  • The Perfect Match : RIS-enabled MIMO Channel Estimation Using Tensor Decomposition

    • The deployment of reconfigurable intelligent surfaces (RISs) in a communication system provides control over the propagation environment, which facilitates the augmentation of a multitude of communication objectives. As these performance gains are highly dependent on the applied phase shifts at the RIS, accurate channel state information at the transceivers is imperative. However, not only do RISs traditionally lack signal processing capabilities, but their end-to-end channels also consist of multiple components. Hence, conventional channel estimation (CE) algorithms become incompatible with RIS-aided communication systems as they fail to provide the necessary information about the channel components, which are essential for a beneficial RIS configuration. To enable the full potential of RISs, we propose to use tensor-decomposition-based CE, which facilitates smart configuration of the RIS by providing the required channel components. We use canonical polyadic (CP) decomposition, that exploits a structured time domain pilot sequence. Compared to other state-of-the-art decomposition methods, the proposed Semi-Algebraic CP decomposition via Simultaneous Matrix Diagonalization (SECSI) algorithm is more time efficient as it does not require an iterative process. The benefits of SECSI for RIS-aided networks are validated with numerical results, which show the improved individual and end-to-end CE accuracy of SECSI.

  • Perceived accuracy of electronic performance appraisal systems : The case of a non‐for‐profit organization from an emerging economy

    • Performance appraisal reports provide vital information for making strategic decisions. Uniting HR practices and IT is an attempt to maximize value creation. Organizations are transforming their performance management systems from traditional to web‐technology based systems to enhance accuracy and objectivity. The present study is an effort to assess employees’ perception of the accuracy of e‐performance appraisal systems. To that effect, 500 close‐ended questionnaires were administered to randomly selected employees working for a non‐for‐profit organization where e‐performance appraisal is practiced. A total of 352 questionnaires were included for analysis and structural equation modeling was used to analyze the data. The results indicate that rater competence and the possibility to challenge performance appraisals judged as unfair or inaccurate highly influence the employees’ perception of accuracy of e‐performance appraisals. Furthermore, a clear communication of standards and the reaction to the last rating moderately affect the employees’ perception of the accuracy of e‐PA.

  • A Compact CPW-Fed Ultra-Wideband Multi-Input-Multi-Output (MIMO) Antenna for Wireless Communication Networks

    • In this article, a compact coplanar waveguide (CPW) technique based ultra-wideband multiple-input-multiple-output (MIMO) antenna is proposed. The design is characterized by a broad impedance bandwidth starting from 3 GHz to 11 GHz. The overall size of the MIMO design is 60 x 60 mm(2) (1.24 x 1.24 lambda(2)(g) @ 3 GHz) with a thickness of 1.6 mm. To make the design ultra-wideband, the proposed MIMO antenna design has four jug-shaped radiating elements. The design is printed on a FR-4 substrate (relative permittivity of epsilon(r) = 4.4 and loss tangent of tan delta = 0.025). The polarization diversity phenomenon is realized by placing four antenna elements orthogonally. This arrangement increases the isolation among the MIMO antenna elements. The simulated results of the ultra-wideband MIMO antenna are verified by measured results. The proposed MIMO antenna has a measured diversity gain greater than 9.98, envelope correlation coefficient (ECC) less than 0.02, and good MIMO performance where the isolation is more than -20dB between the elements. The group delay, channel capacity loss (CCL), and the total active reflection coefficient (TARC) multiplexing efficiency and mean effective gain results are also analyzed. The group delay is found to be less than 1.2ns, CCL values calculated to be less than 0.4 bits/sec/Hz, while the TARC is below -10dB for the whole operating spectrum. The proposed design is a perfect candidate for ultra-wideband wireless communication systems and portable devices.

