    • Reasons for performing study: Neuromuscular electrical stimulation (NMES) is used to increase or maintain muscle strength during rehabilitation. Human studies investigating different protocols show that some treatments induce changes in muscle characteristics. Despite the frequent use of NMES in horses, no studies have been published describing its efficacy.Objectives: To investigate the effects of a NMES protocol on equine fibre types and areas, glycogen concentrations and enzyme activities.Methods: NMES was administrated to m. gluteus medius and m. longissimus dorsi, on one side of 6 healthy Standardbred horses. The contralateral side of each muscle served as a nonstimulated control. The horses were stimulated at 50 Hz a day, with 21-39 mA, for 45-60 min, 5 days a week for 4 weeks. Needle biopsies were obtained from the muscles on both sides before and after the experimental period. Muscle samples were analysed for fibre type proportions and area using histochemical methods and for glycogen and enzyme activities (citrate synthase, 3-OH-acyl CoA dehydrogenase, hexokinase and lactate dehydrogenase) using biochemical methods. Muscle contractions at the location and depth of the muscle biopsy were confirmed by diagnostic ultrasound. Nonparametric tests (Mann-Whitney, Wilcoxon sign-rank) were used for statistical analysis. Results: No significant differences were observed in the percentage of types I, IIA or IIX fibres, fibre areas, glycogen levels or enzyme activities either when comparing stimulated and nonstimulated muscles before and after the NMES treatment, or when comparing the left and right muscle samples.Conclusions: The NMES treatment was well tolerated by the horses, but the present protocol did not induce significant muscle adaptations. Further studies are needed to describe the effect of more intense and/or prolonged NMES treatment protocols on muscles of healthy horses, and to describe if stimulation protocols induce positive changes in atrophied muscles.

  • Atrial fibrillatory rate as predictor of recurrence of atrial fibrillation in horses treated medically or with electrical cardioversion

    • Background: The recurrence rate of atrial fibrillation (AF) in horses after cardioversion to sinus rhythm (SR) is relatively high. Atrial fibrillatory rate (AFR) derived from surface ECG is considered a biomarker for electrical remodelling and could potentially be used for the prediction of successful AF cardioversion and AF recurrence. Objectives: Evaluate if AFR was associated with successful treatment and could predict AF recurrence in horses. Study design: Retrospective multicentre study. Methods: Electrocardiograms (ECG) from horses with persistent AF admitted for cardioversion with either medical treatment (quinidine) or transvenous electrical cardioversion (TVEC) were included. Bipolar surface ECG recordings were analysed by spatiotemporal cancellation of QRST complexes and calculation of AFR from the remaining atrial signal. Kaplan-Meier survival curve and Cox regression analyses were performed to assess the relationship between AFR and the risk of AF recurrence. Results: Of the 195 horses included, 74 received quinidine treatment and 121 were treated with TVEC. Ten horses did not cardiovert to SR after quinidine treatment and AFR was higher in these, compared with the horses that successfully cardioverted to SR (median [interquartile range]), (383 [367-422] vs 351 [332-389] fibrillations per minute (fpm), P <.01). Within the first 180 days following AF cardioversion, 12% of the quinidine and 34% of TVEC horses had AF recurrence. For the horses successfully cardioverted with TVEC, AFR above 380 fpm was significantly associated with AF recurrence (hazard ratio = 2.4, 95% confidence interval 1.2-4.8, P =.01). Main limitations: The treatment groups were different and not randomly allocated, therefore the two treatments cannot be compared. Medical records and the follow-up strategy varied between the centres. Conclusions: High AFR is associated with failure of quinidine cardioversion and AF recurrence after successful TVEC. As a noninvasive marker that can be retrieved from surface ECG, AFR can be clinically useful in predicting the probability of responding to quinidine treatment as well as maintaining SR after electrical cardioversion.

  • Asymmetries of horses walking and trotting on treadmill with and without rider

    • Background Left-right movement symmetry is a highly desirable characteristic in sport horses.Objectives This study compared movement symmetry in well-trained dressage horses in unridden and unrestrained position and ridden in a dressage frame, and investigated possible associations between gaits.Study design Experimental study.Methods Seven sound, high-level dressage horses were measured at walk and sitting trot on a treadmill at several speeds under two conditions: with and without rider. Left-right differences in stance duration, stance protraction and retraction based on longitudinal hoof positions, ipsilateral limb tracking, minimum and maximum vertical positions of the dorsal spinous processes of the sixth thoracic (T6), third sacral vertebrae (S3) and wing of atlas, and vertical ground reaction forces were calculated and analysed in mixed models.Results At walk, five body variables indicated increased asymmetry in the ridden condition compared with unridden condition: forelimb stance duration (unridden/ridden left-right differences 9 vs 13 ms; P = .008), forelimb stance protraction (P = .004), stance retraction (P = .001) and first force peak (P = .003), and hindlimb stance retraction (P = .01). At trot, six body variables were more asymmetrical in the ridden condition: forelimb stance duration (2.5 vs 3.8 ms, P = .004); hindlimb stance protraction (P < .0001) and retraction (P = .01), T6 minimum (4 vs 6 mm, P = .001), T6 maximum (9 vs 11 mm, P = .01) and S3 maximum (6 vs 12 mm, P < .001). Five variables had significant associations between asymmetries at walk and trot, but only three demonstrated a positive slope.Main limitations A limited number of horses and riders were studied. Measurements were performed on a treadmill.Conclusions High-level horses moved slightly more asymmetrically when ridden in a dressage frame than in the unridden condition.

    • Both flocking material and girthing have a significant influence on the saddle pressure and should thus be considered in saddle fitting. Wool seems a better flocking material than foam of the type used in the current study. For girthing, traditional placement seems equally good if not better than the v-system. However, further studies are needed to show if these results are valid for a larger population of riding horses.

Basic knowledge about rider and saddle movements in well-performing horses is likely to be supportive to this task.

