    • BACKGROUND: Coronary atherosclerosis detected by imaging is a marker of elevated cardiovascular risk. However, imaging involves large resources and exposure to radiation. The aim was, therefore, to test whether nonimaging data, specifically data that can be self-reported, could be used to identify individuals with moderate to severe coronary atherosclerosis.METHODS AND RESULTS: We used data from the population-based SCAPIS (Swedish CardioPulmonary BioImage Study) in individuals with coronary computed tomography angiography (n=25 182) and coronary artery calcification score (n=28 701), aged 50 to 64 years without previous ischemic heart disease. We developed a risk prediction tool using variables that could be assessed from home (self-report tool). For comparison, we also developed a tool using variables from laboratory tests, physical examinations, and self-report (clinical tool) and evaluated both models using receiver operating characteristic curve analysis, external validation, and benchmarked against factors in the pooled cohort equation. The self-report tool (n=14 variables) and the clinical tool (n=23 variables) showed high-to-excellent discriminative ability to identify a segment involvement score ≥4 (area under the curve 0.79 and 0.80, respectively) and significantly better than the pooled cohort equation (area under the curve 0.76, P<0.001). The tools showed a larger net benefit in clinical decision-making at relevant threshold probabilities. The self-report tool identified 65% of all individuals with a segment involvement score ≥4 in the top 30% of the highest-risk individuals. Tools developed for coronary artery calcification score ≥100 performed similarly.CONCLUSIONS: We have developed a self-report tool that effectively identifies individuals with moderate to severe coronary atherosclerosis. The self-report tool may serve as prescreening tool toward a cost-effective computed tomography-based screening program for high-risk individuals.

    • RATIONALE: Chronic obstructive pulmonary disease (COPD) includes respiratory symptoms and chronic airflow limitation (CAL). In some cases, emphysema and impaired diffusing capacity for carbon monoxide (DLCO) are present, but characteristics and symptoms vary with smoking exposure.OBJECTIVES: To study the prevalence of CAL, emphysema and impaired DLCO in relation to smoking and respiratory symptoms in a middle-aged population.METHODS: We investigated 28,746 randomly invited individuals (52% women) aged 50-64 years across six Swedish sites. We performed spirometry, DLCO, high-resolution computed tomography (HRCT) and asked for smoking habits and respiratory symptoms. CAL was defined as post-bronchodilator forced expiratory volume in 1 second divided by forced expiratory volume (FEV1/FVC)<0.7.RESULTS: The overall prevalence was for CAL 8.8%, for impaired DLCO (DLCOCONCLUSIONS: In this large population-based study of middle-aged people, CAL and impaired DLCO were associated with common respiratory symptoms. Self-reported asthma was not associated with CAL in never-smokers. Our findings suggest that CAL in never-smokers signifies a separate clinical phenotype that may be monitored and, possibly, treated differently from smoking-related COPD.   • Exertional breathlessness related to medical conditions in middle-aged people: the population-based SCAPIS study of more than 25,000 men and women.

    • Breathlessness is common in the population and can be related to a range of medical conditions. We aimed to evaluate the burden of breathlessness related to different medical conditions in a middle-aged population.Cross-sectional analysis of the population-based Swedish CArdioPulmonary bioImage Study of adults aged 50-64years. Breathlessness (modified Medical Research Council [mMRC]≥2) was evaluated in relation to self-reported symptoms, stress, depression; physician-diagnosed conditions; measured body mass index (BMI), spirometry, venous haemoglobin concentration, coronary artery calcification and stenosis [computer tomography (CT) angiography], and pulmonary emphysema (high-resolution CT). For each condition, the prevalence and breathlessness population attributable fraction (PAF) were calculated, overall and by sex, smoking history, and presence/absence of self-reported cardiorespiratory disease.We included 25,948 people aged 57.5±[SD] 4.4; 51% women; 37% former and 12% current smokers; 43% overweight (BMI 25.0-29.9), 21% obese (BMI≥30); 25% with respiratory disease, 14% depression, 9% cardiac disease, and 3% anemia. Breathlessness was present in 3.7%. Medical conditions most strongly related to the breathlessness prevalence were (PAF 95%CI): overweight and obesity (59.6-66.0%), stress (31.6-76.8%), respiratory disease (20.1-37.1%), depression (17.1-26.6%), cardiac disease (6.3-12.7%), anemia (0.8-3.3%), and peripheral arterial disease (0.3-0.8%). Stress was the main factor in women and current smokers.Breathlessness mainly relates to overweight/obesity and stress and to a lesser extent to comorbidities like respiratory, depressive, and cardiac disorders among middle-aged people in a high-income setting-supporting the importance of lifestyle interventions to reduce the burden of breathlessness in the population.

    • Drug-induced liver injury is a common reason for drug attrition in late clinical phases, and even for post-launch withdrawals. As a consequence, there is a broad consensus in the pharmaceutical industry, and within regulatory authorities, that a significant improvement of the current in vitro test methodologies for accurate assessment and prediction of such adverse effects is needed. For this purpose, appropriate in vivo-like hepatic in vitro models are necessary, in addition to novel sources of human hepatocytes. In this report, we describe recent and ongoing research toward the use of human embryonic stem cell (hESC)-derived hepatic cells, in conjunction with new and improved test methods, for evaluating drug metabolism and hepatotoxicity. Recent progress on the directed differentiation of human embryonic stem cells to the functional hepatic phenotype is reported, as well as the development and adaptation of bioreactors and toxicity assay technologies for the testing of hepatic cells. The aim of achieving a testing platform for metabolism and hepatotoxicity assessment, based on hESC-derived hepatic cells, has advanced markedly in the last 2-3 years. However, great challenges still remain, before such new test systems could be routinely used by the industry. In particular, we give an overview of results from the Vitrocellomics project (EU Framework 6) and discuss these in relation to the current state-of-the-art and the remaining difficulties, with suggestions on how to proceed before such in vitro systems can be implemented in industrial discovery and development settings and in regulatory acceptance.