  • A Wideband Bear-Shaped Compact Size Implantable Antenna for In-Body Communications

    • Biomedical implantable antennas play a vital role in medical telemetry applications. These types of biomedical implantable devices are very helpful in improving and monitoring patients' living situations on a daily basis. In the present paper, a miniaturized footprint, thin-profile bear-shaped in-body antenna operational at 915 MHz in the industrial, scientific, and medical (ISM) band is proposed. The design is a straightforward bear-shaped truncated patch excited by a 50-Omega coaxial probe. The radiator is made up of two circular slots and one rectangular slot at the feet of the patch, and the ground plane is sotted to achieve a broadsided directional radiation pattern, imprinted on a Duroid RT5880 roger substrate with a typical 0.254-mm thickness (epsilon(r) = 2.2, tan delta = 0.0009). The stated antenna has a complete size of 7 mm x 7 mm x 0.254 mm and, in terms of guided wavelength, of 0.027 lambda(g) x 0.027 lambda(g) x 0.0011 lambda(g). When operating inside skin tissues, the antenna covers a measured bandwidth from 0.86 GHz to 1.08 GHz (220 MHz). The simulations and experimental outcomes of the stated design are in proper contract. The obtained results show that the calculated specific absorption rate (SAR) values inside skin of over 1 g of mass tissue is 8.22 W/kg. The stated SAR values are lower than the limitations of the federal communications commission (FCC). Thus, the proposed miniaturized antenna is an ultimate applicant for in-body communications.

    • Regulatory T cells (Tregs) are central in controlling immune responses, and dysregulation of their function can lead to autoimmune disorders or cancer. Despite extensive studies on Tregs, the basis of epigenetic regulation of human Treg development and function is incompletely understood. Long intergenic noncoding RNAs (lincRNA)s are important for shaping and maintaining the epigenetic landscape in different cell types. In this study, we identified a gene on the chromosome 6p25.3 locus, encoding a lincRNA, that was up-regulated during early differentiation of human Tregs. The lincRNA regulated the expression of interleukin-2 receptor alpha (IL2RA), and we named it the lincRNA regulator of IL2RA (LIRIL2R). Through transcriptomics, epigenomics, and proteomics analysis of LIRIL2R-deficient Tregs, coupled with global profiling of LIRIL2R binding sites using chromatin isolation by RNA purification, followed by sequencing, we identified IL2RA as a target of LIRIL2R. This nuclear lincRNA binds upstream of the IL2RA locus and regulates its epigenetic landscape and transcription. CRISPR-mediated deletion of the LIRIL2R-bound region at the IL2RA locus resulted in reduced IL2RA expression. Notably, LIRIL2R deficiency led to reduced expression of Treg-signature genes (e.g., FOXP3, CTLA4, and PDCD1), upregulation of genes associated with effector T cells (e.g., SATB1 and GATA3), and loss of Treg-mediated suppression.

  • Exploring the potential of ligninolytic armory for lignin valorization : A way forward for sustainable and cleaner production

    • Lignin is a key structural constituent of lignocellulosic biomasses that have substantial untapped potential to substitute environmentally unfriendly and non-renewable fossil-based resources. Unfortunately, multifaceted composition, heterogeneity, and structural recalcitrance of the lignin are the biggest technical challenges for its effective deconstruction and bioconversions to an array of bio-based products, e.g., specialty chemicals and biomaterials. Physicochemical methods for lignin depolymerization require strict reaction conditions, high en-ergy to execute processes, and environmental apprehensions. In contrast, biological platforms harnessing the catalytic potentiality of microorganisms and their robust enzymatic armory are thought to be efficient means for lignin decomposition. Enzymes, derived from natural origin, are highly proficient and eco-friendly biocatalysts that manifest high selectivity, require milder reaction conditions, and reduce resource requirements. The utili-zation of enzymes for lignin conversion and pre-treatment of biomass for detergent, textile, pulp and papers, and food sector applications has been investigated for decades. Herein, we reviewed lignin bioconversion by bio-logical means, focusing on ligninolytic enzyme-assisted pretreatment approaches. In the first half, we outlined the lignin as a multipurpose raw feedstock, fixation of CO2 to lignin biosynthesis and tailored lignin approach, and sources and types of lignin. The bio-based pre-treatment approaches for lignin depolymerization, including white-rot fungi, brown-rot fungi, bacteria, and ligninolytic enzymes, i.e., manganese peroxidase (MnP) lignin peroxidase (LiP), Laccase (Lac), versatile peroxidase (VP), and dye-decolorizing peroxidases (DyP) are thor-oughly vetted in the second half.