    • Background To achieve an excellent functional and cosmetic result, primary closure is preferred over leaving wounds to heal by secondary intention. However, traumatic wounds are often under excessive tension during wound closure and incorrect suture technique can compromise microcirculation, leading to skin necrosis and impaired wound healing.Objective To describe an inexpensive and effective tension relief technique that helps the successful primary closure of a variety of equine wounds at high risk of dehiscence.Study design Retrospective case series.MethodsAll wounds that were managed with the Tension Tile System (TTS) at four Equine Hospitals between March 2017 and May 2021 were evaluated. The wounds were classified according to various criteria including anatomical location, time elapsed prior to surgery, depth of wound and post-surgical use of immobilisation. Outcome criteria were based on the success of primary intention healing. The duration of convalescence (weeks) after surgery was also recorded.Results During the study period, the TTS was used in 191/860 (22%) wounds repaired under general anaesthesia or standing sedation. Overall, primary intention healing (Group A) was achieved in 132 of 191 cases (69%, CI 62%-75%), with partial dehiscence (Group B) in a further 30/191 cases (16%, CI 11%-22%). Severe dehiscence (Group C) was recorded in 29/191 cases (15%, CI 11%-21%). The median convalescence time was 4 weeks (Range 3-15, interquartile range 4-6) in Group A.Main limitations Retrospective nature of the study and subjective outcome assessment. The technique was applied to wounds under significant tension; however, this was based on a subjective assessment by the surgeons involved.Conclusions The Tension Tile System is an economical and effective technique for challenging equine wounds under tension, in a variety of anatomical locations.

    • Reasons for performing studyStatic magnetic blankets are often claimed to induce increases in blood flow, reduce muscle tension and tenderness, and be beneficial in both prevention and treatment of musculoskeletal injuries in horses. However, there are no studies that confirm alleged beneficial effects of magnets on muscles of the back in healthy horses. ObjectivesTo investigate whether static magnets sewn into a blanket affect back muscle blood flow, skin temperature, mechanical nociceptive threshold (MNT) and behaviour in healthy horses. Study designProspective, randomised, blinded, placebo-controlled crossover study. MethodsThe following outcome measurements of the back of 10 healthy horses were performed; blood flow by photoplethysmography, skin temperature by use of thermistors in conjunction with digital infrared thermography, and MNTs by algometry. The horses behaviour was filmed during the procedure and scored on an ethogram. Measurements were performed repeatedly for a 30min baseline period. Thereafter a blanket with active, static magnets (900 gauss) or placebo magnets was placed on the horse and measurements were performed for a 60min treatment period and a 30min post treatment period. The study procedure was repeated on the consecutive day, when the horse received the alternative treatment. ResultsBlood flow in muscle, skin temperatures, MNTs and behavioural traits did not differ between active and placebo magnetic blankets. Skin temperature increased similarly during both active and placebo blanket treatment. ConclusionsIn healthy horses, magnetic blankets did not induce additional significant effects on muscle blood flow, skin temperature, MNTs and behaviour when compared with nonmagnetic blankets.

  • Lungeing on hard and soft surfaces: Movement symmetry of trotting horses considered sound by their owners

The difference in movement symmetry, which is a commonly employed lameness measure, has not been quantified between surfaces.ObjectivesTo compare head and pelvic movement symmetry between surfaces and reins during lungeing.Study designQuantitative gait analysis in 23 horses considered sound by their owners.MethodsTwenty-three horses were assessed in-hand and on the lunge on both reins on hard and soft surfaces with inertial sensors. Seven movement symmetry parameters were quantified and used to establish 2 groups, namely symmetrical (n = 9) and forelimb-lame horses (n = 14), based on values from straight-line assessment. Movement symmetry values for left rein measurements were side corrected to allow comparison of the amount of movement symmetry between reins. A mixed model (P<0.05) was used to study effects on movement symmetry of surface (hard/soft) and rein (inside/outside with respect to movement symmetry on the straight).ResultsIn forelimb-lame horses, surface and rein were identified as significantly affecting all head movement symmetry measures (rein, all P<0.0001; surface, all P<0.042). In the symmetrical group, no significant influence of surface or rein was identified for head movement symmetry (rein, all P>0.245; surface, all P>0.073). No significant influence of surface or rein was identified for any of the pelvic movement symmetry measures in either group.ConclusionsWhile more symmetrical horses showed a consistent amount of movement symmetry across surfaces/reins, horses objectively quantified as lame on the straight showed decreased movement symmetry during lungeing, in particular with the lame limb on the inside of a hard circle. The variation within group questions straight-line movement symmetry as a sole measure of lameness without quantification of movement symmetry on the lunge, ideally on hard and soft surfaces to evaluate differences between reins and surfaces. In future, thresholds for lungeing need to be determined using simultaneous visual and objective assessment.

    • Background The increasing popularity of objective gait analysis makes application in prepurchase examinations (PPE) a logical next step. Therefore, there is a need to have more understanding of asymmetry during a PPE in horses described on clinical evaluation as subtly lame.Objectives The objective of this study is to objectively compare asymmetry in horses raising minor vet concerns in a PPE and in horses raising major vet concerns with that found in horses presented with subtle single-limb lameness, and to investigate the effect of age/discipline on the clinicians' interpretation of asymmetry on the classification of minor vet concerns in a PPE.Study Design Clinical case-series.Methods Horses presented for PPE (n = 98) or subjectively evaluated as single limb low-grade (1-2/5) lame (n = 24, 13 forelimb lame, 11 hindlimb lame), from the patient population of a single clinic, were enrolled in the study provided that owners were willing to participate. Horses undergoing PPE were assigned a classification of having minor vet concerns (n = 84) or major vet concerns (n = 14) based on findings during the dynamic-orthopaedic part of the PPE. Lame horses were only included if pain-related lameness was confirmed by an objective improvement after diagnostic analgesia exceeding daily variation determined for equine symmetry parameters using optical motion capture. Clinical evaluation was performed by six different clinicians, each with >= 8 years of equine orthopaedic experience. Vertical movement symmetry was measured using optical motion capture, simultaneously with the orthopaedic examination. Data were analysed using previously described parameters and mixed model analysis and least squares means were used to calculate differences between groups.Results There was no effect of age or discipline on the levels of asymmetry within PPE horses raising minor vet concerns. MinDiff and RUD of the head discriminated between forelimb lame and PPE horses raising minor vet concerns; MinDiff, MaxDiff, RUD of the Pelvis, HHDswing and HHDstance did so for hindlimb lameness. Two lameness patterns differentiated both forelimb and hindlimb lame from PPE horses with minor vet concerns: RUD Poll + MinDiff Withers - RUD Pelvis and RUD Pelvis + RUD Poll - MinDiff Withers. Correcting for vertical range of motion enabled differentiation of PPE horses with minor vet concerns from PPE horses with major vet concerns.Main Limitations Objective data only based on trot on soft surface, limited number of PPE horses with major vet concerns.Conclusions Combinations of kinematic parameters discriminate between PPE horses with minor vet concerns and subtly lame horses, though overlap exists.