    • The results of a recent meta-analysis aroused concern about an increased risk of death associated with the use of paclitaxel-coated angioplasty balloons and stents in lower-limb endovascular interventions for symptomatic peripheral artery disease.We conducted an unplanned interim analysis of data from a multicenter, randomized, open-label, registry-based clinical trial. At the time of the analysis, 2289 patients had been randomly assigned to treatment with drug-coated devices (the drug-coated-device group, 1149 patients) or treatment with uncoated devices (the uncoated-device group, 1140 patients). Randomization was stratified according to disease severity on the basis of whether patients had chronic limb-threatening ischemia (1480 patients) or intermittent claudication (809 patients). The single end point for this interim analysis was all-cause mortality.No patients were lost to follow-up. Paclitaxel was used as the coating agent for all the drug-coated devices. During a mean follow-up of 2.49 years, 574 patients died, including 293 patients (25.5%) in the drug-coated-device group and 281 patients (24.6%) in the uncoated-device group (hazard ratio, 1.06; 95% confidence interval, 0.92 to 1.22). At 1 year, all-cause mortality was 10.2% (117 patients) in the drug-coated-device group and 9.9% (113 patients) in the uncoated-device group. During the entire follow-up period, there was no significant difference in the incidence of death between the treatment groups among patients with chronic limb-threatening ischemia (33.4% [249 patients] in the drug-coated-device group and 33.1% [243 patients] in the uncoated-device group) or among those with intermittent claudication (10.9% [44 patients] and 9.4% [38 patients], respectively).In this randomized trial in which patients with peripheral artery disease received treatment with paclitaxel-coated or uncoated endovascular devices, the results of an unplanned interim analysis of all-cause mortality did not show a difference between the groups in the incidence of death during 1 to 4 years of follow-up. Indicators that ACs can use to recognize sepsis early are beneficial for patient outcomes. Research suggests that elevated point-of-care (POC) plasma glucose and serum lactate levels may help to predict sepsis in the ambulance service (AS) setting. Study Objective:The aim of this study was to test the hypothesis that the elevation of POC plasma glucose and serum lactate levels may help to predict Sepsis-3 in the AS. Methods:A prospective observational study was performed in the AS setting of Gothenburg in Sweden from the beginning of March 2018 through the end of September 2019. The criteria for sampling POC plasma glucose and serum lactate levels in the AS setting were high or intermediate risk according to the Rapid Emergency Triage and Treatment System (RETTS), as red, orange, yellow, and green if the respiratory rate was >22 breaths/minutes. Sepsis-3 were identified retrospectively. A primary and secondary analyses were carried out. The primary analysis included patients cared for in the AS and emergency department (ED) and were hospitalized. In the secondary analysis, patients who were only cared for in the AS and ED without being hospitalized were also included. To evaluate the predictive ability of these biomarkers, the area under the curve (AUC), sensitivity, specificity, and predictive values were used. Results:A total of 1,057 patients were included in the primary analysis and 1,841 patients were included in the secondary analysis. In total, 253 patients met the Sepsis-3 criteria (in both analyses). The AUC for POC plasma glucose and serum lactate levels showed low accuracy in predicting Sepsis-3 in both the primary and secondary analyses. Among all hospitalized patients, regardless of Sepsis-3, more than two-thirds had elevated plasma glucose and nearly one-half had elevated serum lactate when measured in the AS. Conclusions:As individual biomarkers, an elevated POC plasma glucose and serum lactate were not associated with an increased likelihood of Sepsis-3 when measured in the AS in this study. However, the high rate of elevation of these biomarkers before arrival in hospital highlights that their role in clinical decision making at this early stage needs further evaluation, including other endpoints than Sepsis-3.

    • We demonstrate in this study that a single focusing multiple collector inductively coupled plasma massspectrometer (MC-ICP-MS) equipped with a hexapole gas-collision cell (GV-instrument Isoprobe) canprecisely determine the d29Si (2S.D., 0.2&) using a total Si consumption of less than 14 nmole (390 ngSi). Testing and evaluation of background, rinse time, and major matrix effects have been performed ina systematic way to establish a procedure to measure d29Si in small quantities. Chemical purificationprior to analysis is required to remove potential interferences. For data collected during a four-yearperiod, the average d29Si value of IRMM-018 relative to NBS-28 was found to be 0.95& (n ¼ 23,2S.D. 0.16&) with a 95% confidence interval (0.95 0.028&). The mean d29Si value of the Big-Batchstandard was found to be 5.50& (n ¼ 6, 2S.D. 0.26&). Although determination of the d30Simeasurements is not possible, with our current instrument we demonstrate that this system providesa fast and long-term reliable method for the analysis of d29Si in purified samples with low Siconcentration (18 mM Si).

    • The cycling of carbon on the Arctic shelves, including outgassing of CO2 to the atmosphere, is not clearly understood. Degradation of terrestrial organic carbon (OCter) has recently been shown to be pronounced over the East Siberian Arctic Shelf (ESAS), i.e. the Laptev and East Siberian Seas, producing dissolved inorganic carbon (DIC). To further explore the processes affecting DIC, an extensive suite of shelf water samples were collected during the summer of 2008, and assessed for the stable carbon isotopic composition of DIC (delta C-13(DIC)). The delta C-13(DIC) values varied between -7.2 parts per thousand to +1.6 parts per thousand and strongly deviated from the compositions expected from only mixing between river water and seawater. Model calculations suggest that the major processes causing these deviations from conservative mixing were addition of (DIC) by degradation of OCter, removal of DIC during primary production, and outgassing of CO2. All waters below the halocline in the ESAS had delta C-13(DIC) values that appear to reflect mixing of river water and seawater combined with additions of on average 70 +/- 20 mu M of DIC, originating from degradation of OCter in the coastal water column. This is of the same magnitude as the recently reported deficits of DOCter and POCter for the same waters. The surface waters in the East Siberian Sea had higher delta C-13(DIC) values and lower DIC concentrations than expected from conservative mixing, consistent with additions of DIC from degradation of OCter and outgassing of CO2. The outgassing of CO2 was equal to loss of 123 +/- 50 mu M DIC. Depleted delta C-13(POC) values of -29 parts per thousand to -32 parts per thousand in the mid to outer shelf regions are consistent with POC from phytoplankton production. The low delta C-13(POC) values are likely due to low delta C-13(DIC) of precursor DIC, which is due to degradation of OCter, rather than reflecting terrestrial input compositions. Overall, the delta C-13(DIC) values confirm recent suggestions of substantial degradation of OCter over the ESAS, and further show that a large part of the CO2 produced from degradation has been outgassed to the atmosphere.   • Non-conservative behavior of dissolved organic carbon across the Laptev and East Siberian Seas

    • Climate change is expected to have a strong effect on the Eastern Siberian Arctic Shelf (ESAS) region, which includes 40% of the Arctic shelves and comprises the Laptev and East Siberian seas. The largest organic carbon pool, the dissolved organic carbon (DOC), may change significantly due to changes in both riverine inputs and transformation rates; however, the present DOC inventories and transformation patterns are poorly understood. Using samples from the International Siberian Shelf Study 2008, this study examines for the first time DOC removal in Arctic shelf waters with residence times that range from months to years. Removals of up to 10%–20% were found in the Lena River estuary, consistent with earlier studies in this area, where surface waters were shown to have a residence time of approximately 2 months. In contrast, the DOC concentrations showed a strong nonconservative pattern in areas with freshwater residence times of several years. The average losses of DOC were estimated to be 30%–50% during mixing along the shelf, corresponding to a first-order removal rate constant of 0.3 yr−1. These data provide the first observational evidence for losses of DOC in the Arctic shelf seas, and the calculated DOC deficit reflects DOC losses that are higher than recent model estimates for the region. Overall, a large proportion of riverine DOC is removed from the surface waters across the Arctic shelves. Such significant losses must be included in models of the carbon cycle for the Arctic Ocean, especially since the breakdown of terrestrial DOC to CO2 in Arctic shelf seas may constitute a positive feedback mechanism for Arctic climate warming. These data also provide a baseline for considering the effects of future changes in carbon fluxes, as the vast northern carbon-rich permafrost areas draining into the Arctic are affected by global warming.

