    • Optimal growth and development in childhood and adolescence is crucial for lifelong health and well-being1–6. Here we used data from 2,325 population-based studies, with measurements of height and weight from 71 million participants, to report the height and body-mass index (BMI) of children and adolescents aged 5–19 years on the basis of rural and urban place of residence in 200 countries and territories from 1990 to 2020. In 1990, children and adolescents residing in cities were taller than their rural counterparts in all but a few high-income countries. By 2020, the urban height advantage became smaller in most countries, and in many high-income western countries it reversed into a small urban-based disadvantage. The exception was for boys in most countries in sub-Saharan Africa and in some countries in Oceania, south Asia and the region of central Asia, Middle East and north Africa. In these countries, successive cohorts of boys from rural places either did not gain height or possibly became shorter, and hence fell further behind their urban peers. The difference between the age-standardized mean BMI of children in urban and rural areas was <1.1 kg m–2 in the vast majority of countries. Within this small range, BMI increased slightly more in cities than in rural areas, except in south Asia, sub-Saharan Africa and some countries in central and eastern Europe. Our results show that in much of the world, the growth and developmental advantages of living in cities have diminished in the twenty-first century, whereas in much of sub-Saharan Africa they have amplified.

  • Robust estimation of bacterial cell count from optical density

    • Optical density (OD) is widely used to estimate the density of cells in liquid culture, but cannot be compared between instruments without a standardized calibration protocol and is challenging to relate to actual cell count. We address this with an interlaboratory study comparing three simple, low-cost, and highly accessible OD calibration protocols across 244 laboratories, applied to eight strains of constitutive GFP-expressing E. coli. Based on our results, we recommend calibrating OD to estimated cell count using serial dilution of silica microspheres, which produces highly precise calibration (95.5% of residuals <1.2-fold), is easily assessed for quality control, also assesses instrument effective linear range, and can be combined with fluorescence calibration to obtain units of Molecules of Equivalent Fluorescein (MEFL) per cell, allowing direct comparison and data fusion with flow cytometry measurements: in our study, fluorescence per cell measurements showed only a 1.07-fold mean difference between plate reader and flow cytometry data.

  • One thousand plant transcriptomes and the phylogenomics of green plants

    • Green plants (Viridiplantae) include around 450,000-500,000 species(1,2) of great diversity and have important roles in terrestrial and aquatic ecosystems. Here, as part of the One Thousand Plant Transcriptomes Initiative, we sequenced the vegetative transcriptomes of 1,124 species that span the diversity of plants in a broad sense (Archaeplastida), including green plants (Viridiplantae), glaucophytes (Glaucophyta) and red algae (Rhodophyta). Our analysis provides a robust phylogenomic framework for examining the evolution of green plants. Most inferred species relationships are well supported across multiple species tree and supermatrix analyses, but discordance among plastid and nuclear gene trees at a few important nodes highlights the complexity of plant genome evolution, including polyploidy, periods of rapid speciation, and extinction. Incomplete sorting of ancestral variation, polyploidization and massive expansions of gene families punctuate the evolutionary history of green plants. Notably, we find that large expansions of gene families preceded the origins of green plants, land plants and vascular plants, whereas whole-genome duplications are inferred to have occurred repeatedly throughout the evolution of flowering plants and ferns. The increasing availability of high-quality plant genome sequences and advances in functional genomics are enabling research on genome evolution across the green tree of life.

    • Humans rely increasingly on sensors to address grand challenges and to improve quality of life in the era of digitalization and big data. For ubiquitous sensing, flexible sensors are developed to overcome the limitations of conventional rigid counterparts. Despite rapid advancement in bench-side research over the last decade, the market adoption of flexible sensors remains limited. To ease and to expedite their deployment, here, we identify bottlenecks hindering the maturation of flexible sensors and propose promising solutions. We first analyze challenges in achieving satisfactory sensing performance for real-world applications and then summarize issues in compatible sensor-biology interfaces, followed by brief discussions on powering and connecting sensor networks. Issues en route to commercialization and for sustainable growth of the sector are also analyzed, highlighting environmental concerns and emphasizing nontechnical issues such as business, regulatory, and ethical considerations. Additionally, we look at future intelligent flexible sensors. In proposing a comprehensive roadmap, we hope to steer research efforts towards common goals and to guide coordinated development strategies from disparate communities. Through such collaborative efforts, scientific breakthroughs can be made sooner and capitalized for the betterment of humanity.

    • The fourth generation of the Sloan Digital Sky Survey (SDSS-IV) has been in operation since 2014 July. This paper describes the second data release from this phase, and the 14th from SDSS overall (making this Data Release Fourteen or DR14). This release makes the data taken by SDSS-IV in its first two years of operation (2014-2016 July) public. Like all previous SDSS releases, DR14 is cumulative, including the most recent reductions and calibrations of all data taken by SDSS since the first phase began operations in 2000. New in DR14 is the first public release of data from the extended Baryon Oscillation Spectroscopic Survey; the first data from the second phase of the Apache Point Observatory (APO) Galactic Evolution Experiment (APOGEE-2), including stellar parameter estimates from an innovative data-driven machine-learning algorithm known as "The Cannon"; and almost twice as many data cubes from the Mapping Nearby Galaxies at APO (MaNGA) survey as were in the previous release (N = 2812 in total). This paper describes the location and format of the publicly available data from the SDSS-IV surveys. We provide references to the important technical papers describing how these data have been taken (both targeting and observation details) and processed for scientific use. The SDSS web site (www.sdss.org) has been updated for this release and provides links to data downloads, as well as tutorials and examples of data use. SDSS-IV is planning to continue to collect astronomical data until 2020 and will be followed by SDSS-V.