  • Epigenome-Wide Histone Acetylation Changes in Peripheral Blood Mononuclear Cells in Patients with Type 2 Diabetes and Atherosclerotic Disease

    • There is emerging evidence of an association between epigenetic modifications, glycemic control and atherosclerosis risk. In this study, we mapped genome-wide epigenetic changes in patients with type 2 diabetes (T2D) and advanced atherosclerotic disease. We performed chromatin immunoprecipitation sequencing (ChIP-seq) using a histone 3 lysine 9 acetylation (H3K9ac) mark in peripheral blood mononuclear cells from patients with atherosclerosis with T2D (n = 8) or without T2D (ND, n = 10). We mapped epigenome changes and identified 23,394 and 13,133 peaks in ND and T2D individuals, respectively. Out of all the peaks, 753 domains near the transcription start site (TSS) were unique to T2D. We found that T2D in atherosclerosis leads to an H3K9ac increase in 118, and loss in 63 genomic regions. Furthermore, we discovered an association between the genomic locations of significant H3K9ac changes with genetic variants identified in previous T2D GWAS. The transcription factor 7-like 2 (TCF7L2) rs7903146, together with several human leukocyte antigen (HLA) variants, were among the domains with the most dramatic changes of H3K9ac enrichments. Pathway analysis revealed multiple activated pathways involved in immunity, including type 1 diabetes. Our results present novel evidence on the interaction between genetics and epigenetics, as well as epigenetic changes related to immunity in patients with T2D and advanced atherosclerotic disease.

    • Current challenges in production automation requires the involvement of new technologies like Internet of Things (IoT), Systems of Systems and local automation clouds. The objective of this paper is to address one of the challenges involved in establishing and managing a cloud based automation system. Three key capabilities have been identified as required to create the expected benefits of local automation clouds; 1) capturing of plant design 2) capturing and distributing configuration and deployment information 3) coordinating information exchange.This paper addresses the capturing and distribution of configuration and deployment information. For this purpose a system service is proposed, the ConfigurationStore, following the principles of the Arrowhead Framework. The service is accompanied by a deployment methodology and a bootstrapping procedure. These are discussed for several types of automation technology, e.g. controllers, sensors, actuators. A qualitative evaluation of the proposed approach is made for four use cases; Building automation, Manufacturing automation, Process automation and IoT devices. Concluding the usability for large-scale deployment and configuration of Industrial Internet of Things.

    • The emergence and deployment of connected devices in many domains of application (e.g. industrial production, buildings and facilities, urban environment, etc.) have resulted in the need to achieve integration of multiple and more complex systems. This new environment is stressing the intrinsic limits imposed by monolithic standards, data models and integration methods that focus on specific domains of application, types of systems, or specific aspects of a system. This paper describes the Plant Description Service developed as part of the Arrowhead Interoperability framework (EU ECSEL funded project). The manuscript contains a description of the abstract system descriptive model based on which the Plant Description service was implemented, and describes how the service can be used to achieve integration of several industry standards and data models. One use case and one case study is provided that illustrates how the service was practically implemented to support engineering scenarios in the domain of industrial production. The paper concludes with a critical review of the approach and suggestion for future work and developments.

