    • The lattice strain tensor evolution for single bulk grains of austenite and ferrite in a duplex stainless steel during tensile loading to 0.02 applied strain has been investigated using in situ high-energy X-ray measurements and finite-element modeling. Single-grain X-ray diffraction lattice strain data for the eight austenite and seven ferrite grains measured show a large variation of residual lattice strains, which evolves upon deformation to the point where some grains with comparable crystallographic orientations have lattice strains different by 1.5 x 10(-3), corresponding to a stress of similar to 300 MPa. The finite-element simulations of the 15 measured grains in three different spatial arrangements confirmed the complex deformation constraint and importance of local grain environment.

  • Early stage spinodal decomposition and microstructure evolution in TiAlN : A combined in-situ SAXS and phase field study

    • This paper describes in detail the microstructure and phase evolution in Ti0.33Al0.67N and Ti0.50Al0.50N coatings during isothermal annealing, studied by in-situ small angle x-ray scattering (SAXS), in combination with phase field simulations. We show that the isostructural spinodal decomposition occurs in two stages. During the initial stage, the phase segregation proceeds with a constant size of AlN- and TiN-rich domains with an experimentally measured radius of ~0.7 nm for 5 and 20 min at 900 and 850 °C respectively in the Ti0.50Al0.50N alloy. The length of  the initial stage depends on temperature as well as metal composition, and is shorter for the higher Al-content  coating. After the initial stage, the coherent cubic AlN- and TiN-rich domains coarsen. The coarsening process is kinetically limited by diffusion, which allowed us to estimate the diffusivity and activation energies of the metals to 1.4·10-7 m2s-1 and 3.14 eV at-1 respectively.

  • Residual Stress Distributions in Additively Manufactured Parts : Effect of Build Orientation

    • Additive manufacturing (AM) of parts using a layer by layer approach has seen a rapid increase in application for production of net shape or near-net shape complex parts, especially in the field of aerospace, automotive, etc. Due to the superiority of manufacturing complex shapes with ease in comparison to the conventional methods, interest in these kinds of processes has increased. Among various methods in AM, laser powder bed fusion (LPBF) is one of the most widely used techniques to produce metallic components.As in all manufacturing processes, residual stress (RS) generation during manufacturing is a relevant issue for the AM process. RS in AM are generated due to a high thermal gradient between subsequent layers. The impact of residual stresses can be significant for the mechanical integrity of the built parts and understanding the generation of RS and the effect of AM process parameters is therefore important for a broader implementation of AM techniques. The work presented in this licentiate thesis aims to investigate the influence of build orientation on the RS distribution in AM parts. For this purpose, L-shaped Inconel 718 parts were printed by LPBF in three different orientations, 0°, 45°, and 90°, respectively. Inconel 718 was selected because it is a superalloy widely used for making gas turbine components. In addition, IN718 has in general good weldability which renders it a good material for additive manufacturing.Residual stress distributions in the parts removed from the build plate were measured using neutron diffraction technique. A simple finite element model was developed to predict the residual stresses and the effect of RS relaxation due to the separation of the parts and build plate. The trend of residual stress distribution predicted was in good agreement with experimental results. In general, compressive RS at the part center and tensile RS near the surface were found. However, while the part printed in 0° orientation had the least amount of RS in all three principal directions of part, the part built in 90° orientation possessed the highest amount of RS in both compression and tension. The study has shown that residual stress distributions in the parts are strongly dependent on the building process. Further, it has shown that the relaxation of RS associated with the removal of the parts from the build plate after printing has a great impact on the final distribution of residual stress in the parts. These results can be used as guidelines for choosing the orientations of the part during printing.

  • Infliximab reduces bacterial uptake in mucosal biopsies of Crohn’s colitis viamicrotubule-dependent pathway

    • Background: A defective intestinal barrier, shown by increased paracellular permeability is an importantpathogenic factor in Crohn’s disease (CD). TNFα is a key mediator in the regulation of the intestinal barrierfunction. Treatment with antibodies directed against TNFα, such as infliximab, has been established as animportant part of the therapeutic arsenal in severe Crohn’s disease, but the mechanisms of action have yet tobe elucidated. Part of infliximab’s effect has been suggested to be reduced apoptosis of epithelial cells andthereby reduced paracellular permeability. Our aim was to study how infliximab affects uptake of adherent E.coli across the colonic mucosa in CD. Method: Seven patients with active CD colitis were examined before and after a four week treatment withinfliximab. Control biopsies were taken from healthy volunteers (4) and patients undergoing controlexamination for colonic polyps (4), aged 36 (range 25-81), coloscopy. Biopsies were taken from macroscopicallynon-inflamed descending colon and were mounted in Ussing chambers to study barrier function. Transmucosalpassage of green fluorescent protein (GFP) incorporated E. coli HM427, an adherent bacteria isolated from thecolon of a CD patient, was studied by quantifying the translocated fluorescent bacteria using flow cytometry. Results: Bacterial passage across the colonic mucosa was increased in CD (2500 ± 300 arb. units) comparedwith controls (960 ± 280; p<0.05), and was reduced to 500 ± 200 units after the infliximab treatment (p<0.05).In biopsies treated with colchicine (a microtubuline inhibitor) uptake of E. coli HM427 was reduced by 2/3compared to non-treated biopsies. Conclusion: Patients with active Crohn’s disease showed a defect in the barrier to adherent E. coli, which waspartly mediated through a microtubule dependent uptake. The four week treatment with infliximab improvedthe intestinal barrier to these bacteria. This may constitute an important part of infliximab’s mechanisms ofaction in active colitis.

    • Strong compositional-dependent elastic properties have been observed theoretically and experimentally in Ti1−xAlxN alloys. The elastic constant, C11, changes by more than 50% depending on the Al-content. Increasing the Al-content weakens the average bond strength in the local octahedral arrangements resulting in a more compliant material. On the other hand, it enhances the directional (covalent) nature of the nearest neighbor bonds that results in greater elastic anisotropy and higher sound velocities. The strong dependence of the elastic properties on the Al-content offers new insight into the detailed understanding of the spinodal decomposition and age hardening in Ti1−xAlxN alloys.