  • An approach to evaluate wheel-rail match properties considering the flexibility of ballastless track : Comparison of rigid and flexible track models in wheel-rail profile matching

    • Many different wheel/rail profiles are used in the China high-speed railway, and vehicle operation safety and comfort will decrease if the inappropriate wheel-rail profile pair is used. To solve the problem of estimating the wheel-rail match, many numerical models, including vehicle system dynamic models and wheel-rail rolling contact models, have been established to analyse the wheel-rail dynamic responses. Both methods have less consideration of the flexibility and vibration characteristics of ballastless track, leading to deviations in the calculation of middle and high frequency vibration. This paper proposes a vehicle-flexible track coupling model and compares it with the vehicle dynamic model (vehicle-rigid track model). In the rigid track model, only the track irregularities are considered in the track module; the vibrations and deformations of rails, track slab and the foundation are considered in the flexible track model. Taking Chinese CRH3 series wheel profile S1002CN and rail profile CHN60 as examples and considering different track excitations, the two models are compared. The wheel-rail interaction forces, wheel-rail wear depths, wear volumes and vehicle accelerations are chosen as analysis indices for the comparative study.The results show that the wheel-rail forces of the flexible track model are larger than the rigid track model in the frequency range from 70 to 120Hz, while they decrease obviously in the frequency range above 150Hz. The differences in wear depths and volumes between the two models exceed 10%. Therefore, the flexible track model should be considered when studying the match properties of different wheel-rail pairs.

  • A unique gravitational wave signal from phase transition during inflation*

    • We study the properties of gravitational wave (GW) signals produced by first-order phase transitions during the inflation era. We show that the power spectrum of a GW oscillates with its wave number. This signal can be observed directly by future terrestrial and spatial GW detectors and through the B-mode spectrum in the CMB. This oscillatory feature of the GW is generic for any approximately instantaneous sources occurring during inflation and is distinct from the GW from phase transitions after inflation. The details of the GW spectrum contain information about the scale of the phase transition and the later evolution of the universe.

Loci were developed for the following species: Acanthopagrus schlegeli, Anopheles lesteri, Aspergillus clavatus, Aspergillus flavus, Aspergillus fumigatus, Aspergillus oryzae, Aspergillus terreus, Branchiostoma japonicum, Branchiostoma belcheri, Colias behrii, Coryphopterus personatus, Cynogolssus semilaevis, Cynoglossus semilaevis, Dendrobium officinale, Dendrobium officinale, Dysoxylum malabaricum, Metrioptera roeselii, Myrmeciza exsul, Ochotona thibetana, Neosartorya fischeri, Nothofagus pumilio, Onychodactylus fischeri, Phoenicopterus roseus, Salvia officinalis L., Scylla paramamosain, Silene latifo, Sula sula, and Vulpes vulpes.     • As a remarkably powerful analytical technique, surface-enhanced Raman scattering (SERS) continues to find applications from molecular biology and chemistry to environmental and food sciences. In search of reliable and affordable SERS substrates, the development has moved from noble metals to other diverse types of structures, e.g., nano-engineered semiconductor materials, but the cost of the enhancement factors (EF) substantially decreasing. In this work, we employ biocompatible thin films of Ti–Si–Zr–Zn nanometallic glasses as the SERS substrates, while tuning the Zn composition. Aided by quartz crystal microbalance, we find that the composition of 4.3% Zn (Ti–Si–Zr–Zn4.3) gives an ultrasensitive detection of Cytochrome c (Cyt c) with an EF of 1.38 × 104, 10-fold higher than the previously reported EF in the semiconducting metal oxide nanomaterials, such as TiO2, and even comparable to the reported noble-metal-assisted semiconducting tungsten oxide hydrate. Ti–Si–Zr–Zn4.3 exhibits a stronger adhesion force toward Cyt c, which ensures the strong binding of Cyt c to the surface, facilitating the Cyt c adsorption onto the surface and thus enhancing the SERS signal. The high separation efficiency of photoinduced electrons and holes in Ti–Si–Zr–Zn4.3 is also acknowledged for promoting the SERS activity.

    • Being taller is associated with enhanced longevity, and higher education and earnings. We reanalysed 1472 population-based studies, with measurement of height on more than 18.6 million participants to estimate mean height for people born between 1896 and 1996 in 200 countries. The largest gain in adult height over the past century has occurred in South Korean women and Iranian men, who became 20.2 cm (95% credible interval 17.522.7) and 16.5 cm (13.319.7) taller, respectively. In contrast, there was little change in adult height in some sub-Saharan African countries and in South Asia over the century of analysis. The tallest people over these 100 years are men born in the Netherlands in the last quarter of 20th century, whose average heights surpassed 182.5 cm, and the shortest were women born in Guatemala in 1896 (140.3 cm; 135.8144.8). The height differential between the tallest and shortest populations was 19-20 cm a century ago, and has remained the same for women and increased for men a century later despite substantial changes in the ranking of countries.

