    • Tick-borne encephalitis virus (TBEV) is an important European vaccine-preventable pathogen. Discrimination of vaccine-induced antibodies from those elicited by infection is important. We studied anti-TBEV IgM/IgG responses, including avidity and neutralisation, by multiplex serology in 50 TBEV patients and 50 TBEV vaccinees. Infection induced antibodies reactive to both whole virus (WV) and non-structural protein 1 (NS1) in 48 clinical cases, whereas 47 TBEV vaccinees had WV, but not NS1 antibodies, enabling efficient discrimination of infection/vaccination.

  • A Palliative Approach to Dementia Care : Leadership and organisation, existential issues and family support

    • The main purpose of this thesis was to apply the WHO and NHS palliative care approach to dementia care. Thirty-one staff-members in mid-Sweden (studies I and II) and 20 next-of- kin (study IV) were interviewed. In study III, 316 staff-members from dementia care and 121 staff-members from palliative cancer care responded to a questionnaire about family support. The interviews were tape-recorded and analysed with a qualitative phenomenographic (I and II) and a hermeneutic approach (IV). The questionnaires (III) were analysed using qualitative and quantitative content analysis.The staff-members stated almost unanimously that daily leadership was lacking, and consequently clear goal formulations and care planning were rare (I). Proper teamwork between the doctor and the staff who worked on a daily basis with the patients was absent (I). With respect to existential issues, education and staff discussions were lacking (II). The staff were at a loss concerning how to deal with these issues. Nevertheless, these issues are central to family-members who have to deal with an existential crisis (IV). Important questions emerged about obligation and guilt, faithfulness, responsibility, and paying back what you once received. Existential isolation could be identified e.g. in the reversal of roles experienced as "being a parent to your parent" and in the burden of "visiting a living dead person". There were no routines for bereavement visits. The type of support suggested for dementia family members is partly similar to support in palliative cancer care, but it also differs in other respects such as feelings of guilt because the early signs of the disease are misunderstood, the need for respite because of the long trajectory of dementia diseases, and the occurrence of anticipatory grief because in the late phase family members can no longer make any contact at all with the patient (III).A palliative approach can improve the quality of life for the dementia patient and for the family. It can be used as a basis for a clear goal formulation. Some of the suggestions listed in this thesis for improving the quality of care are more a reflection of the need for a change in attitudes rather than the need for substantial budget increases.

  • Citizens´ services : Nordic and Baltic research needs

    • What are the key issues in citizen-centric service development that research should address? People engaged in the development of services for citizens, both public and private, in six countries – Iceland, Sweden, Lithuania, Norway, Estonia and Latvia – have participated in half-day workshops during 2008-2009 to discuss the challenges research projects should take on, which output is needed and how to conduct them in order to better help service developers. This work has been a key part of the project “Citizens’ services – Turning Public Private outside in”. In general, the situation in the six countries is very similar and key findings include: • The need to improve cross- and inter-organizational service development. There are too many “silos”. • The need to better engage citizens in service design, to increase the use and effectiveness of services. • The need for more collaborative research projects with researchers, and public and private service developers in an action-oriented and experimental mode.

  • Creating and sustaining successful knowledge management in purposeful communities : summary of key experiences from pioneers

    • Based on research organized as a number of workshops, case studies and interviews with experienced practitioners as well as academics, we present in this report the most important findings on how to create and sustain successful knowledge management in a community environment. The cases, workshops and interviews deal specifically with the Microsoft Solutions Sharing Network (SSN) program, but the findings, conclusions and preliminary recommendations can be applied more generally to the development of any knowledge management community. A key conclusion is that the bulk of efforts toward creating successful knowledge management communities focus oil less technical, or softer aspects like leadership, culture, social settings and value of participation. However, these are essential, but not sufficient, ingredients for success. Technical issues, issues regarding development and customization of the tools used to facilitate knowledge management (for example, the SSN web portal), and emerging legal issues surrounding the sharing of intellectual property UP) may be perceived as somewhat less important to the participants, but are nevertheless key factors in the long term success of these communities. It is also concluded that the foundation for successful collaboration is primarily laid in the initial phases of community development. A community must make a positive impression oil its participants from the very beginning because most people will not give it a second chance. In this report we have highlighted three important areas to consider when establishing portals for knowledge management: Leadership, Purpose and Process/Infrastructure. A leadership with high credibility in the subject is needed to lead the participants in the right direction, manage the cultural processes and to make sure that relevant content can be found. Initially it is the content that brings people to a specific community. Thus, there has to be some common purpose that not only needs to be in congruence with the professional role of the participants but also be inspiring for them as well. Additionally, the community should have some sort of process that the participants can understand and suits the way they would like to interact. Face-to-face meetings and networking activities create trust which is important to get the process started. Language, IT platform, support and rules governing the contribution, creation and sharing of 'knowledge' for the community are other concerns that need to be considered within the process.   • Turning the Internet Around : e-Me: The Students Ideal e-Service