    • BACKGROUND: The discovery that not all patients who call for the emergency medical service (EMS) require transport to hospital has changed the structure of prehospital emergency care. Today, the EMS clinician at the scene already distinguishes patients with a time-critical condition such as stroke/transitory ischemic attack (TIA) from patients without. This highlights the importance of the early identification of stroke/TIA.AIM: To describe patients with a final diagnosis of stroke/TIA whose transport to hospital was interrupted either due to a lack of suspicion of the disease by the EMS crew or due to refusal by the patient or a relative/friend.METHODS: Data were obtained from a register in Gothenburg, covering patients hospitalised due to a final diagnosis of stroke/TIA. The inclusion criterion was that patients were assessed by the EMS but were not directly transported to hospital by the EMS.RESULTS: Among all the patients who were assessed by the EMS nurse and subsequently diagnosed with stroke or TIA in 2015, the transport of 34 of 1,310 patients (2.6%) was interrupted. Twenty-five of these patients, of whom 20 had a stroke and five had a TIA, are described in terms of initial symptoms and outcome. The majority had residual symptoms at discharge from hospital. Initial symptoms were vertigo/disturbed balance in 11 of 25 cases. Another three had symptoms perceived as a change in personality and three had a headache.CONCLUSION: From this pilot study, we hypothesise that a fraction of patients with stroke/TIA who call for the EMS have their direct transport to hospital interrupted due to a lack of suspicion of the disease by the EMS nurse at the scene. These patients appear to have more vague symptoms including vertigo and disturbed balance. Instruments to identify these patients at the scene are warranted.

The care is given in a complex context which makes prehospital care a potential high-risk activity when it comes to patient safety. Patient safety in the prehospital setting has been only sparsely investigated. The aims of the present study were 1) To investigate the incidence of adverse events (AEs) in prehospital care and 2) To investigate the factors contributing to AEs in prehospital care. Methods: We used a retrospective study design where 30 randomly selected prehospital medical records were screened for AEs each month in three prehospital organizations in Sweden during a period of one year. A total of 1080 prehospital medical records were included. The record review was based on the use of 11 screening criteria. Results: The reviewers identified 46 AEs in 46 of 1080 (4.3%) prehospital medical records. Of the 46 AEs, 43 were classified as potential for harm (AE1) (4.0, 95% CI = 2.9-5.4) and three as harm identified (AE2) (0.3, 95% CI = 0.1-0.9). However, among patients with a life-threatening condition (priority 1), the risk of AE was higher (16.5%). The most common factors contributing to AEs were deviations from standard of care and missing, incomplete, or unclear documentation. The most common cause of AEs was the result of action(s) or inaction(s) by the emergency medical service (EMS) crew. Conclusions: There were 4.3 AEs per 100 ambulance missions in Swedish prehospital care. The majority of AEs originated from deviations from standard of care and incomplete documentation. There was an increase in the risk of AE among patients who the EMS team assessed as having a life-threatening condition. Most AEs were possible to avoid. © 2019 The Author(s).

    • BACKGROUND: TP53 has been described as a prognostic factor in many malignancies, including breast cancer. Whether it also might be a predictive factor with reference to chemo- and endocrine therapy is more controversial. PATIENTS AND METHODS: We investigated relapse-free (RFS), breast cancer-corrected (BCCS) and overall survival (OS) related to TP53 status in node-positive breast cancer patients that had received polychemotherapy [cyclophosphamide, methotrexate, 5-fluorouracil (CMF)] and/or endocrine therapy (tamoxifen). Sequence analyses of the whole TP53 coding region was performed in 376 patients operated on for primary breast cancer with axillary lymph node metastases between 1984 and 1989 (median follow-up time 84 months). RESULTS: TP53 mutations were found in 105 patients (28%). We found 90 (82%) of the 110 mutations in the more frequently analysed exons 5-8, while the other 20 (18%) were located in exons 3-4 and 9-10, respectively. Univariate analyses showed TP53 to be a significant prognostic factor with regard to RFS, BCCS and OS in patients who received adjuvant CMF. CONCLUSIONS: TP53 mutations might induce resistance to certain modalities of breast cancer therapy. Sequence-determined TP53 mutation was of negative prognostic value in the total patient population and in the CMF treated patients.

    • Major depressive disorder (MDD) is highly prevalent in individuals with anorexia nervosa (AN) and is a predictor of greater clinical severity. However, there is a limited amount of evidence supporting the use of psychotropic medications for its management. A systematic scoping review was conducted to assess the current literature on brain stimulation treatments for AN with comorbid MDD, with a specific focus on MDD treatment response and weight restoration. This review was conducted according to Preferred Reporting Items for Systematic Reviews and Meta-Analyses guidelines, and the PubMed, PsycInfo, and MEDLINE databases were searched until July 2022 using specific key words related to AN and brain stimulation treatments. A total of 373 citations were identified, and 49 treatment studies that met the inclusion criteria were included in the review. The initial evidence suggests that electroconvulsive therapy, repetitive transcranial magnetic stimulation, and deep-brain stimulation may be effective in managing comorbid MDD in AN. Emerging evidence suggests that transcranial direct current stimulation may have a positive effect on body mass index in individuals with severe to extreme AN. However, there is a need for the development of better measurement techniques for assessing the severity of depression in the context of AN. Controlled trials that are adequately designed to account for these limitations are highly warranted for deep-brain stimulation, electroconvulsive therapy, and repetitive transcranial magnetic stimulation and hold promise for providing clinically meaningful results.

