  • Effects of Tobacco Smoke on IL-16 in CD8+ Cells from Human Airways and Blood: a Key Role for Oxygen Free Radicals?

    • Chronic exposure to tobacco smoke leads to an increase in the frequency of infections and in CD8(+) and CD4(+)cells as well as the CD4(+) chemo-attractant cytokine IL-16 in the airways. Here, we investigated whether tobacco smoke depletes intracellular IL-16 protein and inhibits de novo production of IL-16 in CD8(+) cells from human airways and blood, while at the same time increasing extracellular IL-16 and whether oxygen free radicals (OFR) are involved. Intracellular IL-16 protein in CD8(+) cells and mRNA in all cells was decreased in bronchoalveolar lavage (BAL) samples from chronic smokers. This was also the case in human blood CD8(+) cells exposed to water-soluble tobacco smoke components in vitro; in which oxidized proteins were markedly increased. Extracellular IL-16 protein was increased in cell-free BAL fluid from chronic smokers and in human blood CD8(+) cells exposed to water-soluble tobacco smoke components in vitro. This was not observed in occasional smokers after short-term exposure to tobacco smoke. A marker of activation (CD69) was slightly increased whereas other markers of key cellular functions (membrane integrity, apoptosis and proliferation) in human blood CD8(+) cells in vitro were negatively affected by water-soluble tobacco smoke components. An OFR scavenger prevented these effects whereas a protein synthesis inhibitor, a beta-adrenoceptor, a glucocorticoid receptor agonist, a phosphodiesterase, a calcineurin phosphatase and a caspase-3 inhibitor did not. In conclusion, tobacco smoke depletes preformed intracellular IL-16 protein, inhibits its de novo synthesis and distorts key cellular functions in human CD8(+) cells. OFR may play a key role in this context.

  • Efficiency of percutaneous core biopsy in pancreatic tumor diagnosis

    • BACKGROUND: Radiologic diagnosis of pancreatic tumors exhibits limited precision. The aim of this study was to investigate the outcome and complications of pancreatic core biopsy in patients with suspected pancreatic neoplasms. METHODS: One hundred patients underwent ultrasonography-guided core biopsy of 1.2 mm external diameter. Medical charts were examined for biochemical and clinical signs of complications. Final diagnosis was settled by operation, autopsy, and clinical signs of the disease including survival with at least 2.3 years of follow-up. RESULTS: Histopathologic biopsy evaluation showed correct discrimination between exocrine and endocrine tumors and nonneoplastic conditions in 89 patients. No false-positive cancer diagnosis was found, and guidance on nature of primary tumors was obtained for eight of eight metastases. The sensitivity was 91% for exocrine and 87% for endocrine pancreatic tumors, and negative predictive values of these diagnoses were 83% and 97%, respectively. No clinically significant complications were noted. CONCLUSIONS: Core biopsy is an attractive alternative to diagnostic laparotomy in unresectable pancreatic cancer and efficiently provides diagnosis of endocrine tumors and pancreatic metastases in conjunction with rare complications. Benign biopsy findings cannot be used to exclude presence of primary or metastatic pancreatic neoplasms.

  • Physical Activity Coaching of Patients with Rheumatoid Arthritis in Everyday Practice : A Long-term Follow-up

    • OBJECTIVES: To investigate the long-term effects on perceived general health, disease activity, pain, activity limitation and cognitive behavioural factors of a one-year coaching programme performed in ordinary physical therapy practice to promote the adoption of health-enhancing physical activity in patients with early rheumatoid arthritis (RA).METHODS: A total of 228 patients with early RA, from 10 rheumatology clinics in Sweden, were randomly assigned to an intervention group (IG; n = 94) or a control group (CG; n = 134). The IG was coached by physical therapists during the first year to adopt health-enhancing levels of physical activity (30 minutes/day, moderately intensive, ≥ 4 days/week). No coaching was given during the subsequent year between post-intervention and follow-up. Follow-up assessment consisted of a postal questionnaire on physical activity and of visual analogue scales for ratings of general health perception and pain. The Health Assessment Questionnaire Disability Index (HAQ) and the Disease Activity Score in 28 joints (DAS 28) were collected at regular medical check-ups.RESULTS: Sixty-five (69%) participants in the IG and 92 (69%) in the CG completed the entire study period by filling in the follow-up questionnaire on physical activity two years after baseline. The intervention seemed to lack any significant influence on long-term outcome. However, different patterns of change in physical activity behaviour were observed in the two groups.CONCLUSIONS: No long-term improvement in perceived general health or other outcomes were found in the follow-up. This may partly be because the intervention lacked several important behavioural elements for physical activity maintenance. Copyright © 2011 John Wiley & Sons, Ltd.