    • Background Quantitative gait analysis offers objective information to support clinical decision-making during lameness workups including advantages in terms of documentation, communication, education, and avoidance of expectation bias. Nevertheless, hardly any data exist comparing outcome of subjective scoring with the output of objective gait analysis systems. Objectives To investigate between- and within-veterinarian agreement on primary lame limb and lameness grade, and to determine relationships between subjective lameness grade and quantitative data, focusing on differences between (1) veterinarians, (2) live vs video assessment, (3) baseline assessment vs assessment following diagnostic analgesia. Study design Clinical observational study. Methods Kinematic data were compared to subjective lameness assessment by clinicians with >= 8 years of orthopaedic experience. Subjective assessments and kinematic data for baseline trot-ups and response to 48 diagnostic analgesia interventions in 23 cases were included. Between and within-veterinarian agreement was investigated using Cohen's Kappa (kappa). Asymmetry parameters for kinematic data ('forelimb lame pattern', 'hindlimb lame pattern', 'overall symmetry', 'vector sum head', 'pelvic sum') were determined, and used as outcome variables in mixed models; explanatory variables were subjective lameness grade and its interaction with (1) veterinarian, (2) live or video evaluation and (3) baseline or diagnostic analgesia assessment. Results Agreement on lame limb between live and video assessment was 'good' between and within veterinarians (median kappa = 0.64 and kappa = 0.53). There was a positive correlation between subjective scoring and measured asymmetry. The relationship between lameness grade and objective asymmetry differed slightly between (1) veterinarians (for all combined parameters, p-values between P < .001 and 0.04), (2) between live and video assessments ('forelimb lame pattern', 'overall symmetry', both P <= .001), and (3) between baseline and diagnostic analgesia assessment (all combined parameters, between P < .001 and .007). Main limitations Limited number of veterinarians (n = 4) and cases (n = 23), only straight-line soft surface data, different number of subjective assessments live vs from video. Conclusions Overall, between- and within-veterinarian agreement on lame limb was 'good', whereas agreement on lameness grade was 'acceptable' to 'poor'. Quantitative data and subjective assessments correlated well, with minor though significant differences in the number of millimetres, equivalent to one lameness grade between veterinarians, and between assessment conditions. Differences between baseline assessment vs assessment following diagnostic analgesia suggest that addition of objective data can be beneficial to reduce expectation bias. The small differences between live and video assessments support the use of high-quality videos for documentation, communication, and education, thus, complementing objective gait analysis data.

  • Effect of circadian rhythm, age, training and acute lameness on serum concentrations of cartilage oligomeric matrix protein (COMP) neo-epitope in horses

    • Background Molecular serum markers that can identify early reversible osteoarthritis (OA) in horses are lacking. Objectives We studied serum concentrations of a novel cartilage oligomeric matrix protein (COMP) neo-epitope in horses subjected to short-term exercise and with acute lameness. The effects of circadian rhythm and age were also evaluated. Study design Longitudinal studies in healthy horses and cross-sectional comparison of lame and non-lame horses. Methods Sera were collected from five horses before and after short-term interval exercise and during full-day box rest. Sera from 32 acutely lame horses were used to evaluate age-related effects. Independent samples from control horses (n = 41) and horses with acute lameness (n = 71) were included. COMP neo-epitope concentrations were analysed using custom-developed inhibition ELISAs validated for equine serum. The presence of COMP neo-epitope was delineated in healthy and osteoarthritic articular cartilage with immunohistochemistry. Results COMP neo-epitope concentrations decreased after speed training but returned to baseline levels post-exercise. No correlations between age and serum COMP neo-epitope concentrations were found (r = 0.0013). The mean (+/- s.d.) serum concentration of COMP neo-epitope in independent samples from non-lame horses was 0.84 +/- 0.38 mu g/mL, and for lame horses was 5.24 +/- 1.83 mu g/mL (P<0.001). Antibodies against COMP neo-epitope did not stain normal articular cartilage, but intracytoplasmic staining was found in superficial chondrocytes of mild OA cartilage and in the extracellular matrix of moderately osteoarthritic cartilage. Main limitations ELISA was based on polyclonal antisera rather than a monoclonal antibody. There is a sex and breed bias within the groups of horses, also it could have been of value to include horses with septic arthritis and tendonitis and investigated joint differences. Conclusions This COMP neo-epitope can be measured in sera, and results indicate that it could be a biomarker for pathologic fragmentation of cartilage in connection with acute joint lameness.

    • Reasons for performing study: One of the most common causes of lameness in racehorses is osteoarthritis (OA). Pathogenesis is not clear and pathological processes of the different joint tissues interact in often progressive events. The interface between cartilage and newly synthesised bone has been shown to be particularly enriched in bone sialoprotein (BSP), a cell-binding matrix protein. Objectives: To establish whether changes in the concentration of BSP may serve as a marker for early biochemical changes of the subchondral bone. Methods: Articular cartilage, cartilage/bone interface and subchondral bone of the proximal third carpal bone from 3 Standardbred trotters were analysed ultrastructurally for the presence of BSP in normal and degenerative areas. Results: A marked increase of BSP in the cartilage/bone interface with degenerative changes of the bone and cartilage compared to the morphologically intact cartilage/bone interface was noted, but levels of the protein were distinctly lower in the distal bone. Conclusions: The results indicate that BSP has the potential to be used as a marker for changes in bone metabolism in the subchondral one. Potential relevance: Tools to monitor early biochemical changes within the connective tissues of the joint in vivo are essential in studies of the pathogenesis of OA. These could be used to monitor and understand such changes in relation to load, exercise, training programmes, inflammation and the development of OA.