    • Because of the flow influx of tributaries, a confluence forms a unique environment carrying interesting hydrodynamic features and other attributes. The understanding of flow behavior here is important, particularly if it is on a tidally influenced channel in a harbor metropolitan. Because of communal requirements, there is a possibility of building wading structures, which may interplay with the flow in this zone. The knowledge of unidirectional river or flume confluences so far is not readily applicable for similar features in channels near coastal areas that have tidal flow in addition to river runoff. In this study, a tidal confluence that has a highly dynamic bidirectional flow is investigated. Near-surface flow patterns in a tidal cycle are simulated by using a numerical model. A field survey provides the bathymetry, time-series boundary conditions, and corresponding verification data. Good agreement is reached between calculated and measured results. Based on the condition of tidal current, four scenarios are selected for which confluence flow patterns are observed, both spatially and temporally. The results indicate that at least one recirculation is always in the tidal confluence for all flow conditions, which rotates counterclockwise for the ebb flow and clockwise for the flood flow. In addition, there is no absolute slack water condition at the tidal junction in the study area. The study also finds that the flows of all three connected channels at the confluence change in a looped pattern with respect to one another. Furthermore, the study reports unique relationships among the ratios of different flows.

    • The incidence of obesity in the developed world is increasing at an alarming rate. Concurrent with the increase in the incidence of obesity is an increase in the incidence of type 2 diabetes. Cyclic AMP (cAMP) and cGMP are key second messengers in all cells; for example, when it comes to processes of relevance for the regulation of energy metabolism, cAMP is a key mediator in the regulation of lipolysis, glycogenolysis, gluconeogenesis and pancreatic β cell insulin secretion. PDE3B, one of several enzymes which hydrolyze cAMP and cGMP, is expressed in cells of importance for the regulation of energy homeostasis, including adipocytes, hepatocytes, hypothalamic cells and β cells. It has been shown, using PDE3 inhibitors and gene targeting approaches in cells and animals, that altered levels of PDE3B result in a number of changes in the regulation of glucose and lipid metabolism and in overall energy homeostasis. This article highlights the complexity involved in the regulation of PDE3B by hormones, and in the regulation of downstream metabolic effects by PDE3B in several interacting tissues.

    • AIM: Tenascin C (TNC) is a large extracellular matrix glycoprotein. It is involved in development and upregulated both during tissue repair and in several pathological conditions, including cardiovascular disease. Extracellular matrix proteins play a role in promoting exercise responses, leading to adaptation, regeneration, and repair. The main goal of this study was to investigate whether a short anaerobic effort leads to increased levels of TNC in serum.METHODS: Thirty-nine healthy men performed a Wingate test followed by a muscle biopsy. Myoblasts were isolated from the muscle biopsies and differentiated to myotubes ex vivo. TNC RNA was quantified in the biopsies, myotubes and myoblasts using RNA sequencing. Blood samples were drawn before and 5 min after the Wingate test. Serum TNC levels were measured using ELISA.RESULTS: After the Wingate test, serum TNC increased on average by 23% [15-33], median [IQR]; P Wilcoxon < 0.0001. This increase is correlated with peak power output and power drop, but not with VO 2max . TNC RNA expression is higher in myoblasts and myotubes compared to skeletal muscle tissue. CONCLUSION: TNC is secreted systemically as a response to the Wingate anaerobic test in healthy males. The response was positively correlated with peak power and power drop, but not with VO 2max which implicates a relation to mechanical strain and/or blood flow. With higher expression in undifferentiated myoblast cells than muscle tissue, it is likely that TNC plays a role in muscle tissue remodelling in humans. Our findings open for research on how TNC contributes to exercise adaptation. This article is protected by copyright. All rights reserved.