  • Tensile strength and young's modulus of polyisoprene/single-wall carbon nanotube composites increased by high pressure cross-linking

    • High-viscosity liquid cis-1,4 polyisoprene (PI), with up to 20 wt % single-wall carbon nanotubes (SWCNTs), has been cross-linked by high pressure and high temperature (HP&HT) treatment at 513 K and pressures in the range 0.5 to 1.5 GPa to yield densified network polymer composites. A composite with 5 wt % SWCNTs showed 2.2 times higher tensile strength σUTS (σUTS = 17 MPa), 2.3 times higher Young’s modulus E (E = 220 MPa) and longer extension at break than pure PI. The improvement is attributed to SWCNT reinforcement and improved SWCNT−PI interfacial contact as a result of the HP&HT cross-linking process, and reduced brittleness despite a higher measured cross-link density than that of pure PI. The latter may originate from an effect similar to crazing, i.e., bridging of microcracks by polymer fibrils. We surmise that the higher cross-link densities of the composites are due mainly to physical cross-links/constraints caused by the SWCNT−PI interaction, which also reflects the improved interfacial contact, and that the CNTs promote material flow by disrupting an otherwise chemically cross-linked network. We also deduce that the PI density increase at HP&HT cross-linking is augmented by the presence of CNTs.

Stora vindkraftsindustriområden är under planering och byggnation i Sverige. Svenska staten såväl som aktörer på energimarknaden, ideella miljöorganisationer och klimataktivister framhåller ofta vindkraften som grön, fossilfri och miljövänlig.Vindkraftsindustriområdena har omfattande negativa konsekvenser för omgivande miljö, renskötsel, djur, fåglar och natur. Vindkraften i dagens design består av uppemot 150 till 300 meter höga stålstrukturer på betongfundament. Konstruktion och underhåll förutsätter nya gruvor, bilvägar, skogsavverkning och stenbrott. Fossila bränslen är en förutsättning i allt detta.   • Sámi Land Free University : An invitation to centre Indigenous Peoples expertise and knowledge in an online open access university