  • The loud and the quiet: Searching for radio counterparts of two radio-weak BL Lac candidates with VLBI

    • BL Lac objects are known to have compact jets inclined to our line of sight at a small angle, showing prominent radio emission. Two radio-weak BL Lac candidates with no counterparts in current radio surveys were recently reported by Massaro et al. Both sources were selected as candidate low-energy counterparts of unassociated Fermi γ -ray sources. We carried out very long baseline interferometry (VLBI) observations with the European VLBI Network (EVN) at 5 GHz to explore their radio properties at the milliarcsecond (mas) scale. One target, J1410+7405, is clearly detected with the EVN. Its measured 5-GHz flux density, 2.4 mJy, is consistent with recent interferometricmeasurementswith theKarlG. JanskyVery LargeArray, suggesting that the radio emission is confined to the inner ≲10-mas region. J1410+7405 is therefore identified as a radio-loud jetted active galactic nucleus, and its brightness temperature exceeds ~109 K. Its properties are similar to those of other γ -ray-detected BL Lac objects. On the other hand, the second target, J0644+6031, remains undetected with the EVN with a 6s brightness upper limit of 0.12 mJy beam-1. This source is thus radio-quiet, confirming its peculiarity, or possibly questioning its BL Lac nature.

    • To develop and test a mobile application that supports the disease self-management of adolescents with type 1 diabetes during their transition to early adulthood.A sequential mixed-methods design was employed.The application content was designed according to previously identified care needs and expectations, followed by application development on the Android operating system. From the outpatient clinic of the Department of Paediatric Endocrinology and Metabolism at a medical centre in northern Taiwan, 35 individuals aged between 16-25years participated in application testing.The overall median score of the QUIS was 4-5, most of the 25% quartile was 4-5, and all of the 75% quartile was 5, indicating adequate user interaction satisfaction.

    • Comparative analysis of multiple genomes in a phylogenetic framework dramatically improves the precision and sensitivity of evolutionary inference, producing more robust results than single-genome analyses can provide. The genomes of 12 Drosophila species, ten of which are presented here for the first time (sechellia, simulans, yakuba, erecta, ananassae, persimilis, willistoni, mojavensis, virilis and grimshawi), illustrate how rates and patterns of sequence divergence across taxa can illuminate evolutionary processes on a genomic scale. These genome sequences augment the formidable genetic tools that have made Drosophila melanogaster a pre-eminent model for animal genetics, and will further catalyse fundamental research on mechanisms of development, cell biology, genetics, disease, neurobiology, behaviour, physiology and evolution. Despite remarkable similarities among these Drosophila species, we identified many putatively non-neutral changes in protein-coding genes, non-coding RNA genes, and cis-regulatory regions. These may prove to underlie differences in the ecology and behaviour of these diverse species.

  • Single-Junction Organic Photovoltaic Cells with Approaching 18% Efficiency

    • Optimizing the molecular structures of organic photovoltaic (OPV) materials is one of the most effective methods to boost power conversion efficiencies (PCEs). For an excellent molecular system with a certain conjugated skeleton, fine tuning the alky chains is of considerable significance to fully explore its photovoltaic potential. In this work, the optimization of alkyl chains is performed on a chlorinated nonfullerene acceptor (NFA) named BTP-4Cl-BO (a Y6 derivative) and very impressive photovoltaic parameters in OPV cells are obtained. To get more ordered intermolecular packing, the n-undecyl is shortened at the edge of BTP-eC11 to n-nonyl and n-heptyl. As a result, the NFAs of BTP-eC9 and BTP-eC7 are synthesized. The BTP-eC7 shows relatively poor solubility and thus limits its application in device fabrication. Fortunately, the BTP-eC9 possesses good solubility and, at the same time, enhanced electron transport property than BTP-eC11. Significantly, due to the simultaneously enhanced short-circuit current density and fill factor, the BTP-eC9-based single-junction OPV cells record a maximum PCE of 17.8% and get a certified value of 17.3%. These results demonstrate that minimizing the alkyl chains to get suitable solubility and enhanced intermolecular packing has a great potential in further improving its photovoltaic performance.

  • Dairy consumption, systolic blood pressure, and risk of hypertension : Mendelian randomization study