A water treadmill provides resistance training on flexor tendon muscles, which might reduce the risk of tendon injury.Objective:To determine the effect of water treadmill training on the properties of the gluteal and superficial digital flexor (SDF) muscles and on cardiocirculatory response to a standardised exercise test.Methods:Five healthy unfit horses were trained on a water treadmill for 5 days/week for 4 weeks, starting with 5 min/day increasing to 20 min/day. Before and after the water treadmill training, an incremental SET was performed on a land treadmill to determine velocity at a heart rate 200 beats/min (V(200)) and resting gluteal and SDF muscle biopsies were obtained for biochemical analyses.Results:There was no measurable difference in resting concentrations of gluteal or SDF muscle glycogen, lactate, ATP or glucose-6-phosphate (G6P), or activities of citrate synthase (CS), 3-hydroxyacyl CoA dehydrogenase (HAD) and lactate dehydrogenase (LDH) after training and no change in V(200), Lactate, glycogen, G6P and ATP concentrations were 50% lower and type 1 fibres 30% higher in SDF compared to gluteal muscles. CS and HAD activities were similar between SDF and gluteal, while LDH was lower in the SDF muscle.Conclusions:A more strenuous water treadmill conditioning protocol may be needed to induce a training effect in gluteal and SDF muscle and heart rate response. The low substrate concentrations and oxidative capacity of SDF may predispose this muscle to catastrophic fatigue during maximal exercise.

High energy forage-only diets may be an alternative to concentrate rich diets and may provide high levels of CP.Objective:To study the effect of feeding 2 forage-only diets, containing either high or moderate CP concentrations on glycogen and free pool amino acid concentrations in the muscle.Materials and methods:Two high energy forage-only diets based on high-energy grass forage were fed for 23 days in a crossover design to 6 Standardbred horses in racing condition. One forage diet provided a high (HP) CP (16.6%) intake and the other diet provided recommended intake (RP) of CP (12.5%). At Day 19 a standardised treadmill test was performed to mimic a race. Blood samples were taken before, during and after (up to 90 min) the treadmill test and muscle biopsies (m. gluteus) were taken before and after exercise and after 90 min. Amino acids were analysed with a HPLC-technique and glycogen with a fluorimetric method.Results:A main effect of the HP diet was that muscle glycogen and leucine concentrations were higher compared to the RP diet. Branched chain amino acid concentrations in plasma remained higher during early recovery from exercise on the HP diet compared to the RP diet. Intense exercise caused a similar decrease in glycogen, aspartate and glutamate concentrations in muscle and increase in alanine concentration on both diets.Conclusion:Feeding a forage-only diet with a high CP intake increases glycogen and leucine concentrations in muscle of horses in training. This may be beneficial for muscle recovery following intensive exercise.

The 2 diets were fed in 2 experimental periods of 3 weeks each in a crossover design. The last day of each period the horses were fasted for 12 h. The horses were weighed and their water intake measured every day during the trial and every hour during the 12 h feed deprivation. During feed deprivation total plasma protein (TPP), insulin, nonesterified fatty acids (NEFA), urea, glucose and acetate concentrations were analysed.Results:Bwt and water intake was higher on FD compared to OD. Bwt loss was higher during feed deprivation on FD compared to OD. TPP was lower before and during the last 8 h of feed deprivation on FD compared to OD. Plasma insulin was lower on FD than on OD at feeding and for 5 h during feed deprivation. Plasma NEFA and urea increased on both diets during feed deprivation. Plasma glucose was not affected by diet or feed deprivation.Conclusion:High energy forage diets could be an alternative to high grain diets for athletic horses. The small increase in bwt on FD diminished with feed deprivation and the low TPP concentration indicate a greater potential to use an internal fluid compartment to maintain plasma volume.

A specific hydrosurgical debridement device (Versajet (TM))(a) is gentle to viable tissues, yet effectively removes macroscopic contaminants and debris. We wished to investigate whether it is effective in reducing bacterial burden and whether it differs from traditional methods. No previous reports compare hydrosurgical debridement and conventional wound debridement with regard to bacterial reduction from in vitro inoculated soft tissue.Objectives: To assess the effectiveness of hydrosurgical debridement in reducing the Staphylococcus aureus load from in vitro inoculated equine muscle compared with conventional wound debridement methods.Study design: In vitro experimental study.Methods: The surface of equine masseter muscle was inoculated with a S. aureus broth and subsequently debrided using one of the following 4 methods: saline irrigation; sharp debridement; saline irrigation and sharp debridement; or hydrosurgical debridement. Tissue samples for quantitative cultures were collected before and after debridement, and the colony-forming units per gram of tissue were calculated and log transformed. The reductions in bacterial counts were analysed statistically using Wilcoxon signed-rank tests and Friedman two-way ANOVA.Results: Hydrosurgical debridement was more effective than conventional debridement methods in reducing the S. aureus load (P<0.05). Hydrosurgical debridement reduced the bacterial load by 99.7%, in comparison to saline irrigation and sharp debridement (87.4%), sharp debridement (82.2%) and saline irrigation (46.0%).Conclusions: Hydrosurgical debridement reduces the S. aureus load from in vitro contaminated equine muscle significantly more than conventional debridement methods.