    • Today students, as many other groups of citizens, are offered, indeed required to use, a rapidly increasing number of e-Services. They range from school and course sites to interactions with authorities as well as companies offering student discounts. This paper reports on a pioneering project in Sweden with a radical approach to this, namely to issue the student with a electronic assistant, an e-Me, that schools, authorities and companies are required to address when interacting with the student. A larger number of students and partners, universities, companies and authorities, have been engaged in the design of such an e-Me. It might be thought of as turning the internet around – rather than having students keep track of sites, they will have to come to the students and interact with them in the way specified by them.

    • Overloading of the emergency departments in hospitals is, in Sweden, a common problem that is often blamed on lack of access to primary care.  We have conducted a cross-sectional study comprising more than 40% of the 347 837  inhabitants of Region Jönköping with access to complete individual data on healthcare consumption, personal doctor continuity, socio-economics, and accessibility data for all of the regions health centres. Individuals with high personal continuity at their own health centre had significantly fewer emergency room visits compared to those with the lowest continuity: for younger adults 55% and for elderly 34% fewer emergency room visits. Access to doctor consultations or to counselling nurses in primary care was not associated with a lower number of emergency room visits. Our results show the importance of personal doctor continuity also for the group of younger adults.

    • This report describes the background, and related theories, results and reflections based on a research - innovation (R&D model comprising multi-stakeholder co-creative workshops. The resulting new concepts, solutions and prototypes produced in a short limited time-scale are extraordinary compared to the outcome of standard research and innovation approaches. The developed prototypes and work-processes and the performed R&I model have in a positive way changed attitudes among many key stakeholders, leading to the start of new R&D programs pushing development of citizen centric services towards building a foundation for positive living areas.

    • This study presents the synthesis and characterization of two fluorescent norbornadiene (NBD) photoswitches, each incorporating two conjugated pyrene units. Expanding on the limited repertoire of reported photoswitchable fluorescent NBDs, we explore their properties with a focus on applications in bioimaging of amyloid beta (Aβ) plaques. While the fluorescence emission of the NBD decreases upon photoisomerization, aligning with what has been previously reported, for the first time we observed luminescence after irradiation of the quadricyclane (QC) isomer. We deduce how the observed emission is induced by photoisomerization to the excited state of the parent isomer (NBD) which is then the emitting species. Thorough characterizations including NMR, UV-Vis, fluorescence, X-ray structural analysis and density functional theory (DFT) calculations provide a comprehensive understanding of these systems. Notably, one NBD-QC system exhibits exceptional durability. Additionally, these molecules serve as effective fluorescent stains targeting Aβ plaques in situ, with observed NBD/QC switching within the plaques. Molecular docking simulations explore NBD interactions with amyloid, unveiling novel binding modes. These insights mark a crucial advancement in the comprehension and design of future photochromic NBDs for bioimaging applications and beyond, emphasizing their potential in studying and addressing protein aggregates.

    • The automotive industry is continuously developing and finding new ways to respond to the incremental demands of higher safety standards and lower environmental impact. As an answer to weight reduction of vehicles, the combination of boron steel and composite material is being developed along with their joining process, self-pierce riveting. Boron steel is an ultra-high strength material that needs to be locally softened before the joining process. However, the joining process deforms the part. This paper investigates factors affecting the geometrical deformation during the tempering process and lists important phenomena that need to be included when simulating the tempering process.

