    • With the publication of The Rise of the Creative Class by Richard Florida in 2002, the ‘creative city’ became the new hot topic among urban policymakers, planners and economists. Florida has developed one of three path-breaking theories about the relationship between creative individuals and urban environments. The economist Åke E. Andersson and the psychologist Dean Simonton are the other members of this ‘creative troika’. In the Handbook of Creative Cities, Florida, Andersson and Simonton appear in the same volume for the first time. The expert contributors in this timely Handbook extend their insights with a varied set of theoretical and empirical tools. The diversity of the contributions reflect the multidisciplinary nature of creative city theorizing, which encompasses urban economics, economic geography, social psychology, urban sociology, and urban planning. The stated policy implications are equally diverse, ranging from libertarian to social democratic visions of our shared creative and urban future.

  • Method development in inverse modeling applied to supercritical fluid extraction of lipids

    • Modeling of the supercritical fluid extraction of solid materials is an important aspect in order to understand and predict the process. A comparison of two empirical models, two semi-empirical models and two mechanistic models is performed using calibration of single experiments. It is concluded that the best fit is obtained using a simple empirical expression. Furthermore, single calibrations did not generate reliable parameters with physical meaning and a methodology is proposed for inverse modeling with complete calibration using several experiments. The experimental dataset contained 29 extractions of lipids from crushed linseeds with varying temperatures, pressures and flow rates. A general rate model and a proposed extension of the hot ball model were evaluated for this purpose. The methodology includes data acquisition, model structure estimation, model calibration and a cross-validation. In general, it was found that the solubility model of Sovová outperformed the other evaluated correlations, and for the general rate model the Toth partition isotherm was also found in the top model structures. However, no generalization could be made regarding the correlations describing the Nernst diffusion layer and diffusivity.

    • The fundamentals of analyte extractable fraction, solubility, partitioning and mass transfer resistance in supercritical fluid extraction were studied using inverse modeling. These phenomena are essential for understanding, predicting and optimizing the supercritical fluid extraction process. Carotenoids, chlorophyll A, ergosterol and total lipids were extracted from the microalgae Chlorella sp. The analytes were measured continuously in-line and on-line using UV/Vis absorption spectroscopy measurements and by evaporative light scattering detection. Various pressures, temperatures, flow rates and fractions of ethanol as a co-solvent were evaluated. The extractable fraction of carotenoids, chlorophyll A and total lipids were dependent on the co-solvent fraction in the extraction phase. The additional amount that could be extracted by using more co-solvent followed a normal distribution, indicating that analytes should not simply be categorized into weakly or strongly bound. The characteristics of diminishing extraction rates over time was accounted for by analyte partitioning rather than intra-particle diffusion limitations.

  • Circulating Biomarkers in Patients with Head and Neck Cancer and the Influence of Cigarette Smoking

    • Head and neck cancer (HNC) is a collective name for heterogeneous tumors located in the head and neck regions for which smoking, alcohol and human papillomavirus (HPV) are documented risk factors. The survival of HNC patients has only improved marginally during the last decade. The most important prognostic factors are tumor size, local spread and distant metastases, tumor node metastasis (TNM) staging. Prognostic biomarkers are needed as a complement to TNM staging.The aim for this thesis was to investigate rapid and low cost blood based biomarkers which could indicate the risk of HNC, recurrence of the disease or the survival of HNC patients. Furthermore, the aim was to examine how cigarette smoking influences the levels of biomarkers.In paper I, a possible role of plasma cytokines or proteins associated with immune response or inflammation, as biomarkers for the survival of HNC patients was investigated. Higher levels of C-reactive protein (CRP) and tumor necrosis factor alpha (TNF-α) were detected in plasma of the patients compared with the levels in the controls. The elevated levels of these two biomarkers detected in patients were associated with decreased survival.In paper II, the influence of 45 single nucleotide polymorphisms (SNPs) located in 41 genes associated with cell cycle progression, cell death, DNA repair or immune response on cancer risk, tumor recurrence and survival in HNC patients were investigated. SNPs in immune response genes were associated with risk for HNC, an elevated risk for recurrence and a decreased survival in HNC patients.In paper III, the influence of cigarette smoking on levels of inflammatory cells, proteins or cytokines/chemokines, microRNAs (miRNAs) and SNPs was analysed in healthy smokers and non-smokers. Higher levels of total white blood cells (WBCs), neutrophils, monocytes, lymphocytes, neutrophil to lymphocyte ratio (NLR), CRP, monocyte chemoattractant protein- 1 (MCP-1) and interferon gamma (IFN-γ) were detected in smokers compared to non-smokers and indicate an inflammatory response. Also, a lower level of oncomiRNA miR-21was detected in smokers. This alteration, in combination with the elevated levels of IFN-γ in smokers could be a protective response to cigarette smoke. The higher levels of IFN-γ in smokers compared to non-smokers were however only detected in individuals with SNP rs2069705 genotype AG/GG. This indicates a genetic association of the levels of IFN-γ.In paper IV, the separate effects of cigarette smoking and HNC on inflammatory or immune biomarkers and the impact of high risk human papillomavirus, age and gender were investigated. Comparisons of circulating levels of WBCs and its subpopulations, plasma proteins or cytokines/chemokines between smoking and non-smoking patients, smoking and non-smoking controls and between the patient and control groups were analysed. Smoking had highest impact on elevated levels of WBCs, IFN-γ and MCP-1, and HNC had highest impact on elevated levels of neutrophils, monocytes, NLR, CRP, macrophage inflammatory protein 1 beta and TNF-α.In conclusion, host immune response associated parameters could be suitable as biomarkers for the risk of HNC, risk of recurrence or in predicting survival of HNC patients. This thesis show that HNC are associated with systemic inflammatory response and upregulated CRP and TNF-α is related to shorter survival in HNC patients. Additionally, SNPs in immune response genes such as rs1800629 in the TNF-α gene indicates a risk for HNC or an elevated risk for recurrence and a decreased survival in HNC patients. These rapid and low cost blood based biomarkers could be used in combination or as a supplement to established biomarkers in the clinic for a more personalized treatment modality.