  • Lion Fire: Extinguishment and mitigation of fires in Li-ion batteries at sea

    • The shipping industry is facing increasing pressure to cut emissions. Diesel-electric hybrid or fully electrical propulsion systems can offer significant savings in fuel consumption and reduce emissions. However, the use of energy storage battery systems on board vessels is introducing new fire hazards and advice on suitable fire extinguishing systems and agents is desired. In a series of tests, both total compartment application water spray and water mist systems and direct injection (using several different agents) into the module were evaluated in fire tests conducted to compare different fire extinguishing approaches for a fire in a battery cell. A test compartment was constructed to simulate a battery room and a commercially available lithium-ion (Li-ion) battery cell was positioned inside a cubic box that mimicked a battery module. By heating the battery cell, combustible gases were generated, and these gases were ignited by a pilot flame inside the simulated battery module. The tests indicated that fire extinguishment of a battery cell fire inside a battery module is unlikely when using total compartment water spray or water mist fire protection systems. The water droplets are simply not able to penetrate the battery module and reach to the seat of the fire. Direct injection of the fire extinguishing agent inside the battery module is necessary. The tests also showed that agents such as water and low-expansion foam, with a high heat capacity, provide rapid cooling and fire extinguishment. The reduced water surface tension associated with low-expansion foam may improve the possibilities for water penetration whilst agents with a high viscosity may not be able to spread to the seat of the fire. Agents with less heat capacity, such as high-expansion foam and nitrogen gas, provide less cooling but fire extinguishment can still be achieved if designed correctly.

    • Objectives: Updated international guideline recommendations for AN inpatient care rely on expert opinions/observational evidence and promote extended inpatient stays, warranting investigation using higher-level ecological evidence.Methods: The study was conducted according to Guidelines for Accurate and Transparent Health Estimates Reporting (GATHER). Data encompassing 13,885 ED inpatients (5336 adolescents and 8549 adults) was retrieved from Swedish public health registries. Variables analyzed included (1) ED inpatient care opportunities, (2) unique number of ED inpatients and (3) mean length of ED-related inpatient stays in age groups 15–19 and 20–88+, across 1998–2020.Results: Mean length of inpatient stays was inversely correlated to relapse to ED-related inpatient care within the same year (p < 0.001, R-squaredadj = 0.5216 and p < 0.00001, R-squaredadj = 0.5090, in the 15–19 and 20–88+ age groups, respectively), independent of number of ED inpatients treated within a year in both age groups. Extending mean adolescent inpatient duration from 35 to 45 days was associated with a ∼30% reduction in the year-wise relapse rate.Conclusions: Mean length of ED-related inpatient treatment stays was associated with reduced relapses to inpatient care within the same year, which could be interpreted as support for recommendations to include a stabilization phase in inpatient ED treatment.

    • A peat deposit from the East European Russian Arctic, spanning nearly 10 000 years, was investigated to study soil organic matter degradation using analyses of bulk elemental and stable isotopic compositions and plant macrofossil remains. The peat accumulated initially in a wet fen that was transformed into a peat plateau bog following aggradation of permafrost in the late Holocene (similar to 2500 cal a BP). Total organic carbon and total nitrogen (N) concentrations are higher in the fen peat than in the moss-dominated bog peat layers. Layers in the sequence that have lower concentrations of total hydrogen (H) are associated with degraded vascular plant residues. C/N and H/C atomic ratios indicate better preservation of organic matter in peat material dominated by bryophytes as opposed to vascular plants. The presence of permafrost in the peat plateau stage and water-saturated conditions at the bottom of the fen stage appear to lead to better preservation of organic plant material. delta 15N values suggest N isotopic fractionation was driven primarily by microbial decomposition whereas differences in delta 13C values appear to reflect mainly changes in plant assemblages. Positive shifts in both delta 15N and delta 13C values coincide with a local change to drier conditions as a result of the onset of permafrost and frost heave of the peat surface. This pattern suggests that permafrost aggradation not only resulted in changes in vegetation but also aerated the underlying fen peat, which enhanced microbial denitrification, causing the observed 15N-enrichment.

    • The discovery of oestrogen receptor beta (ER beta/ESR2) was a landmark discovery. Its reported expression and homology with breast cancer pharmacological target ER alpha (ESR1) raised hopes for improved endocrine therapies. After 20 years of intense research, this has not materialized. We here perform a rigorous validation of 13 anti-ER beta antibodies, using well-characterized controls and a panel of validation methods. We conclude that only one antibody, the rarely used monoclonal PPZ0506, specifically targets ER beta in immunohistochemistry. Applying this antibody for protein expression profiling in 44 normal and 21 malignant human tissues, we detect ER beta protein in testis, ovary, lymphoid cells, granulosa cell tumours, and a subset of malignant melanoma and thyroid cancers. We do not find evidence of expression in normal or cancerous human breast. This expression pattern aligns well with RNA-seq data, but contradicts a multitude of studies. Our study highlights how inadequately validated antibodies can lead an exciting field astray.

    • Atherosclerotic cardiovascular disease is a major problem despite the availability of drugs that influence major risk factors. New treatments are needed, and there is growing interest in therapies that may have multiple actions. Thyroid hormone modulates several cardiovascular risk factors and delays atherosclerosis progression in humans. However, use of thyroid hormone is limited by side effects, especially in the heart. To overcome this limitation, pharmacologically selective thyromimetics that mimic metabolic effects of thyroid hormone and bypass side effects are under development. In animal models, such thyromimetics have been shown to stimulate cholesterol elimination through LDL and HDL pathways and decrease body weight without eliciting side effects. We report here studies on a selective thyromimetic [KB2115, (3-[[3,5-dibromo-4- [4-hydroxy-3-(1-methylethyl)-phenoxy]-phenyl]-amino]-3-oxopropanoic acid)] in humans. In moderately overweight and hypercholesterolemic subjects KB2115 was found to be safe and well tolerated and elicited up to a 40% lowering of total and LDL cholesterol after 14 days of treatment. Bile acid synthesis was stimulated without evidence of increased cholesterol production, indicating that KB2115 induced net cholesterol excretion. KB2115 did not provoke detectable effects on the heart, suggesting that the pharmacological selectivity observed in animal models translates to humans. Thus, selective thyromimetics deserve further study as agents to treat dyslipidemia and other risk factors for atherosclerosis. © 2007 by The National Academy of Sciences of the USA.

  • Systolic Blood Pressure Response to Exercise in Endurance Athletes in Relation to Oxygen Uptake, Work Rate and Normative Values

    • Work rate has a direct impact on the systolic blood pressure (SBP) during aerobic exercise, which may be challenging in the evaluation of the SBP response in athletes reaching high work rates. We aimed to investigate the exercise SBP response in endurance athletes in relation to oxygen uptake (VO2), work rate and to recent reference equations for exercise SBP in the general population. Endurance athletes with a left-ventricular end-diastolic diameter above the reference one performed a maximal bicycle cardiopulmonary exercise test. The increase in SBP during exercise was divided by the increase in VO2 (SBP/VO2 slope) and in Watts, respectively (SBP/W slope). The maximum SBP (SBPmax) and the SBP/W slope were compared to the predicted values. In total, 27 athletes (59% men) were included; mean age, 40 ± 10 years; mean VO2max, 50 ± 5 mL/kg/min. The mean SBP/VO2 slope was 29.8 ± 10.2 mm Hg/L/min, and the mean SBP/W slope was 0.27 ± 0.08 mm Hg/W. Compared to the predicted normative values, athletes had, on average, a 12.2 ± 17.6 mm Hg higher SBPmax and a 0.12 ± 0.08 mm Hg/W less steep SBP/W slope (p < 0.01 and p < 0.001, respectively). In conclusion, the higher SBPmax values and the less steep SBP/W slope highlight the importance of considering work rate when interpreting the SBP response in endurance athletes and suggest a need for specific normative values in athletes to help clinicians distinguish physiologically high maximal blood pressure from a pathological blood pressure response.

