatment were given water to drink before their first and only mating. Our results show that wet larval diets (i.e. containing glycerol) significantly decreased fecundity (total number of eggs laid and the proportion of hatched larvae), whilst it significantly increased male and female longevity. The interaction effect of water access for adult males and females was significant, independent of the glycerol in the larval diet. Longevity in females that were not presented with water as adults was slightly higher if mated with a male that had had access to water, suggesting a mating donation of water. However, females that received water as adults showed a decreased longevity if mated with a male who had also had access to water as an adult, indicating a negative effect of water if received by both males and females. In addition, when the larval diet included glycerol, increased number of eggs laid decreased female longevity, whilst an absence of glycerol in the larval diet resulted in low female longevity that was unlinked with fecundity. Glycerol is used in many artificial insect diets and the fact that it shows a strong effect on key life-history traits (reproductive output and longevity in this species), merits careful re-examination of its effects on these important traits in other laboratory models. We also discuss the possibility that larval diet can affect female reproductive decisions.

    • We have examined the timing of calling behavior in the female Egyptian cotton leafworm, Spodoptera littoralis and its modification by exposure to sex pheromone. The calling rhythm of the female moth was found to be circadian, persistent for at least 4days once it has been entrained, and could be phase shifted by altering the light:dark regime. We also found that female exposure to pheromone affected the rate and duration of calling. A brief exposure to pheromone gland extract increased the proportion of females calling in a constant dim light and this effect persisted for at least 2days. In response to pheromone exposure, significantly more females also called late into scotophase when most unexposed control females had ceased calling. The adaptive significance of responding to conspecific sex pheromone is discussed.

    • Hemocytes and the (prophenol-) phenoloxidase system constitute the immediate innate immune system in insects. These components of insect immunity are present at any post-embryonic life stage without previous infection. Differences between individuals and species in these immune parameters can reflect differences in infection risk, life expectancy, and biological function. In honeybees which show an age-related division of labor within the worker caste, previous studies demonstrated that foragers show a strongly reduced number of hemoctyes compared to the younger nurse bees. This loss of immune competence has been regarded advantageous with respect to an already high mortality rate due to foraging and to redistribution of energy costs at the colony level. Based on the idea that abandoning hemocytes in all adults would be a reasonably direct regulatory mechanism, we posed the hypothesis that abandoning hemocytic immunity is not restricted to worker honeybees. We tested our hypotheses by performing a comprehensive analysis of hemocyte number and phenoloxidase (PO)-activity levels in immunologically naive workers, queens, and drones. We found that in all three adult phenotypes hemocyte number is dramatically reduced in early adult life. In contrast, we found that the dynamics of PO-activity levels have sex and caste-specific characteristics. In workers, PO activity reached a plateau within the first week of adult life, and in queens enzyme levels continuously increased with age and reached levels twice as high as those found in workers. PO-activity levels slightly declined with age in drones. These data support our hypothesis, from which we infer that the previously reported reduction of hemocyte in foragers is not worker specific but represents a general phenomenon occurring in all honeybee adult phenotypes.

    • Metabolic rate, and the flexibility thereof, is a complex trait involving several inter-linked variables that can influence animal energetics, behavior, and ultimately, fitness. Metabolic traits respond readily to ambient temperature variation, in some cases increasing relative or absolute energetic costs, while in other cases, depending on the organism's metabolic and behavioral responses to changing conditions, resulting in substantial energy savings. To gain insight into the rapid recent emergence of the indigenous South African longhorn beetle Cacosceles newmannii as a crop pest in sugarcane, a better understanding of its metabolic rate, feeding response, digestion times, and aerobic scope is required, in conjunction with any behavioral responses to food availability or limitation thereof. Here, we therefore experimentally determined metabolic rate, estimated indirectly as CO2 production using flow-through respirometry, in starved, fasted, and fed C. newmannii larvae, at 20 degrees C and 30 degrees C. We estimated multiple parameters of metabolic rate (starved, standard, active, and maximum metabolic rates) as well as aerobic scope (AS), specific dynamic action (SDA), and the percentage time active during respirometry trials. Additionally, in individuals that showed cyclic or discontinuous gas exchange patterns, we compared rate, volume, and duration of cycles, and how these were influenced by temperature. Standard and active metabolic rate, and AS and SDA were significantly higher in the larvae measured at 30 degrees C than those measured at 20 degrees C. By contrast, starved and maximum metabolic rates and percentage time active were unaffected by temperature. At rest and after digestion was complete, 35% of larvae showed cyclic gas exchange at both temperatures; 5% and 15% showed continuous gas exchange at 20 degrees C and 30 degrees C respectively, and 10% and 0% showed discontinuous gas exchange at 20 degrees C and 30 degrees C respectively. We propose that the ability of C. newmannii larvae to survive extended periods of resource limitation, combined with a rapid ability to process food upon securing resources, even at cooler conditions that would normally suppress digestion in tropical insects, may have contributed to their ability to feed on diverse low energy resources typical of their host plants, and become pests of, and thrive on, a high energy host plant like sugarcane.