    • This roundtable aims at presenting what the platform Sámi Land Free University (SFU) has achieved so far and to invite to a conversation on what SFU may achieve in the practice of decolonising research and education, while inviting interested to join forces for courses and joint or shared research over the colonial borders.Over the last four decades, Indigenous Studies (IS) has been developed as an academic discipline within several settler colonial states and within colonial academia, the strongest presence in North America, Australia and Aotearoa/New Zealand. So far Sweden is lagging behind. Yet, IS mainly turns the gaze towards the Indigenous peoples, while to achieve decolonization and long term sustainability, as well as good relations, there is a need for a 180 degrees shift in vision. What if there were several universities that centre Indigenous knowledges and expertise, as point of departure, while researching and teaching for the benefit of all?In 2015, the SFU [www.samelandsfriauniversitet.com ] was created by Lule and Forest Sámi, Dr May-Britt Ӧhman with a vision and challenge of the mind to include the Sámi community and international Indigenous collaborations, having its base on the Swedish side of Sábme. SFU has co-hosted symposia and workshops. We now envision moving on, producing free to access online educational content, inviting earlier and new collaborators. There are even less opportunities to have a place in academia for Sámi who are open with their Sámi identity. Hence, there is a need for a place to gather, to develop courses, and proposed ways to make this happen, as the (settler colonial) academia so far gives very little space to Sámi perspectives in Sweden. Yet, Swedish legislation states that the Sámi are a people, with rights, and that Sámi history, culture and tradition shall be taught to pupils in school and secondary school.Furthermore, at universities, there is almost no teaching on Indigenous peoples culture, history, traditions, around the world, while the settler colonial perspectives are taught in all university courses.Thus, there is a need for a Sámi university, on Swedish side of Sábme, that will provide both Sámi and other Indigenous peoples history, traditions, culture, expertise, perspectives - the question is mainly how to organize this.2 Frances Wyld, Martu woman (Aboriginal People of the Pilbara region of Australia) living on Kaurna land, Dr of CommunicationJustice and Society, University of South AustraliaAll good thing are wild and free: the wild winds that carried me to Sámi Land Free University.NAISA introduced me to the international cross fertilization of ideas within Indigenous research and education, specifically it created the opportunity to collaborate with Sámi Land Free University as an Australian Aboriginal scholar. The collaboration has included 6 visits to Sweden and Sápmi for symposia and workshops, publications, and freelance work on a climate change and Indigenous sustainability project. I am a storyteller; I write in the moment inspired by nature. I watch the news reporting on the damage done by wild winds on the weekend. The reporter is in front of a house where a tree has fallen onto it, the house is on my road. At the same time, through my open door, I hear the whine of the chainsaw cutting into this majestic ghost gum tree. Only weeks ago, I was riding my bicycle along the same road with a wild sense of freedom as my birds, the Rainbow Lorikeets who live in the avenue of trees, flew alongside me. Sámi Land Free University has given me the same sense of freedom to work as a decolonising scholar within Critical Indigenous Studies, gaining international perspectives and walking on two lands that are both rich is stories. The collaboration has given me opportunities, and now in the spirit of reciprocity, I lend my skills as a curriculum developer to create content for this grassroots university in another land inhabited by Indigenous people who have much in common with my own people, with knowledges to share for a sustainable future.3 Anna Kajsa Aira and Laila Susanna Kuhmunen, Jokkmokk  in collaboration with Alma Linke NilsenDuv gábdde - Du gákti - Din kolt – Your Sámi dressWe are Sámi artisans, duojár. Anna Kajsa makes handicraft – duodje - in the Lule Sámi tradition, Jokkmokk area. Laila Susanna works within the North Sámi duodji tradition of the Karesuando area. We both live in Jokkmokk/Jåhkåmåhkke, a Sámi metropolis. Sámi heritage is passed down from generation to generation.The traditional Sámi dress – gábbde/gákti/kolt - reveals one’s geographical area and is an important link to one’s roots and history. Many Sámi now reclaim their culture by wanting to wear the dress, but there is a gap of one generation in the transmission of knowledge about how to make them. Our project “your Sámi dress” is about providing this knowledge, both by making the gábbde for children and young people, for rent, and through making a digital platform with instructions and advice. It is just as important to be able to wear your gábbde as it is to handle it respectfully.  While there is a university in Sweden for textiles and fashion, there is no such for Sámi culture. We think maybe our work could find a place within the SFU, and thereby receive more support. 4 Henrik Andersson and Hampus Andersson, Gällivare Forest Sámi villageExperiences of and reflections on the needs for education and higher education for and about Sámi reindeer herdersReindeer husbandry is carried out on Sámi territories, on almost 50 percent of the territories of the colonial state of Sweden. It is is a tradition – a way of life – as well as a basis for livelihood, income, since several hundred of years. It is acknowledged as a national interest, to be protected, since 1987.Yet, the knowledge and expertise of reindeer herding is mainly passed within the families. Learning demands spending time together with the older generations, from early childhood. As the school system requests pupils to be in school, this often come into conflict with the need to learn. Furthermore, for a reindeer herder, there are an increasing set of other skills than the actual herding – which is complex enough- that are of importance; economics, law, animal health, and one’s own history and culture etc. In school, secondary school, and at universities, there are no opportunities to study reindeer herding with the other skills thereto related. Another aspect of the education system’s lack of teaching on reindeer and reindeer herding, is that when persons within the reindeer herding meet with all parts of society – health care, police, teachers, social care, etc, they have to carry the burden of constantly educating them, and also facing discrimination and racism. Henrik , 42 years old, and Hampus , 20 years old, are two generations reindeer herders within the Gällivare Forest Sámi village, Flakaberg group. They will share their experiences and reflect on the need for education and higher education for Sámi reindeer herders. 5 Susanne Spik and Gun Aira, Jåhkåmåkke in collaboration with Alma Linke NilsenLule Sámi culture and language on university level ? Experiences,  reflections and ideas Gun Aira is a Lule Sámi teacher and Susanne Spik, is an entrepreneur and innovator. Both live in Jåhkåmåhkke,   and are active within reindeer herding. Both have attended university – teacher’s education- at a Swedish university, in the 1980s. The Lule Sámi language and culture origin in the Lule River valley. Due to the by the Swedish state forced relocation of several North Sámi families, from the Karesuando area during in the 20th century, along with fierce Swedification politics, both the language and culture have become sidestepped. While there is a Lule Sámi centre on Norwegian side, there is no such on the Swedish. In general, there is vastly more support to Sámi culture and language in Norway, than in Sweden. We will present our experiences, reflections and ideas on how SFU could be of use to promote Lule Sámi language and culture, and wish to develop this with the others and the audience.