    • OBJECTIVE To examine whether previous observed inverse associations of dairy intake with systolic blood pressure and risk of hypertension were causal. DESIGN Mendelian randomization study using the single nucleotide polymorphism rs4988235 related to lactase persistence as an instrumental variable. SETTING CHARGE (Cohorts for Heart and Aging Research in Genomic Epidemiology) Consortium. PARTICIPANTS Data from 22 studies with 171 213 participants, and an additional 10 published prospective studies with 26 119 participants included in the observational analysis. MAIN OUTCOME MEASURES The instrumental variable estimation was conducted using the ratio of coefficients approach. Using metaanalysis, an additional eight published randomized clinical trials on the association of dairy consumption with systolic blood pressure were summarized. RESULTS Compared with the CC genotype (CC is associated with complete lactase deficiency), the CT/TT genotype (TT is associated with lactose persistence, and CT is associated with certain lactase deficiency) of LCT-13910 (lactase persistence gene) rs4988235 was associated with higher dairy consumption (0.23 (about 55 g/day), 95% confidence interval 0.17 to 0.29) serving/day; P<0.001) and was not associated with systolic blood pressure (0.31, 95% confidence interval -0.05 to 0.68 mm Hg; P=0.09) or risk of hypertension (odds ratio 1.01, 95% confidence interval 0.97 to 1.05; P=0.27). Using LCT-13910 rs4988235 as the instrumental variable, genetically determined dairy consumption was not associated with systolic blood pressure (beta=1.35, 95% confidence interval -0.28 to 2.97 mm Hg for each serving/day) or risk of hypertension (odds ratio 1.04, 0.88 to 1.24). Moreover, meta-analysis of the published clinical trials showed that higher dairy intake has no significant effect on change in systolic blood pressure for interventions over one month to 12 months (intervention compared with control groups: beta=-0.21, 95% confidence interval -0.98 to 0.57 mm Hg). In observational analysis, each serving/day increase in dairy consumption was associated with -0.11 (95% confidence interval -0.20 to -0.02 mm Hg; P=0.02) lower systolic blood pressure but not risk of hypertension (odds ratio 0.98, 0.97 to 1.00; P=0.11). CONCLUSION The weak inverse association between dairy intake and systolic blood pressure in observational studies was not supported by a comprehensive instrumental variable analysis and systematic review of existing clinical trials.

    • The Southern Ocean surrounding Antarctica is a region that is key to a range of climatic and oceanographic processes with worldwide effects, and is characterised by high biological productivity and biodiversity. Since 2013, the International Bathymetric Chart of the Southern Ocean (IBCSO) has represented the most comprehensive compilation of bathymetry for the Southern Ocean south of 60 degrees S. Recently, the IBCSO Project has combined its efforts with the Nippon Foundation - GEBCO Seabed 2030 Project supporting the goal of mapping the world's oceans by 2030. New datasets initiated a second version of IBCSO (IBCSO v2). This version extends to 50 degrees S (covering approximately 2.4 times the area of seafloor of the previous version) including the gateways of the Antarctic Circumpolar Current and the Antarctic circumpolar frontal systems. Due to increased (multibeam) data coverage, IBCSO v2 significantly improves the overall representation of the Southern Ocean seafloor and resolves many submarine landforms in more detail. This makes IBCSO v2 the most authoritative seafloor map of the area south of 50 degrees S.

    • WISE J224607.56-052634.9 (W2246-0526) is a hyperluminous (L-bol 1.7 x 10(14) L), dust-obscured, and radio-quiet quasar at redshift z = 4.6. It plays a key role in probing the transition stage between dusty starbursts and unobscured quasars in the coevolution of galaxies and supermassive black holes (SMBHs). To search for the evidence of the jet activity launched by the SMBH in W2246-0526, we performed very long baseline interferometry observations of its radio counterpart with the European VLBI Network (EVN) plus the enhanced Multi Element Remotely Linked Interferometer Network (e-MERLIN) at 1.66 GHz and the Very Long Baseline Array (VLBA) at 1.44 and 1.66 GHz. The deep EVN plus e-MERLIN observations detect a compact (size <= 32 pc) sub-mJy component contributing about 10% of its total flux density, which spatially coincides with the peak of dust continuum and [C II] emissions. Together with its relatively high brightness temperature ( >= 8 x 10(6) K), we interpret the component as a consequence of nonthermal radio activity powered by the central SMBH, which likely originates from a stationary jet base. The resolved-out radio emission possibly come from a diffuse jet, quasar-driven winds, or both, while the contribution by star formation activity is negligible. Moreover, we propose an updated geometry structure of its multiwavelength active nucleus and shed light on the radio quasar selection bias toward the blazars at z > 4.

    • BACKGROUND: Associations between dairy intake and body mass index (BMI) have been inconsistently observed in epidemiological studies, and the causal relationship remains ill defined.METHODS: We performed Mendelian randomization (MR) analysis using an established dairy intake-associated genetic polymorphism located upstream of the lactase gene (LCT-13910 C/T, rs4988235) as an instrumental variable (IV). Linear regression models were fitted to analyze associations between (a) dairy intake and BMI, (b) rs4988235 and dairy intake, and (c) rs4988235 and BMI in each study. The causal effect of dairy intake on BMI was quantified by IV estimators among 184802 participants from 25 studies.RESULTS: Higher dairy intake was associated with higher BMI (β = 0.03 kg/m2 per serving/day; 95% CI, 0.00–0.06; P = 0.04), whereas the LCT genotype with 1 or 2 T allele was significantly associated with 0.20 (95% CI, 0.14–0.25) serving/day higher dairy intake (P = 3.15 × 10−12) and 0.12 (95% CI, 0.06–0.17) kg/m2 higher BMI (P = 2.11 × 10−5). MR analysis showed that the genetically determined higher dairy intake was significantly associated with higher BMI (β = 0.60 kg/m2 per serving/day; 95% CI, 0.27–0.92; P = 3.0 × 10−4).CONCLUSIONS: The present study provides strong evidence to support a causal effect of higher dairy intake on increased BMI among adults.