    • Reasons for performing study Lungeing is an important part of lameness examinations as the circular path may accentuate low-grade lameness. Movement asymmetries related to the circular path, to compensatory movements and to pain make the lameness evaluation complex. Scientific studies have shown high inter-rater variation when assessing lameness during straight line movement. Objectives The aim was to estimate inter- and intra-rater agreement of equine veterinarians evaluating lameness from videos of sound and lame horses during lungeing and to investigate the influence of veterinarians' experience and the objective degree of movement asymmetry on rater agreement. Study design Cross-sectional observational study. Methods Video recordings and quantitative gait analysis with inertial sensors were performed in 23 riding horses of various breeds. The horses were examined at trot on a straight line and during lungeing on soft or hard surfaces in both directions. One video sequence was recorded per condition and the horses were classified as forelimb lame, hindlimb lame or sound from objective straight line symmetry measurements. Equine veterinarians (n = 86), including 43 with >5 years of orthopaedic experience, participated in a web-based survey and were asked to identify the lamest limb on 60 videos, including 10 repeats. The agreements between (inter-rater) and within (intra-rater) veterinarians were analysed with statistics (Fleiss, Cohen). Results Inter-rater agreement was 0.31 (0.38/0.25 for experienced/less experienced) and higher for forelimb (0.33) than for hindlimb lameness (0.11) or soundness (0.08) evaluation. Median intra-rater agreement was 0.57. Conclusions Inter-rater agreement was poor for less experienced raters, and for all raters when evaluating hindlimb lameness. Since identification of the lame limb/limbs is a prerequisite for successful diagnosis, treatment and recovery, the high inter-rater variation when evaluating lameness on the lunge is likely to influence the accuracy and repeatability of lameness examinations and, indirectly, the efficacy of treatment.

Identification of training methods that may reduce responses to separation would be useful in practice. Objectives: To investigate whether the initial presence of a familiar companion horse modifies responses to separation from the group, lowers stress levels (as measured by heart rate) and increases training efficiency. Hypothesis: Habituation to separation proceeds more quickly if the horse is first trained with a companion, and heart rate is lower when the horse is subsequently trained alone, compared to control horses trained individually from the start. Methods: Young mares (n = 32), kept in groups of 4 were exposed to social separation: 2 horses of the group were trained singly (S1, n = 16) and the remaining 2 horses (n = 16) were trained first with a companion (P2) and then alone (P1). The training comprised 3 steps whereby distance from the group was gradually increased. The final learning criterion was met when a horse fed calmly alone inside a test arena (Step 3). Horses that were trained in a pair had to succeed in Step 3 together before they repeated the steps alone. Feeding behaviour and heart rate were recorded. Results: There were no significant differences between S1 and P1 horses, indicating that the initial pair-training did not reduce responses in P1 horses. However, heart rate was significantly lower when horses were trained in pairs (P2) compared to when the same horses were subsequently trained alone (P1). Conclusions and potential relevance: It may not be efficient to habituate naïve young horses to social separation initially with a partner as these horses appear to have to relearn being in the test situation alone when switching to the individual training

    • Background Equine recurrent uveitis (ERU) is the leading cause of blindness for horses; previous research implicated the leopard complex spotting allele (LP) as a genetic risk factor for insidious uveitis in the Appaloosa. There is limited information about risk in the Knabstrupper. Objective To evaluate clinical manifestations, disease frequency and potential risk factors for ERU in Knabstrupper horses. Study design Cross-sectional study. Methods Ocular examinations were performed on 116 horses, and based on identified anomalies, horses were classified as suspect, ERU-affected or having no clinical signs. Microagglutination testing (MAT) of serum assessed exposure to Leptospira spp. Clinical signs, age, sex, base colour, coat pattern, LP and PATN1 genotypes, percent white at birth, progressive roaning and Leptospira were assessed as risk factors using multivariable exact logistic regression, accounting for clustering at the barn level. Additionally, a pedigree analysis was performed (n = 20 cases and 21 controls), and coefficients of coancestry (CC) and inbreeding were calculated. Results Prevalence of insidious uveitis in this sample of Knabstruppers was 20.7%. Similar to findings for Appaloosas, LP homozygotes had higher odds of uveitis compared with true solid (N/N) horses (LP/LP OR = 7.64, 95% CI [0.8 to +INF], p = 0.04) and age was also identified as a risk factor. After accounting for LP, the 16-20 age group had higher odds compared with the youngest group (OR = 13.36, 95% CI [1.4-213.4], p = 0.009). The distributions of average CC were significantly different between cases and controls (p = 0.01). Main limitations A relatively small sample size decreased the power for detecting additional associations. The progressive nature of insidious uveitis may have prevented identification of younger affected horses. Conclusions Our data support genotyping for LP to assess risk of ERU in Knabstruppers. Additional studies are necessary to develop more robust risk models across LP breeds for earlier detection and improved clinical management.

    • Background There is a paucity of research describing the gait pattern of lame horses at the walk. Objectives To describe the changes in motion pattern and vertical ground reaction forces (GRFz) in horses with induced forelimb lameness at the walk and compare those changes with the changes observed at the trot. Study design Experimental study. Methods In 10 clinically sound Warmblood horses, moderate forelimb lameness was induced using a sole pressure model followed by trot and walk on a treadmill. Kinematic data were collected using 3D optical motion capture (OMC), and GRFz by an instrumented treadmill. Mixed models were used to compare sound baseline versus forelimb lameness (significance was set atP < .05). Results Lameness induction significantly reduced peak GRFz on the second force peak, and vertical impulse in the lame limb. Stride and stance duration in all limbs were reduced. Lameness significantly affected the vertical movement symmetry of the head and withers. Maximum limb retraction angle, fetlock extension and protraction speed were reduced in the lame limb. Body centre of mass (COM) translation was reduced in the side-to-side direction and increased in the vertical and fore-aft directions. Several compensatory kinetic and kinematic changes were observed in the nonlame limbs. The observed changes in both kinetics and kinematics were generally smaller at walk with fewer variables being affected, compared to the trot. Main limitations Only one degree and type of orthopaedic pain (sole pressure) was studied. Conclusions Compensatory strategies of forelimb lameness at the walk include alteration of several kinetic and kinematic parameters and have some specific patterns and inter-individual differences that are not seen at the trot. However, much like at the trot, head movement and forelimb vertical force symmetry seem to be the most useful parameters to detect forelimb lameness at walk.