    • Title: Ungreen windpower: Sámi Indigenous and scientific perspectives on fossil dependent and environmentally destructive designsContent:This film is produced by Dálkke, a research project and research group at Uppsala University, within the Swedish National Research Programme on Climate, the Swedish research council FORMAS.In the film, Henrik Andersson, reindeer herder within Gällivare Forest Sámi village, Norrbotten County, Sweden, speaks about the environmental destruction caused by the Swedish state, through forestry practices by the Swedish state owned forest company Sveaskog, as well as the planned wind energy industrial area by the Swedish state power company Vattenfall. Associate Professor Eva Charlotta Helsdotter, Uppsala University, tells about how the analysis of windpower is not taking into account all relevant aspects in terms of fossil dependency and environmental consequences. A large part of what is needed to establish, maintain, as well as decommission windpower is never taken into account within the so called LCA analyses. This is what makes it possible to – falsely – claim that windpower is green. However, the windpower designs of today cannot be considered green, nor fossil free.The area in question is one out of two project areas for windpower within Gällivare Forest Sámi village - “Storlandet”, which is the ancestral grounds of Henrik Andersson’s family. Work is currently ongoing with an environmental impact study, planned to be ready for submission in 2022. The other wind power industry area that is planned within the area for the Gällivare Forest Sámi Village is Hällberget, conducted by a private power company Vasavind. In early 2021 it received permission to go ahead with a smaller part of its project. The Hällberget project is currently (August 2021) in court process. The Swedish state, the European Union (EU), as well as actors within the energy market, environmental organizations and climate activists all insist in the promotion of windpower as “green”, fossil free and thereby environmentally friendly. A large part of these constructions are planned for in Sámi territories, on land that has been taken from Sámi during late 19th and early 20th century. Furthermore, the wind power industrial areas have major negative impacts for the local environment. Forests are clear cut and replaced with 100 – 300 meters high steel constructions, on massive foundations made of concrete. Construction and maintenance demands new mines, car and truck access roads and thereby new stone quarries causing major wounds in the landscape.These industrial areas have major negative consequence for the local environment, fresh water, nature, wild life, local inhabitants as well as reindeer herding. This film is part of a supradisciplinary research and documentary project on (un)sustainable power production, researching and documenting the subject matter from Indigenous Sámi reindeer herding point of view, along with analyses of the whole windpower industry. Henrik Andersson, Gällivare Forest Sámi Village, who works to protect - through research and challenging decision makers - the reindeer and other animals from a destructive power production mode along with the Sámi traditional culture in the area is at the front. The Sámi reindeer herding culture is documented to having lived and prospered here since at least two millennia. What can EU, Sweden and EU member countries learn from the knowledges and understandings of the Sámi reindeer herders?   • Alcohol use among university students in Sweden measured by an electronic screening instrument