    • In the aftermath of natural hazards, opportunities often arise for society to make changes that prevent future losses. These changes can also have environmental impacts, increase preparedness, and contribute to sustainable development. Due to the loss of biodiversity and land degradation, there is an urgent need for actions promoting positive environmental changes. Reversing the trend of land degradation and biodiversity loss is motivated by various reasons, such as preserving ecosystem functions and services that offer protection against natural hazards and reducing drivers of climate change. However, due to land degradation, loss of ecosystems, and human settlements in hazard-prone areas, it is also necessary to take actions that reduce the risk of tsunamis by improving preparedness. Improved preparedness from a people-centered perspective is a crucial aspect of sustainable development.The four papers that make up this thesis present and apply two new frameworks— one to assess the environmental effect of human-made societal changes, and one to evaluate the maintenance of a TEWS (Tsunami Early Warning System) over time. The two frameworks capture two aspects of change after a natural hazard: i) environmental impacts and ii) Disaster Risk Reduction (DRR). The thesis focuses particularly on time, a common aspect in both frameworks.The results indicate that changes triggered by natural hazards have a dual nature in that their impact on the environment can be both positive and negative. The thesis also provides evidence that past natural hazards can create a delayed window, which can be used as an argument for nature conservation during the establishment of national parks.The framework for evaluating the maintenance of a TEWS shows how difficult it is to operate all parts of the warning chain over time, from tsunami detection to evacuation. If parts of the chain are not maintained, the warning system may not be reliable.

    • To further explore the extent of structural large-scale variation in the human genome, we assessed copy number variations (CNVs) in a series of 71 healthy subjects from three ethnic groups. CNVs were analyzed using comparative genomic hybridization (CGH) to a BAC array covering the human genome, using DNA extracted from peripheral blood, thus avoiding any culture-induced rearrangements. By applying a newly developed computational algorithm based on Hidden Markov modeling, we identified 1,078 autosomal CNVs, including at least two neighboring/overlapping BACs, which represent 315 distinct regions. The average size of the sequence polymorphisms was approximately 350 kb and involved in total approximately 117 Mb or approximately 3.5% of the genome. Gains were about four times more common than deletions, and segmental duplications (SDs) were overrepresented, especially in larger deletion variants. This strengthens the notion that SDs often define hotspots of chromosomal rearrangements. Over 60% of the identified autosomal rearrangements match previously reported CNVs, recognized with various platforms. However, results from chromosome X do not agree well with the previously annotated CNVs. Furthermore, data from single BACs deviating in copy number suggest that our above estimate of total variation is conservative. This report contributes to the establishment of the common baseline for CNV, which is an important resource in human genetics.

    • A 160 (525 ft) m high asphalt concrete core rockfill dam is developed in an area of very high seismicity in Cetin, Turkey. Topographical constraints of the dam footprint impose a steep outer geometry of the dam, requiring the geotechnical properties of the rockfill to comply with certain minimum limits. Test blasting in a potential quarry and rockfill rolling compaction trials suggest that the tested gneiss/schist rock material to a large extent is weathered and weak. The friction angle of the rockfill is a decisive parameter for both the static and dynamic stability of the dam slopes and especially for the deformation of the dam crest due to strong earthquakes. The suggested rock fill material has therefore undergone detailed testing. To this end, large-scale shear tests with a 1000 mm diameter sample cylinder have been carried out at the Luleå Technical University in Sweden.

    • Recent sediments from the Gotland Deep display enrichments in reactive Fe, associated with elevated Fe/Al ratios and light Fe isotopic signatures of the bulk sediments that are indicative of euxinic (anoxic and sulfidic) conditions. These enrichments can be explained by the Fe shuttle model where benthic Fe is transported from the shelf to the euxinic basin and transferred to the sediments by pyrite precipitation in the sulfidic water. The data provide evidence that the Fe shuttle at present results in accumulations of Fe that are larger compared to Fe enrichments during the Litorina Sea stage in the Gotland Deep probably caused by an increase of the benthic Fe flux from the shelf to the basin. The derived Fe enrichments are also larger compared to those in recent Black Sea sediments, which likely reflects the larger shelf to basin ratio of the Gotland Deep compare to the Black Sea. The Fe isotope data show no correlation with the organic C content of the samples indicating that the negative Fe isotope signatures are not associated with organic materials, as was suggested as an alternative explanation for the origin of the isotopically light Fe in sediments from the Litorina Sea stage. Conversely, pyrites carry the negative Fe isotopic signature of the sediments, which supports the Fe shuttle model. Variations in the abundance and Fe isotopic signature of reactive Fe and pyrite with depth suggest that syngenetically formed pyrite in the sulfidic water column has a less negative Fe isotopic composition compared to diagenetically produced pyrite.

    • Holocene sediments from the Gotland Deep basin in the Baltic Sea were investigated for their Fe isotopic composition in order to assess the impact of changes in redox conditions and a transition from freshwater to brackish water on the isotope signature of iron. The sediments display variations in delta Fe-56 (differences in the Fe-56/Fe-54 ratio relative to the IRMM-14 standard) from -0.27 +/- 0.09 parts per thousand to +0.21 +/- 0.08 parts per thousand. Samples deposited in a mainly limnic environment with oxygenated bottom water have a mean delta Fe-56 of +0.08 +/- 0.13 parts per thousand, which is identical to the mean Fe isotopic composition of igneous rocks and oxic marine sediments. In contrast, sediments that formed in brackish water under periodically euxinic conditions display significantly lighter Fe isotope signatures with a mean delta Fe-56 of -0.14 +/- 0.19 parts per thousand. Negative correlations of the delta Fe-56 values with the Fe/Al ratio and S content of the samples suggest that the isotopically light Fe in the periodically euxinic samples is associated with reactive Fe enrichments and sulfides. This is supported by analyses of pyrite separates from this unit that have a mean Fe isotopic composition of -1.06 +/- 0.20 parts per thousand for delta Fe-56. The supply of additional Fe with a light Fe isotopic signature can be explained with the shelf to basin Fe shuttle model. According to the Fe shuttle model, oxides and benthic ferrous Fe that is derived from dissimilatory iron reduction from shelves is transported and accumulated in euxinic basins. The data furthermore suggest that the euxinic water has a negative dissolved delta Fe-56 value of about -1.4 parts per thousand to -0.9 parts per thousand. If negative Fe isotopic signatures are characteristic for euxinic sediment formation, widespread euxinia in the past might have shifted the Fe isotopic composition of dissolved Fe in the ocean towards more positive delta Fe-56 values.