We aimed to describe their kinematic and temporal adaptation strategies in response to forelimb lameness at walk, trot and tolt.Study designIn vivo experiment.Methods Ten clinically non-lame Icelandic horses were measured before and after reversible forelimb lameness induction. Upper body and limb kinematics were measured using 11 inertial measurement units mounted on the poll, withers, pelvis (tubera sacrale) and all four limbs and hoofs (Equimoves (R), 500 Hz). Horses were measured on a straight line at walk and trot in-hand and at walk, trot and tolt while ridden. Linear mixed models were used to compare baseline and lame conditions (random factor = 'horse'), and results are presented as the difference in estimated marginal means or percentage of change.Results Lameness induction significantly (p < 0.05) increased head vertical movement asymmetry at walk (HDmin/HDmaxHAND: 18.8/5.7 mm, HDmin/HDmaxRIDDEN: 9.8/0.3 mm) and trot (HDmin/HDmaxHAND: 18.1/7.8 mm, HDmin/HDmaxRIDDEN: 24.0/9.3 mm). At the tolt, however, HDmin did not change significantly (1.1 mm), but HDmax increased by 11.2 mm (p < 0.05). Furthermore, pelvis vertical movement asymmetry (PDmax) increased by 4.9 mm, sound side dissociation decreased (-8.3%), and sound diagonal dissociation increased (6.5%). Other temporal stride variables were also affected, such as increased stance duration of both forelimbs at walk, tolt and in-hand trot.Main limitations Only one degree of lameness (mild) was induced with an acute lameness model.Conclusions Classical forelimb lameness metrics, such as vertical head and withers movement asymmetry, were less valuable at tolt compared to walk and trot, except for HDmax. Therefore, it is advised to primarily use the walk and trot to detect and quantify forelimb lameness in the Icelandic horse.

However, when used for the assessment of complex cases of lameness, such as bilateral lameness, discrete variable analysis might overlook relevant functional adaptations. Objectives The overall aim of this paper is to compare continuous and discrete data analysis techniques to evaluate kinematic gait adaptations to lameness. Study design Method comparison. Methods Sixteen healthy Shetland ponies, enrolled in a research programme in which osteochondral defects were created on the medial trochlear ridges of both femurs, were used in this study. Kinematic data were collected at trot on a treadmill before and at 3 and 6 months after surgical intervention. Statistical parametric mapping and linear mixed models were used to compare kinematic variables between and within timepoints. Results Both continuous and discrete data analyses identified changes in pelvis and forelimb kinematics. Discrete data analyses showed significant changes in hindlimb and back kinematics, where such differences were not found to be significant by continuous data analysis. In contrast, continuous data analysis provided additional information on the timing and duration of the differences found. Main limitations A limited number of ponies were included. Conclusions The use of continuous data provides additional information regarding gait adaptations to bilateral lameness that is complementary to the analysis of discrete variables. The main advantage lies in the additional information regarding time dependence and duration of adaptations, which offers the opportunity to identify functional adaptations during all phases of the stride cycle, not just the events related to peak values.

    • Background: Little is known regarding how trotting through curves affects locomotion symmetry in Standardbred trotters.Objectives: To investigate differences in objectively measured Standardbred trotter vertical motion symmetry between straight and non-banked, curved sections of oval trotting tracks during exercise warm-up, using a wireless inertial measurement unit (IMU) system.Study design: Cross-sectional, observational study.Methods: Sixteen horses were included. Mixed models were used to assess associations between symmetry, track segment (straight vs curve) and stride duration.Results: Significant results for forelimb parameters were dependent on interactions between track segments and stride duration. At mean stride duration (0.611 second), during the curved track segment horses showed a lower maximum vertical position of the head after push-off of the outside forelimb (estimate -2.3 mm, P < 0.0001, 95% CI -1.7 to -2.9) and higher minimum vertical position of the head during stance of the outside forelimb (estimate -1.8 mm, P < 0.0001, 95% CI -1.2 to -2.5) compared to straight track, mimicking outside forelimb impact and push-off asymmetry during track curves. For hindlimb parameters, during the curve there was a decreased downward motion of the pelvis during outer hindlimb stance (estimate-0.7 mm, P < 0.0001, 95% CI -0.4 to -1.0), mimicking outside hindlimb impact asymmetry.Main limitations: Horses were evaluated going in one direction only on the track (clockwise).Conclusions: Systematic differences between straight and curved track segments were found but did not fully correspond to previously described findings for horses lunged in circles. Effect sizes were overall small. Data in our study were collected from horses trotting on 1000 m tracks with curve radii of 80-85 m. On non-banked tracks of this size, collecting IMU symmetry data at jogging speeds without distinguishing between straight and curved parts is unlikely to adversely affect clinical decision-making.

Race and jockey race ride population data were used to determine jockey fall and injury incidence rates relative to cause of racehorse death, and were assessed using Poisson regression. Differences in proportions were assessed using Fisher's exact, Pearson's (2), and Cochran-Mantel-Haenszel tests.Results707 racehorses experienced race-related catastrophic injury or sudden death. 199 jockeys had 601 falls with 325 injuries. Jockeys were 162 times more likely to fall (95% confidence interval 137-192; P<0.001) and 171 times more likely to be injured (95% confidence interval 140-208; P<0.001) when they rode a horse that died in a race. We infer that jockeys were more likely to fall or be injured when riding in QH races than in TB races because of a higher incidence of horse fatalities in QH races. Jockey falls occurred with 24% of TB and 36% of QH race-related horse fatalities, and jockey injury occurred in 64% of falls. Jockey falls were most common with TB fetlock injuries and QH carpal, metacarpal and fetlock injuries; and with axial, bilateral and multiple injuries compared with appendicular, unilateral and singular injuries, respectively.ConclusionsPrevention of the most common catastrophic injuries and conditions of the racehorse, e.g. fetlock injuries, may be most effective at decreasing rates of falls and injuries to horseracing jockeys during racing.