    • In this study, Fe-Mn co-doped ceria Fe0.25Mn0.00Ce0.75O2-delta (FMDC1), Fe0.23Mn0.02Ce0.75O2-δ (FMDC2), Fe0.21Mn0.04Ce0.75O2-δ  (FMDC3), and Fe0.19Mn0.06Ce0.75O2-δ  (FMDC4) powders are synthesized by sol gel method and evaluated as cathode materials for intermediate temperature solid oxide fuel cell (IT-SOFC). The combination of Fe and Mn significantly enhanced the ceria catalytic activity and oxygen kinetics for redox-based reactions. The effects of the co-doping mechanism on the phase composition, optical behavior, and electrochemical performance are mainly investigated. The prepared samples are characterized by XRD, SEM, FTIR, UV-vis, and conductivity tests. X-ray diffraction analysis revealed well-developed crystallinity with a single phase cubic structure of synthesized cathode material. SEM depicted the highly porous facet for Fe0.19Mn0.06Ce0.75O2-δ  (FMDC4) resulted in the large triple-phase boundaries for the reduction of ambient air. The inclusion of Mn3+ and Fe3+ ions into CeO2 network created additional oxygen vacancies (Ov) and simultaneously reduced the optical band gap energy from 2.81 eV for FMDC1 (x = 0.00) to 2.54 eV for FMDC4 (x = 0.06). Among the four samples, FMDC4 possessed the highest electrical conductivity (∼0.89 Scm-1) at 650 degrees C and corresponding low activation energy of similar to 0.301 eV, which lead to good catalytic activity with an enhanced electrochemical performance of the SOFC system. The open-circuit voltage (OCV) attained the value of   ∼0.98 V, maximum power density of ∼335 mW cm-2 is obtained at 550 degrees C, which is comparable to previously reported electrodes. The results suggested that the combination of Fe and Mn into ceria can be used as an effective catalytic promoter for oxygen reduction reactions (ORR), and the composition of FMDC4 resulted in the peak conductivity, short term stability and highest power density as compared to other synthesized samples.   • Challenges with Software Verification and Validation Activities in the Space Industry

    • Developing software for high-dependable space applications and systems is a formidable task. With new political and market pressures on the space industry to deliver more software at a lower cost, optimization of their methods and standards need to be investigated. The industry has to follow standards that strictly set quality goals and prescribes engineering processes and methods to fulfill them. The overall goal of this study is to evaluate if current use of the standards from the European Cooperation for Space Standardization (ECSS) is cost efficient and if there are ways to make the process leaner while still maintaining quality and to analyze if their verification and validation (V&V) activities can be optimized. This paper presents results from two industrial case studies of companies in the European space industry that are following ECSS standards in various V&V activities. The case studies reported here focus on how ECSS standards are used by the companies, how that affects their processes and, in the end, how their V&V activities can be further optimized.

This paper describes the results and recommendations based on identified challenges.

