  • Prevalence of coeliac disease and relations to Helicobacter pylori infection and duodenitis in a Swedish adult population sample : A histomorphological and serological survey

    • Aim: The aim of this study was to determine the prevalence of coeliac disease and its relation to duodenitis, H. pylori infection and gastritis in a sample of the adult general population. Methods: A Swedish population sample of 482 subjects (aged 35 to 85 years) were examined with gastro-duodenoscopy with multiple biopsies taken. Circulating antibodies to endomycium, gliadin, and H. pylori were also determined. Results: Based on histomorphological findings, coeliac disease was evident in 9 of 482 subjects giving a prevalence of 1.9 [1.0-4.0, 95% confidence interval] percent. The prevalence of gastritis with or without H. pylori infection did not differ between subjects with and without coeliac disease. Considering subjects without coeliac disease, there was no difference in the serum levels of gliadin antibodies between those with and without duodenitis. However, subjects with positive H. pylori status had significantly higher levels of gliadin antibodies than those with negative H. pylori status. Conclusions: This study confirms that there is a relatively high prevalence of undiagnosed coeliac disease in Swedish adults. There was no association between coeliac disease and H. pylori infection or gastritis, although serum gliadin antibody levels were slightly increased in subjects with positive H. pylori status.

    • The duodenal epithelium secretes bicarbonate at higher rates than does the stomach (or more distal small intestine) and the duodenal secretion is currently accepted as the most important defence mechanism against acid discharged from the stomach. HCO3 - entering the continuous layer of visco-elastic mucus gel on top of the epithelial surface maintains pH in its cell-facing portion at neutrality at acidities encountered in the healthy duodenum. The secretion is decreased in patients with acute and chronic duodenal ulcer disease and is inhibited by non-steroidal anti-inflammatory agents. Studies of the neurohumoral control of the duodenal alkaline secretion and of acid/base transport processes and intracellular signaling in duodenal enterocytes are currently of great research interest.

  • Anti-inflammatory Properties of Doxycycline and Minocycline in Experimental Models : An in Vivo and in Vitro Comparative Study

    • Aims and methods: Minocycline (Mino) and doxycycline (Dox) are second generation tetracyclines known to present several other effects, which are independent from their antimicrobial activities. We studied in a comparative way the anti-inflammatory effects of Mino and Dox, on acute models of peripheral inflammation in rodents (formalin test and peritonitis in mice, and carrageenan-induced paw oedema in rats). Immunohistochemical assays for TNF-alpha and iNOS in rat paws of carrageenan-induced oedema were also carried out as well as in vitro assays for myeloperoxidase (MPO) and lactate dehydrogenase (LDH). Furthermore, antioxidant activities were evaluated by the DPPH assay. Results: In the formalin test although Mino and Dox (1, 5, 10 and 25 mg/kg, i.p.) inhibited the first phase, they acted predominantly on the second phase of the test, where inhibition of the licking time close to 80% were observed. Mino and Dox were very efficacious in reducing the carrageenan-induced paw oedema in rats (10, 25 and 50 mg/kg, i.p.) and carrageenan-induced leucocyte migration (1 and 5 mg/kg, i.p.) to mice peritoneal cavities. Besides, they also significantly inhibited MPO and LDH releases at doses ranging from 0.001 to 1 μg/ml. Thus, in general, the anti-inflammatory activity of Dox was higher as compared to that of Mino, although the radical scavenging activity of Mino was of a magnitude 10 times higher. Conclusions: Our data indicate that anti-inflammatory and antioxidant effects, involve the inhibition of iNOS and TNF-alpha, among other properties, and these encourage clinical studies of these compounds for new therapeutic applications, especially those were inflammation plays a role.

    • OBJECTIVE: Roquinimex is a drug with effects on inflammation and tumors. The pharmacological effect is not fully understood, and the molecular mechanism most characterized in vitro is an increase of plasminogen activator inhibitor type 2 (PAI-2) in human peripheral blood monocytes. The aims were to investigate peripheral pharmacological effects of roquinimex on peripheral blood monocytes and dog gingival fluid (GCF). DESIGN: Six dogs were used in a cross-over study. The amount of GCF was determined with a Periotron. The PAI-2 concentration in GCF was determined with ELISA. Monocytes were isolated from peripheral blood. RESULTS: Dogs treated with the drug had significantly lower GCF flow values and the PAI-2 concentration in GCF was higher, but no effect was seen on peripheral monocytes. CONCLUSION: Roquinimex treatment led to a consistently decreased flow rate of GCF and a higher local concentration of PAI-2 in GCF.

    • Gastric bypass is a clinical option for obesity surgery. An increased susceptibility to Helicobacter pylori infection in the bypassed stomach has been speculated. The aim of the present study was to examine the susceptibility of the bypassed stomach to H. pylori infection in rats and mice. Adult Sprague-Dawley and Wistar rats and NMRI mice were subjected to either gastric bypass or laparotomy only as control. The animals were inoculated with the CagA- and VacA- positive H. pylori strain 67/21 (not mouse-adapted) in the first experiment and with 9 additional isolates in the second, by injection into the bypassed stomach or the control stomach during surgery. The stomach of each animal was collected for H. pylori culture 2-3 weeks later. While all the rats were H. pylori negative, 54% of gastric bypassed mice and 75% of controls were positive (P = 0.4). We conclude that susceptibility to H. pylori infection in the stomach is not increased by gastric bypass surgery.

    • This paper presents a resistor-less high-precision, sub-1 V all-CMOS voltage reference. A curvature-compensation method is used to cancel the logarithmic temperature dependence regardless of mobility temperature exponent (γ). The circuit is simulated in 65 nm CMOS technology and yields an output voltage of 594 mV, temperature coefficient of 7ppm∘C in the range of −40 to 125 °C, a power supply rejection ratio (PSRR) of − 43 dB at of 100 Hz, a line sensitivity of 76μVV in the supply voltage range of 1.2 to 2 V, a power dissipation of 1.4μW at 1.2 V supply and an output noise of 2.8μVHz at 100Hz. The total active area of the design is 0.03mm2. This voltage reference is suitable for low-power, low-voltage applications which also require high precision.

    • In this paper, a low-complexity resistorless high-precision sub-1 V MOSFET-only voltage reference is presented. To obtain an accurate output, a curvature-compensation technique is used, canceling its logarithmic temperature dependence regardless of the value of the mobility temperature exponent (γ). The circuit is realized in 65 nm CMOS technology and yields an output voltage of 574 mV, a temperature coefficient of 3.5 ppm∘C in the range of − 50 to 150 °C, a power supply rejection ratio (PSRR) of − 103 dB at 100 Hz, a line sensitivity of 6μVV in the supply voltage range of 1.3–3 V, a power dissipation of 650nW at 1.3 V supply, and an output noise of 1.7 μV/Hz at 100 Hz. The total active area of the design is 0.03 mm2. This voltage reference is suitable for low-power low-voltage applications which also require high precision.