Blood samples were taken once a week and in Week 5, before and following an exercise test until 22.30 h and analysed for blood sodium (bNa), total plasma protein (TPP), pAldo, troponin I and packed cell volume (PCV). Blood pressure (BP) was measured and pulse wave recorded at rest with high definition oscillometric-technique (HDO). ECG and echocardiography were recorded. Water intake was measured before and on the day of exercise and voluntary saline intake was measured for 2 days after each period. Faecal samples were taken weekly and analysed for sodium and potassium content.Results:The pAldo and the PCV was higher in NaN compared to NaS. There were no differences between diets in BP, ECG, plasma troponin I and echocardiogram but HDO pulse amplitude tended to be smaller on diet NaN. Water intake was lower on diet NaN and saline intake higher. The response to exercise in bNa, pAldo, PCV and TPP was different on the 2 diets. Faecal potassium/sodium ratio was higher on NaN than on NaS.Conclusion:This study shows that 5 weeks of low sodium intake increased plasma aldosterone concentration and PCV but no alterations in heart function was observed. Faecal potassium/sodium ratio could be used to assess sodium status in horses.

    • Conclusions and potential relevance: Obtained prevalences and heritabilities were in accordance with other studies, supporting the hypothesis that data regularly obtained from equine hospitals may be a valuable source in studies of inherited disorders such as OC and POF. There is a need for more standardised documentation of diagnoses and consistent recording of identity of examined horses using passports or breed databases. Compilation of results from major clinics is desired in order to cover most progenies of stallions used in a region or nation.

    • P>Reasons for performing study:Muscular changes caused by training are breed-specific and studies on the Norwegian-Swedish Coldblooded Trotter (NSCT) are limited. Knowledge about lactate-transporters in muscle in this light draught breed used for harness racing is lacking.Objectives:To identify muscular changes associated with training in young NSCTs and investigate muscular distribution of the monocarboxylate transporter 1 (MCT1) and its ancillary protein CD147, which facilitate lactate transport across membranes.Methods:Nine horses were followed from the start of their training period until the end of their 3-year-old season. A biopsy sample of the middle gluteal muscle was collected on 4 occasions. On the last 3 sampling occasions, individual V(La4)-values (the speed corresponding to a blood lactate concentration of 4 mmol/l) were determined in an incremental exercise test on a high-speed treadmill. One horse was excluded due to lameness. Histochemical and immunohistochemical analyses were performed on all muscle samples to determine fibre types (I, IIA, IIAX, IIX), oxidative capacity (NADH) and the expression of MCT1 and CD147. The activity of selected metabolic enzymes in the muscle before and after training was determined.Results:The percentage of type IIX fibres decreased with training while the percentage of type IIAX fibres increased. The activity of citrate synthase and 3-OH-acyl-CoA-dehydrogenase increased with training. The expression of MCT1 was lower in membranes and cytoplasm of type IIX fibres compared to all other fibre types both before and after training. The antibody against CD147 stained membranes and cytoplasm of all fibres. The first V(La4)-value was lower than the last 2 in all horses.Conclusions:Muscular changes with training of NSCTs were similar to those reported in Standardbreds, indicating fibre type transitions and increased oxidative capacity. Expression of MCT1 differed among fibre types and was related to the oxidative capacity of the fibres.

  • Duration of tetanus immunoglobulin G titres following basic immunisation of horses

    • Reasons for performing studyRecommendations for prophylactic vaccination against tetanus in horses vary greatly between countries and have scarce scientific support in the peer-reviewed literature. In human medicine, recommended booster vaccination intervals are also very variable, but are considerably longer than for horses. More information is needed about the duration of immunity induced by modern vaccines.ObjectivesTo investigate if the duration of antibody titres previously determined to be protective against tetanus differ from what is indicated by recommended vaccination intervals for horses.Study designProspective seroconversion study.MethodsThirty-four horses were enrolled for basic immunisation with an ISCOM Matrix-combination vaccine (Equilis Prequenza Te). Horses received the first vaccination at age 5-11 months, and the second dose 4 weeks later. A third vaccine dose was given 15-17 months after the second dose. Serum tetanus antibody titres were analysed by toxin-binding enzyme-linked immunosorbent assay 2 weeks as well as 14-16 months after the second dose. After the third vaccine dose, titres were checked once yearly for 3 years. Results were described by age and level of antibody titre at first sampling.ResultsTwo weeks after the second dose, all horses (34/34) had antibody levels that exceeded the limit of detection, 0.04 iu/ml. After 16 months the levels were above 0.04 iu/ml in 28/33 horses, the remaining 5 horses potentially had suboptimal protection against tetanus. After the third vaccine dose antibody levels remained above 0.04 iu/ml in 25/26 horses for 1 year, 16/16 horses for 2 years, and 8/8 horses for 3 years.ConclusionsHorses that undergo basic immunisation with 3 doses of vaccine after age 5 months are likely to have serum antibody titres consistent with protection against tetanus for more than 3 years. Current guidelines for tetanus prophylaxis should be revised.

  • Detection of early osteoarthritis in the centrodistal joints of Icelandic horses: Evaluation of radiography and low-field magnetic resonance imaging

    • Reasons for performing studyValidated noninvasive detection methods for early osteoarthritis (OA) are required for OA prevention and early intervention treatment strategies.ObjectivesTo evaluate radiography and low-field magnetic resonance imaging (MRI) for the detection of early stage OA osteochondral lesions in equine centrodistal joints using microscopy as the reference standard.Study designProspective imaging of live horses and imaging and microscopy of cadaver tarsal joints.MethodsCentrodistal (distal intertarsal) joints of 38 Icelandic research horses aged 27-29 months were radiographed. Horses were subjected to euthanasia approximately 2 months later and cadaver joints examined with low-field MRI. Osteochondral joint specimens were classified as negative or positive for OA using light microscopy histology or scanning electron microscopy. Radiographs and MRIs were evaluated for osteochondral lesions and results compared with microscopy.ResultsForty-two joints were classified OA positive with microscopy. Associations were detected between microscopic OA and the radiography lesion categories; mineralisation front defect (P<0.0001), joint margin lesion (P<0.0001), central osteophyte (P = 0.03) and the low-field MRI lesion categories; mineralisation front defect (P = 0.01), joint margin lesion (P = 0.02) and articular cartilage lesion (P = 0.0003). The most frequent lesion category detected in microscopic OA positive joints was the mineralisation front defect in radiographs (28/42 OA positive joints, specificity 97%, sensitivity 67%). No significant differences were detected between the sensitivity and specificity of radiography and low-field MRI pooled lesion categories, but radiography was often superior when individual lesion categories were compared.ConclusionsEarly stage centrodistal joint OA changes may be detected with radiography and low-field MRI. Detection of mineralisation front defects in radiographs may be a useful screening method for detection of early OA in centrodistal joints of young Icelandic horses.