    • Background The UN's Sustainable Development Goals (SDGs) are grounded in the global ambition of "leaving no one behind". Understanding today's gains and gaps for the health-related SDGs is essential for decision makers as they aim to improve the health of populations. As part of the Global Burden of Diseases, Injuries, and Risk Factors Study 2016 (GBD 2016), we measured 37 of the 50 health-related SDG indicators over the period 1990-2016 for 188 countries, and then on the basis of these past trends, we projected indicators to 2030. Methods We used standardised GBD 2016 methods to measure 37 health-related indicators from 1990 to 2016, an increase of four indicators since GBD 2015. We substantially revised the universal health coverage (UHC) measure, which focuses on coverage of essential health services, to also represent personal health-care access and quality for several non-communicable diseases. We transformed each indicator on a scale of 0-100, with 0 as the 2.5th percentile estimated between 1990 and 2030, and 100 as the 97.5th percentile during that time. An index representing all 37 health-related SDG indicators was constructed by taking the geometric mean of scaled indicators by target. On the basis of past trends, we produced projections of indicator values, using a weighted average of the indicator and country-specific annualised rates of change from 1990 to 2016 with weights for each annual rate of change based on out-of-sample validity. 24 of the currently measured health-related SDG indicators have defined SDG targets, against which we assessed attainment. Findings Globally, the median health-related SDG index was 56.7 (IQR 31.9-66.8) in 2016 and country-level performance markedly varied, with Singapore (86.8, 95% uncertainty interval 84.6-88.9), Iceland (86.0, 84.1-87.6), and Sweden (85.6, 81.8-87.8) having the highest levels in 2016 and Afghanistan (10.9, 9.6-11.9), the Central African Republic (11.0, 8.8-13.8), and Somalia (11.3, 9.5-13.1) recording the lowest. Between 2000 and 2016, notable improvements in the UHC index were achieved by several countries, including Cambodia, Rwanda, Equatorial Guinea, Laos, Turkey, and China; however, a number of countries, such as Lesotho and the Central African Republic, but also high-income countries, such as the USA, showed minimal gains. Based on projections of past trends, the median number of SDG targets attained in 2030 was five (IQR 2-8) of the 24 defined targets currently measured. Globally, projected target attainment considerably varied by SDG indicator, ranging from more than 60% of countries projected to reach targets for under-5 mortality, neonatal mortality, maternal mortality ratio, and malaria, to less than 5% of countries projected to achieve targets linked to 11 indicator targets, including those for childhood overweight, tuberculosis, and road injury mortality. For several of the health-related SDGs, meeting defined targets hinges upon substantially faster progress than what most countries have achieved in the past. Interpretation GBD 2016 provides an updated and expanded evidence base on where the world currently stands in terms of the health-related SDGs. Our improved measure of UHC offers a basis to monitor the expansion of health services necessary to meet the SDGs. Based on past rates of progress, many places are facing challenges in meeting defined health-related SDG targets, particularly among countries that are the worst off. In view of the early stages of SDG implementation, however, opportunity remains to take actions to accelerate progress, as shown by the catalytic effects of adopting the Millennium Development Goals after 2000. With the SDGs' broader, bolder development agenda, multisectoral commitments and investments are vital to make the health-related SDGs within reach of all populations. Copyright The Authors.     • Pickering emulsions are eco-friendly, stabilized by solid particles, and have an essential role in leading industries. Although Pickering emulations have found several applications, surprisingly few investigations have attempted to explore the effectiveness of various mechanical processes for its production. To fill these gaps, the present investigation comprehensively examined the application of various Pickering emulsion preparation processes such as rotor-stator homogenization emulsification (R-SH), ultrasonic emulsification, and their combined processes by using nano-silica particles. The influences of emulsification time and intensity on emulsion droplets' distribution were analyzed as indicative factors. The kerosene/water nano-silica Pickering emulsion was utilized for all assessments. The obtained results demonstrated that the main distribution peak of the emulsion prepared by R-SH occurred where the chord length was greater than 40 μm. Micro-scale nano-silica-aggregates generated large droplets, while the fine-emulsion fraction was significantly increased after ultrasonic treatment. The experimental results showed that the emulsion prepared only by ultrasound needed substantial power to form a Pickering emulsion since the oil phase was difficult to disperse in the water phase. Finally, it was concluded that preprocessing by R-SH could form a stable and uniform emulsion speedily, which is essential for ultrasound emulsion preparation.

  • Generic Requirements for Inclusive Healthcare Software: Supporting Older Adults and People with Intellectual and Developmental Disability

    • As healthcare technology advances, it is important to ensure that everyone benefits from healthcare software, regardless of age or ability. That is why our research team has been working to identify and encapsulate the unique needs of two groups when developing software systems: older adults and people with mild intellectual and developmental disabilities. Through our research, we have developed design patterns that software engineers can use to create more usable healthcare software to support these groups. These patterns identify the generic requirements for usability including accessibility, crucial for creating software that meets the needs of marginalised populations. In this paper, we describe the steps we undertook to identify and develop these requirements. We discover many overlapping needs of these two cohorts. Our research aims to benefit older adults and people with mild intellectual and developmental disabilities and improve the lives of many others facing similar challenges. As we expand our research to other cohorts, we will be able to identify additional generic requirements that make healthcare software development more inclusive and effective.