  • 231Pa, 230Th and 232Th as tracers of deep water circulation and particle transport : Insights from the Mediterranean Sea and the Arctic Ocean

    • The naturally occurring U and Th-series radionuclides have shown to have a considerable importance for the understanding of biogeochemical processes on Earth and in the ocean. In this thesis, the isotopes 230-thorium (230Th), 232-thorium (232Th) and 231-protactinium (231Pa) are used as tracers of the transport and scavenging of marine particles and water circulation. Pa and Th are particle reactive elements, which makes the production, transport and distribution of Pa and Th key factors for our understanding of the origin, fate and distribution of marine particles in the oceans. This thesis explores the distribution of 231Pa, 230Th and 232Th in two different ocean continental margin environments. In particular, the relative influence of water circulation and particles on the 231Pa, 230Th and 232Th distributions in the Arctic Ocean and Mediterranean Sea was investigated. 231Pa, 230Th and 232Th were analyzed in particles and seawater collected in the Mediterranean Sea during the MedSeA-GA04-S cruise along the GEOTRACES section GA04S and in the Arctic Ocean during the PS94 GN04 ARK-XXIX/3 along the GEOTRACES section GN04.One of the important findings of this thesis was the low fractionation between 231Pa and 230Th in the Mediterranean Sea, contrasting what is observed in the open ocean. Additionally, the observed depth profiles of Pa-Th allowed the identification of deep water convection and ventilation in the Western and Eastern Basins, respectively. Moreover, the particle settling speed was reevaluated to ~500 – 1000 m/y.In the Arctic Ocean, scavenging onto particles derived from hydrothermal activity was producing relatively low F-factors (FTh/Pa ~ 10), while higher values were observed in deep waters (FTh/Pa ~ 20). Additionally, the hydrothermal particles in the Nansen interior produce lower FTh/Pa values compared to FTh/Pa observed at the Nansen continental margin. Application of a boundary scavenging model revealed the importance of 230Th scavenging at the continental margin along the Nansen Basin, hereafter the Nansen margin, and advocate for the advection of 231Pa into the Atlantic Ocean. As the ocean margin was included in this model, a particle settling speed of 600 m/y was obtained at the Nansen margin.Moreover, this thesis includes an inter-comparison of dissolved and particulate 231Pa, 230Th and 232Th measurements between four laboratories of the GEOTRACES community. This comparison was conducted to provide detailed descriptions of various chemical procedures used for Pa-Th analysis and to provide a measure of consistency between the laboratories. Results demonstrated that participating labs can determine concentrations of dissolved 230Th and 231Pa in deep water (below 500 m depth) that are internally consistent within 4 % of the mean values. Analysis of particulate 231Pa, 230Th and 232Th allowed the highlighting of an incomplete Pa dissolution problem with our initial leaching procedure, a problem solved by measuring aliquots of particulate samples at two labs. However, in the present work, consistent particulate 231Pa concentrations as low as ~ 0.002 fg/kg were obtained. Overall, it suggests an improvement of the results consistency compared to the previous GEOTRACES intercalibration exercise.

The distribution of dissolved 231Pa and 230Th in the Arctic Ocean deviates from the linearincrease expected from reversible scavenging. Higher 232Th concentrations were observed at the shelf, slope andin surface waters in the deep basin, pointing at lithogenic sources. Fractionation factors (FTh/Pa) observed at theNansen margin were higher compared to FTh/Pa in the central Nansen Basin, possibly due to the residual occurrenceof hydrothermal particles in the deep central Nansen Basin. Application of a boundary scavengingmodel quantitatively accounts for the dissolved and particulate 230Th distributions in the Nansen Basin.Modelled dissolved 231Pa distributions were largely overestimated, which was attributed to the absence of incorporation of water exchange with the Atlantic Ocean in the model. 231Pa/230Th ratios of the suspended particlesof the Nansen Basin were below the 231Pa/230Th production ratio, but top-core sediments of the Nansenmargin and slope have high 231Pa/230Th-ratios, suggesting that scavenging along the Nansen margin partly actsas a sink for the missing Arctic 231Pa.

    • 231Pa, 230Th and 232Th were analyzed in unfiltered sea water samples (n = 66) and suspended particles (n = 19) collected in the Mediterranean Sea during the MedSeA-GA04-S cruise along the GEOTRACES section GA04S and used to investigate mechanisms controlling the distribution and fractionation of Pa and Th in an ocean margin environment. 231Pa and230Th are particle reactive radionuclides and are often used astracers of processes such as boundary scavenging, particle transport and ocean circulation. The depth profiles of total 231Pa and 230Th concentrations in the Mediterranean Sea displayed non-linear shapes. Higher total 232Th concentrations were observed at the straits and in deepwaters pointing at lithogenic sources. Fractionation factors FTh/Pa ranged from 1.4 to 9. Application of a box-model illustrated that 94 % of the 231Pa and almost all of the 230Th (99.9 %) produced in the Mediterranean Sea is removed to the sediment by scavenging. The negligible export of 230Th to the Atlantic Ocean, leads to a reevaluation of the mean settling speed of the filtered particles, which is now estimated to 500-1000 m/y. The low FTh/Pa fractionation factors are attributed to the efficient scavenging and lack of transport of 231Pa to the Atlantic Ocean.

The paper will focus on the studies and modeling of biomass supply, harvesting and transport and gasification as well as on the system studies. The use of the gas in the metallurgical application is briefly described as background.

The production chain Biomass production and distribution -Gasification-DRI production-DRI use is investigated in four work packages:WP1: Biomass supply: A large amount of Biomass has to be delivered into a single site to exchange a large amount of fossil reductant. It is important to use forestry by- products as a major part of round wood is reserved for other uses. Harvesting, logistics and economics have to be considered. Available data were collected and used to make a system model on harvesting treatment and transport. The simulations indicated that the supply of residuals is possible but will need material from a large part of the north Sweden wood area. WP2: Gasification. The aim is to use to produce hot gas that can be used directly. Pilot experiments are carried out using oxygen in an entrained flow gasifier. WP3: Metallurgical processes. Reduction tests are carried out with gas that can be produced in the gasifier. The limitations of the gas content are studied as well as the effect on DRI. Also the suitability of the DRI product is evaluated WP4: Process integration. A system model is built using the results from work packages 1-3 and used for technical economic optimization the whole system harvesting-transport-gasifier-direct reduction- use of DRI. The process chain is technically possible; however there are problems to be solved, e.g., gas quality vs. demands from DRI process, Biomass supply and logistics. The result is important to evaluate for industrial application, but also to get information of the effect of different governmental control instruments.