    • Bathymetry (seafloor depth), is a critical parameter providing the geospatial context for a multitude of marine scientific studies. Since 1997, the International Bathymetric Chart of the Arctic Ocean (IBCAO) has been the authoritative source of bathymetry for the Arctic Ocean. IBCAO has merged its efforts with the Nippon Foundation-GEBCO-Seabed 2030 Project, with the goal of mapping all of the oceans by 2030. Here we present the latest version (IBCAO Ver. 4.0), with more than twice the resolution (200 x 200m versus 500 x 500m) and with individual depth soundings constraining three times more area of the Arctic Ocean (similar to 19.8% versus 6.7%), than the previous IBCAO Ver. 3.0 released in 2012. Modern multibeam bathymetry comprises similar to 14.3% in Ver. 4.0 compared to similar to 5.4% in Ver. 3.0. Thus, the new IBCAO Ver. 4.0 has substantially more seafloor morphological information that offers new insights into a range of submarine features and processes; for example, the improved portrayal of Greenland fjords better serves predictive modelling of the fate of the Greenland Ice Sheet.     • AR Scorpii (AR Sco), the only-known radio-pulsing white dwarf binary, shows unusual pulsating emission at the radio, infrared, optical, and ultraviolet bands. To determine its astrometric parameters at the radio band independently, we conducted multi-epoch Very Long Baseline Interferometry (VLBI) phase-referencing observations with the European VLBI Network at 5 GHz and the Chinese VLBI Network plus the Warkworth 30-m telescope (New Zealand) at 8.6 GHz. By using the differential VLBI astrometry, we provide high-precision astrometric measurements on the parallax (pi = 8.52(-0.07)(+0.04) mas) and proper motion (mu(alpha) = 9.48(-0.07)(+0.04) mas yr(-1), mu(delta) = -51.32(-0.38)(+0.22) mas yr (-1)). The new VLBI results agree with the optical Gaia astrometry. Our kinematic analysis reveals that the Galactic space velocities of AR Sco are quite consistent with that of both intermediate polars and polars. Combined with the previous tightest VLBI constraint on the size, our parallax distance suggests that the radio emission of AR Sco should be located within the light cylinder of its white dwarf.

    • Newton's gravitational constant G may vary with time at an extremely low level. The time variability of G will affect the orbital motion of a millisecond pulsar in a binary system and cause a tiny difference between the orbital period-dependent measurement of the kinematic distance and the direct measurement of the annual parallax distance. PSR J0437-4715 is the nearest millisecond pulsar and the brightest at radio wavelengths. To explore the feasibility of achieving a parallax distance accuracy of one light-year, comparable to the recent timing result, with the technique of differential astrometry, we searched for compact radio sources quite close to PSR J0437-4715. Using existing data from the Very Large Array and the Australia Telescope Compact Array, we detected two sources with flat spectra, relatively stable flux densities of 0.9 and 1.0 mJy at 8.4 GHz and separations of 13 and 45 arcsec. With a network consisting of the Long Baseline Array and the Kunming 40-m radio telescope, we found that both sources have a point-like structure and a brightness temperature of >= 10(7) K. According to these radio inputs and the absence of counterparts in other bands, we argue that they are most likely the compact radio cores of extragalactic active galactic nuclei, rather than Galactic radio stars. The finding of these two radio active galactic nuclei will enable us to achieve a sub-pc distance accuracy with in-beam phase-referencing very-long-baseline interferometric observations and provide one of the most stringent constraints on the time variability of G in the near future.

    • Hexagonal boron nitride (hBN) has recently been demonstrated to contain optically polarized and detected electron spins that can be utilized for implementing qubits and quantum sensors in nanolayered-devices. Understanding the coherent dynamics ofmicrowave driven spins in hBN is of crucial importance for advancing these emerging new technologies. Here, we demonstrate and study the Rabi oscillation and related phenomena of a negatively charged boron vacancy (V-B(-)) spin ensemble in hBN. We report on different dynamics of the V-B(-) spins at weak and strong magnetic fields. In the former case the defect behaves like a single electron spin system, while in the latter case it behaves like a multi-spin system exhibiting multiple-frequency dynamical oscillation as beat in the Ramsey fringes. We also carry out theoretical simulations for the spin dynamics of V-B(-) and reveal that the nuclear spins can be driven via the strong electron nuclear coupling existing in V-B(-) center, which can be modulated by the magnetic field and microwave field.