    • Background: Electronic-based alcohol screening and brief interventions for university students with problem drinking behaviours forms an important means by which to identify risky drinkers. Methods: In this study an e-SBI project was implemented to assess drinking patterns, and to provide personalised feedback about alcohol consumption and related health problems, to students in a Swedish university. In this study, third semester university students (n = 2858) from all faculties (colleges) at the University were invited to participate in e-SBI screenings. This study employed a randomised controlled trial, with respondents having a equal chance of being assigned to a limited, or full-feedback response. Results: The study shows that high risk drinkers tend to underestimate their own consumption compared to others, and that these high risk drinkers experience more negative consequences after alcohol intake, than other respondents. There was a strong belief, for both high-and low-risk drinkers, that alcohol helped celebrations be more festive. This study also confirms findings from other study locations that while males drank more than females in our study population; females reached the same peak alcohol blood concentrations as males. Conclusion: Obtaining clear and current information on drinking patterns demonstrated by university students can help public health officials, university administration, and local health care providers develop appropriate prevention and treatment strategies.

Den kartläggning som vi har gjort visar bl a att det finns ett stort behov av att systematiskt arbeta med miljöfrågor inom den nordiska träindustrin, särskilt om industrin vill öka sin konkurrenskraft på miljökunniga marknader, t ex EU.

Myndigheten saknar en FoU-strategi som omfattar hela verksamheten. Idag utförs en liten del egen forskningsverksamhet främst runforskning och en större del annan FoU-verksamhet. I stort sett all FoU utförs i samarbete och/eller samverkan med externa parter exempelvis museer, universitet och högskolor. I rapporten har sex kategorier av FoU-verksamhet identifierats. De flesta avdelningar utför FoU-verksamhet främst Kulturvårdsavdelningen, men även Arkiv och bibliotek samt Informationsavdelningen. Kulturmiljöavdelningen arbetar främst operativt med FoU-anslaget och det tillhörande externa FoU-programmet. Staben arbetar strategiskt med FoU-frågor i Sverige och inom EU samt ansvarar strategiskt för det externa FoU-programmet.

    • We present below the first measured data for the thermal diffusivity and the thermal conductivity, λ, of dense, sintered ceramic YBa2Cu4O8 (1-2-4) as a function of temperature T between 40 and 310 K. At 300 K, λ is 6.3 Wm−1K−1, increasing with decreasing T to about 9.5 Wm−1K−1 near Tc. This T dependence is much stronger than in YBa2Cu3O7−δ (1-2-3). Only a very small enhancement is observed in λ below Tc. As in 1-2-3, λ is mainly limited by phonon scattering by defects.

    • Highly dense sintered YBa2Cu4O8 has been produced by hot isostatic pressing (HIP). The electrical resistivity varrho of this material has been measured as a function of temperature T and pressure varrho in the range 40–650 K and 0–0.7 GPa. Both the temperature dependence and the pressure dependence of varrho are found to be well described by a model based on the standard Bloch-Grüneisen theory. It is pointed out that varrho is liner in T only under isobaric conditions, while varrho is strongly nonlinear in all high-Tc superconductors under isochoric (constant volume) conditions. The critical current density of the material is 900 A/cm2 at 4 K, while the resistivity is 630 μΩ cm at 294 K.

    • We have measured the thermal conductivity, lambda, and the thermal diffusivity, a, of dense bulk ceramic YBa2Cu4O8 (1-2-4) between 30 and 300 K. For this material lambda is about 10 W/Km near 100 K, comparable with the in-plane thermal conductivity for single crystals of the more common high-Tc material YBa2Cu3O7 (1-2-3). Also, lambda has a much stronger T dependence in 1-2-4 than in 1-2-3. We find that, for 1-2-4, lambda can be very well described by a semiclassical phonon theory, which includes phonon, electron, and boundary scattering. The difference between the present results and those for 1-2-3 can be explained mainly in terms of a smaller amount of point-defect scattering in 1-2-4 because of the stable oxygen stoichiometry of this material.