    • Syftet med artikeln är att lyfta fram om, och i sådant fall på vilka sätt, en specifik strukturell modell kan utgöra stöd när elever utforskar matematiska strukturer i ekvationer. Artikeln bygger på en empirisk forskningsstudie där elever utforskade matematiska strukturer med stöd av modellen, vilken är avsedd att visualisera strukturer. Lärare och forskare arbetade i en kollaborativ och intervenerande studie i iterativa processer. Sammantaget 149 elever från grundskolans årskurser 3, 8 och 9 deltog i filmade forskningslektioner utifrån forskningsansatsen learning study. Lektionerna designades med inspiration från ramverket lärandeverksamhet och eleverna utmanades i ett teoretiskt arbete. Analysen utfördes utifrån tematisk ansats och två kvalitativt skilda kärnteman identifierades: Formulär respektive Lärandemodell. I analysen framträdde att undervisningen behöver vara tillräckligt utmanande för att eleverna ska finna modellen meningsfull. Undervisningen behöver möjliggöra för eleverna att urskilja relationer mellan alla tal i en ekvation, där relationerna kan beskrivas som en del-helhetsstruktur.

  • More-Than-Two-Parent Families : Displaying Legitimate Parenthood in Swedish Media

    • Norms concerning family formation are generally based on ideals of coupled love and the two-parent-family, however, family practices frequently go beyond these norms. Families consisting of more than two parents that are co-parenting have only been studied to a small extent. Analysing Swedish newspaper and magazine articles on more-than-two-parent families between 1992 and 2016 we ask: How are more-than-two-parent families displayed in Swedish media stories? Are they portrayed as legitimate families, and if so, how is this legitimacy discursively constructed? What role does recognition play in the media stories and how is it negotiated in the narratives? We use the concepts display (Finch 2007) and recognition/redistribution (Fraser 1998; 2003) in exploring the significance that recognition and legitimacy have for the depiction of families with more than two parents in the media material. The display of more-than-two-parent families in the Swedish media stories analysed is generally characterised by repertoires of modern family life, of love and intimacy and responsible and successful parenting. These repertoires are used to display the families as normal, modern, and legitimate. In addition to the repertoires mentioned, there are repertoires of importance of geographical location, of strategies and of critique of current legislation that further emphasise the legitimacy of the more-than-two-parent families in contrast to an outdated legislation that forces these legitimate families to strategise their intimate relationships. Despite several of the people interviewed being described as polyamorous or gay/lesbian, there are no tendencies in the empirical material to motivate the need for rights based on an essentialised polyamorous identity; rather, the focus is on the fact that it is the practical care relations that need to be protected.

  • Phase-contrast MRI volume flow - a comparison of breath held and navigator based acquisitions

    • Background: Magnetic Resonance Imaging (MRI) 2D phase-contrast flow measurement has been regarded as the gold standard in blood flow measurements and can be performed with free breathing or breath held techniques. We hypothesized that the accuracy of flow measurements obtained with segmented phase-contrast during breath holding, and in particular higher number of k-space segments, would be non-inferior compared to navigator phase-contrast. Volumes obtained from anatomic segmentation of cine MRI and Doppler echocardiography were used for additional reference. Methods: Forty patients, five women and 35 men, mean age 65 years (range 53-80), were randomly selected and consented to the study. All underwent EKG-gated cardiac MRI including breath hold cine, navigator based free-breathing phase-contrast MRI and breath hold phase-contrast MRI using k-space segmentation factors 3 and 5, as well as transthoracic echocardiography within 2 days. Results: In navigator based free-breathing phase-contrast flow, mean stroke volume and cardiac output were 79.7 +/- 17.1 ml and 5071 +/- 1192 ml/min, respectively. The duration of the acquisition was 50 +/- 6 s. With k-space segmentation factor 3, the corresponding values were 77.7 ml +/- 17.5 ml and 4979 +/- 1211 ml/min (p = 0.15 vs navigator). The duration of the breath hold was 17 +/- 2 s. K-space segmentation factor 5 gave mean stroke volume 77.9 +/- 16.4 ml, cardiac output 5142 +/- 1197 ml/min (p = 0.33 vs navigator), and breath hold time 11 +/- 1 s. Anatomical segmentation of cine gave mean stroke volume and cardiac output 91.2 +/- 20.8 ml and 5963 +/- 1452 ml/min, respectively. Echocardiography was reliable in 20 of the 40 patients. The mean diameter of the left ventricular outflow tract was 20.7 +/- 1.5 mm, stroke volume 78.3 ml +/- 15.2 ml and cardiac output 5164 +/- 1249 ml/min. Conclusions: In forty consecutive patients with coronary heart disease, breath holding and segmented k-space sampling techniques for phase-contrast flow produced stroke volumes and cardiac outputs similar to those obtained with free-breathing navigator based phase-contrast MRI, using less time. The values obtained agreed fairly well with Doppler echocardiography while there was a larger difference when compared with anatomical volume determinations using SSFP (steady state free precession) cine MRI.

  • Polyamorous Parenthood – Kinship, Gender and Morality

    • Research on polyamorous parenthood has mainly focused on aspects of interactions with schools and authorities, where polyamorous parents have had to develop strategies of openness and concealment and dealing with stigmatization (Palotta-Chiarolla 2010; Riggs 2010; Sheff 2010). Aspects of sex and morality have primarily been explored in general in relation to non-monogamous practice and not specifically in relation to parenthood (Ritchie 2010; Samuels 2010). Based on interviews with 22 persons in Sweden who in different ways have experience of non-monogamous practice, we explore polyamorous parenthood, focusing on kinship practices, gender and sexual morality. The results of these analyses show that there are recurring themes of promiscuity as a risk that polyamorous parents have to handle. This can take the form of negative interactions with family members and society, practical issues with having sex with several partners or identity issues where good parenthood risks being at odds with non-monogamous practices. The boundaries of good parenthood and promiscuity and the negotiations between them are rarely highlighted in research, but have implications for the moral order of kinship for non-monogamous practice today.