    • Many insects have complex life cycles where a drastic ontogenetic change happens between the larval stages and the adult stage, i.e. metamorphosis. Damselflies (order Odonata, suborder Zygoptera) are widely distributed and ecologically important semi-aquatic insects with a complex life cycle. Phenotypic changes over damselfly ontogeny have been documented, however, if and how metabolite profiles are also changing is currently unknown. Here we used a metabolomics methodology to gain insights into the metabolic changes during the life cycle of the Northern damselfly (Coenagrion hastulatum). Hatchlings of wild-caught damselflies were reared in the laboratory and metabolomics analyses using liquid chromatography and gas chromatography coupled to mass spectrometry were carried out at three larval stages and on adult damselflies. Additionally, a subset of larvae was exposed to wastewater effluent to assess how metabolite profiles responded to an environmental stressor. A total of 212 compounds belonging to several classes (e.g. amino acids, fatty acids, sugars) were annotated. Across metamorphosis, we found that damselflies shifted from protein catabolism to lipid catabolism. Wastewater effluent exposure resulted in ontogenetic stage-dependent changes of individual metabolites, but not to a marked extent. Overall, our study is one of the first to describe changes of metabolite profiles during ontogeny of an insect, and it provides a first step towards a greater understanding of the physiological changes occurring during general insect—but especially damselfly—ontogeny.

    • Insects have the capacity to significantly modify their metabolic rate according to environmental conditions and physiological requirement. Consequently, the respiratory patterns can range from continuous gas exchange (CGE) to discontinuous gas exchange (DGE). In the latter, spiracles are kept closed during much of the time, and gas exchange occurs only during short periods when spiracles are opened. While ultimate causes and benefits of DGE remain debated, it is often seen during insect diapause, a deep resting stage that insects induce to survive unfavourable environmental conditions, such as winter. The present study explores the shifts between CGE and DGE during diapause by performing long continuous respirometry measurements at multiple temperatures during key diapause stages in the green-veined white butterfly Pieris napi. The primary goal is to explore respiratory pattern as a non-invasive method to assess whether pupae are in diapause or have transitioned to post-diapause. Respiratory pattern can also provide insight into endogenous processes taking place during diapause, and the prolonged duration of diapause allows for the detailed study of the thermal dependence of the DGE pattern. Pupae change from CGE to DGE a few days after pupation, and this shift coincides with metabolic rate suppression during diapause initiation. Once in diapause, pupae maintain DGE even at elevated temperatures that significantly increase CO2 production. Instead of shifting respiratory pattern to CGE, pupae increase the frequency of DGE cycles. Since total CO2 released during a single open phase remains unchanged, our results suggest that P. napi pupae defend a maximum internal rho CO2 set point, even in their heavily suppressed diapause state. During post-diapause development, CO2 production increases as a function of development and changes to CGE during temperature conditions permissive for development. Taken together, the results show that respiratory patterns are highly regulated during diapause in P. napi and change predictably as diapause progresses.

    • Gene silencing through RNA interference (RNAi) has revolutionized the study of gene function, particularly in non-model insects. However, in Lepidoptera (moths and butterflies) RNAi has many times proven to be difficult to achieve. Most of the negative results have been anecdotal and the positive experiments have not been collected in such a way that they are possible to analyze. In this review, we have collected detailed data from more than 150 experiments including all to date published and many unpublished experiments. Despite a large variation in the data, trends that are found are that RNAi is particularly successful in the family Saturniidae and in genes involved in immunity. On the contrary, gene expression in epidermal tissues seems to be most difficult to silence. In addition, gene silencing by feeding dsRNA requires high concentrations for success. Possible causes for the variability of success in RNAi experiments in Lepidoptera are discussed. The review also points to a need to further investigate the mechanism of RNAi in lepidopteran insects and its possible connection to the innate immune response. Our general understanding of RNAi in Lepidoptera will be further aided in the future as our public database at http://insectacentral.org/RNAi will continue to gather information on RNAi experiments.