    • Adipose tissue plays a major role in regulating whole-body insulin sensitivity and energy metabolism. To accommodate surplus energy, the tissue rapidly expands by increasing adipose cell size (hypertrophy) and cell number (hyperplasia). Previous studies have shown that enlarged, hypertrophic adipocytes are less responsive to insulin, and that adipocyte size could serve as a predictor for the development of type 2 diabetes. In the present study, we demonstrate that changes in adipocyte size correlate with a drastic remodeling of the actin cytoskeleton. Expansion of primary adipocytes following 2 weeks of high-fat diet (HFD)-feeding in C57BL6/J mice was associated with a drastic increase in filamentous (F)-actin as assessed by fluorescence microscopy, increased Rho-kinase activity, and changed expression of actin-regulating proteins, favoring actin polymerization. At the same time, increased cell size was associated with impaired insulin response, while the interaction between the cytoskeletal scaffolding protein IQGAP1 and insulin receptor substrate (IRS)-1 remained intact. Reversed feeding from HFD to chow restored cell size, insulin response, expression of actin-regulatory proteins and decreased the amount of F-actin filaments. Together, we report a drastic cytoskeletal remodeling during adipocyte expansion, a process which could contribute to deteriorating adipocyte function.

    • The ongoing quest to develop single-particle methods for the in situ study of heterogeneous catalysts is driven by the fact that heterogeneity in terms of size, shape, grain structure, and composition is a general feature among nanoparticles in an ensemble. This heterogeneity hampers the generation of a deeper understanding for how these parameters affect catalytic properties. Here we present a solution that in a single benchtop experimental setup combines single-particle plasmonic nanospectroscopy with mass spectrometry for gas phase catalysis under reaction conditions at high temperature. We measure changes in the surface state of polycrystalline platinum model catalyst particles in the 70 nm size range and the corresponding bistable kinetics during the carbon monoxide oxidation reaction via the peak shift of the dark-field scattering spectrum of a closely adjacent plasmonic nanoantenna sensor and compare these changes with the total reaction rate measured by the mass spectrometer from an ensemble of nominally identical particles. We find that the reaction kinetics of simultaneously measured individual Pt model catalysts are dictated by the grain structure and that the superposition of the individual nanoparticle response can account for the significant broadening observed in the corresponding nanoparticle ensemble data. In a wider perspective our work enables in situ plasmonic nanospectroscopy in controlled gas environments at high temperature to investigate the role of the surface state on transition metal catalysts during reaction and of processes such as alloying or surface segregation in situ at the single-nanoparticle level for model catalysts in the few tens to hundreds of nanometer size range.

    • Background and aims Chronic kidney disease leads to uremia and markedly accelerates atherosclerosis. Phenotypic modulation of smooth muscle cells (SMCs) in the arterial media plays a key role in accelerating atherogenesis. The aim of this study was to investigate whether uremia per se modulates the phenotype of aortic SMCs in vivo. Methods Moderate uremia was induced by 5/6 nephrectomy in apolipoprotein E knockout (ApoE-/-) and wildtype C57Bl/6 mice. Plasma analysis, gene expression, histology, and myography were used to determine uremia-mediated changes in the arterial wall. Results Induction of moderate uremia in ApoE-/- mice increased atherosclerosis in the aortic arch en face 1.6 fold (p = 0.04) and induced systemic inflammation. Based on histological analyses of aortic root sections, uremia increased the medial area, while there was no difference in the content of elastic fibers or collagen in the aortic media. In the aortic arch, mRNA and miRNA expression patterns were consistent with a uremia-mediated phenotypic modulation of SMCs; e.g. downregulation of myocardin, α-smooth muscle actin, and transgelin; and upregulation of miR146a. Notably, these expression patterns were observed after acute (2 weeks) and chronic (19 and 30 weeks) uremia, both under normo- and hypercholesterolemic settings. Functionally, aortic constriction was decreased in uremic as compared to non-uremic aorta segments, as measured by myography. Conclusions Uremia modulates the phenotype of aortic SMCs as determined by mRNA/miRNA expression, an increased medial area, and decreased aortic contractility. We propose that this phenotypic modulation of SMCs precedes the acceleration of atherosclerosis observed in uremic mice.