  • Prognostic significance of specific anti-WT1 IgG antibody level in plasma in patients with ovarian carcinoma

    • Ovarian carcinoma (OC) has a poor prognosis and lack early effective screening markers. Wilm's tumor gene 1 (WT1) is overexpressed in OCs. Therefore, it is of great interest to investigate whether WT1-specific antibody (Ab) measurements in plasma can serve as a biomarker of anti-OC response, and is of importance in relation to patient prognosis. Peripheral blood samples were obtained from a total of 103 women with ovarian tumors with median being 1 day (range 0-48 days) before operation. WT1 IgG Ab levels were evaluated using enzyme-linked immunosorbent assay (ELISA). Immunohistochemical analysis of WT1 protein expression was performed on OC tissue samples. We found that low-WT1 Ab level in plasma was related to improved survival in patients diagnosed at stages III-IV and grade 3 carcinomas. Positive WT1 protein staining on OC tissue samples had a negative impact on survival in the entire cohort, both overall survival (OS) (P = 0.046) and progression-free survival (PFS) (P = 0.006), but not in the serous OC subtype. Combining WT1 IgG Ab levels and WT1 staining, patients with high-WT1 IgG Ab levels in plasma and positive WT1 protein staining in cancer tissues had shorter survival, with a significant association in PFS (P = 0.016). These results indicated that WT1 Ab measurements in plasma and WT1 staining in tissue specimens could be useful as biomarkers for patient outcome in the high-risk subtypes of OCs for postoperative individualized therapy.

  • Vaccine failures after active immunisation against tick-borne encephalitis

    • Tick-borne encephalitis (TBE) is a major disease of the central nervous system in Europe and is endemic in Sweden with about 200 notified cases annually. The far most effective protective measure against TBE is active immunisation. The vaccines available today induce a high degree of protection in field studies. However, vaccine failures have occasionally been reported and may be overlooked due to different, and sometimes confusing, antibody kinetics in vaccinees with TBEV infection. In this study, 27 patients with clinical and serological evidences of TBE despite adequate immunisation are presented. Vaccination failure is characterized by a slow, and initially non-detectable, development of the specific TBEV-IgM response, seen together with a rapid rise of IgG and neutralising antibodies in serum. The majority (70%) of the patients were more than 50 years of age, which may implicate a need for a modified immunisation strategy in the elderly.

  • Reduction in WT1 Gene Expression During Early Treatment Predicts the Outcome in Patients With Acute Myeloid Leukemia

    • Wilms tumor gene 1 (WT1) expression has been suggested as an applicable minimal residual disease marker in acute myeloid leukemia (AML). We evaluated the use of this marker in 43 adult AML patients. Quantitative assessment of WT1 gene transcripts was performed using real-time quantitative-polymerase chain reaction assay. Samples from both the peripheral blood and the bone marrow were analyzed at diagnosis and during follow-up. A strong correlation was observed between WT1 normalized with 2 different control genes (beta-actin and ABL1, P < 0.001). WT1 mRNA level at diagnosis was of no prognostic relevance (P > 0.05). A >= 1-log reduction in WT1 expression in bone marrow samples taken < 1 month after diagnosis significantly correlated with an improved overall survival (P = 0.004) and freedom from relapse (P = 0.010) when beta-actin was used as control gene. Furthermore, a reduction in WT1 expression by >= 2 logs in peripheral blood samples taken at a later time point significantly correlated with a better outcome for overall survival (P = 0.004) and freedom from relapse (P = 0.012). This result was achieved when normalizing against both b-actin and ABL1. These results therefore suggest that WT1 gene expression can provide useful information for minimal residual disease detection in adult AML patients and that combined use of control genes can give more informative results.

    • Syftet med denna licentiatuppsats är att pröva en specifik strukturell modell som stöd för elevers utforskande av relationer mellan tal i additiva strukturer även när negativa tal är inkluderade. En intention är att resultatet ska kvalificera undervisningen och utgöra ett stöd vid planering och genomförande av en undervisning avseende ekvationer med additiv struktur utifrån algebraisk undervisning som alternativ till att i första hand finna en lösning med stöd av regler och procedurer. Under studien kartlades, kategoriserades och beskrevs elevernas erfarande av fenomenet relationer mellan kvantiteter. I studien prövades även om, och i sådant fall, på vilka sätt en specifik strukturell modell användes av elever under arbetet med att utforska ekvationers struktur.Elever från årskurserna 3, 8 och 9 deltog i semistrukturerade intervjuer samt i forskningslektioner. Tre grupper av lärare samt två forskare planerade, genomförde och reviderade forskningslektionerna baserat på ansatsen learning study.Fenomenografi var den teoretiska ansatsen för de inledande semistrukturerade intervjuerna. Variationsteori och lärandeverksamhet var de teoretiska ramverken för forskningslektionerna där Davydovs program var en inspirationskälla.För att uppnå syftet formulerades följande forskningsfrågor:Vilka skilda sätt att erfara fenomenet relationer mellan kvantiteter kan urskiljas i elevintervjuer?På vilka sätt använder elever en specifik strukturell modell för att utforska ekvationer?Den första forskningsfrågan besvaras i Artikel 1 som visar att elever erfar relationer mellan kvantiteter som någonting som ska beräknas alternativt någonting som ska relateras. Den andra forskningsfrågan besvaras i Artikel 2 som visar att elever använde sig av den i studien prövade specifika strukturella modellen såsom ett formulär att fylla i alternativt som en lärandemodell och redskap för att identifiera del-helhetsstrukturen mellan tal i en ekvation samt för att välja lämplig operation att lösa ut det obekanta talet.                          Resultaten visar på möjligheten, men även utmaningen, att introducera en algebraisk undervisning med fokus på analys och teoretiska resonemang även för elever med erfarenheter från en alternativ bakgrund.

    • Mycobacterium tuberculosis R2-like ligand-binding oxidase (MtR2lox) belongs to a recently discovered group of proteins that are homologous to the ribonucleotide reductase R2 proteins. MtR2lox carries a heterodinuclear Mn/Fe cofactor and, unlike R2 proteins, a large ligand-binding cavity. A unique tyrosine-valine cross link is also found in the vicinity of the active site. To date, all known structures of R2 and R2lox proteins show a disordered C-terminal segment. Here, we present two new crystal forms of MtR2lox, revealing an ordered helical C-terminal. The ability of alternating between an ordered and disordered state agrees well with bioinformatic analysis of the protein sequence. Interestingly, ordering of the C-terminal helix shields a large positively charged patch on the protein surface, potentially used for interaction with other cellular components. We hypothesize that the dynamic C-terminal segment may be involved in control of protein function in vivo.