Mating activity, female pheromone production/ release and male pheromone responsiveness all show diurnal variations in many species. We found that the response of the male Egyptian cotton leafworm, Spodoptera littoralis, to sex pheromone gland extracts showed a diel rhythm in olfactometer tests, and the variation was persistent for at least 1 day in constant darkness. High male response to sex pheromone was correlated in time with high mating and locomotor activity. Male S. littoralis, maintained in constant darkness and exposed to pheromone gland extracts on a daily basis, showed an induced temporal variation in response after several days, in contrast to unexposed males. This suggests that in the absence of other external zeitgebers, exposure to sex pheromone may function to synchronise circadian behavioural rhythms in male moths. The daily rhythm in mating activity in S. littoralis is also shown to be persistent for at least 2 days in constant darkness. Pairs mated significantly less when either the male or female had been raised in a light:dark cycle 10h out of phase, indicating that the proposed circadian rhythm in mating activity is composed of rhythmic mating preference/ability in both sexes.     • Across a wide range of taxa, individuals within populations exhibit alternative life history strategies (ALHS) where their phenotypes dramatically differ due to divergent investments in growth, reproduction and survivorship, with the resulting trade-offs directly impacting Darwinian fitness. Though the maintenance of ALHS within populations is fairly well understood, little is known regarding the physiological mechanisms that underlie ALHS and how environmental conditions can affect the evolution and expression of these phenotypes. One such ALHS, known as Alba, exists within females of many species in the butterfly genus Colias. Previous works in New World species not only found that female morphs differ in their wing color due to a reallocation of resources away from the synthesis of wing pigments to other areas of development, but also that temperature played an important role in these trade-offs. Here we build on previous work conducted in New World species by measuring life history traits and conducting lipidomics on individuals reared at hot and cold temperatures in the Old World species Colias croceus. Results suggest that the fitness of Alba and orange morphs likely varies with rearing temperature, where Alba females have higher fitness in cold conditions and orange in warm. Additionally shared traits between Old and New World species suggest the Alba mechanism is likely conserved across the genus. Finally, in the cold treatment we observe an intermediate yellow morph that may have decreased fitness due to slower larval development. This cost may manifest as disruptive selection in the field, thereby favoring the maintenance of the two discrete morphs. Taken together these results add insights into the evolution of, and the selection on, the Alba ALHS.

    • The blue gum psyllid, Ctenarytaina eucalypti (Sternorrhyncha: Psyllidae), is an economic threat to Eucalyptus subgenus Symphyomyrtus plantations worldwide. To date, no generally applicable control method is available and the potential for semiochemical-based monitoring or control methods has not yet been investigated. Hence, we conducted the first study on the olfactory sense of C. eucalypti, investigating the specificity and sensitivity of its olfactory sensory neurons (OSNs) to host plant volatiles using single sensillum recordings (SSR). Synthetic compounds were selected from published identifications of Eucalyptus volatiles and after analysis of headspace collections from Eucalyptus cordata. The antenna of C. eucalypti carries four cavities containing olfactory sensilla (S1-S4). Our recordings revealed that each of these sensilla houses three OSNs that could be distinguished electrophysiologically based on spike amplitude differences (A, B, and C neuron with large, intermediate, and small amplitude, respectively). The A neuron in sensillum S1 responded primarily to β-caryophyllene and weaker to β-ocimene, whereas the accompanying B-neuron responded strongly and very specifically to linalool. Furthermore, the B-neuron in both S2 and S3 responded strongly to 1-hexanol, Z3-hexenol, and Z3-hexenyl acetate. OSNs in S4 responded only weakly to a few of the synthetic compounds. Response thresholds in strongly responding OSNs to putative key compounds were close to the 1ng dose on the filter paper and responses exhibited a phasic-tonic profile irrespective of compound dose. C. eucalypti may use the physiologically active compounds for long-range host finding. Future laboratory and field experiments will reveal whether plant volatiles can be used in the management and monitoring of C. eucalypti.

    • In the female turnip moth, Agrotis segetum, a pheromone biosynthesis activating neuropeptide (PBAN) stimulates sex pheromone biosynthesis which exhibits a daily rhythm. Here we show data supporting a circadian rhythm in PBAN release from the corpora cardiaca, which we propose regulates the endogenous rhythm in sex pheromone biosynthesis. This conclusion is drawn as the observed daily rhythm in PBAN-like immunoreactivity, in the hemolymph is persistent in constant darkness and is phase-shifted by an advanced light:dark cycle. PBAN-like immunoreactivity was found in the brain, the optic lobe, the suboesophageal ganglion and in the retrocerebral complex. In each hemisphere ca. 10 immunopositive neurons were observed in the pars intercerebralis and a pair of stained somata in the dorso-lateral protocerebrum. A cluster of cells containing PBAN-like immunoreactive material was found in the tritocerebrum and three clusters of such cells were found in the SOG. Their processes reach the corpora cardiaca via nervi corporis cardiaci and the dorsal surface of the corpora allata via the nervi corporis allati. (C) 2008 Elsevier Ltd. All rights reserved.