  • Vascular smooth muscle–specific YAP/TAZ deletion triggers aneurysm development in mouse aorta

    • Inadequate adaption to mechanical forces, including blood pressure, contributes to development of arterial aneurysms. Recent studies have pointed to a mechanoprotective role of YAP and TAZ in vascular smooth muscle cells (SMCs). Here, we identified reduced expression of YAP1 in human aortic aneurysms. Vascular SMC–specific knockouts (KOs) of YAP/TAZ were thus generated using the integrin α8–Cre (Itga8-Cre) mouse model (i8-YT-KO). i8-YT-KO mice spontaneously developed aneurysms in the abdominal aorta within 2 weeks of KO induction and in smaller arteries at later times. The vascular specificity of Itga8-Cre circumvented gastrointestinal effects. Aortic aneurysms were characterized by elastin disarray, SMC apoptosis, and accumulation of proteoglycans and immune cell populations. RNA sequencing, proteomics, and myography demonstrated decreased contractile differentiation of SMCs and impaired vascular contractility. This associated with partial loss of myocardin expression, reduced blood pressure, and edema. Mediators in the inflammatory cGAS/STING pathway were increased. A sizeable increase in SOX9, along with several direct target genes, including aggrecan (Acan), contributed to proteoglycan accumulation. This was the earliest detectable change, occurring 3 days after KO induction and before the proinflammatory transition. In conclusion, Itga8-Cre deletion of YAP and TAZ represents a rapid and spontaneous aneurysm model that recapitulates features of human abdominal aortic aneurysms.

  • Characterization of Self-Assembled Monolayers of Oligo(phenyleneethynylene) Derivatives of Varying Shapes on Gold : Effect of Laterally Extended ∏-Systems

    • Fully conjugated organic molecules, such as the oligo(phenyleneethynylene) (OPE) systems, are of growing interest within the field of molecular electronics, as is the self-assembly of well-defined molecular thin films with predefined functions. The structure and function of such films are intimately related and governed by the structures of their molecular constituents, through the intermolecular interactions and the interactions between the molecules and the substrate, onto which the film is assembled. Here we report on the synthesis of a series of three OPE derivatives, with the general structure phenylethynylene−aryl−ethynylenephenylene−headgroup, and the structural investigation of the self-assembled monolayers (SAMs) formed from them on Au(111) surfaces. The SAMs were characterized by infrared reflection−absorption spectroscopy, spectroscopic ellipsometry, high-resolution X-ray photoemission spectroscopy, and near-edge X-ray absorption fine structure spectroscopy. The effective thickness of the SAMs was observed to decrease as the π-system of the aryl moiety of the OPE adsorbate was extended perpendicular to its molecular long axis. Changing the aryl moiety from benzene to naphthalene to anthracene resulted in lower molecular surface densities and larger molecular inclination. The average tilt angles for the benzene, naphthalene, and anthracene SAMs were found to be about 30°, 40°, and 42° from the surface normal, respectively. For the largest adsorbate, the anthracene derivative, there is spectroscopic evidence suggesting the existence of nonequivalent binding sites. The differences observed between the SAMs are rationalized in terms of the shape of the adsorbates and the strength of the π−π interactions between them

    • Porphyrin-functionalized oligo(phenyleneethynylene)s (OPE) are promising molecules for molecular electronics applications. Three such molecules (1-3) with the common structure P-OPE-AG (P and AG are a porphyrin and anchor group, respectively) and different anchor groups, viz. an acetyl protected thiol, -S-COCH3 (1), an acetyl protected thiol with methylene linker, -CH2-S-COCH3 (2), and a trimethylsilylethynyl group, -C(triple bond)C-Si(CH3)3 (3) have been synthesized and the corresponding self-assembled monolayers (SAMs) on Au(111) substrates have been prepared. The integrity and structural properties of these films were studied by X-ray photoelectron spectroscopy and near-edge X-ray absorption fine structure spectroscopy. The results suggest that the films formed from 1 have a high orientational order with an almost upright orientation and dense packing of the molecular constituents, i.e. represent a high quality SAM. In contrast, molecule 2 formed disordered molecular layers on Au, even though the molecule-surface bonding (thiolate) is the same as in the case of molecule 1. This suggests that the methylene linker in molecule 2 has a strong impact on the quality of the resulting film, so that a well-ordered SAM cannot be formed. The silane system, 3, is also able to bind to the gold surface but the resulting SAM has a poor quality, being significantly disordered and/or comprised of strongly inclined molecules. The above results suggest that the nature of the anchor group along with a possible linker is an important parameter which, to a high extent, predetermines the entire quality of OPE-based molecular layers