    • Chlamydia trachomatis R2c is the prototype for a recently discovered group of ribonucleotide reductase R2 proteins that use a heterodinuclear Mn/Fe redox cofactor for radical generation and storage. Here, we show that the Mycobacterium tuberculosis protein Rv0233, an R2 homologue and a potential virulence factor, contains the heterodinuclear manganese/iron-carboxylate cofactor but displays a drastic remodeling of the R2 protein scaffold into a ligand-binding oxidase. The first structural characterization of the heterodinuclear cofactor shows that the site is highly specific for manganese and iron in their respective positions despite a symmetric arrangement of coordinating residues. In this protein scaffold, the Mn/Fe cofactor supports potent 2-electron oxidations as revealed by an unprecedented tyrosine-valine crosslink in the active site. This wolf in sheep's clothing defines a distinct functional group among R2 homologues and may represent a structural and functional counterpart of the evolutionary ancestor of R2s and bacterial multicomponent monooxygenases.

    • Tuberculosis is a notorious disease responsible for the deaths of 1.4 million people worldwide. A third of the world's population is infected with Mycobacterium tuberculosis, the bacterium causing the disease. The increase of multi drug-resistant strains worsens the situation, and the World Health Organization has declared tuberculosis to be a global emergency. The bacterium envelopes itself with a unique set of very long-chain lipids that play an important role in virulence and drug resistance. Therefore enzymes involved in lipid metabolism are putative drug targets. To allow entry into different metabolic pathways and transmembrane transport, fatty acids have to be activated. This is done primarily by fatty acyl-CoA synthetases (ACSs). We identified an ACS possibly involved in the bacterium’s virulence and solved its structure. Structural interpretation combined with previously reported data gives us insights into the details of its function. This enzyme is known to harbor lipid substrates longer than the enzyme itself, and we now propose how this peripheral membrane protein accommodates its substrates. Some of the most chemically challenging oxidations are performed by dinuclear metalloproteins belonging to the ferritin-like superfamily. We show that the ferritin-like protein, R2lox, from M. tuberculosis contains a new type of heterodinuclear Mn/Fe cofactor. This protein cofactor is capable of performing potent 2-electron oxidations as demonstrated by a novel tyrosine-valine crosslink observed in the protein. Recently a new subclass of ribonucleotide reductase (RNR) R2 proteins, was identified in the intracellular pathogen Chlamydia trachomatis containing the same type of Mn/Fe cofactor mentioned above. The RNR R2 proteins use their metal site to generate a stable radical, essential for the reduction of ribonucleotides to their deoxy forms, the building blocks of DNA. With this work, we were able to characterize the architecture of this metal cofactor.

    • The essential catalytic radical of Class-I ribonucleotide reductase is generated and delivered by protein R2, carrying a dinuclear metal cofactor. A new R2 subclass, R2c, prototyped by the Chlamydia trachomatis protein was recently discovered. This protein carries an oxygen-activating heterodinuclear Mn(II)/Fe(II) metal cofactor and generates a radical-equivalent Mn(IV)/Fe(III) oxidation state of the metal site, as opposed to the tyrosyl radical generated by other R2 subclasses. The metal arrangement of the heterodinuclear cofactor remains unknown. Is the metal positioning specific, and if so, where is which ion located? Here we use X-ray crystallography with anomalous scattering to show that the metal arrangement of this cofactor is specific with the manganese ion occupying metal position 1. This is the position proximal to the tyrosyl radical site in other R2 proteins and consistent with the assumption that the high-valent Mn(IV) species functions as a direct substitute for the tyrosyl radical.

    • The Mycobacterium tuberculosis acid-induced operon MymA encodes the fatty acyl-CoA synthetase FadD13 and is essential for virulence and intracellular growth of the pathogen. Fatty acyl-CoA synthetases activate lipids before entering into the metabolic pathways and are also involved in transmembrane lipid transport. Unlike soluble fatty acyl-CoA synthetases, but like the mammalian integral-membrane very-long-chain acyl-CoA synthetases, FadD13 accepts lipid substrates up to the maximum length tested (C-26). Here, we show that FadD13 is a peripheral membrane protein. The structure and mutational studies reveal an arginine- and aromatic-rich surface patch as the site for membrane interaction. The protein accommodates a hydrophobic tunnel that extends from the active site toward the positive patch and is sealed by an arginine-rich lid-loop at the protein surface. Based on this and previous data, we propose a structural basis for accommodation of lipid substrates longer than the enzyme and transmembrane lipid transport by vectorial CoA-esterification.

    • Wilms’ tumor gene 1 (WT1) is a zinc finger transcriptional regulator with crucial functions in embryonic development. Originally WT1 was described as a tumor suppressor gene, but later studies have shown oncogenic properties of WT1 in a variety of tumors. Because of its dual functions in tumorigenesis, WT1 has been described as a chameleon gene. In this thesis, the significance of WT1 as a biomarker was investigated in acute myeloid leukemia (AML), clear cell renal cell carcinoma (ccRCC), ovarian carcinoma (OC) and childhood B-cell precursor acute lymphoblastic leukemia (BCP-ALL).Previous studies have suggested that expression of WT1 is a potential marker for detection of minimal residual disease (MRD) in AML. We aimed to define expression of WT1 as an MRD marker in AML. In adult AML patients, we found that a reduction of WT1 expression in bone marrow (≥ 1-log) detected less than 1 month after diagnosis was associated with an improved overall survival (OS) and freedom from relapse (FFR). In peripheral blood, a reduction of WT1 expression (≥ 2-log) detected between 1 and 6 months after treatment initiation was associated with an improved OS and FFR.WT1 harbor pathogenic genetic variants in a considerable proportion of AML and T-lymphoblastic leukemia (T-ALL), but mutations have not been reported in BCP-ALL. We aimed to evaluate the clinical impact of WT1 mutations and single nucleotide polymorphisms (SNPs) in BCP-ALL. Pathogenic mutations in the WT1 gene were rarely seen in childhood BCP-ALL. However, five WT1 SNPs were identified. In survival analyses, WT1 SNP rs1799925 was found to be associated with worse OS, indicating that WT1 SNP rs1799925 may be a useful marker for clinical outcome in childhood BCP-ALL. We also explored whether WT1 mutations and SNPs in ccRCC could be used as biomarkers for risk and treatment stratification. We therefore examined whether SNPs or mutations in WT1 were associated with WT1 expression and clinical outcome. Sequencing analysis revealed that none of the previously reported WT1 mutations were found in ccRCC; however, we identified six different WT1 SNPs. Our data suggest that pathogenic WT1 mutations are not involved in ccRCC, and the prognostic significance of WT1 SNPs in ccRCC is considerably weak. However, a favorable OS and disease-specific survival were found in the few cases harboring the homozygous minor allele.OC has a poor prognosis, and early effective screening markers are lacking. Serous OCs are known to express the WT1 protein. Overexpressed oncogenic proteins can be considered potential candidate antigens for cancer vaccines and T-cell therapy. It was therefore of great interest to investigate whether anti-WT1 IgG antibody (Ab) measurements in plasma could serve as biomarkers of anti-OC response. We found limited prognostic impact, but the results indicated that anti-WT1 IgG Ab measurements in plasma and WT1 staining in tissue specimens could be potential biomarkers for patient outcome in the high-risk subtypes of OCs.In conclusion, the results of this thesis indicate that WT1 gene expression can provide information about MRD of patients with AML, and WT1 SNP rs1799925 may be used as a biomarker for predicting clinical outcome in childhood BCP-ALL. In ccRCC, the prognostic significance of WT1 SNPs is weak and limited to the subgroup of patients that are homozygous for the minor allele. In OCs anti-WT1 IgG Ab measurement in plasma and WT1 staining in tissue specimens could possibly be used as biomarkers for predicting patient outcome in the high-risk subtypes of OCs.