  • Bovine Osteopontin Modifies the Intestinal Transcriptome of Formula-Fed Infant Rhesus Monkeys to Be More Similar to Those That Were Breastfed

    • Background: Osteopontin (OPN) is a multifunctional protein found in human milk at high concentration.Objective: The impact of supplemental bovine OPN on growth, body composition, and the jejunal transcriptome was assessed.Methods: Newborn rhesus monkeys were randomly assigned to be breastfed (n = 4) or to receive formula [formula fed (FF), n = 6] or formula supplemented with 125 mg/L of bovine OPN (bOPN, n = 6) for 3 mo. Jejunal mRNA was extracted and subjected to microarray analysis.Results: Growth was similar among all the treatment groups, but breastfed monkeys were similar to 25% leaner at 3 mo. Pairwise comparisons demonstrated that 1017 genes were differentially expressed between breastfed and FF groups, 217 between breastfed and bOPN groups, and 119 between FF and bOPN groups. The data were also analyzed with the use of weighted gene coexpression network analysis, which revealed 6 modules of coexpressed genes that differed among the 3 treatments. Nearly 50% of genes were assigned to one module in which breastfed differed from FF and bOPN expression was intermediate. This module was enriched for genes related to cell adhesion and motility, cytoskeletal remodeling, wingless and integration site signaling, and neuronal development. Most of these canonical pathways centered on integrins, which are receptors for OPN.Conclusions: The intestinal transcriptome of breastfed and FF monkeys differs, but bovine OPN at levels similar to human milk shifts gene expression profiles to be more similar to breastfed monkeys.

  • Becoming American : Intermarriage during the great migration to the United States

    • Although intermarriage is a common indicator of immigrant integration into host societies, most research has focused on how individual characteristics determine intermarriage. This study uses the 1910 ipums census sample to analyze how contextual factors affected intermarriage among European immigrants in the United States. Newly available, complete-count census microdata permit the construction of contextual measures at a much lower level of aggregation—the county—in this analysis than in previous studies. Our results confirm most findings in previous research relating to individual-level variables but also find important associations between contextual factors and marital outcomes. The relative size and sex ratio of an origin group, ethnic diversity, the share of the native-born white population, and the proportion of life that immigrants spent in the United State are all associated with exogamy. These patterns are highly similar across genders and immigrant generations.

  • Using HTK for Prolonged Pancreas Preservation Prior to Human Islet Isolation

Histidine-tryptophan-ketoglutarate (HTK) has been established as an alternative to University-of-Wisconsin solution (UWS) for abdominal organ preservation, but data about HTK efficiency to preserve pancreata during prolonged cold ischemia time (CIT) are conflicting. In human islet transplantation, HTK provided similar isolation outcomes after short CIT. The present study aimed to investigate whether islets can be successfully isolated from HTK-preserved pancreata after prolonged CIT compared with UWS. Materials and Methods. Sixty-four human pancreata retrieved from donors meeting criteria for kidney donation were perfused utilizing either HTK or UWS and preserved for more or less than 10 h prior to islet isolation. Along with parameters related to isolation and islet quality assessment, the dry-to-wet weight ratio was evaluated. Results. Donor-and procurement-related factors did not vary between HTK- and UWS-perfused pancreata. The dry-to-wet weight ratio was lower in HTK-preserved pancreata indicated tissue edema (21.0% +/- 3.5% versus 24.8% +/- 2.0%, P = 0.007). Isolation-related variables differed between experimental groups after prolonged CIT with respect to purified packed tissue volume (9.1 +/- 5.0 versus 17.2 +/- 8.1 mu L/g, P = 0.004) and islet yield (1910 +/- 980 versus 3150 +/- 1420 IE/g, P = 0.012). Islet purity and survival after culture were similar after HTK or UWS perfusion. The preservation solution did not affect in vitro function and transplantability of isolated islets. Conclusions. Compared with UWS, HTK has similar efficiency to preserve human pancreata for subsequent islet isolation during <10 h CIT but seems to be limited for prolonged cold storage. (C) 2012 Elsevier Inc.