    • We have measured the thermal conductivity κ and the thermal diffusivity a of a dense bulk ceramic polycrystalline sample of YBa2Cu4O8 (1:2:4) in the temperature range 30–300 K. We find κ≊10 W m-1 K-1 at 100 K, significantly higher than in ceramic YBa2Cu3O7-δ (1:2:3) and approaching the in-plane value for single-crystal 1:2:3, and decreasing to 7.6 W m-1 K-1 at 300 K. The data for this sample can be described by standard theories for phonon thermal conductivity of crystalline materials with boundary, phonon, and electron scattering. The higher κ in 1:2:4 as compared to 1:2:3 is, in this model, due to the smaller point defect scattering in the former. The fitted parameters for the three scattering mechanisms all agree with independent estimates based on simple models; inserting data for electric resistivity, grain size, carrier density, and lattice properties we can predict κ and its T dependence to within about 20%. We also discuss models for the phonon and electron thermal conductivities in some detail, including some second-order effects such as inelastic electron scattering and a T-dependent carrier density.

At 100 K, κ is 10 W m-1 K-1, approaching the in-plane κ of single crystals of other high-Tc materials. κ decreases rapidly with increasing T to 7.4 W m-1 K-1 at 300 K. Fitting standard models for κ(T) to the data we find that κ is limited mainly by phonon-phonon interactions. Depending on the model used, the best fit is found for effective values of FTHETADebye between 155 and 185 K, characteristic for the acoustic phonon branches, indicating that such phonons carry most of the heat. Finally, we suggest a possible way to test the electron-phonon model for the electrical and thermal conductivities in high-Tc materials.

    • This study has evaluated the relationship between tumor growth and induction of acute-phase proteins. It has also determined whether an intact cellular immunity is obligatory for a fully expressed acute-phase plasma protein response in the presence of a highly antigenic tumor. Quantitatively, acute-phase responses (protein synthesis, plasma concentrations, hepatic RNA content, anorexia) were proportional to tumor burden. Anti-inflammatory drugs (indomethacin 1 micrograms/g body wt, dexamethasone 0.5 micrograms/g body wt) had no direct effect on the attenuation of the systemic acute-phase responses, but did affect them indirectly by decreasing tumor growth. Immune suppression (cyclosporine A at 20 or 60 micrograms/g body wt) had no effect on either acute-phase reactions or local tumor growth. In endotoxin-stimulated (lipopolysaccharide) normal mice, immune suppression aggravated anorexia and caused high mortality, while dexamethasone partly reversed these effects in endotoxin-stimulated mice. Plasma levels of acute-phase proteins correlated to circulating levels of IL-6 in untreated tumor-bearing mice, but this relationship was not obvious in either drug-treated tumor-bearing or endotoxin-stimulated mice. Tumor tissue induced the synthesis of different acute-phase proteins compared to endotoxin. However, disintegrated normal liver tissue induced the synthesis of serum amyloid protein to the same extent as the growing tumor. This effect was primarily associated with the mitochondrial/lysosomal and microsomal liver cell fractions. In conclusion, the overall acute-phase protein response is not a modulating factor of tumor growth.(ABSTRACT TRUNCATED AT 250 WORDS)

    • Plasma albumin concentration declines in both experimental and clinical cancer. Previous investigations have demonstrated that this is partly explained by increased breakdown of albumin. The present study has identified the tissue sites for increased albumin degradation in a nonmetastasizing sarcoma mouse (C57/BL6J) model. Results have been compared to nontumor-bearing animals either freely fed or food restricted (pair-weighed) so that their body composition was similar to tumor-bearing animals. Tumor-bearing mice had increased albumin degradation (0.13 +/- 0.02 mg/hr/g bw) compared to both freely fed (0.09 +/- 0.007) and pair-weighed control animals (0.05 +/- 0.008). Radioactivity from circulating [3H]raffine aldehyde labeled albumin appeared with maximum peak values in lysosomes isolated from both tumor and nontumor tissues at 48 hr following iv injection. The intralysosomal accumulation of radioactivity was two- to threefold higher in tumor tissue compared to liver tissue, although the specific activity of protease(s) for albumin degradation measured in vitro was not higher in tumor tissue (30.4 +/- 3.6 mg/hr/g tissue) compared to normal liver tissue (36.9 +/- 1.7). Accounting for the entire tumor the proteolytic capacity for albumin breakdown was however much larger in the tumor (161.6 +/- 32.6 mg/organ) compared to both normal liver (37.5 +/- 2.3) and tumor-host liver (56.4 +/- 2.8). Pepstatin inhibited 78 +/- 6% of the proteolytic activity in the tumor measured by 125I-labeled undenatured mouse albumin as the substrate. Leupeptin inhibited 49 +/- 6%. There was a significantly decreased breakdown of albumin in both skeletal muscles and the gastrointestinal tract from tumor-bearing animals.(ABSTRACT TRUNCATED AT 250 WORDS)