    • Polyamory means having a sexual and/or intimate relationship with more than one person at a time. In this study, we use in-depth interviews with 22 persons in Sweden who have experience of polyamorous or non-monogamous relationships to explore how polyamory can include imagining utopian relationships and spaces. Thematic analysis was done which indicated narratives of politically invested attempts to create communal living or societal change that resists capitalist and heteronormative nuclear-family arrangements as well as stories of everyday events that do not explicitly involve political ambitions. The range of utopian dreams and practices of the non-monogamous participants in our study, we argue, are examples of what Muñoz calls concrete utopias, filled with joy and laden with disappointment in the face of potentiality and reality.

    • The aim of this study is to explore the current scientific evidence for using the twelve-step method as a treatment method for sex addiction and compulsive sexual behavior (disorder). Peer-reviewed empirical articles on the twelve-step method and sex addiction and compulsive sexual behavior (disorder) written in English, Danish, Norwegian, or Swedish, retrievable in selected databases were included. No limits were set on publication date or study design. The systematic review resulted in eight empirical studies which were read and assessed according to the Mixed Methods Appraisal Tool. The results were inconclusive, and we found only three articles of high quality, where the samples were composed mainly by men, which indicate that peer-therapy in combination with individual therapy might be beneficial. That twelve-step treatment rests heavily on the idea of sex addiction was unproblematized in most of the publications. Overall, the findings raise issues concerning who benefits from this treatment.

The aim of the study was to investigate the localization of skin tumors after the topical application of ALA, by detecting the PpIX fluorescence either in the spectral or in the time domain.Study Design/Materials and MethodsTwo fluorescence imaging systems were used to identify basal cell carcinomas of the skin in humans, after topical application of 20% ALA ointment. Both systems rely on the comparison between the exogenous and the endogenous fluorescence, performed either in the spectral domain or in the time domain. The first system works by using three images acquired through different spectral filters, whereas the second one measures the spatial map of the average fluorescence lifetime of the sample.ResultsA clear demarcation of skin malignancies was successfully performed in vivo noninvasively with both fluorescence imaging systems.ConclusionThe two complementary approaches considered in the present study show promise for skin tumor detection and delineation based on specific fluorescence features.     • In this work, an on-line process for pressurised hot water extraction (PHWE) of antioxidants from plants as well as drying of the extract in one step by particle formation based on the use of supercritical carbon dioxide (SC-CO2) has been developed. This process has been called WEPO (R), water extraction and particle formation on-line. With this process, dried extracts from onion with the same composition of quercetin derivatives as non-dried extracts have been obtained as a fine powder with spherical particles from 250 nm to 4 mu m in diameter. The major compounds present in the extract were quercetin-3,4'-diglucoside, quercetin-4'-glucoside and quercetin. An auxiliary inert gas (hot N-2) was used to enhance the drying process. Parameters such as temperature (120 degrees C), SC-CO2 and N-2 pressures (80 and 12.5 bar, respectively) and flow rate of SC-CO2 (10 ml/min), have been settled by trial-and-error in order to achieve a fine and constant spray formation. Water content, size and morphology, antioxidant capacity and quercetin content of the particles were studied to evaluate the efficiency of the WEPO process. Results were compared with the ones from extracts obtained by continuous flow PHWE followed by freeze-drying. Results showed that both processes gave similar results in terms of antioxidant capacity, concentration of quercetin derivatives and water content, while only WEPO was able to produce defined spherical particles smaller than 4 mu m.     • Recent geopolitical and economic crises underline the need for a European transition towards a more sustainable food system. Scholars and policymakers have called for a re-territorialization of food production to strike a better balance between local, regional and global value chains. This paper explores the role of re-territorialization through an analysis of the emergence, development and current transformation of the Swedish wild berry value chain. The analysis combines the multi-level perspective on sustainability transitions with a socio-techno-ecological system approach and draws on interviews, informal conversations, participant observations and a range of secondary sources. The resulting case narrative shows how processes of de-territorialization may result in regimes that fail to address sustainability potential and problems. It also highlights that processes of re-territorialization challenge established regimes by promoting niches that represent different, albeit complementary, value chain configurations. Apart from a rich empirical narrative that brings useful knowledge to stakeholders to the Swedish wild berry value chain, the paper contributes to the theoretical understanding re-territorialization, shows how the ecological dimension can be accounted for with the multi-level perspective on sustainability transitions and presents a number of general policy implications.