    • Measurements from the SWERUS-C3 and ISSS-08 Arctic expeditions were used to calibrate and validate a new physical-biogeochemical model developed to quantify key carbon cycling processes on the East Siberian Arctic Shelf (ESAS). The model was used in a series of experimental simulations with the specific aim to investigate the pathways of terrestrial dissolved and particulate organic carbon (DOCter and POCter) supplied to the shelf. Rivers supply on average 8.5 Tg C yr−1 dissolved inorganic carbon (DIC), and further 8.5 and 1.1 Tg C yr−1 DOCter and POCter respectively. Based on observed and simulated DOC concentrations and stable isotope values (δ13CDOC) in shelf waters, we estimate that only some 20 % of the riverine DOCter is labile. According to our model results, an additional supply of approximately 14 Tg C yr−1 eroded labile POCter is however required to describe the observed stable isotope values of DIC (δ13CDIC). Degradation of riverine DOCter and POCter results in a 1.8 Tg C yr−1 reduction in the uptake of atmospheric CO2, while degradation of eroded POCter results in an additional 10 Tg C yr−1 reduction. Our calculations indicate nevertheless that the ESAS is an overall small net sink for atmospheric CO2 (1.7 Tg C yr−1). The external carbon sources are largely compensated by a net export from the shelf to the Arctic Ocean (31 Tg C yr−1), and to a smaller degree by a permanent burial in the sediments (2.7 Tg C yr−1).

However, the characteristics of chemically reactive Fe remain poorly constrained, especially in large Arctic rivers, which drain landscapes highly susceptible to climate change and carbon cycle alteration. The aim of this study was a detailed characterisation (size, mineralogy, and speciation) of riverine Fe-bearing particles (> 0.22 µm) and colloids (1 kDa – 0.22 µm) and their association with organic carbon (OC), in the Lena River and tributaries, which drain a catchment almost entirely underlain by permafrost. Samples fromthe main channel and tributaries representing watersheds that span a wide rangein topography and lithology were taken after the spring flood in June 2013 and summer baseflow in July 2012. Fe-bearing particles were identified, usingTransmission Electron Microscopy, as large (200 nm – 1 µm) aggregates of smaller (20 nm - 30 nm) spherical colloids of chemically-reactive ferrihydrite.In contrast, there were also large (500 nm – 1 µm) aggregates of clay (illite) particles and smaller (100 - 200 nm) iron oxide particles (dominantly hematite) that contain poorly reactive Fe. TEM imaging and Scanning Transmission X-raymicroscopy (STXM) indicated that the ferrihydrite is present as discrete particles within networks of amorphous particulate organic carbon (POC) and attached to the surface of primary produced organic matter and clay particles.Together, these larger particles act as the main carriers of nanoscale ferrihydrite in the Lena River basin.  The chemically reactive ferrihydrite accounts for on average 70 ± 15 % of the total suspended Fe in the Lena River and tributaries. These observations place important constraints on Fe and OC cycling in the Lena River catchment area and Fe-bearing particle transport to the Arctic Ocean.

    • Iron is an important mediator of biotic and abiotic processes on the Earth’s surface, being an electron acceptor in organic matter degradation, a surface for organic matter and trace element adsorption, and a required element for enzymatic processes during primary production. Yet, the role of iron as a mediator of carbon and trace element cycling in high latitude, permafrost-dominated regions remains poorly investigated. The aim of this study was to characterise the chemical reactivity (by size separation, microscopy and spectroscopy) and sources (by isotope composition) of Fe in the Lena River and major tributaries, spanning a wide range in lithology, topography and climate. The Fe transported in the Lena River and major tributaries carries an integrated signal of Fe weathering processes across the permafrost-dominated terrain.A spatial sample set was collected during the post-spring flood period (July 2012, June 2013), from the main channel and tributaries draining contrasting topography and permafrost extent. Across the basin, Fe is mainly transported as chemically reactive ferrihydrite that spans the particulate (> 0.22 µm) and colloidal (0.22 µm – 1 kDa) fractions. The remaining Fe transported as poorly reactive detrital Fe in clays and crystalline oxides. Fe is transported in larger size fractions than the dissolved OM showing that Fe is not a major carrier of DOM. Nano-sized ferrihydrite was attached to OM in the particulate fraction, evidence of a Fe – OM particle association in the Lena River basin.Ferrihydrite shows distinct isotope values in particulate and colloidal fractions, showing that there is a difference in isotopic composition between different size fractions of the same mineral. A conceptual model was developed to understand ferrihydrite formation in the riparian zone of the Lena River and tributaries.  Particulate ferrihydrite has isotope values lower than crustal values resulting from redox and organic-ligand promoted mineral dissolution and precipitation of Fe(II)aq to form coatings of ferrihydrite on particles in the riparian zone.  Ferrihydrite colloids span a wider range of isotope values, higher than Fe particles, resulting from variations in the size and isotope composition of the Fe(II)aq pool transported in soil groundwaters, and the isotope fractionation factor for Fe oxidation and organic complexation.A temporal sample set was collected in the main channel between September 2012 – March 2013 and every three days during May 2015. The colloidal Fe shows distinct seasonal Fe isotope signatures and Fe fluxes, with isotope values lower than crustal during winter baseflow, overlying crustal values during spring flood and higher than crustal values during summer, attributed to changing sources and thus conditions for isotope fractionation. The combined understanding of Fe reactivity and isotope composition allows us to isolate the dominant sources of Fe entering the Lena River.