    • It is essential to obtain biochemically and radioactively pure albumin in studies on albumin metabolism and kinetics in stress and nutrition related conditions. However, published work on albumin metabolism, in both animals and man with acute phase reactions has usually been based on inadequate chemical methods for isolation of homogeneous albumin free from acute phase proteins and other contaminants. Applications of conventional antibody precipitating techniques was usually either not sufficient to give radiopure albumin, or did not allow determination of the true specific radioactivity during in vivo experiments. Thus, the lack of applicable methods to achieve radiopure albumin from small plasma and tissue samples for subsequent analyses and determination of the true specific radioactivity in albumin initiated the present method development. The combination of HPLC ionchromatography (DEAE-sepharose), affinity chromatography (Blue sepharose CL-6B, Con A sepharose) and HPLC based size exclusion chromatography (Protein PAK 300 SW, Waters) was applied. By this procedure we obtained radiopure albumin from both plasma and hepatic samples from individual mice with acute phase response as confirmed by two-dimensional electrophoresis and immune precipitation.

    • Föreliggande studie har undersökt omsorgs- och vårdmönster för ett urval (N = 77) av alla 80 år och äldre som avled i Jönköpings sjukvårdsområde 1999. Information inhämtades om kommunal omsorg och om slutenvård samt genom intervjuer med anhöriga (bortfall 16 %). En fjärdedel av de avlidna bodde hemma tills de avled, men själva dödsfallet inträffade för dem vanligen inom akutvården. Majoriteten av de avlidna (74 %) vistades sista tiden i livet – i genomsnitt 4 år – i ”särskilt boende”. Av dessa avled de allra flesta också där. En liten grupp – och främst bland hemmaboende - använder många av alla slutenvårdstillfällen och än fler av alla vårddagar, men ”vårdkarusell” är inget vanligt fenomen bland de avlidna. Mer än 9 av 10 har fått kommunal omsorg och/eller sjukvård under sitt sista levnadsår. Anhöriga är i allmänhet nöjda med dessa insatser. I regel var de också nöjda med vården i anslutning till dödsfallet.

    • DNA-based HPV assays show high sensitivity but poor specificity in detecting high-grade cervical lesions. Assays detecting mRNA of oncogenic E6/E7 show higher specificity, but lack either detection of all high-risk HPV genotypes or the capacity to specify the detected genotypes. Therefore, a real-time PCR assay detecting type-specific E6/E7 mRNA was developed and the clinical performance evaluated. 210 cervical LBC (liquid based cytology) samples from 204 women were analysed for HPV DNA and mRNA with the in house real-time PCR as well as PreTect HPV-Proofer. The sensitivity of real-time PCR mRNA-detection to detect histologically confirmed CIN2+ (cervical intraepithelial neoplasia grade 2 or higher) were 0.91, compared to 0.95 for DNA-analysis. The specificity was 0.68 compared to 0.38, and the positive predictive value (PPV) was higher for mRNA (0.67 vs 0.52) without any loss in negative predictive value (NPV). The sensitivity of the real-time PCR mRNA-test was somewhat higher than for PreTect HPV-Proofer (0.83 vs 0.75), when analysing for the same genotypes. The specificity was similar (0.76 vs 0.77). When analysing for mRNA of the eight most common genotypes in cervical cancer (HPV16, 18, 31, 33, 35, 45, 52, 58), the sensitivity to detect CIN2+ lesions was 0.87 and the specificity 0.74, with a PPV of 0.70. In conclusion, real-time PCR for detection of HPV E6/E7 mRNA transcripts can be a sensitive and specific tool in screening and investigation of cervical neoplasia. The composition of HPV-types in mRNA-testing needs to be further investigated to optimize sensitivity and specificity.