    • During spring 1998 the course Industrial Case Studies in Control was given at the Department of Automatic Control in Lund. The course consisted of a project at an industry with control applications. This report considers a project at the crude alcohol distillery in Nöbbelöv, where the base product for the liquor Absolut Vodka is produced. The distillery had problems with fluctuations in the concentration of ethanol in the crude alcohol. The purpose of the project was to analyze the cause of the fluctuations, and if possibly eliminate it. The fluctuations were cause by an output limiter on a controller, and were relatively easily eliminated. To examine if control performance could be further improved, a series of experiments was carried out on the plant. The result of the experiments were then used to derive a simple process model, and a suggestion for new controller parameters.

    • A system consisting of a high-pressure tolerant microfluidic glass chip, high-speed absorbance imaging, and image processing has been developed to study rapid dynamic events like pH change in a multiphase flow. The system gives both kinetic and quantitative equilibrated information. By tracking the interactions of aqueous additive mixtures and liquid CO2, at 80 bar and 24 °C, under flow, measurement at a given P, T condition is done in 0.25 s. The acidification rate to steady state was found to be mass transport limited, occurring in less than 1 s. For 30 mM of the additives ammonium acetate and ammonium formate, equilibrium pH of 4.5 and 4.1, respectively, was seen. These additives are of key importance in common mobile phases used in SFC.

    • Preparedness for bioterrorism is based on communication between people in organizations who are educated and trained in several disciplines, including law enforcement, health, and science. Various backgrounds, cultures, and vocabularies generate difficulties in understanding and interpretating terms and concepts, which may impair communication. This is especially true in emergency situations, in which the need for clarity and consistency is vital. The EU project AniBioThreat initiated methods and made a rough estimate of the terms and concepts that are crucial for an incident, and a pilot database with key terms and definitions has been constructed. Analysis of collected terms and sources has shown that many of the participating organizations use various international standards in their area of expertise. The same term often represents different concepts in the standards from different sectors, or, alternatively, different terms were used to represent the same or similar concepts. The use of conflicting terminology can be problematic for decision makers and communicators in planning and prevention or when handling an incident. Since the CBRN area has roots in multiple disciplines, each with its own evolving terminology, it may not be realistic to achieve unequivocal communication through a standardized vocabulary and joint definitions for words from common language. We suggest that a communication strategy should include awareness of alternative definitions and ontologies and the ability to talk and write without relying on the implicit knowledge underlying specialized jargon. Consequently, cross-disciplinary communication skills should be part of training of personnel in the CBRN field. In addition, a searchable repository of terms and definitions from relevant organizations and authorities would be a valuable addition to existing glossaries for improving awareness concerning bioterrorism prevention planning. © 2013, Mary Ann Liebert, Inc.

    • Antibiotic resistance is currently an extensive medical challenge worldwide, with global numbers increasing steadily. Recent data have highlighted wastewater treatment plants as a reservoir of resistance genes. The impact of these findings for human health can best be summarized using a One Health concept. However, the molecular mechanisms impacting resistance spread have not been carefully evaluated. Bacterial viruses, that is bacteriophages, have recently been shown to be important mediators of bacterial resistance genes in environmental milieus and are transferrable to human pathogens. Herein, we investigated the biogeographical impact on resistance spread through river-borne bacteriophages using amplicon deep sequencing of the microbiota, absolute quantification of resistance genes using ddPCR, and phage induction capacity within wastewater. Microbial biodiversity of the rivers is significantly affected by river site, surrounding milieu and time of sampling. Furthermore, areas of land associated with agriculture had a significantly higher ability to induce bacteriophages carrying antibiotic resistance genes, indicating their impact on resistance spread. It is imperative that we continue to analyse global antibiotic resistance problem from a One Health perspective to gain novel insights into mechanisms of resistance spread.

    • Recent advancements in catchment hydrology (such as understanding hydrological processes, accessing new data sources, and refining methods for parameter constraints) make it possible to apply catchment models for ungauged basins over large domains. Here we present a cutting-edge case study applying catchment-modelling techniques at the global scale for the first time. The modelling procedure was challenging but doable and even the first model version show better performance than traditional gridded global models of river flow. We used the open-source code of the HYPE model and applied it for > 130 000 catchments (with an average resolution of 1000 km2), delineated to cover the Earths landmass (except Antarctica). The catchments were characterized using 20 open databases on physiographical variables, to account for spatial and temporal variability of the global freshwater resources, based on exchange with the atmosphere (e.g. precipitation and evapotranspiration) and related budgets in all compartments of the land (e.g. soil, rivers, lakes, glaciers, and floodplains), including water stocks, residence times, interfacial fluxes, and the pathways between various compartments. Global parameter values were estimated using a step-wise approach for groups of parameters regulating specific processes and catchment characteristics in representative gauged catchments. Daily time-series (> 10 years) from 5338 gauges of river flow across the globe were used for model evaluation (half for calibration and half for independent validation), resulting in an average monthly KGE of 0.4. However, the world-wide HYPE (WWH) model shows large variation in model performance, both between geographical domains and between various flow signatures. The model performs best in Eastern USA, Europe, South-East Asia, and Japan, as well as in parts of Russia, Canada, and South America. The model shows overall good potential to capture flow signatures of monthly high flows, spatial variability of high flows, duration of low flows and constancy of daily flow. Nevertheless, there remains large potential for model improvements and we suggest both redoing the calibration and reconsidering parts of the model structure for the next WWH version. The calibration cycle should be repeated a couple of times to find robust values under new fixed parameter conditions. For the next iteration, special focus will be given to precipitation, evapotranspiration, soil storage, and dynamics from hydrological features, such as lakes, reservoirs, glaciers, and floodplains. This first model version clearly indicates challenges in large scale modelling, usefulness of open data and current gaps in processes understanding. Parts of the WWH can be shared with other modellers working at the regional scale to appreciate local knowledge, establish a critical mass of experts and improve the model in a collaborative manner. Setting up a global catchment model has to be a long-term commitment of continuous model refinements to achieve successful and truly useful results.