    • Kashin-Beck disease (KBD) is a chronic endemic osteochondropathy with unclear pathogenesis. It is a degenerative disease similar to osteoarthritis, but with different manifestations of cartilage damage. The aim of this investigation was to show the protein changes in KBD cartilage and to identify the candidate proteins in order to understand the pathogenesis of the disease. Proteins were extracted from the media of primary cell cultures of KBD and normal chondrocytes, and separated by two-dimensional fluorescence difference gel electrophoresis (2-D DIGE). MALDI-TOF/TOF analysis revealed statistically significant differences in 27 proteins from KBD chondrocyte cultures, which consisted of 17 up-regulated and ten down-regulated proteins. The results were further validated by Western blot analysis. The proteins identified are mainly involved in cellular redox homeostasis and stress response (MnSOD, Hsp27, Peroxiredoxin-1, and Cofilin-1), glycolysis (PGK-1, PGM-1, α-enolase), and cell motility and cytoskeletal organization (Actin, Calponin-2, and Keratin). These KBD-associated proteins indicate that cytoskeletal remodeling, glycometabolism, and oxidative stress are abnormal in KBD articular cartilage.

    • Aims/hypothesis: Sugar-sweetened beverages (SSBs) are a major dietary contributor to fructose intake. A molecular pathway involving the carbohydrate responsive element-binding protein (ChREBP) and the metabolic hormone fibroblast growth factor 21 (FGF21) may influence sugar metabolism and, thereby, contribute to fructose-induced metabolic disease. We hypothesise that common variants in 11 genes involved in fructose metabolism and the ChREBP-FGF21 pathway may interact with SSB intake to exacerbate positive associations between higher SSB intake and glycaemic traits. Methods: Data from 11 cohorts (six discovery and five replication) in the CHARGE (Cohorts for Heart and Aging Research in Genomic Epidemiology) Consortium provided association and interaction results from 34,748 adults of European descent. SSB intake (soft drinks, fruit punches, lemonades or other fruit drinks) was derived from food-frequency questionnaires and food diaries. In fixed-effects meta-analyses, we quantified: (1) the associations between SSBs and glycaemic traits (fasting glucose and fasting insulin); and (2) the interactions between SSBs and 18 independent SNPs related to the ChREBP-FGF21 pathway. Results: In our combined meta-analyses of discovery and replication cohorts, after adjustment for age, sex, energy intake, BMI and other dietary covariates, each additional serving of SSB intake was associated with higher fasting glucose (β ± SE 0.014 ± 0.004 [mmol/l], p = 1.5 × 10−3) and higher fasting insulin (0.030 ± 0.005 [loge pmol/l], p = 2.0 × 10−10). No significant interactions on glycaemic traits were observed between SSB intake and selected SNPs. While a suggestive interaction was observed in the discovery cohorts with a SNP (rs1542423) in the β-Klotho (KLB) locus on fasting insulin (0.030 ± 0.011 loge pmol/l, uncorrected p = 0.006), results in the replication cohorts and combined meta-analyses were non-significant. Conclusions/interpretation: In this large meta-analysis, we observed that SSB intake was associated with higher fasting glucose and insulin. Although a suggestive interaction with a genetic variant in the ChREBP-FGF21 pathway was observed in the discovery cohorts, this observation was not confirmed in the replication analysis.     • A tidal disruption event (TDE) involves the shredding of a star in the proximity of a supermassive black hole (SMBH). The nearby (approximate to 230 Mpc) relatively radio-quiet, thermal-emission-dominated source AT2019dsg is the first TDE with a potential neutrino association. The origin of nonthermal emission remains inconclusive; possibilities include a relativistic jet or a subrelativistic outflow. Distinguishing between them can address neutrino production mechanisms. High-resolution very long baseline interferometry 5 GHz observations provide a proper motion of 0.94 +/- 0.65 mas yr(-1) (3.2 +/- 2.2 c; 1 sigma). Modeling the radio emission favors an origin from the interaction between a decelerating outflow (velocity approximate to 0.1 c) and a dense circumnuclear medium. The transition of the synchrotron self-absorption frequency through the observation band marks a peak flux density of 1.19 +/- 0.18 mJy at 152.8 +/- 16.2 days. An equipartition analysis indicates an emission-region distance of >= 4.7 x 10(16) cm, magnetic field strength >= 0.17 G, and number density >= 5.7 x 10(3) cm(-3). The disruption involves a approximate to 2 M (circle dot) star with a penetration factor approximate to 1 and a total energy output of <= 1.5 x 10(52) erg. The outflow is radiatively driven by the accretion of stellar debris onto the SMBH. Neutrino production is likely related to the acceleration of protons to peta-electron-volt energies and the availability of a suitable cross section at the outflow base. The present study thus helps exclude jet-related origins for nonthermal emission and neutrino production, and constrains nonjetted scenarios.

    • To date, PMN J2134–0419 (at a redshift z = 4.33) is the second most distant quasar known with a milliarcsecond-scale morphology permitting direct estimates of the jet proper motion. Based on two-epoch observations, we constrained its radio jet proper motion using the very long baseline interferometry (VLBI) technique. The observations were conducted with the European VLBI Network (EVN) at 5 GHz on 1999 November 26 and 2015 October 6. We imaged the central 10-pc scale radio jet emission and modelled its brightness distribution. By identifying a jet component at both epochs separated by 15.86 yr, a proper motion of μ = 0.035 ± 0.023 mas yr−1 is found. It corresponds to an apparent superluminal speed of βa = 4.1 ± 2.7 c. Relativistic beaming at both epochs suggests that the jet viewing angle with respect to the line of sight is smaller than 20°, with a minimum bulk Lorentz factor Γ = 4.3. The small value of the proper motion is in good agreement with the expectations from the cosmological interpretation of the redshift and the current cosmological model. Additionally we analysed archival Very Large Array observations of J2143−0419 and found indication of a bent jet extending to ∼30 kpc.