    • Arsenic (As) contamination is a well-recognized environmental and health issue, threatening over 200 million people worldwide with the prime cases in South and Southeast Asian and Latin American countries. Rice is mostly cultivated under flooded paddy soil conditions, where As speciation and accumulation by rice plants is controlled by various geo-environmental (biotic and abiotic) factors. In contrast to other food crops, As uptake in rice has been found to be substantially higher due to the prevalence of highly mobile and toxic As species, arsenile (As(Ill)), under paddy soil conditions. In this review, we discussed the biogeochemical cycling of As in paddy soil-rice system, described the influence of critical factors such as pH, iron oxides, organic matter, microbial species, and pathways affecting As transformation and accumulation by rice. Moreover, we elucidated As interaction with organic and inorganic amendments and mineral nutrients. The review also elaborates on As (im) mobilization processes and As uptake by rice under the influence of different mineral nutrients and amendments in paddy soil conditions, as well as their role in mitigating As transfer to rice grain. This review article provides critical information on As contamination in paddy soil-rice system, which is important to develop suitable strategies and mitigation programs for limiting As exposure via rice crop, and meet the UN's key Sustainable Development Goals (SDGs: 2 (zero hunger), 3 (good health and well-being), 12 (responsible consumption and production), and 13 (climate action)).

Therefore, there is a need for themigration of such systems to scalable and modular plug and playplatforms supported by effective engineering tools.This paper outlines a distributed modular service-basedapproach to the implementation of a real-time residentialautomation system application composed of heating, homeautomation and vehicle parking systems. The main focus of thispaper is on the design and simulation of this System of Systems(SoS) focusing on the home parking, heating and vehiclemonitoring application components and their interactions. Theuse of a component-based engineering approach is reported onproviding 3D modelling and simulation capabilities prior to thephysical implementation of the system, and from which theorchestration logic can be automatically generated. A potentialtoolkit for SOA-based application design and deployment issuggested.

    • Requirements validation ensures that the defined requirements conform to the description of the system and are consistent and complete. It is a complex activity that becomes more complicated for global software development (GSD). This study presents the issues associated with requirements validation in GSD and proposes a solution (RVGSD) to overcome these issues. We conducted a literature review and developed a prototype tool to implement RVGSD. The evaluation results show that RVGSD is easy to adopt in the industry and can potentially overcome various issues in the GSD context. Practitioners were satisfied with our framework in all aspects, including validation of requirements, fixing GSD issues, and overall performance.

    • Previously, microRNA‐100 (miR‐100) and its putative mRNA target, insulin‐like growth factor receptor‐1 (IGF1R) were identified as differentially and inversely expressed in bovine longissimus dorsi (LD) muscles with divergent intramuscular fat (IMF) content by our group. While IGF1R signaling is implicated in myogenesis and muscle lipid metabolism, the underlying regulatory mechanisms are poorly understood. In the present study, we aimed to investigate the regulation of IGF1R by miR‐100 during bovine muscle satellite cell (BMSC) myogenesis and lipid deposition. MiR‐100 was confirmed to target the IGF1R 3′‐untranslated region (3′‐UTR) by luciferase reporter assay. Furthermore, expression of miR‐100 and IGF1R was reciprocal during BMSC differentiation, suggesting a crosstalk between the two. Correspondingly, miR‐100 mimic (agomiR) suppressed the levels of IGF1R, PI3K/AKT pathway signaling, myogenic gene MYOG, muscle structural components MYH7 and MYH8, whereas the inhibitor (antagomiR) had no clear stimulating effects. The IGF1R inhibitor (BMS‐754807) curtailed receptor levels and triggered atrophy in muscle myotubes but did not influence miR‐100 expression. AgomiR increased oleic acid‐induced lipid deposition in BMSC myotubes supporting its involvement in intramuscular fat deposition, while antagomiR had no effect. Moreover, mitochondrial beta‐oxidation and long‐chain fatty acid synthesis‐related genes were modulated by agomiR addition. Our results demonstrate modulatory roles of miR‐100 in BMSC development, lipid deposition, and metabolism and suggest a role of miR‐100 in marbling characteristics of meat animals and fat oxidation in muscle.