    • A MeV proton beam with a current in the range of fA-nA is usually measured using a secondary beam signal that is dependent on a sample or requires noise-free accurate charge integration in a Faraday cup. We propose a simple on-line beam current measurement setup that can be used to directly measure beam current during ion beam analysis. A fast beam blanker is used to modulate the beam position after the last set of collimator slits in and out of a mini Faraday cup connected to fA-sensitive electrometer. Less than 100 fA current can be measured during an undisturbed irradiation with kHz sampling frequency. The fast, simple and easily controlled current measurement set-up is going to be routinely used for ion beam analysis and modification at the new Lund sub-micron beam line. (c) 2006 Elsevier B.V.     • Antihypertensive treatment is equally beneficial for reducing cardiovascular risk in both men and women. Despite this, the drug treatment, prevalence and control of hypertension differ between men and women. Men and women respond differently, particularly with respect to the risk of adverse events, to many antihypertensive drugs. Certain antihypertensive drugs may also be especially beneficial in the setting of certain comorbidities - of both cardiovascular and extracardiac nature - which also differ between men and women. Furthermore, hypertension in pregnancy can pose a considerable therapeutic challenge for women and their physicians in primary care. In addition, data from population-based studies and from real-world data are inconsistent regarding whether men or women attain hypertension-related goals to a higher degree. In population-based studies, women with hypertension have higher rates of treatment and controlled blood pressure than men, whereas real-world, primary-care data instead show better blood pressure control in men. Men and women are also treated with different antihypertensive drugs: women use more thiazide diuretics and men use more angiotensin-enzyme inhibitors and calcium-channel blockers. This narrative review explores these sex-related differences with guidance from current literature. It also features original data from a large, Swedish primary-care register, which showed that blood pressure control was better in women than men until they reached their late sixties, after which the situation was reversed. This age-related decrease in blood pressure control in women was not, however, accompanied by a proportional increase in use of antihypertensive drugs and female sex was a significant predictor of less intensive antihypertensive treatment.

    • Denna vägledning förmedlar Trafikverkets syn på hur de båda processerna miljöbedömning och miljöbeskrivning bör tillämpas i planläggning och projektering av väg- och järnvägsprojekt, med utgångspunkt från lagkrav, praxis och Trafikverkets erfarenheter. Den tar även upp innehållet i de båda dokumenten miljökonsekvensbeskrivning och miljöbeskrivning som kommer ut av de båda processerna. Vägledningen syftar till att bidra till integrering av miljöaspekter i planläggning och projektering av vägar och järnvägar samt till en god kvalitet på genomförande och redovisning. Den syftar även till att miljöbedömningar och miljöbeskrivningar ska genomföras på ett likartat sätt i hela Trafikverket och få en jämn kvalitet. Vägledningen riktar sig främst till Trafikverkets konsulter och egna miljöspecialister som gör miljöbedömningar och miljöbeskrivningar i Trafikverkets väg- och järnvägsprojekt. Vägledningen ersätter Trafikverkets Handbok om metodik för miljökonsekvensbeskrivning för vägar och järnvägar (Trafikverket 2011:090). Den ersätter också texter om MKB och miljöbeskrivning i Trafikverkets Rapport planläggning av vägar och järnvägar. Vägledningen innehåller bland annat:relevant lagstiftning och målbeskrivning om hur miljö integreras samt miljöbedömningens roll och moment i planläggningens olika skedensamråd, samrådsunderlag, utredning om betydande miljöpåverkan och miljökonsekvensbeskrivningens (MKB) innehåll Trafikverkets syn på hantering av miljöaspekter, behov av att koppla dessa till miljöintressen för vilka konsekvenser beskrivs

    • Anthropogenic underwater noise is a global pollutant of increasing concern but its impact on reproduction in fish is largely unknown. Hence, a better understanding of its consequences for this important link to fitness is crucial. Working in aquaria, we experimentally tested the impact of broadband noise exposure (added either continuously or intermittently), compared to a control, on the behaviour and reproductive success of the common goby (Pomatoschistus microps), a vocal fish with exclusive paternal care. Compared to the intermittent noise and control treatments, the continuous noise treatment increased latency to female nest inspection and spawning and decreased spawning probability. In contrast, many other female and male pre-spawning behaviours, and female ventilation rate (proxies for stress levels) did not differ among treatments. Therefore, it is likely that female spawning decisions were delayed by a reduced ability to assess male acoustic signals, rather than due to stress per se and that the silent periods in the intermittent noise treatment provided a respite where the females could assess the males. Taken together, we show that noise (of similar frequency range as anthropogenic boat noise) negatively affects reproductive success, particularly under a continuous noise exposure.

    • The known post-glacial isostatic land rise rates at Fårö Island, Central Baltic Sea, allowed for the reliable estimation of frost weathering rates of limestone clasts along five beach profiles. Weathering rates measured by clast dimension change were quantified over periods ranging between 640 and 2090 years. While the deposited beach materials at each profile have varying lithostratigraphic compositions these attributes appear to have no direct influence on the long-term comminution rates. Mean clast size of initially deposited beach materials is suggested to be primarily a function of local wave energy and sourced from local rock platforms. Field observations, climate data and literature point to the beach materials in the upper 10–20 cm being fractured under an active diurnal to short-term frost environment that has persisted over the past 1000–2000 years. Pore water freezing by 9% volumetric expansion and the development of pore water expulsion and hydrofracture is suggested to increase pore volume and pore interconnectedness over time and facilitate ice segregation growth. Observed modes of clast fracture, bursting, flaking and granular disaggregation suggest different frost weathering mechanisms to operate on different limestone lithologies. Long term rates of break down appear however not differentiated by limestone composition.

    • Previous studies on Internet-based treatment with minimal to moderate therapist guidance have shown promising results for a number of specific diagnoses. The aim of this study was to test a new approach to Internet treatment that involves tailoring the treatment according to the patients unique characteristics and comorbidities. A total of 54 participants, regardless of specific anxiety diagnosis, were included after an in-person, semi-structured diagnostic interview and randomized to a 10 week treatment program or to a control group. Treatment consisted of a number of individually-prescribed modules in conjunction with online therapist guidance. Significant results were found for all dependent measures both immediately following treatment and at 1 and 2 year intervals. Mean between-group effect size including measures of anxiety, depression and quality of life was Cohens d = 0.69 at post-treatment, while the mean within-group effect size was d = 1.15 at post-treatment and d = 1.13 and d = 1.04 at 1 and 2 year follow-up respectively. The tentative conclusion drawn from these results is that tailoring the Internet-based therapy can be a feasible approach in the treatment of anxiety in a homogeneous population.