    • We monitored the position of the close-by (about 370 Mpc) gamma-ray burst GRB 190829A, which originated from a massive star collapse, through very long baseline interferometry (VLBI) observations with the European VLBI Network and the Very Long Baseline Array, carrying out a total of nine observations between 9 and 117 days after the gamma-ray burst at 5 and 15 GHz, with a typical resolution of a few milliarcseconds. From a state-of-the art analysis of these data, we obtained valuable limits on the source size and expansion rate. The limits are in agreement with the size evolution entailed by a detailed modeling of the multiwavelength light curves with a forward-plus-reverse shock model, which agrees with the observations across almost 18 orders of magnitude in frequency (including the HESS data at TeV photon energies) and more than 4 orders of magnitude in time. Thanks to the multiwavelength, high-cadence coverage of the afterglow, inherent degeneracies in the afterglow model are broken to a large extent, allowing us to capture some unique physical insights; we find a low prompt emission efficiency of less than or similar to 10(-3), a low fraction of relativistic electrons in the forward shock downstream chi ( e ) < 13% (90% credible level), and a rapid decay of the magnetic field in the reverse shock downstream after the shock crossing. While our model assumes an on-axis jet, our VLBI astrometry is not sufficiently tight as to exclude any off-axis viewing angle, but we can exclude the line of sight to have been more than similar to 2 degrees away from the border of the gamma-ray-producing region based on compactness arguments.

    • Poplar (Populus) is a well-established model system for tree genomics and molecular breeding, and hybrid poplar is widely used in forest plantations. However, distinguishing its diploid homologous chromosomes is difficult, complicating advanced functional studies on specific alleles. In this study, we applied a trio-binning design and PacBio high-fidelity long-read sequencing to obtain haplotype-phased telomere-to-telomere genome assemblies for the 2 parents of the well-studied F1 hybrid “84K” (Populus alba × Populus tremula var. glandulosa). Almost all chromosomes, including the telomeres and centromeres, were completely assembled for each haplotype subgenome apart from 2 small gaps on one chromosome. By incorporating information from these haplotype assemblies and extensive RNA-seq data, we analyzed gene expression patterns between the 2 subgenomes and alleles. Transcription bias at the subgenome level was not uncovered, but extensive-expression differences were detected between alleles. We developed machine-learning (ML) models to predict allele-specific expression (ASE) with high accuracy and identified underlying genome features most highly influencing ASE. One of our models with 15 predictor variables achieved 77% accuracy on the training set and 74% accuracy on the testing set. ML models identified gene body CHG methylation, sequence divergence, and transposon occupancy both upstream and downstream of alleles as important factors for ASE. Our haplotype-phased genome assemblies and ML strategy highlight an avenue for functional studies in Populus and provide additional tools for studying ASE and heterosis in hybrids.

    • We observed 20 Palomar-Green quasars at low redshi ft (z < 0.5) with total flux density >1 mJy, including four radio-loud quasars (RLQs) and 16 radio-quiet quasars (RQQs), using the Very Long Baseline Array (VLBA) at 5 GHz. 10 RQQs are clearly detected in the VLBA images, and a compact radio core is identified in eight of them, indicating the prevalence of active galactic nucleus (AGN)-related radio emission in this flux-density-limited RQQ sample. The RQQs and RLQs in our sample have a division at similar to 30 mJy. The radio emission from RQQs appears to be the result of a combination of star formation and AGN-associated activities. All RQQs in our sample have a 5 GHz flux density ratio of Very Large Array (VLA) A-array to D-array f(c) = S-A(VLA)/S-D(VLA) above 0.2. The RQQs with f(a)(VLBA and VLA flux density ratio S-VLBA/S-A(VLA)) > 0.2 versus f(a) < 0.2 show significant differences in morphology, compactness, and total flux density. f(a) of RQQs is systematically lower than that of RLQs, probably due to the extended jets or relic jets of RQQs on tens to hundreds parsecs that are resolved out in VLBA images. Future larger samples, especially with the addition of milliarcsec resolution radio images of RQQs with total flux densities below 1 mJy, can test the conclusions of this paper and contribute to the understanding of the radio emission mechanism of RQQs, and the dichotomy and physical connection between RQQs and RLQs.

    • Azaleas (Ericaceae) comprise one of the most diverse ornamental plants, renowned for their cultural and economic importance. We present a chromosome-scale genome assembly for Rhododendron simsii, the primary ancestor of azalea cultivars. Genome analyses unveil the remnants of an ancient whole-genome duplication preceding the radiation of most Ericaceae, likely contributing to the genomic architecture of flowering time. Small-scale gene duplications contribute to the expansion of gene families involved in azalea pigment biosynthesis. We reconstruct entire metabolic pathways for anthocyanins and carotenoids and their potential regulatory networks by detailed analysis of time-ordered gene co-expression networks. MYB, bHLH, and WD40 transcription factors may collectively regulate anthocyanin accumulation in R. simsii, particularly at the initial stages of flower coloration, and with WRKY transcription factors controlling progressive flower coloring at later stages. This work provides a cornerstone for understanding the underlying genetics governing flower timing and coloration and could accelerate selective breeding in azalea. Azaleas are one of the most diverse ornamental plants and have cultural and economic importance. Here, the authors report a chromosome-scale genome assembly for the primary ancestor of the azalea cultivar Rhododendro simsi and identify transcription factors that may function in flower coloration at different stages.