    • Background: Skeletal muscle fiber type distribution has implications for human health, muscle function, and performance. This knowledge has been gathered using labor-intensive and costly methodology that limited these studies. Here, we present a method based on muscle tissue RNA sequencing data (totRNAseq) to estimate the distribution of skeletal muscle fiber types from frozen human samples, allowing for a larger number of individuals to be tested. Methods: By using single-nuclei RNA sequencing (snRNAseq) data as a reference, cluster expression signatures were produced by averaging gene expression of cluster gene markers and then applying these to totRNAseq data and inferring muscle fiber nuclei type via linear matrix decomposition. This estimate was then compared with fiber type distribution measured by ATPase staining or myosin heavy chain protein isoform distribution of 62 muscle samples in two independent cohorts (n = 39 and 22). Results: The correlation between the sequencing-based method and the other two were rATPas = 0.44 [0.13–0.67], [95% CI], and rmyosin = 0.83 [0.61–0.93], with p = 5.70 × 10–3 and 2.00 × 10–6, respectively. The deconvolution inference of fiber type composition was accurate even for very low totRNAseq sequencing depths, i.e., down to an average of ~ 10,000 paired-end reads. Conclusions: This new method (https://github.com/OlaHanssonLab/PredictFiberType) consequently allows for measurement of fiber type distribution of a larger number of samples using totRNAseq in a cost and labor-efficient way. It is now feasible to study the association between fiber type distribution and e.g. health outcomes in large well-powered studies.

Here, we aim to understand the metabolic pathways involved in the activation and functional differentiation of human CD4+ T‐cell subsets (Th1, Th2, Th17 and iTregs). We combined genome‐scale metabolic modeling, gene expression data, targeted and non‐targeted lipidomics experiments, together with in vitro gene knockdown experiments and showed that human CD4+ T cells undergo specific metabolic changes during activation and functional differentiation. In addition, we identified and confirmed the importance of ceramide and glycosphingolipid biosynthesis pathways in Th17 differentiation and effector functions. Through in vitro gene knockdown experiments, we substantiated the requirement of serine palmitoyl transferase, a de novo sphingolipid pathway in the expression of proinflammatory cytokine (IL17A and IL17F) by Th17 cells. Our findings may provide a comprehensive resource for identifying CD4+ T‐cell‐specific targets for their selective manipulation under disease conditions, particularly, diseases characterized by an imbalance of Th17/nTreg cells.

    • Structured overlay networks, like any distributed system, use replication to avoid losing data in the presence of failures. In this paper, we discuss the short-comings of existing replication schemes and propose a technique for replication, called ID-Replication. ID-Replication allows different replication degrees for keys in the system, thus allowing popular data to have more copies. We discuss how ID-Replication is less sensitive to churn compared to existing replication schemes, which makes ID-Replication better suited for building consistent services on top of overlays compared to other schemes. Furthermore, we show why ID-Replication is simpler to load-balance and more secure compared to successor-list replication. We evaluate our scheme in detail, and compare it with successor-list replication.

    • Bubble-particle attachment is a key factor in various material processing such as wastewater treatment and flotation separation. Nanobubble’s formation and its stability on hydrophobic surfaces with and without surfactants have been scientifically proven and extensively studied in various investigations. However, the influence of particle roughness on the hydrophilic particle-air bubble attachment, which could be completely different from hydrophobic particle-bubble attachment in the presence of nanobubbles, has not been addressed. For tackling this gap, the present work investigated the impact of nanobubbles on the roughed surfaces of glass bead particles. The temperature rise technique as a known method was used for micro/nanobubble size generation. The glass beads were modified by the commonly applied abrasion method to create different roughness magnitudes. The particle-bubble assessment results indicated that the particle roughness could potentially affect the bubble attachment of hydrophilic glass beads while the attachment area of smooth particles was almost zero. Outcomes also were revealed that the modified attachment rate constant increased from 0.1180 to 2.2802 s-1 with the increase of particle surface roughness, indicating a shortening of attachment performance by enhancing the particle surface roughness. However, it was observed that the temperature rise method could improve the particle-bubble attachment only to a marginal extent.