  • LGBTQ plus Affirmative State Care for Young People in Sweden : New Knowledge and Old Traditions

    • Young LGBTQ+ people are over-represented in various forms of state care. They experience hardships during their placements and staff competence in addressing specific needs among LGBTQ+ youth is lacking. In this article, we investigate whether and how LGTBQ+ issues are considered and described in digital marketing for state care providers. The material consists of the homepages of residential care homes and secure state care institutions, which we analyse using critical discourse analysis. The results show that LGBTQ+ issues are largely invisible. Of the approximately 1,000 existing state care providers, only twenty stated that they worked with or had competence in LGBTQ+ issues. Among these, no secure state care institution offered LGBTQ+ competence at the time of the study. The descriptions of how care providers work with LGBTQ+ issues are characterised by heteronormativity where there is a mix of two types of language on the homepages regarding LGBTQ+ youth; on the one hand, a heteronormative, traditional description based on a binary understanding of gender; and, on the other, an LGBTQ+ inclusive language is used. However, the LGBTQ+ affirmative language has been imposed upon the traditional rather than being integrated into it, which comes across as superficial and unclear. International studies show that LGBTQ+ youth are over-represented in various forms of state care. Using critical discourse analysis, we investigated how LGBTQ+ youth, as a target group, are described and how LGBTQ+ competence is presented on the home pages of residential care homes and secure state care institutions for young people in Sweden. The results show that LGBTQ is largely invisible. Of the approximately 1,000 existing residential care homes, only 20 stated that they worked with or had competence in LGBTQ+ issues. No secure state care institution offered LGBTQ+ competence at the time of the study.

    • This study investigates experiences of polyamory in a society where monogamy is the norm. Polyamory is when more than two people are involved in an intimate and/or sexual relationship. The relationships are known to those involved, and everyone has the opportunity to have multiple relationships at the same time. In-depth interviews were completed with 22 persons in Sweden who identify as polyamorous. Drawing on Ahmed’s phenomenological concepts of turning points and lines and Halberstam’s concept of queer time and temporality, the following questions are explored: What turning points can be seen in the informants’ stories? And what consequences are the informants exposed to when heteronormative expectations are not followed? In the theoretical language of Ahmed, living a life within monogamous boundaries would be considered as being “in line”. Going beyond these monogamous heteronormative lines can result in more relational choices by which one has to find out what kind of relationship works best instead of following a ready-made template. The majority of the informants feel forced to conceal their relationship constellations in several situations and contexts. Living a queer life is seen by others in society as not only incomprehensible but also immature and inexperienced. Interactions with healthcare professionals seldom offer any relief from this; instead, the informants’ stories of these encounters can be interpreted as instances of being stopped and blocked, resulting in stress and shame.

    • Based on interviews and ethnographic fieldwork within BDSM communities in Sweden, this article focuses on links between non-monogamy and BDSM. Drawing on Halberstam´s concept of queer space, the following questions are investigated: What are the connections between BDSM and non-monogamous communities? How does interaction between BDSM and non-monogamous practices create non-normative logic? The transgression of one norm makes it easier to transgress other norms as well, providing opportunities to find new ways of organizing relationships beyond the norms of monogamy. In order for an individual to be able to fully explore kinks in BDSM practices and at the same time respect the boundaries of a partner, nonmonogamy emerges as one logical answer.

    • The adult heart develops hypertrophy to reduce ventricular wall stress and maintain cardiac function in response to an increased workload. Although pathological hypertrophy generally prog-resses to heart failure, physiological hypertrophy may be car-dioprotective. Cardiac-specific overexpression of the lipid-droplet protein perilipin 5 (Plin5) promotes cardiac hypertrophy, but it is unclear whether this response is beneficial. We analyzed RNA -sequencing data from human left ventricle and showed that car-diac PLIN5 expression correlates with up-regulation of cardiac contraction-related processes. To investigate how elevated cardiac Plin5 levels affect cardiac contractility, we generated mice with cardiac-specific overexpression of Plin5 (MHC-Plin5 mice). These mice displayed increased left ventricular mass and cardiomyocyte size but preserved heart function. Quantitative proteomics identified sarcoplasmic/endoplasmic reticulum Ca2+ ATPase 2 (SERCA2) as a Plin5-interacting protein. In situ proximity ligation assay further confirmed the Plin5/SERCA2 interaction. Live imaging showed in-creases in intracellular Ca2+ release during contraction, Ca2+ removal during relaxation, and SERCA2 function in MHC-Plin5 versus WT cardiomyocytes. These results identify a role of Plin5 in improving cardiac contractility through enhanced Ca2+ signaling.

    • CO2-expanded liquids (CXLs) is a class of solvent systems giving relatively low viscosity in comparison to neat organic solvents. The challenge is to achieve a high overall polarity of the CXL, despite the presence of compressed liquid CO2. In this study, the relative static permittivity (εr) was measured for binary and ternary one-phase mixtures of CO2 + green solvent using an in-line microfluidic device. In addition, Kamlet-Taft solvatochromic parameters were experimentally determined, allowing the characterization of the polarizability, acidity, and basicity. Novel data is shown for binary and ternary systems of CO2 expanded ethanol and ethyl lactate, with or without glycerol or water added, at moderate conditions of pressure and temperature (8 MPa and 35 °C). One-phase CXLs systems of a wide relative permittivity range (7.7–75.3) at lowered viscosity were enabled, by mixing different green solvents with CO2.

    • Background: Injury events in homes constitute a major social problem. Falling, cutting and poisoning make up 85 per cent of all injury events in residential areas.This study is based on a unique data set that includes several million cases of falling, cutting and poisoning in Sweden during the period 1990-2013 which lead to either to death, hospitalisation or to a visit to a health care provider, and a control group randomly selected from the population.Three riskgroups are given special attention in the analysis: (1) old people, (2) children, (3) persons with disabilities and or long term illnesses.Methods: Multilevel regression analysis and geographical information systems, GIS.Results: The results show the probability for each riskgroup to be exposed to each type of injury event, and how this probability varies with place (GIS), previous exposure, type of household, socioeconomic status and type of housing.Conclusions: The project is ongoing. Our cross-sectorial group has demonstrated the importance of injury epidemiology as a guiding principle in architectural design, particularly for high-risk groups.