    • The nearby face-on spiral galaxy NGC 2617 underwent an unambiguous 'inside-out' multiwavelength outburst in Spring 2013, and a dramatic Seyfert-type change probably between 2010 and 2012, with the emergence of broad optical emission lines. To search for the jet activity associated with this variable accretion activity, we carried out multiresolution and multiwavelength radio observations. Using the very long baseline interferometric (VLBI) observations with the European VLBI Network at 1.7 and 5.0 GHz, we find that NGC 2617 shows a partially synchrotron self-absorbed compact radio core with a significant core shift, and an optically thin steep-spectrum jet extending towards the north up to about 2 pc in projection. We also observed NGC 2617 with the electronic Multi-Element Remotely Linked Interferometer Network at 1.5 and 5.5 GHz, and revisited the archival data of the Very Large Array (VLA) and the Very Long Baseline Array (VLBA). The radio core had a stable flux density of similar to 1.4 mJy at 5.0 GHz between 2013 June and 2014 January, in agreement with the expectation of a supermassive black hole in the low accretion rate state. The northern jet component is unlikely to be associated with the 'inside-out' outburst of 2013. Moreover, we report that most optically selected changing-look active galactic nuclei (AGN) at z < 0.83 are sub-mJy radio sources in the existing VLA surveys at 1.4 GHz, and it is unlikely that they are more active than normal AGN at radio frequencies.

    • Broad Balmer emission lines in active galactic nuclei (AGN) may display dramatic changes in amplitude, even disappearance and re-appearance in some sources. As a nearby galaxy at a redshift of z = 0.0264, Mrk 590 suffered such a cycle of Seyfert type changes between 2006 and 2017. Over the last 50 yr, Mrk 590 also underwent a powerful continuum outburst and a slow fading from X-rays to radio wavelengths with a peak bolometric luminosity reaching about 10 per cent of the Eddington luminosity. To track its past accretion and ejection activity, we performed very long baseline interferometry (VLBI) observations with the European VLBI Network (EVN) at 1.6 GHz in 2015. The EVN observations reveal a faint (∼1.7 mJy) radio jet extending up to ∼2.8 mas (projected scale ∼1.4 pc) toward north, and probably resulting from the very intensive AGN activity. To date, such a parsec-scale jet is rarely seen in the known changing-look AGN. The finding of the faint jet provides further strong support for variable accretion as the origin of the type changes in Mrk 590.

    • Active galactic nuclei (AGNs) accreting at rates close to the Eddington limit can host radiatively driven mildly relativistic outflows. Some of these X-ray absorbing but powerful outflows can produce strong shocks, resulting in a significant non-thermal emission. This outflow-driven radio emission might be detectable in the radio-quiet quasar PDS 456, as it has a bolometric luminosity that reaches the Eddington limit and a relativistic wide-aperture X-ray outflow with a kinetic power high enough to quench the star formation in its host galaxy. To investigate this possibility, we performed very-long-baseline interferometric (VLBI) observations of the quasar with the European VLBI Network (EVN) at 5 GHz. The full-resolution EVN image reveals two faint and diffuse radio components with a projected separation of about 20 pc and an average brightness temperature of around two million Kelvin. In relation to the optical submas-accuracy position measured by the Gaia mission, the two components are very likely on opposite sides of an undetected radio core. Thus, the VLBI structure at the deca-pc scale could be either a young jet or a bidirectional radio-emitting outflow, launched in the vicinity of a strongly accreting central engine. Two diffuse components at the hecto-pc scale, likely the relic radio emission from past AGN activity, are tentatively detected on each side in the low-resolution EVN image.

    • Highly accreting quasars are quite luminous in the X-ray and optical regimes; while, they tend to become radio quiet and have optically thin radio spectra. Among the known quasars, IRAS F11119+3257 is a supercritical accretion source because it has a bolometric luminosity slightly above the Eddington limit and extremely powerful X-ray outflows. To probe its radio structure, we investigated its radio spectrum between 0.15 and 96.15 GHz and performed very-long-baseline interferometric (VLBI) observations with the European VLBI Network (EVN) at 1.66 and 4.93 GHz. The deep EVN image at 1.66 GHz shows a two-sided jet with a projected separation about 200 pc and a very high flux density ratio of about 290. Together with the best-fitting value of the integrated spectral index of -1.31 +/- 0.02 in the optically thin part, we infer that the approaching jet has an intrinsic speed at least 0.57 times of the light speed. This is a new record among the known all kinds of super-Eddington accreting sources and unlikely accelerated by the radiation pressure in a certain models. We propose a scenario in which IRAS F11119+3257 is an unusual compact symmetric object with a small jet viewing angle and a radio spectrum peaking at 0.53 +/- 0.06 GHz mainly due to the synchrotron self-absorption.