    • Background Osteochondrosis occurs due to failure of the blood supply to growth cartilage. Osteochondrosis lesions have been identified in small tarsal bones and suggested to cause distal tarsal osteoarthritis; however, it has not been determined whether distal tarsal osteochondrosis lesions were the result of vascular failure.Objectives To perform post-mortem arterial perfusion and micro-computed tomography (CT) of the central (CTB) and third tarsal bones (TIII) of fetuses and foals up to 5 months old, to describe tarsal development and any lesions detected.Study design Descriptive, nonconsecutive case series.Methods Twenty-three animals that died or were euthanased from 228 days of gestation to 5 months old were collected, comprising two fetuses and nine foals of miscellaneous breeds and 12 Icelandic Horse foals, a breed with high prevalence of distal tarsal osteoarthritis. One hindlimb from each foal was perfused arterially with barium, and the CTB and TIII were examined with micro-CT.Results Perfusion yielded partial information from 41% of the animals. The CTB and TIII were supplied by nutrient arteries and perichondrial vessels with vertical, transverse and circumferential configurations. Fourteen of the 23 (61%) animals had focal defects in the ossification front, that is, radiological osteochondrosis. The majority of lesions matched the configuration and development of vertical vessels. Additionally, full-thickness, cylindrical defects matched transverse vessels, and the long axes of some dorsal lesions matched circumferential vessels.Main limitations Lack of histological validation.Conclusions Post-mortem perfusion was poor for examination of the blood supply to the growth cartilage of the CTB and TIII. Radiological osteochondrosis lesions were compatible with vascular failure because they were focal, and because lesion geometry matched vessel configuration. The relationship between osteochondrosis and distal tarsal osteoarthritis warrants further investigation.

    • P>Reasons for performing study: Simple objective assessment tools are essential to monitor the clinical efficacy of therapeutic interventions used in equine orthopaedics and rehabilitation. In human medicine, goniometry is a validated tool to quantify restrictions in joint range of motion (ROM); however, the technique is not validated in horses. Objectives: To validate 2 different goniometry techniques for the measurement of passive flexion of the fetlock, carpus and hock by examining; 1) the intra- and inter-tester reliability; 2) the differences between 2 goniometry techniques and 3) differences between standing and anaesthetised horses. Methods: The study is composed of three parts: 1) the intra- and inter-tester reliability was examined on 10 horses, where each horse was assessed by 5 pairs of testers measuring ROM with a universal goniometer; 2) the differences between 2 goniometry techniques were examined on 14 horses, each assessed by 2 investigators (either working in pairs with one investigator holding the limb and the other measuring the joint angle, or working individually at the same time holding and measuring); 3) on 6 horses, the differences between standing and anaesthetised horses were assessed by 2 investigators with the same techniques as described above. Nonparametric tests (Mann-Whitney, Wilcoxon sign-rank) and Intraclass Correlation Coefficient (ICC) were used for statistical analysis (P < 0.05). Results: 1) The intra-tester reliability was high to excellent (ICC 0.8-1) and the inter-tester reliability low to average (ICC 0.1-0.5); 2) significant differences in joint ROM were registered in carpus and hock when measuring in pairs compared to singly and 3) significant differences in joint ROM were registered measuring anaesthetised compared to standing horses. Conclusions: As shown in human studies, goniometry is a promising tool in documenting passive flexion of fetlock, carpus and hock, if used by the same investigator. However, additional studies are needed for further validation.

    • Background Inflammatory bowel disease (IBD) is a well-recognised but poorly understood disease complex in the horse. Clinical signs may vary but often include weight loss, diarrhoea and colic. The effect this disease process may have on the gastrointestinal pacemaker cells (the interstitial cells of Cajal), enteric neurons and glial cells has not been previously evaluated in the horse. Objectives To compare the density of the interstitial cells of Cajal (ICC), enteric neurons and glial cells in horses with IBD to those of normal horses using immunohistochemical markers. Study design Retrospective, quantitative immunohistochemical study. Methods Ileal samples were collected during post-mortem examinations from 14 horses with a clinical and histopathological diagnosis of IBD and from eight normal controls. All horses were Standardbreds 1-15 years of age. Six of the IBD cases had eosinophilic gastroenteritis (EG) while the remaining eight had granulomatous enteritis (GE). Tissue sections were labelled with anti-CD117 (c-Kit), anti-TMEM16 (TMEM16), anti-protein gene product (PGP9.5) and anti-glial fibrillary acidic protein (GFAP) using standard immunohistochemical labelling techniques. Image analysis was performed to quantify the presence of ICC (CD117, TMEM16) as well as neuronal (PGP9.5) and enteroglial (GFAP) networks. Results Interstitial cells of Cajal networks were significantly reduced in the myenteric plexus (MP) region in IBD horses compared with the controls for both markers (P<0.05). There was no significant difference in the density of the neuronal or glial cell markers between the two groups (P>0.05). Main limitations The number of horses included in the study. Conclusions Disruption to ICC networks may contribute to the clinical signs of colic in some horses with IBD. Further studies are needed to establish the pathophysiological mechanisms involved and the functional effects of the reduced ICC networks.