    • Large Arctic rivers are an important source of iron (Fe) to the Arctic Ocean, though seasonal variations in the terrestrial source and supply of Fe to the ocean are unknown. To constrain the seasonal variability, we present Fe concentrations and isotopic compositions (δ56Fe) for particulate (>0.22 µm) and colloidal (<0.22 µm–1 kDa) Fe from the Lena River, NE Russia. Samples were collected every month during winter baseflow (September 2012–March 2013) and every 2–3 days before, during and after river ice break-up (May 2015).Iron in particles have isotope ratios lower than crustal values during winter (e.g., δ56FePart = −0.37 ± 0.16‰), and crustal-like values during river ice break-up and spring flood (e.g., δ56FePart = 0.07 ± 0.08‰), indicating a change in the source of particulate Fe between winter and spring flood. Low isotope values are indicative of mineral dissolution, transport of reduced Fe in sub-oxic, ice-covered sub-permafrost groundwaters and near-quantitative precipitation of Fe as particles. Crustal-like isotopic compositions result from the increased supply of detrital particles from riverbank and soil erosion during river ice break-up and flooding. Iron colloids (<0.22 μm) have δ56Fe values that are comparable to or lower than crustal values during winter (e.g., δ56FeCol = −0.08 ± 0.05‰) but similar to or higher than crustal values during spring flood (e.g., δ56FeCol = +0.24 ± 0.11‰). Low δ56Fe ratios for colloidal Fe during winter are consistent with precipitation from isotopically light Fe(II)aq transported in sub-permafrost groundwaters. Higher colloidal δ56Fe ratios during the spring flood indicate that these colloids are supplied from surface soils, where Fe is fractionated via oxidation or organic carbon complexation, similar to during summer. Approximately half of the annual colloidal Fe flux occurs during spring flood while most of the remaining colloidal Fe is supplied during summer months. The total amount of colloidal Fe transported during winter was relatively low. The seasonal variation in colloidal Fe isotope values may be a useful tool to trace the source of colloidal Fe to the Arctic Ocean and monitor future changes in the sources and supply of Fe from the permafrost landscape to the Lena River basin.

However, the sources of Fe-bearing particles in permafrost-dominated systems and the mechanisms driving this supply of Fe are poorly resolved. Here, Fe isotope ratios were used to determine the sources of Fe-bearing particles and colloids in the Lena River and tributaries. In samples collected after the spring floods, Fe-bearing particles (>0.22 mu m) carried similar to 70% of the Fe and have isotope ratios that are lower than, or similar to that of the continental crust. These particles are composed of a leachable Fe fraction of largely ferrihydrite, with isotope values of -1.40 parts per thousand to -0.12 parts per thousand, and a fraction of clays and Fe oxides with continental crust values. Co-existing Fe-bearing colloids (<0.22 mu m), composed mainly of ferrihydrite, have higher isotope values, of -0.22 parts per thousand to +1.83 parts per thousand. A model is proposed in which soil mineral weathering generates aqueous Fe with lower delta Fe-56 values. During transport, a small fraction of the dissolved Fe is precipitated as colloidal ferrihydrite with higher delta Fe-56 values. Most of the Fe is precipitated onto mineral grains in oxic riparian zones, with the delta Fe-56 values largely generated during weathering. Groundwater discharge and riparian erosion supply the colloids and coated particles to the rivers. The differences between delta Fe-56 values in leachates and detrital grains in Fe-bearing particles agree with values determined in mineral dissolution experiments and in Fe accumulation horizons in soils. The difference in delta Fe-56 values between leachates and colloids reflects isotope fractionation during incremental Fe(III)(aq) precipitation and Fe-OC complexation during transport towards the riparian zone. Overall, the Fe isotope values of riverine particles and colloids reflect processes that occur during mineral dissolution, transport, and secondary mineral formation in permafrost soils.

  • Electronic structure of novel low band gap conjugated polymers

    • The electronic structures of two novel low band gap conjugated polymers have been studied. Both materials are conjugated alternating copolymers based on fluorene units and low band gap donor-acceptor-donor units. The emphasis in this study has been to study the valence and the conduction bands. In particular the degree of localization or delocalization along the polymers and the symmetry of these states have been studied, since these features can be related to their transport properties. The main experimental part of this work is photoelectron spectroscopy, near-edge X-ray photon absorption and resonant photoemission. These techniques have been used to probe the frontier electronic structure of these systems. The experimental results are interpreted with the help of density functional theory calculations. The valence bands are dispersed originating from orbitals delocalized along the polymer chain shile the conduction bands are more flat, as they are derived from orbitals localized on the acceptor units. This band structure would predict good hole transporting properties but poor electron transport.

    • The catalytic oxidation of carbon by molecular oxygen was studied using C/Pt, Pt/C, Pt/Al2O3/C, Pt/CeO2/C, Al2O3/C and CeO2/C model samples prepared by hole-mask colloidal lithography. By this technique the degree of contact between platinum and carbon was controlled with high precision. The oxidation of carbon was monitored using atomic force microscopy and scanning electron microscopy. The results show that Pt in direct contact with carbon catalyzes the oxidation of carbon by spillover of dissociated oxygen from Pt to carbon. By physically separating Pt and carbon with a 10 nm thin spacer layer of Al2O3, the oxygen spillover was entirely blocked. However, through a corresponding spacer layer of CeO2, carbon oxidation was still observed, either by oxygen spillover from Pt to carbon or directly dissociated on the ceria, although at a slower rate compared to the case with no spacer layer between Pt and carbon.

    • The fate of the cell is governed by interactions among proteins, nucleic acids, and other biomolecules. It is vital to look at these interactions in a cellular environment if we want to increase our understanding of cellular processes. Herein we will describe how the in situ proximity ligation assay (in situ PLA) can be used to visualize protein interactions in fixed cells and tissues. In situ PLA is a novel technique that uses DNA, together with DNA modifying processes such as ligation, cleavage, and polymerization, as tools to create surrogate markers for protein interactions of interest. Different in situ PLA designs make it possible not only to detect protein-protein interactions but also post-translational modifications and interactions of proteins with nucleic acids. Flexibility in DNA probe design and the multitude of different DNA modifying enzymes provide the basis for modifications of the method to make it suitable to use in many applications. Furthermore, examples of how in situ PLA can be combined with other methods for a comprehensive view of the cellular activity status are discussed.

    • Secretoneurin is a neuropeptide potentially involved in migration of eosinophils, monocytes, and dendritic cells. Whether secretoneurin is present in the human airway mucosa and whether it is released at ongoing allergic airway inflammation is currently unknown. In patients with allergic rhinitis, we have explored the occurrence of secretoneurin in nasal mucosal biopsies and lavage fluids before and during natural allergen exposure. Immunohistochemical analysis revealed an abundance of nerves displaying secretoneurin immunoreactivity, which were distributed predominantly around blood vessels and submucosal glands. A majority of nerve fibers containing vesicular acetylcholine transporter, tyrosine hydroxylase, calcitonin gene–related peptide, and vasoactive intestinal peptide were also secretoneurin-immunoreactive, indicating a localization of secretoneurin in cholinergic, adrenergic, and sensory nerves. Lavage fluid levels of secretoneurin were increased at allergen exposure (p < 0.01–0.05). Levels of secretoneurin did not correlate with eosinophil cationic protein ({rho} = 0.1, p = 0.7). We conclude that secretoneurin has a widespread occurrence in nasal mucosal nerves of patients with seasonal allergic rhinitis and that increased nasal lavage fluid levels of secretoneurin may characterize ongoing allergen exposure. These data favor a role of secretoneurin in the local traffic of immune cells in human airway mucosa.
